Acetyl-CoA carboxylase inhibition increases retinal pigment epithelial cell fatty acid flux and restricts apolipoprotein efflux

Lipid-rich deposits called drusen accumulate under the retinal pigment epithelium (RPE) in the eyes of patients with age-related macular degeneration (AMD) and Sorsby’s fundus dystrophy (SFD). Drusen may contribute to photoreceptor and RPE degeneration in these blinding diseases. We hypothesize that stimulating β-oxidation of fatty acids could decrease the availability of lipid with which RPE cells can generate drusen. Inhibitors of acetyl-CoA carboxylase (ACC) stimulate β-oxidation and diminish lipid accumulation in fatty liver disease. In this report we test the hypothesis that an ACC inhibitor, Firsocostat, can diminish lipid deposition by RPE cells. We probed metabolism and cellular function in mouse RPE-choroid tissue and in cultured human RPE cells. We used 13C6-glucose, 13C16-palmitate, and gas chromatography-linked mass spectrometry to monitor effects of Firsocostat on glycolytic, Krebs cycle, and fatty acid metabolism. We quantified lipid abundance, apolipoprotein E (ApoE) and vascular endothelial growth factor (VEGF) release using liquid chromatography-mass spectrometry, enzyme-linked immunosorbent assays and localized ApoE deposits by immunostaining. RPE barrier function was assessed by trans-epithelial electrical resistance (TEER). Firsocostat-mediated ACC inhibition increases β-oxidation, decreases intracellular lipid levels, diminishes lipoprotein release, and increases TEER. When human serum or outer segments are used to stimulate lipoprotein release, fewer lipoproteins are released in the presence of either lipid source and Firsocostat. In a culture model of SFD, Firsocostat stimulates fatty acid oxidation, increases TEER, and decreases ApoE release. We conclude that Firsocostat remodels RPE metabolism and can limit lipid deposition. This suggests that ACC inhibition could be an effective strategy for diminishing pathologic drusen in the eyes of patients with AMD or SFD.

Chemical characterization and response of house rats towards faecal odour of conspecifics

Repeated use of rodenticides for rodent pest management leads to the development of bait shyness and resistance in rodents. Therefore, using pheromones for rodent management could be a significant step forward. The present study was conducted to determine the response of house rats towards odour of faecal extracts of conspecifics of different age groups and sexes. Rats of different maturity groups and sexes were exposed to 20% faecal extract of mature and immature male and female rats. Results indicated that faecal extract of estrous stage mature females has a significant attractant/repellent effect on mature rats of opposite/same sex, respectively. Gas chromatography-linked mass spectrometry of the faecal extract of estrous female rats identified 62 compounds, of which 16 were already reported in other animals as pheromonal compounds and might be used in communication by house rats. In the future, these identified compounds can be used in a fixed proportion after testing their efficacy to develop a stable lureto increase the trappability and acceptability of antifertility agents and rodenticide-based baits for the management of house rats.

Semiochemical signatures associated with differential attraction of Anopheles gambiae to human feet.

BackgroundSeveral human-produced volatiles have been reported to mediate the host-seeking process under laboratory conditions, yet no effective lure or repellent has been developed for field application. Previously, we found a gradation of the attractiveness of foot odors of different malaria free individuals to Anopheles gambiae sensu stricto Giles. In this study, foot odor of the individual with the most attractive ‘smelly’ feet to the An. gambiae was collected, analyzed and attractive blend components identified.MethodsThe foot odor of the individual with the most attractive ‘smelly’ feet to the An. gambiae was trapped on Porapak Q and analyzed by gas chromatography-linked mass spectrometry (GC-MS). Specific constituents perceived by the insect olfactory system were then identified by GC-linked to electro-antennography detector (GC-EAD) and characterized by GC-MS. The contribution of each constituent to the behavioral response of An. gambiae was assessed through subtractive assays under semi-field conditions in a screen-house using Counter Flow Geometry (CFG traps) baited with (i) the blend of all the EAD-active and (ii) other blends containing all components with exclusion of one component at a time. The number of mosquitoes trapped in the baited CFG traps were compared with those in the control traps.ResultsEleven major and minor constituents: 2 carboxylic acids, six aldehydes, two ketones and one phenolic compound, were confirmed to be EAD-active. The contribution of each constituent to the behavioral response of An. gambiae was assessed through subtractive assays under semi- field conditions. Exclusion/ subtraction of one of the following compounds: i-butyric acid, i-valeric acid, n-octanal, n-nonanal, n-decanal, n-dodecanal, undecanal or n-tridecanal, from each blend led to reduction in the attractiveness of all the resulting blends, suggesting that all of them are critical/important for the attractiveness of the foot odor to An. gambiae mosquitoes. However, exclusion/subtraction of 4-ethoxyacetophenone, 4-ethylacetophenone and/or 2-methylphenol, led to significant enhancements in the attractiveness of the resulting blends, suggesting that each of these compounds had repellent effect on An. gambiae ss. Undecanal exhibited kairomonal activity at low natural concentrations under semi-field conditions but repellent activity at high unnatural conditions in the laboratory. Furthermore, the comparison of the mean mosquito catches in traps baited with the nine-component blend without 4-ethoxyacetophenone, 4-ethylacetophenone and the complete foot odor collection revealed that the former is significantly more attractive and confirmed the repellent effect of the two carbonyl compounds at low natural concentration levels.ConclusionThese results suggest that differential attractiveness of An. gambiae to human feet is due to qualitative and/or qualitative differences in the chemical compositions of the foot odors from individual human beings and relative proportions of the two chemical signatures (attractants versus repellents) as observed from the ratios of the bioactive components in the foot odors of the most attractive and least attractive individuals. Chemical signature means the ensemble of the compounds released by the organism in a specific physiological state. The chemical signature is emitter-dependent, but does not depend on receiver response. Thus, there is only one chemical signature for one individual or species that may eventually include inactive, attractive and repellent components for another organism. The nine-component attractive blend has a potential as an effective field bait for trapping of malaria vectors in human dwellings.

Analysis of emission profiles from charcoal produced from selected tree species by different pyrolysis methods

The use of charcoal is associated with indoor air pollution where users are continually exposed to combustion pollutants with detrimental effects to their health. This study analyzed the emission profiles of selected domestically used charcoal produced by different pyrolysis methods. Charcoal samples from three tree species Acacia polyacantha, Acacia xanthophloea and Eucalyptus grandis, produced by both efficient and traditional pyrolysis were collected from charcoal producers in rural Kenya. The volatile organic compounds were collected using a fabricated chimney placed on clay cook stove such that smoke was vented to a glass cannula and condensed. Extracted volatiles were analyzed by gas chromatography-linked mass spectrometry. Twenty-two polycyclic aromatic hydrocarbons were identified together with other hydrocarbons. Polycyclic aromatic hydrocarbons found in the smoke extracts with known health effects included naphthalene, 1-methylnaphthalene, 2-methylnaphthalene, fluorene, phenanthrene, anthracene, 1-methylphenanthrene, fluoranthene, benzo(a)anthracene and chrysene. There were significant differences in the concentrations of fluorine, naphthalene and pyrene emitted from Acacia polyacantha and Acacia xanthophloea prepared by traditional pyrolysis. From Acacia polyacantha, concentrations were naphthalene (110 µg/g), fluorine (72 µg/g) and pyrene (60 µg/g), while from Acacia xanthophloea, concentrations were naphthalene (140.42 µg/g), fluorene (97.35 µg/g) and pyrene (71.82 µg/g). Charcoal prepared by traditional pyrolysis emitted the highest number of polycyclic aromatic hydrocarbons and also had higher concentration of polycyclic aromatic hydrocarbons relative to the accepted 0.1–0.2 mg/m3 levels, while those prepared by efficient pyrolysis produced the lowest concentration. Therefore, there is need to use efficient pyrolysis methods of charcoal production.

Influence of enzymatic extrusion liquefaction pretreatment for Chinese rice wine on the volatiles generated from extruded rice.

Volatile compounds in enzymatic extruded rice, produced under different conditions of varying barrel temperature (BT), α-amylase concentration (AC) and moisture content (MC), were extracted and identified by headspace solid phase microextraction (HS-SPME) and gas chromatography-linked mass spectrometry (GC-MS). Statistical analyses reflected that the Maillard reaction could be inhibited both by the mild extrusion conditions and the enhanced hydrolysis caused by thermostable α-amylase. Relative amounts of total volatiles in enzymatic extruded rice were far less than those in severe processed extruded rice. Reverse-phase high-performance liquid chromatography (RP-HPLC) showed that the amino acids (AAs) involved in Maillrad reaction were utmostly preserved in extruded rice with highest amylase concentration by comparison of total AA content of different extrudates. These results suggest that enzymatic extrusion liquefaction is an effective way to control the generation of volatiles from extruded rice for Chinese rice wine production.

Sensor arrays based on nanoparticles for early detection of kidney injury by breath samples

The outcomes of acute kidney injury (AKI) could be severe and even lethal, if not diagnosed in its early stages and treated appropriately. Blood and urine biomarkers, currently in use as indicators for kidney function, are either inaccurate in various cases or not timely. We report on dramatic changes in exhaled breath composition, associated with kidney dysfunction after ischemic insult in rat models. Gas chromatography linked mass spectrometry examination of breath samples indicated significant elevations in the concentration of three exhaled volatile organic compounds, two to six hours after AKI was surgically induced. Relying on these findings, we introduce an array of sensors, based on organic-layer capped gold nanoparticles, sensitive to odor changes. The ability of the array to detect AKI via breath testing was examined and scored a sensitivity of 96%, only one hour after disease induction. From the Clinical EditorIn this study, organic-layer capped gold nanoparticle-based biosensors are used to analyse breath samples in an acute kidney injury model, capitalizing on the observation that specific volatile organic compounds are present in breath samples in that condition. The authors report excellent sensitivity in as little as one hour after acute kidney injury. This method, if commercialized, may replace the current blood and urine sample analysis-based tests with a more convenient, rapid and accurate nanotechnology-based method.

Identification of p-Cresol as an Estrus-Specific Volatile in Buffalo Saliva: Comparative Docking Analysis of Buffalo OBP and β-Lactoglobulin with p-Cresol

Assessment of salivary volatile compounds adopting gas chromatography-linked mass spectrometry (GC-MS) analysis revealed the presence of a total of 11 compounds in the buffalo saliva irrespective of the stages in the reproductive cycle. p-cresol was identified as an estrus-specific volatile compound in the saliva. In addition, modeling of odorant-binding protein (OBP) and β-lactoglobulin revealed that OBP is highly stable and has strong binding affinity with p-cresol. Hydrogen bond interactions indicated that OBP is responsible for pheromone release through saliva. In contrast, β-lactoglobulin, which belongs to the same lipocalin family as OBP, possesses less affinity to p-cresol than OBP, suggesting that it is not involved in p-cresol binding and transport. Phylogenetic characterization revealed that bovine family of OBP is separately clustered. It is suggested that p-cresol has the potential to be developed as a biomarker to detect the reproductive status in the buffalo and for behavioral manipulations.

Differential attractiveness of human foot odours to Anopheles gambiae Giles sensu stricto (Diptera: Culicidae) and variation in their chemical composition

Counter flow geometry (CFG) traps (American Biophysics) baited with foot odours (adsorbed overnight on a combination of a nylon and a cotton socks) from 4 groups of 4 male volunteers were initially used in a screen-house to compare and grade the attractiveness of Anopheles gambiae Giles sensu stricto to these odours. Ten individuals were then selected from the 4 groups for further grading in which mosquito attractiveness to odours adsorbed on socks worn by each of the individuals was compared against a control (clean, un-worn cotton and nylon socks). A gradation of attractiveness was found, with the most attractive foot odour catching 8-fold more mosquitoes than the least attractive one (t-test, p=0.001). For comparison of the chromatographic profiles of the foot odours and identification of their constituents, six adsorbents (tenax, chromsorb, porapak Q, activated charcoal, reverse phase octadecyl and octyl bonded silica) were evaluated for their suitability in trapping the volatiles in a static mode. Of these, porapak Q was found to be the most effective. Comparison of the gas-chromatographic (GC) and gas chromatography–linked mass spectrometry (GC–MS) profiles of blends collected from the most and least attractive feet on porapak Q adsorbent revealed both qualitative and quantitative differences. The implication of our finding on differential attraction of An. gambiae to their preferred feeding site of different human subjects is highlighted.

Gas chromatographic-mass spectrometric analysis of chemical volatiles in buffalo (Bubalus bubalis) urine

Isolation of active fraction and characterization of chemosignals from urine have been attempted in several mammalian species in the recent years. The objective of this study was to identify the urinary volatiles across various reproductive stages of buffalo cow, namely, estrus, diestrus, and pregnancy, and in bull, by chemical extraction followed by gas chromatography–linked mass spectrometry (GC-MS). Urine samples were collected from six buffalo cows at two different phases of estrous cycle, namely, estrus and diestrus. Besides, urinary samples were collected from five pregnant buffalo cows (60–75 days after artificial insemination (AI)) and six adult bulls. Thin-layer chromatography was performed as a preliminary test for qualitative comparison of different compounds extracted by organic solvents. Identification of the urinary compounds was carried out in a gas chromatograph (Perkin Elmer, Autosystem XL) linked to a mass spectrometer (Turbomass). The results of GC-MS analysis indicated the presence of 21 compounds with varying molecular weights and retention time, which were further categorized as diestrus-specific, pregnancy-specific, and bull-specific urinary compounds. No compound, however, could be identified as estrus-specific. We concluded that qualitative differences do exist in estrus, diestrus, and pregnant buffalo cow urine and in bull urine, as evidenced by GC-MS.

<i>In vitro</i> anti-tick properties of the essential oil of <i>Tagetes minuta</i> L. (Asteraceae) on <i>Hyalomma rufipes</i> (Acari: Ixodidae)

In this study we examined the anti-tick properties of the essential oil of Tagetes minuta L. (Asteraceae: Asterales) against Hyalomma rufipes ticks. We obtained the essential oil of T. minuta by hydro-distillation of a combination of fresh flowers, leaves and soft stems, and analysed these by using gas chromatography (GC) and gas chromatography-linked mass spectrometry (GC-MS). The oil had a high percentage of monoterpenes and the major compounds identified were cis-ocimene (28.5%), beta-ocimene (16.83%) and 3-methyl-2-(2-methyl-2-butenyl)-furan (11.94%). Hyalomma rufipes adults displayed a significant (P < 0.05) dose repellent response to the essential oil of T. minuta. Probit analysis indicated a repellent EC50 of T. minuta essential oil for male ticks to be 0.072 mL/mL (CI 0.053 mL/mL to 0.086 mL/mL) and 0.070 mL/mL (CI 0.052 mL/mL to 0.084 mL/mL) for female ticks. There were no significant differences in repellent responses between male and female ticks. The oil also significantly (P < 0.05) delayed moulting of 60% of H. rufipes engorged nymphs. These results suggest that T. minuta may be a potential source of anti-tick agents.

Increased squalene concentrations in the clitoral gland during the estrous cycle in rats: An estrus-indicating scent mark?

Squalene in the rat clitoral gland is reported to be semi-volatile and may serve as a chemo-signal. The objective was to determine squalene concentrations in the clitoral gland throughout the reproductive cycle. Clitoral glands were extracted with dichloromethane; 23 compounds were identified with Gas Chromatography linked Mass Spectrometry (GC-MS). Since squalene concentrations were significantly higher during proestrus and estrus, and remarkably reduced during metestrus and diestrus, we inferred that it could be an ovulation-indicating chemosignal in the female rat, acting as a scent mark for the male. This hypothesis was tested by investigating its efficacy to attract males, including studying the role of the olfactory-vomeronasal system of the male in perceiving squalene. For detection of squalene, males used their conventional olfactory system when at a distance from the female, whereas the vomeronasal organ was used when they were in close proximity to the female. We concluded that squalene was a female-specific chemosignal that attracted males, and furthermore, that the olfactory-vomeronasal system had an important role in the perception of squalene.

1–Iodo-2 methylundecane [1I2MU]: An estrogen-dependent urinary sex pheromone of female mice

In our previous investigations [1], urine of female mice contained specific compounds, namely isocroctylhydrazine, 4-methyl-2-heptanone, and azulene during proestrus, whereas during estrus it contained 1-H-cyclopop.e.azulene, caryophyllene, and copanene. Furthermore, 1-iodo-2 methyl undecane (1I2MU), present during both proestrus and estrus, was regarded as a putative estrus-specific chemo-signal [1]. The primary objective of the present study was to determine the estrogen-dependency of the above-mentioned compounds, including 1I2MU. Furthermore, the effect of these compounds on pre-mating behavior, e.g., sniffing, licking, and grooming, were recorded to determine their role as sex pheromones. Based on gas chromatography linked mass spectrometry (GC-MS) of urine samples, profiles in oophorectomized female mice had 14 major peaks. Furthermore, neither 1I2MU (nor other estrus-specific compounds) were detected in the urine of these mice, although they were detected in urine of proestrus and estrus mice. In addition, 1I2MU was not detected in urine of prepubertal mice. It was noteworthy that both 1I2MU and 4-methyl-2-heptanone reappeared in estrogen-treated females. Based on pre-mating behavioral analysis, 1I2MU was the compound most preferred by males. In conclusion, production of 1I2MU was estrogen-dependent in females, and it enhanced reproductive activities in males.

Discovery and characterization of chemical signals for citrus root weevil, Diaprepes abbreviatus

The tropical root weevil Diaprepes abbreviatus (L. 1758) (Coleoptera: Curculionidae) is a polyphagous insect from the Caribbean Islands and an invasive insect in the southern part of the United States where it is pest of citrus crops and ornamental trees. Adults feed upon foliage where aggregation, mating and oviposition take place. Here, the headspace volatiles from Citrus macrophylla Wester (Rutaceae), D. abbreviatus adults feeding on this plant, adults alone and adult feces, were collected by aeration and solid-phase microextraction (SPME) for analysis by gas chromatography-linked mass spectrometry (GC-MS). Electrophysiological responses of weevil antennal receptors to volatile headspace extracts and synthetic analogues were recorded by gas chromatography-linked electroantennographic detection (GC-EAD) and electroantennograms (EAGs). Antennal responses were recorded to the monoterpenes (R)-(−)-linalool, citronellal, nerol, citral, and geraniol; all present in the headspace of C. macrophylla. Antennal responses were also recorded to carvacrol, present in the headspace of adults and adult feces. The green leaf volatiles cis-3-hexen-1-ol and trans-2-hexen-1-ol, produced by other host plants, elicited reliable responses on the D. abbreviatus antenna. When comparing EAGs between (±)-linalool and (R)-(−)-linalool, no significant difference was found; responses to (R)-(+)-citronellal were larger than for (S)-(−)-citronellal. Among the individual compounds and blends tested using an open T-track dual choice olfactometer, only the blend of (±)-linalool, cis-3-hexen-1-ol and carvacrol (source dose 25:25:2.5 μg) elicited significant attraction of females, the same blend was repellent for males. The biologically active compounds found here likely play a role in host finding by D. abbreviatus and other interactions of the insect with its hostplant.

Essential oil of Ocimum grattissimum (Labiatae) as Sitophilus zeamais (Coleoptera: Curculionidae) protectant

Ocimum grattissimum L. (Labiatae) leaves are widely eaten as a vegetable in Nigeria, and in the eastern parts, are traditionally used in post-harvest protection and relieving stomach aches. The effect of the essential oil of O. grattissimum leaves on Sitophilus zeamais (Motschulsky) (Coleoptera: Curculionidae) was assessed for repellency, mortality, progeny emergence and maize damage in the laboratory. The oil was found to be moderately repellent to the maize weevil and induced high mortality in the weevils. In addition, grains treated with the essential oil showed significant reduction in the number of progeny derived from surviving S. zeamais. There was no observable feeding damage on grains treated with the highest dosage of the essential oil extract. Gas chromatography-linked mass spectrometry (GC-MS) and GC co-injections with authentic samples showed the presence of the following major constituents: thymol (32.7%), paracymene (25.4%),
-terpinene (10.8%),
-selinene (4.5%), phellandrene (3.9%) and
- myrcene (3.1%). The results provide a scientific rationale for the use of the plant in post-harvest protection.

Detection of oestrous-related odour in bovine (Bos taurus) saliva: bioassay of identified compounds

The present study was designed to identify the volatile constituents across the oestrous cycle of bovine in order to detect oestrous-specific chemical signal. The bovine saliva was extracted with diethyl ether (1 : 1 ratio, v/v) and analysed by gas chromatography-linked mass spectrometry. Numerous compounds were identified during oestrous cycle of bovine saliva. Among these, the compounds, namely, trimethylamine, acetic acid, phenol 4-propyl, pentanoic acid and propionic acid were specific to oestrous stage. The behaviour assay revealed that the compound, trimethylamine, is involved in attracting the male animal. The result concludes that the trimethylamine is considered as a putative oestrous-specific salivary chemo-signal in the bovine.

3-Ethyl-2,7-dimethyl octane, a testosterone dependent unique urinary sex pheromone in male mouse ( Mus musculus)

A previous investigation revealed that urine from normal male mice contained five unique volatile constituents; namely: 3-cyclohexene-1-methanol (I); 3-amino triazole (II); 4-ethyl phenol (III); 3-ethyl-2,7-dimethyl octane (IV); 1-iodoundecane (V). The present study was designed to find out whether the production of these male specific urinary compounds was androgen-dependent. Urine of castrated and castrated plus testosterone-treated male mice was analyzed using gas chromatography linked mass spectrometry (GC–MS). Even though castrated male urine contained 10 detectable compounds, the five male specific compounds present in intact males were absent in castrated male mice urine. Only 3-ethyl-2,7-dimethyl octane (IV) reappeared following testosterone treatment into castrated males. Our earlier bioassay revealed that this compound was involved in attracting females. The present study concluded that this compound was a male specific volatile cue that acted as a releaser pheromone and its production was under the control of androgen.

Characterization of urinary volatiles in Swiss male mice (Mus musculus): bioassay of identified compounds.

The present study was carried out to investigate the chemical nature of the urine of male mice and to assess its bioactivity. Urine of mature male mice was extracted with dichloromethane (1 : 1 ratio v/v) and analysed by gas-chromatography linked mass-spectrometry (GC-MS). Ten different compounds such as alkanes, alcohols, etc. were detected in the urine. Among the ten, five compounds are specific to males namely, 3-cyclohexene-1-methanol (I), 3-amino-s-triazole (II), 4-ethyl phenol (III), 3-ethyl-2,7-dimethyl octane (IV) and 1-iodoundecane (V). The compound, 4-ethylphenol, has been previously reported in several strains of male mice. Furthermore, the compounds (II) and (IV) are closely similar to 2-sec-butylthiazole and dehydro-exo-brevicomin compounds which have already been reported in male mice. Bioassay revealed that compounds (II), (III) and (IV) were responsible for attracting females and in inducing aggression towards males, as compared to the other compounds i.e. (I) and (V). The results indicate that these three volatiles (II, III and IV) of male mice appear to act as attractant of the opposite sex.

Odor composition of preferred (buffalo and ox) and nonpreferred (waterbuck) hosts of some Savanna tsetse flies.

A previous study on the feeding responses of tsetse flies, Glossina morsitans morsitans, implicated the existence of allomonal barriers, both volatile and nonvolatile, on the nonpreferred host, waterbuck, Kobus defassa. In the present study, electroantennogram-active compounds in odors from waterbuck were compared with those of two preferred hosts of tsetse flies, buffalo, Syncerus caffer, and ox, Bos indicus. Odors from the three bovids were trapped on activated charcoal and/or reverse-phase (octadecyl bonded) silica and analyzed with a gas chromatography-linked electroantennographic detector (GC-EAD) and, where possible, identified by using gas chromatography-linked mass spectrometry (GC-MS) and chromatographic comparisons with authentic samples. The GC-EAD profiles (with G. m. morsitans antennae) of the odors of the two preferred hosts were comparable, comprising medium-chain, saturated or unsaturated aldehydes and phenols, with buffalo emitting a few more EAG-active aldehydes. Waterbuck odor gave a richer profile, consisting of fewer aldehydes but more phenolic components and a series of 2-ketones (C-C13) and delta-octalactone. This bovid also emits moderate amounts of C5-C9 straight-chain fatty acids, some of which were detected in buffalo and ox only in trace amounts. However, these did not elicit significant GC-EAD responses. Waterbuck profiles from the antennae of G. pallidipes showed broad similarity to those from G. m. morsitans, although the composition of aldehydes and ketones was somewhat different, indicating species-specific difference in the detection of host odors. Certain waterbuck-specific EAG-active components, particularly the 2-ketones and lactone, constitute a candidate allomonal blend in waterbuck odor.

Chemistry of clitoral gland secretions of the laboratory rat: assessment of behavioural response to identified compounds.

The present investigations were carried out to find out the chemical nature of clitoral gland extracts and their involvement in reproductive and social behaviour. Homogenates of clitoral glands of mature estrous female rats were extracted with n-hexane and dichloromethane (1 : 1 ratio v/v) and analysed by gas chromatography linked mass spectrometry (GC-MS). Three peaks were found to be in higher concentration, which were identified as 6,11-dihydro-dibenz-b,e-oxepin-11-one (I); 2,6,10-dodecatrien-1-ol-3,7,11-trimethyl(Z) (II); and 1,2-benzene-dicarboxylic acid butyl(2-ethylpropyl) ester (III). Odour preference tests demonstrated that the first compound attracted conspecifics of the opposite sex. By contrast, the second and third compounds were found to attract both sexes. The results conclude that the clitoral gland of laboratory rat contains three major chemical compounds which have a unique function in maintaining social and reproductive status.
