Chromosomal abnormalities in primary and secondary amenorrhea

Background: Menstruation is an important physiological function of the female reproductive system. The absence of menstruation is called amenorrhea. Many genetic and nongenetic causes are responsible for primary or secondary amenorrhea. This study aimed to determine the types chromosomal abnormalities among patients with primary or secondary amenorrhea. Methods: It was a cross-sectional study conducted in the Department of Pathology, Bangabandhu Sheikh Mujib Medical University, Dhaka, from September 2019 to August 2021. A total of 115 women who had come for a karyotype test with complaints of pathological amenorrhea were purposively selected. One hundred five cases presented with primary amenorrhea, and 10 cases presented with secondary amenorrhea. Karyotype analysis in peripheral blood by G-banding was carried out using the standard method. Results: Among the 105 patients with primary amenorrhea, 53.3% of patients had a normal karyotype (46, XX), and 46.6% had chromosomal abnormalities (numerical or structural). Turner syndrome classic, 45, XO (16.2%) and 46, XY DSD (Disorders of Sexual Development) (11.4%) were the two most frequent chromosomal abnormalities found in the patients with primary amenorrhea. Among the 10 cases with secondary amenorrhea, seven patients had normal karyotype, and three patients had chromosomal abnormalities (2 had sex chromosomal abnormalities, and 1 had a structural abnormality of autosome). Conclusion: The study of chromosomal abnormalities will help in the early and accurate diagnosis of the underlying aetiology of primary or secondary amenorrhea. It will also help in the management and proper counselling of cases.

The profile of steroid hormones in human fetal and adult ovaries.

Reproduction in women is at risk due to exposure to chemicals that can disrupt the endocrine system during different windows of sensitivity throughout life. Steroid hormone levels are fundamental for the normal development and function of the human reproductive system, including the ovary. This study aims to elucidate steroidogenesis at different life-stages in human ovaries. We have developed a sensitive and specific LC-MS/MS method for 21 important steroid hormones and measured them at different life stages: in media from cultures of human fetal ovaries collected from elective terminations of normally progressing pregnancy and in media from adult ovaries from Caesarean section patients, and follicular fluid from women undergoing infertility treatment. Statistically significant differences in steroid hormone levels and their ratios were calculated with parametric tests. Principal component analysis (PCA) was applied to explore clustering of the ovarian-derived steroidogenic profiles. Comparison of the 21 steroid hormones revealed clear differences between the various ovarian-derived steroid profiles. Interestingly, we found biosynthesis of both canonical and "backdoor" pathway steroid hormones and corticosteroids in first and second trimester fetal and adult ovarian tissue cultures. 17α-estradiol, a less potent naturally occurring isomer of 17β-estradiol, was detected only in follicular fluid. PCA of the ovarian-derived profiles revealed clusters from: adult ovarian tissue cultures with relatively high levels of androgens; first trimester and second trimester fetal ovarian tissue cultures with relatively low estrogen levels; follicular fluid with the lowest androgens, but highest corticosteroid, progestogen and estradiol levels. Furthermore, ratios of specific steroid hormones showed higher estradiol/ testosterone and estrone/androstenedione (indicating higher CYP19A1 activity, p < 0.01) and higher 17-hydroxyprogesterone/progesterone and dehydroepiandrosterone /androstenedione (indicating higher CYP17A1 activity, p < 0.01) in fetal compared to adult ovarian tissue cultures. Human ovaries demonstrate de novo synthesis of non-canonical and "backdoor" pathway steroid hormones and corticosteroids. Elucidating the steroid profiles in human ovaries improves our understanding of physiological, life-stage dependent, steroidogenic capacity of ovaries and will inform mechanistic studies to identify endocrine disrupting chemicals that affect female reproduction.

Effects of bariatric surgery on sexual function and fertility: A narrative review.

Obesity has emerged as a prevalent global health concern, with its detrimental effects on the reproductive system and sexual function garnering increasing attention. Both men and women affected by obesity face a heightened risk of fertility challenges and sexual dysfunction. Although fertility and sexual function are distinct topics, they are intricately linked and mutually influential in both medical and societal contexts. Bariatric surgery (BS) has generated promising results in alleviating sexual dysfunction and enhancing fertility, results which are often gender specific. In men, improvements in sexual function can often be attributed to weight loss and subsequent optimizations in sex hormone levels. However, improving female sexual function may be related to a range of factors beyond weight loss. Bariatric procedures have shown limited benefits for male fertility; in fact, in some situations it can even be detrimental, leading to a decrease in sperm count and quality. Conversely, BS may positively impact female fertility, improving pregnancy and neonatal outcomes. Nevertheless, it is essential to consider the potential risks related to the adverse effects of malnutrition and rapid weight loss following BS, making it advisable to wait for 12-18 months before attempting pregnancy.

Therapeutic Potential of Hydrogen Sulfide in Reproductive System Disorders.

Hydrogen sulfide (H2S), previously regarded as a toxic exhaust and atmospheric pollutant, has emerged as the third gaseous signaling molecule following nitric oxide (NO) and carbon monoxide (CO). Recent research has revealed significant biological effects of H2S in a variety of systems, such as the nervous, cardiovascular, and digestive systems. Additionally, H2S has been found to impact reproductive system function and may have therapeutic implications for reproductive disorders. This paper explores the relationship between H2S and male reproductive disorders, specifically erectile dysfunction, prostate cancer, male infertility, and testicular damage. Additionally, it examines the impact of H2S regulation on the pathophysiology of the female reproductive system, including improvements in preterm birth, endometriosis, pre-eclampsia, fetal growth restriction, unexplained recurrent spontaneous abortion, placental oxidative damage, embryo implantation, recovery of myometrium post-delivery, and ovulation. The study delves into the regulatory functions of H2S within the reproductive systems of both genders, including its impact on the NO/cGMP pathway, the activation of K+ channels, and the relaxation mechanism of the spongy smooth muscle through the ROCK pathway, aiming to broaden the scope of potential therapeutic strategies for treating reproductive system disorders in clinical settings.

Comparative Investigation of Genital Self-image and Sexual Function in Women with and Without a History of Female Genital Cosmetic Procedures: A Cross-sectional Study.

Despite the increasing growth of female genital cosmetic procedures, the long-term effects of these procedures are not clearly understood. This study was conducted to compare the genital self-image and sexual function in women with and without female genital cosmetic procedures. This cross-sectional study was conducted on 315 participants (210 women without a history of genital cosmetic surgery and 105 women with it) in Alborz province, Iran, from early February 2023 to mid-May 2023. The sampling was done conveniently. Data collection instruments were Female Genital Self Image Scale and Female Sexual Function Index. Statistical analysis was done in SPSS 16 software using t-test, chi-square, and logistic regression, and P<0.05 was considered statistically significant. The use of laser to tighten the vagina with 77.77% and Perineoplasty with 29.2% were the main cosmetic procedures. The mean duration passed from the surgical procedures was 4.79±3.60 years, while it was 1.13±0.74 years for non-surgical procedures. Women with a history of genital procedures had a higher mean age (39.45±10.38, P=0.023). However, they were lower regarding the level of education (P<0.001), family income (P<0.001), and exercise (P<0.001). Also, they showed a higher number of pregnancies (P<0.001), deliveries (P<0.001), vaginal delivery (P<0.001), episiotomy (P<0.001), and neonates with a weight of ≥3.5 kg (P=0.002). In both groups, midwives and doctors were the most important sources of information about the appearance and function of reproductive system. However, the genital self-image and sexual function of the two groups did not differ significantly (P>0.05). No difference in sexual self-image and lack of difference in sexual function after cosmetic procedures show the need to pay attention to recommending and selecting these procedures. Public awareness about the diverse and natural forms of the female genitalia, education about the variety of the factors affecting sexual function, reduction of unnecessary interventions, increase in physiological births, retraining doctors and midwives, and multidimensional counseling can help to choose more appropriate candidates for cosmetic procedures.

Developmental programming of reproduction in the female animal

Successful reproduction is a cornerstone in food animal industry in order to sustain food production for human. Therefore, various methods focusing on genetics and postnatal environment have been identified and applied to improve fertility in livestock. Yet there is evidence indicating that environmental factors during prenatal and/or neonatal life can also impact the function of reproductive system and fertility in the animals during adulthood, which is called the developmental programming of reproduction. The current review summarizes data associated with the developmental origins of reproduction in the female animals. In this regard, this review focuses on the effect of plane of nutrition, maternal body condition, hypoxia, litter size, maternal age, parity, level of milk production and milk components, lactocrine signaling, stress, thermal stress, exposure to androgens, endocrine disrupting chemicals, mycotoxins and pollutants, affliction with infection and inflammation, and maternal gut microbiota during prenatal and neonatal periods on the neuroendocrine system, puberty, health of reproductive organs and fertility in the female offspring. It is noteworthy that these prenatal and neonatal factors do not always exert their effects on the reproductive performance of the female by compromising the development of organs directly related to reproductive function such as hypothalamus, pituitary, ovary, oviduct and uterus. Since they can impair the development of non-reproductive organs and systems modulating reproductive function as well (e.g., metabolic system and level of milk yield in dairy animals). Furthermore, when these factors affect the epigenetics of the offspring, their adverse effects will not be limited to one generation and can transfer transgenerationally. Hence, pinpointing the factors influencing developmental programming of reproduction and considering them in management of livestock operations could be a potential strategy to help improve fertility in food animals.

Regulation of insect behavior by non-coding RNAs.

The adaptation of insects to environments relies on a sophisticated set of behaviors controlled by molecular and physiological processes. Over the past several decades, accumulating studies have unveiled the roles of non-coding RNAs (ncRNAs) in regulating insect behaviors. ncRNAs assume particularly pivotal roles in the behavioral plasticity of insects by rapidly responding to environmental stimuli. ncRNAs also contribute to the maintenance of homeostasis of insects by fine-tuning the expression of target genes. However, a comprehensive review of ncRNAs' roles in regulating insect behaviors has yet to be conducted. Here, we present the recent progress in our understanding of how ncRNAs regulate various insect behaviors, including flight and movement, social behavior, reproduction, learning and memory, and feeding. We refine the intricate mechanisms by which ncRNAs modulate the function of neural, motor, reproductive, and other physiological systems, as well as gene expression in insects like fruit flies, social insects, locusts, and mosquitos. Furthermore, we discuss potential avenues for future studies in ncRNA-mediated insect behaviors.

Pengaruh Air Rebusan Daun Sirih terhadap Keputihan Fisiologis pada Wanita Usia Subur di Desa Baturono Kecamatan Salam Kabupaten Magelang

Vaginal discharge is one of the reproductive disorders often experienced by women in Indonesia caused by humid weather that affects the development of fungi. Continuous vaginal discharge affects the functioning of the female reproductive system. Overcoming vaginal discharge can be done with non-pharmacological therapy with red betel leaves. This study aims to determine the effect of betel leaf boiled water on physiological vaginal discharge in women of childbearing age in Baturono Village, Salam District, Magelang Regency. The research method used is quantitative research with a Quasy Experiment design using Two Groups Pre-Test and Post-Test with the control group. The population is 60 WUS, and the sample size is 30 respondents. The sampling technique uses Purposive Sampling. Statistical test using Paired T-Test. The result of this study was that the mean value 9pre-test0 given water treatment boiled red betel leaf was 14.80 and the mean value (post-test) was 5.27. So that the provision of red betel leaf boiled water has an effect on overcoming physiological vaginal discharge in Baturono Village based on the results of research p-value 0.000 &lt; 0.5. So it can be concluded that there is an effect of giving red betel leaf boiled water on physiological vaginal discharge in women of childbearing age in Baturono Village, Salam District, Magelang Regency with a p-value of 0.000 &lt; 0.5.

Effects of Pharmaceutical Substances with Obesogenic Activity on Male Reproductive Health.

Obesogens have been identified as a significant factor associated with increasing obesity rates, particularly in developed countries. Substances with obesogenic traits are prevalent in consumer products, including certain pharmaceuticals. Specific classes of pharmaceuticals have been recognized for their ability to induce weight gain, often accompanied by hormonal alterations that can adversely impact male fertility. Indeed, research has supplied evidence underscoring the crucial role of obesogens and therapeutic agents in the normal functioning of the male reproductive system. Notably, sperm count and various semen parameters have been closely linked to a range of environmental and nutritional factors, including chemicals and pharmacological agents exhibiting obesogenic properties. This review aimed to explore studies focused on analyzing male fertility parameters, delving into the intricacies of sperm quality, and elucidating the direct and adverse effects that pharmacological agents may have on these aspects.

D-hormone and its significance for function of prostate gland (literature review and personal observations).

Aim: The aim of this article is to present literature data and personal research of the role of D-hormoneon the functioning of the male reproductive system, and more specifically of the prostate gland, as well as the use of this vitamin D during the complex and independent treatment of benign prostatic hyperplasia in preclinical studies and clinical practice. Materials and Methods: The collection of relevant data were done using the scientific databases Pubmed, Google Scholar. A manual search on reproductive endocrinology and pharmacology sources were also conducted for related published studies . Selected keywords ("benign prostatic hyperplasia" OR "BPH") AND ("prostate") AND ("reproductive system and vitamin D") were used to collect data. The article also presents our personal data of preclinical studies and clinical data of the use of vitamin D as monotherapy and in the complex therapy of reproductive disorders. Conclusions: The effect of vitamin D on prostate volume and BPH has shown perspective results, therefore, it is proposed to conduct further studies on the role of vitamin D in the formation of BPH and reproductive disorders, their prevention and treatment. The use of vitamin D as monotherapy or in the form of pharmaceutical compositions and its inclusion in basic treatment regimens can increase the effectiveness of the prevention and correction of reproductopathies in the presence of or due to BPH and suggests the possibility of restoring the generative potential of individuals with BPH, both with and without D-hypovitaminosis.

Exploring the Diagnostic and Therapeutic Significance of Thyroid Hormones in Female Infertility: A Comprehensive Narrative Review

Thyroid Hormones (TH) are essential for the healthy functioning of the female reproductive system because they regulate ovarian, uterine, and placental tissue metabolism and development. Therefore, hypo-and hyperthyroidism may result in infertility in women. Previous studies have been conducted on women with thyroid dysfunction, including prospective and retrospective studies, in-vitro and in-vivo tests for hypo-and hyperthyroidism using ovarian, uterine, and placental cell culture, and experimental animal models. In order to better understand the physiology of the reproductive system and to develop more effective therapy methods for reproductive dysfunctions that result from thyroid dysfunctions, these studies sought to shed light on the mechanisms by which TH affect reproduction. This comprehensive narrative review investigates the diagnostic and therapeutic implications of TH in female infertility. By scrutinising existing literature, the study aims to elucidate the intricate relationship between thyroid function and reproductive health in women. Such insights are crucial for enhancing diagnostic accuracy and formulating effective therapeutic interventions to address thyroid-related factors influencing female infertility

Pathologies of the male reproductive system, having a different nature, and their impact on reproductive function

This work presents an overview of various pathologies that can affect the health of the male reproductive system and reproductive function. The authors consider such common conditions as varicocele, hypogonadism, epididymitis and prostatitis, and assess their impact on sperm quality, testosterone levels and the patency of the genitourinary tract. The article also discusses the methods of diagnosis and treatment of these pathologies, as well as their possible impact on the reproductive function of men. The study highlights the importance of timely diagnosis and treatment of these conditions in order to maintain the health of the male reproductive system and reproductive ability.

Penerapan Teams Game Tournament untuk Meningkatkan Motivasi Belajar pada Submateri Struktur dan Fungsi Reproduksi Manusia

This research aims to increase students' learning motivation on the sub-material of the structure and function of the human reproductive system, using the Teams Game Tournament learning model. This research is collaborative action research carried out at SMP Negeri 6 Pontianak from July to August 2023. The research was carried out in two learning cycles. Each cycle consists of four stages, namely planning, implementation, observation and reflection. The research subjects came from class IX F students with a total of 28 students consisting of 12 men and 16 women. The instruments used were learning implementation observation sheets and student learning motivation questionnaires. Data analysis uses descriptive statistical techniques. The learning process in cycle 1 was carried out with a percentage of 86% and increased in cycle 2 to 100% with very good criteria. The percentage value of student learning motivation in cycle 1 was 72.73%, and increased in cycle 2 to 80.20% with high criteria. Students with high learning motivation in cycle 1 were 55.85%, and increased in cycle 2 to 83.57%. It can be concluded that the application of the Teams Game Tournament model can increase student learning motivation in the sub-material of the structure and function of the human reproductive system.

Effect Of Zinc And Ingredients Of Protein Supplement On Sperm Parameters And Male Reproductive Hormones- An RCT Study

Objective: The purpose of this study was to assess the protective effects of zinc against injury to sperm parameters and male reproductive hormones brought on by toxic protein supplement constituents. Methods: 30 male Sprague Dawley rats were used in the study, of which 10 were in Group 1 (given standard laboratory food), 10 were in Group 2, fed standard laboratory food nutritional supplement powder and 10 were in Group 3, fed standard laboratory food, nutritional supplement powder and zinc. Semen Analysis of Male Sprague Dawley rats of all 3 groups was evaluated after 8 weeks. Sperm parameters and serum levels of Male Reproductive Hormones were measured at the beginning and the end of the study. Results: There was a significant decrease (P&lt;0.05) in Mean ± SEM of sperm count millions /ml, sperm motility (%), and normal sperm morphology (%) in Group 2 rats when compared with Group 1. Zinc supplement in Group 3 caused a significant increase (P&lt;0.05) in Mean ± SEM of sperm parameters including sperm count millions /ml, sperm motility (%) and normal sperm morphology (%) as compared to Group 2 rats. Mean ± SEM of serum level of testosterone Hormone, Follicle Stimulating Hormone(FSH), and Luteinizing Hormone(LH) of Group 2 rats showed a significant (P&lt;0.05) decrease when compared with Group 1 rats. However, the use of Zinc in Group 3 rats significantly (P&lt;0.05) increased serum Testosterone, FSH, and LH levels when compared to Group 2. No significant difference was noted between serum levels of Testosterone, in Group 1 and Group 3. Serum levels of FSH and LH in Group 3 rats (484.81+ 78.17 ng/ml) were significantly increased (P&lt;0.05) as compared to Group 1 rats. Conclusion: Regular addition of antioxidant Zinc with protein supplements has the potential to restore normal functions of the male reproductive system. Investigation of crocin’s protective effect on cyclophosphamide-induced hypothalamic-pituitary-gonadal axis defects in adult female rats

ABSTRACT Cyclophosphamide is a drug used in chemotherapy. However, it has side effects, including changes in reproductive system functioning. Some herbal compounds can reduce the harmful effects of cyclophosphamide. This study aims to investigate the protective role of crocin against changes caused by Cyclophosphamide in ovarian tissue through changes in the expression of genes involved in the hypothalamic-pituitary-gonadal axis. This experimental study was performed on 24 adult female Wistar rats. Mice were divided into four groups (normal saline, 30 mg/kg cyclophosphamide, 100 mg/kg crocin and 30 mg/kg cyclophosphamide, and 200 mg/kg crocin and 30 mg/kg cyclophosphamide). At the end of the treatment period, the hypothalamus and ovaries were also removed to evaluate ob-Rb, ob-Ra, and NPY genes expression using real-time PCR and histological changes in the ovaries. Data were analyzed by SPSS statistical software. The expression of genes, number of follicles, and follicle diameter significantly decreased in the cyclophosphamide-treated groups compared with the control group. In the crocin and cyclophosphamide-treated groups, drug-induced reproductive complications were mitigated. The current findings indicate that by increasing the expression of genes ob-Rb, ob-Ra, and NPY, crocin could modulate the harmful effects of cyclophosphamide.

Novel Advances in the Role of Selective Estrogen Receptor Modulators in Hormonal Replacement Therapy: A Paradigm Shift.

Estrogen is a key regulatory hormone in the functioning of a female reproductive system. Estrogen hormone regulates many complex physiological processes, which has its role in reproduction and skeletal and cardiovascular systems by acting on estrogen receptors alpha (ERα) and beta (ERβ), which are nuclear transcription factors. Selective estrogen receptor modulators (SERMs) are now being used to treat bone loss, breast carcinoma, and menopausal symptoms, metabolic neurodegenerativebecause of their characteristics that allow them to function as both estrogen agonists and antagonists,depending on the target tissue. First-generation SERMs, such as Tamoxifen, are used in the management protocol for breast cancer, which is estrogen receptor (ER-positive). Raloxifene is a second-generation SERM that is a valuable adjunct used to treat and prevent osteoporosis in postmenopausal women and prevent compression fractures of the vertebral column. Novel SERM molecules are on the horizon, proven more potent and efficacious in preventing and treating osteoporosis. These include Ospemifene, lasofoxifene, bazedoxifene and arzoxifene. The benefits of Raloxifene versus that of Bazedoxifene are under trial. Despite their therapeutic benefits and actions, these medications are not without adverse effects, such as thromboembolic disorders. Increased risk of uterine cancer has been linked to Tamoxifen. This article delves into the world of SERMs, including their development and discovery. The newer SERMs in late development, ospemifene, lasofoxifene, bazedoxifene, and arzoxifene, are described in detail.

Protective Effect of Low-Volume High-Intensity Interval Training on Aspirin-Induced Reproductive Impairments in Adult Male Wistar Rats.

Lifestyle factors such as sedentary behavior and consumption of certain medications can disturb the function of the male reproductive system. In the present study, we investigated the potential co-treatment effects of low-volume high-intensity interval training (HIIT) on markers of reproductive function in adult male Wistar rats under aspirin (ASA) treatment. Eighteen adult male Wistar rats were randomized into three groups: control (C), aspirin treatment (AT), and aspirin treatment + exercise (ATE). Animals in the AT and ATE groups received an oral subchronic dose of aspirin (12.5 mg/kg body mass). The exercise was performed three times per week for 6 weeks (4-6 reps of 10-s sprints). Serum testosterone level, sperm parameters (sperm count, viability, maturity, and DNA fragmentation), histomorphometric (Leydig cell, tubule diameter, thickness of tubular epithelium, and indices of spermatogenesis and spermiogenesis), and histochemical parameter (testicular fat density) were assessed. Results revealed that compared to the C group, ASA consumption led to a negative alteration in serum testosterone levels, sperm parameters, and histomorphometric and histochemical parameters (P < 0.05). However, there were no significant differences between the C and ATE groups in terms of serum testosterone level, number of Leydig cells, epididymal fat density, tubule diameter, epithelium height, immature-to-mature sperm ratio, and DNA breakage (P > 0.05). These findings suggest that ASA treatment is associated with deleterious changes in male reproductive parameters. However, low-volume HIIT may prevent ASA-induced male reproductive impairments and could be considered a potential prophylactic measure in subjects under ASA treatment.

Involvement of toxic metals and PCBs mixture in the thyroid and male reproductive toxicity: In silico toxicogenomic data mining

Toxicological research is mostly limited to considering the effects of a single substance, even though the real exposure of people is reflected in their daily exposure to many different chemical substances in low-doses. This in silico toxicogenomic study aims to provide evidence for the selected environmental (organo)metals (lead, cadmium, methyl mercury) + polychlorinated biphenyls mixture involvement in the possible alteration of thyroid, and male reproductive system function, and furthermore to predict the possible toxic mechanisms of the environmental cocktail. The Comparative Toxicogenomic Database, GeneMANIA online software, and ToppGene Suite portal were used as the main tools for toxicogenomic data mining and gene ontology analysis. The results show that 35 annotated common genes between selected chemicals and endocrine system diseases can interact on the co-expression level. Our study highlighted the disruption of the cytokines, the cell's response to oxidative stress, and the influence of the transcription factors as the potential core of toxicological mechanisms of the discussed mixture's effects. The connected toxicological effects of the tested mixture were abnormal sperm cells, a disrupted level of testosterone, and thyroid hormones. The core mechanisms of these effects were inflammation, oxidative stress, disruption of androgen receptor signaling, and the alteration of the FOXO3-Keap-1/NRF2-HMOX1-NQO1 pathway signaling most likely controlled by the co-expression of overlapped genes among used chemicals. This in silico research can be used as a potential core for the determination of biomarkers that can be monitored in future further in vitro and in vivo experiments.

Steroidogenic Effect of Luteinizing Hormone Receptor Agonists and Metformin in Male Rats with Androgenic Deficiency Caused by Diet-Induced Obesity

In obese men, along with metabolic disorders and insulin resistance, testosterone levels are decreased and reproductive system functions are impaired. One of the ways to correct them may be the use of agonists of luteinizing hormone receptor (LHR) and antidiabetic drugs, but the mechanisms of their effect on the hypothalamic-pituitary-gonadal axis have not been studied enough. The aim of the work was to study the effects of long-term MF therapy (5 weeks, 120 mg/kg) and five-day treatment with LHR agonists, such as human chorionic gonadotropin (hCG, 20 IU/rat/day, s.c.) and allosteric agonist TP03 (15 mg/kg/day, i.p.), on the blood testosterone levels and the expression of testicular and pituitary genes in male rats with long-term diet-induced obesity (DIO). TP03 moderately stimulated testosterone production in male rats with DIO without having an inhibitory effect on LHR expression in the testes and only weakly reducing the expression of the LH β-subunit gene in the pituitary gland. After a single administration into DIO-rats, the steroidogenic effect of TP03 was comparable to that in the control group, but after a five-day administration, it was significantly inferior to it. In DIO-rats, the steroidogenic effect of hCG after a single administration was lower than that in the control, but comparable to that after a five-day administration of hCG, and significantly exceeded the corresponding effects of TP03. Unlike TP03, hCG significantly reduced LHR expression in the testes and more pronouncedly inhibited LH expression in the pituitary gland. MF treatment restored the androgenic status without significantly affecting the expression of steroidogenesis genes in the testes. There was no increase in the steroidogenic effects of both LHR agonists in the MF-treated groups. The results obtained indicate the prospects for the use of TP03 and hCG to stimulate testicular steroidogenesis and the effectiveness of MF therapy to normalize testosterone production in DIO, which can be used to correct reproductive disorders in obesity. At the same time, in DIO, the combined use of MF and LHR agonists seems to be inappropriate.
