Revealing the Hypoglycemic Effect of Red Yeast Rice: Perspectives from the Inhibition of α-Glucosidase and the Anti-Glycation Capability by Ankaflavin and Monascin

Red yeast rice dietary supplements have been proven to ameliorate hyperglycemia, but the mechanism was unclear. In this work, ankaflavin (AK) and monascin (MS), as typical pigments derived from red yeast rice, were found to exert noteworthy inhibitory ability against α-glucosidase, with an IC50 of 126.5 ± 2.5 and 302.6 ± 2.5 μM, respectively, compared with acarbose (IC50 = 341.3 ± 13.6 μM). They also exhibited mixed-type inhibition of α-glucosidase in vitro and caused fluorescence quenching through the static-quenching process. Molecular-docking studies indicated that AK and MS bind to amino acid residues outside the catalytic center, which induces structural changes in the enzyme, thus influencing its catalytic activity. The anti-glycation ability of Monascus-fermented products was evaluated, and they exhibited a high inhibition rate of 87.1% in fluorescent advanced glycation end-product formation at a concentration of 0.2 mg mL−1, while aminoguanidine showed a rate of 75.7% at the same concentration. These results will be significant in broadening the application scope of Monascus pigments, especially AK and MS, in treating type 2 diabetes.

Musa balbisiana seed ameliorates methylglyoxal-induced oxidative stress and advanced glycation end-product formation via modulation of RAGE expression and SIRT1/PGC-1α/TFAM activation in HepG2 cells

Accumulating advanced glycation end-products (AGE) changes proteins' structural and functional properties and elicits many detrimental effects. This study focuses on identifying the phytometabolites present in the bioactive fraction (BF) of Musa balbisiana seed extract by GC-MS/MS. In addition, the potential of BF in mitigating methylglyoxal-induced oxidative stress and AGE formation in HepG2 cells was investigated using western blot and RT-qPCR techniques. The identification of non-targeted phytometabolites revealed the presence of 3-hydroxybenzoic acid, vanillic acid, protocatechuic acid, 4-coumaric acid, 3,4-dihydroxyphenylglycol, and catechin. BF protected fructose-induced protein glycation by reducing crosslink aggregate formation. Furthermore, BF pretreatment alleviated methylglyoxal-induced cytotoxicity in HepG2 cells by increasing the glyoxalase system and preventing AGE formation, thereby downregulating the expression of AGE receptor (RAGE). BF modulated the oxidative stress by upregulating PGC-1α expression, a regulator of mitochondrial biogenesis through activation of SIRT1 and TFAM. These findings suggest that Musa balbisiana seed can be a functional food for managing AGE and oxidative stress.

The Profile and Content of Polyphenolic Compounds and Antioxidant and Anti-Glycation Properties of Root Extracts of Selected Medicinal Herbs.

The objective of our study was to analyse the extracts from six medicinal herb roots (marshmallow, dandelion, liquorice, angelica, burdock, and comfrey) in terms of antioxidant capacity (ABTS, DPPH) and inhibition of advanced glycation end product (AGEs) formation. The quantification of phenolic acids and flavonoids was analysed using the UHPLC-DAD-MS method. Fifteen polyphenolic compounds were detected in the studied herbs. The higher number of polyphenols were found in marshmallows (ten polyphenols), while the lowest was in comfrey (five compounds). Liquorice root revealed the highest individual phenolic concentration (382µg/g dm) with the higher contribution of kaempferol-3-O-rutinoside. Comfrey root extract was characterised by the most abundant TPC (Total Phenolic Content) value (29.79mg GAE/ g dm). Burdock and comfrey showed the strongest anti-AGE activity studies with the BDA-GLU model. Burdock root was also characterised by the highest anti-AGE activity in the BSA-MGO model. The highest antioxidant capacity was determined by ABTS (72.12 µmol TE/g dw) and DPPH (143.01 µmol TE/g dw) assays for comfrey extract. The p-coumaric acid content was significantly correlated with anti-AGE activity determined by the BSA-MGO model. This research sheds new light on the bioactivity of root herbs, explaining the role of p-coumaric acid in preventing diabetes.

Dual Modification of Tau by Pseudophosphorylation and Glycation Does Not Enhance Amorphous Aggregation

Background/Aims: The neurofibrillary tangles consisting of Tau protein are an important pathology in Alzheimer’s disease. The paired helical filaments of Tau form most of the NFTs. These PHFs of Tau are found to carry numerous post-translational modifications, which stabilize them and aid in aggregation. The mechanistic function of Tau is to bind and stabilize the axonal microtubules. Hyperphosphorylation of Tau causes it to compromise its physiological function and accumulate in the neurons in the form of aggregates. Such residue-specific phosphorylation has been studied by employing Tau pseudophosphorylation mutants. But in addition to phosphorylation, several other modifications also aid in stabilizing the Tau PHF. Glycation is one such non-enzymatic PTM caused by sugars and their reactive intermediates. In this study, we employed the pseudophosphorylated Tau double mutants (262/404D, 262/396D, and 231/262) for studying their modification by methyl glyoxal, a reactive intermediate of glucose metabolism. Methods: We studied various biophysical properties like aggregation propensity, Advanced glycation end-product formation, and global conformation of the Tau with dual modifications. Our study includes the use of in vitro techniques e.g., ThS fluorescence assay, electron microscopy, CD spectroscopy, SDS-PAGE. Results: The overall result of the study suggest that the MG-induced Tau aggregation is influenced by the residue-specific Tau phosphorylation. Conclusion: In conclusion, the combinatorial effect of discreet PTMs on Tau function could lead to a better understanding of Tauopathy.

Potential inhibitory effect of highland barley protein hydrolysates on the formation of advanced glycation end-products (AGEs): A mechanism study

Advanced glycation end products (AGEs) can be caused during a glycoxidation reaction. This reaction is associated with complications of diabetes and the consequences of health problems. Therefore, we are exploring the prohibitory effect of highland barley protein hydrolysates (HBPHs) on AGE formation. Herein, first extracted the protein from highland barley with various pH conditions and then hydrolyzed using four different proteolytic enzymes (flavourzyme, trypsin, papain, pepsin) under different degrees of hydrolysis. We assessed three degrees of hydrolysates (lowest, middle, highest) of enzymes used to characterize the antioxidant activity and physicochemical properties. Among all the hydrolysates, flavourzyme-treated hydrolysates F-1, F-2, and F-3 indicated the high ability to scavenge DPPH (IC50 values of 0.97 %, 0.63 %, and 0.90 %), structural and functional properties. Finally, the inhibitory effect of the most active hydrolysates F-1, F-2, and F-3 against the AGEs formation was evaluated in multiple glucose-glycated bovine serum albumin (BSA) systems. Additionally, in a BSA system, F-3 exhibited the strong antiglycation activity, effectively suppressed the non-fluorescent AGE (CML), and the fructosamine level. Moreover, it decreased carbonyl compounds while also preventing the loss of thiol groups. Our results would be beneficial in the application of the food industry as a potential antiglycation agent for several chronic diseases.

Effects of caffeic acid on advanced glycation end product formation and emulsion properties in glycosylated perilla seed meal protein

Protein glycosylation is an effective way to enhance the properties of proteins in food systems. However, the negative effects of glycosylation, such as generation of advanced glycation end products (AGEs), have received less attention; these effects are closely related to the occurrence of various chronic metabolic diseases. In this study, a perilla seed meal protein (PSMP)-fructose/dextran (DEX)-caffeic acid (CA) ternary emulsifying system was constructed. Then, AGEs formation and emulsifying properties were characterized, and the optimal emulsifying system for β-carotene (BC) delivery was investigated. The resulting complex was examined using a high-performance liquid chromatography (HPLC)-ultraviolet (UV) detector, ultra-HPLC-tandem mass spectrometry (UHPLC-MS/MS) system, and nanoparticle size analyzer. The results indicated that CA conjugated with glycosylated PSMP significantly reduced the formation of fructosamine (13.5%), methylglyoxal (33.26%), Nε-carboxymethyl lysine, and Nε-carboxyethyl lysine (>80%). Compared with other conjugates, the PSMP-DEX-CA ternary conjugate showed the lowest AGEs content, smaller particle size, better emulsifying active index, lower polydispersity index, and acceptable ζ-potential. The emulsifying system constructed using abovementioned ternary conjugate revealed better UV and thermal stability in BC delivery, which slowed BC degradation. This study provides a novel approach for constructing a low AGEs emulsion system to deliver BC via PSMP with different modifications.

Inhibition and Mechanism of Protein Nonenzymatic Glycation by Lactobacillus fermentum.

Lactobacillus fermentum (L. fermentum) was first evaluated as a potential advanced glycation end-product (AGE) formation inhibitor by establishing a bovine serum albumin (BSA) + glucose (glu) glycation model in the present study. The results showed that the highest inhibition rates of pentosidine and total fluorescent AGEs by L. fermentum were approximately 51.67% and 77.22%, respectively, which were higher than that of aminoguanidine (AG). Mechanistic analysis showed that L. fermentum could capture methylglyoxal and glyoxal, inhibit carbonyl and sulfhydryl oxidation, reduce the binding of glucose and amino groups, increase total phenolic content and antioxidant activity, and release intracellular substances to scavenge free radicals; these abilities were the basis of the antiglycation mechanism of L. fermentum. In addition, L. fermentum significantly prevented conformational changes in proteins during glycation, reduced protein cross-linking by 35.67%, and protected the intrinsic fluorophore. Therefore, the inhibition of L. fermentum on glycation mainly occurs through antioxidation, the capture of dicarbonyl compounds, and the protection of the BSA structure. These findings collectively suggest that Lactobacillus is an inhibitor of protein glycation and AGE formation and has the potential for nutraceutical applications.

The role of vascular adhesion protein-1 in diabetes and diabetic complications.

Vascular adhesion protein-1 (VAP-1) plays a dual role with its adhesive and enzymatic properties, facilitating leukocyte migration to sites of inflammation and catalyzing the breakdown of primary amines into harmful by-products, which are linked to diabetic complications. Present in various tissues, VAP-1 also circulates in a soluble form in the bloodstream. Diabetes is associated with several complications such as cardiovascular disease, retinopathy, nephropathy, and neuropathy, significantly contributing to disability and mortality. These complications arise from hyperglycemia-induced oxidative stress, inflammation, and the formation of advanced glycation end-products (AGEs). Earlier research, including our own from the 1990s and early 2000s, has underscored the critical role of VAP-1 in these pathological processes, prompting extensive investigation into its contribution to diabetic complications. In this review, we examine the involvement of VAP-1 in diabetes and its complications, alongside its link to other conditions related to diabetes, such as cancer and metabolic dysfunction-associated fatty liver disease. We also explore the utility of soluble VAP-1 as a biomarker for diabetes, its complications, and other related conditions. Since the inhibition of VAP-1 to treat diabetic complications is a novel and promising treatment option, further studies are needed to translate the beneficial effect of VAP-1 inhibitors observed in animal studies to clinical trials recruiting human subjects. Besides, future studies should focus on using serum sVAP-1 levels for risk assessment in diabetic patients, identifying those who need intensive glycemic control, and determining the patient population that would benefit most from VAP-1 inhibitor therapies.

Serum carnosinase 1, an early indicator for incident microalbuminuria in type 1 diabetes

Abstract Aims Carnosinase (CN1) polymorphisms have been linked to diabetic kidney disease (DKD), as CN1 degrades dipeptides which scavenge oxidative metabolites and prevent the formation of advanced glycation end-products. In this work, we studied the association between serum CN1, the systemic redox status and long-term renal outcome in type 1 diabetes. Methods Serum CN1 was measured in a prospective type 1 diabetes cohort (n = 218) with a 16-year follow-up. A total of 218 patients treated at the Diabetes Outpatient Clinic of the Weezenlanden Hospital (nowadays Isala Hospital, Zwolle, The Netherlands) were included in this analysis. We assessed whether serum CN1 was associated with renal function and development of DKD as well as other diabetic complications. Results At baseline, age, systemic redox status and N-terminal pro brain-natriuretic peptide (NT-proBNP) were associated with serum CN1 concentration (p < 0.05). During follow-up, CN1 concentration in the middle tertile was associated with less incident microalbuminuria (odds ratio = 0.194, 95% C.I.: 0.049—0.772, p = 0.02) after adjustment for age, systemic redox status, NT-proBNP and sex. Discussion Serum CN1 could predict incident microalbuminuria and may be used as a novel parameter to identify patients at risk for DKD.

Anti-glycation properties of 05 spices and culinary herbs used in Benin

Diabetes mellitus is a major public health problem characterized by chronic hyperglycemia leading to several other pathologies. It causes oxidative stress, resulting in the overproduction of superoxide radicals and protein glycation, ultimately leading to the formation of Advanced Glycation Endproducts (AGEs). It has been shown elsewhere that spices and herbs commonly used in the culinary arts inhibit glycation and AGE formation. The aim of this study is to evaluate the inhibitory effect of five aromatic herbs and spices commonly used in Benin on protein glycation and AGE formation. Total flavonoids content and polyphenolic compounds were determined with respectively the aluminium chloride method and the Folin-Ciocalteu method. For glycation testing, serum pools from diabetics and non-diabetics were used. Serum fructosamines were measured using the nitro blue tetrazolium colorimetric test and AGEs were detected using a fluorescence method. All five herbs and spices had shown notable ability to inhibit fructosamine and AGE formation. L. nobilis was very rich in flavonoids and exhibited the strongest inhibitory activity against glycation and AGE formation. The results of this study suggested that the investigated five spices and culinary herbs have interesting properties to take in account in the therapy of diabetes and its related complications.

Aldose Reductase as a Key Target in the Prevention and Treatment of Diabetic Retinopathy: A Comprehensive Review.

The escalating global prevalence of diabetes mellitus (DM) over the past two decades has led to a persistent high incidence of diabetic retinopathy (DR), necessitating screening for early symptoms and proper treatment. Effective management of DR aims to decrease vision impairment by controlling modifiable risk factors including hypertension, obesity, and dyslipidemia. Moreover, systemic medications and plant-based therapy show promise in advancing DR treatment. One of the key mechanisms related to DR pathogenesis is the polyol pathway, through which aldose reductase (AR) catalyzes the conversion of glucose to sorbitol within various tissues, including the retina, lens, ciliary body and iris. Elevated glucose levels activate AR, leading to osmotic stress, advanced glycation end-product formation, and oxidative damage. This further implies chronic inflammation, vascular permeability, and angiogenesis. Our comprehensive narrative review describes the therapeutic potential of aldose reductase inhibitors in treating DR, where both synthetic and natural inhibitors have been studied in recent decades. Our synthesis aims to guide future research and clinical interventions in DR management.

Inhibitory effects and mechanisms of phenolic compounds in rapeseed oil on advanced glycation end product formation in chemical and cellular models in vitro

Sinapic acid (SA), canolol (CAO) and canolol dimer (CAO dimer) are the main phenolic compounds in rapeseed oil. However, their possible efficacy against glycation remains unclear. This study aims to explore the impacts of these substances on the formation of advanced glycation end products (AGEs) based on chemical and cellular models in vitro. Based on fluorescence spectroscopy results, three chemical models of BSA-fructose, BSA-methylglyoxal (MGO), and arginine (Arg)-MGO showed that SA/CAO/CAO dimer could effectively reduce AGE formation but with different abilities. After SA/CAO/CAO dimer incubation, effective protection against BSA protein glycation was observed and three different MGO adducts were formed. In MGO-induced HUVEC cell models, only CAO and CAO dimer significantly inhibited oxidative stress and cell apoptosis, accompanied by the regulation of the Nrf2-HO-1 pathway. During the inhibition, 20 and 12 lipid mediators were reversed in the CAO and CAO dimer groups compared to the MGO group.

Hypolipidemic and Hypoglycemic Effects of Silybum marianum (L.) Gaertn.(Milk Thistle) Ethanol Seed Extract in Streptozotocin-induced Diabetes in Rats

Background: Metabolic syndrome (MS) is considered a challenging health problem worldwide. However, the search for alternative treatments to alleviate MS is lacking. Purpose: This study defines Silybum marianum (milk thistle) ethanol seed extract (SMESE) in terms of polyphenol and flavonoid content, antiglycation effect, α-amylase activity, and hypolipidemic and hypoglycemic effects in streptozotocin (STZ)-induced diabetic rats. Materials and Methods: S. marianum seeds were collected, ground, and extracted with 80% ethanol. The α-amylase inhibitory activities of 10, 30, and 100 µg/mL SMESE were evaluated using spectrophotometric methods. The inhibitory activity of SMESE against advanced glycation end-product formation was evaluated using fluorescence spectrophotometry. In vivo, 48 Wistar male albino rats were used. Eight animals served as the normal control (group 1), and 40 rats were injected with 60 mg/kg STZ to induce diabetes. Diabetic rats were divided into five groups: diabetic control (group 2) and positive control that received the hypoglycemic reference drug metformin at 100 mg/kg (group 3). Groups 4, 5, and 6 were administered the seed extract orally at 30, 100, and 300 mg/kg, respectively. Body weight, serum glucose, lipid profile, and malondialdehyde (MDA) levels were measured weekly for five weeks after SMESE administration. Results: SMESE administration inhibited BSA glycation and α-amylase activity. In vivo, SMESE administration increased body weight (BW), decreased serum glucose levels, and reduced triglyceride, total cholesterol, low-density lipoprotein, very low-density lipoprotein, and MDA levels but increased high-density lipoprotein levels. Conclusion: Data indicate the beneficial hypoglycemic and antihyperlipidemic effects of SMESE, suggesting its potential use in controlling lipid metabolism.

The Protective Effect of Ethyl Acetate Fraction and Bioactive Compounds of Mimosa pudica L. Leaves in Methylglyoxal-induced Glucotoxicity

Mimosa pudica L. has been used as a traditional medicine to treat leprosy, dysentery, burning sensation, asthma, inflammation, fatigue, and blood diseases. The purpose of this study was to evaluate the ethyl acetate (EtOAc) fraction and two compounds protocatechuic acid and syringic acid of Mimosa pudica L. against methylglyoxal (MGO)- induced glucotoxicity. The inhibitory effects of EtOAc fraction and two compounds of Mimosa pudica L. on glucotoxicity in Human umbilical vein endothelial cells (HUVECs) and inhibit Advanced glycation end products (AGEs) formation and also breaking effects on MGO-AGEs were measured. Our results demonstrated that EtOAc fraction and two bioactive compounds protocatechuic acid and syringic acid of Mimosa pudica L. leaves inhibit MGO-induced AGEs formation, and break down MGO-AGEs, resulting in the reduction of glucotoxicity. EtOAc fraction and two bioactive compounds protocatechuic acid and syringic acid are promised in effective in preventing or treating MGO-induced endothelial dysfunction.

Impact of Pyridoxine Supplement on Oxidative Stress in Type 2 Diabetic Patients

Background: The physiologically active form of vitamin B6 is pyridoxal 5-phosphate (PLP), which functions as a coenzyme in 150 enzymatic processes such amino acid, carbohydrate, and lipid metabolism and is essential for the production and/or breakdown of neurotransmitters. . It also acts as an antioxidant by quenching Reactive Oxygen Species (ROS) and counteracting the formation of Advanced Glycation End-Product (AGEs). PLP is recycled by mammals from B6 vitamins found in diet and has been linked to a number of clinically significant disorders. This study aim was to evaluate the impact of pyridoxine supplementation on oxidative stress status in type 2 diabetic patients. Method: This prospective controlled randomized open-labeled study of newly diagnosed T2DM patients. The study was conducted from November 2022 to February 2023 at the Mesan Center for Diabetes and Endocrinology under the supervision of a specialist endocrinologist. The total number of participants whose data were collected in this study was one hundred and eight participants, eighty-eight patients newly diagnosed with type 2 diabetes were included in the study, and twenty of the participants were healthy subjects. The patients were allocated into three groups: Group 1: Control group, 20 T2DM patients were treated with non-pharmacological therapy (lifestyle modification) for one month, Group 2: 34 T2DM patients treated with metformin 500 mg/day in addition to non-pharmacological therapy (lifestyle modification) for one month, Group 3: 34 T2DM patients treated with metformin 500 mg/day plus vitamin B6 300 mg/day in addition to non-pharmacological therapy (lifestyle modification) for one month, in addition to Healthy subjects: 20 subjects were taken to compare the study parameters between type 2 diabetic patients and healthy persons at baseline. Measurement of Vitamin B6 (pyridoxine) blood level (PLP), body mass index (BMI) and Serum malondialdehyde level (MDA) was done in this study. Results: The results of the study showed that the use of pyridoxine supplementation plus metformin decreased the level of MDA. There was significant decrease in (MDA) level (P<0.01) pretreatment when compared to after treatment (3.85 μmole/L vs 1.66 μmole/L) respectively. Conclusion: Restoration of vitamin B6 level can leads to a decreased blood level of MDA in diabetic patients.

Effect of brewer's spent grain melanoidins on Maillard reaction products during storage of whey protein model systems

Maillard reaction readily takes place in dairy products because of the association between thermal treatments, extended storage and the matrix composition. Along with the impairment of protein digestion, the formation of glycation and α-dicarbonyl compounds is a concern for quality attributes of whey proteins when used as ingredients. In this paper, we outline the capacity of brewer's spent grain melanoidins in reducing the accumulation of α-dicarbonyl compounds, thus controlling the formation of dietary advanced glycation end-products in accelerated shelf life at 35 °C. Results revealed that brewer's spent grain melanoidins targeted methylglyoxal and glyoxal reactivity leading to the reduction of N-ε-carboxymethyllysine and methylglyoxal-hydroimidazolone up to 27 and 60%, respectively. We here describe that the presence of melanoidins is instrumental in limiting the undesired effects of α-dicarbonyl compounds on whey proteins.

N-acetyl-L-hydroxyproline - A potent skin anti-ageing active preventing advanced glycation end-product formation invitro and exvivo.

Advanced glycation end-products (AGEs) represent a large group of compounds generated by a non-enzymatic reaction between reducing sugars and amino groups. The formation and accumulation of AGEs in the skin lead to protein crosslinking, dermal stiffening and yellowing, which ultimately contribute to cutaneous ageing. Amino acids have been described to exhibit anti-glycation effects. The objective of this study was to understand the inhibitory role of the amino acid derivative N-acetyl-L-hydroxyproline (NAHP) as an anti-glycation active for human skin. A cell-free assay investigating the inhibition of glycation of serum albumin by NAHP was used to determine the capability of NAHP to decrease AGE formation. Also, by assessing the amount of the AGE N-(carboxymethyl)lysine (CML) the anti-glycation abilities of NAHP were investigated utilizing dot blot analysis. The improvement of cell-matrix interaction by NAHP was determined invitro using a glycated fibroblast-populated collagen lattice (FPCL) dermis model. In skin biopsies, AGE autofluorescence was determined after treatment with NAHP and/or glucose exvivo. NAHP significantly and dose-dependently inhibited levels of AGEs, which were induced by the glycation of a protein solution. This decrease could be visualized by showing that the brownish appearance as well as the AGE-specific fluorescence of glucose-treated samples were reduced after the application of increasing amounts of NAHP. Also, CML formation was dose-dependently inhibited by NAHP. In FPCLs, the contractile capacity of fibroblasts was significantly disturbed after glycation. This could be prevented by the addition of NAHP. Compared to glyoxal-treated samples, the co-application of NAHP significantly decreased the diameter as well as the weight of glycated FPCLs. Exvivo application of glucose to skin explants showed a higher AGE fluorescence signal compared to control explants. Co-treatment with NAHP and glucose decreased the level of AGE fluorescence in comparison to glucose-treated explants. These data provide clear evidence that under glycation stress conditions treatment with NAHP inhibited AGE formation invitro and exvivo and prevented the loss of cellular contractile forces in a glycated dermis model. Thus, NAHP obviously provides a beneficial treatment option to counteract AGE-related changes in human skin such as dermal stiffening and yellowish skin appearance.

Novel inhibitory effect of black chokeberry (Aronia melanocarpa) from selected eight berries extracts on advanced glycation end-products formation and corresponding mechanism study

Numerous health hazards have been connected to advanced glycation end products (AGEs). In this investigation, using reaction models including BSA-fructose, BSA- methylglyoxal (MGO), and BSA-glyoxal (GO), we examined the anti-glycation potential of eight different berry species on AGEs formation. Our results indicate that black chokeberry (Aronia melanocarpa) exhibited the highest inhibitory effects, with IC50 values of 0.35 ± 0.02, 0.45 ± 0.03, and 0.48 ± 0.11 mg/mL, respectively. Furthermore, our findings suggest that black chokeberry inhibits AGE formation by binding to BSA, which alleviates the conformation alteration, prevents protein cross-linking, and traps reactive α-dicarbonyls to form adducts. Notably, three major polyphenols, including cyanidin-3-O-galactoside, cyanidin-3-O-arabinoside, and procyanidin B2 from black chokeberry, showed remarkably inhibitory effect on MGO/GO capture, and new adducts formation was verified through LC-MS/MS analysis. In summary, our research provides a theoretical basis for the use of berries, particularly black chokeberry, as natural functional food components with potential anti-glycation effects.

This study aimed to explore the inhibitory effect of pyrogallol on AGE formation in the bovine serum albumin (BSA)/glucose system for 21 days at 37 °C. The AGEs formation was measured in terms of Amadori products, total AGEs, argpyrimidine, and pentosidine. Molecular docking was used to investigate the interaction between pyrogallol and BSA. According to the results, in the presence of pyrogallol, the formation of pentosidine and argpyrimidine AGEs decreased. The molecular interaction studies demonstrated that pyrogallol has a high affinity towards arginine residues of albumin. Finally, results proved pyrogallol is a vigorous antiglycation compound and fruitful for AGE inhibition.
