Phase coding of spatial representations in the human entorhinal cortex.

The grid-like activity pattern of cells in the mammalian entorhinal cortex provides an internal reference frame for allocentric self-localization. The same neurons maintain robust phase couplings with local field oscillations. We found that neurons of the human entorhinal cortex display consistent spatial and temporal phase locking between spikes and slow gamma band local field potentials (LFPs) during virtual navigation. The phase locking maintained an environment-specific map over time. The phase tuning of spikes to the slow gamma band LFP revealed spatially periodic phase grids with environment-dependent scaling and consistent alignment with the environment. Using a Bayesian decoding model, we could predict the avatar’s position with near perfect accuracy and, to a lesser extent, that of heading direction as well. These results imply that the phase of spikes relative to spatially modulated gamma oscillations encode allocentric spatial positions. We posit that a joint spatiotemporal phase code can implement the combined neural representation of space and time in the human entorhinal cortex.

Characterizing the orientation selectivity in V1 and V4 of macaques by quadratic phase coupling

Objective. Orientation selectivity is one of the significant characteristics of neurons in the primary visual cortex (V1). Some neurons in extrastriate visual cortical areas also exhibit certain orientation selectivity. But it is still not well understood that how the orientation selectivity generates. Most previous studies about the orientation selectivity are based on the spike firing rate. However, the spikes are prone to be biased by the detection and sorting algorithms. Then, in this paper, the local field potential (LFP) is adopted to investigate the mechanism of orientation selectivity. Approach. We used the quadratic phase coupling (QPC), which was calculated by wavelet bicoherence, to describe the characteristics of orientation selectivity available in V1 and V4. The raw wideband neural signals were recorded by two chronically implanted multi-electrode arrays, which were placed in V1 and V4 respectively in two macaques performing a selective visual attention task. Main results. There is a strong correlation between the total bicoherence (TotalBic), which is a quantization for the overall QPC of frequency pairs in gamma band, and the grating orientation. Furthermore, the QPC distribution at the non-preferred orientation is mainly concentrated in the low frequencies (30–40 Hz) of gamma; while the QPC distribution at the preferred orientation concentrates in both the low frequencies and high frequencies (60–80 Hz) of gamma. In addition, the TotalBic of the gamma-band LFP between V1 and V4 varies with the grating orientations, indicating that the QPC is available in the feedforward link and the gamma-band LFP in V1 modulates the QPC in V4. Significance. The QPC reflects the orientations of the sinusoidal grating and describes the interaction of gamma-band LFP between different brain regions. Our results suggest that the QPC is an alternative avenue to explore the mechanism for generating orientation selectivity of visual neurons effectively.

Neurovascular Coupling under Chronic Stress Is Modified by Altered GABAergic Interneuron Activity.

Neurovascular coupling (NVC), the interaction between neural activity and vascular response, ensures normal brain function by maintaining brain homeostasis. We previously reported altered cerebrovascular responses during functional hyperemia in chronically stressed animals. However, the underlying neuronal-level changes associated with those hemodynamic changes remained unclear. Here, using in vivo and ex vivo experiments, we investigate the neuronal origins of altered NVC dynamics under chronic stress conditions in adult male mice. Stimulus-evoked hemodynamic and neural responses, especially beta and gamma-band local field potential activity, were significantly lower in chronically stressed animals, and the NVC relationship, itself, had changed. Further, using acute brain slices, we discovered that the underlying cause of this change was dysfunction of neuronal nitric oxide synthase (nNOS)-mediated vascular responses. Using FISH to check the mRNA expression of several GABAergic subtypes, we confirmed that only nNOS mRNA was significantly decreased in chronically stressed mice. Ultimately, chronic stress impairs NVC by diminishing nNOS-mediated vasodilation responses to local neural activity. Overall, these findings provide useful information in understanding NVC dynamics in the healthy brain. More importantly, this study reveals that impaired nNOS-mediated NVC function may be a contributory factor in the progression of stress-related diseases.SIGNIFICANCE STATEMENT The correlation between neuronal activity and cerebral vascular dynamics is defined as neurovascular coupling (NVC), which plays an important role for meeting the metabolic demands of the brain. However, the impact of chronic stress, which is a contributory factor of many cerebrovascular diseases, on NVC is poorly understood. We therefore investigated the effects of chronic stress on impaired neurovascular response to sensory stimulation and their underlying mechanisms. Multimodal approaches, from in vivo hemodynamic imaging and electrophysiology to ex vivo vascular imaging with pharmacological treatment, patch-clamp recording, FISH, and immunohistochemistry revealed that chronic stress-induced dysfunction of nNOS-expressing interneurons contributes to NVC impairment. These findings will provide useful information to understand the role of nNOS interneurons in NVC in normal and pathological conditions.

Persistent Gamma Spiking in SI Nonsensory Fast Spiking Cells Predicts Perceptual Success

Gamma oscillations (30-55Hz) are hypothesized totemporally coordinate sensory encoding, enabling perception. However, fast spiking interneurons (FS), key gamma generators, can be highly sensory responsive, as is the gamma band local field potential (LFP). How can FS-mediated gamma act as an impartial temporal reference for sensory encoding, when the sensory drive itself presumably perturbs the pre-established rhythm? Combining tetrode recording in SI barrel cortex with controlled psychophysics, we found a unique FS subtype that was not sensory responsive and spiked regularly at gamma range intervals (gamma regular nonsensory FS [grnsFS]). Successful detection was predicted by a further increase in gamma regular spiking of grnsFS, persisting from before to after sensory onset. In contrast, broadband LFP power, including gamma, negatively predicted detection and did not cohere with gamma band spiking by grnsFS. These results suggest that a distinct FS subtype mediates perceptually relevant oscillations, independent of the LFP and sensory drive.

Face Repetition Probability Does Not Affect Repetition Suppression in Macaque Inferotemporal Cortex.

Repetition suppression, which refers to reduced neural activity for repeated stimuli, is typically explained by bottom-up or local adaptation mechanisms. However, recent theories have emphasized the role of top-down processes, suggesting that this response reduction reflects the fulfillment of perceptual expectations. To support this, an influential human fMRI study showed that the magnitude of suppression is modulated by the probability of a repetition. No such repetition probability effect was found in macaque inferior temporal (IT) cortex for spiking activity despite the presence of repetition suppression. Contrary to the human fMRI studies that showed an effect of repetition probability, the macaque single-unit study used a large variety of unfamiliar stimuli and the monkeys were not required to attend the stimuli. Here, as in the human fMRI studies, we used faces as stimuli and made the monkeys attend to the stimulus content. We simultaneously recorded spiking activity and local field potentials (LFPs) in the middle lateral face patch (ML) of one monkey (male) and a face-responsive region of another (female). Although we observed significant repetition suppression of spiking activity and high gamma-band LFPs in both animals, there were no effects of repetition probability even when repetitions were task relevant and repetition probability affected behavioral decisions. In conclusion, despite the use of face stimuli and a stimulus-related task, no neural signature of repetition probability was present for faces in a face responsive patch of macaque IT. This further challenges a general perceptual expectation account of repetition suppression.SIGNIFICANCE STATEMENT Repetition suppression is a reduced brain activity for repeated stimuli commonly observed across species. In the predictive coding framework, such suppression is thought to reflect fulfilled perceptual expectations. Although this hypothesis is supported by several human fMRI studies reporting an effect of repetition probability on repetition suppression, this could not be replicated in single-cell recordings in monkey inferior temporal (IT) cortex. Subsequent studies narrowed down the conditions for the effect to requiring attention and being limited to particular stimulus categories such as faces. Here, we show that, even under these conditions, repetition suppression in monkey IT neurons is still unaffected by repetition probability, even in a task with a behavioral effect, challenging the perceptual expectation account of repetition suppression.

Persistent Gamma Spiking in Non-Sensory Fast-Spiking Cells Predicts Perceptual Success

Persistent gamma oscillations (30-55Hz) are hypothesized to temporally coordinate stimulus encoding, enabling perception. This prediction poses a conundrum: How can gamma serve as a template when the stimulus itself drives its mediators, presumably perturbing its maintenance? Specifically, fast-spiking interneurons (FS), a key gamma generator, can be highly sensory responsive. Further, the gamma-band local field potential (LFP) shows properties inconsistent with temporal coordination. Combining tetrode recording with controlled psychophysics revealed an FS subtype (γnsFS) that was not sensory responsive, whose inter-spike intervals peaked at gamma, and that fired with higher periodicity than other FS. Successful detection was predicted by increased regularity in γnsFS spiking at gamma, persisting from before to after sensory onset. In contrast, gamma LFP power negatively predicted detection, and was negatively related to gamma band spiking by γnsFS. These results suggest that a distinct interneuron subgroup, not ‘distracted’ by sensory input, mediates perceptually-relevant oscillations independent of LFP.

A coupled-oscillator model of olfactory bulb gamma oscillations.

The olfactory bulb transforms not only the information content of the primary sensory representation, but also its underlying coding metric. High-variance, slow-timescale primary odor representations are transformed by bulbar circuitry into secondary representations based on principal neuron spike patterns that are tightly regulated in time. This emergent fast timescale for signaling is reflected in gamma-band local field potentials, presumably serving to efficiently integrate olfactory sensory information into the temporally regulated information networks of the central nervous system. To understand this transformation and its integration with interareal coordination mechanisms requires that we understand its fundamental dynamical principles. Using a biophysically explicit, multiscale model of olfactory bulb circuitry, we here demonstrate that an inhibition-coupled intrinsic oscillator framework, pyramidal resonance interneuron network gamma (PRING), best captures the diversity of physiological properties exhibited by the olfactory bulb. Most importantly, these properties include global zero-phase synchronization in the gamma band, the phase-restriction of informative spikes in principal neurons with respect to this common clock, and the robustness of this synchronous oscillatory regime to multiple challenging conditions observed in the biological system. These conditions include substantial heterogeneities in afferent activation levels and excitatory synaptic weights, high levels of uncorrelated background activity among principal neurons, and spike frequencies in both principal neurons and interneurons that are irregular in time and much lower than the gamma frequency. This coupled cellular oscillator architecture permits stable and replicable ensemble responses to diverse sensory stimuli under various external conditions as well as to changes in network parameters arising from learning-dependent synaptic plasticity.

Single-unit activity during natural vision: diversity, consistency, and spatial sensitivity among AF face patch neurons.

Several visual areas within the STS of the macaque brain respond strongly to faces and other biological stimuli. Determining the principles that govern neural responses in this region has proven challenging, due in part to the inherently complex stimulus domain of dynamic biological stimuli that are not captured by an easily parameterized stimulus set. Here we investigated neural responses in one fMRI-defined face patch in the anterior fundus (AF) of the STS while macaques freely view complex videos rich with natural social content. Longitudinal single-unit recordings allowed for the accumulation of each neuron's responses to repeated video presentations across sessions. We found that individual neurons, while diverse in their response patterns, were consistently and deterministically driven by the video content. We used principal component analysis to compute a family of eigenneurons, which summarized 24% of the shared population activity in the first two components. We found that the most prominent component of AF activity reflected an interaction between visible body region and scene layout. Close-up shots of faces elicited the strongest neural responses, whereas far away shots of faces or close-up shots of hindquarters elicited weak or inhibitory responses. Sensitivity to the apparent proximity of faces was also observed in gamma band local field potential. This category-selective sensitivity to spatial scale, together with the known exchange of anatomical projections of this area with regions involved in visuospatial analysis, suggests that the AF face patch may be specialized in aspects of face perception that pertain to the layout of a social scene.

Modelling of neocortical neural dynamics during human focal seizures

Recent advances in microelectrode array (MEA) recordings have made possible the simultaneous recordings of spiking activity in ensembles of neocortical single neurons during human focal seizures. As a result, a much more detailed picture of seizure initiation and spread has begun to emerge [1]. Here, we combine single neuron spiking and local field potential (LFP) data obtained from 96-MEA recordings with mathematical modelling to characterize the synaptic coupling and parameters required to sustain the neural dynamics observed in the two main types of epileptic seizures identified in the studied patients: spike-wave-complex (SWCs ~2-3 Hz) seizures and gamma-band (40-60 Hz) seizures, respectively. We model the neocortex under a 96-MEA (4mm x 4mm) with Izhikevich type neurons [2,3]. There are 28x28 minicolumns in the model. Each minicolumn is 25μm apart from each other and has 12 excitatory neurons and 4 interneurons. The neurons are synaptically connected up to a radius of 75μm and a synaptic delay of 2ms. (Connectivity drops to zero close to the MEA boundaries). Each neuron is driven by an independent Poisson input simulating background synaptic activities (i.e. high Poisson rates close to a few kHz, but individual Poisson events are of small amplitude). We simulate each of the two types of epileptic seizures by controlling the synaptic strengths amongst neurons and the Poisson drive. Dynamics similar to that in gamma-band seizures (Fig. ​(Fig.1)1) were obtained in models that combined strong mutual inhibitory synaptic couplings (gi->i), strong inhibitory to excitatory synaptic couplings (gi->e), and strong Poisson drive to both the pyramidal and interneuronal populations. The resulting single neuron spiking patterns (in both the inhibitory and excitatory cases) remained irregular, despite that they lead to sustained narrow gamma band LFP oscillations, consistent with the observations from the neuronal spiking data recorded from the patients. Dynamics similar to SWC seizures were obtained by decreasing both mutual inhibitory coupling (gi->i ) and inhibitory to excitatory coupling (gi->e), as well as by reducing the Poisson drive to the inhibitory interneuron populations. During each SWC discharge (Figure ​(Figure1),1), we observed the spread of neuronal spiking activity and recruitment of pyramidal cells starting from a focus, accompanied by strong coherent LFP. In the model, the participation of interneurons in SWC generation is minimal due to the weak gi->i,e and low Poisson drive. Our model suggests that gamma-band seizures require strong inhibitory conductance and participation of both excitatory and inhibitory populations, while SWC seizures require weaker inhibitory conductances and are mostly driven by excitatory (pyramidal) neurons. We are working towards a conductance-based network model in which intra and extracellular ionic concentrations are included to explore how disregulation of ionic concentrations can trigger each of the two types of seizures. Theta phase shift in spike timing and modulation of gamma oscillation: a dynamic code for spatial alternation during fixation in rat hippocampal area CA1

Although hippocampus is thought to perform various memory-related functions, little is known about the underlying dynamics of neural activity during a preparatory stage before a spatial choice. Here we focus on neural activity that reflects a memory-based code for spatial alternation, independent of current sensory and motor parameters. We recorded multiple single units and local field potentials in the stratum pyramidale of dorsal hippocampal area CA1 while rats performed a delayed spatial-alternation task. This task includes a 1-s fixation in a nose-poke port between selecting alternating reward sites and so provides time-locked enter-and-leave events. At the single-unit level, we concentrated on neurons that were specifically active during the 1-s fixation period, when the rat was ready and waiting for a cue to pursue the task. These neurons showed selective activity as a function of the alternation sequence. We observed a marked shift in the phase timing of the neuronal spikes relative to the theta oscillation, from the theta peak at the beginning of fixation to the theta trough at the end of fixation. The gamma-band local field potential also changed during the fixation period: the high-gamma power (60-90 Hz) decreased and the low-gamma power (30-45 Hz) increased toward the end. These two gamma components were observed at different phases of the ongoing theta oscillation. Taken together, our data suggest a switch in the type of information processing through the fixation period, from externally cued to internally generated.

Input dependence of local field potential spectra: experiment vs theory

How sensory stimuli are encoded in neuronal activity is a major challenge for understanding perception. A prominent effect of sensory stimulation is to elicit oscillations in EEG and Local Field Potential (LFP) recordings over a broad range of frequencies. Belitski et al. [1] recorded LFPs and spiking activity in the primary visual cortex of anaesthetized macaques presented with naturalistic movies and found that the power of the gamma and low-frequency bands of LFP carried largely independent information about visual stimuli, while the information carried by the spiking activity was largely redundant with that carried by the gamma-band LFPs. To understand better how different frequency bands of the LFP are controlled by sensory input, we computed analytically the power spectrum of the LFP of a theoretical model of V1 (a network composed of two populations of neurons - excitatory and inhibitory), subjected to time-dependent external inputs modelling inputs from the LGN, as a function of the parameters characterizing single neurons, synaptic connectivity, as well as parameters characterizing the statistics of external inputs. Our model consists in a two populations network of excitatory and inhibitory leaky integrate-and-fire neurons. Standard analytical methods using the Fokker-Planck formalism can be used to compute average firing rates of both populations in the asynchronous state of the network, as well as the region of parameters for which this state is stable ([2,3]). The power spectrum of the global activity and the LFP (sum of average excitatory and inhibitory currents onto pyramidal cells of the network) can also be computed in a network of finite size ([2]). Using linear response theory we then calculated the response of the network to a dynamic input ([4]). The final result was an equation giving the LFP power spectrum as a function of the intrinsic parameters of the network and of the parameters characterizing the dynamic input. We then used the analytical expression of the LFP power to fit the experimental data of [1]. The data consists in LFP recordings from primary visual cortex of monkeys, during the presentation of a movie that last several minutes. In order to capture the LFP power changes during the movie presentation, the LFP traces were divided into 2 seconds non-overlapping scenes. We then fitted the LFP power of all the scenes with the same network parameters, but with input parameters free to vary scene-by-scene. We used a simplex method repeatedly applied for different initial conditions. The parameter set that was selected was the one that minimized the reduced chisquare, among sets for which the asynchronous state was stable. The model provided excellent fits of the data. The fitting procedure permitted to extract the values of the firing rates of the excitatory and inhibitory populations and the parameters characterizing the external input for most of the scenes of the movie. These outcomes could be then correlated with experimental firing rates and the features of the movie itself, such as temporal and spatial contrast as well as orientation. We found a significant correlation both between firing rates extracted from fit and the multi-unit activity recorded during the movie and between the parameters characterizing the external input and the features of the movie. These results show how an analytical approach can be used to estimate the key parameters underlying changes in the LFP spectral dynamics.

Non-linear dependency between spiking response and gamma-band power of local field potentials in the human auditory cortex

Recent advancement in recordings of extracellular potentials from multiple sites in the human brain [1] has allowed us to simultaneously record spiking activity and local field potentials (LFP) to examine the brain's response to experimental stimuli. Various mechanisms have been suggested for the generation of the LFP. A point of special interest is that LFP reflects not only the inputs to the tissue, but also local processing. For example, spike contribution to the high-frequency LFP through increased spike rate or increased synchrony has been suggested [2]. Subjects (n = 19 ) were epilepsy patients who needed intracranial electrode implantation and chronic monitoring of for diagnostic and treatment purposes. Protocols were approved by University of Iowa IRB. Recordings were made from micro contacts implanted in the Heschl's gyri during pure tone presentations. Single-trial gamma-band responses were extracted using band-pass filters (40-150 Hz). Spike trains at the same contacts were analyzed with Poisson generalized linear models (GLM) with coded stimulus convolved with finite impulse-response (FIR) function which was estimated from the data, as well as spiking history (up to 80 ms duration, determined by minimum BIC criterion) as covariates. We treated the system as a time-invariant black box and the relationship between spiking activity (rates) and gamma-band LFP power was analyzed with Volterra kernel expansion (memory length of 200 ms) up to the second-order to examine potentially non-linear dependencies [3] (Figure ​(Figure1A).1A). Estimated discharge rates were used as the inputs and gamma LFP power was used as the outputs of the system. Weighted least-squares support-vector machines were utilized for estimating the regression coefficients [4], and the kernels were expanded using Laguerre orthogonal basis. First order kernel (linear filter) typically decreased toward zero within 150 ms lag time (Figure ​(Figure1A1A and ​and1B),1B), and 2nd -order kernels typically contain negative components, indicating non-linear suppressive effect on the gamma. (Figure ​(Figure1B).1B). A separate correlation analysis between spike count and gamma-band power within post-stimulus 200 ms shows strong positive correlation (Figure ​(Figure1C1C). Figure 1

Gamma band LFP in mouse barrel cortex is coupled to respiratory rhythm

The neocortex of mammalian brains shows prominent oscillatory activity in the local field potential (LFP) and EEG signal within a broad range of frequencies from slow (1-8 Hz) delta/theta band to gamma band (40-100 Hz) and higher frequencies [1]. Oscillations in different frequency bands seem to be linked. There is increasing evidence that amplitude modulations in the gamma frequency band are phase-locked to the delta/theta rhythm [2]. Several studies have linked theta-gamma phase-amplitude coupling to cognitive processes [3]. Here we report that in awake mice gamma band LFP power in the barrel cortex is phase-locked to the concurrent theta band LFP oscillation and that this theta band oscillation is strongly correlated to the respiratory rhythm. When the animals were briefly exposed to hypoxic air, the resulting frequency increases in the respiratory rhythm were paralleled in the delta/theta band LFP oscillations (Fig.1A-D). LFP oscillations in sub-bands of the broad (40 – 100 Hz) gamma frequency band were amplitude-modulated in phase with the breathing frequency (Fig. ​(Fig.1E).1E). After removal of the olfactory bulb the frequency profile of the phase-amplitude coupling was significantly changed. Particularly the respiration-locked amplitude modulation in the high gamma band (64-128 Hz), which was prominent in healthy control mice, shifted to lower frequencies in bulbectomized mice. Our findings imply that in mice respiratory activity directly modulates delta/theta band LFP oscillations through respiration-locked olfactory bulb activity and indirectly, through phase-amplitude coupling, gamma band power. Running Speed Alters the Frequency of Hippocampal Gamma Oscillations

Successful spatial navigation is thought to employ a combination of at least two strategies: the following of landmark cues and path integration. Path integration requires that the brain use the speed and direction of movement in a meaningful way to continuously compute the position of the animal. Indeed, the running speed of rats modulates both the firing rate of neurons and the spectral properties of low frequency, theta oscillations seen in the local field potential (LFP) of the hippocampus, a region important for spatial memory formation. Higher frequency, gamma-band LFP oscillations are usually associated with decision-making, increased attention, and improved reaction times. Here, we show that increased running speed is accompanied by large, systematic increases in the frequency of hippocampal CA1 network oscillations spanning the entire gamma range (30-120 Hz) and beyond. These speed-dependent changes in frequency are seen on both linear tracks and two-dimensional platforms, and are thus independent of the behavioral task. Synchrony between anatomically distant CA1 regions also shifts to higher gamma frequencies as running speed increases. The changes in frequency are strongly correlated with changes in the firing rates of individual interneurons, consistent with models of gamma generation. Our results suggest that as a rat runs faster, there are faster gamma frequency transitions between sequential place cell-assemblies. This may help to preserve the spatial specificity of place cells and spatial memories at vastly different running speeds.

Contrast sensitivity of MT receptive field centers and surrounds

Neurons throughout the visual system have receptive fields with both excitatory and suppressive components. The latter are responsible for a phenomenon known as surround suppression, in which responses decrease as a stimulus is extended beyond a certain size. Previous work has shown that surround suppression in the primary visual cortex depends strongly on stimulus contrast. Such complex center-surround interactions are thought to relate to a variety of functions, although little is known about how they affect responses in the extrastriate visual cortex. We have therefore examined the interaction of center and surround in the middle temporal (MT) area of the macaque (Macaca mulatta) extrastriate cortex by recording neuronal responses to stimuli of different sizes and contrasts. Our findings indicate that surround suppression in MT is highly contrast dependent, with the strongest suppression emerging unexpectedly at intermediate stimulus contrasts. These results can be explained by a simple model that takes into account the nonlinear contrast sensitivity of the neurons that provide input to MT. The model also provides a qualitative link to previous reports of a topographic organization of area MT based on clusters of neurons with differing surround suppression strength. We show that this organization can be detected in the gamma-band local field potentials (LFPs) and that the model parameters can predict the contrast sensitivity of these LFP responses. Overall our results show that surround suppression in area MT is far more common than previously suspected, highlighting the potential functional importance of the accumulation of nonlinearities along the dorsal visual pathway.

Attention induces dynamic changes in coherence between monkey area V1 and V4

Neurons receive a vast number of synaptic inputs at the same time. To successfully process a relevant piece of information, a neuron needs to select relevant, while suppressing irrelevant signals. In order to dynamically route a meaningful set of signals through processing pathways, neurons need to have a fast and flexible mechanism to change their effective connectivity. Here, we hypothesize that attention changes effective connectivity by modulating the synchrony between neuronal populations.We investigated whether attention dynamically changes effective connectivity between area V4 neurons and their V1 input. Single unit activity, multi unit activity and local field potentials (LFP) were recorded simultaneously from macaque monkeys’ areas V1 and V4. While maintaining their gaze on a fixation spot, the monkeys had to attend to one of two continuously morphing shapes and respond to reoccurrence of the initial sample shape in the attended stream. Size and position of the shapes were adjusted such that both fitted into a single V4 receptive field (RF) while covering two separate, non-overlapping V1 RFs.We found strong phase-coherence of gamma-band LFPs between the V1 populations responding to the attended stimulus and the V4 population. At the same time, V1 populations responding to the unattended stimulus showed only weak LFP phase-coherence with the V4 population. Furthermore, spike-triggered averages (STAs) of at least 40% of the recording pairs revealed that spikes recorded from area V4 were precisely timed with respect to the phase of the gamma oscillation in the upstream area V1. More importantly, almost all these cases showed an increased effect with attention. These results support the hypothesis that attention induces dynamic changes in the effective connectivity between two anatomically hard-wired neuronal populations by selectively modulating synchrony.

To successfully process a relevant piece of information, a neuron needs to select relevant, while suppressing irrelevant signals. Comparison of spatial integration and surround suppression characteristics in spiking activity and the local field potential in macaque V1

Neurons in primary visual cortex exhibit well documented centre-surround receptive field organization, whereby the centre is dominated by excitatory influences and the surround is generally dominated by inhibitory influences. These effects have largely been established by measuring the output of neurons, i.e. their spiking activity. How excitation and inhibition are reflected in the local field potential (LFP) is little understood. As this can bear on the interpretation of human fMRI BOLD data and on our understanding of the mechanisms of local field potential oscillations, we measured spatial integration and centre-surround properties in single- and multiunit recordings of V1 in the awake fixating macaque monkey, and compared these to spectral power in different frequency bands of simultaneously recorded LFPs. We quantified centre-surround organization by determining the size of the summation and suppression area in spiking activity as well as in different frequency bands of the LFP, with the main focus on the gamma band. Gratings extending beyond the summation area usually inhibited spiking activity while the LFP gamma-band activity increased monotonically for all grating sizes. This increase was maximal for stimuli infringing upon the near classical receptive field surround, where suppression started to dominate spiking activity. Thus, suppressive influences in primary cortex can be inferred from spiking activity, but they also seem to affect specific features of gamma-band LFP activity.
