Rumphellolide J, an Ester of 4β,8β-Epoxycaryophyllan-5-ol and Rumphellaoic acid A, from the Gorgonian Rumphella antipathies

Rumphellolide J (1), a novel sesquiterpenoid derivative, isolated from the gorgonian coral Rumphella antipathies, was determined to be an ester of 4β,8β-epoxycaryophyllan-5-ol and rumphellaoic acid by detailed spectroscopic analysis.

Natural Caryophyllane Sesquiterpenoids from Rumphella antipathies

Two new natural caryophyllane-type sesquiterpenoids, 1 and 2, were isolated from the gorgonian coral Rumphella antipathies. Standard spectroscopic techniques were used to determine the structures of caryophyllanes 1 and 2.

Rumphellaoic acid A, a novel sesquiterpenoid from the formosan gorgonian coral Rumphella antipathies.

A novel sesquiterpenoid, rumphellaoic acid A (1), was isolated from the gorgonian coral Rumphella antipathies, and was found to possess a carbon skeleton that was obtained for the first time from a natural sources. The structure of 1 was elucidated by spectroscopic methods and this compound and was found to exert a moderate inhibitory effect on the release of elastase by human neutrophils.

Rumphellaones B and C, new 4,5-seco-caryophyllane sesquiterpenoids from Rumphellan antipathies.

Two new 4,5-seco-caryophyllane sesquiterpenoids, rumphellaones B (1) and C (2), which were found to possess unprecedented γ-lactone moieties, were obtained from the gorgonian coral Rumphella antipathies. The structures of 1 and 2 were elucidated by spectroscopic methods and compound 2 was found to display modest inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils at a concentration of 10 μg/mL.

Natural Clovanes from the Gorgonian Coral Rumphella Antipathies

Three natural clovane-related sesquiterpenoids, 2beta-acetoxyclovan-9alpha-ol (1), 9alpha-acetoxyclovan-2beta-ol (2) and clovan-2beta,9beta-diol (3), were isolated from the gorgonian coral Rumphella antipathies. The structures of clovanes 1-3 were elucidated by spectroscopic methods and by comparison of the spectral data with those of known clovane analogues. This is the first time that clovanes 1-3 have been isolated from a natural source. Clovanes 1 and 2 displayed inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils.

Rumphellclovanes C–E, new clovane-type sesquiterpenoids from the gorgonian coral Rumphella antipathies

Three new clovane-related sesquiterpenoids, rumphellclovanes C–E (1–3), and a new natural clovane, clovan-2,9-dione (4), were isolated from a gorgonian coral Rumphella antipathies. The structures of clovanes 1–4 were elucidated by spectroscopic methods and by comparison of the spectral data with those of related analogues. Rumphellclovane C (1) was proven to be the first clovane-type sesquiterpenoid containing an ε-lactone moiety. A plausible biosynthetic pathway between clovanes 1 and 2 was proposed. Clovane 4 displayed significant inhibitory effects on the generation of superoxide anion and the release of elastase by human neutrophils.

Synthesis of rumphellaone A via epoxy nitrile cyclization

The first enantioselective total synthesis of rumphellaone A, a cytotoxic 4,5-seco-caryophyllane isolated from the gorgonian coral Rumphella antipathies, has been achieved from a known olefinic alcohol by a 16-step sequence involving Stork's epoxy nitrile cyclization as the key step to concomitantly install a cyclobutane ring and three contiguous stereogenic centers contained in the molecule.

Abstract A novel clovane-type sesquiterpenoid derivative, rumphellclovane B (1), which possesses an unprecedented δ-lactone moiety, and a new natural clovane, 9α-hydroxyclovan-2-one (2), were isolated from the gorgonian coral Rumphella antipathies. The structures of clovanes 1 and 2 were elucidated by interpretations of spectral data. A plausible biosynthetic pathway between these two compounds was proposed. Compound 1 displayed inhibitory effects on superoxide anion generation by human neutrophils.

A new natural caryophyllane-type sesquiterpenoid, rumphellolide H (1), which possesses a tetrahydropyran moiety in structure, was isolated from the gorgonian coral Rumphella antipathies. The structure of 1 was established by spectral data analysis and this metabolite showed moderate inhibitory effects on elastase release by human neutrophils.

Rumphellatins B and C, Two New Caryophyllane-Type Hemiketal Norsesquiterpenoids from the Formosan Gorgonian Coral Rumphella antipathies

Abstract Two new caryophyllane-type hemiketal norsesquiterpenoid alcohols, designated as rumphellatins B (1) and C (2), were obtained from the Formosan gorgonian coral Rumphella antipathies. The structures of natural products 1 and 2 were established by analyzing the spectral data. Rumphellatin B (1) showed antibacterial activity toward the Gram-positive bacterium Staphylococcus aureus.

Rumphellolide G, a New Caryophyllane-type Tetrahydropyran Norsesquiterpenoid from the Gorgonian Coral Rumphella antipathies (Gorgoniidae)

Abstract A new caryophyllane-type tetrahydropyran norsesquiterpenoid, rumphellolide G (1), has been isolated from the gorgonian coral Rumphella antipathies, collected off southern Taiwan coast. The structure of caryophyllane 1 was elucidated by the interpretation of spectral data.

Rumphellatin A, the first chloride-containing caryophyllane-type norsesquiterpenoid from Rumphella antipathies

A novel chloride-containing 14C norsesquiterpenoid, rumphellatin A ( 1), was isolated from the Formosan gorgonian coral Rumphella antipathies. The structure of metabolite 1 was determined on the basis of spectral data analysis and the configuration of 1 was further supported by molecular mechanics calculations. Rumphellatin A ( 1) showed activity against Gram-negative bacteria.

Rumphellolides A-F, Six New Caryophyllane-Related Derivatives from the Formosan Gorgonian Coral Rumphella antipathies

Six new caryophyllane-related natural products, including two carboxylated sesquiterpenoids, rumphellolides A (1) and B (2), and four norsesquiterpene alcohols, rumphellolides C-F (3-6), were isolated from the Formosan gorgonian coral Rumphella antipathies. The structures of the above new natural products were established on the basis of extensive spectral data analysis. Rumphellolides A (1), D (4), E (5), and F (6) showed weak antibacterial activity.
