The Role of Alpha Linolenic Acid on Neuroinflammation: A Systemic Review

Neuroinflammation, an inflammatory response within the central nervous system (CNS), plays a critical role in various neurological disorders, including neurodegenerative diseases. Alpha linolenic acid (ALA), an essential omega-3 polyunsaturated fatty acid, has shown promising health benefits due to its antioxidant and anti-inflammatory properties. This article aims to thoroughly examine the potential effects of ALA in reducing neuroinflammation, emphasizing its roles as an anti-inflammatory and antioxidant agent. The review focuses on studies published between 2018 and 2023, sourced from reputable academic databases such as PubMed, Semantic Scholar, Google Scholar, and ScienceDirect, with nine key papers selected. The findings indicate that ALA significantly mitigates neuroinflammation by decreasing reactive oxygen species (ROS) via the Nrf-2/HO-1/JNK signaling pathway. Additionally, ALA reduces inflammatory cytokines by inhibiting p-JNK activation, disrupting lipid rafts, blocking the pro-inflammatory transcription factor NF-κB, and altering cell membrane phospholipid composition. The review concludes that ALA may have potential therapeutic effects in reducing neuroinflammation through its antioxidant and anti-inflammatory actions, offering possible benefits for various neurological conditions, including Alzheimer's and Parkinson's diseases.

Omega-3 fatty acids-critical for the marine food web and for seabird productivity

Abstract Seabird reproductive success in northern latitudes is often linked with prey abundance, availability, size, or species. Causes are often broadly explained as changes in ocean currents/temperatures, or diets. Few studies trace diets down the food web to primary producers, thus missing what I propose as an underlying cause of seabird colony failure, dietary abundance of essential omega-3 polyunsaturated fatty acids (PUFA). PUFA differ from other nutrients because they are, in part, ligands—critical for reproduction and other physiological processes for the entire marine food web, not just for seabirds. Diatoms are one of the few life forms that can produce PUFA de novo, and a lack of PUFA in zooplankton and fish reduces their abundance and productivity, with consequences up the food web. Since the mid-1980s, historical prey of breeding seabirds has decreased, and frequency of colony failures has increased. In years of failure, prey often had less fat than historically preferred species. Likewise, proportions of alternate species’ PUFA, from published papers, show lower values than in historical prey. Production of PUFA by diatoms is depressed in warmer and more acidic environments, and I hypothesize that these warmer and more acidic seas have affected production of PUFA over the past 3–4 decades. Assuming this is true, I propose that these lower amounts of PUFA have negatively impacted the breeding success of all members of the marine food web, not just seabirds.

Looking into the lipid profile of avocado and byproducts: Using lipidomics to explore value-added compounds.

Consumer priorities in healthy diets and lifestyle boosted the demand for nutritious and functional foods as well as plant-based ingredients. Avocado has become a food trend due to its nutritional and functional values, which in turn is increasing its consumption and production worldwide. Avocado edible portion has a high content of lipids, with the pulp and its oil being rich in monounsaturated fatty acids and essential omega-3 and omega-6 polyunsaturated fatty acids (PUFA). These fatty acids are mainly esterified in triacylglycerides, the major lipids in pulp, but also in minor components such as polar lipids (phospholipids and glycolipids). Polar lipids of avocado have been overlooked despite being recently highlighted with functional properties as well. The growth in the industry of avocado products is generating an increased amount of their byproducts, such as seed and peels (nonedible portions), still undervalued. The few studies on avocado byproducts pointed out that they also contain interesting lipids, with seeds particularly rich in polar lipids bearing PUFA, and thus can be reused as a source of add-value phytochemical. Mass spectrometry-based lipidomics approaches appear as an essential tool to unveil the complex lipid signature of avocado and its byproducts, contributing to the recognition of value-added lipids and opening new avenues for their use in novel biotechnological applications. The present review provides an up-to-date overview of the lipid signature from avocado pulp, peel, seed, and its oils.

Unwrapping the (glyco-)lipidome in the microalgae Microchloropsis gaditana: Effects of eco-friendly extraction methods

Microalgae have recently captivated worldwide attention because of their extraordinary lipid composition, which includes essential omega-3 and omega-6 long-chain polyunsaturated fatty acids (LC-PUFA), neutral lipids, and more complex lipids (glycolipids and phospholipids), as well as eicosanoids. To explore this further, we present a novel (glyco-)lipidomics strategy, including a green chemistry extraction approach, liquid chromatography high-resolution mass spectrometry (LC-HRMS) lipid profiling, and a comprehensive data evaluation workflow. Using untargeted lipidomics based on LC-HRMS approaches, we studied the lipid composition up to the molecular species level of the microalgae Microchloropsis gaditana (formerly Nannochloropsis gaditana), focusing on the distribution of saturated, monounsaturated, and polyunsaturated fatty acids in different lipid classes. Our analysis successfully identified 520 lipid molecular species from 18 lipid classes. Regarding fatty acid composition, our primary focus was on elucidating the distribution of LC-PUFA across the different microalgal lipid classes. This particular strategy holds potential as it may unravel the use of microalgal lipids as innovative sources of food ingredients. Our findings show that LC-PUFA are relatively elevated in membrane lipids, in which cholesterol esters (ChE) have the highest PUFA share (94 %), followed by polar membrane lipids (glycolipids and phospholipids), with phosphatidylethanolamines (PE) up to 67 %, phosphatidylglycerols (PG) up to 47 %, and monogalactosyldiacylglycerols (MGDG) up to 47 % PUFA content. This highlights the high percentage of PUFA in the structural parts of microalgae and the importance of these lipid classes as a reservoir for PUFA. Finally, we also compared a classical Folch extraction method with environmentally friendly ultrasound-assisted extraction approaches and enzymatic pre-treatments to investigate the lipid composition and enrichment of valuable lipid classes. Thus, this extensive analysis of the (glyco-)lipidome of Microchloropsis gaditana provides valuable insights into the existing knowledge about this microalgae species, enhancing our understanding of the complex lipid composition and opening up new opportunities for potential applications in the biotechnology and food industry.

The potential effect of Alpha-linolenic acid against oxidative stress in non-alcoholic fatty liver disease

Non-alcoholic Fatty Liver Disease (NAFLD) is characterized by fat accumulation in hepatocytes without secondary causes. Oxidative stress plays an important role in forming and progressing from NAFLD to non-alcoholic steatohepatitis (NASH). Alpha-linolenic acid (ALA) is an essential omega-3 (n-3) polyunsaturated fatty acid (PUFA) that has a high antioxidant effect. The purpose of writing this article is to determine the potential of ALA on oxidative stress in NAFLD. This article was made using an online search from 2013 to 2023, and six articles were obtained. From this review, we concluded that ALA has an antioxidant effect that can reduce the impact of oxidative stress in NAFLD, especially in liver tissue, which provides a hepatoprotective effect and regulates gene expression that can reduce oxidative stress in NAFLD.

Decreased DHA-containing phospholipids in the neocortex of dementia with Lewy bodies are associated with soluble Aβ42 , phosphorylated α-synuclein, and synaptopathology.

Docosahexaenoic acid (DHA) is an essential omega-3 polyunsaturated fatty acid implicated in cognitive functions by promoting synaptic protein expression. While alterations of specific DHA-containing phospholipids have been described in the neocortex of patients with Alzheimer's disease (AD), the status of these lipids in dementia with Lewy bodies (DLB), known to manifest aggregated α-synuclein-containing Lewy bodies together with variable amyloid pathology, is unclear. In this study, post-mortem samples from the parietal cortex of 25 DLB patients and 17 age-matched controls were processed for phospholipidomics analyses using a liquid chromatography-tandem mass spectrometry (LC-MS/MS) platform. After controlling for false discovery rate, six out of the 46 identified putative DHA-phospholipid species were significantly decreased in DLB, with only one showing increase. Altered putative DHA-phospholipid species were subsequently validated with further LC-MS/MS measurements. Of the DHA-containing phospholipid (DCP) species showing decreases, five negatively correlated with soluble beta-amyloid (Aβ42) levels, whilst three also correlated with phosphorylated α-synuclein (all p < 0.05). Furthermore, five of these phospholipid species correlated with deficits of presynaptic Rab3A, postsynaptic neurogranin, or both (all p < 0.05). Finally, we found altered immunoreactivities of brain lysolipid DHA transporter, MFSD2A, and the fatty acid binding protein FABP5 in DLB parietal cortex. In summary, we report alterations of specific DCP species in DLB, as well as their associations with markers of neuropathological burden and synaptopathology. These results support the potential role of DHA perturbations in DLB as well as therapeutic targets.

The second life of terrestrial and plastic carbon as nutritionally valuable food for aquatic consumers.

Primary production is the basis for energy and biomolecule flow in food webs. Nutritional importance of terrestrial and plastic carbon via mixotrophic algae to upper trophic level is poorly studied. We explored this question by analysing the contribution of osmo- and phagomixotrophic species in boreal lakes and used 13 C-labelled materials and compound-specific isotopes to determine biochemical fate of carbon backbone of leaves, lignin-hemicellulose and polystyrene at four-trophic level experiment. Microbes prepared similar amounts of amino acids from leaves and lignin, but four times more membrane lipids from lignin than leaves, and much less from polystyrene. Mixotrophic algae (Cryptomonas sp.) upgraded simple fatty acids to essential omega-3 and omega-6 polyunsaturated fatty acids. Labelled amino and fatty acids became integral parts of cell membranes of zooplankton (Daphnia magna) and fish (Danio rerio). These results show that terrestrial and plastic carbon can provide backbones for essential biomolecules of mixotrophic algae and consumers at higher trophic levels.

Docosahexaenoic acid promotes M2 microglia phenotype via activating PPARγ-mediated ERK/AKT pathway against cerebral ischemia-reperfusion injury

In ischemia-reperfusion stroke, microglia play a dual role in brain injury as well as brain repair, and promoting their switch from a pro-inflammatory M1 phenotype to an anti-inflammatory M2 phenotype is considered to be a potential therapeutic strategy. Docosahexaenoic acid (DHA) is an essential long-chain omega-3 polyunsaturated fatty acid that exhibits potent anti-inflammatory properties in the acute phase of ischemic stroke, but its effect on microglia polarization is unknown. Thus, the objective of this study was to investigate the neuroprotective effects of DHA on rat brain following ischemia-reperfusion injury, and to investigate the mechanism by which DHA regulates microglia polarization. We administered DHA 5 mg/kg intraperitoneally daily for 3 d following a transient middle cerebral artery occlusion reperfusion model in rats. The protective effects of DHA on cerebral ischemia-reperfusion injury were detected by TTC staining, HE staining, Nissler staining, and TUNEL staining. Quantitative real-time PCR, immunofluorescence, western blot, and enzyme-linked immunosorbent assay were used to detect the expression of M1 and M2 microglia-associated markers and PPARγ-mediated ERK/AKT signaling pathway proteins. We found that DHA significantly improved brain injury by decreasing the expression of the M1 phenotypic marker (iNOS, CD16) and increasing the expression of the M2 phenotypic marker (Arg-1, CD206). DHA also increased the expression of peroxisome proliferator-activated receptor gamma (PPARγ) mRNA and protein, increased the expression of the pathway protein AKT, and decreased the expression of ERK1/2. In addition, DHA promoted the expression of anti-inflammatory factor IL-10 and decreased the expression of pro-inflammatory factors TNF-α and IL-1β. However, the PPARγ antagonist GW9662 greatly blocked these beneficial effects. These results suggest that DHA may activate PPARγ to inhibit ERK and activate AKT signaling pathways to regulate microglia polarization, thereby reducing neuroinflammation and promoting neurological recovery to alleviate cerebral ischemia-reperfusion injury.

Can Glyphosate and Temperature Affect the Nutritional Lipid Quality in the Mussel Mytilus galloprovincialis?

Mussels are an important source of the essential omega-3 polyunsaturated fatty acids (n-3 PUFAs), which play a critical role in human health, preventing a variety of diseases. This study aimed to evaluate, for the first time, the combined effect of glyphosate (Gly) and culturing temperature on the lipid content and fatty acid (FA) profile of the Mediterranean mussel M. galloprovincialis. In addition, a number of lipid nutritional quality indices (LNQIs) were applied as important tools to assess the nutritional value of food. Mussels were exposed for 4 days to two Gly concentrations (1 mg/L, 10 mg/L) and two temperatures (T°: 20-26 °C). Statistical analysis showed significant effects of T°C, Gly, and T°C × Gly interaction (p < 0.05) on the lipid and FA profiles of M. galloprovincialis. Mussels exposed to 10 mg/L Gly at 20 °C showed a decrease in eicosapentaenoic (EPA, from 14.6% to 12% of total FAs) and docosahexaenoic acids (DHA, from 10% to 6.4% of total FAs), compared to the control mussels. Both stressors caused a considerable decrease in n-3 PUFAs, which resulted in a less favorable n-6/n-3 PUFA ratio. Overall, this study demonstrated a decline in the nutritive values of mussels, most prominently in groups exposed to 10 mg/L Gly at a temperature of 20 °C and in those exposed to a temperature of 26 °C. This was confirmed by such LNQIs as EPA + DHA, PUFA/Saturated FAs, atherogenic and thrombogenic indices (AI and TI), the health promoting index (HPI), and the unsaturation index (UI). Further investigations into chronic exposure to both stressors are desirable to predict the impacts on aquatic ecosystems and food quality.

Alpha-linolenic acid enhances the facilitation of GABAergic neurotransmission in the BLA and CA1.

Hyperexcitability is a major mechanism implicated in several neuropsychiatric disorders, such as organophosphate-induced status epilepticus (SE), primary epilepsy, stroke, spinal cord injury, traumatic brain injury, schizophrenia, and autism spectrum disorders. Underlying mechanisms are diverse, but a functional impairment and loss of GABAergic inhibitory neurons are common features in many of these disorders. While novel therapies abound to correct for the loss of GABAergic inhibitory neurons, it has been difficult at best to improve the activities of daily living for the majority of patients. Alpha-linolenic acid (ALA) is an essential omega-3 polyunsaturated fatty acid found in plants. ALA exerts pleiotropic effects in the brain that attenuate injury in chronic and acute brain disease models. However, the effect of ALA on GABAergic neurotransmission in hyperexcitable brain regions involved in neuropsychiatric disorders, such as the basolateral amygdala (BLA) and CA1 subfield of the hippocampus, is unknown. Administration of a single dose of ALA (1500 nmol/kg) subcutaneously increased the charge transfer of inhibitory postsynaptic potential currents mediated by GABAA receptors in pyramidal neurons by 52% in the BLA and by 92% in the CA1 compared to vehicle animals a day later. Similar results were obtained in pyramidal neurons from the BLA and CA1 when ALA was bath-applied in slices from naïve animals. Importantly, pretreatment with the high-affinity, selective TrkB inhibitor, k252, completely abolished the ALA-induced increase in GABAergic neurotransmission in the BLA and CA1, suggesting a brain-derived neurotrophic factor (BDNF)-mediated mechanism. Addition of mature BDNF (20 ng/mL) significantly increased GABAA receptor inhibitory activity in the BLA and CA1 pyramidal neurons similar to the results obtained with ALA. ALA may be an effective treatment for neuropsychiatric disorders where hyperexcitability is a major feature.

Expression of glycerol-3-phosphate acyltransferase increases non-polar lipid accumulation in Nannochloropsis oceanica

Microalgae are considered a suitable production platform for high-value lipids and oleochemicals. Several species including Nannochloropsis oceanica produce large amounts of essential omega-3 polyunsaturated fatty acids (PUFAs) which are integral components of food and feed and have been associated with health-promoting effects. N. oceanica can further accumulate high contents of non-polar lipids with chemical properties that render them a potential replacement for plant oils such as palm oil. However, biomass and lipid productivities obtained with microalgae need to be improved to reach commercial feasibility. Genetic engineering can improve biomass and lipid productivities, for instance by increasing carbon flux to lipids. Here, we report the overexpression of glycerol-3-phosphate acyltransferase (GPAT) in N. oceanica during favorable growth conditions as a strategy to increase non-polar lipid content. Transformants overproducing either an endogenous (NoGPAT) or a heterologous (Acutodesmus obliquus GPAT) GPAT enzyme targeted to the endoplasmic reticulum had up to 42% and 51% increased non-polar lipid contents, respectively, compared to the wild type. Biomass productivities of transformant strains were not substantially impaired, resulting in lipid productivities that were increased by up to 37% and 42% for NoGPAT and AoGPAT transformants, respectively. When exposed to nutrient stress, transformants and wild type had similar lipid contents, suggesting that GPAT enzyme exerts strong flux control on lipid synthesis in N. oceanica under favorable growth conditions. NoGPAT transformants further accumulated PUFAs in non-polar lipids, reaching a total of 6.8% PUFAs per biomass, an increase of 24% relative to the wild type. Overall, our results indicate that GPAT is an interesting target for engineering of lipid metabolism in microalgae, in order to improve non-polar lipid and PUFAs accumulation in microalgae.

Labelling of eicosapentaenoic acid with stable isotope 13C in the marine bacterium Shewanella marinintestina

Eicosapentaenoic acid (EPA) is an essential omega-3 polyunsaturated fatty acid that plays a critical role in marine life. It is present in several marine animals, including fish, but the primary producers of EPA are phytoplankton and specific marine bacteria. Although most of the EPA present in marine animals come from phytoplankton, the bacterial input into the marine EPA food web is still unknown. The labelling of EPA within a bacterial strain could be a viable strategy to help revealing this contribution. In this work, Shewanella marinintestina IRL 567, a marine bacterium isolated from fish guts and known to produce EPA, was labelled with the stable isotope 13C at small (250-mL shake flask), bench (2.5-L shake flask), and pilot scale (50-L stirred tank bioreactor). Growing the bacterium with 13C-acetate in the culture medium demonstrated that EPA was de-novo synthesized utilizing acetate as precursor. 13C incorporation into the EPA molecule resulted in values as high as 95.5% of the synthesized EPA being labelled in small scale, 95.9% in bench scale and 91.5% in pilot scale. This simple method to label EPA proved to be effective and therefore it could be a valuable tool to follow the fate of bacterial EPA into higher trophic levels.

In Vivo Antitumoral Effects of Linseed Oil and Its Combination With Doxorubicin.

Linseed oil (LO) is known for its exceptional nutritional value due to the high content of alpha-linolenic acid (ALA), an essential omega-3 polyunsaturated fatty acid; its anticarcinogenic effect has been established in several experimental and epidemiological studies. As an adjuvant of chemotherapeutic agents, LO and other ALA-rich vegetable oils have been studied in only a handful of studies at the experimental level. However, the efficacy of antitumoral therapy using doxorubicin (Dox) in combination with ALA and ALA-rich substrates has not yet been investigated. In this work, the antitumor activity of LO in a wide dose range was studied with monotherapy and combined with Dox in animal models with Pliss lymphosarcoma (PLS) and Lewis lung adenocarcinoma (LLC). It was founded the daily oral administration of LO (1, 3, and 10 ml per 1 kg) to rats (PLS) and 6 ml/kg to mice (LLC) for 11–12 days from 7 days after subcutaneous transplantation of tumors has a stable statistically significant effect on the dynamics of tumor growth, reducing the intensity of tumor growth and increasing the frequency of complete tumor regressions (CR) compared with the control. LO showed high antimetastatic activity in the LLC model. Furthermore, LO at a dose of 3 ml/kg potentiates the antitumor effect of Dox in the PLS model, reducing the volume of tumors at the end of treatment by 2.0 times (p = 0.013), the value of the tumor growth index by 1.6 times (p < 0.03) and increasing the frequency of CR 60 days after the start of therapy by 3.5 times (p = 0.019) compared with the use of Dox alone. The combination of Dox and LO or fish oil allows growing efficiency therapy of LLC in comparison with Dox alone, increasing the frequency of CR to 73.68% and 94.4%, respectively, and reducing the frequency of metastasis to zero.

Interpreting Clinical Trials With Omega-3 Supplements in the Context of Ancestry and FADS Genetic Variation.

Human diets in developed countries such as the US have changed dramatically over the past 75 years, leading to increased obesity, inflammation, and cardiometabolic dysfunction. Evidence over the past decade indicates that the interaction of genetic variation with changes in the intake of 18-carbon essential dietary omega-6 (n-6) and omega-3 (n-3) polyunsaturated fatty acids (PUFA), linoleic acid (LA) and α-linolenic acid (ALA), respectively, has impacted numerous molecular and clinical phenotypes. Interactions are particularly relevant with the FADS1 and FADS2 genes, which encode key fatty acid desaturases in the pathway that converts LA and ALA to their long chain (≥20 carbons), highly unsaturated fatty acid (HUFA) counterparts. These gene by nutrient interactions affect the levels and balance of n-6 and n-3 HUFA that in turn are converted to a wide array of lipids with signaling roles, including eicosanoids, docosanoids, other oxylipins and endocannabinoids. With few exceptions, n-6 HUFA are precursors of pro-inflammatory/pro-thrombotic signaling lipids, and n-3 HUFA are generally anti-inflammatory/anti-thrombotic. We and others have demonstrated that African ancestry populations have much higher frequencies (vs. European-, Asian- or indigenous Americas-ancestry populations) of a FADS “derived” haplotype that is associated with the efficient conversion of high levels of dietary n-6 PUFA to pro-inflammatory n-6 HUFA. By contrast, an “ancestral” haplotype, carrying alleles associated with a limited capacity to synthesize HUFA, which can lead to n-3 HUFA deficiency, is found at high frequency in certain Hispanic populations and is nearly fixed in several indigenous populations from the Americas. Based on these observations, a focused secondary subgroup analysis of the VITAL n-3 HUFA supplementation trial stratifying the data based on self-reported ancestry revealed that African Americans may benefit from n-3 HUFA supplementation, and both ancestry and FADS variability should be factored into future clinical trials design.

Linotrins: Omega-3 oxylipins featuring an E,Z,E conjugated triene motif are present in the plant kingdom and alleviate inflammation in LPS-challenged microglial cells

Alpha-linolenic acid (ALA), an essential omega-3 polyunsaturated fatty acid found in plants, exerts neuroprotection and anti-inflammatory effects in chronic and acute CNS disease models. However, the underlying mechanisms are not yet understood. Since ALA is not incorporated into the brain, the observed health benefits may result from some of its metabolites. The putative formation of dihydroxylated ALA derivatives (called linotrins) was recently shown in vitro in the presence of lipoxygenases. However, the in vitro biosynthesis of linotrins was neither stereoselective nor quantitatively efficient for studying their physiological roles as enantiomeric pure forms. Herein, we report the first stereo-controlled synthesis that features regio- and stereoselective hydrometalations of alkynes for assembling the sensitive E,Z,E-conjugated trienes, as well as LC-MS investigations that provide evidence of linotrins occurrence in plants. Moreover, strong anti-inflammatory effects on microglia highlight the potential physiological importance of linotrins and open new perspectives in search of CNS therapeutics.

Specialized pro-resolving mediators in respiratory diseases.

Persistent unresolved inflammation results in a number of pathologic respiratory diseases including asthma, cystic fibrosis, acute respiratory distress syndrome (ARDS) and coronavirus disease 2019 (COVID-19)-associated ARDS. Inflammation resolution is an active series of biologic processes orchestrated by a family of bioactive specialized pro-resolving mediators (SPMs) derived from essential omega-3 and omega-6 polyunsaturated fatty acids (PUFAs). In this review, we highlight recent findings on dysregulated inflammation resolution in common respiratory diseases and recent literature on SPM generation with PUFA dietary supplementation with relevance to diseases of respiratory inflammation. Human studies and preclinical models of diseases of lung inflammation have revealed disequilibrium in the levels of pro-inflammatory versus pro-resolving mediators. Recent studies identified actions for SPMs on regulating prophlogistic host responses and stimulating inflammation resolution pathways in inflammatory respiratory diseases. Dietary marine oils are enriched in PUFAs and contain parent omega-3 and omega-6 fatty acids and precursors for conversion to SPMs. Nutritional supplementation with fish oils can boost SPM levels and offer a therapeutic approach targeting inflammation resolution pathways for diseases of lung inflammation.

Health Potential of Chia (Salvia hispanica L.) Seeds-Derived α-linoleic Acid and α-linolenic Acids: A Review

The desirability of functional foods proceeds alongside non-communicable epidemic; diabetes, cardiovascular diseases, obesity and interrelated risk factors. Consequently, research on chia seeds (Salvia hispanica L.) has increased not only as seeds but also as oil due to their rich functional and nutritional proprieties. Chia seed is rich in essential fatty acids such as α-linolenic (ALA) and α-linoleic acids (LA) with ALA being the biological precursor to eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). ALA is a plant-based essential omega-3 polyunsaturated fatty acids that must be obtained through the diet since humans do not possess the enzymes to synthesize the compound. The objective of this review was to identify the beneficial effects of chia seeds derived ALA and LA on human health. From the articles reviewed; it was observed that ALA and LA from chia seed could offer benefits for example; anti-obesity, improvement in glucose homeostasis, athletic performance, and anti-cholesterolemic effects and maintenance of healthy serum lipid level. Furthermore, some articles reported anti-inflammatory, anti-proliferative and apoptotic effects and anti-oxidant activity, which could strengthen the prevention of chronic diseases. Although these benefits are appealing to humans, mechanistic cell culture and animal models are required to enhance our knowledge on their mode of action

Flaxseed oil as omega-3 polyunsaturated fatty acid source modulates cortisol concentrations and social dominance in male and female guinea pigs

Flaxseed oil is an excellent source of the essential omega-3 polyunsaturated fatty acid (PUFA) alpha-linolenic acid (ALA). Omega-3 PUFAs are important neuronal components and can counteract aggressive, depressive, and anxiety-like behavior, reduce glucocorticoid (e.g. cortisol) concentrations under chronic stress but also increase acute glucocorticoid responses. As glucocorticoids per se and glucocorticoid responsiveness can modulate the establishment of dominance hierarchies, we investigated if flaxseed oil high in ALA can promote social dominance through effects on glucocorticoid concentrations. Two male and two female groups of domestic guinea pigs (n = 9 per group) were maintained on a control or a 5% (w/w) flaxseed oil diet for four weeks. Social behaviors, hierarchy indices, locomotion, and saliva cortisol concentrations were determined during basal group housing conditions and stressful social confrontations with unfamiliar individuals of the other groups. Flaxseed groups had increased basal cortisol concentrations and showed no cortisol increase during social confrontations. Cortisol concentrations in control groups significantly increased during social confrontations. Such higher cortisol responses positively affected individual hierarchy indices in control males. However, flaxseed males became dominant irrespective of cortisol concentrations. In females, the opposite was detected, namely a higher dominant status in control compared to flaxseed females. Open-field- and dark-light-tests for anxiety-like behavior revealed no pronounced differences, but flaxseed males showed the highest locomotor activity. Flaxseed oil as an ALA source sex-specifically promoted social dominance irrespective of cortisol concentrations and responses. The underlying neuronal mechanisms remain to be determined, but a sex-specific energetic advantage may have accounted for this effect.

Nutritional and Antioxidant Potential of Fiddleheads from European Ferns.

Ferns are part of the diet and traditional medicine in East Asia, North America, and Oceania, however, their importance has been forgotten in Europe. Here, the nutritional and antioxidant potential of young fern fronds (fiddleheads) of eight families were studied. Most of the tested fern species excelled in high antioxidant capacity when compared to the reference leafy vegetables spinach and rocket. On average, the total phenol content reached 220 mg·g−1 of extract dry weight for all fiddleheads, and 15 out of 24 tested species exceeded 1 g Trolox equivalent per gram of extract dry weight in Oxygen Radical Absorbance Capacity (ORAC) assay. On the other hand, fiddleheads contained a comparable amount of carotenoids and ascorbic acid with the reference vegetables. In the case of fatty acid composition, fiddleheads contained especially high amounts of essential omega-3 (n3) and omega-6 (n6) polyunsaturated fatty acids with a beneficial n6/n3 ratio. The n6/n3 ratio in all tested species was between 2 and 6.4, whereas the ratio in the reference vegetables was below 0.4. All in all, fiddleheads from European ferns are a rich source of valuable antioxidants and essential fatty acids with a desirable n-6/n-3 ratio and may thus form an alternative source of these compounds, especially for those people not consuming fish and fish products.

INFLUENCE OF COMPLEX TREATMENT ON BIOCHEMICAL BLOOD PARAMETERS OF PATIENTS WITH NON-ALCOHOLIC FATTY LIVER DISEASE AND CONCOMITANT PRE-DIABETES

The aim: Of this research is to evaluate laboratory changes in the liver blood tests, carbohydrate and lipid metabolism in NAFLD patients with concomitant pre-diabetes, and to study the feasibility of their complex treatment with the inclusion of omega-3 polyunsaturated fatty acids and essential phospholipids. Materials and methods: We have examined 55 patients with non-alcoholic fatty liver disease on the background of pre-diabetes aged 40 to 75 years. Modification of lifestyle was recommended to all patients as a basic treatment. In addition, the patients were prescribed essential phospholipids in 2 capsules 3 times a day and omega-3 polyunsaturated fatty acids 1000 mg per day for 28 patients (group 1) or rosuvastatin 10 mg per day for 27 persons (group 2). The effectiveness of the treatment was evaluated in 3 months, and the long-term outcomes were evaluated in 12 months. Results: Under the influence of the prescribed treatment, a hypolipidemic effect was observed in both groups, but a significant decline in the activity of alanine aminotransferase and aspartate aminotransferase occurred only under the influence of a combination of essential phospholipids and omega-3 polyunsaturated fatty acids. Conclusions: Thus, the described results allow to recommend this combination of medicines to patients with non-alcoholic fatty liver disease and concomitant pre-diabetes.
