Response of endophyte‐infected and endophyte‐free tall fescue plants to partial submergence at two developmental stages

AbstractTall fescue (Festuca arundinacea Schreb.) is a perennial C3 grass species usually associated with the endophyte fungus Epichloë coenophiala that enhances tolerance to biotic and abiotic stresses. Given that the proportion of infected tall fescue plants in the grasslands of the Flooding Pampa (Argentina) has been progressively increasing, it is proposed that endophytes confer tolerance to the stresses characteristic of that environment, especially flooding. Plants from a naturalized population and a commercial cultivar were grown, both with and without endophyte (wild type and AR584 respectively). The plants were subjected to two submergence treatments (i.e., control and partial submergence) in two development stages (5‐leaf stage and beginning of the elongation of internodes). Plant performance (aerial and root biomass, root aerenchyma formation, total root length and root diameter, proportion of reproductive tillers, number of panicles and seeds produced) and endophyte transmission to progeny were evaluated. Endophytes did not alter the ecological fitness of plants under partial submergence. Independently of the water condition, the symbiosis was linked to low biomass in plants from the naturalized population, while the opposite occurred in plants from the cultivar. Partial submergence did not affect the germination of the seeds produced, nor the transmission of the endophyte to the seedlings that originated from them. Our work suggests that the invasion ability of tall fescue in the Flooding Pampa grasslands is not related to an endophyte‐mediated improvement of plants to tolerate water excess.

Sub1 QTL confers submergence tolerance in rice through nitro-oxidative regulation and phytohormonal signaling

Constant change in global climate has become the most important limiting factor to crop productivity. Asymmetrical precipitations are causing recurrent flood events around the world. Submergence is one of the most detrimental abiotic stresses for sustainable rice production in the rainfed ecosystems of Southeast Asia. Therefore, the development of submergence-tolerant rice is an essential requirement to encounter food security. Submergence tolerance in rice is governed by the major quantitative trait locus (QTL) designated as Submergence1 (Sub1) near the centromere of chromosome 9. The introduction of the Sub1 in high-yielding rice varieties producing near-isogenic lines (NILs) has shown extreme submergence tolerance. The present study aimed to understand the responses of rice genotype IR64 and its Sub1 NIL IR64 Sub1 following one week of complete submergence treatment. Submergence imposed severe nitro-oxidative stress in both the rice genotypes, consequently disrupting the cellular redox homeostasis. In this study, IR64 exhibited higher NADPH oxidase activity accompanied by increased reactive oxygen species, reactive nitrogen species, and malondialdehyde buildups and cell death under submergence. Higher accumulations of 1-Aminocyclopropane-1-carboxylic acid, gibberellic acid, and Indole-3-acetic acid were also observed in IR64 which accelerated the plant growth and root cortical aerenchyma development following submergence. In contrast, IR64 Sub1 had enhanced submergence tolerance associated with an improved antioxidant defense system with sustainable morpho-physiological activities and restricted root aerenchyma formation. The comprehensive analyses of the responses of rice genotypes with contrasting submergence tolerance may demonstrate the intricacies of rice under complete submergence and may potentially contribute to improving stress resilience by advancing our understanding of the mechanisms of submergence tolerance in rice.

The uptake, transportation, and chemical speciation of Sb(III) and Sb(V) by wetland plants Arundinoideae (Phragmites australis) and Potamogetonaceae (Potamogeton crispus)

Antimony (Sb) is increasingly released and poses a risk to the environment and human health. Antimonite (Sb(III)) oxidation can decrease Sb toxicity, but the current knowledge regarding the effects of Sb(III) and antimonate (Sb(V)) exposure is limited to wetland plants, especially the Sb speciation in plants. In this study, Phragmites australis and Potamogeton crispus were exposed to 10 and 30 mg/L Sb(III) or Sb(V) for 20 days. The total concentration, subcellular distribution, and concentration in the iron plaque of Sb were determined. The Sb speciation in plants was analyzed by HPLC-ICP-MS. It illustrated that Sb(III) exposure led to more Sb accumulation in plants than Sb(V) treatments, with the highest Sb concentration of 405.35 and 3218 mg/kg in Phragmites australis and Potamogeton crispus, respectively. In the subcellular distribution of Sb, accumulation of Sb mainly occurred in cell walls and cell cytosol. In Phragmites australis, the transport factor in the Sb(V) treatments was about 3 times higher than the Sb(III) treatments, however, it was lower in the Sb(V) treatments than Sb(III) treatments for Potamogeton crispus. Sb(V) was detected in the plants of Sb(III) treatments with different Sb(V)-total Sb vitro (Phragmites australis: 34 % and, Potamogeton crispus: 15 %), moreover, Sb(V) was also detected in the nutrient solution of Sb(III) treatments. Antimony exposure caused a reduction of the iron plaque formation, at the same time, the root aerenchyma formation was disrupted, and this phenomenon is more pronounced in the Sb(III) treatments. Moreover, the iron plaque has a higher sorption potential to Sb under Sb(III) exposure than that under Sb(V) exposure. The results can fill the gap for antinomy speciation in wetland plants and expand the current knowledge regarding the Sb translocation in wetland systems.

Rice receptor kinase FLR7 regulates rhizosphere oxygen levels and enriches the dominant Anaeromyxobacter that improves submergence tolerance in rice.

Oxygen is one of the determinants of root microbiome formation. However, whether plants regulate rhizosphere oxygen levels to affect microbiota composition and the underlying molecular mechanisms remain elusive. The receptor-like kinase (RLK) family member FERONIA modulates the growth-defense tradeoff in Arabidopsis. Here, we established that rice FERONIA-like RLK 7 (FLR7) controls rhizosphere oxygen levels by methylene blue staining, oxygen flux, and potential measurements. The formation of oxygen-transporting aerenchyma in roots is negatively regulated by FLR7. We further characterized the root microbiota of 11 FLR mutants including flr7 and wild-type Nipponbare (Nip) grown in the field by 16S ribosomal RNA gene profiling and demonstrated that the 11 FLRs are involved in regulating rice root microbiome formation. The most abundant anaerobic-dependent genus Anaeromyxobacter in the Nip root microbiota was less abundant in the root microbiota of all these mutants, and this contributed the most to the community differences between most mutants and Nip. Metagenomic sequencing revealed that flr7 increases aerobic respiration and decreases anaerobic respiration in the root microbiome. Finally, we showed that a representative Anaeromyxobacter strain improved submergence tolerance in rice via FLR7. Collectively, our findings indicate that FLR7 mediates changes in rhizosphere oxygen levels and enriches the beneficial dominant genus Anaeromyxobacter and may provide insights for developing plant flood prevention strategies via the use of environment-specific functional soil microorganisms.

Strong plastic responses in aerenchyma formation in F1 hybrids of Imperata cylindrica under different soil moisture conditions.

Hybrids can express traits plastically, enabling them to occupy environments that differ from parental environments. However, there is insufficient evidence demonstrating how phenotypic plasticity in specific traits mediates hybrid performance. Two parental ecotypes of Imperata cylindrica produce F1 hybrids. The E-type in wet habitats has larger internal aerenchyma than the C-type in dry habitats. This study evaluated relationships between habitat utilisation, aerenchyma plasticity, and growth of I. cylindrica accessions. We hypothesize that plasticity in expressing parental traits explains hybrid establishment in habitats with various soil moisture conditions. Aerenchyma formation was examined in the leaf midribs, rhizomes and roots of two parental ecotypes and their F1 hybrids in their natural habitats. In common garden experiments, we examined plastic aerenchyma formation in leaf midribs, rhizomes and roots of natural and artificial F1 hybrids and parental ecotypes and quantified vegetative growth performance. In the natural habitats where soil moisture content varied widely, the F1 hybrids showed larger variation in aerenchyma formation in rhizomes than their parental ecotypes. In the common garden experiments, F1 hybrids showed high plasticity of aerenchyma formation in rhizomes, and their growth was similar to that of C-type and E-type under drained and flooded conditions, respectively. The results demonstrate that F1 hybrids of I. cylindrica exhibit plasticity in aerenchyma development in response to varying local soil moisture content. This characteristic allows the hybrids to thrive in diverse soil moisture conditions.

Peroxynitrite is essential for aerenchyma formation in rice roots under waterlogging conditions

In this study, we report that peroxynitrite is necessary for ethylene-mediated aerenchyma formation in rice roots under waterlogging conditions. Plants under waterlogging stress face anoxygenic conditions which reduce their metabolism and induce several adaptations. The formation of aerenchyma is of paramount importance for the survival of plants under waterlogging conditions. Though some studies have shown the involvement of ethylene in aerenchyma formation under waterlogging conditions, the implication of peroxynitrite (ONOO-) in such a developmental process remains elusive. Here, we report an increase in aerenchyma formation in rice roots exposed to waterlogging conditions under which the number of aerenchyma cells and their size was further enhanced in response to exogenous ethephon (a donor of ethylene) or SNP (a donor of nitric oxide) treatment. Application of epicatechin (a peroxynitrite scavenger) to waterlogged plants inhibited the aerenchyma formation, signifying that ONOO- might have a role in aerenchyma formation. Interestingly, epicatechin and ethephon co-treated waterlogged plants were unable to form aerenchyma, indicating the necessity of ONOO- in ethylene-mediated aerenchyma formation under waterlogging conditions. Taken together, our results highlight the role of ONOO- in ethylene-mediated aerenchyma formation in rice and could be used in the future to develop waterlogging stress-tolerant varieties of rice.

High-yielding nitrate transporter cultivars also mitigate methane and nitrous oxide emissions in paddy.

Development of high yield rice varieties is critical to ensuring global food security. However, the emission of greenhouse gases (GHG) such as Methane (CH4) and Nitrous oxide (N2O) from paddy fields threatens environmental sustainability. In this study, we selected overexpressed high-affinity nitrate transporters (NRT2.3 along with their partner protein NAR2.1) cultivars, which are effective nitrogen use efficient transgenic lines pOsNAR2.1: OsNAR2.1 (Ox2) and p35S:OsNRT2.3b (O8). We used high (270 kg N/ha) and low (90 kg N/ha) nitrogen (N) fertilizers in paddy fields to evaluate morphophysiological traits, including GHG emission. We found that Ox2 and O8 reduced CH4 emissions by 40% and 60%, respectively, compared to their wild type (WT). During growth stages, there was no consistent N2O discharge pattern between WT and transgenics (Ox2, O8) in low and high N application. However, total cumulative N2O in a cropping season reduced in O8 and increased in Ox2 cultivars, compared to WT. Root aerenchyma formation reduced by 30-60% in transgenic lines. Methanogens like mcrA in low and high N were also reduced by up to 50% from rhizosphere of Ox2 and O8. However, the nitrifying bacterial population such as nosZ reduced in both transgenics significantly, but nirK and nirS did not show a consistent variation. The high yield of transgenic rice with limited aerenchyma mitigates the discharge of CH4 and N2O by reducing root exudates that provide substrates for GHG. Our results improve understanding for breeders to serve the purpose of sustainable development.

Effects of different water conditions on the biomass, root morphology and aerenchyma formation in bermudagrass (Cynodon dactylon (L.) Pers)

BackgroundThe bermudagrass (Cynodon dactylon (L.) Pers) roots responded differently in terms of morphological and anatomical characteristics under diverse submergence conditions, and they developed aerenchyma under non-flooding condition. In order to understand these mechanisms, bermudagrass cuttings were used as experimental material to examine their biomass, root morphology, and aerenchyma formation under three different water treatments, including control (CK), shallow submergence (SS), and deep submergence (DS).ResultsThe total root length, root volume, root surface area, and biomass of bermudagrass were largest in CK, followed by SS and DS. However, the average root diameter was greater in each of DS and SS than that in CK. Root aerenchyma formation was observed in CK, and submergence boosted the aerenchyma formation and the root cavity rate. Furthermore, our study found that the process of aerenchyma formation began with the increase of cell volume and cell separation to form a narrow space, and these cells gradually died to form matured aerenchyma cavity, which belongs to schizo-lysigenous aerenchyma. Meanwhile, typical biomarkers of programmed cell death were also observed.ConclusionOverall, these results suggested that submergence inhibited the accumulation of biomass and root growth, but facilitated aerenchyma formation by increasing root diameter.

Modeling-based age-dependent analysis reveals the net patterns of ethylene-dependent and -independent aerenchyma formation in rice and maize roots

Plants require oxygen for the functioning of roots, and thus the establishment of a long-distance diffusion path from above-water tissues to the submerged roots is essential to survive flooding. Rice (Oryza sativa) constitutively forms aerenchyma (gas spaces) under aerobic conditions, and induces its formation in response to low-oxygen conditions. Constitutive aerenchyma formation in rice roots is regulated by the phytohormone auxin, whereas ethylene stimulates inducible aerenchyma formation. However, the net patterns of the ethylene-dependent and -independent (auxin-dependent) aerenchyma formation remain unclear. In the present study, we used a modeling approach to determine age-dependent aerenchyma formation in the wild-type rice and reduced culm number 1 mutant, in which ethylene production is reduced, to reveal the net patterns of ethylene-dependent and -independent aerenchyma formation. Subsequent comparison of age-dependent aerenchyma formation between rice and maize roots suggested that more rapid induction of ethylene-dependent aerenchyma formation and more aerenchyma in rice roots are essential to achieve efficient oxygen diffusion under low-oxygen conditions. Moreover, rice roots showed rapid increase in the expression levels of ethylene biosynthesis and responsive genes, suggesting that the local ethylene production at an early time point after root-cell emergence contributes to the rapid induction of the ethylene-dependent aerenchyma formation in rice. Plants growing in hypoxic conditions need to adapt to oxygen deficiency in substrate, including with the formation of lysigenous aerenchyma formation. Although lysigenous aerenchyma formation is well understood, its creation on different plant age is not well studied. In current article is described aerenchyma formation in roots of barley growing in hydroponic conditions up to 30 day after germination (DAG). It is shown that aerenchyma in primary roots developed on 7 DAG, and its volume in root cortex significantly enlarged, especially in basal root part, up to 30 DAG. H2O2 content increased from basal to apical part of roots and rose in 3-4 times on 30 DAG in all root parts. Guaiacol peroxidase (GP) demonstrated high activity on 27-30 DAG, and it coincided with aerenchymal lacunae origin. Superoxide dismutase activated earlier than GP and its activity may be involved in initial stage of aerenchyma formation. A clear correlation between H2O2 and antioxidative enzymes was not found, probably due to activation of the other processes of synthesis and degradation of H2O2.

A novel 3D X‐ray computed tomography (CT) method for spatio‐temporal evaluation of waterlogging‐induced aerenchyma formation in barley

AbstractIncreased precipitation during winter months in Western Europe is predicted from climate change. This is expected to increase the frequency of flooding events, with waterlogged conditions becoming more prevalent. A consequence of waterlogging is hypoxic conditions and dying back of roots, thereby negatively affecting plant nutrient uptake, growth, and development. Barley (Hordeum vulgare L.) forms lysigenous aerenchyma, which are air‐filled pockets within the root to facilitate O2 transport and support root respiration. Current methods for visualizing aerenchyma formation in roots are largely 2D and involve embedding a fixed root segment in resin, followed by destructive sectioning for microscopy. Here, we report the development of an X‐ray computed tomography (CT) acquisition and analytical method for spatio‐temporal identification and quantitation of aerenchyma in 2D and 3D in barley roots without the requirement for chemical fixation, resin embedding, followed by physical sectioning of the root. We demonstrated that the method we have developed can facilitate the identification and quantitation of aerenchyma in barley as early as 9 d post‐waterlogging. The method we have developed can be potentially applied to different plant species and has the potential to support the selection of breeding markers through X‐ray CT phenotyping of aerenchyma.

Aerenchyma Formation in Adventitious Roots of Tall Fescue and Cocksfoot under Waterlogged Conditions

The formation of aerenchyma in adventitious roots is one of the most crucial adaptive traits for waterlogging tolerance in plants. Pasture grasses, like other crops, can be affected by waterlogging, and there is scope to improve tolerance through breeding. In this study, two summer-active cocksfoot (Dactylis glomerata L.) cultivars, Lazuly and Porto, and two summer-active tall fescue (Lolium arundinaceum Schreb., syn. Festuca arundinacea Schreb.) cultivars, Hummer and Quantum II MaxP, were selected to investigate the effects of waterlogging on root growth and morphological change. Cultivars were subjected to four periods of waterlogging treatments (7, 14, 21 and 28 days), while comparable plants were kept under free drained control conditions. The experiment was arranged as a split–split plot design, with waterlogging treatments (waterlogged, control) considered as main plots, time periods (days of waterlogging) as subplots and cultivars as sub-subplots. Plants began to show signs of waterlogging stress 14–21 days after the onset of waterlogging treatments. There were no significant differences in shoot biomass between the waterlogged and control plants of any cultivar. However, waterlogging significantly reduced root dry matter in all cultivars, with greater reduction in cocksfoot (56%) than in tall fescue (38%). Waterlogging also led to increased adventitious root and aerenchyma formation in both species. Cocksfoot cultivars showed a greater increase in adventitious roots, while tall fescue cultivars had a greater proportion of aerenchyma. Both cultivars within each species showed similar responses to waterlogging treatments. However, an extended screening program is needed to identify whether there are varietal differences within species, which could be used to discover genes related to aerenchyma or adventitious root formation (waterlogging tolerance) for use in breeding programs.

Longitudinal Pattern of Aerenchyma Formation Using the Ti-Gompertz Model in Rice Adventitious Roots.

The longitudinal pattern of root aerenchyma formation of its relationship with the function of adventitious roots in rice remains unclear. In this study, the percentage of the aerenchyma area to the cross-sectional area (i.e., aerenchyma percentage) was fit with four non-linear models, namely, W0-Gompertz, Ti-Gompertz, logistic, and von Bertalanffy. Goodness-of-fit criteria such as the R2, the Akaike information criterion (AIC), and the Bayesian information criterion (BIC) were used to select the model. The bias of the parameters was evaluated using the difference between the ordinary least squares-based parameter estimates and the mean of 1,000 bootstrap-based parameter estimates and the symmetry of the distributions of these parameters. The results showed that the Ti-Gompertz model, which had a high goodness-of-fit with an R2 close to 1, lower AIC and BIC values, parameter estimates close to being unbiased, and good linear approximation, provided the best fit for the longitude pattern of rice aerenchyma formation with different root lengths among the competing models. Using the second- and third-order derivatives according to the distance from the root apex, the critical points of Ti-Gompertz were calculated. The rapid stage for aerenchyma formation was from the maximum acceleration point (1.38–1.76 cm from the root apex) to the maximum deceleration point (3.13–4.19 cm from the root apex). In this stage, the aerenchyma percentage increased by 5.3–15.7% per cm, suggesting that the cortical cells tended to die rapidly for the aerenchyma formation rather than for the respiration cost during this stage. Meanwhile, the volume of the aerenchyma of the entire roots could be computed using the integral function of the Ti-Gompertz model. We proposed that the longitudinal pattern of root aerenchyma formation modeled by the Ti-Gompertz model helped to deeply understand the relationship between the anatomical traits and physiological function in rice adventitious roots.

Aerenchyma formation in the root of leaf-vegetable sweet potato: Programmed cell death initiated by ethylene-mediated H2 O2 accumulation.

Sweet potato, commonly planted in Southeast Asia and South America with abundant rainfall, often suffers from waterlogging. The aerenchyma formation in roots is an effective way for plants to facilitate gas exchange. In the present study, tolerant and sensitive varieties, respectively, designated NC1 and C211, were evaluated under water oxygen content at 2.0mg·L-1 (hypoxia treatment) and 8.0mg·L-1 (control). The results showed that NC1 variety has a relatively higher root growth rate under low oxygen condition. In NC1 plants, aerenchyma was observed in the mid-section of the main adventitious root and spread to the proximal and distal ends, forming a complete channel in the cortex. However, in C211 plants, the aerenchyma occurred relatively later and could not turn into a whole channel. Ethylene synthesis-related (ACS1, ACS4, ACS5, etc.) and signal transduction-related (ETR1, ERS1, EIN2, etc.) genes were upregulated in the NC1 plants and led to changes in the reactive oxygen species-related genes (RBOHA, SOD, CAT, etc.) and enzyme activities. It was found that programmed cell death was induced by H2 O2 accumulation. A regulatory model of lysigenous aerenchyma formation in the root of sweet potato was constructed. Our study enriches the understanding of the mechanisms of the aerenchyma formation in plants.

Cation transporters in cell fate determination and plant adaptive responses to a low-oxygen environment.

Soil flooding creates low-oxygen environments in root zones and thus severely affects plant growth and productivity. Plants adapt to low-oxygen environments by a suite of orchestrated metabolic and anatomical alterations. Of these, formation of aerenchyma and development of adventitious roots are considered very critical to enable plant performance in waterlogged soils. Both traits have been firmly associated with stress-induced increases in ethylene levels in root tissues that operate upstream of signalling pathways. Recently, we used a bioinformatic approach to demonstrate that several Ca2+ and K+ -permeable channels from KCO, AKT, and TPC families could also operate in low oxygen sensing in Arabidopsis. Here we argue that low-oxygen-induced changes to cellular ion homeostasis and operation of membrane transporters may be critical for cell fate determination and formation of the lysigenous aerenchyma in plant roots and shaping the root architecture and adventitious root development in grasses. We summarize the existing evidence for a causal link between tissue-specific changes in oxygen concentration, intracellular Ca2+ and K+ homeostasis, and reactive oxygen species levels, and their role in conferring those two major traits enabling plant adaptation to a low-oxygen environment. We conclude that, for efficient operation, plants may rely on several complementary signalling pathway mechanisms that operate in concert and 'fine-tune' each other. A better understanding of this interaction may create additional and previously unexplored opportunities to crop breeders to improve cereal crop yield losses to soil flooding.

Lysigenous aerenchyma formation: responsiveness to waterlogging in oil palm roots

Oil palm (Elaeis guineensis Jacq.) responds to waterlogging stress by induction of lysigenous aerenchyma tissues, which facilitates the flow of oxygen through their root tissues for survival under waterlogged conditions. Thus, the morphological and genetic adaptation involved in lysigenous aerenchyma formation in the roots of the oil palm genotype Deli × Calabar under waterlogging stress was evaluated. This study found the highest number of dead cells after waterlogging stress for 2 d in the secondary root, while the percentage of root porosity was increased with increasing of time in both roots, especially at 1.0 - 2.0 cm from the root tip. This change in cell morphology implied the formation of lysigenous aerenchyma in oil palm roots under waterlogging stress. At the same time, most of the candidate genes involved in lysigenous aerenchyma formation revealed a higher mRNA expression after waterlogging stress for 3 d. Genes of ethylene synthesis group ACS3, ACO, and ACO1 were highly up-regulated in both types of roots, while XTH22, XTH23, and CEL12 in the cell wall modification group were more highly up-regulated in the primary roots than in the secondary roots. CML11, CAMTA4, TCTP, and CPI1 in a signaling group were up-regulated in the primary roots, but they were down-regulated in the secondary roots. NAC29, ERF1, ERF113, and HSFA2C in a transcription factor group were strongly up-regulated in the oil palm roots. However, there have been no previous reports on the expression of CAMTA4, bHLH79, and bHLH94 under waterlogging conditions. Our findings confirm gene expression during lysigenous aerenchyma development in oil palm roots under waterlogging. It can also be stated that primary roots are an important part of the adaptation mechanism of oil palm roots for survival under waterlogging stress. Furthermore, the molecular markers of all expressed genes will be developed and applied in our oil palm breeding project for selection of waterlogging tolerance.

Nitro-oxidative stress induces the formation of roots' cortical aerenchyma in rice under osmotic stress.

Drought stress induces the formation of cortical aerenchyma in roots, providing drought tolerance by reducing respiration. However, unrestricted aerenchyma formation impedes the radial transport of water through the root's central cylinder; thereby decreasing the water uptake under drought stress. Therefore, exploring the root architectural and anatomical alterations in rice under drought is essential for targeting crop improvement. Drought stress-induced accumulation of reactive oxygen species (ROS) plays a key role in the lysigenous aerenchyma development. However, the influence of nitric oxide (NO) and reactive nitrogen species (RNS) in the development of lysigenous aerenchyma under drought has never been studied in rice. The present study examined the effect of ROS and RNS, generated by progressive drought stress, on the lysigenous aerenchyma formation in the roots of contrasting rice genotypes of the Eastern Indo-Gangetic plains (EIGP). As expected, the PEG-induced drought stress stimulated the expression of NADPH oxidase (NOX), thereby promoting the ROS generation in roots of the rice seedlings. Excessive ROS and RNS accumulations in roots affected the membrane lipids, promoting the tissue-specific programmed cell death (PCD) in rice. The activation of the antioxidant defense system played a major role in the ROS and RNS detoxification, thereby restricting the root aerenchyma formation in rice under drought stress. The results also displayed that drought tolerance in rice is associated with the formation of the Casparian strip, which limits the apoplastic flow of water in the water-deficient roots. Overall, our study revealed the association of nitro-oxidative metabolism with PCD and lysigenous aerenchyma formation in the cortical cells of root under drought stress in rice.

Heterotrimeric G protein γ subunit DEP1 is involved in hydrogen peroxide signaling and promotes aerenchyma formation in rice roots

ABSTRACT Heterotrimeric G-protein α and β-subunits regulate H2O2-mediated aerenchyma formation. The rice G-protein γ-subunit, dense and erect panicle 1 (DEP1), is known to interact with the α-subunit and regulate nitrogen utilization and yield. However, it is unclear whether DEP1 regulates cell death for aerenchyma formation in rice roots. Using wild-type WYJ8 and its transgenic line WYJ8(DEP1), we confirmed that DEP1 is involved in H2O2-mediated aerenchyma formation. The rates of aerenchyma formation varied in different parts of the roots in both varieties, with the highest rate in the 4–7 cm segments, reaching a plateau in the 7–8 cm segments. Compared with WYJ8, the aerenchyma area and H2O2 content in WYJ8(DEP1) were increased by 55.98% and 53.37%, respectively; however, the responses of aerenchyma formation to exogenous H2O2 were basically the same in the two varieties. Diphenylene iodonium (DPI) treatment had no effect on H2O2 production and elimination processes in WYJ8, but significantly reduced the activity of the key enzyme that catalyzes H2O2 biosynthesis in WYJ8(DEP1). Importantly, exogenous H2O2 treatment did not offset the effect of the decrease in endogenous H2O2 level caused by DPI on aerenchyma formation. These results indicated that DEP1 enhanced H2O2 biosynthesis and promoted the cell death of the root cortex, thus contributing to aerenchyma development in WYJ8(DEP1).

Root length is proxy for high-throughput screening of waterlogging tolerance in Urochloa spp. grasses.

C4 perennial Urochloa spp. grasses are widely planted in extensive areas in the tropics. These areas are continuously facing waterlogging events, which limits plant growth and production. However, no commercial cultivar combining excellent waterlogging tolerance with superior biomass production and nutritional quality is available. The objective of this study was to identify root traits that can be used for selecting waterlogging tolerant species of Urochloa. Root respiration, root morphological, architectural and anatomical traits were evaluated in eight contrasting Urochloa spp. genotypes grown under aerated or deoxygenated stagnant solutions. Moreover, modelling of internal aeration was used to relate differences in root traits and root growth in waterlogged soils. Increased aerenchyma formation in roots, reduced stele area and development of a fully suberised exodermis are characteristics improving internal aeration of roots and therefore determining waterlogging tolerance in these C4 forage grasses. Waterlogging-tolerant genotypes had steeper root angles and greater root lengths than the waterlogging-sensitive genotypes. In stagnant conditions, waterlogging-tolerant genotypes had a greater proportion of aerenchyma and reduced stele area in root cross-sections, had deeper roots, steeper root angle and larger root biomass, which in turn, allowed for greater shoot biomass. Total root length had the strongest positive influence on shoot dry mass and can therefore be used as proxy for selecting waterlogging tolerant Urochloa genotypes.

Reactive oxygen species and reactive nitrogen species induce lysigenous aerenchyma formation through programmed cell death in rice roots under submergence

Nitric oxide (NO) plays a significant role in plants under different abiotic stress. However, the influence of NO and other reactive nitrogen species (RNS) in employing nitrosative stress in rice seedlings under submergence remains unexplored. Submergence also stimulates the development of lysigenous aerenchyma, facilitating oxygen supply to roots but the contribution of NO and RNS in aerenchyma formation under submergence is not known. Present study investigated the major components of the nitro-oxidative stress and their association with lysigenous aerenchyma development in the Sub1 near isogenic line of rice under submergence. Following submergence, Swarna showed increased NADPH oxidase (NOX) activity with excess reactive oxygen species (ROS) production in roots. Submergence also caused increased NO content and membrane lipid peroxidation in Swarna roots. Submergence-induced ROS and RNS accumulation in roots disturbed the redox homeostasis leading to the formation of lysigenous aerenchyma through programmed cell death (PCD). PCD was also accompanied by altered cytoplasmic streaming and DNA damage. In the present study Swarna Sub1 exhibited increased SOD, CAT, POX, APX, GR and GSNOR activity with subsequent detoxification of ROS and RNS; eventually decreasing the aerenchyma formation in root under submerged conditions. Overall, the study established ROS and RNS-mediated unique mechanism in lysigenous aerenchyma formation in rice roots under submergence.
