Hemocyanin-derived phenoloxidase reaction products display anti-infective properties

Hemocyanin is a multi-functional protein located in the hemolymph (blood) of certain arthropods and molluscs. In addition to its well-defined role in oxygen transport, hemocyanin can be converted into a phenoloxidase-like enzyme. Herein, we tested the antimicrobial properties of horseshoe crab (Limulus polyphemus) hemocyanin-derived phenoloxidase reaction products using broad ranges of phenolic substrates (e.g. l-DOPA) and microbial targets (Gram-positive/negative bacteria, yeast). The enzyme-catalysed turnover of several substrates generated (by)products that reduced significantly the number of colony forming units. Microbicidal effects of hemocyanin-derived phenoloxidase were thwarted by the inhibitor phenylthiourea. Data presented here further support a role for hemocyanin in invertebrate innate immunity.

Hemocyanin-derived phenoloxidase activity is dependent on dodecameric structure in shrimp Litopenaeus vannamei

Hemocyanin (Hc) is a multifunctional protein in both mollusks and arthropods. Phenoloxidase (PO) activities are the most important physiological functions for Hcs after conversion. In shrimp, Hc occurs as two oligomer forms, dodecamers and hexamers. Differences in the transport oxygen capacity and agglutination activity between the two oligomers of shrimp Hc have been found. In the present study, we investigated the differences in the Hc-derived PO activity between the dodecameric and hexameric Hc forms of the shrimp Litopenaeus vannamei. The two oligomers were separated by non-denaturing polyacrylamide gel electrophoresis, converted by trypsin cleavage and their PO activities were determined by oxidation of L-DOPA. The dodecamers exhibited PO activity after enzymatic conversion while the hexamers did not exhibit PO activity. This result provides new insight into the structural/functional relationships of Hcs.

Effects of phosphatidyl serine on immune response in the shrimp Litopenaeus vannamei

Abstract Phosphatidyl serine plays an important role in animal innate immunity. Given its important functions, numerous investigations have been carried out on its immunological function in many animals. However, studies of phosphatidyl serine in the white shrimp Litopenaeus vannamei, an economically important animal, are rare. In this paper, we demonstrated influences of injecting phosphatidyl serine (PS) on immune response including some parameters from pro-phenol oxidase activating system (pro-PO system) and hemocyanin-derived phenol oxidase activity (Hd-PO) along with antibacterial and bacteriolytic activities in the white shrimp Litopenaeus vannamei with different PS concentrations (5, 10 and 20 μg mL−1). The results showed that PS could affect immune response of L. vannamei significantly (P<0.05), including total hemocyte counts (THC), PO activity from hemocyte, phenol oxidase (PO) activity from plasma, hemocyanin concentration, Hd-PO activity as well as antibacterial and bacteriolytic activities in the plasma. Among the lines, 20 μg mL−1 PS had the strongest effect on the above parameters, whereas 5 μg mL−1 had the least effect. The experimental results indicated that PS was able to activate exocytosis of pro-PO and formation of Hd-PO in white shrimp after injection, further regulating the immune process reflected by variation of antibacterial and bacteriolytic activities in a certain way.

A putative link between phagocytosis-induced apoptosis and hemocyanin-derived phenoloxidase activation

Apoptosis and phagocytosis are crucial processes required for developmental morphogenesis, pathogen deterrence and immunomodulation in metazoans. We present data showing that amebocytes of the chelicerate, Limulus polyphemus, undergo phagocytosis-induced cell death after ingesting spores of the fungus, Beauveria bassiana, in vitro. The observed biochemical and morphological modifications associated with dying amebocytes are congruent with the hallmarks of apoptosis, including: extracellularisation of phosphatidylserine, intranucleosomal DNA fragmentation and an increase in caspase 3/7-like activities. Previous studies have demonstrated that phosphatidylserine is a putative endogenous activator of hemocyanin-derived phenoloxidase, inducing conformational changes that permit phenolic substrate access to the active site. Here, we observed extracellular hemocyanin-derived phenoloxidase activity levels increase in the presence of apoptotic amebocytes. Enzyme activity induced by phosphatidylserine or apoptotic amebocytes was reduced completely upon incubation with the phosphatidylserine binding protein, annexin V. We propose that phosphatidylserine redistributed to the outer plasma membrane of amebocytes undergoing phagocytosis-induced apoptosis could interact with hemocyanin, thus facilitating its conversion into a phenoloxidase-like enzyme, during immune challenge.

Hemocyanin-derived phenoloxidase activity: A contributing factor to hyperpigmentation in Nephrops norvegicus

The phenomenon of hyperpigmentation (melanosis) in shellfish has long been attributed to phenoloxidase enzymes. Over the last number of years, the oxygen carrier hemocyanin, has demonstrated several immune- and physiological functionalities, most notably, inducible phenoloxidase activity. In this study, hemocyanin purified from the hemolymph of Nephrops norvegicus displays diphenoloxidase activity in the presence of a number of elicitors and retains structural and functional integrity throughout the process of freeze–thawing (at −25°C). Conversely, cellular phenoloxidase activity (present in cell-lysates), demonstrates >98% reduction in activity after freeze–thawing. We present evidence that hemocyanin may act as a causative agent of hyperpigmentation in N. norvegicus. The inhibition of hemocyanin-derived phenoloxidase activity is discussed, and for the first time, the biophysical interactions of shellfish hemocyanin with known phenoloxidase inhibitors are presented.

Hemocyanin synthesis and hemocyanin-derived phenol oxidase in white shrimp(Litopenaeus vannamei)injected with phosphatidyl serine

In order to investigate the effects of injecting phosphatidyl serine(PS)on hemocyanin synthesis and hemocyanin-derived phenol oxidase(Hd-PO)and probe into the pathway and activating mechanism of PS as a possible native elicitor for Hd-PO in white shrimp(Litopenaeus vannamei),we injected adult white shrimps different PS with 5,10,20 μg/mL,and assayed hemocyanin concentration in plasma,expression of hemocyanin subunits(p75 and p77)and mRNA in hepatopancreas,along with Hd-PO activity in plasma from white shrimp.They were investigated by spectrophotometry,native-PAGE,SDS-PAGE and RT-PCR in vivo with different sampling time at 0,6,12,24,36,48 and 60 h respectively,and we set the control group and parallel group.The results showed that hemocyanin concentration declined slightly during 6 h after injecting PS and afterwards changed as a peak by 36th h after injection with a maximum at 24th h(P0.05);expression of hemocyanin subunits and mRNA all had peak changes during 36 h after injecting PS,and likewise,all reaching their ceiling values at 12th h(P0.05);Hd-PO activity had a peak change within 36 h after injecting PS,with its climax at 12th h(P0.05).The research revealed that PS could activate hemocyanin to show phenol oxidase activity with significant time-dose effects,and meanwhile brought down hemocyanin concentration in a short time at first and subsequently started the hemocyanin synthesis,indicating that PS is the elicitor of Hd-PO in white shrimp,which lays the theoretical foundation for the study on immunological function of hemocyanin in shrimp.

Effects of known phenoloxidase inhibitors on hemocyanin-derived phenoloxidase from Limulus polyphemus

Inhibitors of phenoloxidase are used routinely to characterise the structural and functional properties of phenoloxidases. Hemocyanin-derived phenoloxidase activity is also sensitive to standard phenoloxidase inhibitors. In this study, we characterise the effects of a number of phenoloxidase inhibitors on hemocyanin-derived phenoloxidase activity from the chelicerate, Limulus polyphemus. Both inhibition type and K(i) values were similar to those observed for hemocyanin-derived phenoloxidase from another chelicerate, Eurypelma californicum. In addition, substrate inhibition was observed at concentrations above 2mM dopamine. The conformation in which two of the inhibitors, namely tropolone and kojic acid, would bind near the Cu(II) centre of hemocyanin is proposed.

Hemocyanin-derived phenoloxidase activity with broad temperature stability extending into the cold environment in hemocytes of the hair crab Erimacrus isenbeckii

Phenoloxidase (PO) activity is a major component of the innate immune response in arthropods. In this study, we characterized PO activity from the hair crab Erimacrus isenbeckii, which inhabits very cold regions (2.4-3.4°C) of the Bering Sea. Hemocyte lysate supernatant (HLS) prepared from E. isenbeckii was inactive HLS until activated by nonspecific agents such as sodium dodecyl sulfate and trypsin, and elicitors such as lipopolysaccharide and lipoteichoic acid from the cell wall constituent of bacteria. The PO activity was maximal at 4°C, decreased slightly at temperatures up to 60°C, and fell rapidly at 80°C. Both L-DOPA and catechol were efficient substrates for the PO (EC 1.10.3.1), with K(m) values of 0.96 and 1.15mM, respectively, whereas tyrosine and hydroquinone were not. We isolated a protein fraction from HLS as a hexamer of 75kDa units with 216.7-fold higher PO activity than that of the HLS. The N-terminal amino acid analysis of an isolated protein revealed 80% sequence identity to hemocyanins from other crabs. These results suggest that cold-adapted hemocyanin-derived PO activity is important to the survival of these crabs. This is the first report of a crab PO activity with broad temperature stability extending into the cold environment.

Possible role of phosphatidylserine–hemocyanin interaction in the innate immune response of Limulus polyphemus

Phenoloxidase enzymes and the associated pro-phenoloxidase activation cascade play an essential role in the immune response of arthropods. Phenoloxidase activity can be elicited in the oxygen carrier, hemocyanin, by the addition of the artificial inducer, SDS. There is some evidence to support hemocyanin acting as a phenoloxidase in vivo; however, the identity of natural activators remains unclear. This study explores the role of the phospholipid, phosphatidylserine, as a possible natural activator of hemocyanin-derived phenoloxidase activity. Characterisation of the structural changes associated with activation of hemocyanin-derived phenoloxidase suggests that phosphatidylserine induces similar conformational changes to those caused by the artificial inducer, SDS. We propose that anionic phospholipids, in particular phosphatidylserine, may act as natural activators of hemocyanin-derived phenoloxidase.

Identification and characterization of a hemocyanin-derived phenoloxidase from the crab Charybdis japonica

To determine effective activators of crab hemocyanin (Hc) and the properties of Hc-derived phenoloxidase (HdPO), Hc, for the first time, was purified from hemolymph of Charybdis japonica, and the properties of activated HdPO were studied by using L-DOPA as a substrate. Three distinct subunits were isolated, and each had a molecular mass of about 80, 75 and 70 kDa, respectively. SDS and HLS were much effective in conversion of Hc into HdPO whose PO activity was optimal at pH 7.0 and temperature of 40 degrees C. The Km value of the HdPO was 2.90 mM for L-DOPA and 7.33 mM for tyrosine. The PO activity of HdPO was most sensitive to 1-phenyl-2-thiourea, cysteine and ascorbic acid, and much sensitive to thio urea and sodium sulfite. Based on its inhibition characteristics and the substrate specificity, this HdPO could be classified as a kind of tyrosinase-type phenoloxidase. The PO activity of HdPO was also strongly inhibited by Cu(2+), Zn(2+), ethylenediaminetetraacetic acid (EDTA) and diethyldithiocarbamate (DETC). The results with EDTA, DETC, and some metal ions, combined with the perfect recovery effect of Cu(2+) on DETC-inhibited PO activity, indicate that the HdPO is a kind of copper-containing metalloenzyme. All these imply that the Hc, as an oxygen carrier, can be activated to have PO activities by SDS or HLS, and the activated HdPO has the properties of a tyrosinase-type copper-containing phenoloxidase. This study makes us to understand more easily the multifunctions of crustacean Hc in oxygen carrier and melaninization at certain stresses in host defence as well.

Hemocyanin-derived phenoloxidase activity in the spiny lobster Panulirus argus (Latreille, 1804)

Hemocyanin and phenoloxidase belong to the type-3 copper protein family, sharing a similar active center whereas performing different roles. In this study, we demonstrated that purified hemocyanin (450 kDa) from the spiny lobster Panulirus argus shows phenoloxidase activity in vitro after treatment with trypsin, chymotrypsin and SDS (0.1% optimal concentration), but it is not activated by sodium perchlorate or isopropanol. The optimal pHs of the SDS-activated hemocyanin were 5.5 and 7.0. Hemocyanin from spiny lobster behaves as a catecholoxidase. Kinetic characterization using dopamine, L-DOPA and catechol shows that dopamine is the most specific substrate. Catechol and dopamine produced substrate inhibition above 16 and 2 mM respectively. Mechanism-based inhibition was also evidenced for the three substrates, being less significant for L-DOPA. SDS-activated phenoloxidase activity is produced by the hexameric hemocyanin. Zymographic analysis demonstrated that incubation of native hemocyanin with trypsin and chymotrypsin, produced bands of 170 and 190 kDa respectively, with intense phenoloxidase activity. Three polypeptide chains of 77, 80 and 89 kDa of hemocyanin monomers were identified by SDS-PAGE. Monomers did not show phenoloxidase activity induced by SDS or partial proteolysis.

A 160‐kDa Protein Is Essential for Hemocyanin‐derived Melanosis of Prawn

ABSTRACT A purified 160‐kDa protein, isolated from the cuticle of kurama prawn and named melanosis collaborating factor (MCF), was a key factor in promoting melanosis by a cooperative reaction with hemocyanin‐derived phenoloxidase but hemocyanin itself was incapable of producing black pigment. The enzymatic activity of MCF in the body of the prawn was very stable against the process of freezing and thawing; the enzyme maintained more than 80% of its activity after 3 mo of storage at −2 5 °C. These results indicated that MCF, acting in conjunction with hemocyanin‐derived phenoloxidase, played a crucial role in postharvest melanogenesis in prawn.
