Comparison of High n-3 PUFA Levels and Cyclic Heat Stress Effects on Carcass Characteristics, Meat Quality, and Oxidative Stability of Breast Meat of Broilers Fed Low- and High-Antioxidant Diets

This study investigated the individual and combined effects of a high dietary n-3 PUFA intake and cyclic heat stress (HS) on the carcass characteristics, meat quality, and oxidative stability of broiler breast meat and the potential of antioxidant supplementation (vitamins E, C, and selenium) to mitigate these effects. A total of 192 one-day-old male Ross 308 broilers were randomly assigned to 24 pens within two controlled environment chambers and fed with the following diets: a basal diet low in antioxidants according to NRC recommendations (NRC group), a basal diet according to Aviagen recommendations additionally supplemented with 200 IU/kg vitamin E, 250 mg/kg vitamin C, and 0.15 mg/kg selenium (HAOX group), and these two diets further supplemented with 5% linseed oil (NRC N-3 and HAOX N-3 groups). On day 22, the broilers were exposed to the following two environmental conditions: thermoneutral (TN, 21 °C) or cyclic HS (HS, 34 ± 1 °C, 7 h/d) in a 2 × 2 × 2 factorial design. A high intake of n-3 PUFAs significantly decreased growth performance, dressing percentage, and breast yield, while the incidence of pale, soft, and exudative (PSE) meat characteristics and malondialdehyde (MDA) levels increased. Cyclic HS reduced body weight (BW) and average daily feed intake (ADFI), but had limited effects on meat quality. No interactions between n-3 PUFAs and HS were observed for any measurements. High antioxidant supplementation increased breast yield, improved meat quality, and reduced oxidative stress, as evidenced by an enhanced antioxidant activity and lower MDA levels. In conclusion, n-3 PUFAs had a negative effect on both the carcass characteristics and meat quality of broilers, while HS primarily affected only carcass characteristics, with neither stressor having severe adverse effects. High levels of antioxidants could mitigate the negative effects of dietary- and heat-induced oxidative stress by enhancing the oxidative stability of broiler meat.

Effects of Heat-Induced Oxidative Stress and Astaxanthin on the NF-kB, NFE2L2 and PPARα Transcription Factors and Cytoprotective Capacity in the Thymus of Broilers.

The thymus, a central lymphoid organ in animals, serves as the site for T cell development, differentiation and maturation, vital to adaptive immunity. The thymus is critical for maintaining tissue homeostasis to protect against tumors and tissue damage. An overactive or prolonged immune response can lead to oxidative stress from increased production of reactive oxygen species. Heat stress induces oxidative stress and overwhelms the natural antioxidant defense mechanisms. This study's objectives were to investigate the protective properties of astaxanthin against heat-induced oxidative stress and apoptosis in the chicken thymus, by comparing the growth performance and gene signaling pathways among three groups: thermal neutral, heat stress, and heat stress with astaxanthin. The thermal neutral temperature was 21-22 °C, and the heat stress temperature was 32-35 °C. Both heat stress groups experienced reduced growth performance, while the astaxanthin-treated group showed a slightly lesser decline. The inflammatory response and antioxidant defense system were activated by the upregulation of the NF-kB, NFE2L2, PPARα, cytoprotective capacity, and apoptotic gene pathways during heat stress compared to the thermal neutral group. However, expression levels showed no significant differences between the thermal neutral and heat stress with antioxidant groups, suggesting that astaxanthin may mitigate inflammation and oxidative stress damage.

Use of Microalgae-Derived Astaxanthin to Improve Cytoprotective Capacity in the Ileum of Heat-Induced Oxidative Stressed Broilers.

The gastrointestinal tract has a pivotal role in nutrient absorption, immune function, and overall homeostasis. The ileum segment of the small intestine plays respective roles in nutrient breakdown and absorption. The purpose of this study was to investigate the impact of heat-induced oxidative stress and the potential mitigating effects of an astaxanthin antioxidant treatment on the ileum of broilers. By comparing the growth performance and gene expression profiles among three groups-thermal neutral, heat stress, and heat stress with astaxanthin-thermal neutral temperature conditions of 21-22 °C and heat stress temperature of 32-35 °C, this research aims to elucidate the role of astaxanthin in supporting homeostasis and cellular protection in the ileum. Results showed both treatments under heat stress experienced reduced growth performance, while the group treated with astaxanthin showed a slightly lesser decline. Results further showed the astaxanthin treatment group significantly upregulated in the cytoprotective gene expression for HSF2, SOD2, GPX3, and TXN, as well as the upregulation of epithelial integrity genes LOX, CLDN1, and MUC2. In conclusion, our experimental findings demonstrate upregulation of cytoprotective and epithelial integrity genes, suggesting astaxanthin may effectively enhance the cellular response to heat stress to mitigate oxidative damage and contribute to cytoprotective capacity.

Assessment of Cinnamaldehyde's Potency on Heat Stress-induced Testicular Impairments in Wistar Rats.

Male sterility results from high testicular temperatures, which affect mammalian spermatogenesis. High testicular temperatures affect sperm motility, morphology and fertility according to their magnitude and duration. The aim of the current study is to examine the effects of heat-induced oxidative stress and cinnamaldehyde on Wistar rat testicular structure and function. The rats used in this experiment were Wistar albino rats. This research has six animals per group. Male Wistar albino rats of 2.5-3 months old and 275-300 g. (I) control, (II) heat stress (HS) in a closed chamber at 41°C for 14 days and (III) HS with cinnamaldehyde (CA) 50 mg/kg body weight for 14 days. (IV) CA alone. After the study, the animals were euthanised, and test samples were taken for sperm count, morphology, haematoxylin and eosin stain for normal cellular morphology, antioxidants and DNA integrity assessments. The data were analysed statistically using one- and two-way ANOVA tests for comparisons between groups. The stress group had significantly lower sperm counts and poor sperm morphology. The stress group's antioxidant capacity is much lower than that of the control group. Animals under stress have fragmented DNA. Treatment with cinnamaldehyde increased overall antioxidant capacity and seminal parameters, and rats behaved most like controls. CA restores malondialdehyde levels, total antioxidant capacity, sperm characteristics and mitigates testicular damage in rats exposed to experimental HS.

Advancements in Genetic Biomarkers and Exogenous Antioxidant Supplementation for Safeguarding Mammalian Cells against Heat-Induced Oxidative Stress and Apoptosis.

Heat stress represents a pervasive global concern with far-reaching implications for the reproductive efficiency of both animal and human populations. An extensive body of published research on heat stress effects utilizes controlled experimental environments to expose cells and tissues to heat stress and its disruptive influence on the physiological aspects of reproductive phenotypic traits, encompassing parameters such as sperm quality, sperm motility, viability, and overall competence. Beyond these immediate effects, heat stress has been linked to embryo losses, compromised oocyte development, and even infertility across diverse species. One of the primary mechanisms underlying these adverse reproductive outcomes is the elevation of reactive oxygen species (ROS) levels precipitating oxidative stress and apoptosis within mammalian reproductive cells. Oxidative stress and apoptosis are recognized as pivotal biological factors through which heat stress exerts its disruptive impact on both male and female reproductive cells. In a concerted effort to mitigate the detrimental consequences of heat stress, supplementation with antioxidants, both in natural and synthetic forms, has been explored as a potential intervention strategy. Furthermore, reproductive cells possess inherent self-protective mechanisms that come into play during episodes of heat stress, aiding in their survival. This comprehensive review delves into the multifaceted effects of heat stress on reproductive phenotypic traits and elucidates the intricate molecular mechanisms underpinning oxidative stress and apoptosis in reproductive cells, which compromise their normal function. Additionally, we provide a succinct overview of potential antioxidant interventions and highlight the genetic biomarkers within reproductive cells that possess self-protective capabilities, collectively offering promising avenues for ameliorating the negative impact of heat stress by restraining apoptosis and oxidative stress.

Research Note: Heat stress affects immune and oxidative stress indices of the immune organs of broilers by changing the expressions of adenosine triphosphate-binding cassette subfamily G member 2, sodium-dependent vitamin C transporter-2, and mitochondrial calcium uniporter

This study aimed to determine the mechanisms of heat-induced oxidative stress in the thymus and spleen of broilers. After 28 d, 30 broilers were randomly divided into the control (25°C ± 2°C; 24 h/d) and heat-stressed (36°C ± 2°C; 8 h/d) groups; the experiment lasted for 1 wk. The broilers in each group were euthanized, and some samples were collected and analyzed at 35 d. The results showed that the birds subjected to heat stress reduced the weight (P < 0.01) and the indices of thymus (P < 0.01), the activities of T-AOC (P < 0.01) and SOD (P < 0.05) of spleen, and levels of IL-10 (P < 0.05) and the GSH-PX (P < 0.05) in thymus and spleen, and increased the IL-6 content of thymus (P < 0.05), the MDA content (P < 0.01), and the reactive oxygen species (ROS) levels (P < 0.01) in thymus and spleen. Moreover, the expression of the IgG gene in the thymus and spleen of heat-stressed broilers was increased (P < 0.05); however, the expression of the IgM gene in the spleen was increased (P < 0.05), with no difference (P > 0.05) in the thymus of heat-stressed broilers compared with the control. Furthermore, the relative expression of adenosine triphosphate-binding cassette subfamily G member 2 (ABCG2) in the thymus and spleen both increased (P < 0.05). The sodium-dependent vitamin C transporter-2 (SVCT-2) (P < 0.01) and mitochondrial calcium uniporter (MCU) (P < 0.01) mRNA levels in the thymus of heat-stressed broilers increased, and the expression of ABCG2 (P < 0.05), SVCT-2 (P < 0.01), and MCU (P < 0.01) proteins in the thymus and spleen of heat-stressed broilers increased compared with the control group. This study confirmed that heat stress-induced oxidative stress in the immune organs of broilers, further reducing immune function.

Rice FERONIA-LIKE RECEPTOR 3 and 14 affect grain quality by regulating redox homeostasis during endosperm development.

Chalky endosperm negatively affects the appearance, milling, and eating qualities of rice (Oryza sativa L.) grains. Here, we report the role of two receptor-like kinases, FERONIA-LIKE RECEPTOR 3 (FLR3) and FERONIA-LIKE RECEPTOR 14 (FLR14), in grain chalkiness and quality. Knockouts of FLR3 and/or FLR14 increased the number of white-core grains caused by aberrant accumulation of storage substances, resulting in poor grain quality. Conversely, the overexpression of FLR3 or FLR14 reduced grain chalkiness and improved grain quality. Transcriptome and metabolome analyses showed that genes and metabolites involved in the oxidative stress response were significantly up-regulated in flr3 and flr14 grains. The content of reactive oxygen species was significantly increased in flr3 and flr14 mutant endosperm but decreased in overexpression lines. This strong oxidative stress response induced the expression of programmed cell death (PCD)-related genes and caspase activity in endosperm, which further accelerated PCD, resulting in grain chalkiness. We also demonstrated that FLR3 and FLR14 reduced grain chalkiness by alleviating heat-induced oxidative stress in rice endosperm. Therefore, we report two positive regulators of grain quality that maintain redox homeostasis in the endosperm, with potential applications in breeding rice for optimal grain quality.

Effects of Dietary Glutamine Supplementation on Heat-Induced Oxidative Stress in Broiler Chickens: A Systematic Review and Meta-Analysis.

In avian species, heat stress (HS) is usually the result of being exposed to high ambient temperatures, whereas oxidative stress (OS) results from the overproduction of reactive oxygen species. The current literature suggests that HS often leads to OS. Therefore, this systematic review and meta-analysis was conducted to assess the effects of dietary supplementation of glutamine on the antioxidant status and growth performances in heat-stressed broilers. A total of 13 studies were deemed eligible after an exhaustive search of the literature from Google Scholar, PubMed, and Scopus. Briefly, the following criteria were used to select the studies: trials performed on broilers; publication in peer-review journals using English as the text language; and sufficient details about the design and inclusion of dietary glutamine as a treatment for HS. Two main categories of outcomes were extracted from the studies included in the review: growth parameters and OS markers. For the meta-analysis, a random effect model was used when the heterogeneity was higher than 50%, and a fixed effect model was applied otherwise. Pooled standardized mean differences (SMD), and mean differences (MD) with their confidence intervals (CI) from the studies revealed that dietary glutamine could increase body weight gain (SMD = 0.70, CI = 0.50 to 0.90, p < 0.05), and feed intake (FI) (SMD = 0.64, CI = 0.43 to 0.86, p < 0.05), and reduce the feed conversion ratio (MD = -0.05, CI = -0.07 to -0.02, p < 0.05) in heat-exposed birds. Additionally, higher glutamine (SMD = 1.21, CI = 1.00 to 1.43, p < 0.05), glutathione (SMD = 1.25, CI = 0.88 to 1.62, p < 0.05), superoxide dismutase (SOD) (SMD = 0.97, CI = 0.58 to 1.36, p < 0.05), and catalase (SMD = 0.94, CI = 0.72 to 1.16, p < 0.05) levels were recorded in the serum, breast, and thigh muscle after supplementation of glutamine. Furthermore, the subgroup analysis revealed that malondialdehydes levels were decreased only in the serum (SMD = -0.83, CI = -1.25 to -0.41, p < 0.001) and thigh muscle (SMD = -1.30, CI = -1.86 to -0.35, p < 0.001) while glutathione peroxidase (GPX) activity was increased in the breast (SMD = 1.32, CI = 0.95 to 1.68, p < 0.05) and thigh muscle (SMD = 1.53, CI = 1.06 to 1.99, p < 0.05). Meta-regression models indicated that longer periods of heat exposure were inversely associated with the effectiveness of dietary glutamine in increasing FI, GPX, and SOD (p < 0.05). Besides, increasing the dietary concentration of glutamine led to higher GPX and SOD levels (p < 0.05). Taken together, results suggest that dietary supplementation of glutamine can effectively mitigate the deleterious effects of HS by enhancing the antioxidant status and increasing growth performances in broilers.

Exogenous Application of Salicylic acid Mitigates heat-induced Oxidative Stress in Barley (Hordeum vulgare L.)

Due to climate change, heat stress is becoming one of the most disastrous abiotic stresses and responsible for huge crop losses in different regions of the world. Adverse effects of heat stress can be encountered by the use of phytohormones and salicylic acid (SA) is one of those hormones having capability to protect plants from heat stress. Current research work was planned to explore the defensive role of exogenously applied SA in barley cultivars under high temperature. For this purpose, two weeks old plants of four barley cultivars (Jau-87, B-10007, B-14003 and B-14037) were foliar sprayed with SA (2 mM and 5 mM) for 24 hours and then were exposed to heat stress (42 °C). Our results showed that heat exposure made a prominent decrease in chlorophyll contents, carotenoids, relative water contents, antioxidant enzymes activities (superoxide dismutase, peroxidase and catalase) and anthocyanin activity while increased lipid peroxidation (malondialdehyde), hydrogen peroxide (H2O2) contents and proline accumulation. Foliar application of SA enhanced plants photosynthetic pigments (chlorophyll a, b and total), RWC, proline, and anthocyanin under control and stressed conditions. SA pre-treatment under heat stress reduced H2O2 and MDA levels whereas activities of reactive oxygen species (ROS) scavenging enzymes were elevated by all tested SA concentrations. These results suggest that SA can efficiently remove H2O2 by increasing activity of above mentioned enzymes hence reducing oxidative stress and malondialdehyde (MDA) contents. Proline, an important amino acid that acts as an anti-oxidative system in plants also highly accumulated in SA co-treated heat-stressed plants. Our results also showed that 2mM SA is more effective in protecting barley plants under heat stress as compared to 5mM SA. This concluded that defensive nature of SA regulates different physiological and biochemical processes to induce heat tolerance in barley.

Procyanidin B2 Alleviates Heat-Induced Oxidative Stress through the Nrf2 Pathway in Bovine Mammary Epithelial Cells.

The objective of this study was to investigate the protective effects and potential molecular mechanisms of procyanidin B2 (PB2) in MAC-T (mammary alveolar cells-large T antigen) cells during heat stress (HS). The MAC-T cells were divided into three treatment groups: control (37 °C), HS (42 °C), and PB2 + HS (42 °C). Compared with MAC-T cells that were consistently cultured at 37 °C, acute HS treatment remarkably decreased cell viability, reduced activities of catalase (CAT), superoxide dismutase (SOD), and total antioxidant capacity (T-AOC), and elevated intracellular levels of malondialdehyde (MDA) and reactive oxygen species (ROS). Additionally, nuclear factor erythroid 2-related factor 2 (Nrf2) was activated and translocated to the nucleus, in accompaniment with upregulation of Nrf2, heme oxygenase 1 (HO-1), thioredoxin reductase 1 (Txnrd1), and heat shock protein 70 (HSP70). In parallel, both mRNA transcript and actual protein secretion of pro-inflammatory cytokines, including tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β), were increased by heat stress. Pretreatment of MAC-T cells with 0~25 μM PB2 alleviated the decline of cell viability by HS in a dose-dependent fashion and protected cells against HS-induced oxidative stress, as evidenced by significantly improved CAT, SOD, and T-AOC activity, as well as with decreased MDA and ROS generation. Furthermore, PB2 further activated the Nrf2 signaling pathway and reversed the inflammatory response induced by HS. Silencing of Nrf2 by si-Nrf2 transfection not only exacerbated HS-induced cell death and provoked oxidative stress and the inflammatory response, but also greatly abolished the cytoprotective effects under HS of PB2. In summary, PB2 protected MAC-T cells against HS-induced cell death, oxidative stress, and inflammatory response, partially by operating at the Nrf2 signal pathway.

Melatonin: A Vital Pro-Tectant for Crops against Heat Stress: Mechanisms and Prospects

Heat stress (HS) is a serious environmental stress that negatively affects crop growth and productivity across the globe. The recent increase in atmospheric temperature caused by global warming has increased its intensity, which is a serious challenge that needs to be addressed. Plant growth and development involves a series of physiological, metabolic, and biochemical processes that are negatively affected by heat-induced oxidative stress, disorganization of cellular membranes and disturbed plant water relations, nutrient uptake, photosynthetic efficiency, and antioxidant activities. Plant tolerance to abiotic stresses can be substantially increased by the application of bio-stimulants, without posing a threat to the ecosystem. Melatonin (MT) is a multi-functional signaling molecule that has the potential to protect plants from the adverse impacts of HS. MT protects the cellular membranes, maintains the leaf water content, and improves the water use efficiency (WUE) and nutrient homeostasis; thereby, improving plant growth and development under HS. Moreover, MT also improves gene expression, crosstalk of hormones, and osmolytes, and reduces the accumulation of reactive oxygen species (ROS) by triggering the antioxidant defense system, which provides better resistance to HS. High endogenous MT increases genes expression and antioxidant activities to confer HS tolerance. Thus, it is important to understand the detailed mechanisms of both exogenous and endogenous MT, to induce HS tolerance in plants. This review highlights the versatile functions of MT in various plant responses, to improve HS tolerance. Moreover, we also discussed the MT crosstalk with other hormones, antioxidant potential of MT, and success stories of engineering MT to improve HS tolerance in plants. Additionally, we also identified various research gaps that need to be filled in future research using this important signaling molecule. Thus, this review will help the readers to learn more about MT under changing climatic conditions and will provide knowledge to develop heat tolerance in crops.

Wheat variety carrying 2NvS chromosomal segment provides yield advantage through lowering terminal heat-induced oxidative stress.

A 2NvS chromosomal segment carrying bread wheat variety, BARI Gom 33 ('BG33'), showed tolerance to terminal heat stress andhigher yield over a heat-tolerant non-2NvS BARI Gom 26 ('BG26') and a heat-susceptible Pavon 76 ('Pavon'). This study aimed to ascertain the potential of the 2NvS 'BG33' in terminal heat-induced oxidative stress tolerance compared to non-2NvS 'BG26' and heat-susceptible 'Pavon' under two heat regimes at the reproductive stages viz. control (optimum sowing time) and heat stress (late sowing). We found that both 'BG26' and 'BG33' showed significantly higher tolerance to oxidative stress by limiting the generation of reactive oxygen species (ROS), methylglyoxal under heat stress. During terminal heat stress, both 'BG33' and 'BG26' exhibited greater cellular homeostasis than heat-susceptible 'Pavon', which was maintained by the increased accumulation of osmolytes, nonenzymatic antioxidants, and enzymes associated with ROS scavenging, ascorbate-glutathione cycle, and glyoxalase system. Lesser cellular damage in 'BG26' and 'BG33' was eventually imitated in a smaller reduction in grain yield (15 and 12%, respectively) than in 'Pavon', which had a 33% reduction owing to heat stress. Collectively, our findings revealed that the chromosomal segment 2NvS provides yield advantage to 'BG33' under terminal heat stress by lowering oxidative damage. As 2NvS translocation contains multiple nucleotide-binding domain leucine-rich repeat containing, cytochrome P450, and other gene families associated with plant stress tolerance, further studies are warranted to dissect the underlying molecular mechanisms associated with higher heat stress tolerance of 2NvS carrying 'BG33'.

Acute exposure to perfluorooctane sulfonate exacerbates heat-induced oxidative stress in a tropical coral species.

Perfluorooctane sulfonate (PFOS) is among the most commonly per- and poly-fluoroalkyl substances (PFAS) found in environmental samples. Nevertheless, the effect of this legacy persistent organic contaminant has never been investigated on corals to date. Corals are the keystone organisms of coral reef ecosystems and sensitive to rising ocean temperatures, but it is not understood how the combination of elevated temperature and PFOS exposure will affect them. Therefore, the aims of the present study were (1) to evaluate the time-dependent bioconcentration and depuration of PFOS in the scleractinian coral Stylophora pistillata using a range of PFOS exposure concentrations, and (2) to assess the individual and combined effects of PFOS exposure and elevated seawater temperature on key physiological parameters of the corals.Our results show that the coral S. pistillata rapidly bioconcentrates PFOS from the seawater and eliminates it 14 days after ceasing the exposure. We also observed an antagonistic effect between elevated temperature and PFOS exposure. Indeed, a significantly reduced PFOS bioconcentration was observed at high temperature, likely due to a loss of symbionts and a higher removal of mucus compared to ambient temperature. Finally, concentrations of PFOS consistent with ranges observed in surface waters were non-lethal to corals, in the absence of other stressors. However, PFOS increased lipid peroxidation in coral tissue, which is an indicator of oxidative stress and enhanced the thermal stress-induced impairment of coral physiology. This study provides valuable insights into the combined effects of PFOS exposure and ocean warming for coral's physiology. PFOS is usually the most prevalent but not the only PFAS defected in reef waters, and thus it will be also important to monitor PFAS mixture concentrations in the oceans and to study their combined effects on aquatic wildlife.

L-Theanine mediates the p38MAPK signaling pathway to alleviate heat-induced oxidative stress and inflammation in mice.

L-Theanine, an active ingredient in the tea plant (Camellia sinensis) associated with calming, is widely used as a functional ingredient and dietary supplement. In this study, a heat stress mouse model was used to evaluate the anti-heat stress effect of L-theanine and its possible mechanism of action. Mice subjected to heat stress (40 °C) that were administered L-theanine at various doses (100, 200, and 400 mg kg-1 d-1) had reduced oxidative stress and inflammatory factors when L-theanine was administered both long-term and as a preventative treatment. Our L-theanine intervention countered the reduction in growth and feed intake of mice under heat stress and reversed liver and jejunum tissue damage. Moreover, L-theanine countered the increase in inflammatory factors TNF-α, IL-6, and IL-1β and antioxidant enzymes SOD and CAT; it also counteracted GSH-Px inactivation, the upregulation of AST and ALT enzyme activity, and MDA production. The mechanism of action may involve mediation of the P38 signaling pathway, inhibition of MK2 overexpression, and downregulation of p-P65/P65 caused by the overexpression of downstream HSP27. This would inhibit the heat stress-induced imbalance in oxidative stress and inflammatory responses.

ITRAQ-based quantitative proteomic analyses the cycle chronic heat stress affecting liver proteome in yellow-feather chickens

Heat stress (HS) is one of the main environmental factors affecting the efficiency of poultry production. The yellow-feather chickens (YFC) as an indigenous strain of chicken is a popular poultry breed in China. Our previous study used the RNA-seq to analyze the gene expression profiles of male YFC under HS and showed that the lipid and energy metabolism pathways are activated in livers of YFC exposed to acute HS (38°C, 4 h and 25°C recovery 2 h). In this study, we used quantitative proteome analysis based on iTRAQ to study the liver response of YFC to cycle chronic HS (38 ± 1°C, 8 h/d, 7 d, CyCHS). The male YFCs treatment used the CyCHS from 22 to 28 days of age. The liver tissue samples were collected at 28 d old. A total of 39,327 unique peptides matches were detected using iTRAQ analysis and 4,571 proteins exhibited a false discovery rate of 1% or less. Forty-six significant differentially expressed proteins (DEPs) were detected in the CyCHS group compared with the control group for the liver samples, including up- and down-regulated DEPs were 18 and 28, respectively. We found that the enriched biological process terms of the DEPs expressed in the liver were related to DNA metabolic process, oxidation-reduction process, oxidative stress and gluconeogenesis. In KEGG pathway analysis. Most of the hepatic DEPs were annotated to glutathione metabolism and TCA cycle in response to CyCHS. The up-regulation of 5 DEPs (GPX1, GSTT1, GSTT1L, RRM2, and LOC100859645) in the glutathione metabolism pathway likely reflects an attempt to deal with oxidative damage by CyCHS. The down-regulation of 3 DEPs (Isocitrate dehydrogenase [IDH3A], IDH3B, and phosphoenolpyruvate carboxykinase 1) in the TCA cycle pathway contributes to the regulation mechanism of energy metabolism and probably to cope with the balance of heat production and dissipation during CyCHS in order to adapt to high temperature environments. Our results provide insights into the potential molecular mechanism in heat-induced oxidative stress and energy in YFCs and future studies will investigate the functional genes associated with the response to HS.

Melatonin alleviates heat stress-induced oxidative stress and apoptosis in human spermatozoa

Oxidative stress generates a large amount of reactive oxygen species (ROS) and affects sperm quality via damaging sperm DNA and compromising the intracellular homeostasis in human spermatozoa. In assisted reproductive technology (ART), it is substantial to prevent spermatozoa from ROS attack. The pineal hormone melatonin has the natural antioxidant capacity and can scavenge ROS. To the best of our knowledge, however, there are presently no studies investigating if melatonin can protect human spermatozoa from heat-induced oxidative damage. Herein, we induced oxidative stress in human spermatozoa with heat treatment, and determined that melatonin could protect human spermatozoa from heat-induced oxidative stress. We first confirmed that heat stress-induced oxidative stress damaged human spermatozoa by decreasing sperm motility and viability. Furthermore, the pretreatment of human spermatozoa by melatonin was able to alleviate such damage by suppressing sperm mitochondrial ROS generation, increasing mitochondrial membrane potential, reducing the formation of the lipid peroxidation product, 4-HNE, and reducing sperm DNA damage and apoptosis. Collectively, these findings suggest that melatonin is useful as a potential treatment option for male infertility caused by heat-induced oxidative stress.

Heat-responsive and time-resolved transcriptome and metabolome analyses of Escherichia coli uncover thermo-tolerant mechanisms

Current understanding of heat shock response has been complicated by the fact that heat stress is inevitably accompanied by changes in specific growth rates and growth stages. In this study, a chemostat culture was successfully performed to avoid the physico-chemical and biological changes that accompany heatshock, which provided a unique opportunity to investigate the full range of cellular responses to thermal stress, ranging from temporary adjustment to phenotypic adaptation at multi-omics levels. Heat-responsive and time-resolved changes in the transcriptome and metabolome of a widely used E. coli strain BL21(DE3) were explored in which the temperature was upshifted from 37 to 42 °C. Omics profiles were categorized into early (2 and 10 min), middle (0.5, 1, and 2 h), and late (4, 8, and 40 h) stages of heat stress, each of which reflected the initiation, adaptation, and phenotypic plasticity steps of the stress response. The continued heat stress modulated global gene expression by controlling the expression levels of sigma factors in different time frames, including unexpected downregulation of the second heatshock sigma factor gene (rpoE) upon the heat stress. Trehalose, cadaverine, and enterobactin showed increased production to deal with the heat-induced oxidative stress. Genes highly expressed at the late stage were experimentally validated to provide thermotolerance. Intriguingly, a cryptic capsular gene cluster showed considerably high expression level only at the late stage, and its expression was essential for cell growth at high temperature. Granule-forming and elongated cells were observed at the late stage, which was morphological plasticity occurred as a result of acclimation to the continued heat stress. Whole process of thermal adaptation along with the genetic and metabolic changes at fine temporal resolution will contribute to far-reaching comprehension of the heat shock response. Further, the identified thermotolerant genes will be useful to rationally engineer thermotolerant microorganisms.

Effects of supplementation of DL-methionine on tissue and plasma antioxidant status during heat-induced oxidative stress in broilers

Exposure to high ambient temperature has been shown to impair growth performance and to cause oxidative stress in broilers. This study investigated the hypothesis that supplementation with methionine (Met) as DL-Met (DLM) more than the National Research Council recommendations improves growth performance and alleviates oxidative stress in broilers exposed to high ambient temperature. One-day-old male Cobb-500 broilers (n = 68) were allotted to 4 groups and phase-fed 3 basal diets during days 1 to 10, 11 to 21, and 22 to 35. One group was kept under thermoneutral temperature conditions and received the basal diets with Met + cysteine (Cys) concentrations according to recommendations of NRC. The other 3 groups were kept in a room with an increased ambient temperature from week 3 to 5 and were fed either the basal diet or the basal diets supplemented with 2 levels of DLM in which Met + Cys concentrations exceeded NRC recommendations by around 20% (group DLM1) and 40% (group DLM2), respectively. As expected, the broilers exposed to high ambient temperature showed a lower feed intake, lower body weight gains, a higher feed:gain ratio, and biochemical indications of oxidative stress in comparison to broilers kept under thermoneutral temperature conditions. Supplementation of DLM did not improve the growth performance in broilers exposed to high ambient temperature. However, the broilers supplemented with DLM had increased concentrations of glutathione in liver and breast muscle (groups DLM1 and DLM2), increased concentrations of tocopherols in the liver (group DLM2), and reduced concentrations of 7α-hydroxycholesterol and 7-ketocholesterol in heat-processed thigh muscle (groups DLM1 and DLM2) in comparison to the control group exposed to high ambient temperature. Concentrations of thiobarbituric acid-reactive substances and vitamin C in plasma, liver, and muscle were not different between the 3 groups exposed to heat stress. Nevertheless, the study shows that supplementation of DLM in slight excess of the Met concentration required for maximum growth performance improved the antioxidant status in tissues and reduced the susceptibility of muscle toward oxidation in heat-stressed broilers.

Silicon and Gibberellins: Synergistic Function in Harnessing ABA Signaling and Heat Stress Tolerance in Date Palm (Phoenix dactylifera L.).

Date palm is one of the most economically vital fruit crops in North African and Middle East countries, including Oman. A controlled experiment was conducted to investigate the integrative effects of silicon (Si) and gibberellic acid (GA3) on date palm growth and heat stress. The exogenous application of Si and GA3 significantly promoted plant growth attributes under heat stress (44 ± 1 °C). The hormonal modulation (abscisic acid [ABA] and salicylic acid [SA]), antioxidant accumulation, and the expression of abiotic stress-related genes were evaluated. Interestingly, heat-induced oxidative stress was markedly reduced by the integrative effects of Si and GA3 when compared to their sole application, with significant reductions in superoxide anions and lipid peroxidation. The reduction of oxidative stress was attributed to the enhancement of polyphenol oxidase, catalase, peroxidase, and ascorbate peroxidase activities as well as the upregulation of their synthesis related genes expression viz. GPX2, CAT, Cyt-Cu/Zn SOD, and glyceraldehyde3-phosphate dehydrogenase gene (GAPDH). The results showed the activation of heat shock factor related genes (especially HsfA3) during exogenous Si and GA3 as compared to the control. Furthermore, the transcript accumulation of ABA signaling-related genes (PYL4, PYL8, and PYR1) were significantly reduced with the combined treatment of Si and GA3, leading to reduced production of ABA and, subsequently, SA antagonism via its increased accumulation. These findings suggest that the combined application of Si and GA3 facilitate plant growth and metabolic regulation, impart tolerance against stress, and offers novel stress alleviating strategies for a green revolution in sustainable food security.
