Wheat Germ Agglutinin-Conjugated Disulfide Cross-Linked Alginate Nanoparticles as a Docetaxel Carrier for Colon Cancer Therapy.

PurposeIn chemotherapy, oral administration of drug is limited due to lack of drug specificity for localized colon cancer cells. The inability of drugs to differentiate cancer cells from normal cells induces side effects. Colonic targeting with polymeric nanoparticulate drug delivery offers high potential strategies for delivering hydrophobic drugs and fewer side effects to the target site. Disulfide cross-linked polymers have recently acquired high significance due to their potential to degrade in reducing colon conditions while resisting the upper gastrointestinal tract’s hostile environment. The goal of this project is, therefore, to develop pH-sensitive and redox-responsive fluorescein-labeled wheat germ agglutinin (fWGA)-mounted disulfide cross-linked alginate nanoparticles (fDTP2) directly targeting docetaxel (DTX) in colon cancer cells.MethodsfDTP2 was prepared by mounting fWGA on DTX-loaded nanoparticles (DTP2) using the two-step carbodiimide method. Morphology of fDTP2 was examined using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Dynamic light scattering (DLS) study was carried out to determine the mean diameter, polydispersity index (PDI) and zeta potential of fDTP2. Cellular uptake efficiency was examined using fluorescence microplate reader. Biocompatibility and active internalization of fDTP2 were conducted on HT-29.ResultsfDTP2 was found to exhibit a DTX loading efficiency of 19.3%. SEM and TEM tests revealed spherical nanoparticles. The in vitro DTX release test showed a cumulative release of 54.7%. From the DLS study, fDTP2 reported a 277.7 nm mean diameter with PDI below 0.35 and −1.0 mV zeta potential. HT-29 which was fDTP2-treated demonstrated lower viability than L929 with a half maximal inhibitory concentration (IC50) of 34.7 µg/mL. HT-29 (33.4%) internalized fDTP2 efficiently at 2 h incubation. The study on HT-29 active internalization of nanoparticles through fluorescence and confocal imaging indicated such.ConclusionIn short, fDTP2 demonstrated promise as a colonic drug delivery DTX transporter.

Docetaxel-Loaded Disulfide Cross-Linked Nanoparticles Derived from Thiolated Sodium Alginate for Colon Cancer Drug Delivery.

In this study, fluorescein-labelled wheat germ agglutinin (fWGA)-conjugated disulfide cross-linked sodium alginate nanoparticles were developed to specifically target docetaxel (DTX) to colon cancer cells. Different amounts of 3-mercaptopropionic acid (MPA) were covalently attached to sodium alginate to form thiolated sodium alginate (MPA1–5). These polymers were then self-assembled and air-oxidised to form disulfide cross-linked nanoparticles (MP1–5) under sonication. DTX was successfully loaded into the resulting MP1–5 to form DTX-loaded nanoparticles (DMP1–5). DMP2 had the highest loading efficiency (17.8%), thus was chosen for fWGA surface conjugation to form fWGA-conjugated nanoparticles (fDMP2) with a conjugation efficiency of 14.1%. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) analyses showed spherical nanoparticles, and an in vitro drug release study recorded a cumulative drug release of 48.6%. Dynamic light scattering (DLS) analysis revealed a mean diameter (MD) of 289 nm with a polydispersity index (PDI) of 0.3 and a zeta potential of −2.2 mV for fDMP2. HT-29 human colon cancer cells treated with fDMP2 showed lower viability than that of L929 mouse fibroblast cells. These results indicate that fDMP2 was efficiently taken up by HT-29 cells (29.9%). Fluorescence and confocal imaging analyses also showed possible internalisation of nanoparticles by HT-29 cells. In conclusion, fDMP2 shows promise as a DTX carrier for colon cancer drug delivery.

Assessment of wheat germ agglutinin staining of the ocular surface in contact lens wearers and non-contact lens wearers

Purpose: Fluorescein-labeled wheat germ agglutinin (F-WGA) can act as a clinical marker for the presence of the ocular surface glycocalyx. This study sought to investigate whether the degree of F-WGA fluorescence observed at the ocular surface differed between symptomatic contact lens wearers, asymptomatic contact lens wearers and non-contact lens wearers.

FvSNF1, the sucrose non-fermenting protein kinase gene of Fusarium virguliforme, is required for cell-wall-degrading enzymes expression and sudden death syndrome development in soybean

Fusarium virguliforme is a soil-borne pathogenic fungus that causes sudden death syndrome (SDS) in soybean. Its pathogenicity is believed to require the activity of cell-wall-degrading enzymes (CWDEs). The sucrose non-fermenting protein kinase 1 gene (SNF1) is a key component of the glucose de-repression pathway in yeast, and a regulator of gene expression for CWDEs in some plant pathogenic fungi. To elucidate the functional role of the SNF1 homolog in F. virguliforme, FvSNF1 was disrupted using a split-marker strategy. Disruption of FvSNF1 in F. virguliforme abolishes galactose utilization and causes poor growth on xylose, arabinose and sucrose. However, the resulting Fvsnf1 mutant grew similar to wild-type and ectopic transformants on glucose, fructose, maltose, or pectin as the main source of carbon. The Fvsnf1 mutant displayed no expression of the gene-encoding galactose oxidase (GAO), a secretory enzyme that catalyzes oxidation of D-galactose. It also exhibited a significant reduction in the expression of several CWDE-coding genes in contrast to the wild-type strain. Greenhouse pathogenicity assays revealed that the Fvsnf1 mutant was severely impaired in its ability to cause SDS on challenged soybean plants. Microscopy and microtome studies on infected roots showed that the Fvsnf1 mutant was defective in colonizing vascular tissue of infected plants. Cross and longitudinal sections of infected roots stained with fluorescein-labeled wheat germ agglutinin and Congo red showed that the Fvsnf1 mutant failed to colonize the xylem vessels and phloem tissue at later stages of infection. Quantification of the fungal biomass in inoculated roots further confirmed a reduced colonization of roots by the Fvsnf1 mutant when compared to the wild type. These findings suggest that FvSNF1 regulates the expression of CWDEs in F. virguliforme, thus affecting the virulence of the fungus on soybean.

Determination of the glycosylation-pattern of the middle ear mucosa in guinea pigs

In the present study the glycosylation pattern of the middle ear mucosa (MEM) of guinea pigs, an approved model for middle ear research, was characterized with the purpose to identify bioadhesive ligands which might prolong the contact time of drug delivery systems with the middle ear mucosa (MEM). To assess the utility of five fluorescein labeled plant lectins with different carbohydrate specificities as bioadhesive ligands, viable MEM specimens were incubated at 4°C and the lectin binding capacities were calculated from the MEM-associated relative fluorescence intensities. Among all lectins under investigation, fluorescein-labeled wheat germ agglutinin (F-WGA) emerged as the highest bioadhesive lectin. In general, the accessibility of carbohydrate moieties of the MEM followed the order: sialic acid and N-acetyl-d-glucosamine (WGA)>>mannose and galactosamine (Lensculinaris agglutinin)>N-acetyl-d-glucosamine (Solanumtuberosum agglutinin)>fucose (Ulexeuropaeus isoagglutinin I)>>terminal mannose α-(1,3)-mannose (Galanthusnivalis agglutinin). Competitive inhibition studies with the corresponding carbohydrate revealed that F-WGA-binding was inhibited up to 90% confirming specificity of the F-WGA–MEM interaction. The cilia of the MEM were identified as F-WGA binding sites by fluorescence imaging as well as a z-stack of overlays of transmission, F-WGA- and nuclei-stained images of the MEM. Additionally, co-localisation experiments revealed that F-WGA bound to acidic mucopolysaccharides of the MEM. All in all, lectin-mediated bioadhesion to the MEM is proposed as a new concept for drug delivery to prolong the residence time of the drug in the tympanic cavity especially for successful therapy for difficult-to-treat diseases such as otitis media.

Fluorophotometric Analysis of the Ocular Surface Glycocalyx in Soft Contact Lens Wearers

Purpose: Unstable tear film characterized by shorting of tear break-up time (BUT) is associated with discomfort and dryness in contact lens wearers. The glycocalyx is thought to be crucial in maintaining the wettability and lubrication of the ocular surface. We evaluated the ocular surface glycocalyx in soft contact lens (SCL) wearers using a fluorescein-labeled wheat germ agglutinin (F-WGA) as a marker to demonstrate the ocular surface glycoconjugates in vivo.Methods: Twenty experienced SCL wearers and 20 healthy volunteers with no history of CL wearing (controls) were enrolled in the study. After applying a 5% F-WGA solution to the eyes of study individuals, fluorescent intensities in their respective central corneas were measured by fluorophotometry. The relationship between F-WGA intensity in the corneal surface and clinical parameters associated with contact lens wear were analyzed.Results: F-WGA fluorescence intensity in the SCL group was 418.5 ± 103.3, which was significantly lower than that of the controls (825.0 ± 179.8; p < 0.0001, Mann–Whitney test). F-WGA fluorescence intensity was not correlated with Schirmer’s test values or age, whereas a statistically significant correlation between F-WGA fluorescence intensity and tear film BUT was observed (r = 0.77, p < 0.0001). The decrease in F-WGA fluorescence intensity could be reversed by discontinuation of SCL use.Conclusion: Reduction and/or compositional alteration of ocular surface glycocalyx may be one of the causative factors of SCL-induced eye dryness.

Preparation, characterization and application of artificial Caco-2 cell surfaces in the silver nanoparticle enhanced fluorescence technique

To approach in vivo conditions during real time monitoring of biorecognitive interactions, biomimetic surfaces were prepared by fusion of purified plasma membrane fractions from Caco-2 cells with self-assembled monolayers attached to silver colloid coated standard microplates. Proper orientation and integrity of the membrane coating was confirmed by binding of fluorescein-labelled wheat germ agglutinin (F-WGA) which interacts with certain carbohydrates of the glycocalyx. Additionally, the competition with the complementary carbohydrate decreased the F-WGA binding to 15%. The assay setup offers information about the real time binding kinetics and the affinity of the interaction with the cell membrane excluding interfering events such as internalization and metabolism. As exemplified by F-WGA-binding, the mean velocity of the interaction is 627.07 mFU/s and the working range is 40–240 nM with a detection limit of 1.6 pmol F-WGA. The storage stability of the ready-to-use plates exceeded at least 1 month. The real time monitoring of lectin-prodrug binding to the biomimetic membranes revealed that high conjugation numbers reduces the affinity.The assays are simple and fast one-step reactions without any washing steps as this new technique discriminates between membrane bound and bulk fluorescence. Thus, biomimetic membranes on silver colloid layers represent a versatile tool for high throughput screening at early stages of drug discovery and development.

Hemocyte surface changes in the migratory grasshopper, Melanoplus sanguinipes in response to wounding and infection with Beauveria bassiana

AbstractSurface changes of hemocytes from the migratory grasshopper, Melanoplus sanguinipes (Fab.), following wounding and/or infection with Beauveria bassiana (Bals.) Vuillemin were assessed using fluorescein labelled wheat germ agglutinin (WGA) and concanavalin A (Con A). Binding was observed on outer wall of hemocytes when either Con A or WGA conjugates were tested. After infection there was an increase in the percentage of hemocytes which bound to WGA and which remained so for 24 h. However, after wounding this increase in binding subsided after only 1 h. Furthermore, it was found that the increase in WGA‐binding occurred within the first 15 min of wounding or infection. In addition, ConA binding in males only increased 3‐fold in fungus‐injected insects within 1 h of treatment and remained high for 24 h. These results indicated the presence of mannose (and/or glucose) residues (Con A specificity) and N‐acetyl‐D‐glucosamine residues (wheat germ agglutinin; WGA specificity) on outer wall surfaces of the hemocytes. The implications of these results in the immune response of M. sanguinipes to infection with B. bassiana are discussed.

Chitin: a cell-surface component of Phytomonas françai.

The occurrence of chitin as a structural component of the surface of the phytopathogenic protozoan Phytomonas françai was demonstrated by paper and gas-liquid chromatographic analysis of the products of enzymatic and chemical hydrolysis of alkali-resistant polysaccharides, lectin binding, glycosidase digestion, and infrared spectra. Chitin was characterized by its insolubility in hot alkali and chromatographic immobility as well as by the release of glucosamine on hydrolysis with strong acid and of N-acetylglucosamine (GlcNAc) on hydrolysis with chitinase. The presence of chitin was also shown directly by binding of wheat-germ agglutinin (WGA), which recognizes GlcNAc units, to the parasite surface. Fluorescein-labeled WGA binding was completely abolished by treatment with chitinase. This effect was specific since it could be prevented by incubating the enzyme with chitin before treatment of the phytomonads. These findings indicate that chitin is an exposed cell-surface polysaccharide in Phytomonas françai. The data were confirmed by the infrared spectrum of an alkali-insoluble residue, which showed a pattern typical of chitin.

Single photon radioluminescence. II. Signal detection and biological applications

A quantitative theory for excitation of fluorescent molecules by beta decay electrons is reported in the accompanying manuscript; experimental detection methods and biological applications are reported here. The single photon signals produced by an excited fluorophore (single photon radioluminescence, SPR) provide quantitative information about the distance between radioisotope and fluorophore. Instrumentation was constructed for SPR signal detection. Photons produced in a 0.5-ml sample volume were detected by a cooled photomultiplier and photon counting electronics. To minimize electronic noise and drift for detection of very small SPR signals, a mechanical light chopper was used for gated-signal detection, and a pulse height analyzer for noise rejection. SPR signals of approximately 1 cps were reproducibly measurable. The influence of inner filter effect, sample turbidity, and fluorophore environment (lipid, protein, and carbohydrate) on SPR signals were evaluated experimentally. SPR was then applied to measure lipid exchange kinetics, ligand binding, and membrane transport, and to determine an intermolecular distance in an intact membrane. (a. Lipid exchange kinetics.) Transfer of 12-anthroyloxystearic acid (12-AS) from sonicated lipid vesicles and micelles to vesicles containing 3H-cholesterol was measured from the time course of increasing SPR signal. At 22 degrees C, the half-times for 12-AS transfer from vesicles and micelles were 3.3 and 1.1 min, respectively. (b. Ligand binding.) Binding of 3H-oleic acid to albumin in solution, and 3H-2,2'-dihydro-4,4'-diisothiocyanodisulfonic stilbene (3H-H2DIDS) to band 3 on the erythrocyte membranes were detected by the radioluminescence of the intrinsic tryptophans. The SPR signal from 5 microCi 3H-oleic acid bound to 0.3 mM albumin decreased from 13 +/- 2 cps to 3 +/- 2 cps upon addition of nonradioactive oleic acid, giving 2.7 high affinity oleic acid binding sites per albumin. The SPR signal from 1 microCi 3H-H2DIDS bound selectively to erythrocyte band 3 in erythrocyte ghosts (1.5 mg protein/ml) was 2.2 +/- 0.8 cps. (c. Membrane transport). Dilution of J774 macrophages loaded with 3H-3-O-methylglucose and BCECF gave a decreasing SPR signal with a half-time of 81 s due to methylglucose efflux; the SPR measurement of the efflux rate was in agreement with a conventional tracer efflux rate determination by filtration. 20 microM cytochalasin B inhibited efflux by 97%. (d. Distance determination.) The SPR signal from erythrocyte membranes labeled with 27 microCi 3H-oleic acid and 10 microM of fluorescein-labeled wheat germ agglutinin was 5.7 +/- 0.5 cps, giving an average glycocalyx-to-bilayer distance of 5 nm. The results establish methods for experimental detection of SPR signals and demonstrate the applications of radioluminescence to the measurement of lipid exchange kinetics, ligand binding, membrane transport, and submicroscopic distances in intact membranes in real time.

Tvichostrongylus colubriformis: analysis of monoclonal antibody and lectin binding to the larval cuticle

A monoclonal antibody (VRI 86-1) raised against third stage Trichostrongylus colubriformis preferentially bound to the excretory pore of living exsheathed larvae, with little or no binding to other sites on the parasite surface. A similar binding pattern was observed with fluorescein-labelled wheat germ agglutinin (WGA), although the anterior end of the parasite was also stained. To gain information at the molecular level regarding the parasite components at these sites, the residues recognized by WGA (i.e. N-acetylglucosamine and N-acetylneuraminic acid) were radiolabelled on the surface of living larvae. After homogenization and detergent extraction of the larvae, four dominant bands and a number of minor bands were revealed by SDS-PAGE and fluorography. None of these bands was specifically immunoprecipitated or recognized on a Western blot by VRI 86-1, suggesting that the epitope recognized by this antibody either resides on a different molecule or is destroyed or changed during the radiolabelling procedures. These results provide further evidence that the nematode cuticle is not uniform at the molecular level, and that the excretory pore contains molecules and antigens that may be unique to that site.

Hemocyte Cell Surface Changes in Aedes aegypti in Response to Microfilariae of Dirofilaria immitis

This study involved the assessment of surface changes on hemocytes of Aedes aegypti black-eyed Liverpool strain in association with the melanization response against intrathoracically inoculated Dirofilaria immitis microfilariae (mff). Surface changes on hemocytes were identified using fluorescein-labeled wheat germ agglutinin (WGA). In mosquitoes eliciting a melanization response against inoculated mff, there was a 5-fold increase in the percentages of hemocytes exhibiting WGA binding compared with saline inoculated controls. Relationships of this hemocyte activation in relation to cell-mediated melanization responses of adult mosquitoes against mff are discussed.

Rearrangement of Con A and WGA receptors on murine mastocytoma cell membranes: relationship to cell cycle.

Incubation of interphase murine mastocytoma cells with fluorescein-labeled wheat germ agglutinin (FITC-WGA) or with rhodamine-labeled concanavalin A (Rh-Con A) shows binding of the lectins to all the cells in a ring form distribution. Treatment of the cells with trypsin before addition of FITC-WGA, or with colchicine before addition of Rh-Con A, promoted rearrangement of the lectin binding sites into cap formation in 27-29% of the cells. Trypsin had no effect on cap formation by Rh-Con A, and colchicine had no effect on cap formation by FITC-WGA. When the same cells were pretreated consecutively with trypsin and colchicine followed by incubation with FITC-WGA and Rh-Con A, no increase in the percentage of cells bearing caps was observed. Moreover, the same cells showed co-capping with both lectins. Experiments with synchronous cultures of mastocytoma cells have indicated that capping induced by either lectin is maximal in the G2 phase and minimal in the G1 and S phases of the cell cycle. At the G2 phase 60% of the cells show cap formation either after trypsin-WGA treatment or after colchicine-Con A treatment. These data indicate that rearrangement of membrane receptors for Con-A and WGA in murine mastocytoma cells can occur only at certain phases of the cell cycle.

Lectin binding by Giardia lamblia.

Surface carbohydrates of Giardia lamblia were examined using six plant lectins chosen because of their specificity for major carbohydrates moieties. The binding to axenically grown G. lamblia trophozoites was assessed in both a quantitative microagglutination assay and a fluorescence assay. Of the six lectins tested, wheat germ agglutinin (WGA) agglutinated the highest percentage (22.9 +/- 3.7%) of live trophozoites, and fluorescein-labeled WGA (100 micrograms/ml) bound to 98 +/- 5% of them. Since the carbohydrate specificity of WGA includes both N-acetyl-D-glucosamine (GlcNAc) and sialic acid, inhibition experiments were performed. GlcNAc inhibited he binding of WGA to G. lamblia in both assays to a greater extent than did sialic acid. Binding of WGA was not altered by prior treatment of trophozoites with neuraminidase, suggesting that WGA was binding to GlcNAc moieties on G. lamblia and not to sialic acid. The remaining five lectins either bound nonspecifically or exhibited low percentages of binding. The apparent presence of GlcNAc but not sialic acid or other exposed surface carbohydrates may be important in the interaction of G. lamblia with its human host.

Purification and characterization of plasma membrane fractions from cultured pituitary cells

Highly purified plasma membrane fractions have been prepared from GH 3 pituitary cells grown in suspension cultures. These membrane fractions have been obtained by differential and sucrose gradient centrifugation and were characterized in terms of their lipid content, marker enzyme analysis and the binding of 3H-labelled thyrotropin-releasing hormone (TRH) to its receptor. Alkaline phosphatase and 5′-nucleotidase activities were enriched 12- to 15-fold in the plasma membrane fraction with somewhat greater enrichment (28-fold) of the specific binding component for [ 3H]TRH, with a specific activity of 2286 fmol [ 3H]TRH bound per mg protein. A single class of binding sites for TRH was observed with an apparent dissociation constant of 18 nM, a value similar to that observed for intact cells. No detectable TRH binding to the nuclear fraction was observed that could not be ascribed to residual plasma membrane contamination. By electron microscopy, these fragments appeared to be sealed vesicles with an average diameter of approximately 1800 Å. The binding of 125I-labelled wheat germ agglutinin was used as a marker for plasma membrane purification. In addition to specific binding to this membrane fraction, specific binding was also observed in the nuclear fraction. Studies with fluorescein-labelled wheat germ agglutinin revealed that, in fixed cells, fluorescence was restricted to the plasma membrane. However, if the cells were treated with Triton before labelling, most of the fluorescence was then associated with the cell nucleus. Hence, the use of wheat germ agglutinin binding as a specific plasma membrane marker must be reevaluated.
