Optimization of supercritical CO2 fluid extraction process, component analysis, and antioxidant spectral activity correlation of Huoluo Xiaoling Dan

ABSTRACT This study elucidated the optimization conditions, GC-MS analysis conditions, total antioxidant activity, and components closely related to antioxidant activity for supercritical extraction of active ingredients in Huoluo Xiaoling Dan (HLXLD). The yield, octyl acetate content, and total antioxidant activity of HLXLD under different conditions were obtained through orthogonal experiments, and the optimal supercritical extraction conditions were determined to be 45°C, 25 MPa, and 4 h. The supercritical extract was subjected to GC-MS analysis and matched with the WILEY and NIST databases, resulting in the qualitative identification of 37 chemical components with a matching degree above 60. The similarity evaluation system of traditional Chinese medicine fingerprint was used to evaluate all orthogonal experimental samples, with a similarity ranging from 0.522 to 0.989. Thirteen common characteristic peaks were selected, and the quantification of these peaks was performed using peak area normalization method. The combination of the content of common characteristic peaks in different samples and their total antioxidant activity was analyzed using multivariate statistical methods such as principal component analysis and multiple regression equation. It was found that five components, F3, F2, F5, F7 and F12, among the common characteristic peaks were closely related to antioxidant activity.

A high-permeability method for extracting purple yam saponins based on ultrasonic-assisted natural deep eutectic solvent

The extraction of active ingredients from traditional Chinese medicine has received considerable attentions. In this study, 16 kinds of natural deep eutectic solvent (NADES) with ultrasonic were selected to extract saponins from purple yam root and the extraction mechanism was investigated. The results showed that chloride/acrylic acid (1:2; n/n) had the highest extraction yield for saponins. The optimal extraction process parameters were 24% water content, 20 mL/g liquid-solid ratio, and ultrasonic extraction for 85 min (81 °C, 600 W). The extraction rate (ER) of purple yam saponins was 0.935%, close to the fitted result of 96.5 mg/g. Molecular dynamics simulations and FT-IR results showed that the NADES may extract the saponin constituents from purple yam through hydrogen bonding. Compared with traditional extraction methods and molecularly imprinted polymer methods, NADES has a higher ER and lower cost (1.53 $/g), which provides a reference for subsequent industrial quantitative production.

Enzymatic and ultrasound assisted β-cyclodextrin extraction of active ingredients from Forsythia suspensa and their antioxidant and anti-inflammatory activities

With the proposal of the 2030 Agenda for Sustainable Development, the Chinese medicine extraction technology has been innovatively improved to prioritize low energy consumption, sustainability, and minimized organic solvent utilization. Forsythia suspensa (FS) possesses favorable pharmacological properties and is extensively utilized in traditional Chinese medicine. However, due to the limitations of the composition and extraction methods, its potential has not been fully developed. Thus, a combination of ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and β-cyclodextrin extraction (β-CDE) was employed to isolate and purify rutin, phillyrin, and forsythoside A from FS. The results demonstrated that the efficiency of extracting enzymatic and ultrasound assisted β-cyclodextrin extraction (EUA-β-CDE) was highly influenced by the temperature and duration of hydrolysis, as well as the duration of the extraction process. According to the results of the single-factor experiment, Box-Behnken design (BBD) in Response surface method (RSM) was used to optimize the experimental parameters to achieve the maximum comprehensive evaluation value (CEV) value. The EUA-β-CDE compared with other extraction methods, has good extraction effect and low energy consumption by high performance liquid chromatography (HPLC), scanning electron microscopy (SEM), calculation of power consumption and CO2 emission The EUA-β-CDE compared with other extraction methods, has good extraction effect and low energy consumption by HPLC, SEM, calculation of power consumption and CO2 emission. Then, the structural characteristics of EUA-β-CDE of FS extract had significant interaction with β-CD by Fourier infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC). In addition, EUA-β-CDE extract has good antioxidant and anti-inflammatory activities. The establishment of EUA-β-CDE of FS provides a new idea for the development and application of other sustainable extraction methods of traditional Chinese medicine.

The anti-depression effect and potential mechanism of the petroleum ether fraction of CDB: Integrated network pharmacology and metabolomics

The combination of Chaidangbo (CDB) is an antidepressant traditional Chinese medicine (TCM) prescription simplified by Xiaoyaosan (a classic antidepressant TCM prescription) through dismantling research, which has the effect of dispersing stagnated liver qi and nourishing blood in TCM theory. Although the antidepressant effect of CBD has been confirmed in animal studies, the material basis and possible molecular mechanism for antidepressant activity in CBD have not been clearly elucidated. Herein, we investigated the effects and potential mechanisms of CDB antidepressant fraction (petroleum ether fraction of CDB, PEFC) on chronic unpredictable mild stress (CUMS)-induced depression-like behavior in mice using network pharmacology and metabolomics. First, a UPLC-QE/MS was employed to identify the components of PEFC. To extract active ingredients, SwissADME screening was used to the real PEFC components that were found. Potential PEFC antidepressant targets were predicted based on a network pharmacology approach, and a pathway enrichment analysis was performed for the predicted targets. Afterward, a CUMS mouse depression model was established and LC-MS-based untargeted hippocampal metabolomics was performed to identify differential metabolites, and related metabolic pathways. Finally, the protein expressions in mouse hippocampi were determined by Western blot to validate the network pharmacology and metabolomics deduction. A total of 16 active compounds were screened in SwissADME that acted on 73 core targets of depression, including STAT3, MAPKs, and NR3C1; KEGG enrichment analysis showed that PEFC modulated signaling pathways such as PI3K-Akt signaling pathway, endocrine resistance, and MAPK to exert antidepressant effects. PEFC significantly reversed abnormalities of hippocampus metabolites in CUMS mice, mainly affecting the synthesis and metabolism of glycine, serine, and threonine, impacting catecholamine transfer and cholinergic synapses and regulating the activity of the mTOR signaling pathway. Furthermore, Western blot analysis confirmed that PEFC significantly influenced the main protein levels of the PI3K/Akt/mTOR signaling pathways in the hippocampus of mice subjected to CUMS. This study integrated metabolomics, network pharmacology and biological verification to explore the potential mechanism of PEFC in treating depression, which is related to the regulation of amino acid metabolism dysfunction and the activation of PI3K/Akt/mTOR signaling pathways in the hippocampus. The comprehensive strategy also provided a reasonable way for unveiling the pharmacodynamic mechanisms of multi-components, multi-targets, and multi-pathways in TCM with antidepressant effect.

The Glycyrrhiza glabra L. crude extract alleviates lipid accumulation in NAFLD by activating Nrf2 and promoting autophagy

Glycyrrhiza glabra L. is a common heritage herbal medicine rich in antioxidant compounds such as flavonoids and triterpenoids. Non-alcoholic fatty liver disease has been the global prevalent chronic liver disorder. To investigate the curative role and mechanism of the Glycyrrhiza glabra L. crude extract on NAFLD, in vitro, we used oleic acid, palmitic acid, ML-385 and 3-MA to stimulate HepG2 cells. In vivo, we used Tyloxapol to inject mice intraperitoneally to establish an acute model, and high-fat diet to feed wild-type, Nrf2-/- and ATG5flox/flox mice to establish a chronic model. We used high performance liquid chromatography to analyse the content of major bioactive components in the Glycyrrhiza glabra L. crude extract. Our study found the Glycyrrhiza glabra L. crude extract could effectively alleviate lipid accumulation in non-alcoholic fatty liver disease, activate Nrf2 and promote autophagy. However, this alleviating effect disappeared when the Nrf2 and autophagy were inhibited separately. Our study demonstrates that crude extraction of active ingredients from traditional herbal medicines may be a potentially inexpensive, rapid and safe approach to alleviate lipid accumulation in non-alcoholic fatty liver disease.

Mass transfer performance of pulsed disk and doughnut column (PDDC) in the extraction of active ingredients in peppermint extract obtained by hot percolation

AbstractThe active ingredients in peppermint are widely used in the pharmaceutical, food, cosmetic, and other industries. The traditional methods for producing peppermint extract through hydrodistillation (HD) and percolation are long process with low efficiency. The aim of this study is the improvement of the extraction performance and the efficiency of the extract obtained from hot percolation via pulsed disk and doughnut columns (PDDC), which is one of the most important pieces of liquid–liquid extraction (LLE) equipment. Peppermint extract was used as the continuous phase in the PDDC. n‐Hexane was considered as the dispersed phase in this column. In this study, the effects of three operating parameters were investigated, including the pulse intensity, continuous phase height, and dispersed phase flow rate, on the extraction efficiency of active ingredients in peppermint. The response surface method (RSM) was used to investigate the effect of these parameters on the extraction efficiency. The efficiency of this column in the extracting of active ingredients from peppermint was measured by UV–vis spectrophotometry, via the absorption rate of the main active ingredients of peppermint (menthol) in the samples, and the efficiency was 75% on average. The experimental and theorical of dispersed phase mass transfer coefficient were evaluated and compared. The results showed that the PDDC improved the extraction and mass transfer. By increasing the flow rate of the dispersed phase and the height of the continuous phase, the extraction efficiency and the mass transfer coefficient were increased.

The Conversion of Pistachio and Walnut Shell Waste into Valuable Components with Subcritical Water

Pistachio and walnut shells accumulate in large quantities as waste during food processing and represent a promising lignocellulosic biomass for the extraction of valuable components. Subcritical water technology was used as an environmentally friendly technique to study the extraction of active ingredients and other valuable degradation products from walnut and pistachio waste. Subcritical water extraction (SWE) was carried out under different process conditions (temperature (150–300 °C) and short reaction times (15–60 min)) and compared with conventional extraction using different organic solvents (acetone, 50% acetone and ethanol). The extracts obtained from pistachio and walnut shell waste are rich in various bioactive and valuable components. The highest contents of total phenols (127.08 mg GA/g extract at 300 °C for 15 min, from walnut shells), total flavonoids (10.18 mg QU/g extract at 200 °C for 60 min, from pistachio shells), total carbohydrates (602.14 mg TCH/g extract at 200 °C for 60 min, from walnut shells) and antioxidant activity (91% at 300 °C, for 60 min, from pistachio shells) were determined when the extracts were obtained via subcritical water. High contents of total phenols (up to 86.17 mg GA/g extract) were also determined in the conventional extracts obtained with ethanol. Using the HPLC method, sugars and their valuable derivatives were determined in the extracts, with glucose, fructose, furfurals (5-hydroxymethylfurfural (5-HMF) and furfural) and levulinic acid being the most abundant in the extracts obtained by subcritical water. The results show that subcritical water technology enables better exploitation of biowaste materials than conventional extraction methods with organic solvents, as it provides a higher yields of bioactive components such as phenolic compounds and thus extracts with high antioxidant activity, while at the same time producing degradation products that are valuable secondary raw materials.

Unlocking the Medicinal Potential of Plant-Derived Extracellular Vesicles: current Progress and Future Perspectives.

Botanical preparations for herbal medicine have received more and more attention from drug researchers, and the extraction of active ingredients and their successful clinical application have become an important direction of drug research in major pharmaceutical companies, but the complexity of extracts, multiple side effects, and significant individual differences have brought many difficulties to the clinical application of herbal preparations. It is noteworthy that extracellular vesicles as active biomolecules extracted from medicinal plants are believed to be useful for the treatment of a variety of diseases, including cancer, inflammation, regenerative-restorative and degenerative diseases, which may provide a new direction for the clinical utilization of herbal preparations. In this review, we sort out recent advances in medicinal plant extracellular vesicles and discuss their potential as disease therapeutics. Finally, future challenges and research directions for the clinical translation of medicinal plant extracellular vesicles are also discussed, and we expect that continued development based on medicinal plant extracellular vesicles will facilitate the clinical application of herbal preparations.

Research Progress on the Antitumor Molecular Mechanism of Ginsenoside Rh2.

Cancer has evolved into a substantial public health concern as the second-leading cause of mortality globally. Radiotherapy and chemotherapy have been the two most widely used cancer therapies in recent years; however, both have drawbacks. Therefore, the focus has shifted to the creation of herbal medicines, the extraction of active ingredients, replacement therapy, and the adverse effects of these medications. Ginsenoside Rh2, which is extracted from ginseng, has been identified in many cancer cells. The immune system of the body is strengthened by ginsenoside Rh2, which can also cause the proliferation, death, and differentiation of tumor cells through various pathways. For instance, it inhibits the expression of the NF-[Formula: see text]B signaling pathway and induces cell apoptosis, affects the expression levels of mitochondrial apoptosis proteins Bcl-2 and Bax, and cooperates with the PD-1 blockade to reactivate T cells to promote an antitumor immune response. Furthermore, ginsenosides Rh2 has the effect of reversing the toxic effect of chemotherapy drugs on normal cells, reducing myocardial damage, and relieving bone marrow function suppression. For clinical applications, it is mainly used as an adjuvant drug for preoperative neoadjuvant chemotherapy, postoperative adjuvant chemotherapy, and rescue treatment of advanced cancer. This paper summarizes the pharmacological action and mechanism of ginsenosides Rh2 in all kinds of cancer and looks forward to its future development and application.

ABSTRACT As a traditional Chinese medicine, Angelica dahurica has significant medicinal and nutritional value. To optimize the extraction of flavonoids from Angelica dahurica and enhance its medicinal value, this study was conducted to optimize the process using response surface methodology and investigated the effects of ultrasonication time, ethanol concentration, and solid-liquid ratio on the extraction rate of flavonoids in Angelica dahurica based on single-factor tests. The ultrasound-assisted extraction process can significantly improve the extraction rate of flavonoids in Angelica dahurica, and the flavonoids exhibit antioxidant capacities, which provides a practical basis for the extraction of active ingredients from Angelica dahurica.

Efficient extraction and optimization procedures of polyphyllins from Paris polyphylla var chinensis by deep eutectic solvent coupled with ultrasonic-assisted extraction

As a rare and valuable Chinese herb, Paris polyphylla var chinensis has good pharmacological effects, among which Polyphyllins (PNs) are widely studied as the main active ingredients. Recently, alternative solvents namely, ionic liquids and deep eutectic solvents (DESs) have been used to extract active ingredients with remarkable efficiency. Compared to ionic liquids, deep eutectic solvents (DESs) were found to be effective with the added advantages of ease of production, economy and environment-protecting solvents to substitute common organic ones and have gradually become the research focus of green chemistry. In the present research, twelve varieties of DESs were tailored and PNs were successfully extracted by ultrasound assistance. Among them, combined with single-factor experiments and optimized using response surface methodology (RSM) and artificial neural network (ANN), a liquid–solid ratio of 41.72 mL/g, extraction temperature of 55.97 °C, extraction time of 30.21 min were the optimum extraction method with the overall PNs extraction yield of 52.56 mg/g. Moreover, DESs displayed superior extraction performance in PNs extraction than those traditional solvents. Overall, this study implied that DESs were more conducive to the full utilization and development of the resources of Paris polyphylla, which also confirmed the ability of DESs to effectively extract biologically active ingredients for next step adhibition.

Screening the Extract of Laportea bulbifera (Sieb. et Zucc.) Wedd. Based on Active Component Content, Its Antioxidant Capacity and Exploration of Hepatoprotective Activity in Rats.

Laportea bulbifera (Sieb. et Zucc.) Wedd., a plant with a long history of medicinal use, possesses uncertainly defined medicament portions while its antioxidant capacity remains largely unexplored. To gain a better understanding of its medicinal value, this study focused on investigating the Laportea bulbifera aboveground part (LBAP) and the Laportea bulbifera root (LBR). Through an assessment of the bioactive compound content, a significant finding emerged: the LBR exhibited notably higher levels of these bioactive phytochemicals compared to the LBAP. This observation was further reinforced by the antioxidant assays, which demonstrated the superiority of the LBR's antioxidant capacity. The experimental results unequivocally indicate that the root is the optimal medicament portion for Laportea bulbifera. Furthermore, it was discovered that the presence of alcohol in the extraction solvent significantly enhanced the extraction of active ingredients, with the methanol extract of LBR performing the best among the extracts tested. Consequently, this extract was selected for further research. Leveraging cutting-edge UHPLC-ESI-Q-TOF-MS technology, the methanol extract of LBR was meticulously analyzed, revealing the presence of 41 compounds, primarily belonging to the phenolics and fatty acids. Remarkably, stability experiments demonstrated that the phenolics in the methanol extract maintained their stability across various pH values and during in vitro simulations of the human digestive system, albeit showing gradual degradation under high temperatures. Furthermore, the oxidative stability tests conducted on oils revealed the potential of the methanol extract as a stabilizer for olive oil and sunflower oil. Moreover, oral acute toxicity studies confirmed the low toxicity of the methanol extract, further supporting its safe use for medicinal purposes. Of particular note, histopathological examination and biochemical analysis affirmed the remarkable protective effects of the methanol extract against d-galactosamine-induced liver damage. These findings underscore the therapeutic potential of the methanol extract from the LBR in the treatment of diseases associated with oxidative imbalance.

Green extraction of active ingredients from finger citron using subcritical water and assessment of antioxidant activity

In this study, subcritical water extraction (SWE) was used to extract active compounds, such as total phenols, total flavonoids, and essential oil, from finger citron. Response surface methodology (RSM) was applied to determine the optimal extraction conditions (total phenols: liquid-to-solid ratio (LSR) 40.9 mL/g, extraction time (ET) 2.7 h, subcritical water temperature (SWT) 183 °C; total flavonoids: LSR 38.8 mL/g, ET 2.4 h, SWT 176 °C; essential oil: LSR 42.3 mL/g, ET 2.5 h, SWT 182 °C). Under these conditions, the extraction yields of total phenols, total flavonoids, and essential oil were 45.65 ± 0.21 mg/g, 33.12 ± 0.19 mg/g, and 4.36 %, respectively. The extracts had remarkable antioxidant activity in vitro, especially those from SWE, which outperformed other methods. For the extracts, the IC50 values for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, 2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonat) (ABTS) radical scavenging, and (2-Phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl) (PTIO) radical scavenging were 0.32 mg/mL, 0.05 mg/mL, and 6.14 mg/mL, respectively. Finger citron essential oil (FCEO) was also analyzed by GC-MS and 37 constituents were identified. The major component was D-limonene at 23.59 %. The essential oil extracted by subcritical water also showed notable antioxidant activity, with IC50 values of 0.97 mg/mL, 0.08 mg/mL, and 5.35 mg/mL for DPPH radical scavenging, ABTS radical scavenging, and PTIO radical scavenging, respectively. These findings show that SWE has a promising potential to extract active compounds from natural products and enhance their biological activity.

Construction of coaxial liquid centrifugal platform for the extraction of active ingredients from natural products.

In the classical natural product extraction and separation process, it is tedious and requires large amounts of reagents and time. In this study, an efficient coaxial liquid centrifugal oil-water-oil triple-liquid-phase system with a simple structure and convenient operation was successfully constructed and used to extract flavonoids from Platycladi Cacumen. The results showed that the coaxial liquid centrifugal platform constructed in this study had good stability and 6ml was the minimum volume of the middle phase for 1000rpm to stabilize the system. Besides, it was easy to repeat the operation: the relative standard deviations of the extraction yields of flavonoids and sugar in six parallel operations were all less than 10%. Moreover, it was only one-tenth of the time required for this method as traditional liquid-liquid extraction while reducing the use of volatile organic reagents. Finally, the new method was more selective than the traditional method for the extraction of flavonoids. Therefore, this study provides a possibility for the coaxial liquid centrifugal platform to be used in multi-liquid phase systems to achieve the simultaneous extraction of different parts of natural products by different liquid phases. It is expected to provide a reliable reference for further expansion of small-scale experimental operations to industrial production.

Online pharmaceutical process analysis of Chinese medicine using a miniature mass spectrometer: Extraction of active ingredients as an example

The automation of traditional Chinese medicine (TCM) pharmaceuticals has driven the development of process analysis from offline to online. Most of common online process analytical technologies are based on spectroscopy, making the identification and quantification of specific ingredients still a challenge. Herein, we developed a quality control (QC) system for monitoring TCM pharmaceuticals based on paper spray ionization miniature mass spectrometry (mini-MS). It enabled real-time online qualitative and quantitative detection of target ingredients in herbal extracts using mini-MS without chromatographic separation for the first time. Dynamic changes of alkaloids in Aconiti Lateralis Radix Praeparata (Fuzi) during decoction were used as examples, and the scientific principle of Fuzi compatibility was also investigated. Finally, the system was verified to work stably at the hourly level for pilot-scale extraction. This mini-MS based online analytical system is expected to be further developed for QC applications in a wider range of pharmaceutical processes.

Integrated Proteomics and Metabolomics Analysis to Explore the Amelioration Mechanisms of Rosa roxburghii Tratt Fruit Polyphenols on Lipopolysaccharide-Induced Acute Lung Injury Mice

Acute lung injury (ALI) is the main cause of death for the elderly and children due to its high morbidity and mortality rates. Plant-derived functional foods are becoming increasingly important to the healthcare and food industries for adjunctive and alternative treatments of ALI. Polyphenols have been regarded to be beneficial to the prevention and amelioration of ALI. Rosa roxburghii Tratt fruit polyphenols (RRTP) has potential to prevent ALI, but mechanism remains unclear. This study was set up to systematically analyze the RRTP extract active ingredients, comprehensively evaluate its protective effects via lung histopathological examination, protein concentration, and cytokines production in ALI mice induced by lipopolysaccharide (LPS), and finally revealed alleviation mechanisms of the regulatory effects of RRTP by proteomics and metabolomics approach. The results demonstrated RRTP could synergistically exert significant preventive effects against ALI by notably ameliorating lung histopathological damage and pulmonary capillary permeability in ALI mice, inhibiting lung tissue inflammatory response and acute phase proteins and S-100 calcium binding proteins, suppressing excessive activation of complement and coagulation cascades, and regulating disordered lipids metabolism and amino acid metabolism. This study illustrated that RRTP has obvious advantages in ALI adjunctive therapy and revealed the complicated amelioration mechanisms, which provides a breakthrough for the development and demonstration of RRTP as a nutritional compound additive for complementary therapy of ALI.

Recycling utilization of Chinese medicine herbal residues resources: systematic evaluation on industrializable treatment modes.

Traditional Chinese medicine (TCM) is an indispensable part of the world health and medical system and plays an important role in treatment, prevention, and health care. These TCM produce a large amount of Chinese medicine herbal residues (CHMRs) during the application process, most of which are the residues after the decoction or extraction of botanical medicines. These CMHRs contain a large number of unused components, which can be used in medical, breeding, planting, materials, and other industries. Considering the practical application requirements, this paper mainly introduces the low-cost treatment methods of CHMRs, including the extraction of active ingredients, cultivation of edible fungi, and manufacture of feed. These methods not only have low upfront investment, but also have some income in the future. Furthermore, other methods are briefly introduced. In conclusion, this paper can provide a reference for people who need to deal with CMHRs and contribute to the sustainable development of TCM.

Chemical Compositions, Extraction Optimizations, and In Vitro Bioactivities of Flavonoids from Perilla Leaves (Perillae folium) by Microwave-Assisted Natural Deep Eutectic Solvents.

Natural deep eutectic solvents (NADESs) have been gradually applied to green extraction of active ingredients. In this study, microwave-assisted NADESs were applied to the extraction of flavonoid compounds from perilla leaves. Through comparative experiments, NADES-3 (choline chloride and malic acid at a molar ratio of 1:1) was found to have the highest extraction efficiency of total flavonoids, including apigenin 7-O-caffeoylglucoside, scutellarein 7-O-diglucuronide, luteolin 7-O-diglucuronide, and scutellarein 7-O-glucuronide by HPLC-MS. The following optimal extraction parameters were obtained based on response surface design: water content in NADES of 23%, extraction power of 410 W, extraction time of 31 min, and solid-liquid ratio of 75 mg/mL, leading to the extraction yield of total flavonoids of 72.54 mg/g. Additionally, the strong antimicrobial and antiallergic activity, inhibition of nitrosation, and antioxidant activity of total flavonoids by using NADESs were confirmed. This new extraction method provides a reference for the further exploration of NADES systems and may be widely used for the green extraction of natural active ingredients.
