Real-Time Chlorophyll-a Pigment Monitoring of Chlamydomonas reinhardtii in a Controlled Environment Using Pulsed LED Fluorescence LiDAR System

The real-time chlorophyll-a pigment monitoring of C. reinhardtii is studied using our developed LED fluorescence light detection and ranging (LiDAR) system. It features a portable set-up that uses a pulsed LED module with an excitation wavelength of 385 nm. We were able to monitor the different growth phases of C. reinhardtii with specific cultivation parameters. The developed fluorescence LiDAR system showed the linear correlation of its chlorophyll-a signal with the optical density and EEM fluorescence measurements at 680 nm emission wavelength. Water quality and weather parameters were also measured, which explains the variation in the growth dynamics of C. reinhardtii during the sampling period. The results from the monitoring demonstrated a different technique that can be used in estimating algal biomass in the environment.

Laser Remote Sensing of Lake Kinneret by Compact Fluorescence LiDAR.

Harmful algal blooms in freshwater reservoirs became a steady phenomenon in recent decades, so instruments for monitoring water quality in real time are of high importance. Modern satellite remote sensing is a powerful technique for mapping large areas but cannot provide depth-resolved data on algal concentrations. As an alternative to satellite techniques, laser remote sensing is a perspective technique for depth-resolved studies of fresh or seawater. Recent progress in lasers and electronics makes it possible to construct compact and lightweight LiDARs (Light Detection and Ranging) that can be installed on small boats or drones. LiDAR sensing is an established technique; however, it is more common in studies of seas rather than freshwater reservoirs. In this study, we present an experimental verification of a compact LiDAR as an instrument for the shipborne depth profiling of chlorophyll concentration across the freshwater Lake Kinneret (Israel). Chlorophyll depth profiles of 3 m with a 1.5 m resolution were measured in situ, under sunlight conditions. A good correlation (R2 = 0.89) has been established between LiDAR signals and commercial algae profiler data. A non-monotonic algae depth distribution was observed along the boat route during daytime (Tiberias city–Jordan River mouth–Tiberias city). The impact of high algal concentration on water temperature laser remote sensing has been studied in detail to estimate the LiDAR capability of in situ simultaneous measurements of temperature and chlorophyll concentration.

Daytime measurements of bioaerosol simulants using a hyperspectral laser-induced fluorescence LIDAR for biosphere research

A new compact home-built hyper-spectral laser-induced fluorescence (LIF) light detection and ranging (LIDAR) system is presented that can detect and identify bioaerosol clouds in an open atmosphere in daylight up to a distance of 3−4 km. The system is based on a 266 nm laser source and a dual-stage micro-channel plate intensified charge-coupled device sensor, which provides a high-resolution fluorescence spectrum with the sensitivity of a single photon counting imaging detection at a 10 Hz repetition rate. Controlled remote sensing experiments on biological simulants in the aerosol-phase within an enclosed chamber (ΔR=1.2 m) were conducted from a short distance of ∼170 m. A meticulous comparison between the remote fluorescence signal detected by the system and the instantaneous concentration and particle size distribution of the bioaerosol inside the chamber detected by an aerodynamic particle sizer yielded a threshold of ∼20 and 137 μgr s m-3 for the spectral identification of tryptophan and ovalbumin, respectively. This showed that the LIDAR can measure environmental airborne biological aerosol remotely in the daytime and can be utilized as a real-time biosphere research tool. Next, the system was tested in open atmospheric field campaigns in the Israeli desert during the day. It successfully detected bioaerosol clouds at up to 2.5 km in strong wind and sunny conditions. These empirical detection and identification results provide robust estimates that a biological cloud can be sensed at a distance of ∼ 4 km.

Active 3D Imaging of Vegetation based on Multi-Wavelength Fluorescence LiDAR.

Comprehensive and accurate vegetation monitoring is required in forestry and agricultural applications. The optical remote sensing method could be a solution. However, the traditional light detection and ranging (LiDAR) scans a surface to create point clouds and provide only 3D-state information. Active laser-induced fluorescence (LIF) only measures the photosynthesis and biochemical status of vegetation and lacks information about spatial structures. In this work, we present a new Multi-Wavelength Fluorescence LiDAR (MWFL) system. The system extended the multi-channel fluorescence detection of LIF on the basis of the LiDAR scanning and ranging mechanism. Based on the principle prototype of the MWFL system, we carried out vegetation-monitoring experiments in the laboratory. The results showed that MWFL simultaneously acquires the 3D spatial structure and physiological states for precision vegetation monitoring. Laboratory experiments on interior scenes verified the system’s performance. Fluorescence point cloud classification results were evaluated at four wavelengths and by comparing them with normal vectors, to assess the MWFL system capabilities. The overall classification accuracy and Kappa coefficient increased from 70.7% and 0.17 at the single wavelength to 88.9% and 0.75 at four wavelengths. The overall classification accuracy and Kappa coefficient improved from 76.2% and 0.29 at the normal vectors to 92.5% and 0.84 at the normal vectors with four wavelengths. The study demonstrated that active 3D fluorescence imaging of vegetation based on the MWFL system has a great application potential in the field of remote sensing detection and vegetation monitoring.

Bioaerosol detection over Athens, Greece using the laser induced fluorescence technique

The challenge in today's bioaerosol monitoring is to retrieve real-time information on the qualitative and quantitative composition of the ambient air in bioparticles implicated to human health. A pilot study was conducted during March–May 2018 in Athens, Greece in order to detect bioparticles within the Planetary Boundary Layer (PBL) by implementing the LIF LiDAR (Laser-Induced Fluorescence Light Detection and Ranging) technique at an excitation wavelength of 266 nm in order to determine the major components' contribution on the total fluorescence LiDAR signals aloft (30–100 m above our site). The laboratory characterization of the prevalent pollen grains and fungal spores fluorescence signatures enabled through deconvolution the breaking down of the retrieved LIF LiDAR signals and unravelled each bioparticle's contribution. The bioaerosol occurrence and concentration, as determined by the concurrent sampling with a volumetric particle sampler, verified that the detected fluorescence is related to the fungal and pollen aerosol concentration. The results of this study are very promising for the implementation of remote sensing technology in routine detection and quantification of airborne bioparticles in real-time which is important for allergy sufferers and physicians.

Compact HLIF LiDAR for marine applications

ABSTRACTThe accidental discharge or accumulated pollution in seawater constitutes high risk of damage to the marine environment. Early detection of deviations in water quality allows taking preventive measures in order to minimize harmful influence of pollution or to implement adequate response actions before the pollution becomes a major spill. This requires regular monitoring of risk areas with effective, reliable, and economically sustainable solutions. The Hyperspectral Laser Induced Fluorescence (HLIF) LiDAR (light detection and ranging) combines highly sensitive and selective oil-in-water detection with characterization capabilities based on feature extraction and pattern recognition in HLIF spectra. Therefore, this technique is equally effective for oil detection in open and coastal waters. Operated in unattended mode as a payload of marine vehicles, it delivers the real-time analytical capabilities directly on site. This article describes the development of HLIF LiDAR and its application on board of operational vessels in the Norwegian Sea, Barents Sea, and the Baltic Sea during the period 2014–2015.

Automated Underwater Object Recognition by Means of Fluorescence LIDAR

This paper focuses on automated recognition of underwater objects by means of light detection and ranging (LIDAR) systems. Differently from most works involved in underwater object recognition with LIDAR, where objects are recognized by their shape, here the interest is distinguishing objects on the basis of physical/chemical properties of object materials. To this aim, laser-induced fluorescence (LIF) spectroscopy is exploited, and an ad hoc signal processing chain is presented to effectively analyze the LIF spectra extracted at the detected object-range. Specifically, the goal is that of automatically recognizing the detected object with respect to a database (DB) of objects of interest, which have been previously spectrally characterized by means of laboratory fluorescence measurements. To this aim, suitable physics-based methodologies are proposed to compensate the signal for water-column effects. A decision-theory-based framework is developed to approach spectral recognition of the detected object with respect to the object DB. Experimental results from a laboratory test-bed show that the proposed processing chain is effective at automatically recognizing objects submerged in an artificial water column at different depths, based on a diverse DB of sample materials. The presented approach is shown to provide great potential for automated object recognition in marine and other water environments.

Ultraviolet Fluorescence LiDAR (UFL) as a Measurement Tool for Water Quality Parameters in Turbid Lake Conditions

Despite longstanding contributions to oceanography, similar use of fluorescence light detection and ranging (LiDAR) in lake settings is not routine. The potential for ship-mounted, multispectral Ultraviolet Fluorescence LiDAR (UFL) to provide rapid, high-resolution data in variably turbid and productive lake conditions are investigated here through a series of laboratory tank and field measurements carried out on Lake Balaton, Hungary. UFL data, calibrated empirically to a set of coinciding conventionally-analyzed samples, provide simultaneous estimates of three important parameters-chlorophyll a(chla), total suspended matter (TSM) and colored dissolved organic matter (CDOM). Successful UFL retrievals from both laboratory and field measurements were achieved for chla (0.01–378 mg∙m−3; R = 0.83–0.92), TSM (0.1–130 g∙m−3; R = 0.90–0.96) and CDOM (0.003–0.125 aCDOM(440); R = 0.80–0.97). Fluorescence emission at 685 nm is shown through tank measurements to display robust but distinct relationships with chla concentration for the two cultured algae species investigated (cyanobacteria, Cylindrospermopsis raciborskii, and chlorophyta, Scenedesmus armatus). The ratio between fluorescence emissions measured at 650 nm, related to the phycocyanin fluorescence maximum, to that at 685 nm is demonstrated to effectively distinguish these two species. Validation through both laboratory measurements and field measurements confirmed that site specific calibration is necessary. This study presents the first known assessment and application of ship-mounted fluorescence LiDAR in freshwater lake conditions and demonstrates the use of UFL in measuring important water quality parameters despite the more complicated hydro-optic conditions of inland waters.

Airborne Laser-Induced Fluorescence (LIF) Light Detection and Ranging (LiDAR) for the Quantification of Dissolved Organic Matter Concentration in Natural Waters

Analysis of dissolved organic matter (DOM) concentration and composition is essential to quantifying biological and chemical oxygen demand and atmosphere–ocean heat flux exchange in natural waters. However, manual water sampling is costly and time consuming over large areas. The purpose of this research was to analyze the applicability of airborne laser-induced fluorescence light detection and ranging (LiDAR) for the detection of DOM in estuarine ecosystems impacted by agriculture. A fluorescence LiDAR system (Airborne Marine) (FLS-AM) was used to assess the DOM concentration of the Annapolis River and Basin, Nova Scotia, Canada, as well as three rivers and their estuaries in Prince Edward Island, Canada. Two FLS-AM flight missions were conducted in the summers of 2008 and 2009 and the resulting datasets were compared with spectral fluorescence signature (SFS DOM) and dissolved organic carbon (DOC) analysis of in situ water samples. Significant positive correlations were found at five of seven sites between the FLS-AM DOM and SFS DOM relationship which indicates that the FLS-AM sensor is a good surrogate for traditional sample collection of DOM data in estuaries in this region. Positive correlations were also found at all sites between FLS-AM DOM values and DOC. FLS-AM DOM patterns show that DOM values are significantly higher in rivers and estuaries that drain watersheds which are heavily impacted by agricultural practices. The results of this study show that the FLS-AM can be used efficiently as a general indicator for how estuaries are affected by runoff from agricultural watersheds in real time and thus reduce the requirement for traditional water sample collection and laboratory analysis methods.

LIDAR for Detection of Chemical and Biological Warfare Agents

Remote detection of chemical and biological warfare agents and toxic gases in the atmosphere is of current interest to both the military and civilian agencies. Out of all currently available techniques, no single technique provides efficient detection against such threats at significant standoff distances. Light detection and ranging (LIDAR) technologies, based on the transmission of laser pulses and analysis of the return signals, have demonstrated impressive capabilities in remote detection of such toxic chemicals. LIDAR is a highly sensitive tool to detect the extremely low concentrations of various toxic agents present in the form of thin clouds at distances of few kilometer. The detection of these toxic clouds is based on the approach of first detecting and measuring the range of the clouds using the scattering phenomena and subsequently identifying the composition of toxic clouds using absorption and fluorescence phenomena. Laser Science and Technology Centre (LASTEC), Delhi has been working on the design and development of LIDAR systems for detection of chemical and biological warfare (CBW) agents. In this paper, theoretical analysis of differential absorption LIDAR (DIAL) for detection of chemical agents and fluorescence LIDAR for detection of biological agents has been discussed. For some typical parametric conditions, the received power levels from different ranges to detect specific concentrations of chemical or biological clouds have been computed and discussed. The technical details of the indigenously developed backscattering LIDAR, which detects and measures the distance of cloud layers up to 5 km is also presented. Spectroscopic Calibration Correlation of Field and Lab-Sized Fluorescence LIDAR Systems

A method has been developed to correlate spectral signatures obtained with various fluorescence LIght Detection And Ranging (LIDAR) systems. A calibrated fluorescence reference target was used to calibrate the spectral response of the LIDAR transmitter channels and obtain their transfer functions. Two LIDAR systems have been spectrally characterized, and corrected signatures for two bioaerosols are presented. The first LIDAR system is the Standoff Integrated Bioserosol Active Hyperspectral Detection field LIDAR developed by Defence R&D Canada. This standoff system uses a 351-nm pulsed laser in a monoaxial design. The second system is a lab-sized aerosol chamber designed to characterize fluorescent aerosols under controlled environmental conditions. The chamber was designed according to classical short-range biaxial LIDAR principles, with the purpose of duplicating the results obtained with field LIDAR systems. Aerosols generated within the chamber are probed by a 355-nm pulsed laser, and autofluorescence spectra are measured with a spectrometer and an intensified charge-coupled device camera. This chamber is used to collect the reference spectra of various fluorescing aerosols and simulants of biological agents. One of the main objectives in using this chamber is to produce and compile a library of instrument-free fluorescing spectra that can be transferred to other LIDAR-based bioaerosol sensors with known optical transfer functions.

Standoff sensing of bioaerosols using intensified range-gated spectral analysis of laser-induced fluorescence

In atmospheric sensing, one application that has demonstrated several impressive successes over the last two decades is the light detection and ranging (LIDAR). Elastic LIDAR has shown an important capability in providing aerosol density and spatial distribution from a standoff position. However, it provides limited information on the material composition of the aerosol component. On the other hand, inelastic LIDARs (including laser-induced fluorescence and Raman LIDARs) measure the spectrally distributed returned signal that may contain important clues about the nature of the scatterers. In order to investigate the capability of these LIDARs in characterizing bioaerosols from a standoff position, Defence Research & Development Canada initiated a three-year program in spring 1999, named SINBAHD (Standoff Integrated Bioaerosol Active Hyperspectral Detection). The aim of the program was to investigate the sensitivity and discrimination capabilities of an inelastic LIDAR based on the intensified range-gated spectral detection of laser-induced fluorescence. An exploratory prototype based on this technique has shown sensitivity of a few living bioaerosol particles per liter of air for a range of 1.4 km at night. Furthermore, based on spectral signatures measured during open-air releases, good discrimination capabilities were obtained between Bacillius subtilis var globiggi (BG) and Erwinia herbicola (EH). These results agree well with a performance model using Raman returns from atmospheric nitrogen as a calibration tool.
