Current progress towards the metabolic engineering of plant seed oil for hydroxy fatty acids production.

Hydroxy fatty acids produced in plant seed oil are important industrial material. This review focuses on the use of metabolic engineering approaches for the production of hydroxy fatty acids in transgenic plants. Vegetable oil is not only edible but can also be used for industrial purposes. The industrial demand for vegetable oil will increase with the continued depletion of fossil fuels and ensuing environmental issues such as climate change, caused by increased carbon dioxide in the air. Some plants accumulate high levels of unusual fatty acids in their seeds, and these fatty acids (FAs) have properties that make them suitable for industrial applications. Hydroxy fatty acids (HFAs) are some of the most important of these industrial FAs. Castor oil is the conventional source of HFA. However, due to the presence of toxin ricin in its seeds, castor is not cultivated on a large scale. Lesquerella is another HFA accumulator and is currently being developed as a new crop for a safe source of HFAs. The mechanisms of HFA synthesis and accumulation have been extensively studied using castor genes and the model plant Arabidopsis. HFAs accumulated to 17% in the seed oil of Arabidopsis expressing a FA hydroxylase gene from castor (RcFAH12), but its seed oil content and plant growth decreased. When RcFAH12 gene was coexpressed with additional castor gene(s) in Arabidopsis, ~30% HFAs were accumulated and the seed oil content and plant growth was almost restored to the wild-type level. Further advancement of our understanding of pathways, genes and regulatory mechanisms underlying synthesis and accumulation of HFAs is essential to developing and implementing effective genetic approaches for enhancing HFA production in oilseeds.

Reconstitution of EPA and DHA biosynthesis in Arabidopsis: Iterative metabolic engineering for the synthesis of n−3 LC-PUFAs in transgenic plants

An iterative approach to optimising the accumulation of non-native long chain polyunsaturated fatty acids in transgenic plants was undertaken in Arabidopsis thaliana. The contribution of a number of different transgene enzyme activities was systematically determined, as was the contribution of endogenous fatty acid metabolism. Successive iterations were informed by lipidomic analysis of neutral, polar and acyl-CoA pools. This approach allowed for a four-fold improvement on levels previously reported for the accumulation of eicosapentaenoic acid in Arabidopsis seeds and also facilitated the successful engineering of the high value polyunsaturated fatty acid docosahexaenoic acid to 10-fold higher levels. Our studies identify the minimal gene set required to direct the efficient synthesis of these fatty acids in transgenic seed oil.

Enhancing the accumulation of omega-3 long chain polyunsaturated fatty acids in transgenic Arabidopsis thaliana via iterative metabolic engineering and genetic crossing

The synthesis and accumulation of long chain polyunsaturated fatty acids such as eicosapentaenoic acid has previously been demonstrated in the seeds of transgenic plants. However, the obtained levels are relatively low, indicating the need for further studies and the better definition of the interplay between endogenous lipid synthesis and the non-native transgene-encoded activities. In this study we have systematically compared three different transgenic configurations of the biosynthetic pathway for eicosapentaenoic acid, using lipidomic profiling to identify metabolic bottlenecks. We have also used genetic crossing to stack up to ten transgenes in Arabidopsis. These studies indicate several potential approaches to optimize the accumulation of target fatty acids in transgenic plants. Our data show the unexpected channeling of heterologous C20 polyunsaturated fatty acids into minor phospholipid species, and also the apparent negative metabolic regulation of phospholipid-dependent Δ6-desaturases. Collectively, this study confirms the benefits of iterative approaches to metabolic engineering of plant lipid synthesis.

The role of Δ6‐desaturase acyl‐carrier specificity in the efficient synthesis of long‐chain polyunsaturated fatty acids in transgenic plants

The role of acyl-CoA-dependent Δ6-desaturation in the heterologous synthesis of omega-3 long-chain polyunsaturated fatty acids was systematically evaluated in transgenic yeast and Arabidopsis thaliana. The acyl-CoA Δ6-desaturase from the picoalga Ostreococcus tauri and orthologous activities from mouse (Mus musculus) and salmon (Salmo salar) were shown to generate substantial levels of Δ6-desaturated acyl-CoAs, in contrast to the phospholipid-dependent Δ6-desaturases from higher plants that failed to modify this metabolic pool. Transgenic plants expressing the acyl-CoA Δ6-desaturases from either O.tauri or salmon, in conjunction with the two additional activities required for the synthesis of C20 polyunsaturated fatty acids, contained higher levels of eicosapentaenoic acid compared with plants expressing the borage phospholipid-dependent Δ6-desaturase. The use of acyl-CoA-dependent Δ6-desaturases almost completely abolished the accumulation of unwanted biosynthetic intermediates such as γ-linolenic acid in total seed lipids. Expression of acyl-CoA Δ6-desaturases resulted in increased distribution of long-chain polyunsaturated fatty acids in the polar lipids of transgenic plants, reflecting the larger substrate pool available for acylation by enzymes of the Kennedy pathway. Expression of the O.tauriΔ6-desaturase in transgenic Camelina sativa plants also resulted in the accumulation of high levels of Δ6-desaturated fatty acids. This study provides evidence for the efficacy of using acyl-CoA-dependent Δ6-desaturases in the efficient metabolic engineering of transgenic plants with high value traits such as the synthesis of omega-3 LC-PUFAs.

Castor Phospholipid:Diacylglycerol Acyltransferase Facilitates Efficient Metabolism of Hydroxy Fatty Acids in Transgenic Arabidopsis

Producing unusual fatty acids (FAs) in crop plants has been a long-standing goal of green chemistry. However, expression of the enzymes that catalyze the primary synthesis of these unusual FAs in transgenic plants typically results in low levels of the desired FA. For example, seed-specific expression of castor (Ricinus communis) fatty acid hydroxylase (RcFAH) in Arabidopsis (Arabidopsis thaliana) resulted in only 17% hydroxy fatty acids (HFAs) in the seed oil. In order to increase HFA levels, we investigated castor phospholipid:diacylglycerol acyltransferase (PDAT). We cloned cDNAs encoding three putative PDAT enzymes from a castor seed cDNA library and coexpressed them with RcFAH12. One isoform, RcPDAT1A, increased HFA levels to 27%. Analysis of HFA-triacylglycerol molecular species and regiochemistry, along with analysis of the HFA content of phosphatidylcholine, indicates that RcPDAT1A functions as a PDAT in vivo. Expression of RcFAH12 alone leads to a significant decrease in FA content of seeds. Coexpression of RcPDAT1A and RcDGAT2 (for diacylglycerol acyltransferase 2) with RcFAH12 restored FA levels to nearly wild-type levels, and this was accompanied by a major increase in the mass of HFAs accumulating in the seeds. We show the usefulness of RcPDAT1A for engineering plants with high levels of HFAs and alleviating bottlenecks due to the production of unusual FAs in transgenic oilseeds.

식물 종자오일의 지방산은 인간에 필수 지방산을 공급하는 식용 및 생필품 생산에 필요한 다양한 산업원료로 사용된다. 식물 오일의 식용 및 산업용 적합성과 경제성을 극대화하기 위해 유전공학에 의한 종자오일의 양과 지방산 조성 변형을 위한 대사조절연구가 계속 진행되고 있다. 하지만 식물에 일반적으로 존재하지 않는, 산업적으로 유용한 특이지방산의 합성과 종자오일로의 축적은 한계가 있음이 알려져 있다. 그 이유는 재배가 용이하며 생산성이 높은 오일식물의 acyltransferase가 특이지방산에 대한 기질특이성이 떨어지며 또한 특이지방산에 대한 세포막지질에서 종자오일로 전환시키는 편집기작 (editing mechanism)이 없기 때문으로 사료된다. 최근에 모델식물의 종자오일의 축적에 관여하는 acyltransferase에 관한 유전자들이 클로닝되었고, 특이지방산이 합성되는 인지질에서의 편집기작에 관여하는 acyltransferase 유전자들이 밝혀져 이들 유전자들의 정보를 이용하여 특이지방산을 효과적으로 생산.증진할 수 있는 기술이 개발될 수 있을 것으로 기대한다. 피마자오일의 주성분인 산업용 특이지방산인 리시놀레인 지방산을 오일식물에서 생산하기 위해 이에 관여할 것으로 추정되는 11개의 acyltransferase 유전자를 피마자 유전체 데이터베이스에서 존재함을 확인하였다. 이들 유전자들의 도입에 의해 형질전환 식물이 갖고 있지 않은 리시놀레인산에 대한 기질 특이성을 부여하여 종자오일 내의 특이지방산의 생산을 증가시킬 것으로 기대된다. Fatty acids in seed oil from plants are essential for human nutrients and have been used for industrial purpose. The growing demands of seed oil as food resources and feedstocks for industrial uses have attempted to modify fatty acid composition and to increase oil content in transgenic plants. However, production of unusual fatty acids in transgenic plants are limited, which is not synthesized the level same as original plants. This bottleneck was common for production of several unusual fatty acids in transgenic plants and suggests that there is different for substrate preference in oil metabolic pathway enzymes between host oil plants and original wild plants. Review of acyltransferases involved in acyl-editing and seed oil accumulation of oil plant and wild-plant producing unusual fatty acids will design strategies to maximize the production of unusual fatty acids in transgenic plants. In here, we identified eleven acyltransferase genes in castor based on sequence homology, which will be useful to increase hydroxy unusual fatty acids in transgenic plants.

The Production of Unusual Fatty Acids in Transgenic Plants

The ability to genetically engineer plants has facilitated the generation of oilseeds synthesizing non-native fatty acids. Two particular classes of fatty acids are considered in this review. First, so-called industrial fatty acids, which usually contain functional groups such as hydroxyl, epoxy, or acetylenic bonds, and second, very long chain polyunsaturated fatty acids normally found in fish oils and marine microorganisms. For industrial fatty acids, there has been limited progress toward obtaining high-level accumulation of these products in transgenic plants. For very long chain polyunsaturated fatty acids, although they have a much more complex biosynthesis, accumulation of some target fatty acids has been remarkably successful. In this review, we consider the probable factors responsible for these different outcomes, as well as the potential for further optimization of the transgenic production of unusual fatty acids in transgenic plants.

Relief for fish stocks: oceanic fatty acids in transgenic oilseeds

Three recent reports (Baoxiu Qi et al., Amine Abbadi et al. and Anthony J. Kinney et al.) describe the production of very long-chain polyunsaturated fatty acids in transgenic plants. This might lead to a sustainable source of these valuable fatty acids for use in human food and animal feed. At present they are mainly available via consumption of fish, which is a limited and endangered resource.

Progress toward the production of long‐chain polyunsaturated fatty acids in transgenic plants

Long-chain PUFA such as eicosapentaenoic and docosahexaenoic acids are prevalent in fish oils, and these compounds have been demonstrated to play important roles in human health and nutrition. In particular, these n-3/omega-3 long-chain PUFA provide protection from cardiovascular disease and a collection of symptoms (termed metabolic syndrome) associated with progression toward type 2 diabetes and obesity. Within Western populations, a large increase in the occurrence of these conditions represents a major public health concern. Unfortunately, both marine fish stocks and (consequentially) consumption of fish oils are in steep decline, limiting the protective role of long-chain PUFA in human health. One alternative approach to the provision of these health-beneficial FA is via their synthesis in transgenic plants. This review will describe recent advances in the production of transgenic plant oils nutritionally enhanced to produce long-chain PUFA.

The production of long chain polyunsaturated fatty acids in transgenic plants by reverse-engineering

The nutritional importance of long chain polyunsaturated fatty acids (LC-PUFAs) is now well-established, with these lipids playing roles in human development and the prevention of various diseases. In particular, n-3 polyunsaturated fatty acids are protective against cardiovascular disease (CVD) and risks associated with metabolic syndrome. In view of the decline in marine fish stocks, which represent the predominant natural reserves of n-3 long chain polyunsaturates, alternative sources are urgently required. One approach may be to express the LC-PUFA-biosynthetic pathway in transgenic plants. Recent progress in validating this approach has now emerged, demonstrating the feasibility of using transgenic plants to synthesise these important human nutrients.

The production of long chain polyunsaturated fatty acids in transgenic plants.

Very long chain polyunsaturated fatty acids (VLC-PUFAs) are valuable commodities currently sourced either from vulnerable natural (marine) stocks or via expensive microbial fermentation. The advent of plant genetic engineering has raised the possibility of producing PUFAs in transgenic plants. This technology allows the transfer of a particular gene or trait into a non-native (heterologous) host, and has the potential to revolutionize modern agriculture. In the case of PUFAs, it is possible to envisage “designer” oilseeds in which the crop plant has been engineered to make very long chain PUFAs, even though these fatty acids do usually occur in higher plant oil seeds (1). Considering the cost-benefits of genetically-engineered oilseeds as an alternative supply to conventional sources, it is perhaps unsurprising that considerable resources have been focused on achieving this goal. We would therefore like to review the progress in this field, focusing on the underpinning molecular biology which is enabling this (potential) biotechnology revolution.

What limits production of unusual monoenoic fatty acids in transgenic plants?

Unusual monounsaturated fatty acids are major constituents (greater than 80%) in seeds of Coriandrum sativum L. (coriander) and Thunbergia alata Bojer, as well as in glandular trichomes (greater than 80% derived products) of Pelargonium x hortorum (geranium). These diverged fatty acid structures are produced via distinct plastidial acyl-acyl carrier protein (ACP) desaturases. When expressed in Arabidopsis thaliana (L.) Heynh. under strong seed-specific promoters the unusual acyl-ACP desaturases resulted in accumulation of unusual monoene fatty acids at 1-15% of seed fatty acid mass. In this study, we have examined several factors that potentially limit higher production of unusual monoenes in transgenic oilseeds. (i) Immunoblots indicated that the introduced desaturases were expressed at levels equivalent to or higher than the endogenous delta9 18:0-ACP desaturase. However, the level of unusual fatty acid produced in transgenic plants was not correlated with the level of desaturase expression. (ii) The unusual desaturases were expressed in several backgrounds, including antisense 18:0-ACP desaturase plants, in fab1 mutants, and co-expressed with specialized ACP or ferredoxin isoforms. None of these experiments led to high production of expected products. (iii) No evidence was found for degradation of the unusual fatty acids during seed development. (iv) Petroselinic acid added to developing seeds was incorporated into triacylglycerol as readily as oleic acid, suggesting no major barriers to its metabolism by enzymes of glycerolipid assembly. (v) In vitro and in situ assay of acyl-ACP desaturases revealed a large discrepancy of activity when comparing unusual acyl-ACP desaturases with the endogenous delta9 18:0-ACP desaturase. The combined results, coupled with the sensitivity of acyl-ACP desaturase activity to centrifugation and low salt or detergent suggests low production of unusual monoenes in transgenic plants may be due to the lack of, or incorrect assemble of, a necessary multi-component enzyme association.

Towards the production of pharmaceutical fatty acids in transgenic plants

AbstractRecent advances in plant molecular biology have facilitated the introduction of novel traits into a range of plant species. It is now possible to modify the fatty acid composition of plant oilseeds, which opens up the possibility of improving the nutritional qualities of plant oils. The fatty acid composition of an oilseed determines its end use and therefore its commercial value. Value can be added to certain crop species by manipulations which would allow production of rare and valuable oils. Genetic engineering also provides novel opportunities for the production of pharmaceutical fatty acids such as γ‐linolenic acid and arachidonic acid which are important for human health and nutrition.© 2001 Society of Chemical Industry

Expression of a cyanobacterial delta 6-desaturase gene results in gamma-linolenic acid production in transgenic plants.

Gamma-linolenic acid (GLA), a nutritionally important fatty acid in human and animal diets, is not produced in oil seed crops. Many oil seed plants, however, produce significant quantities of linoleic acid, a fatty acid that could be converted to GLA by the enzyme delta 6-desaturase if it were present. As a first step to producing GLA in oil seed crops, we have cloned a cyanobacterial delta 6-desaturase gene. Expression of this gene in transgenic tobacco resulted in GLA accumulation. Octadecatetraenoic acid, a highly unsaturated, industrially important fatty acid, was also found in transgenic tobacco plants expressing the cyanobacterial delta 6-desaturase. This is the first example of engineering the production of 'novel' polyunsaturated fatty acids in transgenic plants.
