A one health approach for monitoring antimicrobial resistance: developing a national freshwater pilot effort

Antimicrobial resistance (AMR) is a world-wide public health threat that is projected to lead to 10 million annual deaths globally by 2050. The AMR public health issue has led to the development of action plans to combat AMR, including improved antimicrobial stewardship, development of new antimicrobials, and advanced monitoring. The National Antimicrobial Resistance Monitoring System (NARMS) led by the United States (U.S) Food and Drug Administration along with the U.S. Centers for Disease Control and U.S. Department of Agriculture has monitored antimicrobial resistant bacteria in retail meats, humans, and food animals since the mid 1990’s. NARMS is currently exploring an integrated One Health monitoring model recognizing that human, animal, plant, and environmental systems are linked to public health. Since 2020, the U.S. Environmental Protection Agency has led an interagency NARMS environmental working group (EWG) to implement a surface water AMR monitoring program (SWAM) at watershed and national scales. The NARMS EWG divided the development of the environmental monitoring effort into five areas: (i) defining objectives and questions, (ii) designing study/sampling design, (iii) selecting AMR indicators, (iv) establishing analytical methods, and (v) developing data management/analytics/metadata plans. For each of these areas, the consensus among the scientific community and literature was reviewed and carefully considered prior to the development of this environmental monitoring program. The data produced from the SWAM effort will help develop robust surface water monitoring programs with the goal of assessing public health risks associated with AMR pathogens in surface water (e.g., recreational water exposures), provide a comprehensive picture of how resistant strains are related spatially and temporally within a watershed, and help assess how anthropogenic drivers and intervention strategies impact the transmission of AMR within human, animal, and environmental systems.

Assessing the Interpretability–Performance Trade-Off of Artificial Neural Networks Using Sentinel Fish Health Data

A number of sentinel species are regularly sampled from the environment near the Oil Sands Region (OSR) in Alberta, Canada. In particular, trout-perch are sampled as a proxy for the health of the aquatic ecosystem. As the development of the OSR began before the environmental monitoring program was in place, there is currently no established measure for the baseline health of the local ecosystem. A common solution is to calculate normal ranges for fish endpoints. Observations found to be outside the normal range are then flagged, alerting researchers to the potential presence of stressors in the local environment. The quality of the normal ranges is dependent on the accuracy of the estimates used to calculate them. This paper explores the use of neural networks and regularized regression for improving the prediction accuracy of fish endpoints. We also consider the trade-off between the prediction accuracy and interpretability of each model. We find that neural networks can provide increased prediction accuracy, but this improvement in accuracy may not be worth the loss in interpretability in some ecological studies. The elastic net offers both good prediction accuracy and interpretability, making it a safe choice for many ecological applications. A hybridized method combining both the neural network and elastic net offers high prediction accuracy as well as some interpretability, and therefore it is the recommended method for this application.

Analysis of Thorium (Th) and Uranium (U) in radioactive wastewater sample in accordance with American Public Health Association (APHA) 3125 method

The main issue with thorium (Th) and uranium (U) in radioactive wastewater is the potential of these elements to contaminate the water bodies. The unregulated radioactive wastewater release can affect the ecosystem and potentially harm aquatic life. Therefore, there is a need to establish the analysis of Th and U using standard method to obtain better sensitivity and good confidence in the final report. In this study, the determination of uranium and thorium was employed using the American Public Health Association (APHA) method 3125 standard method. The method performance and method detection level (MDL) samples for APHA 3125 were prepared in the deionized water and digested using hot block (APHA 3030E) and microwave digester (APHA 3030K). The quantification of 238U, 235U and 232Th in the method performance and MDL samples was performed using Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). From the results, the method performance samples for both digestion methods were in accordance with the APHA requirement. The MDL obtained from APHA 3030E for 232Th, 238U, and 235U were 5, 7, and 8 μg L−1 respectively. Meanwhile, the MDL obtained from the microwave digester for 232Th, 238U, and 235U, and were 2, 3, and 3 μg L−1 respectively. The APHA 3030K digestion technique gives lower and better detection limit compared to the APHA 3030E technique. Based on APHA 3030K MDL, 232Th was detected in the radioactive wastewater sample while 238U and 235U were less than MDL. Meanwhile, for APHA 3030E, all elements were below MDL. This study will enhance the detection of uranium and thorium in the radioactive wastewater sample towards better environmental monitoring programs at the national and global levels.

Hepatic transcriptome, transcriptional effects and antioxidant responses in Poecilia vivipara exposed to sanitary sewage

Aquatic environments are subject to threats from multiple human activities, particularly through the release of untreated sanitary sewage into the coastal environments. These effluents contain a large group of natural or synthetic compounds referred to as emerging contaminants. Monitoring the types and quantities of toxic substances in the environment, especially complex mixtures, is an exhausting and challenging task. Integrative effect-based tools, such as biomarkers, are recommended for environmental quality monitoring programs. In this study, fish Poecilia vivipara were exposed for 24 and 96 h to raw untreated sewage diluted 33 % (v/v) in order to identify hepatic genes to be used as molecular biomarkers. Through a de novo hepatic transcriptome assembly, using Illumina MiSeq, 54,285 sequences were assembled creating a reference transcriptome for this guppy species. Transcripts involved in biotransformation systems, antioxidant defenses, ABC transporters, nuclear and xenobiotic receptors were identified and evaluated by qPCR. Sanitary sewage induced transcriptional changes in AhR, PXR, CYP2K1, CYP3A30, NQO1, UGT1A1, GSTa3, GSTmu, ST1C1, SOD, ABCC1 and SOX9 genes from liver of fish, particularly after 96 h of exposure. Changes in hepatic enzyme activities were also observed. The enzymes showed differences in fish exposed to both periods, while in the gills there was a prevalence of significant results after 96 h. The observed differences were associated to gender and/or to sewage exposure. The obtained results support the use of P. vivipara as sentinel and model organism for ecotoxicological studies and evidence the importance of understanding the differential responses associated to gender.

Use of Unmanned Surface Vehicles (USVs) in Water Chemistry Studies.

Unmanned surface vehicles (USVs) equipped with integrated sensors are a tool valuable to several monitoring strategies, offering enhanced temporal and spatial coverage over specific timeframes, allowing for targeted examination of sites or events of interest. The elaboration of environmental monitoring programs has relied so far on periodic spot sampling at specific locations, followed by laboratory analysis, aiming at the evaluation of water quality at a catchment scale. For this purpose, automatic telemetric stations for specific parameters have been installed by the Institute of Marine Biological Resources and Inland Waters of Hellenic Centre for Marine Research (IMBRIW-HCMR) within several Greek rivers and lakes, providing continuous and temporal monitoring possibilities. In the present work, USVs were deployed by the Athens Water and Sewerage Company (EYDAP) as a cost-effective tool for the environmental monitoring of surface water bodies of interest, with emphasis on the spatial fluctuations of chlorophyll α, electrical conductivity, dissolved oxygen and pH, observed in Koumoundourou Lake and the rivers Acheloos, Asopos and Kifissos. The effectiveness of an innovative heavy metal (HM) system installed in the USV for the in situ measurements of copper and lead was also evaluated herewith. The results obtained demonstrate the advantages of USVs, setting the base for their application in real-time monitoring of chemical parameters including metals. Simultaneously, the requirements for accuracy and sensitivity improvement of HM sensors were noted, in order to permit full exploitation of USVs' capacities.

Recent progresses in analytical method development for 210Pb in environmental and biological samples.

As a decay product of uranium series, 210Pb spreads widely in the nature and imposes strong radiological and chemical toxicity. It is vital to establish reliable and efficient radioanalytical methods for 210Pb determination to support environment and food radioactivity monitoring programs. This article critically reviews analytical methods developed for determining 210Pb in environmental and biological samples, especially new development in recent years. Techniques applied throughout different analytical steps including sample pretreatment, separation, purification, and detection are summarized and their pros and cons are discussed to provide a holistic overview for 210Pb environmental and biological assay.

Radiation-hygienic situation in the area of sublimate production waste storage of JSC “AECC”

In accordance with the fundamentals of state policy in the field of ensuring nuclear and radiation safety, State Corporation «Rosatom» carries out the remediation of JSC “AECC” nuclear legacy sites. The article presents the results of environmental impact assessment carried out in 2021-2023 in the vicinity of sublimation waste storage facilities that are being decommissioned. Health physics studies involved ambient dose rate measurements, sampling (soil, groundwater) and analysis of radiological and non radiological environmental pollutants. The external gamma dose rate, as well as content of natural and artificial radionuclides in soil, don’t exceed background levels. While uranium presence in groundwater is found to be 10 times and higher then background level. The data obtained indicate that the accumulated wastes of sludge sumps (bottom sediments, pulp) сome into contact with natural water. Flooded storage facilities form a plume of contamination spreading out in the direction of groundwater flow. Given the current situation optimization of the existing environmental monitoring programs in the JSC “AECC” health protection zone is recommended and reference levels of uranium proposed.

Antimicrobial resistance in rural rivers: Comparative study of the Coquet (Northumberland) and Eden (Cumbria) River catchments

Many studies have characterised resistomes in river microbial communities. However, few have compared resistomes in parallel rural catchments that have few point-source inputs of antimicrobial genes (ARGs) and organisms (i.e., AMR) – catchments where one can contrast more nebulous drivers of AMR in rural rivers. Here, we used quantitative microbial profiling (QMP) to compare resistomes and microbiomes in two rural river catchments in Northern England, the Coquet and Eden in Northumberland and Cumbria, respectively, with different hydrological and geographical conditions. The Eden has higher flow rates, higher annual surface runoff, and longer periods of soil saturation, whereas the Coquet is drier and has lower flowrates. QMP analysis showed the Eden contained significantly more abundant microbes associated with soil sources, animal faeces, and wastewater than the Coquet, which had microbiomes like less polluted rivers (Wilcoxon test, p < 0.01). The Eden also had greater ARG abundances and resistome diversity (Kruskal Wallis, p < 0.05), and higher levels of potentially clinically relevant ARGs. The Eden catchment had greater and flashier runoff and more extensive agricultural land use in its middle reach, which explains higher levels of AMR in the river. Hydrological and geographic factors drive AMR in rural rivers, which must be considered in environmental monitoring programmes.

Advancing environmental monitoring across the water continuum combining biomarker analysis in multiple sentinel species: A case study in the Seine-Normandie Basin (France)

Nowadays, biomarkers are recognized as valuable tools to complement chemical and ecological assessments in biomonitoring programs. They provide insights into the effects of contaminant exposures on individuals and establish connections between environmental pressure and biological response at higher levels. In the last decade, strong improvements in the design of experimental protocols and the result interpretation facilitated the use of biomarker across wide geographical areas, including aquatic continua. Notably, the statistical establishment of reference values and thresholds enabled the discrimination of contamination effects in environmental conditions, allowed interspecies comparisons, and eliminated the need of a reference site.The aim of this work was to study freshwater-estuarine-coastal water continua by applying biomarker measurements in multi-species caged organisms. During two campaigns, eight sentinel species, encompassing fish, mollusks, and crustaceans, were deployed to cover 25 sites from rivers to the sea. As much as possible, a common methodology was employed for biomarker measurements (DNA damage and phagocytosis efficiency) and data interpretation based on guidelines established using reference values and induction/inhibition thresholds (establishment of three effect levels).The methodology was successfully implemented and allowed us to assess the environmental quality. Employing multiple species per site enhances confidence in observed trends. The results highlight the feasibility of integrating biomarker-based environmental monitoring programs across a continuum scale. Biomarker results align with Water Framework Directive indicators in cases of poor site quality. Additionally, when discrepancies arise between chemical and ecological statuses, biomarker findings offer a comprehensive perspective to elucidate the disparities. Presented as a pilot project, this work contributes to gain insights into current biomonitoring needs, providing new questions and perspectives.

American crocodiles (Crocodylus acutus: Reptilia: Crocodilidae) visiting the facilities of a freshwater aquaculture of the Northern Pacific region, Costa Rica, carry tetracycline-resistant Escherichia coli.

Apex predators are exposed to antimicrobial compounds and resistant microbes, which accumulate at different trophic levels of the related ecosystems. The study aimed to characterize the presence and the antimicrobial resistance patterns of fecal Escherichia coli isolated from cloacal swab samples obtained from wild-living American crocodiles (Crocodylus acutus) (n = 53). Sampling was conducted within the distinctive context of a freshwater-intensive aquaculture farm in Costa Rica, where incoming crocodiles are temporarily held in captivity before release. Phenotypic antimicrobial susceptibility profiles were determined in all isolates, while resistant isolates were subjected to whole-genome sequencing and bioinformatics analyses. In total, 24 samples contained tetracycline-resistant E. coli (45.3%). Isolates carried either tet(A), tet(B), or tet(C) genes. Furthermore, genes conferring resistance to ß-lactams, aminoglycosides, fosfomycin, sulfonamides, phenicol, quinolones, trimethoprim, and colistin were detected in single isolates, with seven of them carrying these genes on plasmids. Genome sequencing further revealed that sequence types, prevalence of antibiotic resistance carriage, and antibiotic resistance profiles differed between the individuals liberated within the next 24 h after their capture in the ponds and those liberated from enclosures after longer abodes. The overall presence of tetracycline-resistant E. coli, coupled with potential interactions with various anthropogenic factors before arriving at the facilities, hinders clear conclusions on the sources of antimicrobial resistance for the studied individuals. These aspects hold significant implications for both the aquaculture farm's biosecurity and the planning of environmental monitoring programs using such specimens. Considering human-crocodile conflicts from the One Health perspective, the occurrence of antimicrobial resistance underscores the importance of systematical surveillance of antibiotic resistance development in American crocodiles.

Innovative analytical methodologies for characterizing chemical exposure with a view to next-generation risk assessment

The chemical burden on the environment and human population is increasing. Consequently, regulatory risk assessment must keep pace to manage, reduce, and prevent adverse impacts on human and environmental health associated with hazardous chemicals. Surveillance of chemicals of known, emerging, or potential future concern, entering the environment-food-human continuum is needed to document the reality of risks posed by chemicals on ecosystem and human health from a one health perspective, feed into early warning systems and support public policies for exposure mitigation provisions and safe and sustainable by design strategies. The use of less-conventional sampling strategies and integration of full-scan, high-resolution mass spectrometry and effect-directed analysis in environmental and human monitoring programmes have the potential to enhance the screening and identification of a wider range of chemicals of known, emerging or potential future concern. Here, we outline the key needs and recommendations identified within the European Partnership for Assessment of Risks from Chemicals (PARC) project for leveraging these innovative methodologies to support the development of next-generation chemical risk assessment.

Agriculturally Improved and Semi-Natural Permanent Grasslands Provide Complementary Ecosystem Services in Swedish Boreal Landscapes

Permanent grasslands cover more than a third of European agricultural land and are important for a number of ecosystem services. Permanent grasslands used for agriculture are broadly separated into agriculturally improved and semi-natural grasslands. High cultural and natural values linked to semi-natural grasslands are well documented. However, in boreal and hemi-boreal agricultural landscapes, less information is available about the areal coverage of improved permanent grasslands and their role for ecosystem service provision and biodiversity. In Sweden, grasslands are administratively separated into semi-natural (i.e., land that cannot be ploughed) or arable (i.e., improved temporary or permanent grassland on land that can be ploughed). We used data from a large-scale environmental monitoring program to show that improved permanent grassland (i.e., permanent grasslands on arable fields) may be a previously unrecognised large area of the agricultural land use in Sweden. We show that improved permanent grasslands together with semi-natural grasslands are both comparable but also complementary providers of a range of ecosystem services (plant species richness, plant resources for pollinators and forage amount for livestock production). However, as expected, semi-natural grasslands with the highest-level AESs (special values) show high species richness values for vascular plants, plants indicating traditional semi-natural management conditions and red-listed species. Improved permanent grasslands on arable fields are likely an underestimated but integral part of the agricultural economy and ecological function in boreal landscapes that together with high nature value semi-natural grasslands provide a broad range of ecosystem services.

Intensive Environmental Sampling and Whole Genome Sequence-based Characterization of Listeria in Small- and Medium-sized Dairy Facilities Reveal Opportunities for Simplified and Size-appropriate Environmental Monitoring Strategies

Small- and medium-sized dairy processing facilities (SMDFs) may face unique challenges with respect to controlling Listeria in their processing environments, e.g., due to limited resources. The aim of this study was to implement and evaluate environmental monitoring programs (EMPs) for Listeria control in eight SMDFs in a ∼1-year longitudinal study; this included a comparison of pre-operation (i.e., after cleaning and sanitation and prior to production) and mid-operation (i.e., at least 4 h into production) sampling strategies. Among 2,072 environmental sponge samples collected across all facilities, 272 (13%) were positive for Listeria. Listeria prevalence among pre- and mid-operation samples (15% and 17%, respectively), was not significantly different. Whole genome sequencing (WGS) performed on select isolates to characterize Listeria persistence patterns revealed repeated isolation of closely related Listeria isolates (i.e., ≤20 high-quality single nucleotide polymorphism [hqSNP] differences) in 5/8 facilities over >6 months, suggesting Listeria persistence and/or reintroduction was relatively common among the SMDFs evaluated here. WGS furthermore showed that for 41 sites where samples collected pre- and mid-operation were positive for Listeria, Listeria isolates obtained were highly related (i.e., ≤10 hqSNP differences), suggesting that pre-operation sampling alone may be sufficient and more effective for detecting sites of Listeria persistence. Importantly, our data also showed that only 1/8 of facilities showed a significant decrease in Listeria prevalence over 1 year, indicating continued challenges with Listeria control in at least some SMDFs. We conclude that options for simplified Listeria EMPs (e.g., with a focus on pre-operation sampling, which allows for more rapid identification of likely persistence sites) may be valuable for improved Listeria control in SMDFs.

Using iterative 2δ‐technique and 4δ‐outlier test for evaluating the geochemical background concentrations of trace elements in topsoil at Cao Bang province, Vietnam

AbstractThis study investigated the geochemical background values for seven trace elements in topsoil samples collected from highland areas within Cao Bang province, Vietnam. Total concentrations of these elements were determined, revealing the following order of mean concentration: Zn &gt; Cr &gt; Pb &gt; Cu &gt; As &gt; Ni &gt; Co. The background values were calculated using two different methods including iterative 2δ‐technique and 4δ‐outlier test. The 4δ‐outlier test yielded the following background concentrations (dry weight) for the seven elements: As (24.41 ± 12.42 mg kg−1), Co (15.64 ± 10.57 mg kg−1), Cr (49.31 ± 33.41 mg kg−1), Cu (29.77 ± 27.14 mg kg−1), Ni (22.52 ± 13.33 mg kg−1), Pb (38.06 ± 31.48 mg kg−1), Zn (71.79 61.09 mg kg−1). The 2δ‐technique, on the other hand, resulted in the following background concentrations (dry weight) for the same elements: As (8.41 ± 8.57 mg kg−1), Co (5.06 ± 4.31 mg kg−1), Cr (26.72 ± 26.20 mg kg−1), Cu (25.31 ± 24.81 mg kg−1), Ni (8.02 ± 7.83 mg kg−1), Pb (28.12 ± 27.58 mg kg−1), Zn (51.91 ± 49.44 mg kg−1). These results provide valuable baseline data for future environmental assessments and monitoring programs in the region. They also highlight the potential impact of human activities, such as mining, on the geochemical landscape of highland areas.

Improved sampling and DNA extraction procedures for microbiome analysis in food-processing environments.

Deep investigation of the microbiome of food-production and food-processing environments through whole-metagenome sequencing (WMS) can provide detailed information on the taxonomic composition and functional potential of the microbial communities that inhabit them, with huge potential benefits for environmental monitoring programs. However, certain technical challenges jeopardize the application of WMS technologies with this aim, with the most relevant one being the recovery of a sufficient amount of DNA from the frequently low-biomass samples collected from the equipment, tools and surfaces of food-processing plants. Here, we present the first complete workflow, with optimized DNA-purification methodology, to obtain high-quality WMS sequencing results from samples taken from food-production and food-processing environments and reconstruct metagenome assembled genomes (MAGs). The protocol can yield DNA loads >10 ng in >98% of samples and >500 ng in 57.1% of samples and allows the collection of, on average, 12.2 MAGs per sample (with up to 62 MAGs in a single sample) in ~1 week, including both laboratory and computational work. This markedly improves on results previously obtained in studies performing WMS of processing environments and using other protocols not specifically developed to sequence these types of sample, in which <2 MAGs per sample were obtained. The full protocol has been developed and applied in the framework of the European Union project MASTER (Microbiome applications for sustainable food systems through technologies and enterprise) in 114 food-processing facilities from different production sectors.

Survey of Antifungal in Surface- and Groundwater: A Portuguese Environmental Case Study

Antifungals support modern medical and agricultural practices, and extensive screening environmental data are missing. This work aims to survey antifungals for medical and agricultural purposes in surface- and groundwater in Portugal. A passive sampling technique and a high-resolution chromatographic system were used. Antifungals applied in medical practice were not detected as only antifungals for agricultural purposes (fungicides). Thirty-nine different fungicide molecules were found; its detection frequency in surface- and groundwater was dominated by rabenzazole (61%, 92%) and ethoxyquin (54%, 77%); among the five fungicides with similar surface- and groundwater catching rates, ferimzone was the most polluting (54%, 54%); oxadixyl (61%), kresoxim-methyl (61%) and fenamidone (46%) were primarily designated surface water contaminants; for azoles, the occurrence in surface water ranged from a residual (10%) to a moderate detection rate for two compounds (31%—propiconazole and tebuconazole). Surprisingly, only 51% of detected fungicides are authorised. The frequently detected fungicides that are either not authorised (ethoxyquin and its impurity, furmecyclox, oxadixyl), without data in the EU (rabenzazole, ferimzone), or authorised at the national level (fenamidone), should be included in environmental monitoring programmes and followed as emerging (micro)contaminants. Policy makers should gather their efforts to allow the implementation of proper risk management and effective contamination control strategies to achieve Sustainable Development Goals.

An overview of automated flow systems for total and isotopic analysis of strontium and yttrium in samples of environmental interest

Due to the different uses of radioactivity during the last decades, there has been an increase in the concentration of natural and artificial radionuclides in the environment. This, along with some accidents with a high affect public opinion (for example, Chernobyl and Fukushima), have led to the growth and establishment of environmental radioactivity monitoring programs. Currently, trends in legislation and research are focused on the development of accurate, precise, reliable and fast analytical methods with low limits of detection (LOD) for radionuclides determination, such as strontium and yttrium, in environmental samples. In this paper, two comprehensive reviews and four automated analytical systems for total and isotopic determination of yttrium and strontium are presented. The developed methods have been applied in the analysis of environmental samples with low concentrations of these analytes. These methodologies have been automated by exploiting flow analysis techniques, such as multi-syringe flow injection analysis (MSFIA), Sequential injection analysis (SIA) and laboratory-on-valve (LOV) systems, achieving a minimal handling and low consumption of samples and reagents, a significant reduction in waste generation and a high frequency of analysis. In the developed methodologies, some spectrometric methods such as ICP-OES and ICP-MS have been implemented as detection techniques instead of radiometric detectors obtaining a fully automated, low-cost and fast yttrium and strontium determinations.

Analysis of Alternative Methods of Environmental Monitoring for Listeria in Food Production Facilities

Validated alternative test methodologies may be used in place of culture-based methods recommended for environmental monitoring programs (EMPs) for Listeria in food production facilities. In order to help guide decisions on which testing method to use to simplify Listeria EMP implementation in food production facilities, alternative methods were compared to the culture-based method in actual EMPs for Listeria. Seventy-two samples collected from two facilities of souzai production businesses that use meat and meat products as ingredients, one facility of processed meat product production business, and one facility of processed meat product and souzai production business were applied to EMPs for Listeria using the culture-based method, 3MTM Molecular Detection System (MDS), and InSite L. mono Glo (InSite). The kappa coefficient in MDS was 0.65 for Listeria monocytogenes and 0.74 for Listeria spp., both of which were deemed substantial compared with the culture-based method. The kappa coefficient in InSite was −0.01 for L. monocytogenes and 0.50 for Listeria spp., which indicated poor and moderate reproducibility, respectively. When the medium of InSite was smeared on agar medium, 7 of the 19 samples tested positive only for Listeria spp. (negative for L. monocytogenes) but L. monocytogenes was cultured, indicating that the sensitivity of detecting L. monocytogenes via fluorescence may be low. MDS was considered a useful alternative for both L. monocytogenes and Listeria spp. as targets, and InSite was not possible as a substitute for detecting L. monocytogenes; however, it is considered a helpful alternative method for detecting Listeria spp. EMPs for Listeria often target Listeria spp. as an indicator of L. monocytogenes. The alternative methods studied in this study are rapid, simple, and useful in EMPs for Listeria. However, the data in this study were a comparatively small sample set and impacted by variability, so more robust comparisons are desirable in the future.

Novel yeast-based biosensor for environmental monitoring of tebuconazole

Due to increasing demand for high and stable crop production, human populations are highly dependent on pesticide use for growing and storing food. Environmental monitoring of these agrochemicals is therefore of utmost importance, because of their collateral effects on ecosystem and human health. Even though most current-use analytical methods achieve low detection limits, they require procedures that are too complex and costly for routine monitoring. As such, there has been an increased interest in biosensors as alternative or complementary tools to streamline detection and quantification of environmental contaminants. In this work, we developed a biosensor for environmental monitoring of tebuconazole (TEB), a common agrochemical fungicide. For that purpose, we engineered S. cerevisiae cells with a reporter gene downstream of specific promoters that are expressed after exposure to TEB and characterized the sensitivity and specificity of this model system. After optimization, we found that this easy-to-use biosensor consistently detects TEB at concentrations above 5 μg L−1 and does not respond to realistic environmental concentrations of other tested azoles, suggesting it is specific. We propose the use of this system as a complementary tool in environmental monitoring programs, namely, in high throughput scenarios requiring screening of numerous samples.Key points• A yeast-based biosensor was developed for environmental monitoring of tebuconazole.•The biosensor offers a rapid and easy method for tebuconazole detection ≥ 5 μg L−1.•The biosensor is specific to tebuconazole at environmentally relevant concentrations.

Secchi Disk Depth or Turbidity, Which Is Better for Assessing Environmental Quality in Eutrophic Waters? A Case Study in a Shallow Hypereutrophic Reservoir

Water transparency is widely used in environmental monitoring programs and in assessing water quality in aquatic environments. The purpose of this study was to determine which of two water transparency-measuring tools, a Secchi disk or a water turbidity meter, is better to assess environments in shallow eutrophic waters. Measurements of the Secchi disk depth (Dsd) and water turbidity (Turb) were carried out simultaneously at eight stations of the hypereutrophic Sestroretsky Razliv reservoir in 2015–2018. In May, October, and December, Dsd varied around 0.6 m but was significantly lower in August during algal blooms. Turbidity ranged from 10 to 20 nephelometric turbidity units (NTU) in different seasons but increased to almost 70 NTU in August. Principal component analysis revealed that Dsd was inversely proportional to Turb, total suspended solids, and chlorophyll concentrations. The data showed that at turbidities below 20 NTU, the Secchi disk clearly distinguishes differences in water transparency, but when Turb exceeds 40 NTU, measuring transparency using the Secchi disk no longer allows for water differentiation. In this case, it is preferable to use water turbidity measurements, which remain an effective indicator even in highly turbid waters. This should be taken into account when assessing the environment in eutrophic waters.
