Computational simulations and Ca2+ imaging reveal that slow synaptic depolarizations (slow EPSPs) inhibit fast EPSP evoked action potentials for most of their time course in enteric neurons.

Transmission between neurons in the extensive enteric neural networks of the gut involves synaptic potentials with vastly different time courses and underlying conductances. Most enteric neurons exhibit fast excitatory post-synaptic potentials (EPSPs) lasting 20-50 ms, but many also exhibit slow EPSPs that last up to 100 s. When large enough, slow EPSPs excite action potentials at the start of the slow depolarization, but how they affect action potentials evoked by fast EPSPs is unknown. Furthermore, two other sources of synaptic depolarization probably occur in enteric circuits, activated via GABAA or GABAC receptors; how these interact with other synaptic depolarizations is also unclear. We built a compartmental model of enteric neurons incorporating realistic voltage-dependent ion channels, then simulated fast EPSPs, slow EPSPs and GABAA or GABAC ligand-gated Cl- channels to explore these interactions. Model predictions were tested by imaging Ca2+ transients in myenteric neurons ex vivo as an indicator of their activity during synaptic interactions. The model could mimic firing of myenteric neurons in mouse colon evoked by depolarizing current during intracellular recording and the fast and slow EPSPs in these neurons. Subthreshold fast EPSPs evoked spikes during the rising phase of a slow EPSP, but suprathreshold fast EPSPs could not evoke spikes later in a slow EPSP. This predicted inhibition was confirmed by Ca2+ imaging in which stimuli that evoke slow EPSPs suppressed activity evoked by fast EPSPs in many myenteric neurons. The model also predicted that synchronous activation of GABAA receptors and fast EPSPs potentiated firing evoked by the latter, while synchronous activation of GABAC receptors with fast EPSPs, potentiated firing and then suppressed it. The results reveal that so-called slow EPSPs have a biphasic effect being likely to suppress fast EPSP evoked firing over very long periods, perhaps accounting for prolonged quiescent periods seen in enteric motor patterns.

Electrophysiological alterations in subthalamic neurons after unilateral dopamine depletion in the rat

The present study examined the effects of unilateral 6-hydroxydopamine (6-OHDA) lesions of the substantia nigra pars compacta (SNc) on electrophysiological properties of subthalamic neurons (STN) in adult rats. Most neurons displayed regular spontaneous tonic firing patterns in both control and lesioned animals; however, the percentage of neurons with spontaneous burst firing at hyperpolarized membrane potentials was increased significantly in lesioned animals compared with controls (45% vs. 14% respectively). In the presence of bicuculline, a gamma-aminobutyric acid type A (GABAA) receptor antagonist, electrical stimulation of the internal capsule produced monosynaptic excitatory postsynaptic potentials (EPSPs) in almost all recorded neurons. DA (50 microM) increased the amplitude and/or duration of the EPSPs in neurons from both groups, whereas the DA D1 receptor agonist SKF 81297 (10 microM) produced a significant increase in amplitude and/or duration of EPSPs in neurons from the lesioned group only. This latter increase was blocked by pretreatment with the DA D1 antagonist SCH 23390 (10 microM). These data suggest that unilateral degeneration of DA neurons in the SNc changes firing properties and enhances electrophysiological responsiveness of STN neurons to activation of DA D1 receptors.

Long-term potentiation of thalamic input to the motor cortex induced by coactivation of thalamocortical and corticocortical afferents

1. Intracellular recordings were obtained from neurons in the motor cortex (MCx), in which excitatory postsynaptic potentials (EPSPs) were evoked by microstimulation of the somatosensory cortex (SCx) and the ventrolateral nucleus (VL) of the thalamus. The effects of combined tetanic stimulation of SCx and VL on the amplitudes of these EPSPs were studied. 2. Amplitudes of both corticocortical (CC) and thalamocortical (TC) EPSPs were potentiated after combined tetanic stimulation. This potentiation occurred exclusively in neurons that were located in the superficial layers (II/III) and that received direct input from both the SCx and VL, with both inputs synapsing in close proximity to each other. In all cases, the potentiation lasted until the electrode went out of the cell (21 +/- 25 min, mean +/- SD) the longest being 90 min. We therefore refer to this potentiation as long-term potentiation (LTP). 3. Tetanic stimulation of the thalamus only did not produce LTP in neurons receiving direct input from the VL. 4. LTP was not induced in either CC or TC EPSPs in neurons located in layer V and/or in neurons receiving long-latency CC EPSPs. 5. It is concluded that TC input from the VL to the MCx is potentiated only when coactivated with the CC input from the SCx.

Statistical study on reverberation between two mutually excitatory neuron groups

The reverberation that occurs between two neuron groups, which have excitatory mono-synaptic random connections with each other can be studied theoretically by employing a model neuron, which expresses well the characters of a real neuron. In this model we consider three effects, which are; the effect of the summation of the excitatory post-synaptic potential (EPSP) of neurons; the effect of the spontaneous firing of neurons as a noise in groups and the effect of the relative refractory period of neurons. As a result, it is shown that under the effect of the summation of the EPSP of neurons and the effect of the noise, the systematic threshold p ϑ takes the same value as is observed in practice. The effect of the relative refractory period has been considered in order to explain the low speed of the increase in firing activity, as observed in the reverberating system. It suppresses slightly the speed of the increase in firing activity ( p i ) in the system. Moreover, the speed can be suppressed by making the refractory effect strong according to the increase of p i . However, the initial increase of p i at a high speed that was observed in the experiment cannot be explained simply by the effect of the refractoriness, even if it were the absolute refractoriness.
