Ultrasound-assisted enzymatic extraction of jujube (Ziziphus jujuba Mill.) polysaccharides: Extraction efficiency, antioxidant activity, and structure features

This study investigated the effect of ultrasound-assisted enzymatic extraction (UAEE) on the extraction efficiency, antioxidant activity, and structural properties of jujube polysaccharide (JPS), with hot water extraction (HWE), ultrasound-assisted extraction (UAE), and enzymatic-assisted extraction (EAE) serving as controls. Optimal extraction conditions were determined through a multi-index weighted scoring method that comprehensively accounted for yield, duration, and antioxidant activity. Results demonstrated that the JPS yield obtained by UAEE at 22/33 kHz was 10.5 % to 16.3 % higher than those achieved by the other methods, significantly enhancing antioxidant activity. Monosaccharide composition analysis revealed that UAEE increased the content of key mono-sugars in JPS. Additionally, assessments of molecular weight distribution, zeta potential, and rheological properties showed that UAEE reduced the molecular weight and apparent viscosity of JPS, resulting in a looser structural configuration. These structural modifications were observed in scanning electron microscope (SEM) images, which revealed a filamentous branched morphology in JPS obtained through UAEE. Further observations using the atomic force microscope (AFM) indicated that the polysaccharide chains extracted by UAEE were shorter in length, lower in height, and free from aggregation

Antioxidant and antimicrobial activities of different extracts of Tragopogon dubius and Tragopogon porrifolium L. subsp. longirostris: Determination of their phytochemical contents by UHPLC-Orbitrap®-HRMS analysis

In this study, dichloromethane (DCM), n-hexane, ethanol, methanol, and natural deep eutectic solvents (NADES) were used to extract Tragopogon porrifolium L. subsp. longirostris (Sch. Bip.) Greuter and Tragopogon dubius L. Antioxidant properties (Cu2+ reduction capacity, DPPH, ABTS and Fe3+ reduction) and antimicrobial activities of these extracts were determined. Also, metabolite profiles were determined using UHPLC-Orbitrap®-HRMS. NADES extracts of both species (MIC: 0.18-0.71 mg/ml) showed significantly better antimicrobial activity against Klebsiella pneumoniae, Bacillus subtilis, and Staphylococcus aureus compared to other solvents (MIC: 25 mg/ml). The study showed that T. dubius methanol and NADES extracts had better inhibitory effects on DPPH (EC50:45.17 μg/mL, EC50:108.73 μg/mL) and ABTS (EC50:59.54 μg/mL, EC50:95.98 μg/mL) radical scavenging activities than other extracts. Similarly, T. porrifolium methanol and NADES extracts had better inhibitory effects on DPPH (EC50:48.33 μg/mL, EC50:38.72 μg/mL) and ABTS (EC50:77 μg/mL, EC50:58.93 μg/mL) radical scavenging activities than other extracts. Furthermore, Fe3+ and Cu2+ reducing capacities were highest in methanol and NADES extracts of both plants.UHPLC-Orbitrap®-HRMS was used to profile the chemical composition, validating 71 phytochemicals, with the R2 value ranging from 0.9903 to 0.9993. The method's LOD values ranged from 0.45 to 2.94, and LOQ values ranged from 1.50 to 9.78. Recovery percentages at 10, 60, and 100 ng/mL concentrations ranged from 71.73% to 116.91%, 85.36% to 115.38%, and 97.08% to 109.65%, respectively. The analysis identified 29 phytochemicals, with major compounds including chlorogenic acid, quinic acid, apigenin 7-glucuronide, vitexin, schaftoside, orientin+isoorientin, luteoloside, and isoquercitrin. The significant biological impacts of these plants as a compelling reservoir of phytochemicals with therapeutic properties.

Effects of Cultivation Temperature on Phenolic Compound Content and Antioxidant Capacity in the Leaves and Stems of the Vegetable Gynura bicolor (Roxb. ex Willd.) DC

The leafy vegetable Gynura bicolor (Roxb. ex Willd.) DC. is cultivated in Taiwan and is popular because of its abundant antioxidant components. In this study, the effects of cultivation temperature on the antioxidant levels and antioxidant activity of methanolic extract from G. bicolor leaves and stems were studied. In cultivated plants, total phenolics were the most abundant components of the methanol extract. The levels of antioxidant compounds from the aboveground portions were highest when cultivated at 25 °C, followed by those when cultivation at 20, 30, and 35 °C. The antioxidant capacity of the leaf extract was highest when G. bicolor was grown at 25 °C, scavenging 69% of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical. The antioxidant capacity was lower in stems, scavenging 26% to 41% of the DPPH. A principal component analysis (PCA) was performed to explore the correlation between phenolic compound levels and antioxidant capacity under different cultivation temperatures. The PCA confirmed that growth at 25 °C resulted in the highest levels of phenolic compounds in the leaves G. bicolor.

Terminalia catappa Kernel Flour Characterization as a Functional and Bioactive Ingredient for Cookies Formulation

Terminalia catappa (tropical almond) represents an underutilized resource with potential applications in functional food development. This study investigated the technological properties and bioactive characteristics of T. catappa kernel flour and its application in cookie formulation. The research examined the techno-functional properties, pasting behavior, and bioactive profile of T. catappa flour and its blends with different sweeteners (erythritol and cane sugar at 5% and 15% concentrations). Cookies were formulated using optimized ingredients, and their quality parameters were evaluated through physical, chemical, and sensory analyses. T. catappa flour demonstrated significant water holding capacity (4.48 g H2O/g DM) and notable antioxidant activity in both aqueous and ethanolic extracts (DPPH: 1.95–3.35 mg TE/g DM). The addition of sweeteners influenced pasting properties, with higher concentrations generally reducing peak viscosity and pasting temperature. Developed cookies exhibited stable water activity (0.294–0.320) over one month of storage and contained substantial dietary fiber (5.018 g/100 g). Sensory evaluation revealed superior acceptability for thicker (10 mm) cookies, particularly in texture and appearance attributes. This study establishes T. catappa kernel flour as a promising functional ingredient for gluten-free bakery applications, offering both technological functionality and bioactive properties suitable for health-conscious product development.

Poultry manure application enhances the phytochemical contents, antioxidant, and fixed oil of Momordica charantia L

Due to its rich sources of bioactive compounds, Momordica charantia (bitter melon) is used as a food and natural medicine, resuting in many beneficial health effects. Poultry manure is a valuable source of organic fertilizer used to enrich soil fertility to obtain high yields from agricultural products. It contains high levels of essential macro and micronutrients. The present study aimed to determine the effects of chicken and turkey manure applications on the protein content, extract content, antioxidant activity, total phenolic and flavonoid contents, and mineral matter productivity of bitter melon fruits. In addition, the study evaluated the chicken and turkey manure applications on the fixed oil quality of bitter melon seeds. In this context, different doses of chicken (T1:10, T2:12.5, T3:15, T4:17.5 t/ha) and turkey (T5: 10, T6: 12.5, T7:15, T8:17.5 t/ha) manures were applied by sowing. The crude protein contents values of the bitter melon fruits ranged from 4.79 to 7.80 %, and a maximum crude protein content of bitter melon fruits was obtained from T8 application. The highest DPPH activity was observed with the T7 (88.43 %) and T4 (86.23 %) applications, while the highest FRAP value was obtained from T4 application (73.52 mg TE/g). In addition, the highest total phenolic content (21.13 mg GAE/g) and total flavonoid amount (11.44 mg QE/g) of bitter melon fruits were obtained from T6 application. Maximum capric acid (50.40 %) was recorded in the seed oil after applying the T1, while the highest concentration of lauric acid (30.09 %) were reflected by applying the T5. The highest Fe, Cu and Mn concentrations were found in the T4, while the highest concentration of Zn and B recorded in the fruit extract appeared after applying the T6. The PCA analysis showed 56.2 % of total variation. Thus, the results suggest that the application of poultry manure could be employed to get maximum benefits regarding protein and extract contents, antioxidant activity, total phenolic and flavonoid contents, fixed oil component and mineral content values.

Eutectic solvents for the extraction of phenolic compounds from the Moldovan Dragonhead

Introduction. Deep eutectic solvents (DES) are of interest to modern pharmaceutical technology as alternative extractants to traditional organic solvents. The prospects for their use are due to low toxicity, biodegradability, the possibility of selective extraction of individual substances and regeneration of the extractant. The purpose of the study is to develop and optimize a technology for ultrasonic extraction of phenolic acids from the Moldovan dragonhead using DES, and to study the antioxidant activity of the resulting extracts. Results. 11 DES compositions with different molar ratios of components were obtained; the optimal composition one was selected based on choline and lactic acid in a molar ratio of 1:3. The dependence of the extraction of phenolic compounds on the composition of DES, the influence of water content in the composition of DES on the process of extraction of phenolic compounds have been studied. Data were obtained on the antioxidant activity of the Moldavian snakehead extract obtained using DES. Conclusion. A method has been developed for the extraction of phenolic acids from the Moldavian snakehead with deep eutectic solvents. The extraction efficiency of the DES-3 mixture based on choline chloride and lactic acid in a molar ratio of 1:3 has been proven. The content of phenolic compounds in terms of rosmarinic acid was 7.72±0.12%, which is 0.23% more than when using 50% ethyl alcohol as an extractant. Using cyclic voltammetry, it was established that the antioxidant activity of extracts obtained using DES is due to the content of rosmarinic acid.

Antioxidant, antimicrobial and antiproliferative activities of alcoholic extracts from Piper aequale Vahl leaves

Piper is a large plant genus containing essential oils rich in mono and sesquiterpenes and other secondary metabolites showing different biological activities. Piper aequale, cordoncillo, is used in México for urinary and prostate ailments, suggesting a potential therapeutic effect against prostate cancer. Due to the lack of chemical and pharmacological information on this species, in this work antimicrobial and antiproliferative activities were evaluated. Leaves ethanol and methanol extracts were used to assess antioxidant activity (DPPH, FRAP), antimicrobial activity (Kirby-Bauer) against clinically relevant strains and antiproliferative activity on prostate cancer cell line PC-3 (MTT assay). Methanolic extract exhibited the highest antioxidant activity, with 69.08% (DPPH) and inhibitory effects on pathogenic bacterial strains associated with urinary tract infections. Ethanolic extract displayed moderate antiproliferative activity (IC50 81.28 μg/mL), showing cytotoxicity from 100 μg/mL. This study demonstrates P. aequale exhibits inhibitory effects against bacteria associated with urinary problems and antiproliferative properties in prostate cancer cells.

Tailored recovery of antioxidant fractions enriched in caffeine and phenolic compounds from coffee pulp using ethanol-modified supercritical carbon dioxide

Coffee pulp (CP) is the by-product of coffee processing that urgently needs to be revalorized using sustainable technologies. This work applied a design of experiment (DoE) for modeling the extraction of bioactive compounds from CP using supercritical carbon dioxide (sc-CO2) with ethanol as a co-solvent under variable conditions (temperature, pressure, and ethanol percentage). Considering extraction efficiency (per unit of CP) and extraction selectivity (per unit of extract), results showed that ethanol percentage significantly enhanced the efficiency of total phenolic content, as well as the selectivity of chlorogenic acid and protocatechuic acid (p < 0.05). The effects of temperature and pressure in EtOH-modified sc-CO2 extraction exhibited non-linear patterns, highlighting the interaction between temperature and ethanol. Total phenolic content was responsible for the antioxidant activity of the EtOH-modified sc-CO2 extracts, which was also affected by caffeine in the extracts. The extraction condition that balances both efficiency and selectivity was found at 65 °C, 23 MPa, and 9 % ethanol as co-solvent. The extraction yield (5.8 mg/g CP) was 3.8 times and 6.7 times lower than that of aqueous and ethanolic extraction, respectively, indicating a relatively lower efficiency of sc-CO2. However, the selectivity of caffeine (106.3 mg/g extract) and sum of identified phenolic compounds (16.3 mg/g extract) such as chlorogenic, protocatechuic, and caffeic acids, especially for protocatechuic acid (14.9 mg/g extract), was superior to conventional extraction, resulting in the better ABTS chemical (182.3 mg TE/g extract) and cellular antioxidant (384.9 U/mg extract) activities. The notable enrichment of caffeine and protocatechuic acid, together with the antioxidant potential observed in the EtOH-modified sc-CO2 extracts of CP could be promising for functional food, cosmetic, and pharmaceutical industries.

The study of the total antioxidant capacity of St. John’s Wort (Hypericum perforatum L.) herb

Aim. To determine the total antioxidant capacity of St. John’s Wort herb. Materials and methods. The content of phenolic compounds, anthraquinone derivatives, flavonoids, and derivatives of hydroxycinnamic acids was determined by the spectrophotometric analysis, while organic acids were identified by the alkalimetric method; the antioxidant activity of extracts obtained was evaluated by the potentiometric method. Results and discussion. The total antioxidant capacity of St. John’s Wort herb was 91.35 mmol-equiv/m dry weight, the total content of phenolic compounds was 22.70 mg of gallic acid/mL, anthraquinone derivatives – 0.46 mg of hyperecin/mL, flavonoids – 8.10 mg of rutin/mL, derivatives of hydroxycinnamic acids – 7.90 mg of chlorogenic acid/mL and organic acids – 10.80 mg of citric acid/mL in St. John’s Wort herb extracts obtained during the sequential exhaustive extraction. The correlation analysis showed a very high positive correlation between the antioxidant activity and the total content of phenolic compounds, flavonoids, hydroxycinnamic acid derivatives in St. John’s Wort herb extracts. Conclusions. The total antioxidant capacity of H. perforatum herb was 91.35 mmol-equiv./m dry weight. The results of the analysis of the BAS content and the antioxidant activity of H. perforatum herb extracts revealed that the aqueous extract had a significant content of phenolic compounds, flavonoids, hydroxycinnamic acids, organic acids and the antioxidant activity, while anthraquinone derivatives dominated in 96 and 60 % EtOH extracts. The quantitative analysis showed that flavonoids and hydroxycinnamic acids dominated among phenolic compounds. The results can be applied in developing optimal technologies for obtaining drugs based on the extract of H. perforatum herb.

Phytochemical analysis, antioxidant activity and modulatory effect of aqueous leaf extract of Pistacia lentiscus L on oxaliplatin-induced oxidative stress in isolated rat liver mitochondria

This study evaluates the antioxidant effects of aqueous leaf extract of Pistacia lentiscus (ALEPL) and its potential to counteract oxaliplatin (OXA)-induced mitochondrial oxidative stress in rat livers, a common side effect of chemotherapy in cancer treatment. Bioactive compounds were identified using High-Performance Liquid Chromatography coupled with Tandem Mass Spectrometry (HPLC-MS and MS), with Fourier-Transform Infrared Spectroscopy (FTIR) and Atomic Absorption Spectrophotometry (AAS) for chemical and mineral analysis. ALEPL showed notable antioxidant activity, with IC50 values of 4.30 ± 0.27 μg/mL for DPPH (2,2-Diphenyl-1-picrylhydrazyl) scavenging, 13.64 ± 0.51 μg/mL for reducing power, 32.62 ± 5.32 μg/mL for hydroxyl radical scavenging, and 205.08 ± 25.77 μg/mL for superoxide anion radical scavenging. In ex vivo experiments, mitochondria isolated from Wistar rat livers were treated with OXA and ALEPL in a dose-dependent manner. ALEPL pretreatment effectively restored mitochondrial antioxidant enzyme activities, increased glutathione (GSH) levels, and reduced lipid peroxidation (MDA) caused by OXA. These findings suggest that ALEPL has the potential to act as a natural antioxidant to support cancer treatment by mitigating chemotherapy-induced oxidative stress. Future studies could explore its application as an adjuvant in clinical settings to enhance the efficacy of chemotherapy while reducing its side effects.

Effect of in vitro Digestion on Anticancer and Antioxidant Activity of Phenolic Extracts from Latex of Fig Fruit (Ficus carica L.).

In this study, changes in total phenolic content, phenolic profile and antioxidant activity and anticancer activity against cervix cancer and colorectal cancer cell lines of phenolic extracts of black and white fig (Ficus carica L.) latex (milk) were investigated during in vitro gastro-intestinal digestions for the first time. The findings indicated that in vitro digestion process exerted a significant effect on total phenolic content (TPC) of the phenolic extract from white fig milk (WFM-PE) and phenolic extract from black fig milk (BFM-PE) and TPC tended to a decrease after in vitro digestion (p<0.05). As consistent with these findings, antioxidant activity (by CUPRAC method) of the samples decreased (p<0.05) during in vitro digestion. The IC50 value of the undigested BFM-PE was significantly lower than that of the undigested WFM-PE (p<0.05). The undigested and the digested WFM-PE and BFM-PE did not show any cytotoxic activity against normal cells. However, anticancer activity of WFM-PE on cervix and colorectal cancer cell lines (p<0.05) and anticancer activity of BFM-PE against colorectal cancer cell lines cells decreased after in vitro digestion (p<0.01). On the other hand, the dominant phenolic was catechine hydrate and was syringic acid.

Development of Herbal Ultrasound Gel Prototype from Zingiber cassumunar Roxb., Curcuma longa L. and Zingiber officinale Roscoe for Antioxidant and Anti-Inflammation

This research aimed to determine the total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity of phlai, turmeric, ginger and combined extract of phlai, turmeric, and ginger at recipe 1:2:2 (PTG). In addition, the formula of herbal ultrasound gel for antioxidant and anti-inflammatory properties was developed. The TPC, flavonoid, antioxidant and anti-inflammation were investigated by using the Folin-Ciocalteu method, flavonoid, DPPH, ABTS and nitric oxide (NO) scavenging assay. The PTG extract was tested for the phytochemicals by GC-MS technique. The microbial contamination standard of herbal ultrasound gel prototype was determined. The result show that the PTG extract has the TPC and flavonoid at 105.11 ± 16.93 mg GAE/g extract and 63.58 ± 2.29 mg QE/g extract, respectively. Moreover, the DPPH and ABTS assay show that PTG extract has the half maximal inhibitory concentration (IC50) at 0.82 ± 0.11 and 0.98 ± 1.56 mg/mL, respectively. In addition, PTG extract has a high percentage of NO inhibition at 53.38 ± 0.16. Furthermore, the GC-MS method found that PTG extract has 7 active ingredients. The highest amount of phytochemical is AR-turmerone. The safety assessment for external use found that the herbal ultrasound gel prototype product passed standard criteria for microbial contamination at the laboratory level. To conclude, the PTG extract has a synergistic effect and a high ability to act as an antioxidant and anti-inflammation. Moreover, the PTG herbal ultrasound gel has a microbial contamination safety to use. Therefore, the prototype herbal ultrasound gel product may be further developed into health products to treat pain and inflammation efficiently in clinical practice.

Antioxidant, anticoagulant and antithrombotic properties of ethanolic extract of Centaurea diluta subsp. algeriensis (Coss. &amp; Durieu) Maire: in vitro study and novel therapeutic approach

The aim of this study is to estimate the antioxidant, anticoagulant and antithrombotic activities of the ethanolic extract of Centaurea diluta subsp. Algeriensis (Coss. &amp; Durieu) Maire, an endemic Algerian plant belonging to Asteraceae family. The antioxidant activity was determined by DPPH radical scavenging activity and metal chelating test. The plasma of healthy volunteers was used to evaluate the clotting time, prothrombin time, and thrombolysis activity. C. diluta ethanolic extract (CEE) showed a good ability to scavenge DPPH radical with an IC50 value of 0.115 ±0.005 mg/ml. In addition, this extract exhibited a high iron chalating capacity with IC50 of 0.36 ± 0.07 mg / ml. Also, the results showed that CEE exhibited anticoagulant activity by prolonging the clotting time at the concentration of 50 mg/ml. For prothrombin time, the concentration of 50 mg/ml has a strong effect with value of 13.14±1.91 sec. In the thrombolysis test, the CEE extract showed a moderate thrombolytic activity with the best effect at a concentration of 50 mg/ml (20.86±0.31%).These findings could pave the way for the development of natural antioxidant and anticoagulant agents that can be used in the prevention and treatment of thrombotic disorders.

Evaluation of Toxicity and Antioxidant Activity of the Ethanolic Extract from Ulva Lactuca

Ulva lactuca (sea lettuce) is a popular food for consumers and has high nutritional value. It has also been found to be a source of health-promoting substances, including a high amount of proteins which can be utilized as a future plant-based functional ingredient in the food industry. However, before extensive applications in the aforementioned fields, it is crucial to ensure the extracts’ biological-pharmacological activities and their safety. In this study, we evaluated the cytotoxicity, genotoxicity, and antioxidant activity of the ethanolic extract of the sea lettuce extract.We tested cytotoxicity by MTT assays on 5 cell lines, including fibroblast (L929), macrophages (RAW 264.7), hepatocytes (FL83B), keratinocytes (HaCaT) and normal colon (CCD-18co) cells. Genotoxicity of the extract was tested by comet and micronucleus assays on human lymphoblast cells (TK6). Our results demonstrated at a concentration lower than 50 µg/mL the extract did not show any cytotoxicity as well as genotoxicity. The extract was shown to possess high antioxidant capacity by DPPH and ABTS scavenging at the EC50 value of U. lactuca extracts was 631.84 ± 5.64 and 330.35 ± 19.49 μg/mL, respectively. Therefore, the extract may be safely used as an ingredient for food, that should help to ensure food security for humans in the future, as well as for cosmetic, pharmaceutical and other health-promoting products. HIGHLIGHTS Ulva lactuca extract displayed strong antioxidant activity. Neither cytotoxicity nor genotoxicity to cell lines are seen in the ethanol extract of lactuca. GRAPHICAL ABSTRACT

Metabolic and stress resistance effects in Macrobrachium rosenbergii juveniles supplemented with Curcuma longa extract.

This work studied the effects of C. longa extract as a dietary supplement for Macrobrachium rosenbergii. Shrimp juvenile (n = 960, initial average weight 0.06 ± 0.042g and initial stocking density 0.4 animals/L) received diets with different concentrations of turmeric extract (0.05, 0.2, 1%) for 60 days, and zootechnical (weight gain, specific growth rate, initial and final condition factor) and metabolic (activities of digestive enzymes, amino acid metabolism enzymes, and energy metabolism enzymes) parameters were evaluated. A transport simulation (TS) test was conducted and, in challenged animals, survival and antioxidant status were studied. Phenolic compound content and in vitro antioxidant potential of the turmeric extract were evaluated. Turmeric extract (0.05%) significantly increased weight gain and the activity of digestive enzymes. An increase in citrate synthase activity for all concentrations of turmeric was also observed. The glutamate dehydrogenase activity in the 0.05% group was also increased by turmeric extract. The in vitro antioxidant activity of the turmeric extract was like butylated hydroxytoluene (BHT), a common antioxidant, and the total phenolic content was 0.98 ± 0.02 mmolAG/g. In animals from the TS test, turmeric extract promoted a dose-dependent increase in animal survival. Furthermore, turmeric supplementation increased the activity of the enzymes catalase, glutathione reductase, glutathione peroxidase, glutathione S-transferase, and it reduced glutathione (GSH) content. A reduction in the levels of lipid peroxidation (TBARS) and protein carbonyl was also observed in the hepatopancreas of all experimental groups. The addition of low concentrations of turmeric extract to the diet of M. rosenbergii juvenile presents great potential for application in commercial shrimp farming, since the extract (at concentrations as low as 0.05 to 1%) increased weight gain and improved the physiological and metabolic conditions of the animals, improving the use of nutrients and consequently the animals' responses to stressful conditions, such as transport.

Optimization of aqueous extraction methods for Japanese taro (Colocasia esculenta (L) Schott)) tuber: analysis of antioxidant activity, total flavonoid content, and luteolin content

Japanese taro (Colocasia esculenta (L) Schott)) is one of the edible plants that contain phytochemicals such as flavonoids, phenolics, triterpenoids, tannins, and vitamin C which are beneficial to the health of the human body. The compound is also known to have antioxidant activity. Proper extraction can produce quality extracts. This study aims to optimize the extraction method on Japanese taro tubers. Taro tuber was extracted using water with three different temperatures (room temperature, 40℃, and 60℃). The ABTS (2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) method was used to determine the antioxidant activity of the aqueous extract, in addition to determining the total flavonoid content by colorimetric test, and also analyzing the luteolin content by Thin Layer Chromatography (TLC) Densitometry. The total flavonoid content of Taro extracts at room temperature (TERT), 40℃ (TE 40), and 60℃ (TE 60) were 13.97±3.52 mg QE/gr extract; 7.99±2.10 mg QE/gr extract; and 5.98±0.49 mg QE/gr extract, respectively. Luteolin content of TERT, TE40, and TE60 were 0.27%±0.08; 0.25%±0.06; and 0.31%±0.05, respectively. Antioxidant IC50 values of TERT, TE40, and TE60 were 120.32, 137.03, and 159.37 µg/mL, respectively. The study shows that the greater the temperature in the extraction process, the less flavonoid and antioxidant compounds content. While there is a slight difference in determining luteolin levels, optimum luteolin content is obtained at 60°C for 6 hours.

The polysaccharides from blackened jujube with ultrasonic assistance extraction: Optimization of extraction conditions, antioxidant activity and structural analysis

Jujube is a widely grown agricultural product. In order to avoid the waste of resources and improve the added value of the blackened jujube, the blackened jujube was processed under certain temperature and humidity conditions. At present, there are relatively few researches on polysaccharides of blackened jujube. The selection of extraction method is very important for the study and utilization of plant polysaccharides. Ultrasonic assisted extraction technology has been widely used in polysaccharide extraction for its advantages of environmental protection, economy and high extraction rate. In order to fully tap the utilization potential of polysaccharides from blackened jujube, the ultrasonic assisted extraction technology of polysaccharides from blackened jujube was optimized. Based on the optimal conditions obtained by response surface analysis, the extraction rate of polysaccharide was as high as 12.75 ± 0.05%. The IC50 values of hydroxyl radical, DPPH radical and superoxide anion were 2.604 mg/mL, 0.754 mg/mL and 8.069 mg/mL, respectively. The blackened jujube polysaccharide (BJP) was mainly composed of rhamnose (Rha), arabinose (Ara), galactose (Gal) and galacturonic acid (Gal-UA), with a molecular weight of 1.42 × 106 Da, and the surface was a dense network structure with voids. The results showed that ultrasound-assisted extraction could extract polysaccharides more economically and efficiently. In this study, the polysaccharide extracted from blackened jujube has natural antioxidant effect, which may be related to the high content of galacturonic acid in polysaccharide and the molecular chain structure of polysaccharide can be destroyed by ultrasonic wave. As a kind of natural antioxidant, blackened jujube polysaccharide has important application value in functional products in food and medicine industries.

Determination of Antioxidant Activity and Biochemical Content of Homalothecium philippeanum (Spruce) Schimp.

Living organisms produce reactive oxygen species (ROS) during the oxygen processing process. ROS damage biomolecules and causes oxidative stress. Antioxidants prevent cellular damage against these harmful effects of ROS by neutralizing free radicals. Medicinal plants provide a rich source of antioxidants to reduce oxidative stress and play an important role in the treatment of diseases. Bioactive compounds, especially polyphenols and flavonoids, protect cells against oxidative damage by neutralizing free radicals. Bryophytes, especially mosses, are one of the plant groups that attract attention in this area. Mosses show biologically active properties with secondary metabolites such as terpenes and flavonoids they contain. In this study, extraction of Homalothecium philippeanum moss with ethanol, methanol and n-hexane solvents was carried out and the biochemical content analysis of the extracts was investigated. 1,2-Benzenedicarboxylic acid, bis(2-methylpropyl) ester was detected as the major component in the ethanol and methanol extracts of the H. philippeanum. Biologically active alkanes such as Tetradecane and Hexadecane were found in the n-hexane extract. The antioxidant activity of moss ethanol extract was investigated using the DPPH method and the EC50 value was determined as 7.084 mg/ml.

Chemical constituents and biological activities of endophytic fungi from Fagopyrum dibotrys.

Fagopyrum dibotrys is an important wild food and feed germplasm resource. It has high nutritional and medicinal value and is rich in natural products, including flavonoids, phenolic acids, coumarins, and alkaloids. Endophytic fungi in F. dibotrys have emerged as valuable sources of natural products. However, studies on the biological activity and chemical composition of these endophytic fungi remain limited. In this paper, a new method to obtain natural active ingredients by fermentation of endophytic fungi from medicinal plants was proposed. Then the antioxidant and pathogenic activities of the endophytic fungi extracts were determined in vitro. In addition, secondary metabolites produced by endophytic fungi with medicinal activity were analyzed by high performance liquid chromatography-tandem mass spectrometry (LC-MS). Among the 95 endophytic fungal strains in F. dibotrys, four strains with high phenol yields were selected by reaction: Alternaria alstroemeriae (J2), Fusarium oxysporum (J15), Colletotrichum karsti (J74), and Colletotrichum boninense (J61). Compared with those of various extracts, the ethyl acetate fractions of A. alstroemeriae (J2), F. oxysporum (J15), and C. boninense (J61) exhibited superior antioxidant and antibacterial properties. The results indicated that the fungal extract was an excellent natural antioxidant and might be a potential antibacterial agent. The DPPH free radical clearance of A. alstroemeriae was 94.96 ± 0.004%. These findings indicated that A.alstroemeriae had strong antioxidant activity. In addition, the extract of A.alstroemeriae had good antibacterial activity against Escherichia coli and Staphylococcus aureus, with MICs of 0.5 and 0.05 mg/mL, respectively. The chemical constituents of the ethyl acetate extract from A. alstroemeriae were further analyzed by liquid chromatography-mass spectrometry (LC-MS). We noted that A. alstroemeriae can create a variety of medicinal substances that have high value in medicine, such as caffeic acid (884.75 ng/mL), 3-phenyllactic acid (240.72 ng/mL) and norlichexanthone (74.36ng/mL). In summary, many valuable active substances and medicinal substances can be obtained through the study of endophytic fungi of F. dibotrys.
