Immunohistochemical localization of histamine neuropeptide in the brain neurosecretory cells of tasar silkworm, Antheraea mylitta (DRURY)

Histamine is a putative neurotransmitter in the mammalian central nervous system where it plays a modulatory function. Recently, there has also been growing interest in histamine as a putative neurotransmitter in invertebrates and it has now been implicated as a neurotransmitter in molluscs. In the present study, the distribution and localization of histamine immunoreactive cells were investigated by using polyclonal antibody against the histamine during the larval development of tropical tasar silkworm, Antheraea mylitta. The result confirmed the presence of histamine immunoreactive cells in the brain of larvae. During the development of first to fifth larval instars, a pair of four groups of neurosecretory cells (MNC, LNC, PNC and VNC) were observed in the brain of larvae. In the present study it has been found that only the MNC group shows positive reactivity with anti-histamine antibody, during immunohistochemical reaction.

Insulin-like peptides involved in photoperiodism in the aphid Acyrthosiphon pisum

Aphids were the first animals reported as photoperiodic as their life cycles are strongly determined by the photoperiod. During the favourable seasons (characterised by long days) aphid populations consist exclusively of viviparous parthenogenetic females (known as virginoparae). Shortening of the photoperiod in autumn is perceived by aphids as the signal that anticipates the harsh season, leading to a switch in the reproductive mode giving place to the sexual morphs (oviparae females and males) that mate and lay winter-resistant (diapause-like) eggs. The molecular and cellular basis governing the switch between the two reproductive modes are far from being understood. Classical experiments identified a group of neurosecretory cells in the pars intercerebralis of the aphid brain (the so called group I of neurosecretory cells) that were essential for the development of embryos as parthenogenetic females and were thus proposed to synthesise a parthenogenesis promoting substance that was termed “virginoparin”. Since insulin-like peptides (ILPs) have been implicated in the control of diapause in other insects, we investigated their involvement in aphid photoperiodism. We compared the expression of two ILPs (ILP1 and ILP4) and an Insulin receptor coding genes in A. pisum aphids reared under long- and short-day conditions. The three genes showed higher expression in long-day reared aphids. In addition, we localised the site of expression of the two ILP genes in the aphid brain. Both genes were found to be expressed in the group I of neurosecretory cells. Altogether, our results suggest that ILP1 and ILP4 play an important role in the control of the aphid life-cycle by promoting the parthenogenetic development during long-day seasons while their repression by short days would activate the sexual development. Thus we propose these ILPs correspond to the so called “virginoparin” by early bibliography. A possible connection with the circadian system is also discussed.

Fine structure of neurosecretory cells and sinus gland in the eyestalk of the freshwater crab Travancoriana schirnerae Bott, 1969 (Decapoda: Gecarcinucidae)

This study elucidated the fine structure of neurosecretory cells and sinus gland in the optic ganglia of the freshwater crab Travancoriana schirnerae Bott, 1969 (Decapoda: Gecarcinucidae). The eyestalk ganglion showed the presence of four well defined ganglia arranged below the ommatidium: lamina ganglionaris, medulla externa, medulla interna and medulla terminalis of which the lamina ganglionaris, was devoid of neurosecretory cells. Groups of neurosecretory cells seen distributed along the medulla externa, interna and terminalis regions constitute the X-organs. Electron microscopic observations of the eyestalk ganglia revealed ten types of neurosecretory cells, mostly apolar with a few unipolar and bipolar cells classified according to the size, shape and density of the cell and nucleus, cell organelles/inclusions, together with the arrangement and properties of chromatin. These cells were characterized by the presence of large nuclei with unusually condensed chromatin, inclusions like vacuoles and vesicles of varying size, shape and density and organelles like Golgi, endoplasmic reticulum, ribosomes and mitochondria and neurosecretory material. The sinus gland of T. schirnerae was positioned laterally between the externa and interna regions, composed of axonal endings of the neurosecretory cells of the optic ganglia with interspersed glial cells. The axon terminals were enclosed with several small to large membrane bound homogenously dense neurosecretory granules which also occur in the preterminal areas of the axons. Based on size, shape and density of granules and axoplasmic matrix, seven terminal types could be distinguished in the sinus gland of T. schirnerae. Mostly, the granules contained in a terminal were of the same type; rarely, the same terminal enclosed granules of varying size, shape and density. The neurosecretory cell types and axon terminal types represent the types of neurohormones they contained. A precise knowledge of the morphology and cytology of neurosecretory cells in the XO-SG complex of the eyestalk that secrete neurohormones controlling major physiological processes such as growth and reproduction is imperative for successful captive breeding of a species of aquaculture potential.

Local neuropeptide signaling modulates serotonergic transmission to shape the temporal organization of C. elegans egg-laying behavior.

Animal behaviors are often composed of distinct alternating behavioral states. Neuromodulatory signals are thought to be critical for establishing stable behavioral states and for orchestrating transitions between them. However, we have only a limited understanding of how neuromodulatory systems act in vivo to alter circuit performance and shape behavior. To address these questions, we have investigated neuromodulatory signaling in the context of Caenorhabditis elegans egg-laying. Egg-laying activity cycles between discrete states–short bursts of egg deposition (active phases) that alternate with prolonged quiescent periods (inactive phases). Here using genetic, pharmacological and optogenetic approaches for cell-specific activation and inhibition, we show that a group of neurosecretory cells (uv1) located in close spatial proximity to the egg-laying neuromusculature direct the temporal organization of egg-laying by prolonging the duration of inactive phases. We demonstrate that the modulatory effects of the uv1 cells are mediated by peptides encoded by the nlp-7 and flp-11 genes that act locally to inhibit circuit activity, primarily by inhibiting vesicular release of serotonin from HSN motor neurons. This peptidergic inhibition is achieved, at least in part, by reducing synaptic vesicle abundance in the HSN motor neurons. By linking the in vivo actions of specific neuropeptide signaling systems with the generation of stable behavioral outcomes, our study reveals how cycles of neuromodulation emanating from non-neuronal cells can fundamentally shape the organization of a behavioral program.

Localization and identification of crustacean hyperglycemic hormone producing neurosecretory cells in the eyestalk of blue swimmer crab, Portunus pelagicus.

This study intensely focuses on to the localization and identification of crustacean hyperglycemic hormone (CHH) producing neurosecretory cells in the eyestalk of the blue swimmer crab Portunus pelagicus. Anti-Carcinus maenas-CHH was used to identify the location of CHH neurosecretory cells by immunohistochemistry. Ten pairs of eyestalks were collected from intact adult intermoult female crab and fixed in Bouin's fixative. Eyestalks were serially sectioned and stained with chrome-hematoxylin-phloxine stain. Histological studies show the presence of different types of neurosecretory cells namely A (multipolar), B (tripolar), C (bipolar), D (unipolar), E (oval), and F (spherical) in the medulla interna, externa, and terminalis regions based on their size, shape, and tinctorial properties. The neurohemal organ, sinus gland (SG) was observed laterally between medulla interna and terminalis regions. Immunohistochemical studies showed the presence of distinct CHH-like immunoreactivity in the optic ganglia. Divergent group of neurosecretory cells with varying degree of immunoreactivity with Anti-Carcinus maenas-CHH (low, moderate, and intense reactivity) were identified in medulla terminalis, medulla interna, medulla externa, and sinus gland. The present study maps the various types of neurosecretory cells in the optic ganglia and also shows the presence of CHH-like immunoreactivity in various regions of optic ganglia in P. pelagicus. The presence of these unique neurosecretory cell types with larger cell diameter in medulla terminalis, a region that bears the neurosecretory cell bodies, suggest high secretory activity.

Ultrastructural studies on the neurosecretory cells in the pars intercerebralis of larvae of tasar silkworm, Antheraea mylitta (D) eco-race Bhandara (Lepidoptera: Saturniidae)

The cerebral neurosecretory cells (NSC) constitute four paired groups, medial (MNC), lateral (LNC-I, LNC-II) and posterior (PNC) in the brain of larvae of tasar silkworm Antheraea mylitta (D) Eco-race Bhandara. The MNC is the largest groups of peptidergic neurosecretory cells and are located in the pars intercerebralis region. The transmission electron microscopic (TEM), ultrastructure of the NSC confirmed the presence of mitochondria, endoplasmic reticulum, Golgi bodies, lysosomes and neurosecretory granules. The median neurosecretory cells shows secretory activity and release of secretory products, the neurosecretory granules (NSG) in the axons of NSC as well as the blood sinus.

A Small Group of Neurosecretory Cells Expressing the Transcriptional Regulatoraponticand the NeuropeptidecorazoninMediate Ethanol Sedation in Drosophila

In the fruit fly Drosophila melanogaster, as in mammals, acute exposure to a high dose of ethanol leads to stereotypical behavioral changes beginning with increased activity, followed by incoordination, loss of postural control, and eventually, sedation. The mechanism(s) by which ethanol impacts the CNS leading to ethanol-induced sedation and the genes required for normal sedation sensitivity remain largely unknown. Here we identify the gene apontic (apt), an Myb/SANT-containing transcription factor that is required in the nervous system for normal sensitivity to ethanol sedation. Using genetic and behavioral analyses, we show that apt mediates sensitivity to ethanol sedation by acting in a small set of neurons that express Corazonin (Crz), a neuropeptide likely involved in the physiological response to stress. The activity of Crz neurons regulates the behavioral response to ethanol, as silencing and activating these neurons affects sedation sensitivity in opposite ways. Furthermore, this effect is mediated by Crz, as flies with reduced crz expression show reduced sensitivity to ethanol sedation. Finally, we find that both apt and crz are rapidly upregulated by acute ethanol exposure. Thus, we have identified two genes and a small set of peptidergic neurons that regulate sensitivity to ethanol-induced sedation. We propose that Apt regulates the activity of Crz neurons and/or release of the neuropeptide during ethanol exposure.

Gene dosage effects on hypothalamic visceromotor cell types (Commentary on Duplan et al.)

The paraventricular nucleus of the hypothalamus (PVN) is a mosaic of anatomically and chemically specified cell types, in which each of the major avenues by which the brain regulates visceromotor function is represented amply, and in a topographically ordered manner (Swanson & Sawchenko, 1983). These include (i) magnocellular neurosecretory neurons that synthesize the peptide hormones, oxytocin (OT) and vasopressin (AVP), for release into the general circulation via the posterior pituitary, (ii) parvicellular neurosecretory cells that serve as the central arms of the hypothalamo-pituitary-adrenal and -thyroid axes, which are broadly involved in adaptations to stress and metabolic challenges, respectively, and (iii) neurons that project within the CNS to cell groups involved in central autonomic control, including vagal and sympathetic preganglionic neurons and the nucleus of the solitary tract (NTS), the principal CNS recipient of primary visceral sensory information. This latter, ‘pre-autonomic,’ contingent is not known to possess a distinctive neurochemical signature; instead, small subsets of it share phenotypes with adjoining neurosecretory populations, with OT, AVP and corticotropin-releasing factor predominating. As these output neuron classes may be called into play in various combinations to promote adaptation to specific physiologic conditions, the PVN has become an obligatory focus for studying integrative aspects of hypothalamic function. A key factor in the specification of PVN cellular phenotypes during development is Sim1, a basic helix-loop-helix-PAS nuclear transcription factor identified by homology with a Drosophila gene, single-minded, which regulates CNS midline development. Murine Sim1 is required for the development of virtually all neurons in the PVN and the related supraoptic nucleus (Michaud et al., 1998), a pure magnocellular neurosecretory cell group. Whereas sim1 null mice die shortly after birth, heterozygous sim1 mutants are viable, and develop early onset hyperphagia and obesity (Michaud et al., 2001). Sim1 is, in fact, one of a handful of genes linked with monogenic human obesity. In this issue of EJN, Duplan and colleagues (2009) further explore the developmental and functional consequences of sim1 haploinsufficiency, finding that two major classes of PVN visceromotor neurons are compromised. They report that sim1+/− mice display substantially reduced complements of AVP- and, particularly, OT-immunoreactive cells, and reduced mRNA expression, while major parvicellular neurosecretory PVN phenotypes are ostensibly unaffected (cf. (Kublaoui et al., 2008). Sim1 heterozygotes are as capable as wild type mice of defending plasma osmolality under milder dehydration conditions (substituting hypertonic saline for drinking water, but not more strenuous ones, presumably as a result of diminished natriuretic and water-retaining capabilities deriving from partial loss (or dysfunction) of magnocellular OT and AVP populations. The effects of reduced sim1 gene dosage were not limited to magnocellular cells, as heterozygotes also displayed a >70% reduction in PVN neurons labeled following retrograde tracer injections in a major target of autonomic-related PVN outputs, the dorsal vagal complex (NTS and dorsal motor nucleus of the vagus). These new findings direct particular attention to the role of Sim1 in the development of preautonomic PVN outputs, their potential role in the obese phenotype of sim1+/− mice, and more generally in the control of food intake and obesity. The projection to the dorsal vagal complex, and particularly its OT component, has been implicated in feeding inhibition, by modulating the sensitivity of the NTS to visceral sensory cues (Blevins et al., 2004), including the anorexia associated with chronic dehydration (Rinaman et al., 2005). This may well relate to the new finding that sim1+/− mice eat less than wild type counterparts under dehydration conditions that similarly affect plasma osmolality in the two groups. In this light, recent evidence that central administration of OT can normalize food intake and weight gain of sim1+/− mice (Kublaoui et al., 2008) supports an involvement of such a mechanism in the obese phenotype associated with sim1 haploinsufficiency. Additional contributors remain to be identified. Only fragmentary information is available as to the extent to which the phenotypic diversity that characterizes autonomic-related PVN projections may reflect a degree of chemical coding within them. In addition, because of the extensive degree of axon collateralization within these pathways (Swanson & Kuypers, 1980), the involvement of other targets, notably vagal and sympathetic preganglionic neurons, remains to be evaluated. Interpreting the effects of reduced sim1 dosage by germ line manipulation is complicated by the findings that viral vector-mediated modulation of sim1 expression in the PVN can increase or decrease food intake in mature wild type mice (Yang et al., 2006). As noted by Duplan et al. (2009), conditional gene disruption in postnatal animals will be required to isolate the postnatal role(s) of Sim1, whose transcriptional target genes are not well characterized. Similar approaches, likely exploiting axonal transport methods to achieve differential targeting of functionally differentiated PVN populations that may share neurochemical phenotypes, will be needed to parse the roles of paraventriculo-autonomic projections in visceral sensory and motor functions involved in regulating appetitive behavior and metabolism.

Sex pheromone composition of the cotton caterpillar, Palpita indica (Lepidoptera: Pyralidae), in Korea

This study was conducted to investigate the sex pheromone composition of Palpita (= Diaphania) indica (Saunders) (Lepidoptera: Pyralidae) in Korea. Two sex pheromone components, E11-16:Al and E,E-10,12-16:Al, were identified by GC and GC-EAD analysis of sex pheromone gland extracts. The mean ratios of the two components, E11-16:Al and E,E-10,12-16:Al, were about 6.5:3.5 and 7.5:2.5 in gland extracts and in SPME collection, respectively. In field bioassays, maximum attraction occurred in traps baited with a 7:3 ratio of E11-16:Al and E,E-10,12-16:Al. The head extracts of P. indica stimulated the sex pheromone production of P. indica, as well as Hez-PBAN and PssPT, indicating that a PBAN-like substance exists in the head extracts of P. indica. Whole-mount immunocytochemistry showed that three groups of neurosecretory cells showed PBAN-like immunoreactivity in the SEG of P. indica. The PBAN-like immunoreactivity connected to the Corpora Cardiaca, a neurohemal organ. Also, PBAN-like immunoreactivity was found in the brain and in the thoracic and third/fourth abdominal ganglia. The addition of sex pheromone components of Peridroma saucia to the sex pheromone of P. indica greatly improved the attraction of P. indica males. The mixing of the sex pheromone components of S. exigua did not significantly increase the catches of P. indica, while the sex pheromone of H. armigera completely inhibited the attractiveness.

Identification of novel neuropeptides in the ventral nerve cord ganglia and their targets in an annelid worm, Eisenia fetida

AbstractPeriviscerokinins (PVKs) and pyrokinins (PKs) are neuropeptides known in several arthropod species. Sequence homology of these peptides with the molluscan small cardioactive peptides reveals that the occurrence of PVKs and PKs is not restricted to arthropods. Our study focuses on the biochemical and immunocytochemical identification of neuropeptides with sequence homology to PVKs and PKs in the central and peripheral nervous system of the earthworm Eisenia fetida. By means of affinity chromatography, nanoflow liquid chromatography, and high accuracy mass spectrometry, six peptides, SPFPR(L/I)amide, APFPR( L/I)amide, SPLPR( L/I)amide, SFVR( L/I)amide, AFVR( L/I)amide, and SPAFVR( L/I)amide, were identified in the central nervous system with the common‐XR( L/I)amide C‐terminal sequence. The exact anatomical position of 13 labeled XR( I/L)amide expressing neuron groups and numerous peptide‐containing fibers were determined by means of immunocytochemistry and confocal laser scanning microscopy in whole‐mount preparations of ventral nerve cord ganglia. The majority of the stained neurons were interneurons with processes joining the distinct fine‐fibered polysegmental tracts in the central neuropil. Some stained fibers were seen running in each segmental nerve that innervated metanephridia and body wall. Distinct groups of neurosecretory cells characterized by small round soma and short processes were also identified. Based on immunoelectron microscopy six different types of labeled cells were described showing morphological heterogeneity of earthworm peptides containing elements. Our findings confirm that the sequence of the identified earthworm neuropeptides homologous to the insect PVKs and PKs suggesting that these peptides are phylogenetically conservative molecules and are expressed in sister‐groups of animals such as annelids, mollusks, and insects. Neurosecretory cells in the optic lobes of the brain and activity rhythms in Lycosa tarentula (Araneae: Lycosidae)

Several paired groups of neurosecretory cells (NS) were identified in the dorsal cortical neurons of the optic lobes of the brain of Lycosa tarentula (Araneae). Two large bottle-shaped cells (NS A1, A2) and a cluster of ca. 20 smaller cells (NS B) were found between the lamina and medulla of the anterior median eyes (AM). The forward oriented bundles of NS B axons run alongside large fibres linked to the synaptic zones of the indirect eyes. In front of the arcuate body, an islet of about 10 fusiform cells (NS C1) sends short axons close to the internal cortical border. Other large cells (NS C2, C3) are found from the medulla of the AM to the anterior border of the central body. Their long axons end deeply in the brain neuropil. NS B and C1 function synchronously. The secretory cycles of NS A1 and A2 seem to be in opposition. The activity of these three types of NS depends on the phase of the day. Anatomical relationships of NS A, B and C1 with visual afferent/efferent fibres via synaptic buttons indicate a role of these cells in the modulation of circadian rhythms of visual and locomotor activity. On the other hand, NS C2 and C3, the functioning of which is not synchronous, might be involved in the modulation or control of the elementary movements of L. tarentula when active or at rest.

Enhanced fos expression in the zebra finch (Taeniopygia guttata) brain following first courtship.

Young zebra finch males that court a female for the first time develop a stable preference for the females of that species. On the neuronal level, consolidation of the imprinted information takes place. Here we demonstrate that first courtship or being chased around in the cage leads to enhanced fos expression in forebrain areas implicated in learning and imprinting in zebra finch males compared with birds reared in isolation or in the aviary. Two of the forebrain areas highly active during first courtship (as demonstrated by the 14C-2-deoxyglucose technique), the imprinting locus latral neo/hyperstriatum ventrale (LNH) and the secondary visual area hyperstriatum accessorium/dorsale (HAD), demonstrate enhanced fos expression. Two other imprinting-related areas, the medial neo/hyperstriatum ventrale (MNH) and archistriatum/neostriatum caudale (ANC), do show c-fos induction; however, the areas are not congruous with those demarcated by the 2-DG autoradiographic studies. Additional telencephalic areas include the olfactory lobe, the information storage site lobus parolfactorius (LPO), the memory site hippocampus, the auditory caudomedial neostriatum implicated in the strength of song learning, and the caudolateral neostriatum, which is comparable to the mammalian prefrontal cortex. In addition, c-fos is induced by first courtship and chasing in neurosecretory cell groups of the preoptic area and hypothalamus associated with the repertoire of sexual behavior and stress or enhanced arousal. Enhanced fos expression is also observed in brainstem sources of specific (noradrenergic, catecholaminergic) and nonspecific (reticular formation) activating pathways with inputs to higher brain areas implicated in the imprinting process. Birds reared in isolation or alternatively in the aviary with social and sexual contact to conspecifics showed attenuated or no fos expression in most of the above-mentioned areas. First courtship and chasing both lead to enhanced uptake of 2-DG in the four imprinting areas, as well as subsequent changes in spine density-an anatomical manifestation of the imprinting process. fos expression in the imprinting and other telencephalic, preoptic, hypothalamic, and mesencephalic brain regions indicates processing of stimuli originating from exposure (like chasing) and the analysis of stimuli in a behaviorally relevant, sexually explicit context (like first courtship). c-fos induction in these brain areas indicates its involvement in the triggering of neural changes that accompany the learning process of imprinting, leading eventually to alterations in dendritic spine density in the zebra finch.

Cerebroid Ganglion is the Presumptive Pacemaker of the Circadian Rhythm of Electrical Response to Light in the Crayfish

One of the most widely studied circadian rhythms in invertebrates is that of light responsiveness whose underlying mechanisms seem to involve different groups of oscillators which act as pacemakers. Although, in crayfish, there are clear circadian rhythms in the electroretinogram (ERG) amplitude, the precise location of the pacemaker system driving this rhythm is uncertain. Some data suggest that the circadian pacemaker could be located in a group of neurosecretory cells of the supraesophageal ganglion (the cerebroid ganglion or brain) and that the sinus gland plays a determinant role in the generation and expression of this rhythm through periodic release of pigment-dispersing hormone (PDH). The aim of this work is to examine the role of the brain in the expression of the ERG circadian activity. The hypothesis we test is that the electrical activity at the brain level has a circadian behavior in the firing pattern of spontaneous multiunit activity (MUA) and in visual evoked potentials (VEPs). The results indicate that there are robust circadian rhythms in both MUA, recorded from several regions of the brain, and in the averaged VEPs recorded from the protocerebrum area. These rhythms are 180° out of phase to one another. The rhythm of VEPs showed a main peak at midnight which was in close phase relationship with the ERG amplitude rhythm.

Cardioacceleratory action of tachykinin-related neuropeptides and proctolin in two coleopteran insect species ☆,☆☆

Several cardioactive peptides have been identified in insects and most of them are likely to act on the heart as neurohormones. Here we have investigated the cardioactive properties of members of a family of insect tachykinin-related peptides (TRPs) in heterologous bioassays with two coleopteran insects, Tenebrio molitor and Zophobas atratus. Their effects were compared with the action of the pentapeptide proctolin. We tested the cardiotropic activity of LemTRP-4 isolated from the midgut of the cockroach Leucophaea maderae, CavTK-I and CavTK-II isolated from the blowfly Calliphora vomitoria. The semi-isolated hearts of the two coleopteran species were strongly stimulated by proctolin. We observed a dose dependent increase in heartbeat frequency (a positive chronotropic effect) and a decrease in amplitude of contractions (a negative inotropic effect). In both beetles the TRPs are less potent cardiostimulators and exert lower maximal frequency responses than proctolin. LemTRP-4 applied at 10 −9–10 −6 M was cardiostimulatory in both species inducing an increase of heart beat frequency. The amplitude of contractions was stimulated only in Z. atratus. CavTK-I and CavTK-II also exerted cardiostimulatory effects in Z. atratus at 10 −9–10 −6 M. Both peptides stimulated the frequency, but only CavTK-II increased the amplitude of the heart beat. In T. molitor, however, the CavTKs induced no significant effect on the heart. Immunocytochemistry with antisera to the locust TRPs LomTK-I and LomTK-II was employed to identify the source of TRPs acting on the heart. No innervation of the heart by TRP immunoreactive axons could detected, instead it is possible that TRPs reach the heart by route of the circulation. The likely sources of circulating TRPs in these insects are TRP-immunoreactive neurosecretory cells of the median neurosecretory cell group in the brain with terminations in the corpora cardiaca and endocrine cells in the midgut. In conclusion, LemTRP-4, CavTK-I and CavTK-II are less potent cardiostimulators than proctolin and also exert stimulatory rather than inhibitory action on amplitude of contractions. The differences in the responses to proctolin and TRPs suggest that the peptides regulate heart activity by different mechanisms.

Baratin, a nonamidated neurostimulating neuropeptide, isolated from cockroach brain: Distribution and actions in the cockroach and locust nervous systems

During the purification of tachykinin-related peptides from the brain of the cockroach Leucophaea maderae, a few other peptides were collected in adjacent high-performance liquid chromatography fractions. Edman degradation, mass spectrometry, and chemical synthesis revealed that one of these peptides had the sequence DNSQWGGFA. This nonamidated nonapeptide was designated baratin and appears not to be related to any known insect peptide. Baratin was not found to be bioactive in the L. maderae hindgut or oviduct muscle contraction assay. (Both synthetic nonamidated and amidated baratin were tested.) To screen for possible sites of action, we raised a rabbit antiserum to baratin. We found baratin-immunoreactive (BAR-IR) interneurons throughout the cockroach central nervous system. Some prominent brain neuropils were supplied by BAR-IR neuron processes: the central body, the calyx, and lobes of the mushroom bodies, parts of the optic lobe, and the tritocerebral neuropil. Additionally we found BAR-IR neurosecretory cells in the median neurosecretory cell group with processes supplying the storage lobe of the corpora cardiaca. In each of the thoracic and abdominal ganglia processes of BAR-IR projection neurons and local neurons were seen. The baratin antiserum also labeled neurons in the brain of the locust Locusta migratoria, some of which are similar to those of the cockroach. A prominent system of interganglionic BAR-IR processes was found in the locust subesophageal, thoracic, and abdominal ganglia. This was formed by four large projection neurons with cell bodies in the abdominal ganglia A1-2. The processes of these BAR-IR neurons are distributed dorsally and laterally in each of the ventral nerve cord ganglia. When baratin (10(-6)-10(-4) M) was applied to desheathed abdominal ganglia of locusts and cockroaches, we could monitor bursts of action potentials in neurons with axons in the anterior abdominal nerve (nerve 1), but not in the posterior nerve (nerve 2). In ganglia displaying spontaneous rhythmic firing in units of nerve 1, baratin strengthened the rhythmic pattern. Thus baratin appears to have a role in modulation of motor patterns in abdominal ganglia. The immunocytochemical findings suggest further modulatory actions of baratin in different circuits of the brain and ventral nerve cord.

Subcortical projections of area 25 (subgenual cortex) of the macaque monkey

In several species, including primates, stimulation studies indicate that the infralimbic cortex, the most caudal part of the ventromedial prefrontal cortex, functions as a visceral motor region. In addition, recent positron emission tomography studies implicate the subgenual region in depression and mania. To determine the subcortical projections of this region in primates, injections of Phaseolus vulgaris leukoagglutinin, biotinylated dextran amine, or rhodamine-labeled dextran amine were placed in area 25 in three monkeys. In contrast to the efferents from area 25 previously described in the rat, there were no projections to autonomic effector regions, such as the nucleus of the solitary tract, magnocellular neurosecretory cell groups in the hypothalamus, ventrolateral medulla, or intermediolateral column of the spinal cord. However, projections were shown to a number of structures with probable roles in autonomic function and direct connections to some of the abovementioned autonomic effector regions, including bed nucleus of the stria terminalis, perifornical and anterior hypothalamus, periaqueductal gray, and lateral parabrachial nucleus. In addition, there were projections to several forebrain structures that receive projections from other components of the medial prefrontal network including the medial part of the caudate nucleus, lateral septum, midline and mediodorsal thalamic nuclei, the lateral parvocellular part of the basal accessory amygdaloid nucleus, and the magnocellular part of the basal amygdaloid. None of the injections resulted in labeling in the medulla. These connections support the idea of a role for cortical area 25 in emotional and autonomic responses, albeit less direct than that described in rodents.

Ultrastructural analysis of neurosecretory cells in the antennae of the mosquito, Culex salinarius (Diptera: Culicidae).

An antiserum raised against the peptide, culetachykinin II, immunocytochemically detected a group of neurosecretory cells in the first flagellar segment of the antennae of both males and females of the mosquito, Culex salinarius. This is the first insect species in which neurosecretory cells have been found in the antennae. The ultrastructure of these antennal neurosecretory cells (ANC) is described, as well as their relationship to other neurons in the antennae and antennal lobe of the mosquito. These tachykinin-reactive cells contain relatively small (140-220 nm) elementary neurosecretory granules. Not only do the ANC have axons that terminate on specific glomeruli of the deutocerebrum, but these neurons also have collaterals that form neurohemal terminals in the receptor lymph channels of the dendrites of the sensory neurons. Thus, the ANC not only influence higher centers of the brain that interpret signals from the antennal sensillae, but also modulate the response of the sensory receptors. To our knowledge, this is the first report of neurosecretory cells directly affecting the signal reception of sensory neurons.

A novel peptide-processing activity of insect peptidyl-dipeptidase A (angiotensin I-converting enzyme): the hydrolysis of lysyl-arginine and arginyl-arginine from the C-terminus of an insect prohormone peptide.

Insect peptidyl-dipeptidase A [angiotensin I-converting enzyme (ACE)] is a soluble single-domain peptidyl-dipeptidase that has many properties in common with the C-domain of mammalian somatic ACE and with the single-domain mammalian germinal ACE. Mammalian somatic ACE is important in blood homoeostasis, but the role of ACE in insects is not known. Immunocytochemistry has been used to localize ACE in the neuroendocrine system of the locust, Locusta migratoria. Staining was observed in five groups of neurosecretory cells in the brain and suboesophageal ganglion, in the nervi corpori cardiaci, the storage part of the corpora cardiaca and in the nervi corpori allati. In three groups of neurosecretory cells, ACE co-localized with locustamyotropins, suggesting a possible role for the enzyme in the metabolism of these neuropeptides. We demonstrate in vitro a novel activity of ACE that removes pairs of basic amino acid residues from a locustamyotropin peptide extended at the C-terminus with either Gly-Lys-Arg or Gly-Arg-Arg, corresponding to a consensus recognition sequence for endoproteolysis of prohormone proteins by prohormone convertases. The low Km and high kcat values (Km 7.3 and 5.0 microM, kcat 226 and 207 s-1 for the hydrolysis of Phe-Ser-Pro-Arg-Leu-Gly-Lys-Arg and Phe-Ser-Pro-Arg-Leu-Gly-Arg-Arg, respectively) obtained for the hydrolysis of these two peptides by insect ACE means that these peptides, along with mammalian bradykinin, are the most favoured in vitro ACE substrates so far identified. The discovery of this in vitro prohormone-processing activity of insect ACE provides a possible explanation for the intracellular co-localization of the enzyme with locustamyotropin peptides, and provides evidence for a new role for ACE in the biosynthesis of peptide hormones and transmitters.
