Simple Coumarins from Peucedanum luxurians Fruits: Evaluation of Anxiolytic Activity and Influence on Gene Expression Related to Anxiety in Zebrafish Model.

Anxiety is one of the most common central nervous system disorders, affecting at least one-quarter of the worldwide population. The medications routinely used for the treatment of anxiety (mainly benzodiazepines) are a cause of addiction and are characterized by many undesirable side effects. Thus, there is an important and urgent need for screening and finding novel drug candidates that can be used in the prevention or treatment of anxiety. Simple coumarins usually do not show side effects, or these effects are much lower than in the case of synthetic drugs acting on the central nervous system (CNS). This study aimed to evaluate the anxiolytic activity of three simple coumarins from Peucedanum luxurians Tamamsch, namely officinalin, stenocarpin isobutyrate, and officinalin isobutyrate, in a 5 dpf larval zebrafish model. Moreover, the influence of the tested coumarins on the expression of genes involved in the neural activity (c-fos, bdnf) or dopaminergic (th1), serotoninergic (htr1Aa, htr1b, htr2b), GABA-ergic (gabarapa, gabarapb), enkephalinergic (penka, penkb), and galaninergic (galn) neurotransmission was assessed by quantitative PCR. All tested coumarins showed significant anxiolytic activity, with officinalin as the most potent compound. The presence of a free hydroxyl group at position C-7 and the lack of methoxy moiety at position C-8 might be key structural features responsible for the observed effects. In addition, officinalin and its isobutyrate upregulated the expression of genes involved in neurotransmission and decreased the expression of genes connected with neural activity. Therefore, the coumarins from P. luxurians might be considered as promising drug candidates for the therapy of anxiety and related disorders.

Background: Anxiety disorders are most common mental illness, characterized by changes in mood, behavior, somatic function, and cognition. At present, benzodiazepines are most commonly used drugs for treating anxiety and are associated with many side effects. Medicinal plants are inexhaustible source for searching new drug or new activity, with comparatively lower side effects and drug interactions. Aim and Objectives: The aim of the study was to evaluate the anxiolytic activity of Ajwain oil alone and combination of Diazepam and Ajwain oil (in subthreshold doses), in Swiss albino mice. Materials and Methods: A total of 24 Swiss albino mice of either sex weighing 20–30 g were included in the study and were divided into four groups of six animals each. Among which, Group 1 (control) received – tween 80 (10 mg/kg), Group 2 (standard) – Diazepam (1 mg/kg), Group 3 (test-1) – Ajwain oil (50 mg/kg), and Group 4 (test-2) – Diazepam (0.5 mg/kg) + Ajwain oil (25 mg/kg). After 60 min of oral administration of drugs, anxiolytic activity was assisted using elevated plus maze (EPM) and Digital Actophotometer. Results: Statistical analysis was performed using ANOVA, followed by post hoc Tukey’s test. Ajwain oil showed significant increase in number of entries and time spent in open arm in EPM and in Digital Actophotometer there was decrease in locomotor activity when compared to control. In Group 4, where we used subthreshold dose of Ajwain oil and diazepam, number of entries in EPM and decrease in locomotor activity in Actophotometer showed significant results when compared to standard. Conclusion: Hence, with the above results, we suggest Ajwain oil possesses significant anxiolytic activity.

Evaluation of Anxiolytic Activity of Ixora coccinea Linn. Ethanolic Extract in Swiss Albino Mice

Ixora coccinea Linn. (Family: Rubiaceae), a small to medium sized hardy shrub is cultivated for ornamental purpose and also it finds place in traditional Indian medicine. The objective of this study was to evaluate the anxiolytic activity of Ixora coccinea ethanolic extract. The methods used for this study are elevated plus maze paradigm test and Hole board test. The ethanolic extract ICEE shows a significant (P<0.01) anxiolytic effect during the elevated plus maze test and the hole board test in a dose dependent manner when compared with the standard dose of diazepam.
