WITHDRAWN: Grape defense against gray mold involves VvBZR1-mediated autophagy

Brassinosteroids (BRs) are a class of polyhydroxylated steroidal phytohormones that effectively affect the disease resistance in grape. However, the molecular mechanism of BR signaling networks responsible for activation of host defense against gray mold infection remained to be elucidated. The induced resistance mechanism of eBR treatment on table grapes (Vitis vinifera L. cv. Thompson Seedless) was conducted by an artificial inoculation trial. Grape berries were soaked in 0.5 mg L-1 24-epibrassinolide solution, and Botrytis cinerea suspension was injected after 6 hours. We reported a novel defense mechanism that BR-regulated autophagy in grape berry against B. cinerea. Exogenous application of 24-epibrassinolide (eBR) enhanced the grape disease resistance. The eBR-treated berries activated an oxidation-reduction (redox), polyamines metabolism, and autophagy during Botrytis infection. Transcript of BR signaling pathway genes were up-regulated within 12 h. In addition, transcriptome analysis in EBR-treated grapes infected with gray mold showed that the differentially expressed genes (DEGs) were enriched in the metabolic pathway of BR signaling pathway and autophagy. Meanwhile, the endogenous BR, SA and JA accumulated transiently. Overexpressing and silencing VvBZR1 in grape significantly affected oxidation, autophagy, and defense response. DNA affinity purification sequencing (Dap-seq), Yeast one-hybrid assay (Y1H) and dual luciferase assays (LUC) verified VvBZR1 transcriptionally regulated the autophagy related gene VvATG18a. Overexpressing VvATG18a improved the resistance of grapes to gray mold. Overall, this study sheds light on the immune mechanisms underlying the involvement of the BR signal transduction pathway in grape innate immunity, highlighting the pivotal role of VvBZR1-mediated autophagy in enhancing disease resistance.

24-epibrassinolide enhances drought tolerance in grapevine (Vitis vinifera L.) by regulating carbon and nitrogen metabolism.

Exogenous application of 24-epibrassinolide can alleviate oxidative damage, improve photosynthetic capacity, and regulate carbon and nitrogen assimilation, thus improving the tolerance of grapevine (Vitis vinifera L.) to drought stress. Brassinosteroids (BRs) are a group of plant steroid hormones in plants and are involved in regulating plant tolerance to drought stress. This study aimed to investigate the regulation effects of BRs on the carbon and nitrogen metabolism in grapevine under drought stress. The results indicated that drought stress led to the accumulation of superoxide radicals and hydrogen peroxide and an increase in lipid peroxidation. A reduction in oxidative damage was observed in EBR-pretreated plants, which was probably due to the improved antioxidant concentration. Moreover, exogenous EBR improved the photosynthetic capacity and sucrose phosphate synthase activity, and decreased the sucrose synthase, acid invertase, and neutral invertase, resulting in improved sucrose (190%) and starch (17%) concentrations. Furthermore, EBR pretreatment strengthened nitrate reduction and ammonium assimilation. A 57% increase in nitrate reductase activity and a 13% increase in glutamine synthetase activity were observed in EBR pretreated grapevines. Meanwhile, EBR pretreated plants accumulated a greater amount of proline, which contributed to osmotic adjustment and ROS scavenging. In summary, exogenous EBR enhanced drought tolerance in grapevines by alleviating oxidative damage and regulating carbon and nitrogen metabolism.

The Exogenous Application of 24-Epibrassinolide (24-EBL) Increases the Cd and Pb Resilience in Zea mays (L.) by Regulating the Growth and Physiological Mechanism.

Heavy metals (HMs) at a concentration above the thresholdlevel act as environmental pollutants and very often threaten theagricultural productivity globally. Finding affordable and environmentally sustainable deliverables to address this issue is therefore a top focus. Phytohormones alleviate the HMs-induced toxicity and positively influence the plant growth. Considering the importance of phytohormones, the present study aimed to assess the effect of 24-epibrassinolide (24-EBL; 10 µM) as seed soaking treatment on growth performance of Zea mays (L.) contaminated separately with increasing concentrations (50-400 mg.kg-1) of lead (Pb) and cadmium (Cd). With increasing metal concentrations, growth and plant biometric criteria were reduced. For instance, Cd at 400 mg.kg-1 soil reduced the germination efficiency (56%), root (77%) and shoot (69%) dry weight, total chlorophyll (64%), and carotenoid content (45%). Contrarily, both HMs caused increase in stress biomarkers and antioxidant enzymes in seedling. However, exogenous administration of 24-EBL significantly enhanced the growth attributes, photosynthetic pigments, proline, MDA, and antioxidant enzyme activity while reducing the harmful effects of HMs stress on Z. mays. For instance, 24-EBL (10 µM) improved the germination percentage, root biomass, chl a, chl b, total chlorophyll, and carotenoid content by 16, 21, 17, 34, 18, and 15%, respectively, in 50 mg.Pb.kg-1 soil-treated Z. mays plants. Furthermore, the amounts of proline, MDA, and antioxidant enzymes in foliage of Z. mays were interestingly and dramatically lowered by 24-EBL application. Uptake of metals in plant organs was significantly reduced when 24-EBL was applied to Pb- and Cd-treated Z. mays. The recent findings help us better understand how 24-EBL regulates growth and development of Z. mays as well as how it boosts HMs' resilience, which could increase the possibility of employing 24-EBL to increase Z. mays productivity. Thus, the present findings confirmed the potentiality of pre-soaking the seed in 24-EBL solution that neutralizes the toxic effects of heavy metals in Z. mays plants. Therefore, it is suggested that applying phytohormones including 24-EBL in removal of heavy metal stress in plants is the best possible solution in sustainable agriculture.

Accumulation of Galactinol and ABA Is Involved in Exogenous EBR-Induced Drought Tolerance in Tea Plants.

Drought stress severely limits growth and causes losses in the yield of tea plants. Exogenous application of 24-epibrassinolide (EBR) positively regulates drought responses in various plants. However, whether EBR could contribute to drought resistance in tea plants and the underlying mechanisms has not been investigated. Here, we found that EBR application is beneficial for the drought tolerance of tea plants. The transcriptome results revealed that EBR could contribute to tea plant drought resistance by promoting galactinol and abscisic acid (ABA) biosynthesis gene expression. The content of galactinol was elevated by EBR and EBR-responsive CsDof1.1 positively regulated the expression of the galactinol synthase genes CsGolS2-1 and CsGolS2-2 to contribute to the accumulation of galactinol by directly binding to their promoters. Moreover, exogenous EBR was found to elevate the expression of genes related to ABA signal transduction and stomatal closure regulation, which resulted in the promotion of stomatal closure. In addition, EBR-responsive CsMYC2-2 is involved in ABA accumulation by binding to the promoters CsNCED1 and CsNCED2 to activate their expression. In summary, findings in this study provide knowledge into the transcriptional regulatory mechanism of EBR-induced drought resistance in tea plants.

The Mechanisms Underlying Salt Resistance Mediated by Exogenous Application of 24-Epibrassinolide in Peanut.

Peanut is one of the most important oil crops in the world, the growth and productivity of which are severely affected by salt stress. 24-epibrassinolide (EBL) plays an important role in stress resistances. However, the roles of exogenous EBL on the salt tolerance of peanut remain unclear. In this study, peanut seedlings treated with 150 mM NaCl and with or without EBL spray were performed to investigate the roles of EBL on salt resistance. Under 150 mM NaCl conditions, foliar application of 0.1 µM EBL increased the activity of catalase and thereby could eliminate reactive oxygen species (ROS). Similarly, EBL application promoted the accumulation of proline and soluble sugar, thus maintaining osmotic balance. Furthermore, foliar EBL spray enhanced the total chlorophyll content and high photosynthesis capacity. Transcriptome analysis showed that under NaCl stress, EBL treatment up-regulated expression levels of genes encoding peroxisomal nicotinamide adenine dinucleotide carrier (PMP34), probable sucrose-phosphate synthase 2 (SPS2) beta-fructofuranosidase (BFRUCT1) and Na+/H+ antiporters (NHX7 and NHX8), while down-regulated proline dehydrogenase 2 (PRODH). These findings provide valuable resources for salt resistance study in peanut and lay the foundation for using BR to enhance salt tolerance during peanut production.

Effect of Exogenously Applied 24-Epibrassinolide and Brassinazole on Xylogenesis and Microdistribution of Cell Wall Polymers in Leucaena leucocephala (Lam) De Wit

Plant growth regulators play a key role in cell wall structure and chemistry of woody plants. Understanding of these regulatory signals is important in advanced research on wood quality improvement in trees. The present study is aimed to investigate the influence of exogenous application of 24-epibrassinolide (EBR) and brassinosteroid inhibitor, brassinazole (BRZ) on wood formation and spatial distribution of cell wall polymers in the xylem tissue of Leucaena leucocephala using light and immuno electron microscopy methods. Brassinazole caused a decrease in cambial activity, xylem differentiation, length and width of fibres, vessel element width and radial extent of xylem suggesting brassinosteroid inhibition has a concomitant impact on cell elongation, expansion and secondary wall deposition. Histochemical studies of 24-epibrassinolide treated plants showed an increase in syringyl lignin content in the xylem cell walls. Fluorescence microscopy and transmission electron microscopy studies revealed the inhomogenous pattern of lignin distribution in the cell corners and middle lamellae region of BRZ treated plants. Immunolocalization studies using LM10 and LM 11 antibodies have shown a drastic change in the micro-distribution pattern of less substituted and highly substituted xylans in the xylem fibres of plants treated with EBR and BRZ. In conclusion, present study demonstrates an important role of brassinosteroid in plant development through regulating xylogenesis and cell wall chemistry in higher plants.

Study the effect of 24-epibrassinolide application on the Cu/Zn-SOD expression and tolerance to drought stress in common bean.

To study the possibility of increasing the drought tolerance of common bean with the exogenous application of 24-epibrassinolide (EBL), an experiment was conducted in 2016 and 2017. In this experiment, two irrigation levels (optimal irrigation and drought stress) were applied to the main plots and two common bean genotypes (Kusha cultivar and COS16 genotype) and four EBL concentrations (0, 2, 4, and 6μM) were allocated to sub-plots as factorial. In the flowering stage, drought stress was applied and plants were sprayed with EBL. The results showed that drought stress reduced relative water content (RWC) and increased proline content, malondialdehyde (MDA) content, and antioxidant enzymes activity. However, exogenous application of EBL reduced the seed yield loss and increased the drought stress tolerance in both common bean genotypes by decreasing the MDA content and increasing the RWC, proline content, antioxidant enzymes activity, and nitrate reductase activity. It can beconcluded that foliar spray of 4µM EBL as the best concentration may increase the seed yield and enhance the drought stress tolerance of common bean. Also, Cu/Zn-SOD was up-regulated in response to the drought stress and exogenous EBL. The COS16 genotype showed better response to the drought stress and exogenous EBL than the Kusha cultivar, because of the higher up-regulation of Cu/Zn-SOD in this genotype compared to the Kusha cultivar. Therefore, EBL can be used as a plant growth regulator to enhance drought stress tolerance and minimize the seed yield loss of common bean caused by water deficit.

Improvement in photosynthesis, seed yield and protein content of common bean (Phaseolus vulgaris) by foliar application of 24-epibrassinolide under drought stress

Brassinosteroids (BRs), as a class of plant growth regulators, have been shown to affect different physiological traits of plants and counteract various stresses. We studied the possibility of increasing seed and protein yields of two genotypes of common bean (Phaseolus vulgaris L.) with the exogenous application of 24-epibrassinolide (EBL) in an experiment conducted during 2016 and 2017. Two levels of irrigation (optimal and drought stress) were applied to the main plots, and two genotypes of common bean (cv. Kusha and genotype COS16) and four concentrations of EBL (0, 2, 4 and 6 μm) were allocated to subplots in a factorial arrangement. At the flowering stage, drought stress was applied and bean plants were sprayed with EBL. The results indicate that drought stress reduced leaf area, yield components, seed yield and protein content. Moreover, substantial increase in intercellular CO2 concentration and decrease in transpiration rate, stomatal conductance and net photosynthetic rate were also recorded. However, exogenous application of EBL remarkably improved gas exchange attributes, leaf area, yield components, seed yield and protein content both under optimal irrigation and drought-stress conditions. Analysis of regression showed that, under both water-supply conditions, genotype COS16 would have highest seed yield when receiving 4.05 and 4.52 µm EBL, and cv. Kusha would have the highest seed yield by receiving 3.27 and 3.62 µm EBL. Therefore, EBL can be used as a plant growth regulator to enhance drought tolerance and minimise yield loss of common bean caused by water deficits.

Brassinosteroids (BRs) are steroid hormones that modulate numerous physiological processes in plants. However, few studies have focused on the involvement of BRs in sensing and responding to the stress of mineral nutrient deficiency. In the present study, we evaluated the roles of BRs in the response of rice (Oryza sativa) to iron (Fe) deficiency during Fe uptake, transport, and translocation. Exogenous application of 24-epibrassinolide (EBR) to wild-type (WT) plants exaggerated leaf symptoms of Fe deficiency and suppressed growth. EBR increased and decreased Fe concentrations in roots and shoots, respectively, under both Fe-deficient and Fe-sufficient conditions. Transcripts involved in Fe homeostasis, including OsIRT1, OsYSL15, OsYSL2, OsNAS1, and OsNAS2, were enhanced by EBR under Fe-deficient conditions. EBR depressed expression of OsNAS1, OsNAS2, and OsYSL2 in shoots, and inhibited Fe transport and translocation via the phloem. Rice mutant d2-1, which is defective in BR biosynthesis, was more tolerant to Fe deficiency than the WT, and accumulated greater amounts of Fe in roots than the WT under Fe-sufficient conditions. A greater upregulation of OsIRT1, OsYSL15, OsYSL2, OsNAS1, and OsNAS2 in the d2-1 mutant compared to the WT was found under Fe-sufficient conditions, while expression of these genes in the d2-1 mutant was lower than in the WT under Fe-deficient conditions. The greater tolerance of the d2-1 mutant could be partly mitigated by exogenous application of EBR. These novel findings highlight the important role of BR in mediating the response of strategy II plants to Fe deficiency by regulating Fe uptake and translocation in rice.

Plant steroidal hormones, brassinosteroids, play a pivotal role in variety of plant developmental processes and adaptation to various environmental stresses. The present work investigates the response of various stress markers upon exogenous application of 24-epibrassinolide (EBL) on Pusa Basmati-1, a commercially important rice variety, under salt stress conditions. Rice seeds after treatment with different concentrations of NaCl alone or in combination with different concentrations of 24-epibrassinolide (EBL) were analysed for various growth parameters, protein, proline and malondialdehyde content (MDA) and antioxidant enzymes activities. The seedlings exposed to NaCl exhibited a significant decline in growth parameters and changes in the levels of antioxidant enzymes, however, treatment with EBL showed an improvement in growth, levels of protein and proline content and antioxidant enzymes activity. The enhanced levels of MDA content during salt stress in rice seedlings was decreased with EBL treatment. Further, the treatment with EBL increased the expression of various oxidative stress marker genes, although to different levels. Expression of various brassinosteroids (OsBRI1, OsDWF4) and salt (SalT) responsive genes, revealed the down regulation of OsDWF4 with application of EBL and upregulation of SalT in presence of salt stress thereby confirming the efficacy of the treatments. Interestingly, a significant down regulation of SalT gene was observed on application of EBL along with salt compared to salt treatment alone. On the other hand, the application of EBL alone and in combination with salt has resulted in upregulation of OsBRI1.
