Transpiration Responses of Herbicide-Resistant and -Susceptible Palmer Amaranth (Amaranthus palmeri (S.) Wats.) to Progressively Drying Soil

Drought events are predicted to increase in the future. Evaluating the response of herbicide-resistant and -susceptible weed ecotypes to progressive drought can provide insights into whether resistance traits affect the fitness of resistant weed populations. Two experiments were conducted in the greenhouse between January and May 2021 to evaluate drought tolerance differences between Palmer amaranth accessions resistant to S-metolachlor or glyphosate and their susceptible counterparts. The accessions used were S-metolachlor-resistant (17TUN-A), a susceptible standard (09CRW-A), and glyphosate-resistant (22–165 EPSPS copies) and glyphosate-susceptible (3–10 EPSPS copies) plants from accession 16CRW-D. Daily transpiration of each plant was measured. The daily transpiration rate was converted to normalized transpiration ratio (NTR) using a double-normalization procedure. The daily soil water content was expressed as a fraction of transpirable soil water (FTSW). The threshold FTSW (FTSWcr), after which NTR decreases linearly, was estimated using a two-segment linear regression analysis. The data showed differences between S-metolachlor-resistant and -susceptible accessions (p ≤ 0.05). The FTSW remaining in the soil at the breakpoint for the S-metolachlor-susceptible accession (09CRW-A) was 0.17 ± 0.007. The FTSW remaining in the soil at the breakpoint for the S-metolachlor-resistant accession (17TUN-A) was 0.23 ± 0.004. The FTSW remaining in the soil at the breakpoint for the glyphosate-resistant and glyphosate-susceptible plants (16CRW-D) was 0.25 ± 0.007 and 0.25 ± 0.008, respectively. Although the mechanism endowing resistance to S-metolachlor might have contributed to increased drought tolerance, follow-up experiments are needed in order to verify this finding. Increased EPSPS copy numbers did not improve the drought tolerance of Palmer amaranth. As droughts are predicted to increase in frequency and severity, these results suggest that S-metolachlor-resistant and glyphosate-resistant Palmer amaranth populations will not be at a competitive disadvantage compared to susceptible genotypes. Alternative and diverse management strategies will be required for effective Palmer amaranth control, regardless of herbicide resistance status.

Enhancement of glyphosate efficacy on Amaranthus palmeri by exogenous quinate application

• Quinate is an alternative source of carbon for the shikimate pathway and is converted into tyrosine and phenylalanine. • In glyphosate-susceptible plants, the application of one-quarter of the recommended dose was as lethal as the recommended dose if quinate was sprayed one day after. • In glyphosate-resistant plants, quinate application after glyphosate did not change the lethality provoked by the herbicide but induced metabolic changes in the shikimate pathway. • The results raise the possibility of the application of quinate after glyphosate to improve the efficacy of the herbicide in the control of A. palmeri .

Interaction of Chemical Pesticides and Their Formulation Ingredients with Microbes Associated with Plants and Plant Pests.

Chemical pesticides and their formulation ingredients can have unintended effects on microbes associated with plants and plant pests. These effects can be due to direct effects on the microbes or to effects on crops or weeds that subsequently affect the microbes. In addition to fungicides, some insecticides, herbicides, and formulation compounds are toxic to plant pathogenic microbes, as well as to potentially beneficial microbes, such as those that infect insect pests. These chemicals, especially herbicides, can also indirectly affect microbes through their effects on crops and weeds. For example, glyphosate strongly impairs shikimic acid pathway-based plant defenses to microbial diseases in glyphosate-susceptible plants, significantly increasing its efficacy as an herbicide. Some herbicides induce plant defenses against plant pathogens. For a complete understanding of integrated pest management and overall cost/benefit of pesticide use, much more information is needed on microbial/pesticide interactions.

Temperature Effects on Glyphosate Resistance in California Populations of Junglerice

Temperature effects glyphosate resistance. Glyphosate is more effective when applied at cooler temperatures. Air temperature is important in glyphosate application. This study determined the effect of glyphosate on a glyphosate‐resistant (GR) and glyphosate‐susceptible (GS) biotype of junglerice (Echinochloa colona L.). Potted plants at the 4‐ to 5‐leaf stage were acclimatized for 72 h in growth chambers programmed for day/night temperatures of 15/10, 25/20, and 35/30°C. On the fourth day, three plants of each biotype were treated with glyphosate at 840 g ae ha−1 and immediately placed back in the respective growth chambers and grown for seven additional days. Three non‐treated plants of each biotype were used as controls for each temperature regime. On the eighth day after glyphosate treatment, the plants were moved to a greenhouse set at 22°C, grown for 21 d, and aboveground dry weights recorded after evaluating phytotoxicity. None of the treated GS plants survived at any of the temperatures while all the non‐treated plants grew well at all temperatures. All treated and non‐treated GR plants showed no phytotoxicity in the 25/20 and 35/30°C temperature regimes. However, all the treated GR plants subjected to 15/10°C died. The aboveground biomass of the non‐treated GR plants was reduced under cooler temperatures but was unaffected in the GS plants. Therefore, this study showed that GR junglerice was susceptible to glyphosate when grown under 15/10°C suggesting that the prevalence of GR plants in the fields could be reduced if glyphosate applications were made in spring when temperatures were at or below 15°C when the plants were just emerging or had developed less than six leaves.

Differential Tolerance of Glyphosate-Susceptible and Glyphosate-Resistant Biotypes of Junglerice (Echinochloa colona) to Environments during Germination, Growth, and Intraspecific Competition

AbstractMuch of the agricultural area in California’s southwestern San Joaquin Valley (SJV) is prone to moisture stress and high soil-salinity conditions. Increased prevalence of glyphosate-resistant (GR) biotypes of junglerice [Echinochloa colona (L.) Link] in these environments and their ecological implications need to be further explored. Studies were conducted on GR and glyphosate-susceptible (GS) biotypes of E. colona to compare the effects of moisture and salinity stress on seed germination and salinity stress alone on growth and seed production. Intraspecific competition between the GR and GS plants was also assessed in a replacement series design experiment. With respect to germination, both biotypes were tolerant to moisture and salinity stress at germination; however, the GR biotype was more tolerant than the GS biotype. Water potential and electrical conductivity (EC) levels that reduced germination by 50% were estimated as −1.5 and −2.3 MPa and 8.5 and 12 dS m−1 for the GS and GR biotypes, respectively. The EC levels that reduced aboveground biomass by 50% were estimated as 9 and 11.5 dS m−1 for the GS and GR biotypes, respectively. Seed production was generally greater in the GR than the GS plants below 10 dS m−1. All plants produced up to 140 seeds plant−1, even at 20 dS m−1. The GR plants were more competitive and produced more aboveground dry biomass and seeds than the GS plants when grown together or alone. In conclusion, differences between these particular GR and GS biotypes to environmental stresses and intraspecific competition were noted that could have ecological implications for their prevalence in the southwestern SJV. The results also suggested that there could be high genetic variability and phenotypic plasticity in E. colona populations in the SJV and further population shifts could occur due to selection pressure from glyphosate.

Hyperspectral Imaging for Differentiating Glyphosate-Resistant and Glyphosate-Susceptible Italian Ryegrass

Glyphosate is widely used in row crop weed control programs of glyphosate-resistant (GR) crops. With the accumulation of glyphosate use, several weeds have evolved resistance to glyphosate. In order to control GR weeds for profitable crop production, it is critical to first identify them in crop fields. Conventional method for identifying GR weeds is destructive, tedious and labor-intensive. This study developed hyperspectral imaging for rapid sensing of Italian ryegrass (Lolium perenne ssp. multiflorum) plants to determine if each plant is GR or glyphosate-susceptible (GS). In image analysis, a set of sensitive spectral bands was determined using a forward selection algorithm by optimizing the area under the receiver operating characteristic between GR and GS plants. Then, the dimensionality of selected bands was reduced using linear discriminant analysis. At the end the maximum likelihood classification was conducted for plant sample differentiation of GR Italian ryegrass from GS ones. The results indicated that the overall classification accuracy is between 75% and 80%. Although the accuracy is lower than the classification of Palmer amaranth (Amaranthus palmeri S. Wats.) in our previous study, this study provides a rapid, non-destructive approach to differentiate between GR and GS Italian ryegrass for improved site-specific weed management.

Distribution of Herbicide-Resistant Giant Ragweed (Ambrosia trifida) in Indiana and Characterization of Distinct Glyphosate-Resistant Biotypes

Giant ragweed is a highly competitive weed that continually threatens crop production systems due to evolved resistance to acetolactate synthase–inhibiting herbicides (ALS-R) and glyphosate (GR). Two biotypes of GR giant ragweed exist and are differentiated by their response to glyphosate, termed here as rapid response (RR) and non–rapid response (NRR). A comparison of data from surveys of Indiana crop fields done in 2006 and 2014 showed that GR giant ragweed has spread from 15% to 39% of Indiana counties and the NRR biotype is the most prevalent. A TaqMan®single-nucleotide polymorphism genotyping assay was developed to identify ALS-R populations and revealed 47% of GR populations to be ALS-R as well. The magnitude of glyphosate resistance for NRR populations was 4.6 and 5.9 based on GR50and LD50estimates, respectively. For RR populations, these values were 7.8 to 9.2 for GR50estimates and 19.3 to 22.3 for LD50estimates. A novel use of the Imaging-PAM fluorometer was developed to discriminate RR plants by assessing photosystem II quantum yield across the entire leaf surface. H2O2generation in leaves of glyphosate-treated plants was also measured by 3,3′-diaminobenzidine staining and quantified using imagery analysis software. Results show photo-oxidative stress of mature leaves is far greater and occurs more rapidly following glyphosate treatment in RR plants compared with NRR and glyphosate-susceptible plants and is positively associated with glyphosate dose. These results suggest that under continued glyphosate selection pressure, the RR biotype may surpass the NRR biotype as the predominant form of GR giant ragweed in Indiana due to a higher level of glyphosate resistance. Moreover, the differential photo-oxidative stress patterns in response to glyphosate provide evidence of different mechanisms of resistance present in RR and NRR biotypes.

Biology and Management of Glyphosate-Resistant and Glyphosate-Susceptible Palmer Amaranth (Amaranthus palmeri) Phenotypes from a Segregating Population

Palmer amaranth is the most economically damaging glyphosate-resistant (GR) weed in the southern United States. An understanding of the basic biology, including relative growth and competitiveness of GR and glyphosate-susceptible (GS) Palmer amaranth phenotypes from a segregating population collected from the same geographical location, may yield information helpful in the management of resistant populations. A segregating population of Palmer amaranth collected in North Carolina during 2010 was used as a plant source for both GR and GS traits. Research was conducted in the greenhouse to compare the following: level of resistance and shikimate accumulation in GR and GS phenotypes following glyphosate application; interference from GR and GS phenotypes on early-season vegetative growth of corn, cotton, and peanut; effect of various durations of imposed drought stress on GR and GS phenotypes; and response of GR and GS phenotypes to POST-applied herbicides. The GR50(glyphosate rate providing 50% reduction in shoot dry biomass) was 17 times greater with the GR phenotype compared with the GS phenotype. Shikimate accumulated in both GR and GS phenotypes following glyphosate application, but greater concentrations were found in GS plants. The GR and GS phenotypes responded similarly when subjected to drought stress; grown with corn, cotton, and peanut; or treated with 2,4-D, atrazine, dicamba, fomesafen, glufosinate, paraquat, tembotrione, and thifensulfuron. These results indicate that in the absence of glyphosate selection pressure, resistance to glyphosate does not influence the growth and competitiveness of GR and GS Palmer amaranth phenotypes collected from the same geographical location.

The history and current status of glyphosate.

Glyphosate is the only herbicide to target the enzyme 5-enolpyruvyl-3-shikimate phosphate synthase (EPSPS). It is a high use rate, non-selective herbicide that translocates primarily to metabolic sinks, killing meristematic tissues away from the application site. Its phloem-mobile properties and slow action in killing weeds allow the herbicide to move throughout the plant to kill all meristems, making it effective for perennial weed control. Since commercialization in 1974, its use has grown to dominate the herbicide market. Much of its use is on transgenic, glyphosate-resistant crops (GRCs), which have been the dominant transgenic crops worldwide. GRCs with glyphosate provided the most effective and inexpensive weed management technology in history for a decade or more. However, as a consequence of the rapid increase in glyphosate-resistant (GR) weeds, the effectiveness of glyphosate use in GRCs is declining. Critics have claimed that glyphosate-treated GRCs have altered mineral nutrition and increased susceptibility to plant pathogens because of glyphosate's ability to chelate divalent metal cations, but the complete resistance of GRCs to glyphosate indicates that chelating metal cations do not contribute to the herbicidal activity or significantly affect mineral nutrition. The rates of increases in yields of maize, soybean, and cotton in the USA have been unchanged after high adoption rates of GRCs. Glyphosate is toxic to some plant pathogens, and thereby can act as a fungicide in GRCs. Ultra-low doses of glyphosate stimulate plant growth in glyphosate-susceptible plants by unknown mechanisms. Despite rapid and widespread increases in GR weeds, glyphosate use has not decreased. However, as GR weeds increase, adoption of alternative technologies will eventually lead to decreased use. Confirmation and Control of Annual Bluegrass (Poa annua) with Resistance to Prodiamine and Glyphosate

Poor annual bluegrass control was reported in golf course roughs following treatment with prodiamine (1120 gaiha−1) and glyphosate (840 gaeha−1) during hybrid bermudagrass dormancy. Research was conducted to determine if this annual bluegrass phenotype was resistant to both prodiamine and glyphosate and to determine the efficacy of herbicide mixtures for controlling this phenotype in the field. In PRE or POST dose-response experiments, 9 to 31 times more prodiamine or glyphosate was needed to control (or reduce dry biomass of) this resistant phenotype by 50% compared to an herbicide-susceptible phenotype. Moreover, glyphosate-susceptible plants accumulated 50% more shikimic acid (898 mgkg−1) 6 d after treatment than those resistant to glyphosate (394 mgkg−1). October (fall) applications of herbicide mixtures containing trifloxysulfuron, simazine,S-metolachlor, or mesotrione controlled this resistant annual bluegrass phenotype 84 to 98% in April (spring), with no differences detected among treatments. Our findings document the second instance of annual bluegrass evolving multiple resistance in a managed turfgrass system. However, several herbicide mixtures can be used to effectively manage this resistant phenotype.

Environmental Conditions Influence Induction of Key ABC-Transporter Genes Affecting Glyphosate Resistance Mechanism in Conyza canadensis.

Conyza canadensis has been reported to be the most frequent weed species that evolved resistance to glyphosate in various parts of the world. The objective of the present study was to investigate the effect of environmental conditions (temperature and light) on the expression levels of the EPSPS gene and two major ABC-transporter genes (M10 and M11) on glyphosate susceptible (GS) and glyphosate resistant (GR) horseweed populations, collected from several regions across Greece. Real-time PCR was conducted to determine the expression level of the aforementioned genes when glyphosate was applied at normal (1×; 533 g·a.e.·ha−1) and high rates (4×, 8×), measured at an early one day after treatment (DAT) and a later stage (four DAT) of expression. Plants were exposed to light or dark conditions, at three temperature regimes (8, 25, 35 °C). GR plants were made sensitive when exposed to 8 °C with light; those sensitized plants behaved biochemically (shikimate accumulation) and molecularly (expression of EPSPS and ABC-genes) like the GS plants. Results from the current study show the direct link between the environmental conditions and the induction level of the above key genes that likely affect the efficiency of the proposed mechanism of glyphosate resistance.

Glyphosate Resistance in Perennial Ryegrass (Lolium perenneL.) is Associated with a Fitness Penalty

The net selection effect of herbicides on herbicide-resistance traits in weeds is conditioned by the fitness benefits and costs associated with resistance alleles. Fitness costs play an important evolutionary role preventing the fixation of adaptive alleles and contributing to the maintenance of genetic polymorphisms within populations. Glyphosate is widely used in world agriculture, which has led to the evolution of widespread glyphosate resistance in many weed species. The fitness of glyphosate-resistant and -susceptible perennial ryegrass plants selected from within a single population were studied in two field experiments conducted during 2011 and 2012 under different soil water availability. Glyphosate-resistant plants showed a reduction in height of 12 and 16%, leaf blade area of 16 and 33%, shoot biomass of 45 and 55%, seed number of 33 and 53%, and total seed mass of 16 and 5% compared to glyphosate-susceptible plants in 2011 and 2012, respectively. The reduction in seed number per plant resulted in a 40% fitness cost associated with the glyphosate-resistance trait in perennial ryegrass. Fitness costs of glyphosate-resistant plants were expressed under both conditions of water availability. These results could be useful for designing management strategies and exploiting the reduced glyphosate-resistant perennial ryegrass fitness in the absence of glyphosate selection.

Growth stage, temperature, and time of year affects the control of glyphosate-resistant and glyphosate-paraquat resistant Conyza bonariensis with saflufenacil

Conyza bonariensis (L.) Cronq. is a problematic weed in crop and non-crop areas of California. This problem has been further aggravated by the discovery of herbicide-resistant biotypes. Experiments were conducted in 2012 and 2013 in fall and spring, respectively, to determine the efficacy of glyphosate, saflufenacil, and a tank-mixture of saflufenacil + glyphosate on glyphosate-susceptible (GS), glyphosate-resistant (GR), and glyphosate-paraquat-resistant (GPR) biotypes of C. bonariensis. Efficacy of the herbicides were evaluated at three growth stages (5- to 8-leaf seedling, rosette, and bolting); and three day/night temperature regimes (15/10 °C, 25/20 °C, 35/30 °C). Results differed between experiments conducted in the fall and spring. Saflufenacil-alone was more effective in the fall than in spring. All the GS, GR and GPR plants were controlled by saflufenacil-alone at the 5-to 8-leaf stage and rosette stage, but level of control declined at the bolting stage. Better control with saflufenacil-alone and glyphosate-alone was obtained at the 15/10 °C and 25/20 °C than at the 35/30 °C temperature regime. However, a tank-mixture of saflufenacil + glyphosate provided good control of the plants at 35/30 °C. Efficacy of saflufenacil-alone was inconsistent in spring and varied between the biotypes, but the control with saflufenacil + glyphosate was excellent and consistent between seasons. Glyphosate-alone provided good control of all three biotypes at the 5- to 8-leaf stage in the fall, but the control was poor in spring. Therefore, saflufenacil-alone can provide excellent control of C. bonariensis plants prior to the bolting stage in the fall; but in spring, it will be more effective when applied with glyphosate.

Glyphosate-resistant population of Lolium perenne loses resistance at winter temperatures

In New Zealand vineyards, evolution of glyphosate resistance in perennial ryegrass (Lolium perenne) has been reported recently, and restricted translocation of glyphosate has been found in these resistant plants. Past research with other plant species has found that restricted glyphosate translocation is temperature dependent. Glasshouse dose-response experiments were conducted with resistant perennial ryegrass at different temperatures to investigate if this population becomes susceptible to glyphosate under cooler conditions. Glyphosate-susceptible plants (population SP) grown in either warm (average of 20 °C) or cool (average of 9 °C) temperatures following treatment with several rates of glyphosate responded similarly to the herbicide regardless of temperature. In contrast, plants of population J (glyphosate resistant) were significantly more resistant than population SP to glyphosate when treated under warm conditions, but at the cooler temperature, population J was as susceptible as population SP. Thus the mechanism of resistance in population J appeared to be suppressed under cool conditions, suggesting that application of glyphosate during winter might improve control of glyphosate-resistant perennial ryegrass infestations.

Distribution of glyphosate-resistant and glyphosate-susceptible hairy fleabane (Conyza bonariensis) in central California and their phenological development

Glyphosate-resistant (GR) hairy fleabane was first documented in California in 2007. A preliminary study comparing plants from this population to a glyphosate-susceptible (GS) population determined that they developed more rapidly than the GS plants. However, it was not known if this was a general characteristic of all GR populations. Therefore, seeds of hairy fleabane were collected from 122 locations in central California. Plants from each location were grown and tested for resistance using a shikimate enzyme assay. Seeds from 90 locations germinated of which 27 % were determined as GR, 21 % as GS, and 52 % as intermediate/mixed. Plants from five GR and five GS populations, as determined by the assay, were randomly selected for comparisons and were grown in 2010/2011. Time taken by each plant to reach key phenological stages was recorded and converted into growing degree day (GDD) using a base temperature of 4.2 °C. The biomass of each plant was recorded at seed set. The five GR and GS plants had similar phenological development rates and biomass. Therefore, the differences previously identified between the two populations were not present in the additionally tested populations. The phenological development was hence described by a common GDD model with the plants requiring 374, 711, 970, 1289, and 1551 GDD, to reach the rosette, bolting, first bud, first flower, and first seed stages, respectively. This study shows widespread distribution of GR hairy fleabane in central California, provides useful information on biology of this weed for management, and suggests possible label changes to herbicide manufacturers.

Physiology of Glyphosate-Resistant and Glyphosate-Susceptible Palmer Amaranth (Amaranthus palmeri) Biotypes Collected from North Carolina

Glyphosate-resistant (GR) biotypes of Palmer amaranth are now commonly found across the southern United States. Experiments were conducted to characterize physiological differences between a GR biotype and a glyphosate-susceptible (GS) biotype from North Carolina. The GR biotype had an 18-fold level of resistance based upon rates necessary to reduce shoot fresh weight 50%. Shikimate accumulated in both biotypes following glyphosate application, but greater concentrations were found in GS plants. Absorption and translocation of14C-glyphosate were studied in both biotypes with and without an overspray with commercial glyphosate potassium salt (840 g ae ha−1) immediately prior to14C-glyphosate application. Greater absorption was noted 6 h after treatment (HAT) in GS compared with GR plants, but no differences were observed at 12 to 72 HAT. Oversprayed plants absorbed 33 and 61% more14C by 48 and 72 HAT, respectively, than plants not oversprayed.14C distribution (above treated leaf, below treated leaf, roots) was similar in both biotypes. Together, these results suggest that resistance in this biotype is not due to an altered target enzyme or translocation but may be in part due to the rate of glyphosate absorption. These results also are consistent with resistance being due to increased gene copy number for the target enzyme.

Bases for Interactions between Saflufenacil and Glyphosate in Plants

Buckwheat (Fagropyrum esculentum Moench.), cabbage (Brassica oleracea L), and conventional and glyphosate-resistant varieties of canola (Brassica napus L.) were used to study the bases of saflufenacil and glyphosate interactions. Compared to the addition of Merge (surfactant), the addition of both Transorb (i.e., commercial product, Transorb formulation with glyphosate) and Merge increased the cuticular absorption of [(14)C] saflufenacil in cabbage plants with thick epicuticular wax layers. However, in all cases, the addition of glyphosate reduced the translocation of [(14)C]saflufenacil in glyphosate-susceptible plants, while translocation was not affected in glyphosate-resistant canola. Moreover, the phytotoxicity of saflufenacil reduced the activity of glyphosate, possibly by reducing its translocation in all plant species studied. Increased absorption of saflufenacil by the addition of Transorb (i.e., Transorb formulation with glyphosate) plus Merge appears to increase its contact activity, thus the interaction of saflufenacil and glyphosate involves two separate processes, absorption and translocation.

A simple chromatographic assay to discriminate between glyphosate-resistant and susceptible soybean ( Glycine max) cultivars

In glyphosate-susceptible soybean ( Glycine max L. Merrill), the herbicide glyphosate [( N-phosphonomethyl)glycine] inhibits the enzyme 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase, causing a massive accumulation of the metabolite shikimate. This phenomenon does not occur in glyphosate-resistant soybean due the presence of a gene encoding glyphosate-insensitive EPSP synthase. This study proposes a simple and reliable assay as an alternative tool for differentiating glyphosate-resistant from susceptible soybean cultivars. The assay is based on a single extraction of leaf or root tissue. Shikimate is quantified by reversed-phase high–performance liquid chromatography at 220 nm and isocratic elution with phosphoric acid. After glyphosate treatment, tissues of glyphosate-susceptible plants were found to massively accumulate shikimate, whereas the tissues of glyphosate-resistant plants did not accumulate the metabolite.

Manganese uptake and redistribution in soybean as affected by glyphosate

Detrimental effects of glyphosate on plant mineral nutrition have been reported in the literature, particularly on Mn uptake and redistribution. However, in most of the experiments conducted so far glyphosate-susceptible plants were used. Effects of glyphosate on Mn absorption kinetics, accumulation, and distribution within the plant, as well as soybean response to Mn as affected by glyphosate were studied in three experiments. In the first experiment, in nutrient solution, the effect of glyphosate on soybean Mn uptake kinetic parameters (Imax, Km and Cmin) was determined. In a second experiment, also in nutrient solution, differential Mn accumulation and distribution were studied for a conventional soybean cultivar and its near-isogenic glyphosate-resistant counterpart as affected by glyphosate. In a third experiment, response of glyphosate-resistant soybean cultivars to Mn application was studied in the presence of glyphosate, in pots with Mn-deficient soil. Maximum Mn influx (Imax) was higher in the herbicide-resistant (GR) cultivar than in its conventional counterpart. Glyphosate applied to nutrient solution at low rates decreased Km and Cmin. A few days after herbicide treatment, RR soybean plants developed yellowish leaves, a symptom which, in the field, could be misinterpreted as Mn deficiency, but herbicide application had no effect on Mn uptake or distribution within the plant. In the soil experiment, soybean Mn uptake was increased by Mn application, with no effect of glyphosate. Under greenhouse conditions, there was no evidence of deleterious effects of glyphosate on Mn absorption, accumulation and distribution in the plant and on soybean cultivars response to Mn application.

Transformation and segregation of GFP fluorescence and glyphosate resistance in horseweed (Conyza canadensis) hybrids

The goal of this research was to generate a breeding population of horseweed segregating for glyphosate resistance. In order to generate a marker to select between hybrids of glyphosate resistant (GR) and glyphosate susceptible (GS) horseweed, a GR horseweed accession from western Tennessee was transformed with a green fluorescent protein (GFP) transgene. The GFP marker allowed for the simple and accurate determination of GR hybrid plants by visual observation. GR plants were shown to be transgenic via the green fluorescence under UV light, and resistant to glyphosate when sprayed with the field-use-rate 0.84 kg acid equivalent ha(-1) of glyphosate (i.e. Roundup) herbicide. An in vitro screen for glyphosate resistance in seedlings was developed, and a 5 microM glyphosate concentration was found to reduce dry weight in GS seedlings but not in GR seedlings. The GR plants containing GFP were then hand-crossed with GS plants from eastern Tennessee under greenhouse conditions, with GS plants acting as the pollen acceptor. Resulting seed was collected and germinated for GFP fluorescence screening. Seedlings that exhibited the transgenic GFP phenotype were selected as F(1) hybrids between GR and GS horseweed. Thirty GSxGR hybrids were produced on the basis of a green-fluorescent GFP phenotype of GR plants. GSxGFP/GR F(1) hybrids produced F(2) seeds, and F(2) plants were shown to segregate for GFP fluorescence and glyphosate resistance independently. Both traits segregated at a Mendelian 3:1 ratio, indicating a single gene is responsible for each phenotype.
