Differential Effect of Hydroxen Peroxide οn Toxic Cyanobacteria of Hypertrophic Mediterranean Waterbodies

Cyanobacterial blooms have been known since ancient times; however, they are currently increasing globally. Human and ecological health risks posed by harmful cyanobacterial blooms have been recorded around the world. These risks are mainly associated with their ability to affect the ecosystem chain by different mechanisms like the production of cyanotoxins, especially microcystins. Their expansion and their harmful effects have led many researchers to seek techniques and strategies to control them. Among them, hydrogen peroxide could be a promising tool against cyanobacteria and cyanotoxins and it is well-established as an environmentally friendly oxidizing agent because of its rapid decomposition into oxygen and water. The aim of the present study was to evaluate the effect of hydrogen peroxide on phytoplankton from two hypertrophic waterbodies in Greece. The effect of hydrogen peroxide on concentration of microcystins found in the waterbodies was also studied. Treatment with 4 mg/L hydrogen peroxide was applied to water samples originated from the waterbodies and Cyanobacterial composition and biomass, phycocyanin, chlorophyll-a, and intra-cellular and total microcystin concentrations were studied. Cyanobacterial biomass and phycocyanin was reduced significantly after the application of 4 mg/L hydrogen peroxide in water treatment experiments while chlorophytes and extra-cellular microcystin concentrations were increased. Raphidiopsis (Cylindrospermopsis) raciborskii was the most affected cyanobacterial species after treatment of the water of the Karla Reservoir in comparison to Aphanizomenon favaloroi, Planktolyngbya limnetica, and Chroococcus sp. Furthermore, Microcystis aeruginosa was more resistant to the treatment of Pamvotis lake water in comparison with Microcystis wesenbergii and Microcystis panniformis. Our study showed that hydrogen peroxide differentially impacts the members of the phytoplankton community, affecting, thus, its overall efficacy. Different effects of hydrogen peroxide treatment were observed among cyanobacerial genera as well as among cyanobacterial species of the same genus. Different effects could be the result of the different resistance mechanisms of each genus or species to hydrogen peroxide. Hydrogen peroxide could be used as a treatment for the mitigation of cyanobacterial blooms in a waterbody; however, the biotic and abiotic characteristics of the waterbody should be considered.

Comparison of UV-A photolytic and UV/TiO2 photocatalytic effects on Microcystis aeruginosa PCC7813 and four microcystin analogues: A pilot scale study

To date, the high cost of supplying UV irradiation has prevented the widespread application of UV photolysis and titanium dioxide based photocatalysis in removing undesirable organics in the water treatment sector. To overcome this problem, the use of UV-LEDs (365 nm) for photolysis and heterogeneous photocatalysis applying TiO2 coated glass beads under UV-LED illumination (365 nm) in a pilot scale reactor for the elimination of Microcystis aeruginosa PCC7813 and four microcystin analogues (MC-LR, -LY, -LW, -LF) with a view to deployment in drinking water reservoirs was investigated. UV-A (365 nm) photolysis was shown to be more effective than the UV/TiO2 photocatalytic system for the removal of Microcystis aeruginosa cells and microcystins. During photolysis, cell density significantly decreased over 5 days from an initial concentration of 5.8 × 106 cells mL−1 until few cells were left. Both intra- and extracellular microcystin concentrations were significantly reduced by 100 and 92 %, respectively, by day 5 of the UV treatment for all microcystin analogues. During UV/TiO2 treatment, there was great variability between replicates, making prediction of the effect on cyanobacterial cell and toxin behavior difficult.

Oxidative stress in the cyanobacterium Microcystis aeruginosa PCC 7813: Comparison of different analytical cell stress detection assays

Cyanobacterial blooms are observed when high cell densities occur and are often dangerous to human and animal health due to the presence of cyanotoxins. Conventional drinking water treatment technology struggles to efficiently remove cyanobacterial cells and their metabolites during blooms, increasing costs and decreasing water quality. Although field applications of hydrogen peroxide have been shown to successfully suppress cyanobacterial growth, a rapid and accurate measure of the effect of oxidative stress on cyanobacterial cells is required. In the current study, H2O2 (5 and 20 mg L−1) was used to induce oxidative stress in Microcystis aeruginosa PCC 7813. Cell density, quantum yield of photosystem II, minimal fluorescence and microcystin (MC-LR, -LY, -LW, -LF) concentrations were compared when evaluating M. aeruginosa cellular stress. Chlorophyll content (determined by minimal fluorescence) decreased by 10% after 48 h while cell density was reduced by 97% after 24 h in samples treated with 20 mg L−1 H2O2. Photosystem II quantum yield (photosynthetic activity) indicated cyanobacteria cell stress within 6 h, which was considerably faster than the other methods. Intracellular microcystins (MC-LR, -LY, -LW and -LF) were reduced by at least 96% after 24 h of H2O2 treatment. No increase in extracellular microcystin concentration was detected, which suggests that the intracellular microcystins released into the surrounding water were completely removed by the hydrogen peroxide. Thus, photosynthetic activity was deemed the most suitable and rapid method for oxidative cell stress detection in cyanobacteria, however, an approach using combined methods is recomended for efficient water treatment management.

Cyanobacteria and cyanotoxins in estuarine water and sediment

While transfer of freshwater cyanobacteria to estuaries has been observed worldwide, the associated transfer of cyanotoxins is less often reported, in particular the sediment contribution. During fall 2018, we monitored the co-occurrence of cyanobacteria and microcystin (MC) in both the water column and in surface sediments at five stations along a river continuum, from a freshwater reservoir to the coastal area in Brittany, France. Cyanobacteria dominated the phytoplankton community in the water column with high densities at the freshwater sites. Microcystis cells and intracellular MC transfer to estuarine and marine sites were observed with decreasing concentrations in accordance with flow dilution. Extracellular MC showed the opposite trend and increased from upstream to downstream in accordance with the lysing of the cells at elevated salinities. Surface sediment samples contained high densities of colonial Microcystis in freshwater and with decreasing concentrations along the salinity gradient, similar to cells concentrations in the water column. Intracellular MC was detected in sediment at all sites except at the marine outlet suggesting the survival of intact cells. Extracellular MC concentrations in sediment were up to five times higher than intracellular concentrations suggesting incomplete MC degradation. mcyB genes were present at all sites, while mcyA genes were absent at the marine outlet suggesting the presence of toxic strains along the estuary. The high densities of intact colonies of potentially toxic Microcystis in the estuarine sediment strongly suggest that sediments can act as an inoculum of cyanobacteria and cyanotoxins in estuaries.

Suppressing Cyanobacteria with Hydrogen Peroxide Is More Effective at High Light Intensities.

Hydrogen peroxide (H2O2) can be used as an emergency method to selectively suppress cyanobacterial blooms in lakes and drinking water reservoirs. However, it is largely unknown how environmental parameters alter the effectiveness of H2O2 treatments. In this study, the toxic cyanobacterial strain Microcystis aeruginosa PCC 7806 was treated with a range of H2O2 concentrations (0 to 10 mg/L), while being exposed to different light intensities and light colors. H2O2 treatments caused a stronger decline of the photosynthetic yield in high light than in low light or in the dark, and also a stronger decline in orange than in blue light. Our results are consistent with the hypothesis that H2O2 causes major damage at photosystem II (PSII) and interferes with PSII repair, which makes cells more sensitive to photoinhibition. Furthermore, H2O2 treatments caused a decrease in cell size and an increase in extracellular microcystin concentrations, indicative of leakage from disrupted cells. Our findings imply that even low H2O2 concentrations of 1–2 mg/L can be highly effective, if cyanobacteria are exposed to high light intensities. We therefore recommend performing lake treatments during sunny days, when a low H2O2 dosage is sufficient to suppress cyanobacteria, and may help to minimize impacts on non-target organisms.

Long-term exposure to antibiotic mixtures favors microcystin synthesis and release in Microcystis aeruginosa with different morphologies

The ecological risks of antibiotics in aquatic environments have raised great concerns worldwide, but the chronic effect of antibiotic contaminants on cyanotoxin production and release remains unclear. This study investigated the long-term combined effects of spiramycin (SP) and ampicillin (AMP) on microcystin (MC) production and release in both unicellular and colonial Microcystis aeruginosa (MA) through semi-continuous exposure test. At exposure concentration of 300 ng L−1, MA growth rates were stimulated till the end of exponential phase accompanied with the up-regulation of photosynthesis-related gene. The exponential growth phases of unicellular and colonial MA were prolonged for 2 and 4 days, respectively. The stimulation rate of growth rate and MC content in unicellular MA were significantly higher than that in colonial MA. The highest concentrations of intracellular MC (IMC) and extracellular MC (EMC) were observed in the binary mixture at equivalent SP/AMP ratio (1:1). The promotion of IMC concentration was in consistent with the stimulated expression of MC-synthesis-related gene and nitrogen-transport-related gene. The malondialdehyde content and activities of superoxide dismutase and catalase in unicellular MA were significantly higher than those in colonial MA. The EMC concentration and the antioxidant responses of both unicellular and colonial MA significantly increased with exposure time. Long-term exposure to mixture of SA and AMP at environmentally relevant concentrations would aggravate the disturbance to aquatic ecosystem balance through the stimulation of MA proliferation as well as the promotion of MC production and release.

Hot and toxic: Temperature regulates microcystin release from cyanobacteria

The mechanisms regulating toxin release by cyanobacteria are poorly understood despite the threat cyanotoxins pose to water quality and human health globally. To determine the potential for temperature to regulate microcystin release by toxin-producing cyanobacteria, we evaluated seasonal patterns of water temperature, cyanobacteria biomass, and extracellular microcystin concentration in a eutrophic freshwater lake dominated by Planktothrix agardhii. We replicated seasonal variation in water temperature in a concurrent laboratory incubation experiment designed to evaluate cause-effect relationships between temperature and toxin release. Lake temperature ranged from 3 to 27°C and cyanobacteria biomass increased with warming up to 18°C, but declined rapidly thereafter with further increases in temperature. Extracellular microcystin concentration was tightly coupled with temperature and was most elevated between 20 and 25°C, which was concurrent with the decline in cyanobacteria biomass. A similar trend was observed in laboratory incubations where productivity-specific microcystin release was most elevated between 20 and 25°C and then declined sharply at 30°C. We applied generalized linear mixed modeling to evaluate the strength of water temperature as a predictor of cyanobacteria abundance and microcystin release, and determined that warming≥20°C would result in a 36% increase in microcystin release when Chlorophyll a was ≤50μgl−1. These results show a temperature threshold for toxin release in P. agardhii, which demonstrates a potential to use water temperature to forecast bloom severity in eutrophic lakes where blooms can persist year-round with varying degrees of toxicity.

Effects of two copper compounds on Microcystis aeruginosa cell density, membrane integrity, and microcystin release

Microcystin release following Microcystis aeruginosa cell lysis after copper-based algaecide treatment is often cited as a concern leading to restricted use of algaecide in restoration of natural water resources. To examine this concern, bench-scale experiments were conducted to study responses of M. aeruginosa to 8-day copper exposures as copper sulfate and copper–ethanolamine (Cu–EA). M. aeruginosa UTEX 2385 was cultured in BG11 medium to cell density of 106cells/mL with total and extracellular microcystin of 93 and 53μg/L, respectively. Exposures of copper concentration ranged from 40 to 1000μgCu/L. Cell membrane integrity was indicated by erythrosine B. In the end of experiment, total microcystin and cell density in untreated control (313μg/L and 107cells/mL) was 3.3 and 10 times greater than pretreatment value, respectively. Minimum amount of copper required to reduce M. aeruginosa population within 8 days was 160μgCu/L as copper sulfate and 80μgCu/L as Cu–EA, where total and extracellular microcystin concentrations (47 and 44μg/L for copper sulfate; 56 and 44μg/L for Cu–EA) were degraded with degradation rate coefficient 0.1 day−1 and were less than pretreatment values. Given a copper concentration at 80µgCu/L as Cu–EA, M. aeruginosa cells were intact and less microcystin were released compared to treatments at 160–1000µgCu/L, where lysed cells and relatively greater microcystin release were observed. Based on the laboratory results, a minimum amount of copper required for reducing M. aeruginosa population could decrease total microcystin concentration and not compromise cells and minimize microcystin release.

Environmental influence on cyanobacteria abundance and microcystin toxin production in a shallow temperate lake

The increasing frequency of harmful cyanobacterial blooms in freshwater systems is a commonly recognized problem due to detrimental effects on water quality. Vancouver Lake, a shallow, tidally influenced lake in the flood plain of the Columbia River within the city of Vancouver, WA, USA, has experienced numerous summertime cyanobacterial blooms, dominated by Aphanizomenon sp. and Anabaena sp. Cyanobacteria abundance and toxin (microcystin) levels have been monitored in this popular urban lake for several years; however, no previous studies have identified which cyanobacteria species produce toxins, nor analyzed how changes in environmental variables contribute to the fluctuations in toxic cyanobacteria populations. We used a suite of molecular techniques to analyze water samples from Vancouver Lake over two summer bloom cycles (2009 and 2010). Both intracellular and extracellular microcystin concentrations were measured using an ELISA kit. Intracellular microcystin concentrations exceeded WHO guidelines for recreational waters several times throughout the sampling period. PCR results demonstrated that Microcystis sp. was the sole microcystin-producing cyanobacteria species present in Vancouver Lake, although Microcystis sp. was rarely detected in microscopical counts. qPCR results indicated that the majority of the Microcystis sp. population contained the toxin-producing gene (mcyE), although Microcystis sp. abundance rarely exceeded 1 percent of overall cyanobacteria abundance. Non-metric multidimensional scaling (NMDS) revealed that PO4–P was the main environmental variable influencing the abundance of toxic and non-toxic cyanobacteria, as well as intracellular microcystin concentrations. Our study underscores the importance of using molecular genetic techniques, in addition to traditional microscopy, to assess the importance of less conspicuous species in the dynamics of harmful algal blooms.

Seasonal dynamics of water bloom-forming Microcystis morphospecies and the associated extracellular microcystin concentrations in large, shallow, eutrophic Dianchi Lake

The increasing occurrence of Microcystis blooms is of great concern to public health and ecosystem due to the potential hepatotoxic microcystins (MCs) produced by these colonial cyanobacteria. In order to interpret the relationships between variations of Microcystis morphospecies and extracellular MC concentrations, the seasonal dynamics of phytoplankton community composition, MC concentrations, and environmental parameters were monitored monthly from August, 2009 to July, 2010. The results indicated that Microcystis dominated total phytoplankton abundance from May to December (96%–99% of total biovolume), with toxic Microcystis viridis and non-toxic Microcystis wesenbergii dominating after July (constituting 65%–95% of the Microcystis population), followed by M. viridis as the sole dominant species from November to January (49%–93%). Correlation analysis revealed that water temperature and nutrient were the most important variables accounting for the occurrence of M. wesenbergii, while the dominance of M. viridis was related with nitrite and nitrate. The relatively low content of MCs was explained by the association with a large proportion of M. viridis and M. wesenbergii, small colony size of Microcystis populations, and low water temperature, pH and dissolved oxygen. The extracellular MC (mean of 0.5±0.2μg/L) of water samples analyzed by enzyme-linked immunosorbent assay (ELISA) demonstrated the low concentrations of MC in Dianchi Lake which implied the low potential risk for human health in the basin. The survey provides the first whole lake study of the occurrence and seasonal variability of Microcystis population and extracellular MCs that are of particular interest for water quality monitoring and management.

Effects of limonene stress on the growth of and microcystin release by the freshwater cyanobacterium Microcystis aeruginosa FACHB-905

The effects of limonene exposure on the growth of Microcystisaeruginosa and the release of toxic intracellular microcystin (MCY) were tested by evaluating the results obtained from the batch culture experiments with M. aeruginosa FACHB-905. The time series of cell as well as intracellular and extracellular MCY concentrations were evaluated during 5d of the incubation. After exposure to limonene, the number of cells gradually diminished; the net log cell reduction after 5d of the exposure was 3.0, 3.6, and 3.8log when the initial cell densities were set at 1.6×107, 1.1×106 and 4.1×105cell/mL, respectively. Limonene was found to significantly influence the production and release of MCY. As the limonene exposure could inhibit the increase in the number of cells, the increase in the total MCY concentration in the medium was also inhibited. In the presence of limonene, the intracellular MCY was gradually released into the medium through a gradual reduction in the number of cells. The extracellular MCY concentration in the medium was significantly higher in the limonene-exposed samples than in the control samples, which confirmed that limonene cannot decompose the extracellular MCY.

Effects of d-menthol stress on the growth of and microcystin release by the freshwater cyanobacterium Microcystis aeruginosa FACHB-905

The effects of d-menthol on the growth of Microcystis aeruginosa FACHB-905 and microcystin (MCY) concentration were evaluated by batch culture experiments. The algal biomass and the intracellular and extracellular MCY concentrations were evaluated during 5d incubation. After the d-menthol exposure, the dry weight of the cells gradually decreased; the decrease in the dry weight after 5d exposure was 29, 12, and 2mgL−1 when the initial cell densities were 1.4×107, 1.2×106, and 2.9×105cellmL−1, respectively. The results indicate that the d-menthol exposure inhibited the cellular growth, thus also inhibiting the increase of the total MCY concentration. In the presence of d-menthol, the intracellular MCY was gradually released into the medium after the cell lysis. The extracellular MCY concentration in the medium was significantly higher in the d-menthol-exposed samples than in the control samples, confirming that d-menthol cannot decompose the extracellular MCY.

Development of an Efficient Extraction Method to Quantify Microcystin–LR from Natural Microcystis Bloom Samples from the Colombo Lake, Sri Lanka

Microcystins are a diverse group of hepatotoxic secondary metabolites produced bycyanobacteria (blue-green algae). Up to date more than 90 different Microcystins wereidentified. Among them, Microcystin-LR (MC-LR) is one of the most toxic and mostcommonly encountered Microcystin variants worldwide. Although detection and extractionmethods for Microcystins have been previously reported, there are no such techniquesavailable in Sri Lanka regarding the water safety monitoring. Therefore in this study, we aimto develop an efficient extraction and quantification method of intracellular and extracellularMicrocystins from natural Microcystis bloom samples. Collection of bloom material from theColombo Lake, Sri Lanka was carried out for a period of six months from January to June2013. Samples were filtered through GF-C filters to retain cyanobacterial cells (intracellularMicrocystins) and extracellular Microcystins to pass through. Intracellular Microcystins (GFCfilters) were extracted repeatedly (x2) in 80% methanol, rotary evaporated and the residuewas reconstituted in 100% methanol. Extracellular Microcystins were extracted by C18 solidphaseextraction cartridges and eluted in 80% methanol. The cool-dry method carried out wassimilar to the GFC-filter extraction; however, the filter was subjected to heating at 550C for15 min followed by cooling (15 min) prior to the 80% methanol extraction. During thecentrifugation method, 1 litre of bloom material was centrifuged (3,000 g, 20 min). The pelletwas extracted in 100% methanol, rotary evaporated (40 0C) and the residue was reconstitutedin 80% methanol. The supernatant was solid phase extracted using C18 cartridges and elutedin 80% methanol. Identification and determination of the intracellular and extracellularMicrocystin concentration were performed by photodiode array – high performance liquidchromatography. Recovery of 17.57±0.73, 15.81±0.44 and 14.66±1.04 ppm of totalmicrocystin-LR (intracellular and extracellular) was recorded for the filter method. Incontrast, total MC-LR extracted by the cool-dry method was 0.48±0.005, 0.32±0.03 and0.22±0.07 ppm. However, compared to other two methods, centrifugation method achievedthe highest total recovery of MC-LR (16.86±2.11, 15.71±1.73 and 13.77±0.04 ppm). Thus, itreveals that the filter method and centrifugation method are the most efficient extractionmethods for MC-LR from natural bloom samples. Further optimization of these methodswould enhance the monitoring and safety measures for the drinking and recreational waterreservoirs in Sri Lanka.

A systematic study on spatial and seasonal patterns of eight taste and odor compounds with relation to various biotic and abiotic parameters in Gonghu Bay of Lake Taihu, China

A systematic study was conducted on seasonal and spatial patterns of taste and odor (T&O) compounds with relation to biotic and abiotic parameters at fifteen sites in Gonghu Bay of Lake Taihu in 2008. We developed a sensitive and automated method to simultaneously analyze eight T&O compounds (boiling points ranging from 38 °C to 239 °C) by using Purge-and-Trap (P&T) coupled with GC/MS. Maximum particulate dimethyl trisulfide (DMTS, 69.6 ng/L) exceeded its odor threshold concentrations (OTC, 10 ng/L) and maximum dissolved DMTS was 6.1 ng/L, but still far below concentration in the drinking water pollution incident of Wuxi City in 2007 when DMTS reached 1768–11,399 ng/L. Geosmin (GEO), 2-methylisoborneol (MIB), β-cyclocitral, β-ionone and 2-isobutyl-3-methoxypyrazine (IBMP) occasionally or frequently exceeded their OTCs, whereas 2-isopropyl-3-methoxypyrazine (IPMP) and dimethyl sulfide (DMS) did not. We found for the first time significant correlations between particulate β-cyclocitral and β-ionon concentrations and intracellular and extracellular microcystin concentrations. Spatially, Nanquan Waterworks faced more risk by T&O contamination than Xidong Waterworks. High concentrations of NO 3–N, TDN and TN could be risky signs of taste and odor events by DMS, DMTS, IPMP, IBMP and GEO.

Factors Affecting Temporal and Spatial Variations of Microcystins in Gonghu Bay of Lake Taihu, with Potential Risk of Microcystin Contamination to Human Health

A field survey of the seasonal variation of microcystin (MC) concentration was performed in Gonghu Bay (a total of 15 sampling sites) of Lake Taihu from January to December 2008. Microcystis spp. biomass and intra-/extracellular MCs were significantly correlated with water temperature, suggesting the importance of temperature in cyanobacterial blooming in the lake. Higher MC concentration was found in summer and autumn, and peaks of Microcystis biomass and intra-/extracellular MC concentrations were all present in October. Spatially, risk of MCs was higher in littoral zones than in the pelagic area. There were significant correlations between N or P concentrations, and Microcystis biomass or MC content, suggesting that N and P levels affected MC production through influencing Microcystis biomass. Intra-/extracellular MCs and Microcystis biomass had negative exponential relationships with TN:TP, and the maximum values all occurred when TN:TP was <25. Multivariate analyses by pcca indicated that intra- and extracellular MC concentrations had better correlations with biological factors (such as Microcystis biomass and chl-a) than physicochemical factors. The maximum concentration reached up to 17 µg/L MC-Lreq, considerably higher drinking water safety standard (1 µg/L) recommended who. So it is necessary take measures reduce exposure risk of cyanobacterial toxins human beings.

Diversity of planktonic cyanobacteria and microcystin occurrence in Polish water bodies investigated using a polyphasic approach

Microscopic measurements of fresh biomass and 16S rRNA gene sequences from clone libraries and denaturing gradient gel electrophoresis (DGGE) were used to investigate cyanobacter- ial diversity in Polish water bodies in 2002. In addition, measurements of microcystin (MC) con- centrations were made. Thirty water samples were taken from 11 water bodies; of these samples, 18 were obtained from the Sulejow Reservoir during regular monitoring from June to October. Intra- and extracellular MC concentrations in Sulejow samples were measured by high performance liquid chromatography (HPLC). The extracellular MC concentration was assessed using a protein phos- phatase inhibition assay (PPIA) in additional lakes. Additionally, physicochemical parameters were measured (total nitrogen (TN), total phosphorus (TP), TN:TP ratio, chlorophyll a concentration, temperature). In Sulejow, high intracellular MC concentrations corresponded to large cyanobacterial biovolumes and to low TN:TP ratios. In the other lakes, extracellular MCs were not linked to any measured parameters. The combination of the microscopic and molecular data showed that Aphani- zomenon and Microcystis were the dominant genera during the summer period in the Sulejow Reser- voir. At the genetic level, there was a succession of 2 different operational taxonomic units (OTUs) belonging to the lineage Anabaena/Aphanizomenon. In the other water bodies, the most frequent populations were Aphanizomenon, Anabaena, Microcystis and Planktothrix. Small populations of Romeria, Snowella, Woronichinia, Limnothrix and Pseudanabaena were observed, and an enigmatic cluster affiliated with Prochlorothrix was genetically retrieved. Anabaena and Microcystis were presumed to be the main genera responsible for the MC production.

Microcystin production by Microcystis aeruginosa exposed to different stages of herbivorous zooplankton

Microcystin (MC) production by four monoclonal Microcystis aeruginosa strains was evaluated in response to infochemicals (indirect exposure) released from different stages of herbivorous zooplankton (neonate/juvenile and adult Daphnia magna and Moina macrocopa). The intracellular MC and extracellular MC concentrations were significantly different among the control and treatments with zooplankton culture media filtrates ( p<0.05), and in most cases MC production was significantly higher ( p<0.05) in strains exposed to infochemicals released from adult zooplankton rather than those of neonate/juvenile zooplankton in four strains of M. aeruginosa. Compared to intracellular MC (385.0–5598.6 μg g −1 DW), very low concentrations of extracellular MC (9.9–737.6 μg ml −1) were released, but both showed similar temporal patterns over the course of the experiment. This result might be attributed to the fact that adult zooplankton produced more infochemical signals than equal numbers of smaller juveniles and neonates. It is the first study to provide evidence that MC production might be impacted by infochemicals released from different stages of zooplankton, mediated with physiological characteristics, body size, and feeding habits.

Changes in microcystin production in cyanobacteria exposed to zooplankton at different population densities and infochemical concentrations

We investigated microcystin (MC) production by four cyanobacterial strains (three Microcystis aeruginosa and one Planktothrix agardhii) in response to different grazer densities (direct exposure: zero, two, four, or eight individuals per 300 mL) and infochemical concentrations (indirect exposure: 0%, 10%, 25%, and 50%) of Daphnia magna and Moina macrocopa. MC production increased after direct exposure to both zooplankton species and was higher with increasing concentration of infochemicals. This MC production was significantly different among the control and three zooplankton treatment levels. Upon direct and indirect exposure of cyanobacteria to zooplankton, intracellular MC peaked on days 3 and 4. In most cyanobacterial strains, the peak MC contents were significantly higher in direct treatment with the highest zooplankton density and in indirect treatment with the highest concentration of zooplankton culture media filtrate than with treatments with the lowest density and concentration, respectively. Extracellular MC concentrations were much lower than intracellular ones, but both showed similar temporal patterns over the course of the experiment. Cyanobacteria directly exposed to Daphnia released greater amounts of extracellular MC than did those exposed to Moina. This is the first study to provide evidence of an induced defense of increased MC production by cyanobacteria in response to increasing zooplankton grazer density and increased concentrations of infochemicals released by zooplankton. In addition to the induction of tolerance in Daphnia to toxic Microcystis, we discuss how these reciprocal defenses may explain the coexistence of zooplankton and toxic cyanobacteria in eutrophic freshwaters.

Elucidation and short-term forecasting of microcystin concentrations in Lake Suwa (Japan) by means of artificial neural networks and evolutionary algorithms

Non-supervised artificial neural networks (ANN) and hybrid evolutionary algorithms (EA) were applied to analyse and model 12 years of limnological time-series data of the shallow hypertrophic Lake Suwa in Japan. The results have improved understanding of relationships between changing microcystin concentrations, Microcystis species abundances and annual rainfall intensity. The data analysis by non-supervised ANN revealed that total Microcystis abundance and extra-cellular microcystin concentrations in typical dry years are much higher than those in typical wet years. It also showed that high microcystin concentrations in dry years coincided with the dominance of the toxic Microcystis viridis whilst in typical wet years non-toxic Microcystis ichthyoblabe were dominant. Hybrid EA were used to discover rule sets to explain and forecast the occurrence of high microcystin concentrations in relation to water quality and climate conditions. The results facilitated early warning by 3-days-ahead forecasting of microcystin concentrations based on limnological and meteorological input data, achieving an r 2=0.74 for testing.

Cosmopolitan freshwater cyanobacteria of the Microcystis genus can produce the toxin microcystin, and present a potential health risk in many eutrophic lakes. In this study, M. aeruginosa Strain PCC 7806 was grown in semi-continuous turbidostats to investigate the effect of increasing salinity on growth rate, microcystin cell quota, microcystin production and extracellular microcystin concentration. Specific growth rate, microcystin cell quota and microcystin production remained more or less unaffected by salinity levels up to 10 g l–1. Specific growth rate collapsed when salinity was increased beyond 10 g l–1 for several weeks. Cell size and microcystin cell quota decreased while extracellular microcystin concentrations increased at salinities above 10 g l–1, indicating leakage and/or cell lysis. Salt-shock experiments revealed that M. aeruginosa can temporarily endure salinities as high as 17.5 g l–1. These results indicate that, for a freshwater species, M. aeruginosa has a high salt tolerance. Rising salinities in freshwater ecosystems are therefore unlikely to suppress M. aeruginosa blooms, and may in fact enhance the exposure of aquatic organisms to elevated concentrations of extracellular microcystins.
