Guiding seed movement: environmental heterogeneity drives genetic differentiation in Plathymenia reticulata, providing insights for restoration.

Forest and landscape restoration is one of the main strategies for overcoming the environmental crisis. This activity is particularly relevant for biodiversity-rich areas threatened by deforestation, such as tropical forests. Efficient long-term restoration requires understanding the composition and genetic structure of native populations, as well as the factors that influence these genetic components. This is because these populations serve as the seed sources and, therefore, the gene reservoirs for areas under restoration. In the present study, we investigated the influence of environmental, climatic and spatial distance factors on the genetic patterns of Plathymenia reticulata, aiming to support seed translocation strategies for restoration areas. We collected plant samples from nine populations of P. reticulata in the state of Bahia, Brazil, located in areas of Atlantic Forest and Savanna, across four climatic types, and genotyped them using nine nuclear and three chloroplast microsatellite markers. The populations of P. reticulata evaluated generally showed low to moderate genotypic variability and low haplotypic diversity. The populations within the Savanna phytophysiognomy showed values above average for six of the eight evaluated genetic diversity parameters. Using this classification based on phytophysiognomy demonstrated a high predictive power for genetic differentiation in P. reticulata. Furthermore, the interplay of climate, soil and geographic distance influenced the spread of alleles across the landscape. Based on our findings, we propose seed translocation, taking into account the biome, with restricted use of seed sources acquired or collected from the same environment as the areas to be restored (Savanna or Atlantic Forest).

Diversity and genetic structure of natural populations of the palm tree Euterpe precatoria (Arecaceae)

Introduction: The natural ecosystems of northern Mato Grosso, Brazil, are in process of fragmentation, mainly due to population growth and the expansion of agriculture. This endangers the palm Euterpe precatoria (locally known as açaí), used for construction, palm hearts, juices and ice cream. Objective: To evaluate the local diversity and genetic structure in native populations of E. precatoria. Methods: We collected leaves from 106 fruiting palms from five populations in Mato Grosso State, for analysis of microsatellite markers with Polymerase Chain Reaction. Results: The five SSR loci revealed a total of 30 alleles, ranging from 5 (EE23 and EE43) to 7 (EE2 and EE15), with an average of 6 alleles per locus. The mean PIC was 0.74 and confirmed low heterozygosity and inbreeding. The UPGMA dendrogram produced two groups and molecular variance revealed greater genetic differentiation within populations. The high levels of homozygous microsatellite loci indicate low genetic diversity. Conclusions: These populations have low gene diversity, high average number of alleles per locus, and rare and exclusive alleles. We recommend the establishment of permanent conservation units with corridors among them.

Honey Bee Diversity Is Swayed by Migratory Beekeeping and Trade Despite Conservation Practices: Genetic Evidence for the Impact of Anthropogenic Factors on Population Structure

The intense admixture of honey bee (Apis melliferaL.) populations at a global scale is mostly attributed to the widespread migratory beekeeping practices and replacement of queens and colonies with non-native races or hybrids of different subspecies. These practices are also common in Anatolia and Thrace, but their influence on the genetic make-up of the five native subspecies of honey bees has not been explored. Here, we present an analysis of 30 microsatellite markers from honey bees from six different regions in Anatolia and Thrace (N= 250 samples), with the aim of comparing the impact of: (1) migratory beekeeping, (2) queen and colony trade, and (3) conservation efforts on the genetic structure of native populations. Populations exposed to migratory beekeeping showed less allegiance than stationary ones. We found genetic evidence for migratory colonies, acting as a hybrid zone mobile in space and time, becoming vectors of otherwise local gene combinations. The effect of honey bee trade leaves very high introgression levels in native honey bees. Despite their narrow geographic range, introgression occurs mainly with the highly commercial Caucasian bees. We also measured the direction and magnitude of gene flow associated with bee trade. A comparison between regions that are open and those closed to migratory beekeeping allowed the evaluation of conservation sites as centers with limited gene flow and demonstrated the importance of establishing such isolated regions. Despite evidence of gene flow, our findings confirm high levels of geographically structured genetic diversity in four subspecies of honey bees in Turkey and emphasize the need to develop policies to maintain this diversity. Our overall results are of interest to the wider scientific community studying anthropogenic effects on the population diversity of honey bees and other insects. Our findings on the effects of migratory beekeeping, replacement of queens and colonies have implications for the conservation of honey bees, other pollinators, and invertebrates, in general, and are informative for policy-makers and other stakeholders in Europe and beyond.

Spatial patterns of genetic diversity, community composition and occurrence of native and non-native amphipods in naturally replicated tributary streams.

BackgroundWorldwide, natural communities are invaded by non-native species, with potentially devastating effects on the native communities. A large part of past research aimed at finding traits and characteristics of the invading species or the invaded community explaining observed invasions. Only recently, the focus shifted on the spatial patterns during invasions per se. Empirical data, however, are limited, as invasions are often unique incidences of a complex spatio-temporal process. In order to identify generalities of invasion patterns, we studied 13 naturally replicated tributary streams draining into Lake Constance, and studied the occurrence of native and non-native amphipods along linear transects from the stream outlets to the upstream headwater reaches.ResultsWe found repeated spatial patterns of community composition and the occurrence of native and non-native amphipod species across two different years. Specifically, occurrence as well as abundance of two non-native amphipod species decreased from the stream outlets at the lake site towards upstream headwater reaches. Populations of the most common native amphipod species were largest at the uppermost headwater reaches. All populations of this native species, however, showed significant signals of recent genetic bottlenecks, irrespective of the stream position and occurrence of non-native species. Contrary to our expectations, this native species also showed no longitudinal genetic differentiation within individual tributaries as postulated for headwater versus outlet populations.ConclusionsOur results indicate that invasions of river-systems may overall follow predictable patterns on the level of spatial distributions and community composition. Higher genetic diversity in introduced than in native populations of the mussel Mytella charruana: evidence of population admixture at introduction sites

AbstractAim Levels of genetic diversity can be used to determine haplotype frequency, population size and patterns of invasive species distribution. In this study, we sought to investigate the genetic structure of the invasive marine mussel Mytella charruana and compare variation from invasive populations with variation found within three native populations.Location Invaded areas in the USA (Florida, Georgia); native areas in Ecuador, Colombia and Brazil.Methods We sequenced 722 bp of the mitochondrial COI gene from 83 M. charruana samples from four invasive populations (USA) and 71 samples from two natural populations (Ecuador, Columbia). In addition, we sequenced 31 individuals of a congeneric species, Mytella guyanensis, from Salvador, Brazil. We constructed the phylogenetic relationship among all haplotypes and compared diversity measures among all populations.Results We found significantly higher levels of nucleotide diversity in invasive populations than in native populations, although the number of haplotypes was greater in the native populations. Moreover, mismatch distribution analyses resulted in a pattern indicative of population admixture for the invasive populations. Conversely, mismatch distributions of native populations resulted in a pattern indicative of populations in static equilibrium.Main conclusion Our data present compelling evidence that the M. charruana invasion resulted from admixture of at least two populations, which combined to form higher levels of genetic diversity in invasive populations. Moreover, our data suggest that one of these populations originated from the Caribbean coast of South America. Overall, this study provides an analysis of genetic diversity within invasive populations and explores how that diversity may be influenced by the genetic structure of native populations and how mass dispersal may lead to invasion success.

Microsatellite markers for the Amazon peacock bass (Cichla piquiti)

A set of primers to amplify 10 microsatellite DNA loci was developed for the Neotropical fish Cichla piquiti, one of the largest sized cichlids in the Amazon Basin. These loci were used to genotype individuals from two populations, one native population from the Tocantins River, the other an introduced population in southeast Brazil, Upper Paraná River. Cross-amplification was also successful for another species of peacock bass, C. kelberi. An average of 4.4 alleles per locus (2-9 alleles) was detected. These markers will be useful for the characterization of genetic structure of native populations, and also for invasive biology studies since Cichla species have been introduced in many river basins outside their native ranges.

Plant breeding in black chokeberry (Aronia melanocarpa) is based mainly on seedlings derived from domesticated Russian plants. Previous studies have, however, demonstrated very low levels of phenotypic variation within this gene pool. The present investigation was undertaken in order to study the genetic structure in native populations growing in North America. Random amplified polymorphic DNA (RAPD) marker variation was analysed in eight populations (three or five mother plants/population and five seedlings/mother plant) and compared with the variation in four cultivars and 15 seedlings derived from a Russian plantation. The four cultivars and all the Russian seedlings turned out to have identical RAPD profiles. In the native plant material, there were two types of mother plants: diploid plants that produced very heterogeneous offspring and tetraploid plants that produced homogeneous offspring. Partitioning of variability based on Shannon's diversity index attributed approx. 22% of the variation to the among-population level in diploids, compared to approx. 55% in the tetraploids. However, the diploid populations and the tetraploid populations did not differ significantly in within-population variation. These results prompted a second set of RAPD analyses, which were carried out on offspring obtained through open pollination of the initially examined material when growing in an experimental field. The analyses showed that tetraploid plants produced tetraploid offspring that, with few exceptions, were identical, indicating apomixis, whereas offspring of diploid plants were diploid or triploid, and highly heterogeneous, indicating outcrossing. Presumably, the tetraploid form of Aronia is an allopolyploid, with A. melanocarpa as one of the parents.

Lack of detectable nepotism in multiple-queen colonies of the fire ant Solenopsis invicta (Hymenoptera: Formicidae)

Multiple-queen (polygyne) colonies of the introduced fire ant Solenopsis invicta present a paradox for kin selection theory. Egg-laying queens within these societies are, on average, unrelated to one another, and the numbers of queens per colony are high, so that workers appear to raise new sexuals that are no more closely related to them than are random individuals in the population. This paradox could be resolved if workers discriminate between related and unrelated nestmate sexuals in important fitness-related contexts. This study examines the possibility of such nepotism using methods that combine the following features: (1) multiple relevant behavioral assays, (2) colonies with an unmanipulated family structure, (3) multiple genetic markers with no known phenotypic effects, and (4) a statistical technique for distinguishing between nepotism and potentially confounding phenomena. We estimated relatedness between interactants in polygyne S. invicta colonies in two situations, workers tending egg-laying queens and workers feeding maturing winged queens. In neither case did we detect a significant positive value of relatedness that would implicate nepotism. We argue that the non-nepotistic strategies displayed by these ants reflect historical selection pressures experienced by native populations, in which nestmate queens are highly related to one another. The markedly different genetic structure in native populations may favor the operation of stronger higher-level selection that effectively opposes weaker individual-level selection for nepotistic interactions within nests.
