A convergent fragment coupling strategy to access quaternary stereogenic centers.

The formation of quaternary stereogenic centers via convergent fragment coupling is a longstanding challenge in organic synthesis. Here, we report a strategy for the formation of quaternary stereogenic centers in polycyclic systems based upon the semi-pinacol reaction. In the key transformation, two fragments of a similar size and complexity are joined by a 1,2-addition of an alkenyl lithium to an epoxy ketone, and the resulting epoxy silyl ether undergoes a semi-pinacol rearrangement catalyzed by N-(trimethylsilyl)bis(trifluoromethanesulfonyl)imide (TMSNTf2) or trimethylsilyl trifluoromethanesulfonate (TMSOTf). Polycyclic scaffolds were generated in high yields and the reaction conditions tolerated a variety of functional groups including esters, silyl ethers, enol ethers, and aryl triflates. This method provides a useful strategy for the synthesis of complex polycyclic natural product-like scaffolds with quaternary stereogenic centers from simplified fragments.

Carbene‐Catalyzed Enantioselective Aldol Reaction: Post‐Aldol Stereochemistry Control and Formation of Quaternary Stereogenic Centers

AbstractThe dominated approaches for asymmetric aldol reactions have primarily focused on the aldol carbon–carbon bond‐forming events. Here we postulate and develop a new catalytic strategy that seeks to modulate the reaction thermodynamics and control the product enantioselectivities via post‐aldol processes. Specifically, an NHC catalyst is used to activate a masked enolate substrate (vinyl carbonate) to promote the aldol reaction in a non‐enantioselective manner. This reversible aldol event is subsequently followed by an enantioselective acylative kinetic resolution that is mediated by the same (chiral) NHC catalyst without introducing any additional substance. This post‐aldol process takes care of the enantioselectivity issues and drives the otherwise reversible aldol reaction toward a complete conversion. The acylated aldol products bearing quaternary/tetrasubstituted carbon stereogenic centers are formed in good yields and high optical purities.

Enantioselective Construction of Quaternary Stereogenic Centers by the Addition of an Acyl Anion Equivalent to 1,3-Dienes.

We report the enantioselective formation of quaternary stereogenic centers by the intermolecular addition of malononitrile, an acyl anion equivalent, and related pronucleophiles to several 1,3-disubstituted acyclic 1,3-dienes in the presence of a Pd-PHOX catalyst. Products are obtained in up to 88% yield and 99:1 er and in most cases are formed as a single regioisomer. The products' malononitrile unit undergoes oxidative functionalization to afford β,γ-unsaturated carbonyls bearing internal olefins and α-quaternary stereogenic centers.

Stereoselective formation of quaternary stereogenic centers via alkylation of α-substituted malonate-imidazolidinones.

A new stereoselective alkylation methodology is presented for formation of chiral, nonracemic quaternary centers via a chiral auxiliary protocol involving α-alkylated malonate imidazolidinones. Based on two X-ray structures of quaternized products, the diastereoselectivity observed may be rationalized via a transition-state involving an s-transC-N conformation of the C-N bond of the auxiliary, with the metal cation (K(+)) chelated into the malonate six-membered hole as a Z-enolate. A deprotection protocol involving ethanethiolate exchange of the imide to the corresponding thioester, followed by a standard Fukuyama reduction and a borohydride reduction, furnishes α,α'-quaternized β-hydroxypropionates in high ee overall.

Ruthenium Hydride/Brønsted Acid-Catalyzed Tandem Isomerization/N-Acyliminium Cyclization Sequence for the Synthesis of Tetrahydro-β-carbolines

This paper describes an efficient tandem sequence for the synthesis of 1,2,3,4-tetrahydro-β-carbolines (THBCs) relying on a ruthenium hydride/Brønsted acid-catalyzed isomerization of allylic amides to N-acyliminium ion intermediates which are trapped by a tethered indole nucleophile. The methodology provides not only a convenient "aldehyde-free" alternative to the classical Pictet-Spengler reaction but also attractive possibilities for total synthesis, including rapid generation of molecular complexity and formation of quaternary stereogenic centers. TBHCs can also be accessed by harnessing the Suzuki cross-coupling reaction to the isomerization/N-acyliminium cyclization sequence. Finally, diastereo- and enantioselective versions of the title reaction have been examined using substrate control (with dr >15: 1) and asymmetric catalysis (ee up to 57%), respectively.

Formation of Quaternary Stereogenic Centers by Copper‐Catalyzed Asymmetric Conjugate Addition Reactions of Alkenylaluminums to Trisubstituted Enones

Alkenylaluminums undergo asymmetric copper-catalyzed conjugate addition (ACA) to β-substituted enones allowing the formation of stereogenic all-carbon quaternary centers. Phosphinamine-copper complexes proved to be particularly active and selective compared with phosphoramidite ligands. After extensive optimization, high enantioselectivities (up to 96% ee) were obtained for the addition of alkenylalanes to β-substituted enones. Two strategies for the generation of the requisite alkenylaluminums were explored allowing for the introduction of aryl- and alkyl-substituted alkenyl nucleophiles. Moreover, alkyl-substituted phosphinamine (SimplePhos) ligands were identified for the first time as highly efficient ligands for the Cu-catalyzed ACA.

Formation of Quaternary Stereogenic Centers by NHC–Cu‐Catalyzed Asymmetric Conjugate Addition Reactions with Grignard Reagents on Polyconjugated Cyclic Enones

The copper-catalyzed conjugate addition of various Grignard reagents to polyconjugated enones (dienone and enynone derivatives) is reported. The catalyst system, composed of copper triflate and an NHC ligand, led to the unusual selective formation of the 1,4-addition products. This reaction allows for the creation of all-carbon chiral quaternary centers with enantiomeric excesses up to 99%. The remaining unsaturation on the 1,4 adducts give access to valuable synthetic transformations.

Enantioselective Conjugated Addition of Vinylaluminums to Cyclic Enones

The development of transition-­metal-catalyzed enantioselective reactions allowing the formation of quaternary stereogenic centers is a challenging task. Herein, the authors report a highly enantioselective conjugated addition of Si-substituted vinylaluminum reagents to five- and six-membered cyclic β-substituted enones of type 1. A chiral bidentate NHC-copper complex has been identified to facilitate the desired transformation of 1 into 2 with good to excellent yields and enantioselectivities.

Formation of Quaternary Stereogenic Centers by Wagner-Meerwein Rearrangement – Synthesis of Optically Active Cyclopentadienyl Complexes from Borneol and Fenchol

The development of optically active cyclopentadienyl complexes as enantioselective catalysts calls for simple synthetic procedures for cyclopentadienes with optically active alkyl substituents. While exo-bornyl chloride and exo-fenchyl bromide do not react or exclusively eliminate hydrogen halide with cyclopentadienylmetal compounds in ether solvents or ammonia, they undergo Wagner- Meerwein rearrangement and substitution with cyclopentadienylmagnesium chloride in toluene. The bornyl cation yields racemic exo-bornylcyclopentadiene and partially racemized isocamphylcyclopentadiene, but for the fenchyl cation no racemization pathway is available, and the main diastereomer among the lithium salts of the ensuing substituted cyclopentadienes can be isolated in 95% diastereomeric purity by solvent extraction. This material with the IUPAC name lithium (2R)- 2,5,5-trimethylbicyclo[2.2.1]hept-2-ylcyclopentadienide carries an alkyl substituent having no trivial name so far. Exo-norbornylcyclopentadiene could be synthesized in high yield with a similar procedure. The same protocol works with 1-bromoadamantane. The novel alkylcyclopentadienes have been converted to ferrocenes and molybdenum complexes of the type [CpRMo(CO)3CH3]. (2R)- 2,5,5-Trimethylbicyclo[2.2.1]hept-2-ylcyclopentadiene with an optical purity of 78% ee (the optical purity of the starting material fenchol) was converted into an optically active titanocene dichloride and tested in the catalytic hydrogenation of 2-phenyl-1-butene. The hydrogenation product was obtained with 31% ee, which compares favorably with results obtained with other group 4 metallocene dichlorides with one optically active alkyl substituent on each ring ligand. Facile procedures for the synthesis of the starting compounds exo-bornyl chloride and exo-fenchyl bromide based on the tosylate method have been developed with a tosylate melt or with toluene serving as solvents.

Enantioselective Catalytic Formation of Quaternary Stereogenic Centers

AbstractEnantioselective catalytic formation of tertiary stereogenic centers has nowadays reached an impressive level of maturity, as is reflected in the large variety of available methods that afford high yields and high stereoselectivities. However, the development of stereoselective approaches for the formation of quaternary stereogenic centers still represents an enormous challenge for synthetic chemists. On the other hand, biologically active molecules containing quaternary stereogenic centers provide an incentive for the development of new, selective, and useful processes. Over the last few years, breakthrough work relating to the formation of fully substituted carbon centers has appeared in the literature. In this review we discuss recent highlights of this new direction in catalysis research: the formation of quaternary stereogenic centers by enantioselective catalytic methodologies.(© Wiley‐VCH Verlag GmbH & Co.
