Diffusion time-related structure-function coupling reveals differential association with inter-individual variations in body mass index

Body mass index (BMI) is an indicator of obesity, and recent neuroimaging studies have demonstrated that inter-individual variations in BMI are associated with altered brain structure and function. However, the mechanism underlying the alteration of structure-function correspondence according to BMI is under-investigated. In this study, we studied structural and functional connectivity derived from diffusion MRI tractography and inter-regional correlations of functional MRI time series, respectively. We combined the structural and functional connectivity information using the Riemannian optimization approach. First, the low-dimensional principal eigenvectors (i.e., gradients) of the structural connectivity were generated by applying diffusion map embedding with varying diffusion times. A transformation was identified so that the structural and functional embeddings share the same coordinate system, and subsequently, the functional connectivity matrix was simulated. Then, we generated gradients from the simulated functional connectivity matrix. We found the most apparent cortical hierarchical organization differentiating between low-level sensory and higher-order transmodal regions in the middle of the diffusion time, indicating that the hierarchical organization of the brain may reflect the intermediate mechanisms of mono- and polysynaptic communications. Associations between the functional gradients and BMI were strongest when the hierarchical structure was the most evident. Moreover, the gradient-BMI association map was related to the microstructural features, and the findings indicated that the BMI-related structure-function coupling was significantly associated with brain microstructure, particularly in higher-order transmodal areas. Finally, transcriptomic association analysis revealed the potential biological underpinnings specifying gene enrichment in the striatum, hypothalamus, and cortical cells. Our findings provide evidence that structure-function correspondence is strongly coupled with BMI when hierarchical organization is the most apparent and that the associations are related to the multiscale properties of the brain, leading to an advanced understanding of the neural mechanisms related to BMI.

Identifying subgroups of eating behavior traits unrelated to obesity using functional connectivity and feature representation learning.

Eating behavior is highly heterogeneous across individuals and cannot be fully explained using only the degree of obesity. We utilized unsupervised machine learning and functional connectivity measures to explore the heterogeneity of eating behaviors measured by a self-assessment instrument using 424 healthy adults (mean ± standard deviation [SD] age = 47.07 ± 18.89 years; 67% female). We generated low-dimensional representations of functional connectivity using resting-state functional magnetic resonance imaging and estimated latent features using the feature representation capabilities of an autoencoder by nonlinearly compressing the functional connectivity information. The clustering approaches applied to latent features identified three distinct subgroups. The subgroups exhibited different levels of hunger traits, while their body mass indices were comparable. The results were replicated in an independent dataset consisting of 212 participants (mean ± SD age = 38.97 ± 19.80 years; 35% female). The model interpretation technique of integrated gradients revealed that the between-group differences in the integrated gradient maps were associated with functional reorganization in heteromodal association and limbic cortices and reward-related subcortical structures such as the accumbens, amygdala, and caudate. The cognitive decoding analysis revealed that these systems are associated with reward- and emotion-related systems. Our findings provide insights into the macroscopic brain organization of eating behavior-related subgroups independent of obesity.

Extracting functional connectivity brain networks at the resting state from pulsed arterial spin labeling data

IntroductionFunctional connectivity in the brain is often studied with blood oxygenation level dependent (BOLD) resting state functional magnetic resonance imaging (rsfMRI), but the BOLD signal is several steps removed from neuronal activity. Arterial spin labeling (ASL), particularly pulsed ASL (PASL), has also the capacity to measure the blood-flow changes in response to activity. In this paper, we investigated the feasibility of extracting major brain networks from PASL data, in contrast with rsfMRI analsyis. Materials and methodsIn this retrospective study, we analyzed a cohort dataset that consists of 21 mild traumatic brain injury (mTBI) patients and 29 healthy controls, which was collected in a previous study. By extracting 10 major brain networks from the data of both PASL and rsfMRI, we contrasted their similarities and differences in the 10 networks extracted from both modalities. ResultsOur data demonstrated that PASL could be used to extract all 10 major brain networks. Eight out of 10 networks demonstrated over 60 ​% similarity to rsfMRI data. Meanwhile, there are similar but not identical changes in networks detected between mTBI patients and healthy controls with both modalities. Notably, the PASL-extracted default mode network (DMN), other than the rsfMRI-extracted DMN, includes some regions known to be associated with the DMN in other studies. It demonstrated that PASL data can be analyzed to identify resting state networks with reasonable reliability, even without rsfMRI data. ConclusionOur analysis provides an opportunity to extract functional connectivity information in heritage datasets in which ASL but not BOLD was collected.

Prediction of behavioral deficits in acute stroke from lesion and structural disconnection mapping.

Prediction of behavioral deficits in stroke relies on understanding the distribution of focal damage as well as the distribution of the underlying functional anatomy. Using structural or functional magnetic resonance imaging, previous studies investigated the predictive performance of imaging biomarkers for behavioral deficits in stroke patients. However, only focal lesion information or functional connectivity information alone was used in the modelling, with a small sample size and on a specific behavioral deficit domain. In this study, we investigated the prediction of behavioral deficits in acute stroke using both focal lesion patterns and structural disconnection mapping on a cohort of 551 ischemic stroke patients within one week post symptom onset. Five behavioral deficits domains, including motor, cognitive, visual, somatosensory and coordination deficits, were investigated. A probabilistic map of lesion-induced structural "disconnectome" map was created to estimate the degree of structural disconnection due to lesions. In the predictive modelling, both lesion volume and location and distant structural disconnections were included in combination with the clinical information. The results showed that improved prediction performance was achieved when considering both focal lesion patterns and global lesion-induced structural disconnections for all five behavioral deficits groups. Distinct lesion maps were obtained for each behavioral deficit, providing insights into neurobiological mechanisms of stroke functional impairment.

Spatial Optimization with Morphological Spatial Pattern Analysis for Green Space Conservation Planning

Conservation areas are essential for preserving green spaces and biological diversity. Although previous studies have demonstrated that spatial optimization techniques are effective for balancing the relationship between ecological importance and spatial pattern during conservation practices, the design of ecological corridors still requires an efficient, intelligent, and flexible workflow. In addition, functional connectivity information is usually unavailable or very difficult to obtain. To alleviate these problems, this paper has developed a new spatial optimization-based model that combines morphological spatial pattern analysis (MSPA) with ecological importance assessment. The consideration of MSPA can guarantee enough ecological corridors in the conservation plan, while the regions with higher ecological importance can be discovered through an ecological importance assessment. This method has been applied to the planning of conservation areas in a highly developed city. Several experiments have indicated that our proposed model could achieve much better performance than conventional models in terms of spatial pattern. Therefore, this new model is expected to assist decision processes during the planning and regulation of green spaces in fragmented urban ecosystems. Furthermore, it can be applied to ecological management and planning in many other aspects because the above-mentioned research gaps are not unique to only Asian or less-developed countries.

Brainprint based on functional connectivity and asymmetry indices of brain regions: A case study of biometric person identification with non‐expensive electroencephalogram headsets

AbstractBrain‐computer interface applications for biometric person identification have increased their interest in recent years since they are potentially more secure and more difficult to counterfeit than traditional biometric techniques. However, it is necessary to consider how brain waves are acquired for this purpose, not only in terms of efficiency but also of practical comfort for the user and the affordability degree of the biosignal acquisition device so that their everyday application can become a realistic possibility. In this context, this paper presents the capabilities of using a non‐expensive wireless electroencephalogram (EEG) device to extract spectral‐related and functional connectivity information of brain activity. The proposed method achieved a sufficient biometric identification with two datasets of 13 and 109 subjects when comparing the performance of a sizeable classification algorithm set. In addition, a novel feature in EEG biometric identification, called asymmetry index, is introduced here. Furthermore, this is the first study in this field to consider the effect of the time‐lapse between different recording sessions on the system's behaviour when using a low‐cost EEG device with identification accuracy rates of up to 100%.

A Spatiotemporal Graph Attention Network Based on Synchronization for Epileptic Seizure Prediction.

Accurate early prediction of epileptic seizures can provide timely treatment for patients. Previous studies have mainly focused on a single temporal or spatial dimension, making it difficult to take both relationships into account. Therefore, the effective properties of electroencephalograms (EEGs) may not be fully evaluated. To solve this problem, we propose a spatiotemporal graph attention network (STGAT) based on synchronization. The spatial and functional connectivity information between EEG channels was extracted by using the phase locking values (PLVs) first, which allowed multichannel EEG signals to be modeled as graph signals. Afterward, the STGAT model was used to dynamically learn the temporal correlation properties of EEG sequences and explore the spatial topological structure information of multiple channels. Experimental results demonstrated that the STGAT model was able to obtain spatiotemporal correlations and achieve good results on two benchmark datasets. The accuracy, specificity and sensitivity were 98.74%, 99.21% and 98.87%, respectively, on the CHB-MIT dataset. Moreover, all evaluation indices of the private dataset had reached more than 98.8%, with the area under the curve (AUC) reaching 99.96% . The proposed method is superior or comparable to the state-of-the-art models. Extensive experiments demonstrate that our end-to-end automatic seizure prediction model can be extended to design clinical assistant decision systems.

Coherence based graph convolution network for motor imagery-induced EEG after spinal cord injury.

Spinal cord injury (SCI) may lead to impaired motor function, autonomic nervous system dysfunction, and other dysfunctions. Brain-computer Interface (BCI) system based on motor imagery (MI) can provide more scientific and effective treatment solutions for SCI patients. According to the interaction between brain regions, a coherence-based graph convolutional network (C-GCN) method is proposed to extract the temporal-frequency-spatial features and functional connectivity information of EEG signals. The proposed algorithm constructs multi-channel EEG features based on coherence networks as graphical signals and then classifies MI tasks. Different from the traditional graphical convolutional neural network (GCN), the C-GCN method uses the coherence network of EEG signals to determine MI-related functional connections, which are used to represent the intrinsic connections between EEG channels in different rhythms and different MI tasks. EEG data of SCI patients and healthy subjects have been analyzed, where healthy subjects served as the control group. The experimental results show that the C-GCN method can achieve the best classification performance with certain reliability and stability, the highest classification accuracy is 96.85%. The proposed framework can provide an effective theoretical basis for the rehabilitation treatment of SCI patients.

Graph-generative neural network for EEG-based epileptic seizure detection via discovery of dynamic brain functional connectivity

Dynamic complexity in brain functional connectivity has hindered the effective use of signal processing or machine learning methods to diagnose neurological disorders such as epilepsy. This paper proposed a new graph-generative neural network (GGN) model for the dynamic discovery of brain functional connectivity via deep analysis of scalp electroencephalogram (EEG) signals recorded from various regions of a patient’s scalp. Brain functional connectivity graphs are generated for the extraction of spatial–temporal resolution of various onset epilepsy seizure patterns. Our supervised GGN model was substantiated by seizure detection and classification experiments. We train the GGN model using a clinically proven dataset of over 3047 epileptic seizure cases. The GGN model achieved a 91% accuracy in classifying seven types of epileptic seizure attacks, which outperformed the 65%, 74%, and 82% accuracy in using the convolutional neural network (CNN), graph neural networks (GNN), and transformer models, respectively. We present the GGN model architecture and operational steps to assist neuroscientists or brain specialists in using dynamic functional connectivity information to detect neurological disorders. Furthermore, we suggest to merge our spatial–temporal graph generator design in upgrading the conventional CNN and GNN models with dynamic convolutional kernels for accuracy enhancement.

Altered functional and directed connectivity in propofol-induced loss of consciousness: A source-space resting-state EEG study

ObjectiveGeneral anesthesia might disrupt neuronal network communications measured by functional connectivity (FC; undirected connectivity) and directional information flow (directed connectivity). We sought to characterize the state-dependent effects of propofol on cortico-cortical undirected and directed FC. MethodsWe collected 256-channel high-density EEGs from 14 patients undergoing surgery while awake (AWA) or in propofol-induced moderate sedation (SED) or loss of consciousness (LOC) states. Using source-space EEG, we estimated neuronal oscillatory activity for 68 cortical regions of interest. FC was analyzed using the weighted phase lag index. Directed connectivity was computed using directed phase transfer entropy (dPTE) as a measure of information flow in the bilateral prefrontal, frontal, parietal, and occipital areas. ResultsFC strength evidently reduced during LOC compared with those during the AWA and SED states. The dPTE analysis showed significant propofol-induced changes in directed connectivity. In the alpha band, the prefrontal-to-frontal information flow was significantly stronger in the AWA than in the SED (p = 0.033) and LOC states (p = 0.033). The parietal-to-frontal dPTE was significantly weaker during LOC than during the AWA (p = 0.033) and SED states (p = 0.007). Finally, a loss of occipital-to-frontal connectivity occurred during LOC but not the AWA state (p = 0.001). In the beta band, the dominant occipital-to-frontal direction of information flow in the AWA state was gradually converted to a frontal-to-occipital direction during LOC. ConclusionsPropofol-induced unconsciousness is marked by a decrease in FC and posterior-to-anterior (feedforward) directed connectivity, which may be useful as a measure to discriminate different conscious states caused by propofol administration. SignificanceThe study demonstrates that propofol produces state-dependent effects on cortico-cortical undirected and directed FC, supporting the idea that propofol induces loss of consciousness may through disrupting network interactions and cortical coordination.

Ultra-high-resolution time-frequency analysis of EEG to characterise brain functional connectivity with the application in Alzheimer’s disease

Objective. This study aims to explore the potential of high-resolution brain functional connectivity based on electroencephalogram, a non-invasive low-cost technique, to be translated into a long-overdue biomarker and a diagnostic method for Alzheimer’s disease (AD). Approach. The paper proposes a novel ultra-high-resolution time-frequency nonlinear cross-spectrum method to construct a promising biomarker of AD pathophysiology. Specifically, using the peak frequency estimated from a revised Hilbert–Huang transformation (RHHT) cross-spectrum as a biomarker, the support vector machine classifier is used to distinguish AD from healthy controls (HCs). Main results. With the combinations of the proposed biomarker and machine learning, we achieved a promising accuracy of 89%. The proposed method performs better than the wavelet cross-spectrum and other functional connectivity measures in the temporal or frequency domain, particularly in the Full, Delta and Alpha bands. Besides, a novel visualisation approach developed from topography is introduced to represent the brain functional connectivity, with which the difference between AD and HCs can be clearly displayed. The interconnections between posterior and other brain regions are obviously affected in AD. Significance. Those findings imply that the proposed RHHT approach could better track dynamic and nonlinear functional connectivity information, paving the way for the development of a novel diagnostic approach.

Abstract WMP64: Multimodal Bayesian Modeling Of Stroke Severity

Introduction: Understanding the relations between lesions and outcomes is a particularly promising avenue to support tailored stroke care. We here employed a novel Bayesian framework integrating lesion location and functional lesion connectivity, i.e., lesion network mapping data, aiming to augment the prediction of stroke severity and the interpretability of lesion-symptom associations in a multi-center cohort of acute ischemic stroke (AIS) patients. Methods: Analyses relied on 1,077 AIS patients of the MRI-GENIE study [age: 64.2(14.7), 38%women]. NIHSS-based stroke severity was modeled via Bayesian linear regression. Structural location and functional connectivity information was represented by ten unique patterns each. We compared the out-of-sample predictive performances of structural vs. structural-functional models in Bayesian model comparisons. In ancillary analyses, we extracted functional connectivity pattern relevant for stroke severity and repeated analyses after stratifying for lesion size (50% of patients with larger lesions). Models were generally adjusted for age, sex, comorbidities and total DWI lesion volume. Results: Model comparison indicated a benefit of the combined structural-functional stroke severity prediction model compared to the structural one (model weights: 82% vs. 18%). This benefit of the combined model became even more apparent in case of patients with large lesions (97% vs. 3%). Positive lesion connectivity to the bilateral ventral attention (VA) and somatomotor networks, as well as temporal default mode network (DMN) predicted higher severity, while positive connectivity to more medial parts of the DMN and the visual network predicted lower severity. Additionally, negative lesion connectivity to the bilateral DMN, somatomotor, dorsal attention, and control networks predicted higher, and negative connectivity to the right control and bilateral VA networks predicted lower severity. Conclusions: Model comparisons indicated a benefit of integrating structural and functional information for the prediction of stroke severity, especially in case of larger lesions. Lesion connectivity data can enhance our understanding of why specific lesion locations can lead to symptoms post-stroke.

Auto-weighted centralised multi-task learning via integrating functional and structural connectivity for subjective cognitive decline diagnosis.

Early diagnosis and intervention of mild cognitive impairment (MCI) and its early stage (i.e., subjective cognitive decline (SCD)) is able to delay or reverse the disease progression. However, discrimination between SCD, MCI and healthy subjects accurately remains challenging. This paper proposes an auto-weighted centralised multi-task (AWCMT) learning framework for differential diagnosis of SCD and MCI. AWCMT is based on structural and functional connectivity information inferred from magnetic resonance imaging (MRI). To be specific, we devise a novel multi-task learning algorithm to combine neuroimaging functional and structural connective information. We construct a functional brain network through a sparse and low-rank machine learning method, and also a structural brain network via fibre bundle tracking. Those two networks are constructed separately and independently. Multi-task learning is then used to identify features integration of functional and structural connectivity. Hence, we can learn each task's significance automatically in a balanced way. By combining the functional and structural information, the most informative features of SCD and MCI are obtained for diagnosis. The extensive experiments on the public and self-collected datasets demonstrate that the proposed algorithm obtains better performance in classifying SCD, MCI and healthy people than traditional algorithms. The newly proposed method has good interpretability as it is able to discover the most disease-related brain regions and their connectivity. The results agree well with current clinical findings and provide new insights into early AD detection based on the multi-modal neuroimaging technique.

Functional Brain Connectivity Hyper-Network Embedded with Structural Information for Epilepsy Diagnosis

Automatic diagnosis of brain diseases based on brain connectivity network (BCN) classification is one of the hot research fields in medical image analysis. The functional brain network reflects the brain functional activities and structural brain network reflects the neural connections of the main brain regions. It is of great significance to explore and explain the inner mechanism of the brain and to understand and treat brain diseases. In this paper, based on the graph structure characteristics of brain network, the fusion model of functional brain network and structural brain network is designed to classify the diagnosis of brain mental diseases. Specifically, the main work of this paper is to use the Laplacian graph embed the information of diffusion tensor imaging, which contains the characteristics of structural brain networks, into the functional brain network with hyper-order functional connectivity information built based on functional magnetic resonance data using the sparse representation method, to obtain brain network with both functional and structural characteristics. Projection of the brain network and the two original modes data to the kernel space respectively and then classified by the multi-task learning method. Experiments on the epilepsy dataset show that our method has better performance than several state-of-the-art methods. In addition, brain regions and connections that are highly correlated with disease revealed by our method are discussed.

Cigarette craving modulation is more feasible than resistance modulation for heavy cigarette smokers: empirical evidence from functional MRI data.

Modulation of cigarette craving and neuronal activations from nicotine-dependent cigarette smokers using real-time functional MRI (rtfMRI)-based neurofeedback (rtfMRI-NF) has been previously reported. The aim of this study was to evaluate the efficacy of rtfMRI-NF training in reducing cigarette cravings using fMRI data acquired before and after training. Treatment-seeking male heavy cigarette smokers (N = 14) were enrolled and randomly assigned to two conditions related to rtfMRI-NF training aiming at resisting the urge to smoke. In one condition, subjects underwent conventional rtfMRI-NF training using neuronal activity as the neurofeedback signal (activity-based) within regions-of-interest (ROIs) implicated in cigarette craving. In another condition, subjects underwent rtfMRI-NF training with additional functional connectivity information included in the neurofeedback signal (functional connectivity-added). Before and after rtfMRI-NF training at each of two visits, participants underwent two fMRI runs with cigarette smoking stimuli and were asked to crave or resist the urge to smoke without neurofeedback. Cigarette craving-related or resistance-related regions were identified using a general linear model followed by paired t-tests and were evaluated using regression analysis on the basis of neuronal activation and subjective craving scores (CRSs). Visual areas were mainly implicated in craving, whereas the superior frontal areas were associated with resistance. The degree of (a) CRS reduction and (b) the correlation between neuronal activation and CRSs were statistically significant (P < 0.05) in the functional connectivity-added neurofeedback group for craving-related ROIs. Our study demonstrated the feasibility of altering cigarette craving in craving-related ROIs but not in resistance-related ROIs via rtfMRI-NF training.

Morphometric and Functional Brain Connectivity Differentiates Chess Masters From Amateur Players.

A common task in brain image analysis includes diagnosis of a certain medical condition wherein groups of healthy controls and diseased subjects are analyzed and compared. On the other hand, for two groups of healthy participants with different proficiency in a certain skill, a distinctive analysis of the brain function remains a challenging problem. In this study, we develop new computational tools to explore the functional and anatomical differences that could exist between the brain of healthy individuals identified on the basis of different levels of task experience/proficiency. Toward this end, we look at a dataset of amateur and professional chess players, where we utilize resting-state functional magnetic resonance images to generate functional connectivity (FC) information. In addition, we utilize T1-weighted magnetic resonance imaging to estimate morphometric connectivity (MC) information. We combine functional and anatomical features into a new connectivity matrix, which we term as the functional morphometric similarity connectome (FMSC). Since, both the FC and MC information is susceptible to redundancy, the size of this information is reduced using statistical feature selection. We employ off-the-shelf machine learning classifier, support vector machine, for both single- and multi-modality classifications. From our experiments, we establish that the saliency and ventral attention network of the brain is functionally and anatomically different between two groups of healthy subjects (chess players). We argue that, since chess involves many aspects of higher order cognition such as systematic thinking and spatial reasoning and the identified network is task-positive to cognition tasks requiring a response, our results are valid and supporting the feasibility of the proposed computational pipeline. Moreover, we quantitatively validate an existing neuroscience hypothesis that learning a certain skill could cause a change in the brain (functional connectivity and anatomy) and this can be tested via our novel FMSC algorithm.

BCI Training Effects on Chronic Stroke Correlate with Functional Reorganization in Motor-Related Regions: A Concurrent EEG and fMRI Study

Brain–computer interface (BCI)-guided robot-assisted training strategy has been increasingly applied to stroke rehabilitation, while few studies have investigated the neuroplasticity change and functional reorganization after intervention from multimodality neuroimaging perspective. The present study aims to investigate the hemodynamic and electrophysical changes induced by BCI training using functional magnetic resonance imaging (fMRI) and electroencephalography (EEG) respectively, as well as the relationship between the neurological changes and motor function improvement. Fourteen chronic stroke subjects received 20 sessions of BCI-guided robot hand training. Simultaneous EEG and fMRI data were acquired before and immediately after the intervention. Seed-based functional connectivity for resting-state fMRI data and effective connectivity analysis for EEG were processed to reveal the neuroplasticity changes and interaction between different brain regions. Moreover, the relationship among motor function improvement, hemodynamic changes, and electrophysical changes derived from the two neuroimaging modalities was also investigated. This work suggested that (a) significant motor function improvement could be obtained after BCI training therapy, (b) training effect significantly correlated with functional connectivity change between ipsilesional M1 (iM1) and contralesional Brodmann area 6 (including premotor area (cPMA) and supplementary motor area (SMA)) derived from fMRI, (c) training effect significantly correlated with information flow change from cPMA to iM1 and strongly correlated with information flow change from SMA to iM1 derived from EEG, and (d) consistency of fMRI and EEG results illustrated by the correlation between functional connectivity change and information flow change. Our study showed changes in the brain after the BCI training therapy from chronic stroke survivors and provided a better understanding of neural mechanisms, especially the interaction among motor-related brain regions during stroke recovery. Besides, our finding demonstrated the feasibility and consistency of combining multiple neuroimaging modalities to investigate the neuroplasticity change.

Effect of APOE \u03b54 on multimodal brain connectomic traits: a persistent homology study

BackgroundAlthough genetic risk factors and network-level neuroimaging abnormalities have shown effects on cognitive performance and brain atrophy in Alzheimer’s disease (AD), little is understood about how apolipoprotein E (APOE) ε4 allele, the best-known genetic risk for AD, affect brain connectivity before the onset of symptomatic AD. This study aims to investigate APOE ε4 effects on brain connectivity from the perspective of multimodal connectome.ResultsHere, we propose a novel multimodal brain network modeling framework and a network quantification method based on persistent homology for identifying APOE ε4-related network differences. Specifically, we employ sparse representation to integrate multimodal brain network information derived from both the resting state functional magnetic resonance imaging (rs-fMRI) data and the diffusion-weighted magnetic resonance imaging (dw-MRI) data. Moreover, persistent homology is proposed to avoid the ad hoc selection of a specific regularization parameter and to capture valuable brain connectivity patterns from the topological perspective. The experimental results demonstrate that our method outperforms the competing methods, and reasonably yields connectomic patterns specific to APOE ε4 carriers and non-carriers.ConclusionsWe have proposed a multimodal framework that integrates structural and functional connectivity information for constructing a fused brain network with greater discriminative power. Using persistent homology to extract topological features from the fused brain network, our method can effectively identify APOE ε4-related brain connectomic biomarkers.

Iterative consensus spectral clustering improves detection of subject and group level brain functional modules

Specialized processing in the brain is performed by multiple groups of brain regions organized as functional modules. Although, in vivo studies of brain functional modules involve multiple functional Magnetic Resonance Imaging (fMRI) scans, the methods used to derive functional modules from functional networks of the brain ignore individual differences in the functional architecture and use incomplete functional connectivity information. To correct this, we propose an Iterative Consensus Spectral Clustering (ICSC) algorithm that detects the most representative modules from individual dense weighted connectivity matrices derived from multiple scans. The ICSC algorithm derives group-level modules from modules of multiple individuals by iteratively minimizing the consensus-cost between the two. We demonstrate that the ICSC algorithm can be used to derive biologically plausible group-level (for multiple subjects) and subject-level (for multiple subject scans) brain modules, using resting-state fMRI scans of 589 subjects from the Human Connectome Project. We employed a multipronged strategy to show the validity of the modularizations obtained from the ICSC algorithm. We show a heterogeneous variability in the modular structure across subjects where modules involved in visual and motor processing were highly stable across subjects. Conversely, we found a lower variability across scans of the same subject. The performance of our algorithm was compared with existing functional brain modularization methods and we show that our method detects group-level modules that are more representative of the modules of multiple individuals. Finally, the experiments on synthetic images quantitatively demonstrate that the ICSC algorithm detects group-level and subject-level modules accurately under varied conditions. Therefore, besides identifying functional modules for a population of subjects, the proposed method can be used for applications in personalized neuroscience. Functional connectivity of thalamus in children with primary nocturnal enuresis: results from a resting-state fMRI study.

Primary nocturnal enuresis (PNE) is characterized by a low cure rate and a high reoccurrence rate, since its underlying mechanism remains unclear. Based on the recent studies that thalamus plays an important role in waking up a sleeping person, here we further investigate the functional connectivity (FC) information between thalamus and other brain regions, in order to make better understanding of the PNE's pathogenesis. In this study, we enrolled 30 children diagnosed with PNE and 30 typically developing children that are age and sex matched, the thalamus-based FC estimates were extracted at the resting-state. Experiments showed that for children with PNE, there were four brain regions found with a reduced connection efficiency with thalamus, that were cerebellum posterior lobe, frontal lobe, parietal lobe and precentral gyrus. It can be concluded that these relevant regions might induce an arousal disorder, and therefore further lead to PNE. This finding also provides a new insight in the pathophysiology of PNE.
