Mito-Specific Nutri-Hijacker Synergizing Mitochondrial Metabolism and Glycolysis Intervention for Enhanced Antitumor Bioenergetic Therapy.

Metabolic rewiring, a dynamic metabolic phenotype switch, confers that tumors exist and proliferate after fitness (or preadaptation) in harsh environmental conditions. Glycolysis deprivation was considered to be a tumor's metabolic Achilles heel. However, metabolic configuration can flexibly retune the mitochondrial metabolic ability when glycolysis is scared, potentially resulting in more aggressive clones. To address the challenge of mitochondrial reprogramming, an antiglycolytic nanoparticle (GRPP NP) containing a novel mitochondrial-targeted reactive oxygen species (ROS) generator (diIR780) was prepared to hijack glucose and regulate mitochondria, thus completely eliminating tumorigenic energy sources. In this process, GRPP NPs@diIR780 can catalyze endogenous glucose, leading to significantly suppressed glycolysis. Moreover, diIR780 can be released and selectively accumulated around mitochondria to generate toxic ROS. These combined effects, in turn, can hamper mitochondrial metabolism pathways, which are crucial for driving tumor progression. This synchronous intervention strategy enables utter devastation of metabolic rewiring, providing a promising regiment to eradicate tumor lesions without recurrence.

Assessment of a Full-Electric Floating Liquefied Natural Gas Concept for Harsh Sea State Conditions at Remote Ultra-Deepwater Locations

_ This work describes an offshore gas monetization solution for harsh environmental conditions at ultra-deepwater and remote locations, as an alternative to conventional onshore plants associated with long subsea pipelines for Greenfield projects. Although gas-to-liquids and marine compressed natural gas are initially evaluated, associated gas is supposed to be exported from deepwater floating production storage and offloading units to a central floating liquefied natural gas (FLNG) facility for additional treatment, fractionation, and liquefaction, producing liquefied natural gas (LNG) and liquefied petroleum gas (LPG) for further tandem offloading. To demonstrate the technical feasibility of the proposed FLNG, its key technologies are reviewed, including liquefaction process selection, cargo containment system, station-keeping, offloading, and power generation. A full-electric FLNG is proposed, replacing conventional feed gas power generation systems with inner hull nuclear power modules, envisaging neutral CO2 emissions and earlier first LNG drop milestone achievement. The FLNG general arrangement is developed, along with its internal cargo tanks layout, for preliminary LNG and LPG production rundown and feed gas throughput. Numerical analyses are carried out to show that static and dynamic tilt criteria can be met, even for harsh sea states. Gas import riser configurations for the challenging ultra-deepwater location are outlined, after appropriate FLNG station-keeping analyses, in addition to a feasibility assessment of seawater intake risers, aiming for an overall gas plant efficiency increase. International Biological Flora: Festuca dolichophylla†

Abstract This account provides information on all aspects of the biology of Festuca dolichophylla J. Presl (Chilliwa, Chilligua) that are relevant to understanding its ecological characteristics and behaviour. The main topics are presented within the standard framework of the International Biological Flora including distribution, habitat, communities, responses to biotic and environmental factors, structure and physiology, phenology, floral and seed characters, herbivores and disease, history, conservation and global heterogeneity. Festuca dolichophylla is a tall perennial tussock grass that thrives at high elevations. It is native and widely distributed along the Andes of South America. It also occurs in North and Central America, especially in Mexico and Costa Rica. Festuca dolichophylla plays a significant role in Andean ecosystems and communities of Ecuador, Perú, Bolivia and northern Argentina. It serves various purposes, primarily as food source for South American camelids such as llama and alpaca. Festuca dolichophylla is dominant in Altiplano (Andean high plains) grasslands and is also present in Bofedales (Andean peatlands), shrublands and possibly in Ecuador and Colombia paramos. Festuca dolichophylla reproduces through seeds and vegetative tillers. Flowering tillers bear panicles with multiple spikelets and florets. An ungrazed plant can produce as much as 15.6 g of seeds during May and June, and seeds can germinate in 7 days or more. Festuca dolichophylla exhibits physiological and ecological traits that enable it to thrive in harsh environmental conditions, including extreme temperatures, drought and nutrient‐poor soils. It thrives in a range of soil types from strongly acid to slightly alkaline, encompassing textures that include loamy, sandy clay loam, clay loam and clay, and which are influenced by the cold and semiarid Andean climate. Fertilizers with different NPK formulations enhance fresh aerial biomass, plant height and volume. Additionally, F. dolichophylla forms mycorrhizal associations, stores large amounts of fixed carbon in its stems, and bioaccumulates mercury, lead and arsenic. Despite its importance, Festuca dolichophylla remains poorly studied. Further research is needed to explore its ecology, breeding systems, diseases or ecotypes. Moreover, population and demographic studies are crucial to assess the impact of climate change on this species. Adaptive, strategic and organized management and conservation efforts are essential, particularly in Indigenous Andean settlements, where F. dolichophylla offers multiple applications and benefits. Furthermore, utilizing this species in grassland revegetation and restoration initiatives highlights the need for ongoing research and management endeavours to ensure its conservation and sustainable use.

Durability analysis of polymer electrolyte membrane fuel Cell's gas diffusion layer based on distribution relaxation time analysis: Influence of the presence or absence of a micro-porous layer

The U.S. Department of Energy accelerated stress test carbon protocol emphasizes improving the durability of both the catalyst layer and the micro-porous layer (MPL) carbon support. This underscores the need for research focused solely on the degradation of the Gas Diffusion Layer (GDL). Traditionally, electrochemical impedance spectroscopy impedance data used to analyze resistance due to GDL durability has been interpreted through equivalent circuit models. However, this method simplifies simultaneous response and multiple models for the same data, which reduces the objectivity of the analysis. Therefore, this study analyzed the durability characteristics of GDLs with and without MPLs using distribution of relaxation times, a method less prone to subjectivity. In short, the results showed that substrates without MPL are more susceptible to performance degradation and loss, and highlighted the emergence of Warburg resistance due to limited diffusion under harsh environmental conditions.

Green Fabrication of Superhydrophilic/Underwater Superoleophobic Composite Membrane for High-Efficiency Oil/Water Separation in Harsh Environments.

Due to the high oil spill incidence and industrial wastewater discharge including oil and emulsified oil, designing and synthesizing oil-water separation materials which can maintain stability under harsh environmental conditions with high separation efficiencies remains a great challenge. The present work developed an easy, green, cost-effective, and easily scaled-up approach for fabricating cellulose-based membranes. First, we coated polydopamine (PDA) onto fibers of filter membrane (FM). Then, the PDA-FM membrane was immersed into the mixed solution of poly(vinyl alcohol)/poly(acrylic acid) (PVA/PAA) and further thermally cross-linked at 150 °C to create a superhydrophilic/underwater superoleophobic membrane (PVA/PAA@PDA-FM) to separate oil/water mixtures. The simple thermally cross-linking process promotes multiple covalent chemical bonds generation between cellulose filter membrane, PAA, PDA, and PVA, endowing membranes with excellent stability and resistance to acidity, alkalinity, and salinity. The PVA/PAA@PDA-FM membrane not only demonstrates great separation performance (>99.8%) and great flux (>1000 L m-2 h-1) in oil-water immiscible mixtures but also maintains high separation efficiency under conditions of high acidity, alkalinity, and salinity. Additionally, the PVA/PAA@PDA-FM membrane exhibits excellent separation capacity in oil-water emulsions, which can maintain the >99.6% separation efficiency even after 40 cycles in harsh environments, showing outstanding reusability. Thus, due to the multiple cross-linked networks in the membrane, the excellent performance makes the PVA/PAA@PDA-FM membrane a good application prospect in water purification and oily wastewater treatment.

Laboratory Demonstration of Grating Embedded Mirrors for Single-Shot Heliostat Optical Metrology

Abstract Concentrated solar power (CSP) plants need to monitor the surface slope error of thousands of heliostats with sub-milliradian accuracy. Large numbers of heliostats installed in harsh environments mean that measurement speed and durability are key design considerations for metrology systems. We present a compact, accurate, and high-speed heliostat slope error metrology system that is robust to the harsh environmental conditions at CSP plants. The system is composed of (1) grating embedded mirrors (GEMs), which have multiple different phase diffraction gratings written within a glass mirror substrate, and (2) a compact optical system, the diffractive auto-stigmatic Hartmann camera (DASHCam). Using focused ultrafast laser pulses, we write phase gratings within second-surface float glass mirror without damaging the reflective coating on the backside. The gratings direct a small fraction of incident light to non-specular directions, which the DASHCam senses from a virtual center of curvature to measure the mirror slopes at each grating patch. In a CSP plant, each heliostat mirror would be a GEM, and the DASHCam would rapidly measure slope and canting errors during heliostat manufacturing, installation, or operation. We fabricated 0.1 meter-diameter GEMs with 24 grating patches, built a prototype DASHCam system, and tested the system repeatability and accuracy by comparing against a Fizeau interferometer. Our results show, in a laboratory environment, 24μrad root mean square (RMS) measurement repeatability and 47μrad RMS accuracy, with single-shot image capture. The combination of GEMs and DASHCam is a promising metrology approach that could lead to improved optical accuracy of heliostats throughout their life cycle. This work serves as a proof of principle for this system.

Therapeutic Potential of Bovine Milk-Derived Extracellular Vesicles.

Milk is a fundamental component of the human diet, owing to its substantial nutritional content. In addition, milk contains nanoparticles called extracellular vesicles (EVs), which have indicated their potential beneficial roles such as cell-to-cell communication, disease biomarkers, and therapeutics agents. Amidst other types of EVs, milk EVs (MEVs) have their significance due to their high abundance, easy access, and stability in harsh environmental conditions, such as low pH in the gut. There have been plenty of studies conducted to evaluate the therapeutic potential of bovine MEVs over the past few years, and attention has been given to their engineering for drug delivery and targeted therapy. However, there is a gap between the experimental findings available and clinical trials due to the many challenges related to EV isolation, cargo, and the uniformity of the material. This review aims to provide a comprehensive comparison of various techniques for the isolation of MEVs and offers a summary of the therapeutic potential of bovine MEVs described over the last decade, analyzing potential challenges and further applications. Although a number of aspects still need to be further elucidated, the available data point to the role of MEVs as a potential candidate with therapeutics potential, and the supplementation of MEVs would pave the way to understanding their in-depth effects.

Different machine learning methods based on maxillary sinus in sex estimation for northwestern Chinese Han population.

Sex estimation is a critical aspect of forensic expertise. Some special anatomical structures, such as the maxillary sinus, can still maintain integrity in harsh environmental conditions and may be served as a basis for sex estimation. Due to the complex nature of sex estimation, several studies have been conducted using different machine learning algorithms to improve the accuracy of sex prediction from anatomical measurements. In this study, linear data of the maxillary sinus in the population of northwest China by using Cone-Beam Computed Tomography (CBCT) were collected and utilized to develop logistic, K-Nearest Neighbor (KNN), Support Vector Machine (SVM) and random forest (RF) models for sex estimation with R 4.3.1. CBCT images from 477 samples of Han population (75 males and 81 females, aged 5-17 years; 162 males and 159 females, aged 18-72) were used to establish and verify the model. Length (MSL), width (MSW), height (MSH) of both the left and right maxillary sinuses and distance of lateral wall between two maxillary sinuses (distance) were measured. 80% of the data were randomly picked as the training set and others were testing set. Besides, these samples were grouped by age bracket and fitted models as an attempt. Overall, the accuracy of the sex estimation for individuals over 18 years old on the testing set was 77.78%, with a slightly higher accuracy rate for males at 78.12% compared to females at 77.42%. However, accuracy of sex estimation for individuals under 18 was challenging. In comparison to logistic, KNN and SVM, RF exhibited higher accuracy rates. Moreover, incorporating age as a variable improved the accuracy of sex estimation, particularly in the 18-27 age group, where the accuracy rate increased to 88.46%. Meanwhile, all variables showed a linear correlation with age. The linear measurements of the maxillary sinus could be a valuable tool for sex estimation in individuals aged 18 and over. A robust RF model has been developed for sex estimation within the Han population residing in the northwestern region of China. The accuracy of sex estimation could be higher when age is used as a predictive variable.

Metabolic profile and gene expression pattern of cytokines and antioxidants markers during different physiological stages in Barki ewes

BackgroundIn livestock, identifying the physiological and reproductive stages is valuable in guiding management decisions related to nutrition, veterinary procedures, and breeding programs. To achieve this goal, a cohort of Barki ewes in this research underwent observation across three pivotal physiological conditions: pre-pregnancy, late pregnancy, and early lactation. Blood samples were collected to investigate the changes in serum metabolic profile as well as gene expression pattern of cytokines and antioxidants markers during these stages.ResultsOur results showed that during late pregnancy, there was a significant (P < 0.05) increase in red blood cells (11.9 ± 0.5 1012/L), hemoglobin (10.8 ± 0.4 g/dl) and neutrophils count (7 ± 0.1 109/L) with significant decrease (P < 0.05) of total white blood cell count (9.1 ± 0.05 109/L). The packed cell volume (%) and monocyte count showed a significant (P < 0.05) decrease during both late pregnancy and early lactation stages. The serum concentrations of glucose, cholesterol, GSH, GPx, SOD and catalase displayed significant (P < 0.05) decrease during late pregnancy and early-lactation. Notably, during late pregnancy, there was a significant (P < 0.05) increase in the serum concentrations of albumin, globulin, urea, IGF-1, and malondialdehyde with significant decrease (P < 0.05) of total protein (4.9 ± 0.08 g/dl). Additionally, during early lactation, there was a significant (P < 0.05) increase in the serum levels of non-esterified fatty acids, triiodothyronine (T3), and thyroxin (T4). The gene expression profiles of cytokines (IL-4, IL-6, IL-8, and NFKB) were decreased in the ewes during late pregnancy compared to pre-pregnant and early lactation stages. In addition, the expression profile of antioxidant genes (SOD, CAT, GPX, and Nrf2) was significantly upsurged in the non-pregnant ewes compared to late pregnancy and early lactation ones.ConclusionsThe results concluded that different physiological status significantly affects the blood metabolic profile and gene expression pattern in Barki sheep. Our findings can be helpful in monitoring animal health and applying in breeding programs of Barki sheep under harsh environmental conditions.

Unlocking the biosynthetic potential and taxonomy of the Antarctic microbiome along temporal and spatial gradients.

Extreme environments, such as Antarctica, select microbial communities that display a range of evolutionary strategies to survive and thrive under harsh environmental conditions. These include a diversity of specialized metabolites, which have the potential to be a source for new natural product discovery. Efforts using (meta)genome mining approaches to identify and understand biosynthetic gene clusters in Antarctica are still scarce, and the extent of their diversity and distribution patterns in the environment have yet to be discovered. Herein, we investigated the biosynthetic gene diversity of the biofilm microbial community of Whalers Bay, Deception Island, in the Antarctic Peninsula and revealed its distribution patterns along spatial and temporal gradients by applying metagenome mining approaches and multivariable analysis. The results showed that the Whalers Bay microbial community harbors a great diversity of biosynthetic gene clusters distributed into seven classes, with terpene being the most abundant. The phyla Proteobacteria and Bacteroidota were the most abundant in the microbial community and contributed significantly to the biosynthetic gene abundances in Whalers Bay. Furthermore, the results highlighted a significant correlation between the distribution of biosynthetic genes and taxonomic diversity, emphasizing the intricate interplay between microbial taxonomy and their potential for specialized metabolite production.IMPORTANCEThis research on antarctic microbial biosynthetic diversity in Whalers Bay, Deception Island, unveils the hidden potential of extreme environments for natural product discovery. By employing metagenomic techniques, the research highlights the extensive diversity of biosynthetic gene clusters and identifies key microbial phyla, Proteobacteria and Bacteroidota, as significant contributors. The correlation between taxonomic diversity and biosynthetic gene distribution underscores the intricate interplay governing specialized metabolite production. These findings are crucial for understanding microbial adaptation in extreme environments and hold significant implications for bioprospecting initiatives. The study opens avenues for discovering novel bioactive compounds with potential applications in medicine and industry, emphasizing the importance of preserving and exploring these polyextreme ecosystems to advance biotechnological and pharmaceutical research.

Mitigation of UV-B Radiation Stress in Tobacco Pollen by Expression of the Tardigrade Damage Suppressor Protein (Dsup).

Pollen, the male gametophyte of seed plants, is extremely sensitive to UV light, which may prevent fertilization. As a result, strategies to improve plant resistance to solar ultraviolet (UV) radiation are required. The tardigrade damage suppressor protein (Dsup) is a putative DNA-binding protein that enables tardigrades to tolerate harsh environmental conditions, including UV radiation, and was therefore considered as a candidate for reducing the effects of UV exposure on pollen. Tobacco pollen was genetically engineered to express Dsup and then exposed to UV-B radiation to determine the effectiveness of the protein in increasing pollen resistance. To establish the preventive role of Dsup against UV-B stress, we carried out extensive investigations into pollen viability, germination rate, pollen tube length, male germ unit position, callose plug development, marker protein content, and antioxidant capacity. The results indicated that UV-B stress has a significant negative impact on both pollen grain and pollen tube growth. However, Dsup expression increased the antioxidant levels and reversed some of the UV-B-induced changes to pollen, restoring the proper distance between the tip and the last callose plug formed, as well as pollen tube length, tubulin, and HSP70 levels. Therefore, the expression of heterologous Dsup in pollen may provide the plant male gametophyte with enhanced responses to UV-B stress and protection against harmful environmental radiation.

Eco-evolutionary dynamics of cooperative antimicrobial resistance in a population of fluctuating volume and size

Abstract Antimicrobial resistance to drugs (AMR), a global threat to human and animal health, is often regarded as resulting from a cooperative behaviour. Moreover, microbes generally evolve in volatile environments that, together with demographic fluctuations (birth and death events), drastically alter population size and strain survival. Motivated by the need to better understand the evolution of AMR, we study a population of time-varying size consisting of two competing strains, one drug-resistant and one drug-sensitive, subject to demographic and environmental variability. This is modelled by a binary carrying capacity randomly switching between mild and harsh environmental conditions, and driving the fluctuating volume (total amount of nutrients and antimicrobials at fixed concentration), and thus the size of the community (number of resistant and sensitive cells). We assume that AMR is a shared public good when the concentration of resistant cells exceeds a fixed {\it concentration cooperation threshold}, above which the sensitive strain has a growth advantage, whereas resistant cells dominate below it. Using computational means, and devising an analytical treatment (built on suitable quenched and annealed averaging procedures), we fully characterise the influence of fluctuations on the eco-evolutionary dynamics of AMR, and notably obtain specific strain fixation and long-lasting coexistence probabilities as a function of the environmental variation rate and cooperation threshold. We find that microbial strains tend to coexistence, but demographic fluctuations eventually lead to the extinction of resistant or sensitive cells for small or large values of the concentration cooperation threshold, respectively. This also holds for dynamic environments, whose specific properties determine the extinction timescale.

Short-term extreme value prediction for the structural responses of the IEA 15 MW offshore wind turbine under extreme environmental conditions

As offshore wind turbines enter deep seas, large-scale models ranging from 15 MW to 20 MW are becoming the mainstream choice for the future market. The dynamic responses of 15 MW wind turbines under extreme environmental conditions should be considered in the design process. With increasing structure sizes, the mooring systems and tower base structures play essential roles in maintaining position and supporting wind turbines under harsh environmental conditions. Accurate estimation of the extreme mooring loads and dynamic responses at the tower base is crucial, particularly for the ultimate limit state (ULS) analysis. The dynamic responses of the offshore wind turbine are calculated by Open Fast and validated with other numerical tools. For the safety of deep-sea structures, it is necessary to consider the extreme environmental conditions with a 100-year return period, which is beyond the current standards. An environmental contour-based approach is proposed to select the short-term extreme conditions, enabling approximate prediction of the extreme values of the dynamic responses during the preliminary design stage. The Gumbel method and the ACER method are used to predict the short-term extreme dynamic responses. Comparative evaluations against the relevant standards for floating wind turbines are conducted to assess structural safety.

Food ethnobotany of forest resource in the high-altitude Himalaya Mountains: Enhancing the food sovereignty of ethnic groups

The vital knowledge of traditional foraging techniques used in indigenous peoples' cuisines may be preserved through gastronomical field studies before it is lost forever. Certain plants are appreciated for their gastronomic, social, economic and ecological importance in the High Himalaya because they play such a significant role in the daily life of the local indigenous populations. To determine the plant cultural indicators for each ethnic group, the data were analyzed using an indicator species analysis. Semi-structured interviews of 167 participants, focus group discussion, and field observations were used to gather information from February 2019 to August 2022 for a comprehensive and systematic gastronomical survey. Across four ethnic groups, a total of (N = 99) wild edible plant species and (N = 9) fungi species from 38 families were documented. Tender leaves were the most commonly used, while tubers were the least commonly used. The majority of the species reported were used as vegetables, followed by species used as fruits and herbal tea. For the first time, we examined cross-cultural similarity and dissimilarity for gastronomic usage: vegetables and savoring plants,fruits, and herbal tea across the cultures studied from the perspective of food ethnobotany. We discovered a clear difference in how different cultures used forest resources, with Gujjar and Pahari showing the most species similarity. This is because these ethic groups are culturally linked with one another and share a common social structure, geography, and language. We calculated plant cultural markers for each species, and 19 of them had significant p-values among the four ethnic groups. The findings revealed a positive relationship between the Gujjar and Pahari ethnic groups and the gastronomical indicator values, while the Kashmiri ethnic group had a negative relationship. The Kashmiri ethnic group experienced the loss of traditional knowledge because of the economic transition that caused people to prefer purchasing vegetables from the market rather than foraging for them in the wild. Traditional knowledge was also dwindling between the Gujjar and Pahari ethnic groups because of social changes brought on by developmental activities that changed their way of life. The identified plant cultural markers were significant to local residents and frequently had adaptations to extremely harsh climatic and environmental conditions. Our finding that 47% of species had market value suggests that these species have a sizable market demand and a promising economic value. They have the potential to boost food security, local community sovereignty, and people's living standards. Food production systems may become more resilient and sustainable by incorporating these indicator species into conventional agriculture. The current study raises awareness about forest resources and enables ethnic groups to live in a natural environment without food scarcity.

Its use can reduce the demand for natural resources, extend the service life of structures, and reduce waste generation, aligning with the direction of sustainable development and green building. This makes it an ideal choice for constructing large-span lightweight structures and structures operating in harsh environmental conditions, attracting extensive research attention in the international engineering materials field. In summary, RPC, as a new type of concrete material, has broad application prospects in the construction field. With continuous research and technological development, RPC will undoubtedly bring more innovations and new solutions to the construction industry, promoting its continuous progress and development.

Neanderthal resilience and adaptability: insights from the Abric Pizarro faunal assemblage during the MIS 4

The examination of faunal assemblages through zooarchaeological analyses constitutes a fundamental approach for gaining insight into the intricate behaviours of Neanderthals. Previous investigations have primarily focused on periods of relative environmental stability, and this has provided a wealth of relevant archaeological data. However, our understanding of Neanderthal resilience during the MIS 4, a period presumably characterised by harsh environmental conditions, remains limited. This study presents the first comprehensive analysis of the faunal assemblages from Levels M and P at Abric Pizarro. The geographic location of Abric Pizarro in the southeast Pre-Pyrenees, combined with chronometric dating, offers a unique opportunity to explore Neanderthal behaviours during a poorly known chronological period. The detailed zooarchaeological analysis comprised taxonomic identification, taphonomic analysis and age-at-death profiling to explore the adaptability and flexibility in the Neanderthal diet. The findings indicate that Neanderthal groups incorporated a diverse range of protein resources from small herbivores (e.g., caprids) to very large herbivores (e.g., Bos/Bison). These results not only demonstrate an adaptability to changing environments in an area traditionally deemed unsuitable for long-term occupation, but also contributes significantly to our understanding of the complex behaviours exhibited by Neanderthals.

Nutritional Quality and Socio-Ecological Benefits of Mare Milk Produced under Grazing Management.

This review discusses the scientific evidence that supports the nutritional value of mare milk and how its properties are essentially achieved when mares are managed under grazing conditions. Mare milk's similarity with the chemical composition of human milk makes this food and its derived products not only suitable for human consumption but also an interesting food regarding human health. The contribution of horse breeding under grazing management to other socio-ecological benefits generated by equine farms is also highlighted. Both the high added value of mare milk and the socio-ecological benefits derived from pasture-based systems could be explored to improve the performance of equine farms located in arid and semi-arid areas or in regions with moderately harsh environmental conditions as equids have a strong adaptation capacity.

Characterization of a CZT-based spectrometer for underwater operation via simulations and experiments

The harsh environmental conditions in the marine environment pose variousconstraints on developing efficient instruments to carry out long-term, in situradioactivity measurements. In addition, the strong attenuation of γ-raysin the water medium, makes remote sensing of such radiation a challenging task.In the present work, we report on the efforts to find the optimal characteristicsand deployment scenarios of a new prototype γ-ray instrument based on asmall-size CZT crystal enclosed in seal-tight housing to be deployed for operationin large depths. Lab experiments and detailed Monte Carlo simulations were combinedto validate the actual crystal dimensions, determine its efficiency and energyresolution, as well as establish the minimum detectable activity values of theinstrument in different configurations and scenarios.

The feasibility of in-situ resource utilisation binder systems for construction materials on Mars: A review

The harsh environmental conditions on Mars require building materials that are highly resilient. The cost of transporting binder ingredients for construction materials to Mars is very high. In-situ resource utilisation (ISRU) of Martian resources can help minimise the transportation costs. This review discusses the proposed direction of future investigations to achieve high resiliency and minimal cost for Martian ISRU-based binders. The Martian ISRU-based binders can be bio-based, magnesium-based, polymer-based, geopolymer-based, metal-based or sulphur-based. The review focuses strictly on studies related to the investigation of binder systems produced by utilising simulated in-situ Martian resources. It is divided into sections based on the characteristics of the binder system. Important properties that are investigated include rheological properties, mechanical properties, radiation-shielding properties, and abrasion resistance. The recommended research directions for each Martian ISRU-based binder and other future works related to the key properties are described at the end of this manuscript. This review provides an overview of the potential directions of future research and strongly advocates for additional studies that simulate the behaviour of potential Martian ISRU-based binder systems for use in construction applications in the harsh environment of Mars.

OsPRMT6a-mediated arginine methylation of OsJAZ1 regulates jasmonate signaling and spikelet development in rice

Although both protein arginine methylation (PRMT) and jasmonate (JA) signaling are crucial for regulating plant development, the relationship between these processes in the control of spikelet development remains unclear. In this study, we used CRISPR/Cas9 technology to generate two OsPRMT6a loss-of-function mutants that exhibit various abnormal spikelet structures. Interestingly, we found that OsPRMT6a can methylate arginine residues in JA signal repressors OsJAZ1 and OsJAZ7. We showed that arginine methylation of OsJAZ1 enhances the binding affinity of OsJAZ1 with the JA receptors OsCOI1a and OsCOI1b in the presence of JAs, thereby promoting the ubiquitination of OsJAZ1 by the SCFOsCOI1a/OsCOI1b complex and degradation via the 26S proteasome. This process ultimately releases OsMYC2, a core transcriptional regulator in the JA signaling pathway, to activate or repress JA-responsive genes, thereby maintaining normal plant (spikelet) development. However, in the osprmt6a-1 mutant, reduced arginine methylation of OsJAZ1 impaires the interaction between OsJAZ1 and OsCOI1a/OsCOI1b in the presence of JAs. As a result, OsJAZ1 proteins become more stable, repressing JA responses, thus causing the formation of abnormal spikelet structures. Moreover, we discovered that JA signaling reduces the OsPRMT6a mRNA level in an OsMYC2-dependent manner, thereby establishing a negative feedback loop to balance JA signaling. We further found that OsPRMT6a-mediated arginine methylation of OsJAZ1 likely serves as a switch to tune JA signaling to maintain normal spikelet development under harsh environmental conditions such as high temperatures. Collectively, our study establishes a direct molecular link between arginine methylation and JA signaling in rice.
