Literatur Review: Siderophore Activity and Campability of Bacillus as Pathogen Controller of Eggplant Plant

Solanum melongena (eggplant) is an important cultivated vegetable crop with a very high consumer level, both in tropical and subtropical regions. Eggplant is widely favored by the public because in addition to having a good taste, it can also be made pickled and candied eggplant, and is good as a source of nutrition in supporting public health. Siderophores are metal-coating agents with low molecular mass (200-2000 Da) produced by microorganisms and plants, especially under Fe-limiting conditions. Antagonistic bacteria generally live colonizing roots an d are beneficial, because they are able to increase plant growth and induce systemic resistance. one species of the genus Bacillus spp. namely B. subtilis is able to produce amylase, protease, pullunase, chitinase, xylanase and lipase enzymes which are secondary metabolites to control pathogens and spur plant growth. This article is sourced from relevant literature studies related to the title. The purpose of writing the article is to determine the benefits of siderophores as pathogen disease control in eggplant plants. Results successfully inhibited the growth of pathogens.

Antimicrobial Delivery Using Metallophore-Responsive Dynamic Nanocarriers.

The increasing prevalence of multidrug-resistant (MDR) pathogens has promoted the development of innovative approaches, such as drug repurposing, synergy, and efficient delivery, in complement to traditional antibiotics. In this study, we present an approach based on biocompatible nanocarriers containing antimicrobial cations and known antibiotics. The matrices were prepared by coordinating GaIII or InIII to formulations of chitosan/tripolyphosphate or catechol-functionalized chitosan with or without encapsulated antibiotics, yielding particles of 100-200 nm in hydrodynamic diameter. MDR clinical isolates of Pseudomonas aeruginosa were found to be effectively inhibited by the nanocarriers under nutrient-limiting conditions. Fractional inhibitory concentration (FIC) indices revealed that cation- and antibiotic-encapsulated nanomatrices were effective against both Gram-negative and Gram-positive pathogens. Metallophores, such as deferoxamine (DFO), were probed to facilitate the sequestration and transport of the antimicrobial cations GaIII or InIII. Although the antimicrobial activities were less significant with DFO, the eradication of biofilm-associated bacteria showed promising trends against P. aeruginosa and Staphylococcus epidermidis. Interestingly, indium-containing compounds showed enhanced activity on biofilm formation and eradication, neutralizing P. aeruginosa under Fe-limiting conditions. In particular, InIII-cross-linked catechol-modified chitosan matrices were able to inhibit pathogenic growth together with DFO. The nanocarriers showed low cytotoxicity toward A549 cells and improvable CC50 values with NIH/3T3 cells.

Convergent gene pair dSH3 and irr regulate Pi and Fe homeostasis in Bradyrhizobium diazoefficiens USDA110 and symbiotic nitrogen fixation efficiency

The nitrogen-fixing bacteroids inhabit inside legume root nodules must manage finely the utilization of P and Fe, the two most critical elements, due to their antagonistic interactions. While the balance mechanism for them remains unclear. A double SH3 domain-containing protein (dSH3) in the Bradyrhizobium diazoefficiens USDA110 was found to inhibit the alkaline phosphatase activity, thereby reducing P supply from organophosphates. The dSH3 gene is adjacent to the irr gene, which encodes the iron response repressor and regulates Fe homeostasis under Fe-limited conditions. Their transcription directions converge to a common intergenic sequence (IGS) region, forming a convergent transcription. Extending the IGS region through Tn5 transposon or pVO155 plasmid insertion significantly down-regulated expression of this gene pair, leading to a remarkable accumulation of P and an inability to grow under Fe-limited conditions. Inoculation of soybean with either of the insertion mutants resulted in N2-fixing failure. However, the IGS-deleted mutant showed no visible changes in N2-fixing efficiency on soybean compared to that inoculated with wild type. These findings reveal a novel regulative strategy in the IGS region and its flanking convergent gene pair for antagonistic utilization of P and Fe in rhizobia and coordination of N2-fixing efficiency.

Stimulating and toxic effect of chromium on growth and photosynthesis of a marine chlorophyte.

Marine phytoplankton can interchange trace metals in various biochemical functions, particularly under metal-limiting conditions. Here, we investigate the stimulating and toxicity effect of chromium (Cr) on a marine Chlorophyceae Osetreococcus tauri under Fe-replete and Fe-deficient conditions. We determined the growth, photosynthesis, and proteome expressions of Osetreococcus tauri cultured under different Cr and Fe concentrations. In Fe-replete conditions, the presence of Cr(VI) stimulated significantly the growth rate and the maximum yield of photochemistry of photosystem II (Fv /Fm ) of the phytoplankton, while the functional absorption cross-section of photosystem II (σPSII ) did not change. Minor additions of Cr(VI) partially rescued phytoplankton growth under Fe-limited conditions. Proteomic analysis of this alga grown in Fe-replete normal and Fe-replete with Cr addition media (10 μM Cr) showed that the presence of Cr significantly decreased the expression of phosphate-transporting proteins and photosynthetic proteins, while increasing the expression of proteins related to carbon assimilation. Cr can stimulate the growth and photosynthesis of O. tauri, but the effects are dependent on both the Cr(VI) concentration and the availability of Fe. The proteomic results further suggest that Cr(VI) addition might significantly increase starch production and carbon fixation.

Abiotic and Biotic Reduction of Iodate Driven by Shewanella oneidensis MR-1.

Iodate (IO3-) can be abiotically reduced by Fe(II) or biotically reduced by the dissimilatory Fe(III)-reducing bacterium Shewanella oneidensis (MR-1) via its DmsEFAB and MtrCAB. However, the intermediates and stoichiometry between the Fe(II) and IO3- reaction and the relative contribution of abiotic and biotic IO3- reduction by biogenic Fe(II) and MR-1 in the presence of Fe(III) remain unclear. In this study, we found that abiotic reduction of IO3- by Fe(II) produced intermediates HIO and I- at a ratio of 1:2, followed by HIO disproportionation to I- and IO3-. Comparative analyses of IO3- reduction by MR-1 wild type (WT), MR-1 mutants deficient in DmsEFAB or MtrCAB, and Shewanella sp. ANA-3 in the presence of Fe(III)-citrate, Fe(III) oxides, or clay minerals showed that abiotic IO3- reduction by biogenic Fe(II) predominated under iron-rich conditions, while biotic IO3- reduction by DmsEFAB played a more dominant role under iron-poor conditions. Compared to that in the presence of Fe(III)-citrate, MR-1 WT reduced more IO3- in the presence of Fe(III) oxides and clay minerals. The observed abiotic and biotic IO3- reduction by MR-1 under Fe-rich and Fe-limited conditions suggests that Fe(III)-reducing bacteria could contribute to the transformation of iodine species and I- enrichment in natural iodine-rich environments.

Identification and Functional Analysis of Two Mitoferrins, CsMIT1 and CsMIT2, Participating in Iron Homeostasis in Cucumber

Mitochondria are one of the major iron sinks in plant cells. Mitochondrial iron accumulation involves the action of ferric reductase oxidases (FRO) and carriers located in the inner mitochondrial membrane. It has been suggested that among these transporters, mitoferrins (mitochondrial iron transporters, MITs) belonging to the mitochondrial carrier family (MCF) function as mitochondrial iron importers. In this study, two cucumber proteins, CsMIT1 and CsMIT2, with high homology to Arabidopsis, rice and yeast MITs were identified and characterized. CsMIT1 and CsMIT2 were expressed in all organs of the two-week-old seedlings. Under Fe-limited conditions as well as Fe excess, the mRNA levels of CsMIT1 and CsMIT2 were altered, suggesting their regulation by iron availability. Analyses using Arabidopsis protoplasts confirmed the mitochondrial localization of cucumber mitoferrins. Expression of CsMIT1 and CsMIT2 restored the growth of the Δmrs3Δmrs4 mutant (defective in mitochondrial Fe transport), but not in mutants sensitive to other heavy metals. Moreover, the altered cytosolic and mitochondrial Fe concentrations, observed in the Δmrs3Δmrs4 strain, were recovered almost to the levels of WT yeast by expressing CsMIT1 or CsMIT2. These results indicate that cucumber proteins are involved in the iron transport from the cytoplasm to the mitochondria.

Role of Serine Coordination in the Structural and Functional Protection of the Nitrogenase P-Cluster.

Nitrogenase catalyzes the multielectron reduction of dinitrogen to ammonia. Electron transfer in the catalytic protein (MoFeP) proceeds through a unique [8Fe-7S] cluster (P-cluster) to the active site (FeMoco). In the reduced, all-ferrous (PN) state, the P-cluster is coordinated by six cysteine residues. Upon two-electron oxidation to the P2+ state, the P-cluster undergoes conformational changes in which a highly conserved oxygen-based residue (a Ser or a Tyr) and a backbone amide additionally ligate the cluster. Previous studies of Azotobacter vinelandii (Av) MoFeP revealed that when the oxygen-based residue, βSer188, was mutated to a noncoordinating residue, Ala, the P-cluster became redox-labile and reversibly lost two of its eight Fe centers. Surprisingly, the Av strain with a MoFeP variant that lacked the serine ligand (Av βSer188Ala MoFeP) displayed the same diazotrophic growth and in vitro enzyme turnover rates as wild-type Av MoFeP, calling into question the necessity of this conserved ligand for nitrogenase function. Based on these observations, we hypothesized that βSer188 plays a role in protecting the P-cluster under nonideal conditions. Here, we investigated the protective role of βSer188 both in vivo and in vitro by characterizing the ability of Av βSer188Ala cells to grow under suboptimal conditions (high oxidative stress or Fe limitation) and by determining the tendency of βSer188Ala MoFeP to be mismetallated in vitro. Our results demonstrate that βSer188 (1) increases Av cell survival upon exposure to oxidative stress in the form of hydrogen peroxide, (2) is necessary for efficient Av diazotrophic growth under Fe-limiting conditions, and (3) may protect the P-cluster from metal exchange in vitro. Taken together, our findings suggest a structural adaptation of nitrogenase to protect the P-cluster via Ser ligation, which is a previously unidentified functional role of the Ser residue in redox proteins and adds to the expanding functional roles of non-Cys ligands to FeS clusters.

Effect of the Interaction between Elevated Carbon Dioxide and Iron Limitation on Proteomic Profiling of Soybean.

Elevated atmospheric CO2 (eCO2) and iron (Fe) availability are important factors affecting plant growth that may impact the proteomic profile of crop plants. In this study, soybean plants treated under Fe-limited (0.5 mM) and Fe-sufficient (20 mM) conditions were grown at ambient (400 μmol mol-1) and eCO2 (800 μmol mol-1) in hydroponic solutions. Elevated CO2 increased biomass from 2.14 to 3.14 g plant-1 and from 1.18 to 2.91 g plant-1 under Fe-sufficient and Fe-limited conditions, respectively, but did not affect leaf photosynthesis. Sugar concentration increased from 10.92 to 26.17 μmol g FW-1 in roots of Fe-sufficient plants and from 8.75 to 19.89 μmol g FW-1 of Fe-limited plants after exposure to eCO2. In leaves, sugar concentration increased from 33.62 to 52.22 μmol g FW-1 and from 34.80 to 46.70 μmol g FW-1 in Fe-sufficient and Fe-limited conditions, respectively, under eCO2. However, Fe-limitation decreases photosynthesis and biomass. Pathway enrichment analysis showed that cell wall organization, glutathione metabolism, photosynthesis, stress-related proteins, and biosynthesis of secondary compounds changed in root tissues to cope with Fe-stress. Moreover, under eCO2, at sufficient or limited Fe supply, it was shown an increase in the abundance of proteins involved in glycolysis, starch and sucrose metabolism, biosynthesis of plant hormones gibberellins, and decreased levels of protein biosynthesis. Our results revealed that proteins and metabolic pathways related to Fe-limitation changed the effects of eCO2 and negatively impacted soybean production.

MbHY5-MbYSL7 mediates chlorophyll synthesis and iron transport under iron deficiency in Malus baccata.

Iron (Fe) plays an important role in cellular respiration and catalytic reactions of metalloproteins in plants and animals. Plants maintain iron homeostasis through absorption, translocation, storage, and compartmentalization of iron via a cooperative regulative network. Here, we showed different physiological characteristics in the leaves and roots of Malus baccata under Fe sufficiency and Fe deficiency conditions and propose that MbHY5 (elongated hypocotyl 5), an important transcription factor for its function in photomorphogenesis, participated in Fe deficiency response in both the leaves and roots of M. baccata. The gene co-expression network showed that MbHY5 was involved in the regulation of chlorophyll synthesis and Fe transport pathway under Fe-limiting conditions. Specifically, we found that Fe deficiency induced the expression of MbYSL7 in root, which was positively regulated by MbHY5. Overexpressing or silencing MbYSL7 influenced the expression of MbHY5 in M. baccata.

Iron redistribution induces oxidative burst and resistance in maize against Curvularia lunata.

ΔClnps6 induced iron redistribution in maize B73 leaf cells and resulted in reactive oxygen species (ROS) burst to enhance plant resistance against Curvularia lunata. Iron is an indispensable co-factor of various crucial enzymes that are involved in cellular metabolic processes and energy metabolism in eukaryotes. For this reason, plants and pathogens compete for iron to maintain their iron homeostasis, respectively. In our previous study, ΔClnps6, the extracellular siderophore biosynthesis deletion mutant of Curvularia lunata, was sensitive to exogenous hydrogen peroxide and virulence reduction. However, the mechanism was not studied. Here, we report that maize B73 displayed highly resistance to ΔClnps6. The plants recruited more iron at cell wall appositions (CWAs) to cause ROS bursts. Intracellular iron deficiency induced by iron redistribution originated form up-regulated expression of genes involved in intracellular iron consumption in leaves and absorption in roots. The RNA-sequencing data also showed that the expression of respiratory burst oxidase homologue (ZmRBOH4) and NADP-dependent malic enzyme 4 (ZmNADP-ME4) involved in ROS production was up-regulated in maize B73 after ΔClnps6 infection. Simultaneously, jasmonic acid (JA) biosynthesis genes lipoxygenase (ZmLOX), allene oxide synthase (ZmAOS), GA degradation gene gibberellin 2-beta-dioxygenase (ZmGA2OX6) and ABA degradation genes abscisic acid hydroxylase (ZmABH1, ZmABH2) involved in iron homeostasis were up-regulated expression. Ferritin1 (ZmFER1) positive regulated maize resistance against C. lunata via ROS burst under Fe-limiting conditions. Overall, our results showed that iron played vital roles in activating maize resistance in B73-C. lunata interaction.

Glutathione regulates transcriptional activation of iron transporters via S-nitrosylation of bHLH factors to modulate subcellular iron homoeostasis.

Glutathione (GSH) is known to regulate iron (Fe) deficiency response in plants but its involvement in modulating subcellular Fe homoeostasis remains elusive. In this study, we report that the GSH-deficient mutants, cad2-1 and pad2-1 displayed increased sensitivity to Fe deficiency with significant downregulation of the vacuolar Fe exporters, AtNRAMP3 and AtNRAMP4, and the chloroplast Fe importer, AtPIC1. Moreover, the pad2-1 mutant accumulated higher Fe levels in vacuoles but lower Fe levels in chloroplasts compared to wild type (Columbia ecotype [Col-0]) under Fe limited conditions. Exogenous GSH treatment enhanced chloroplast Fe contents in Col-0 but failed to do so in the nramp3nramp4 double mutants demonstrating that GSH plays a role in modulating subcellular Fe homoeostasis. Pharmacological experiments, mutant analysis, and promoter assays revealed that this regulation involves the transcriptional activation of Fe transporter genes by a GSH-S-nitrosoglutathione(GSNO) module. The Fe responsive bHLHtranscription factors (TFs), AtbHLH29, AtbHLH38, and AtbHLH101 were found to interact with the promoters of these genes, which were, in turn, activated via S-nitrosylation (SNO). Taken together, the present study highlights the role of the GSH-GSNO module in regulating subcellular Fe homoeostasis by transcriptional activation of the Fe transporters AtNRAMP3, AtNRAMP4, and AtPIC1 via SNO of bHLH TFs during Fe deficiency.

Host Genotype and Edaphic Factors Cumulatively Influence the Occurrence of Siderophore-producing Bacteria Associated with Rice (Oryza sativa L.)

Seed-borne rice endophytes are capable of disseminating into host plant tissues as well as to their rhizosphere. Here, we investigated the occurrence of siderophore-producing bacteria (SPB) in the seed endospheres of two distinct rice (Oryza sativa L.) cultivars, TK8 (ssp. japonica) and TCN1 (ssp. indica), and their dissemination into the rhizospheres through culture-dependent methods. Their patterns of occurrence in the rhizospheres as well as in the root and shoot tissues of 30 day-old cultivars grown in three different kinds of soils were tested. The significance of SPB on Fe sequestration of TCN1 was studied using Enterobacter sp. LS-756. TK8 seeds were found to be not only abundant in endopsheric SPB (> 10-fold), but also exhibited enhanced SPB dissemination into the rhizosphere (1.3-fold) as compared to TCN1. The proportion of endophytic SPB was consistently higher in roots than in shoots, and it was found to decline with decreasing soil pH. A similar declining trend was further evident through the analysis of SPB composition in the rhizospheric and bulk soils. LS-756-inoculated TCN1 seedlings under low availability of Fe showed 32%, 178%, and 368% increases in Fe, chlorophyll, and chlorophyll b contents as compared to the uninoculated controls. Thus, the occurrence of seed-borne endophytic SPB and their dissemination into the rhizosphere vary significantly according to the rice genotype. Higher co-occurrence of SPB in the rhizosphere and internal root tissues of rice plants grown under Fe-limited conditions and the enhanced Fe uptake due to SPB inoculation substantiated their potential involvement in Fe sequestration.

Functional characterization of a high-affinity iron transporter (PiFTR) from the endophytic fungus Piriformospora indica and its role in plant growth and development.

Iron (Fe) is a micronutrient required for plant growth and development; however, most Fe forms in soil are not readily available to plants, resulting in low Fecontents in plants and, thereby, causing Fe deficiency in humans. Biofortification through plant-fungal co-cultivation might be a sustainable approach to increase crop Fe contents. Therefore, we aimed to examine the role of a Piriformospora indica Fe transporter on rice Fe uptake under low Fe conditions. A high-affinity Fe transporter (PiFTR) from P. indica was identified and functionally characterized. PiFTR fulfilled all criteria expected of a functional Fe transporter under Fe-limited conditions. Additionally, PiFTR expression was induced when P. indica was grown under low Fe conditions, and PiFTR complemented a yeast mutant lacking Fe transport. A knockdown (KD) P. indica strain was created via RNA interference to understand the physiological role of PiFTR. We observed that the KD-PiFTR-P. indica strain transported a significantly lower amount of Feto colonized rice (Oryza sativa) than the wild type(WT) P. indica. WT P. indica-colonized rice plants were healthier and performed significantly better than KD-PiFTR-P. indica-colonized rice plants.Our study offers potential avenues for an agronomically sound amelioration of plant growth in low Fe environments.

Interplay between Coumarin Accumulation, Iron Deficiency and Plant Resistance to Dickeya spp.

Coumarins belong to a group of secondary metabolites well known for their high biological activities including antibacterial and antifungal properties. Recently, an important role of coumarins in plant resistance to pathogens and their release into the rhizosphere upon pathogen infection was discovered. It is also well documented that coumarins play a crucial role in the Arabidopsis thaliana growth under Fe-limited conditions. However, the mechanisms underlying interplay between plant resistance, accumulation of coumarins and Fe status, remain largely unknown. In this work, we investigated the effect of both mentioned factors on the disease severity using the model system of Arabidopsis/Dickeya spp. molecular interactions. We evaluated the disease symptoms in Arabidopsis plants, wild-type Col-0 and its mutants defective in coumarin accumulation, grown in hydroponic cultures with contrasting Fe regimes and in soil mixes. Under all tested conditions, Arabidopsis plants inoculated with Dickeya solani IFB0099 strain developed more severe disease symptoms compared to lines inoculated with Dickeya dadantii 3937. We also showed that the expression of genes encoding plant stress markers were strongly affected by D. solani IFB0099 infection. Interestingly, the response of plants to D. dadantii 3937 infection was genotype-dependent in Fe-deficient hydroponic solution.

Iron and sulfur isotopes of sulfides from the Wocan hydrothermal field on the Carlsberg Ridge, Indian Ocean

The basalt-hosted Wocan hydrothermal field (WHF) was identified in 2013 during Chinese Dayang 28th cruise, which is located on the slow-spreading Carlsberg Ridge, northwest Indian Ocean. Few investigation has been carried out on the hydrothermal activities and associated sulfide deposit in this area. Herein, we report Fe and S isotope compositions of sulfide minerals including chalcopyrite, bornite, pyrite and sphalerite collected from the WHF. The results showed that all sulfide minerals display small δ34S variations (2.44‰ to 5.37‰, with an average of 4.00‰), and a wide δ56Fe range (−2.05‰ to 0.10‰, with an average of −0.92‰). Sulfur and iron isotopes in paired pyrite and chalcopyrite minerals reveal that it did not reach the equilibrium fractionation during pyrite precipitation, and support that the FeS precipitates first during the pyrite formation. The δ56Fe values of chalcopyrite and concurrent bornite are characterized by a well-defined negative correlation, which may be related to the repeated dissolution-reprecipitation of these copper-bearing sulfides under Fe-limited conditions. Most pyrites from the WHF demonstrate a positive correlation between δ56Fe and δ34S values, in accord with those from other hydrothermal fields, which can be explained by the temperature gradient and similar degrees of isotope exchange in the stable hydrothermal system. Conversely, the δ56Fe and δ34S values of chalcopyrite exhibit a slightly negative correlation, in which the temperature gradient can explain the negative correlation, but cannot cover the wide ranges of δ56Fe and δ34S values. More work on fractionation parameters and chalcopyrite formation mechanisms is still needed. In summary, the coupling of Fe and S isotopes is a useful tool in helping to delineate processes of sulfide mineralization in hydrothermal systems

Iron limitation – A perspective on a growth-restricted cultivation strategy for a H2 production system using the diazotrophic cyanobacterium Nostoc PCC 7120 ΔhupW

H2 is a promising carbon-free alternative to fossil fuels, and can be photoautotrophically produced by cyanobacteria. Despite efforts to increase cyanobacterial H2 production by genetic engineering and technical optimization the yield remains economically non-viable. Recent studies have therefore demanded the impairment of growth to reallocate more carbohydrates into biofuel biosynthesis, and nutrient limitation may be a strategy to achieve that. Here we show that limitation of Fe in the filamentous and heterocystous mutant strain Nostoc PCC 7120 ΔhupW resulted in a 5.3-fold lower chlorophyll a concentration as compared to the controls during four-week cultivation. The Fe-limited filaments contained a heterocyst frequency at ~6% and a 4-fold higher specific carbohydrate concentration as compared to the control cultures. We discuss whether Fe-limiting conditions leading to an accumulation of carbon storage compounds could be used as a heterocyst based H2 production system.

The interplay between iron limitation, light and carbon in the proteorhodopsin-containing Photobacterium angustum S14

Iron (Fe) limitation is known to affect heterotrophic bacteria within the respiratory electron transport chain, therefore strongly impacting the overall intracellular energy production. We investigated whether the gene expression pattern of the light-sensitive proton pump, proteorhodopsin (PR), is influenced by varying light, carbon and Fe concentrations in the marine bacterium Photobacterium angustum S14 and whether PR can alleviate the physiological processes associated with Fe starvation. Our results show that the gene expression of PR increases as cells enter the stationary phase, irrespective of Fe-replete or Fe-limiting conditions. This upregulation is coupled to a reduction in cell size, indicating that PR gene regulation is associated with a specific starvation-stress response. We provide experimental evidence that PR gene expression does not result in an increased growth rate, cell abundance, enhanced survival or ATP concentration within the cell in either Fe-replete or Fe-limiting conditions. However, independent of PR gene expression, the presence of light did influence bacterial growth rates and maximum cell abundances under varying Fe regimes. Our observations support previous results indicating that PR phototrophy seems to play an important role within the stationary phase for several members of the Vibrionaceae family, but that the exact role of PR in Fe limitation remains to be further explored.

Fullerenol increases effectiveness of foliar iron fertilization in iron-deficient cucumber.

The water-soluble fullerenols are novel carbon-based nanomaterials with unique properties, which afford them with wide agricultural applications. Iron (Fe) deficiency is the most common and widespread nutrition disorder affecting plants. Foliar Fe treatments of plants have been carried out with solutions devoid of fullerenol. In this study, the role of fullerenol [C60(OH)22–24] in alleviation of Fe deficiency in Cucumis sativus (a Strategy I plant) via foliar fertilization was investigated. Cucumber plants were grown hydroponically, either with (Fe) or in Fe-free (−Fe) nutrient solution. The following foliar spray treatments were applied: fullerenol at final concentrations of 1 (F1) and 10 (F10) mg L-1; Fe(II)SO4·7H2O; Fe(II)-EDTA (ethylenediaminetetraacetic acid); and Fe(II)-F1 and Fe(II)-F10. The best used compound was a combination of Fe(II)-sulfate with fullerenol, especially Fe-F1. The addition of fullerenol to Fe(II)-sulfate solutions significantly increased leaf-active Fe (extracted by an Fe(II) chelator) and re-greening at the site of application. The fullerenol-induced mutual influences did not appear when fullerenol was sprayed alone, suggesting a beneficial role of Fe(II)–fullerenol interactions in the penetration of Fe(II) in the leaves and re-greening under Fe-limited conditions. The results are of importance to enhancing the potential of foliar Fe fertilization as the commonly used strategy for ameliorating Fe deficiency and improving crop yield and quality.

Copper-only superoxide dismutase enzymes and iron starvation stress in Candida fungal pathogens

Copper (Cu)-only superoxide dismutases (SOD) represent a newly characterized class of extracellular SODs important for virulence of several fungal pathogens. Previous studies of the Cu-only enzyme SOD5 from the opportunistic fungal pathogen Candida albicans have revealed that the active-site structure and Cu binding of SOD5 strongly deviate from those of Cu/Zn-SODs in its animal hosts, making Cu-only SODs a possible target for future antifungal drug design. C. albicans also expresses a Cu-only SOD4 that is highly similar in sequence to SOD5, but is poorly characterized. Here, we compared the biochemical, biophysical, and cell biological properties of C. albicans SOD4 and SOD5. Analyzing the recombinant proteins, we found that, similar to SOD5, Cu-only SOD4 can react with superoxide at rates approaching diffusion limits. Both SODs were monomeric and they exhibited similar binding affinities for their Cu cofactor. In C. albicans cultures, SOD4 and SOD5 were predominantly cell wall proteins. Despite these similarities, the SOD4 and SOD5 genes strongly differed in transcriptional regulation. SOD5 was predominantly induced during hyphal morphogenesis, together with a fungal burst in reactive oxygen species. Conversely, SOD4 expression was specifically up-regulated by iron (Fe) starvation and controlled by the Fe-responsive transcription factor SEF1. Interestingly, Candida tropicalis and the emerging fungal pathogen Candida auris contain a single SOD5-like SOD rather than a pair, and in both fungi, this SOD was induced by Fe starvation. This unexpected link between Fe homeostasis and extracellular Cu-SODs may help many fungi adapt to Fe-limited conditions of their hosts.

Proteomic responses to ocean acidification of the marine diazotroph Trichodesmium under iron-replete and iron-limited conditions.

Growth and dinitrogen (N2) fixation of the globally important diazotrophic cyanobacteria Trichodesmium are often limited by iron (Fe) availability in surface seawaters. To systematically examine the combined effects of Fe limitation and ocean acidification (OA), T. erythraeum strain IMS101 was acclimated to both Fe-replete and Fe-limited concentrations under ambient and acidified conditions. Proteomic analysis showed that OA affected a wider range of proteins under Fe-limited conditions compared to Fe-replete conditions. OA also led to an intensification of Fe deficiency in key cellular processes (e.g., photosystem I and chlorophyll a synthesis) in already Fe-limited T. erythraeum. This is a result of reallocating Fe from these processes to Fe-rich nitrogenase to compensate for the suppressed N2 fixation. To alleviate the Fe shortage, the diazotroph adopts a series of Fe-based economic strategies (e.g., upregulating Fe acquisition systems for organically complexed Fe and particulate Fe, replacing ferredoxin by flavodoxin, and using alternative electron flow pathways to produce ATP). This was more pronounced under Fe-limited-OA conditions than under Fe limitation only. Consequently, OA resulted in a further decrease of N2- and carbon-fixation rates in Fe-limited T. erythraeum. In contrast, Fe-replete T. erythraeum induced photosystem I (PSI) expression to potentially enhance the PSI cyclic flow for ATP production to meet the higher demand for energy to cope with the stress caused by OA. Our study provides mechanistic insight into the holistic response of the globally important N2-fixing marine cyanobacteria Trichodesmium to acidified and Fe-limited conditions of future oceans.
