Quantitative proteogenomic characterization in MUC1 and MUC4 in oral squamous cell carcinoma, oral potentially malignant disorders, and normal oral mucosa in carcinogenesis.

Mucins are glycoproteins with a significant molecular weight that have a diverse range of biological functions. MUC1 & MUC4 are transmembrane mucin family members that are expressed in airway epithelial cells and body fluids. It is expressed excessively in numerous carcinomas in addition to their alteredexpression. A thorough review of the literature reveals very limited research about MUC1 & 4 expressionsin oral malignancies and oralpremalignant disorders. The aim of the research is to assess the pattern and quantity of gene expression in MUC1 & MUC4 individually in various grades of oral squamous cell carcinoma (OSCC), oral premalignant disorders (OPMDs)- oral epithelial dysplasia (OED), oral submucous fibrosis (OSF), oral lichen planus (OLP), and compared with normal oral mucosa (NOM). Immunohistochemistry and qRTPCR evaluation of MUC1 & 4 on sixty-three cases of OSCC, OPMD, and NOM was accomplished. The one-way ANOVA test and Chi-square test were done for statistical analysis. The overall immunoexpression of MUC1 & 4 increased significantly from NOM to OPMDs to OSCC. In the subgrades of OSCC and OPMDs, the staining intensity varied. For MUC4, the intensity was strongest in well-differentiated grades and MUC1, exhibiting a higher expression in poorly differentiated grades of OSCC and OPMDs. The study's results indicate that MUC1 & 4 individually are crucial formonitoringOSCC and OPMD pathogenesis as the former gives an idea of highly undifferentiated grades while the latter indicated more differentiated tumors and perhaps a better prognosis. Therefore, the two can be useful tumor markers for determining the severity and eliminating it in its early phases.

Immunohistochemical Expression of Bcl-2, E-cadherin, CD34 and CD105 in Basaloid Squamous Cell Carcinoma - An In Vitro Study

Abstract Introduction: Basaloid squamous cell carcinoma (BSCC) is a rare aggressive variant of oral squamous cell carcinoma (OSCC) with a high propensity for distant metastasis. In this article, we present clinicopathological and survival data of eight cases of BSCC and further analyse the behaviour of these tumours with the help of E-cadherin, CD34, CD105 and B cell lymphoma-2 (Bcl-2) immunoexpression. Materials and Methods: Histopathologically confirmed cases of BSCC were retrieved from the department archives. Clinicopathological details and survival data of these patients were collected. Immunohistochemical analysis was performed with Bcl-2, E-cadherin, CD34 and CD105 on these cases and compared with different grades of OSCC (well differentiated, moderately differentiated and poorly differentiated). The statistical analysis was done using IBM SPSS software version 23. Results: BSCC was seen commonly in males of age group 49–71 years and predominantly reported in the retromolar trigone. Bcl-2 expression was significantly lower in BSCCs when compared to the conventional OSCC groups (P < 0.05). E-cadherin expression showed no significant difference between BSCC and well-differentiated OSCC group (P = 0.487). The overall mean survival for patients with BSCC was 6.37 months. Discussion: BSCCs of the oral cavity show increased CD105, CD34, E-cadherin and low Bcl-2 labelling. A substantial relationship between the tumour neo-angiogenesis, collective cell migration and apoptotic property could be related to the aggressive nature of this tumour and its poor overall survival rate. BSCCs are common in middle to older aged male and show increased expression of CD105, CD34 and E-cadherin.

Evaluation of Tumor-Associated Macrophages and Micro-Vessel Density in Verrucous Carcinoma and Squamous Cell Carcinoma of the Oral Cavity.

Tumor-associated macrophages (TAMs) are a crucial cellular component of human tumors, but their exact roles in tumor growth and angiogenesis during cancer progression are still subject to debate. The present study aimed to explore the impact of TAMs on tumor progression and angiogenesis in oral squamous cell carcinoma (OSCC) and oral verrucous carcinoma (OVC). For this cross-sectional study, 60 samples consisting of 40 OSCC and 20 OVC samples were chosen. Immunohistochemical staining was performed on the samples using anti-CD68 and anti-CD31 monoclonal antibodies to evaluate the frequency of TAMs and micro-vessel density (MVD), respectively. The data were analyzed using statistical software SPSS (v. 23) and through the independent samples Student t-test, one-way analysis of variance, and Pearson's correlation coefficient. The results of our study revealed that the mean number of TAMs and MVD were significantly greater in OSCC samples compared to OVC samples (P = 0.001) (P = 0.004). Also, the average number of TAMs and MVD increased in the high grade of OSCC; however, the correlation between both parameters and histopathological grades of OSCC was significant only for MVD. Our study found no significant correlation between TAMs and MVD (P = 0.005). Based on the results of our study, both TAMs and MVD were found to be significantly higher in OSCC samples compared to OVC samples. These findings suggest that the progression of oral cancer is associated with an increase in TAMs and vascular density. Additionally, the use of CD68 and CD31 markers may help in differentiating between these two types of tumors.

To evaluate the role of mast cells on angiogenesis in various grades of oral squamous cell carcinoma: A histochemical study

Abstract Background: Oral cancer is the sixth most common cancer, and 90% of them are oral squamous cell carcinomas (OSCC). As most OSCC are asymptomatic and are only detected at an advanced stage, the 5-year survival rate is only 50%. Thus, using novel prognosticators can minimise mortality and morbidity associated with OSCC. This study aims to evaluate the relationship between mast cells and angiogenesis in different grades of OSCC to analyse their role in its progression. Material and Methods: A total of 45 cases were included, comprising 10 well-differentiated SCCs (WDOSCC), 10 moderately differentiated SCCs (MDOSCC), and 10 poorly differentiated SCCs (PDOSCC). Additionally, five normal buccal mucosae (NBM) samples served as negative controls for OSCC. Five cases of neurofibroma and pyogenic granuloma were used as positive controls for mast cells and angiogenesis, respectively. Results: The mean MCD in WDOSCC, MDOSCC, and PDOSCC were 3.2620 ± 2.65177, 3.0310 ± 1.38276, and 4.1580 ± 2.49482, respectively. The MVD in WDOSCC, MDOSCC, and PDOSCC were 10.2850 ± 4.35032, 9.9240 ± 2.72533, and 7.1520 ± 2.26966, respectively. Discussion: MCD was the highest in PDOSCC, followed by WDOSCC and MDOSCC. These results indicate a redundant role of mast cells in OSCC, or they might jumpstart malignancy but are retarded with OSCC progression. The MVD decreased with higher grades, in contrast to the prevalent literature. The correlation analysis between MVD and MCD revealed no significant correlation between them. Conclusion: We found a non-significant role of mast cells in tumour biology and a decrease in vascularity with advancing grades. These results indicate a lower need for mast cell activation to augment vascularisation. A study with a larger sample size is needed to confirm our results.

Quantitative correlation of serum and salivary trace elements in oral squamous cell carcinoma and oral potentially malignant disorders: An institution-based biochemical analysis

Abstract Background: Trace elements are the important components that play a crucial role in various functions of the human body at cellular and molecular levels. Considering the role of the trace elements in precancerous and cancerous conditions, estimation of their levels in these conditions could aid in understanding the disease process and progression. The purpose of this study is to determine the alteration in salivary electrolyte concentration of oral potentially malignant disorders (OPMD) and oral squamous cell carcinoma (OSCC) patients to correlate the variations with the severity and biological behaviour. Material and Method: A total of 70 subjects were included in this study, and they were divided into three groups: patients with OSCC (30), OPMDs (30) and apparently healthy individuals (10). An informed consent was obtained, following which blood and saliva samples were collected from the participants. Salivary and serum levels of copper, zinc, lead, cadmium, calcium and magnesium were measured and compared between the groups. Results: The levels of biochemical elements in both serum and saliva were in perfect correlation. The amount of all the estimated metallic ions was found to be significantly (P 0.001) increased in OSCC followed by OPMDs and normal mucosa. The levels of copper, cadmium and magnesium were gradually increased in increasing grades of OSCC (P 0.001). Conclusion: Salivary trace element levels could possibly have diagnostic significance in the early evaluation of OPMDs and OSCC as well. Increased levels of these elements might be used as a marker of disease progression and predictor of prognosis.

Association of Cellular Cannibalism with Immunohistochemical Expression of CD31, CD68 and BCL2 in Oral Squamous Cell Carcinoma: An Observational Study.

Cellular cannibalism (CC) is a prime metabolic event to determine the aggressive potential of oral squamous cell carcinoma (OSCC). However, the etiology and mechanism behind this degradation are still ambiguous. The aim of the study was to explore the etiopathogenetic mechanism behind CC, along with its association with degree of differentiation, angiogenic, phagocytic and antiapoptotic activity in OSCC. Seventy-three tissue sections of various histological grades of OSCC were retrieved from departmental archives and scanned for cannibalistic cells. Immunohistochemical analysis using CD31, CD68, and BCL2 was performed. The data obtained were analyzed using Chi-square, Spearman's correlation test and multiple regression analysis (p < 0.05). CCs were present significantly in various grades of OSCC (p < 0.00). Immunohistochemical analysis revealed a significant difference in CD68, BCL2 (p < 0.05 in both), and CD31 (p < 0.001) expression with CC. The internalized cells showed positivity for CD68 and negativity for BCL2. Regression analysis revealed that tumor grade, CD31 and BCL2 immunoreactivity were significant predictors of frequency of CC. The association of CC with degree of differentiation, CD31, CD68, and BCL2 expression could predict the biological behavior of OSCC and might serve as a promising histopathological parameter in future.

Correlation of Clinical and Pathological TNM Staging With Histopathological Grading in Oral Squamous Cell Carcinoma.

Background Oral squamous cell carcinoma (OSCC) is the most common type of head-neck cancer. The staging and grading of OSCC play an important role in disease management. Accurate staging helps in patient counseling, treatment planning, and prognostication in head-neck SCC. However, discrepancies between pathological and clinical staging have been stated, which affect disease prognosis. Method A retrospective review of 60 surgically treated patients with OSCC was done. Tumor-nodal-metastasis staging, both clinically and pathologically, was equated and tabulated to determine upstaging,downstaging, and cases where no stage change occurred. Additionally, the clinical and pathological TNM (tumor, node, metastasis) staging were correlated with the evaluation of histopathological grading. Results This study comprised 60 surgically operated OSCC patients. The T and N stages showed significant differences when compared clinically and pathologically. There was no significant correlation between histopathological grading and the disparities in TNM staging. Conclusion Some discrepancies exist between TNM staging evaluated clinically and pathologically for OSCC, which may show its effect on treatment planning and the prognosis of affected individuals. The histopathological analysis is the gold standard for the categorization of staging and grading in OSCC for proper treatment planning.

Assessment of immunohistochemical expression of trophoblast cell surface antigen 2 (Trop 2) in oral squamous cell carcinoma and its clinicopathological correlation

Trophoblast cell surface antigen 2 (Trop 2) is a transmembrane glycoprotein that is involved in a variety of oncogenic cell signaling pathways. Its heterogeneous expression in normal and cancerous tissues makes it a diagnostic and prognostic marker for various malignancies, including Oral Squamous Cell Carcinoma (OSCC). To assess the immunohistochemical expression of Trop 2 in OSCC and evaluate its clinicopathological correlation. Histopathologically diagnosed 40 cases of OSCC and their respective clinical records were retrieved and subsequent clinical staging and histopathological grading was done. Formalin fixed parrafin embedded sections were subjected to immunohistochemistry with anti Trop 2 antibody using standard DAB technique. The immunohistochemical expression of Trop 2 was evaluated using immunostaining intensity and mean integrated score followed by its comparison with age, gender, anatomical site, clinical stages and histopathological grades of OSCCTrop 2 expression was positive in 97.5% of cases. There was a statistically significant difference (p value&amp;#60;0.001) between immunostaining intensity and integrated score of Trop 2 among various histopathological grades of OSCC. As grade of OSCC increased, integrated score decreased. Trop 2 is one of the biomarkers that may serve as a useful tool to detect the changes undergoing at the molecular level which have prognostic or therapeutic significance.

Immunohistochemical (IHC) Expression of p16 in Various Grades of Oral Squamous Cell Carcinoma (OSCC) with Snuff Use in Tertiary Care Hospitals of Peshawar

Oral squamous cell carcinoma (OSCC) is a significant health concern, with various factors influencing its development and progression. Understanding these factors, including p16 expression and clinicopathologic features, is crucial for improved diagnosis and treatment. Objective: To compare various grades of OSCC based on immunohistochemical expression of p16 and clinicopathologic parameters. Methods: The comparative cross-sectional study was conducted at the Department of Pathology, Peshawar Medical College (PMC), and Peshawar Dental College (PDC) from August 2020 to August 2021. It included 53 cases of OSCC with documented snuff use history. Tumor sections were stained with Hematoxylin and Eosin and underwent immunohistochemical staining for p16 expression. Sample size calculation utilized G Power software. Statistical analysis was performed using SPSS version 20.0, employing the Chi-Square test to assess categorical variables. Results: Among the 53 OSCC cases, the majority were male (66.0%) with ages ranging from 26 to 85 years, the most common age group being 51-70 years. The tongue was the primary site for OSCC development. Well-differentiated cases were predominant (64.2%), followed by moderate (20.8%) and poor grade cases (15.1%). However, no significant association was found between p16 expression and OSCC grades. Notably, p16 expression tended to be higher in snuff users and well-differentiated OSCC cases, although not statistically significant. Conclusions: Well-differentiated OSCC cases exhibited the highest expression of p16, followed by moderate and poorly differentiated cases. However, no significant correlation was observed between p16 expression and OSCC in snuff users.

A Quantitative Assessment of Apoptotic Index with Varying Histological Degree of Dysplasia and Squamous Cell Carcinoma of Oral Cavity - A Light Microscopic Study.

The aim of the present study was to quantitatively assess the apoptotic index at a light microscopic level in hyperkeratosis with varying histological degrees of dysplasia and different grades of oral squamous cell carcinoma (OSCC) and its correlation with prognostic significance. The study included the histo-pathologically diagnosed cases of hyperkeratosis with varying degrees of dysplasia and different grades of OSCC, which were retrieved from the archives of the department, and clinical details were retrieved from the case records of the patients. The sample size consisted of 80 biopsied cases, divided into seven groups of 10 experimental cases and one group of 10 cases of normal mucosa as a control group. The age range of 50-59 shows maximum patients of hyperkeratosis with varying degrees of dysplasia and OSCC. Maximum apoptotic bodies were seen in cases of severe dysplasia and well-differentiated cases of OSCC. There was an observational difference noticed in apoptotic bodies between the patients who had less than 1-year disease-free survival rate. The apoptotic cells are being seen using routine light microscopy techniques with hematoxylin and eosin staining. In different grades of squamous cell carcinoma, a decrease in the apoptotic count was noticed from well-differentiated squamous cell carcinoma toward poorly differentiated squamous cell carcinoma cases. Increased mean apoptotic bodies were observed in patients showing less than 1 year of survival.

Correlation of prognosis of oral squamous cell carcinoma with CD44 expression: A retrospective immunohistochemical analysis

ABSTRACT Background: Cluster of differentiation (CD44) is a cancer stem cell marker responsible for angiogenesis, invasion, and migration in oral squamous cell carcinoma (OSCC). Objectives: The primary objective of the study was to analyze the difference in expression of CD44 among different grades of OSCC. The secondary objective was to correlate the prognosis with the expression. Materials and Methods: Based on histopathological diagnosis, we retrieved OSCC paraffin-embedded blocks from the archives of the Department of Oral Pathology, Sri Ramaswamy Memorial (SRM) Dental College, Chennai, India, reported from January 2012 to May 2022. We evaluated sections from 15 tissues of normal oral mucosa, 15 paraffin blocks of well-differentiated OSCC, 15 moderately differentiated OSCC, and 15 poorly differentiated OSCC for CD44 immunostaining. Immunohistochemical expression of CD44 was studied in these tissues and graded based on the intensity of staining. The difference in immune expression of CD44 between different grades was quantitatively analyzed using image analysis by Fiji image analysis software. Analysis of variance (ANOVA) was used to compare the mean difference in expression between different groups by using the Statistical Package for Social Sciences (SPSS) software version 16. Statistical significance was set at P &lt; 0.05. Results: We identified that 10% (n = 6) of the normal mucosa showed strong expression of CD44, whereas 13.3% (n = 8) of well-differentiated OSCC, 10% (n = 6) of moderately differentiated OSCC, and 0% (n = 0) of poorly-differentiated OSCC were strongly positive for CD44 expression. The difference in comparison was statistically significant (P, 0.003). By quantitative analysis, the mean expression of CD44-positive cells in well-differentiated OSCC was 38258 ± 4762, moderately-differentiated OSCC was 27353 ± 1357, and poorly-differentiated OSCC was 10265 ± 1685. Upon comparison, the difference in mean was found to be significant (P, 0.002). Conclusion: The expression of CD44 was noted to decrease from well-differentiated to poorly-differentiated OSCC.

Immunohistochemical Evaluation of E- Cadherin in Oral Epithelial Dysplasia and Oral Squamous Cell Carcinoma.

Cancers arising in the oral cavity are more commonly of squamous cell carcinomas. E-cadherin is a calcium-dependant transmembrane glycoprotein of the type-1 cadherin superfamily is an invasion/tumor suppressor gene, which plays a vital role in epithelial cell-cell adhesion. Epithelial E-cadherin expression loss increases tumor invasiveness and metastasis. To determine the expression of E-cadherin in oral epithelial dysplasia (OED) and oral squamous cell carcinoma (OSCC). Analysis of E-cadherin expression in 10 cases of normal mucosa, 15 cases of various grades of OED, 15 cases of OSCC. The data were calculated using Chi-square test and analysis of variance test (ANOVA). An intragroup comparison of staining intensity and staining location for OED showed a highly significant difference between mild and moderate grade (P < 0.001). A significant difference of staining intensity was noted among well and moderately differentiated grades, and well and poorly differentiated grades of OSCC. A comparison of staining location among well and poorly differentiated grades of OSCC was found to be significant. Expression loss is observed as the severity of the lesion progresses in both OSCC and OED. The increased loss of expression in oral squamous cell carcinoma poorer the prognosis.

Expression of IGF-1R in oral submucous fibrosis and oral squamous cell carcinoma – An RT-PCR study

ObjectivesWith the rising number of cases of oral submucous fibrosis (OSMF) undergoing malignant transformation into oral squamous cell carcinoma (OSCC), it is needed to detect the changes at the earliest. Several molecular markers have been used for this purpose, and insulin-like growth factor receptor 1 (IGF-1R) is one of the markers in vogue for several human malignancies. Thus, we compared the expression of IGF-1R mRNA in normal oral mucosa (NOM), OSMF, and OSCC. Materials and methodsThirty biopsy tissue samples for each of the three groups, NOM, OSMF, and OSCC, were taken and analyzed histopathologically and then subjected to RT-PCR for evaluation of the expression of IGF-1R mRNA in each of them. ResultsThere was an upregulation of IGF-1R mRNA in OSMF tissue samples as compared to NOM, but a downregulation as compared to OSCC. There was a 3.24-fold upregulation of IGF-1R mRNA expression in OSMF as compared to NOM and a 1.31-fold downregulation as compared to OSCC. Likewise, there was a 4.2-fold upregulation of IGF-1R mRNA expression in OSCC as compared to NOM and 1.31-fold upregulation as compared to OSMF. Moreover, the expression was more in OSMF patients with interincisal opening (IIO) < 27 mm as compared to those with IIO >27 mm and also was found to increase with the grade of OSCC. ConclusionThe study reflected a potential role of IGF-1R in the pathogenesis of OSMF and OSCC, thus implying its possible exploitation as a diagnostic and prognostic marker and also as a therapeutic target in future.

Histomorphological Evidence of Complex Cannibalism-An Uncharted Territory in Oral Squamous Cell Carcinoma.

Complex cannibalism (CxC) is an emerging new seeming entity in oral squamous cell carcinoma (OSCC) embody as a prime metabolic event in determining the aggressive potential. Owing to paucity in literature regarding it, the aim of the present study to deciphering the occurrence of CxC in OSCC. Further, the expression of cluster of differentiation 68 (CD68) was studied in tumor cells and correlated with CxC to ascertain the biological behaviour of OSCC. 30 Hematoxylin and Eosin stained sections of various grades of OSCC were scanned for CxC and correlated with clinicopathological parameters. Immunohistochemical analysis using CD68 was performed. While observing CxC in OSCC, statistically significant in age (p-0.048) and histological grades (p-0.004). CD68 expression in tumor cells was statistically significant in histopathological grades (p < 0.001) whereas on correlating with CxC (p - 0.171) was non-significant. The analogous rise in CxC and CD68 with increasing histopathological grades could aid in recognising CxC as a precise histopathological parameter to assess the aggressive biological potential in OSCC.

The clinical implication and translational research of OSCC differentiation.

The clinical value and molecular characteristics of tumor differentiation in oral squamous cell carcinoma (OSCC) remain unclear. There is a lack of a related molecular classification prediction system based on pathological images for precision medicine. Integration of epidemiology, genomics, experiments, and deep learning to clarify the clinical value and molecular characteristics, and develop a novel OSCC molecular classification prediction system. Large-scale epidemiology data (n = 118,817) demonstrated OSCC differentiation was a significant prognosis indicator (p < 0.001), and well-differentiated OSCC was more chemo-resistant than poorly differentiated OSCC. These results were confirmed in the TCGA database and in vitro. Furthermore, we found chemo-resistant related pathways and cell cycle-related pathways were up-regulated in well- and poorly differentiated OSCC, respectively. Based on the characteristics of OSCC differentiation, a molecular grade of OSCC was obtained and combined with pathological images to establish a novel prediction system through deep learning, named ShuffleNetV2-based Molecular Grade of OSCC (SMGO). Importantly, our independent multi-center cohort of OSCC (n = 340) confirmed the high accuracy of SMGO. OSCC differentiation was a significant indicator of prognosis and chemotherapy selection. Importantly, SMGO could be an indispensable reference for OSCC differentiation and assist the decision-making of chemotherapy.

Study of expression of GLUT-1 in oral potentially malignant disorders and oral squamous cell carcinoma: An immuno-histochemical analysis.

Glucose is the chief source of energy for cells, and glucose transporter 1 (GLUT-1) is one of the most common glucose transporters in humans. Tumour cells are known to express hypoxia-related protein, and these may allow tumour cells to survive under a sustained hypoxic environment. Surviving cells develop a more aggressive phenotype, which results in poor prognosis. Expression and comparison of GLUT-1 in normal tissues, potentially malignant disorders (PMDs), and oral squamous cell carcinoma (OSCC) and comparison of expression in different grades of OSCC. A total of 57 cases (10 normal, 17 PMD, and 30 cases of OSCC) were stained immuno-histochemically with GLUT-1. The expression was scored as 0, 1, 2, 3, and 4 for negative, mild, moderate, severe, and intense staining, respectively. GLUT-1 expression was detected in all grades of OSCC. A significant correlation was found on comparing normal and OSCC, normal and PMDs, and PMD and OSCC. The expression of GLUT-1 was significant when compared with different histopathological grades of OSCC. Expression of GLUT-1 increased from normal to PMDs to increasing grades of OSCC and hence can be used as a prognostic predictive marker for OSCC.

Evaluation of Collagen Morphology and Eosinophils in Different Grades of Oral Squamous Cell Carcinoma Using Hematoxylin and Eosin and Congo Red Stains – A Pilot Study

Abstract Background: Oral squamous cell carcinoma (OSCC) exhibits dysplastic features in the epithelium and connective tissue which constitute tumor microenvironment. Eosinophils are granulocytic cells which impart inflammatory response and cell death, thereby producing a great role in the regulation and progression of tumors. The morphology of collagen fibers also varies on the progression of oral cancer. Aims: The present study aims at evaluating collagen morphology and amount of eosinophil in different grades of OSCC using hematoxylin and eosin (H and E) and Congo red stains. Settings and Design: The present study includes histopathologically proven cases of 15 OSCC subclassified into five cases per each group. Subjects and Methods: Both H and E and Congo red staining were done for five cases per grades of OSCC and viewed by two observers in zigzag pattern. Statistical Analysis Used: Statistical analysis was done using ANOVA test and interobserver variability by kappa statistics. Results: P &lt; 0.001 was obtained based on the ANOVA test and P value of eosinophils among different grades was statistically significant. Conclusions: The present study concludes that stroma will change from dense to loose and increase in number of eosinophils as carcinoma progresses.

The role of these miRNAs together in clinical application is not always consistent and, its prognostic value in oral squamous cell carcinoma (OSCC) is still elusive. This study was performed to investigate the correlation of these miRNAs expression with histological grades of OSCC according to Bryne's histological grading system, to predict prognosis and to evaluate their relationship with clinico-pathological data. MethodsReal-time quantitative Reverse Transcription Polymerase Chain Reaction (qRT-PCR) was done to evaluate the expression levels of miRNA-196a and miRNA-196b in 75 pairs of OSCC tissue matched with adjacent normal mucosa, used as a control. ResultsmiRNA-196a and 196b expression in OSCC was significantly higher than that in corresponding adjacent normal tissues (p > 0.001). Also, a significant differential correlation was found in between the expression levels of these two miRNAs (Pearson correlation test r = 0.676, p-value<0.0001). The increased expression of these miRNAs was more frequently observed in OSCC tissues with advanced clinical and pathological TNM stages (IVa and IVb, pIVb respectively, p-value<0.0001). Significant correlation was found between miRNA-196a upregulation and moderate prognostic score (p-value<0.0001) in comparison with good and poor prognostic score of histological grades of OSCC. Sensitivity and specificity for miRNA-196a were 95 % and 85 %, respectively (AUC = 1, 95 % CI = 0.617–0.850; p 0.001), while for miRNA-196b were 94 % and 86 %, respectively (AUC = 0.808, 95 % CI = 0.701–0.916; p0.0001). ConclusionsThese findings suggest that the increased expression of miRNA-196a and 196b may play an important role in tumor progression in OSCC. miRNA-196a might be a useful marker for predicting the clinical outcome of OSCC, especially for advanced stages. In conclusion, our data demonstrate for the first time that these miRNAs may serve as a potent prognostic marker for tumor progression. We further highlight miRNA-196a and miRNA-196b as a promising predictor of prognostic assessment in OSCC.

Immunohistochemical Evaluation of Cytokeratin 17 Expression in Oral Squamous Cell Carcinoma

Oral squamous cell carcinoma (OSCC), which makes up 80–90% of all malignant neoplasms of the mouth, is the mostcommon type of oral cancer. Immunohistochemistry plays a crucial role in diagnostic pathology by aiding in identifying andclassifying tumors IHC is utilized to study the distribution and localization of specific proteins. Identifying diagnostic markers forOSCC is essential for early diagnosis and patient-specific treatment. Cytokeratins (CK) are a family of intermediate filamentspredominantly expressed in epithelial cells. The study aimed to evaluate immunohistochemical expression cytokeratin 17 in 50cases of microscopically confirmed OSCC. The present study was observational, with retrospective and prospective sampling.The study was carried out for two years, from 2020 to 2022. The present study was designed to determine CK 17 expressionby IHC and to correlate CK 17 expression with Broders grading of OSCC and other clinical pathologic parameters.Immunostaining for CK 17 was performed. The statistical analysis was done by chi-square test. In this study, there was asignificant correlation between CK 17 expression and Broders grading, and CK 17, when correlated with other clinic pathologicparameters, showed no significant correlation. It was concluded that Cytokeratin17 expression could correlate with theBroders' Grading, thus contributing to early detection and more accurate determination of tumor aggressiveness to determineappropriate therapy.

Protocol for the evaluation of cyclin D1 expression in tumor tissue and surgical margins in oral squamous cell carcinoma – a retrospective study

Oral squamous cell carcinoma (OSCC) is a malignant epithelial tumor with poor prognosis. There is an increase in the proliferation of epithelial cells in OSCC which is control by cell cycle regulators like Cyclin D1. Cyclin D1 is linked to a poor prognosis for patients, including aggressive malignancies, recurrence, protracted remission, etc. To evaluate Cyclin D1 immunoexpression in different grades and surgical margins in OSCC. All tissue sections will be processed for Cyclin D1 IHC. After that light microscopic evaluation of Cyclin D1 expression was classified as weak (scoring 1-4), moderate (score 5-8), or strong based on a final expression score that was obtained by multiplying the labelling index score by the intensity score (score 9–12). The present study will find the immunohistochemical expression of Cyclin D1 in OSCC. There will be variation in the immunoexpression of Cyclin D1 among different grades of OSCC its surgical margin and normal oral mucosa. We hypothesize that: As the disease condition goes towards poor grade, intensity of cyclin D1 expression immunohistochemically may increase. Hence, the intensity can be used as a prognostic marker.
