Recycling of expired cow milk for constructing multifunctional biomass nonfluorinated chromatic paint with superhydrophobicity

Through denaturation and amination of the mixture, consisted of expired cow milk, kaolin and gardenia yellow, in the hot organic solvent that contained alkaline aminosilane coupling agent, then adsorption of diverse acid dyes in the acid condition, thus obtaining active matrixes with different colors. By virtue of the non-catalyzed in-situ reaction among the acrylate groups of pentaerythritol ester compounds and the amino groups of obtained matrixes as well as long-chain alkyl amines, the biomass fluoride-free superhydrophobic doped paints was prepared, however the poor stability was reflected in its anti-wetting coating surface. Furthermore, in order to improve durable superhydrophobicity and multifunctionality of the expired cow milk-based water-repellent paint, the thermally denatured protein, originated from expired cow milk heating, was blended with nanoscale inorganic pigments to accept the before-mentioned procedures of amination and in-situ growth, consequently constructing the multifunctional biomass nonfluorinated chromatic paint with robust superhydrophobic function, moreover its coated fabrics had 155.7° of average contact angle and 1.5° of average sliding angle. Additionally, by only using synergy of complex denaturation and chemical coupling, the expired cow milk could be transformed into UV-photoluminescent nonfluorinated superhydrophobic color paint, the contact angle of such paint-coated fabric was 153.3° and its sliding angle was 2°. Based on the additional function of as-prepared biomass paints, the applications of non-wetted switchable fluorescence imaging as well as photothermal deicing were realized. We believe that the expired cow milk-based multifunctional paints can expand into the fields of outdoor protective textiles, decorative building materials, fluorescent falsification-preventing and ice protection.

Use of intra-operative fluorescence imaging in periprosthetic joint infection: State of the art and future perspectives.

In periprosthetic joint infections (PJIs), the surgeon's role becomes pivotal in addressing the infection locally, necessitating the surgical removal of infected and necrotic tissue. Opportunity to enhance the visualization of infected tissue during surgery could represent a game-changing innovation. The aim of this narrative review is to delineate the application of intraoperative fluorescence imaging for targeting infected tissues in PJIs. A systematic review, adhering to the Preferred Reporting Items for Systematic Review and Meta-Analyses (PRISMA) guidelines, was carried out. The search included multiple online database; MEDLINE, Scopus, and Web of Science. For data extraction the following were evaluated: (i) diagnosis of musculoskeletal infection; (ii) use of intraoperative fluorescence imaging; (iii) infected or necrotic tissues as target. Initially, 116 studies were identified through online database searches and reference investigations. The search was narrowed down to a final list of 5 papers for in-depth analysis at the full-text level. Subsequently, 2 studies were included in the review. The study included a total of 13 patients, focusing on cases of fracture-related infections of the lower limbs. The primary and crucial role for orthopedic surgeons in PJIs is the surgical debridement and precise removal of necrotic and infected tissue. Technologies that enable clear and accurate visualization of the tissue to be removed can enhance the eradication of infections, thereby promoting healing. A promising avenue for the future involves the potential application of intraoperative fluorescence imaging in pursuit of this objective.

Two-Photon Absorption Aggregation-Induced Emission Luminogen/Paclitaxel Nanoparticles for Cancer Theranostics.

Multifaceted nanoplatforms integrating fluorescence imaging and chemotherapy have garnered acknowledgment for their potential potency in cancer diagnosis and simultaneous in situ therapy. However, some drawbacks remain for traditional organic photosensitizers, such as poor photostability, short excitation wavelength, and shallow penetration depth, which will greatly lower the chemotherapy treatment efficiency. Herein, we present lipid-encapsulated two-photon active aggregation-induced emission (AIE) luminogen and paclitaxel (PTX) nanoparticles (AIE@PTX NPs) with bright red fluorescence emission, excellent photostability, and good biocompatibility. The AIE@PTX NPs exhibit dual functionality as two-photon probes for visualizing blood vessels and tumor structures, achieving penetration depth up to 186 and 120 μm, respectively. Furthermore, the tumor growth of the HeLa-xenograft model can be effectively prohibited after the fluorescence imaging-guided and PTX-induced chemotherapy, which shows great potential in the clinical application of two-photon cell and tumor fluorescence imaging and cancer treatment.

In-depth Photo-physical Properties, Characterization and Cell Imaging Applications of 2, 6-dicyanoanilines Based on Iso-phthalaldehyde and Tere-phthalaldehyde Frame

In current manuscript we have reported the interesting fluorescence properties, characterization and cell imaging applications of 2, 6-dicyanoanilines based on iso-phthalaldehyde and tere-phthalaldehyde frame. Earlier preliminary reported work was further extended to in-depth study of photo-physical properties like quantum yield, Fluorescence decay, stock shifts etc, and screening of representative compounds (1b and 2b) for use in fluorescent cell imaging applications. The 2, 6-dicyanoanilines based on iso-phthalaldehyde and tere-phthalaldehyde frame were showing very interesting photo-physical properties and promising prospectus for cell imaging applications.

Two spirobifluene-based turn-on fluorescent probes for highly selective detection of Cysteine and the applications in cells two-photon fluorescence imaging

Two spirobifluene-based fluorescent probes SPF1 and SPF2, were designed and synthesized. The probes displayed “turn-on” fluorescence response for Cysteine. One of the challenges in developing a Cysteine probe is to secure high selectivity. SPF1/SPF2 can discriminate Cysteine from GSH as well as Hcy, and showed high substrate selectivity. The detection limit of SPF1 is 36 nM, which is excellent comparing with other optical sensors for Cysteine. The sensing mechanism of SPF1/SPF2 was verified by experimental data and theoretical calculations. There was a good linear relationship between the fluorescence intensity of SPF1/SPF2 and the concentration of Cysteine. The MTT tests indicated that SPF1/SPF2 had low cytotoxicity and good biocompatibility. Theoretical calculations demonstrated that SPF1, SPF2, and their related reaction products with Cysteine exhibited good two-photon absorption properties. Finally, SPF1/SPF2 had been successfully applied to the imaging of Cysteine in living cells under two-photon excitation.

Charge Transfer-Promoted Excited State of a Heavy-Atom-Free Photosensitizer for Efficient Application of Mitochondria-Targeted Fluorescence Imaging and Hypoxia Photodynamic Therapy.

Conventional photosensitizers (PSs) used in photodynamic therapy (PDT) have shown preliminary success; however, they are often associated with several limitations including potential dark toxicity in healthy tissues, limited efficacy under acidic and hypoxic conditions, suboptimal fluorescence imaging capabilities, and nonspecific targeting during treatment. In response to these challenges, we developed a heavy-atom-free PS, denoted as Cz-SB, by incorporating ethyl carbazole into a thiophene-fused BODIPY core. A comprehensive investigation into the photophysical properties of Cz-SB was conducted through a synergistic approach involving experimental and computational investigations. The enhancement of intersystem crossing (kISC) and fluorescence emission (kfl) rate constants was achieved through a donor-acceptor pair-mediated charge transfer mechanism. Consequently, Cz-SB demonstrated remarkable efficiency in generating reactive oxygen species (ROS) under acidic and low-oxygen conditions, making it particularly effective for hypoxic cancer PDT. Furthermore, Cz-SB exhibited good biocompatibility, fluorescence imaging capabilities, and a high degree of localization within the mitochondria of living cells. We posit that Cz-SB holds substantial prospects as a versatile PS with innovative molecular design, representing a potential "one-for-all" solution in the realm of cancer phototheranostics.

Shedding light on imaging safety: Decoding the origin of photocytotoxicity in RhB-assisted fluorescence imaging.

Photocytotoxicity represents a significant limitation in the application of dye-assisted fluorescence imaging (FI), often resulting in undesirable cellular damage or even cell death, thereby restricting their practical utility. The prevalence of Rhodamine B (RhB) in FI underscores the importance of elucidating its photocytotoxicity effects to minimize photodamage. This study identifies the primary cause of photocytotoxicity stems from the generation of cytotoxic singlet oxygen in RhB, utilizing femtosecond transient absorption spectroscopy coupled with quantum chemical calculations. The Laser power-dependent cellular viability reveals a threshold at about 50 mW cm-2, surpassing which produces pronounced photocytotoxicity in vitro and in vivo. Notably, this threshold significantly falls below the safety limits (<200 mW cm-2) for laser use in health care, implying a huge risk of photodamage. This study provides valuable insights into the photocytotoxicity and offers essential guidelines for developing safer imaging protocols.

Fresnel incoherent compressive holography toward 3D videography via dual-channel simultaneous phase-shifting interferometry.

Fresnel incoherent correlation holography (FINCH) enables high-resolution 3D imaging of objects from several 2D holograms under incoherent light and has many attractive applications in motionless 3D fluorescence imaging. However, FINCH has difficulty implementing 3D imaging of dynamic scenes since multiple phase-shifting holograms need to be recorded for removing the bias term and twin image in the reconstructed scene, which requires the object to remain static during this progress. Here, we propose a dual-channel Fresnel noncoherent compressive holography method. First, a pair of holograms with π phase shifts obtained in a single shot are used for removing the bias term noise. Then, a physic-driven compressive sensing (CS) algorithm is used to achieve twin-image-free reconstruction. In addition, we analyze the reconstruction effect and suitability of the CS algorithm and two-step phase-shift filtering algorithm for objects with different complexities. The experimental results show that the proposed method can record hologram videos of 3D dynamic objects and scenes without sacrificing the imaging field of view or resolution. Moreover, the system refocuses images at arbitrary depth positions via computation, hence providing a new method for fast high-throughput incoherent 3D imaging.

Functionalized Green Carbon dots for Specific Detection of Copper in Human Serum Samples and Living Cells.

Intracellular copper ion (Cu2+) is irreplaceable and essential in regulation of physiological and biological processes, while excessive copper from bioaccumulation may cause potential hazards to human health. Hence, effective and sensitive recognition is urgently significant to prevent over-intake of copper. In this work, a novel highly sensitive and green carbon quantum dots (Green-CQDs) were synthesized by a low-cost and facile one-step microwave auxiliary method, which utilized gallic acid, carbamide and PEG400 as carbon source, nitrogen source and surface passivation agent, respectively. The decreased fluorescence illustrated excellent linear relationship with the increasing of Cu2+ concentration in a wide range. Substantial surface amino and hydroxyl group introduced by PEG400 significantly improved selectivity and sensitivity of Green-CQDs. The surface amino chelation mechanism and fluorescence internal filtration effect were demonstrated by the restored fluorescence after addition of EDTA. Crucially, the nanosensor illustrated good cell permeability, high biocompatibility and recovery rate, significantly practical application in fluorescent imaging and biosensing of intracellular Cu2+ in HepG-2 cells, which revealed a potential and promising biological applications in early diagnosis and treatment of copper ion related disease.

Excimer emission from polycyclic arenes bearing triphenylmethyl group: Solid-state fluorescence, mechanofluorochromism, aggregation-induced emission and cell imaging application

The excimer emission based on discrete π-stacked dimers of polycyclic π-systems has generated significant interest in the structure-luminescence relationship of excimers owing to their ultra-large Stokes shift. Herein, a series of excimer emissive luminogens were obtained by conjugating different polycyclic aromatic aldehydes (anthraldehyde, pyrenealdehyde and perylenealdehyde) with triphenylmethylamine. In crystalline states, all the molecules were arranged in the form of π-stacked arene dimers which were spatially isolated from each other by the bulky triphenylmethyl groups, and thus emitted bright excimer emission. The anthracene and pyrene derivatives showed fluorescence enhancement responses to grinding and the enhanced fluorescence could recover to the original state upon heating. The aggregation-induced emission (AIE) properties of them were dependent on the shapes and sizes of the polycyclic aromatic groups. The pyrene derivative showed the most excellent excimer-based AIE behavior among them. All of them were more apt to exhibit the excimer emission when formed nanoparticles with pluronic F-127 than that without pluronic F-127. Furthermore, PETP was utilized for bioimaging of living Hela cells and the high-resolution image was observed.

Mosaic-PICASSO: accurate crosstalk removal for multiplex fluorescence imaging.

Ultra-multiplexed fluorescence imaging has revolutionized our understanding of biological systems, enabling the simultaneous visualization and quantification of multiple targets within biological specimens. A recent breakthrough in this field is PICASSO, a mutual-information-based technique capable of demixing up to 15 fluorophores without their spectra, thereby significantly simplifying the application of ultra-multiplexed fluorescence imaging. However, this study has identified a limitation of mutual information (MI)-based techniques. They do not differentiate between spatial colocalization and spectral mixing. Consequently, MI-based demixing may incorrectly interpret spatially co-localized targets as non-colocalized, leading to overcorrection. We found that selecting regions within a multiplex image with low-spatial similarity for measuring spectroscopic mixing results in more accurate demixing. This method effectively minimizes overcorrections and promises to accelerate the broader adoption of ultra-multiplex imaging. Enhanced theranostic efficacy of epirubicin-loaded SPION@MSN through co-delivery of an anti-miR-21-expressing plasmid and ZIF-8 hybridization to target colon adenocarcinoma.

Using targeted drug delivery systems has emerged as a promising approach to increase the efficacy of chemotherapy, particularly in combination with gene therapy. The overexpression of miR-21 plays a crucial role in colorectal cancer (CRC) progression, and targeted inhibition of miR-21 offers significant potential for enhancing CRC chemotherapy outcomes. In this study, a theranostic system based on mesoporous silica and superparamagnetic iron oxide nanoparticles (SPION@MSNs) was synthesized as a core-shell structure. After loading epirubicin (EPI) in the open pores of MSN, the plasmid expressing anti-miR-21 (pDNA) covered the outer surface with the help of a ZIF-8 (zeolitic imidazolate framework-8) film. Afterward, polyethylene glycol (PEG) and AS1411 aptamer were conjugated to the surface to improve the protective, biocompatibility, and targeting abilities of the nanocarrier. Moreover, the physicochemical characteristics as well as the loading capacity and release profile of EPI and pDNA were fully evaluated. The uptake of the nanoparticles by CRC and normal cell lines in addition to the anticancer effects related to targeted combinational therapy were investigated in vitro. Finally, in vivo tests were performed on BALB/c mice bearing colorectal tumors to evaluate the effectiveness of the targeted nanoparticles, their possible side effects, and also their application in fluorescence and magnetic imaging in vivo. The successful synthesis of SPION@MSN-EPI/pDNA-ZIF-8-PEG-Apt nanoparticles (∼68 nm) and good loading efficiency and controlled release of EPI and pDNA were confirmed. Moreover, hemolysis and gel retardation assays demonstrated the biocompatibility and plasmid protection. Cellular uptake and expression of copGFP illustrated selective entry and transient transfection of targeted nanoparticles, consistent with the cytotoxicity results that indicated the synergistic effects of chemo-gene therapy. The results of animal studies proved the high antitumor efficiency of targeted nanoparticles with minimal tissue damage, which was in line with fluorescence and magnetic imaging results. The novel synthesized nanoparticles containing SPION@MSN-ZIF-8 were suitable for CRC theranostics, and the combined approach of chemo-gene therapy suppressed the tumor more effectively.

Mitochondria-targeting biocompatible fluorescent BODIPY probes.

An increase in the mitochondrial membrane potential (MMP) is a characteristic feature of cancer and cardiovascular disease. Therefore, it remains of crucial importance to develop new and improved fluorescent probes that are sensitive to the MMP, to report on mitochondrial health and function. Reported here are the design, synthesis, photophysical properties and biological characterisation of a series of BODIPY dyes, BODIPY-Mito-n, for mitochondria-targeted fluorescence imaging applications. Six BODIPY-Mito-n analogues were synthesised under mild conditions, and displayed excellent fluorescence quantum yields of between 0.59 and 0.72 in aqueous environments at physiological pH (pH = 7.4). The incorporation of poly(ethylene glycol) (PEG) chains to the triarylphosphonium cation moiety significantly improved the biocompatibility of the probes (BODIPY-Mito-6, IC50 > 50 μM). All BODIPY-Mito-n compounds demonstrated a high MMP-sensitive localisation in the mitochondria, with Pearson's correlation coefficients (PCC) of between 0.76 and 0.96. Compounds BODIPY-Mito-2 and BODIPY-Mito-6 revealed the highest sensitivity to the MMP, with a decrease in the emission intensity of 62% and 75%, respectively following MMP depolarisation. It is anticipated that the highest MMP sensitivity and enhanced biocompatibility of BODIPY-Mito-6 could lead to the development of new probes for mitochondrial imaging in the future.

A near-infrared AIE fluorescent probe for accurate detection of sulfur dioxide derivatives and visualization of fingerprints

In most biophysiological processes, sulfur dioxide (SO2) is an important intracellular signaling molecule that plays an important role. The change of SO2 in cells are closely related to various diseases such as neurological disorders and lung cancer, so it is necessary to develop fluorescent probes with the ability to accurately detect SO2 during physiological processes. In this work, we designed and synthesized a multifunctional fluorescent probe TIS. TIS has excellent properties such as near-infrared emission, large stokes shift, excellent SO2 detection capabilities, low detection limit, high specificity and visualization of color change before and after reaction. Simultaneously, TIS has low cytotoxicity, good biocompatibility, clear cell imaging capability and mitochondrial targeting ability. In addition, the ability of TIS to be applied to different material surfaces for latent fingerprint fluorescence imaging was also explored. TIS provides an excellent method for the accurate detection of SO2 derivatives and shows great potential applications in near-infrared cellular imaging and latent fingerprint fluorescence imaging.

Synthesis of a super-low detection limit fluorescent probe for Al3+ and its application in fluorescence imaging of zebrafish and cells

A novel fluorescent probe N'-(2-hydroxybenzylidene)-indole-3-formylhydrazine (JHK) was designed and synthesized based on the condensation reaction of indole-3-formylhydrazine and salicylaldehyde. The probe JHK solution could highly selectively recognize Al3+ by the obvious fluorescence enhancement (288-fold) after adding Al3+. And the probe solution with Al3+ had a very high fluorescence quantum yield (89.29 %). The detection limit was calculated to be 1.135 nM, which was significantly lower than many reported detection limits, indicating that the probe JHK had pretty good sensitivity. The ratio of JHK to Al3+ (1:1) and the sensing mechanism were determined by Job's plot, 1H NMR spectra, FTIR spectra, ESI-MS and Gaussian calculation. The probe solution and medium-speed filter paper were successfully used to make test papers for more convenient detection of Al3+. Furthermore, the probe JHK had been successfully applied to the detection of Al3+ in real water, zebrafish and living cells.

Multiplane HiLo microscopy with speckle illumination and non-local means denoising.

HiLo microscopy synthesizes an optically sectioned image from two images, one obtained with uniform and another with patterned illumination, such as laser speckle. Speckle-based HiLo has the advantage of being robust to aberrations but is susceptible to residual speckle noise that is difficult to control. We present a computational method to reduce this residual noise without undermining resolution. In addition, we improve the versatility of HiLo microscopy by enabling simultaneous multiplane imaging (here nine planes). Our goal is to perform fast, high-contrast, multiplane imaging with a conventional camera-based fluorescence microscope. Multiplane HiLo imaging is achieved with the use of a single camera and z-splitter prism. Speckle noise reduction is based on the application of a non-local means (NLM) denoising method to perform ensemble averaging of speckle grains. We demonstrate the capabilities of multiplane HiLo with NLM denoising both with synthesized data and by imaging cardiac and brain activity in zebrafish larvae at 40Hz frame rates. Multiplane HiLo microscopy aided by NLM denoising provides a simple tool for fast optically sectioned volumetric imaging that can be of general utility for fluorescence imaging applications.

EqPhotochemistry of a water-soluble coumarin-based photoswitch

A novel water-soluble stilbene-type molecular photoswitch (CP) has been synthesized by attaching an N-methylpyridinium unit to 7-diethylamino-4-methylcoumarin (coumarin 1) via an ethylene bridge. The photoisomerization cycle of CP and the spectroscopic properties of the two isomers were studied in acetonitrile and in water. In both solvents, the illumination of trans-CP with green light leads to a photostationary state (PSS) with 90% cis isomer and irradiating then this PSS mixture with near UV light, the trans isomer is almost fully recovered. In water, the quantum yield for the trans-cis photoisomerization is 0.042, for the cis-trans photoisomerization it is 0.23. In acetonitrile, the photoisomerization quantum yields are even higher, the respective values are 0.12 and 0.56. The trans isomer of CP is fluorescent with red emission and it has a large Stokes shift, whereas the cis isomer is non-fluorescent. The two isomers are thermally stable at room temperature. The efficient photoisomerization of CP in both direction, the thermal stability of both isomeric forms and the distinct fluorescence of the trans isomer suggest that some water-soluble coumarin-based photoswitches may be promising candidates for applications in fluorescence imaging of biological samples.

Single Atom Stabilization of Phosphinate Ester-Containing Rhodamines Yields Cell Permeable Probes for Turn-On Photoacoustic Imaging.

Photoacoustic imaging (PAI) is an emerging imaging technique that uses pulsed laser excitation with near-infrared (NIR) light to elicit local temperature increases through non-radiative relaxation events, ultimately leading to the production of ultrasound waves. The classical xanthene dye scaffold has found numerous applications in fluorescence imaging, however, xanthenes are rarely utilized for PAI since they do not typically display NIR absorbance. Herein, we report the ability of Nebraska Red (NR) xanthene dyes to produce photoacoustic (PA) signal and provide a rational design approach to reduce the hydrolysis rate of ester containing dyes, affording cell permeable probes. To demonstrate the utility of this approach, we construct the first cell permeable rhodamine-based, turn-on PAI imaging probe for hypochlorous acid (HOCl) with maximal absorbance within the range of commercial PA instrumentation. This probe, termed SNR700 -HOCl, is capable of detecting exogenous HOCl in mice. This work provides a new set of rhodamine-based PAI agents as well as a rational design approach to stabilize esterified versions of NR dyes with desirable properties for PAI. In the long term, the reagents described herein could be utilized to enable non-invasive imaging of HOCl in disease-relevant model systems.

Tetraphenylethylene-based AIE nanoprobes for labeling lysosome by two-photon imaging in living cells

Lysosomes are essential cellular organelles, serving vital functions in cellular metabolism and degradation. The design of specifically targeting lysosomes probes with aggregation-induced emission (AIE) characteristics using two-photon excitation techniques is significance and challenging work. Here we designed and synthesized two tetraphenylethylene (TPE)-based AIE fluorescence probes, naming TPE-Ma and TPE-Py, with TPE as the matrix and morpholine (Ma) or pyrrolidone (Py) as the targeting group. These probes exhibit a significant Stokes shift, low cytotoxicity, two-photo fluorescence imaging and lysosome-specific targeting capability ensuring their suitability for fluorescence imaging applications. To enhance the water solubility and cellular accumulation of TPE-Ma and TPE-Py in tumor cells, we employed a biocompatible polymer 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000] (DSPE-mPEG2000) as a nanocarrier. By encapsulating TPE-Ma and TPE-Py within DSPE-mPEG2000, we successfully developed two AIE fluorescent nanoprobes known as DSPE@ TPE-Ma and DSPE@ TPE-Py. The results demonstrated that fluorescent nanoprobes DSPE@ TPE-Ma and DSPE@ TPE-Py possess excellent cell permeability, biocompatibility, superior photostability and specific targeting towards lysosomes in MCF-7 cells. Our findings highlight the potential of these fluorescent nanoprobes as effective tools for two-photon fluorescence imaging and targeted detection of lysosomes in cancer cells.
