Carotenoid biosynthesis genes LcLCYB, LcLCYE, and LcBCH from wolfberry confer increased carotenoid content and improved salt tolerance in tobacco

Carotenoids play essential roles in plant growth and development and provide plants with a tolerance to a series of abiotic stresses. In this study, the function and biological significance of lycopene β-cyclase, lycopene ε-cyclase, and β-carotene hydroxylase, which are responsible for the modification of the tetraterpene skeleton procedure, were isolated from Lycium chinense and analyzed. The overexpression of lycopene β-cyclase, lycopene ε-cyclase, and β-carotene hydroxylase promoted the accumulation of total carotenoids and photosynthesis enhancement, reactive oxygen species scavenging activity, and proline content of tobacco seedlings after exposure to the salt stress. Furthermore, the expression of the carotenoid biosynthesis genes and stress-related genes (ascorbate peroxidase, catalase, peroxidase, superoxide dismutase, and pyrroline-5-carboxylate reductase) were detected and showed increased gene expression level, which were strongly associated with the carotenoid content and reactive oxygen species scavenging activity. After exposure to salt stress, the endogenous abscisic acid content was significantly increased and much higher than those in control plants. This research contributes to the development of new breeding aimed at obtaining stronger salt tolerance plants with increased total carotenoids and vitamin A content.

S-ABA Enhances Rice Salt Tolerance by Regulating Na+/K+ Balance and Hormone Homeostasis.

In order to explore the regulating role and the physiological and biochemical mechanisms of trans-abscisic acid (hereinafter referred as S-ABA) in the process of rice growth and development under salt stress, we took Chaoyou 1000 and Yuxiangyouzhan as materials and set up three salt concentration treatments, CK0 (Control treatment), N1 (50 mmol L-1 NaCl), and N2 (100 mmol L-1 NaCl), in potted trials; we aimed to study the mechanism of rice's response to salt stress from the perspective of agricultural traits and physiological biochemicals and to improve rice's resistance to salt stress through exogenously applying the regulating technology of S-ABA. The following results were obtained: Under salt stress, the growth of rice was significantly suppressed compared to CK0, exhibiting notable increases in agricultural indicators, photosynthesis efficiency, and the NA+ content of leaves. However, we noted a significant decrease in the K+ content in the leaves, alongside a prominent increase in NA+/K+ and a big increase in MDA (malondialdehyde), H2O2 (hydrogen peroxide), and O2- (superoxide anion). This caused the cytomembrane permeability to deteriorate. By applying S-ABA under salt stress (in comparison with salt treatment), we promoted improvements in agronomic traits, enhanced photosynthesis, reduced the accumulation of NA+ in leaves, increased the K+ content and the activity of antioxidant enzymes, and reduced the active oxygen content, resulting in a sharp decrease in the impact of salt stress on rice's development. The application of S-ABA decreased the endogenous ABA (abscisic acid) content under salt stress treatment but increased the endogenous GA (gibberellin) and IAA (indole acetic acid) contents and maintained the hormonal homeostasis in rice plants. To summarize, salt stress causes damage to rice growth, and the exogenous application of S-ABA can activate the pouring system mechanism of rice, suppress the outbreak of active oxygen, and regulate NA+/K+ balance and hormone homeostasis in the blades, thus relieving the salt stress.

Effects of exogenous glycine betaine on growth and development of tomato seedlings under cold stress.

Low temperature is a type of abiotic stress affecting the tomato (Solanum lycopersicum) growth. Understanding the mechanisms and utilization of exogenous substances underlying plant tolerance to cold stress would lay the foundation for improving temperature resilience in this important crop. Our study is aiming to investigate the effect of exogenous glycine betaine (GB) on tomato seedlings to increase tolerance to low temperatures. By treating tomato seedlings with exogenous GB under low temperature stress, we found that 30 mmol/L exogenous GB can significantly improve the cold tolerance of tomato seedlings. Exogenous GB can influence the enzyme activity of antioxidant defense system and ROS levels in tomato leaves. The seedlings with GB treatment presented higher Fv/Fm value and photochemical activity under cold stress compared with the control. Moreover, analysis of high-throughput plant phenotyping of tomato seedlings also supported that exogenous GB can protect the photosynthetic system of tomato seedlings under cold stress. In addition, we proved that exogenous GB significantly increased the content of endogenous abscisic acid (ABA) and decreased endogenous gibberellin (GA) levels, which protected tomatoes from low temperatures. Meanwhile, transcriptional analysis showed that GB regulated the expression of genes involved in antioxidant capacity, calcium signaling, photosynthesis activity, energy metabolism-related and low temperature pathway-related genes in tomato plants. In conclusion, our findings indicated that exogenous GB, as a cryoprotectant, can enhance plant tolerance to low temperature by improving the antioxidant system, photosynthetic system, hormone signaling, and cold response pathway and so on.

Progress in Fruit Cracking Control of Gibberellic Acid and Abscisic Acid

Fruit cracking or splitting is a severe physiological disease that significantly affects appearance and quality, compromising the commodity value of fruit and causing substantial economic losses to the producers of several fleshy fruit crops. The growth-promoting plant hormone gibberellins (GAs) and growth-inhibiting abscisic acid (ABA) antagonistically regulate numerous processes throughout the plant life cycle. The homeostasis of GA and ABA plays a significant role in the normal growth and development of fruits, and the imbalance of them may lead to the occurrence of cracking or splitting during the process of fruit growth, development, ripening and postharvest storage. The pathways of GA and ABA metabolism and signaling have been studied widely, and the major components are well characterized, including the genes encoding major biosynthesis and catabolism enzymes and the key signaling components. Nevertheless, our knowledge of the mechanisms of GA and ABA governing fruit cracking is not comprehensive enough. In this review, we summarize the advances in understanding the effects of endogenous GAs and ABA contents in fruits and exogenous GAs and ABA treatments on fruit cracking, and we endeavor to provide some genetic cues on the function of GAs and ABA responsible for fruit cracking modulation. The progress in understanding the molecular bases underlying the actions of GAs and ABA in fruit cracking coordination control will facilitate breeding strategies of cracking-resistant ideotypes of fruits, and also carry great theoretical significance in guiding the establishment of integrated prevention and control measures in fruit cracking.

Elevated CO2-conferred sodium homeostasis is linked to abscisic acid-mediated stomatal closure and signalling pathway in tomato plants

Soil salinity hampers plant performance. Elevated atmospheric CO2 (e[CO2]) could alleviate the detrimental effect of salinity on plants but whether abscisic acid (ABA) is involved in this process is unclear. To address this issue, three tomato (Solanum lycopersicum) genotypes with varying endogenous ABA concentrations (wild-type AC, ABA-deficient mutant flacca and ABA-overproduction line SP5) were grown in pots under ambient (400 μmol · mol–1) or elevated (800 μmol · mol–1) CO2 with or without the addition of 100 mmol · L−1sodium chloride (NaCl). The results showed that e[CO2] favored ion homeostasis by decreasing root-to-shoot delivery of Na+, which was mainly attributed to lowered transpiration rate rather than altered xylem-sap Na+ concentration. In AC and SP5, the low transpiration rate of e[CO2]-plants under salinity was accompanied by enhanced endogenous ABA levels, which might play a role in upregulating the abundance of specific transcripts related to Na+ homeostasis (i.e., SALT OVERLY SENSITIVE) under salt stress. In flacca, e[CO2]-induced Na+ homeostasis was abolished, which could be ascribed to the low and unaltered ABA levels, albeit the ethylene biosynthesis was enhanced in flacca under salt stress, indicating an antagonistic relationship between ABA and ethylene. Furthermore, e[CO2] inhibited ethylene biosynthesis under salt stress in all three genotypes. The results enrich our comprehension of the fundamental processes of e[CO2]-conferred salt tolerance in tomato.

Physiological, molecular, and genetic mechanism of action of the biostimulant Quantis™ for increased thermotolerance of potato (Solanum tuberosum L.)

BackgroundRaising global temperatures limit crop productivity and new strategies are needed to improve the resilience of thermosensitive crops such as potato (Solanum tuberosum L.). Biostimulants are emerging as potential crop protection products against environmental stress, however their mechanism of action remains largely unknown, hindering their wider adoption. We used comprehensive physiological, molecular, and mass spectrometry approaches to develop understanding of the mechanism of plant thermotolerance exerted by the biostimulant, Quantis™, under heat stress. Using orthologues gene mutations in Arabidopsis thaliana we report heat-defence genes, modified by Quantis™, which were also investigated for potential overlapping functions in biotic stress defence to Sclerotinia sclerotiorum and Rhizoctonia solani.ResultsQuantis™ enhanced PSII photochemical efficiency and decreased thermal dissipation of potato grown under heat stress. These effects were associated with upregulation of genes with antioxidant function, including PR10, flavonoid 3′‐hydroxylase and β-glucosidases, and modulation of abscisic acid (ABA) and cytokinin (CK) activity in leaves by Quantis™. The biostimulant modulated the expression of the heat-defence genes, PEN1, PR4 or MEE59, with functions in leaf photoprotection and root thermal protection, but with no overlapping function in biotic stress defence. Protective root growth under heat stress, following the biostimulant application, was correlated with enhanced CK signalling in roots. Increased endogenous concentrations of ABA and CK in potato leaves and significant upregulation of StFKF1 were consistent with tuberisation promoting effects. Quantis™ application resulted in 4% tuber weight increase and 40% larger tuber size thus mitigating negative effects of heat stress on tuber growth.ConclusionsQuantis™ application prior to heat stress effectively primed heat tolerance responses and alleviated temperature stress of S. tuberosum L. and A. thaliana by modulating the expression and function of PR4 and MEE59 and by regulating CK activity above and below ground, indicating that the mechanism of action of the biostimulant is conserved, and will be effective in many plant species. Thus, a biostimulant application targeting the most susceptible crop developmental stages to heat disorders can be effectively integrated within future agronomy practices to mitigate losses in other thermosensitive crops.Graphical

E3 ubiquitin ligase TaSDIR1-4A activates membrane-bound transcription factor TaWRKY29 to positively regulate drought resistance.

Drought is a deleterious abiotic stress factor that constrains crop growth and development. Post-translational modification of proteins mediated by the ubiquitin-proteasome system is an effective strategy for directing plant responses to stress, but the regulatory mechanisms in wheat remain unclear. In this study, we showed that TaSDIR1-4A is a positive modulator of the drought response. Overexpression of TaSDIR1-4A increased the hypersensitivity of stomata, root length and endogenous abscisic acid (ABA) content under drought conditions. TaSDIR1-4A encodes a C3H2C3-type RING finger protein with E3 ligase activity. Amino acid mutation in its conserved domain led to loss of activity and altered the subcellular localization. The membrane-bound transcription factor TaWRKY29 was identified by yeast two-hybrid screening, and it was confirmed as interacting with TaSDIR1-4A both in vivo and in vitro. TaSDIR1-4A mediated the polyubiquitination and proteolysis of the C-terminal amino acid of TaWRKY29, and its translocation from the plasma membrane to the nucleus. Activated TaWRKY29 bound to the TaABI5 promoter to stimulate its expression, thereby positively regulating the ABA signalling pathway and drought response. Our findings demonstrate the positive role of TaSDIR1-4A in drought tolerance and provide new insights into the involvement of UPS in the wheat stress response.

A 14-3-3 Protein-Encoding Gene, BdGF14g, Confers Better Drought Tolerance by Regulating ABA Biosynthesis and Signaling

Abscisic acid (ABA), a phytohormone, enacts a cardinal function in coping with abiotic stress. 14-3-3 proteins can interact with ABA-responsive-element-binding transcription factors (ABFs), a chief constituent of ABA signaling, and play critical roles in the dehydration response involving ABA signaling. Meanwhile, whether and how 14-3-3 proteins regulate ABA signaling to respond to aridity stress is yet to be fully investigated. Herein, BdGF14g, a 14-3-3 gene induced by ABA, H2O2, and PEG treatments, was identified in Brachypodium distachyon (B. distachyon). Overexpression of BdGF14g improved drought stress tolerance in tobacco plants, with a higher survival rate, longer root length, enhanced cell membrane stability, and increased antioxidase activity compared with non-transgenic controls in coping with dehydration. Both drought and exogenous ABA treatments resulted in smaller stomatal apertures in BdGF14g-transgenic lines. Additionally, when an ABA biosynthesis inhibitor was added, the better growth statuses, less H2O2 accumulation, and higher activities of catalase and superoxide dismutase under mannitol stress disappeared. Moreover, BdGF14g interacted with NtABF2, upregulated the endogenous ABA content, and enhanced the transcription of ABA-related genes, including NtNCED1, a crucial ABA biosynthesis gene, under drought conditions. In conclusion, BdGF14g acts as a positive factor in the water deficiency response by affecting ABA biosynthesis and signaling in tobacco plants.

OsNCED3 and OsPYL1 promote the closure of rice florets by regulating sugar transporters through endogenous abscisic acid

As the world's largest food crop, rice often encounters flowering asynchronization problems during hybrid rice seed production. In addition, the slow closure of female florets leads to seed mildew and affects the quality. Plant hormone abscisic acid (ABA) plays a crucial role in plant responses to abiotic stresses. Previous studies showed that exogenous ABA promotes floret closure. However, the molecular mechanisms and effects of endogenous ABA on floret closure remain unknown. In this study, the effect of endogenous ABA on floret closure and the molecular mechanism of ABA promoting floret closure through sugar transporters were investigated by changing the expression level of OsNCED3 and OsPYL1 in rice. The results show that OE-OsNCED3 increases the endogenous ABA level of florets. Florets close 5.91 min earlier and CR-OsNCED3 delays the closure of florets by 5.08 min compared with the wild type. In addition, OsPYL1 regulates the endogenous ABA content and changes the sensitivity to ABA such that the floret closure time for OE and CR is 9.84 min earlier and 12.78 min later, respectively, resulting in an increase in the split husk rate to 15.4%. The gene expression levels of some sugar transporters (STs) changed. OsPYL1 and OsSWEET4 proteins could interact on the cell membrane. These results indicate that ABA promotes the closure of rice florets and an enhanced sensitivity to ABA promotes this effect more. The molecular mechanism is mainly related to downstream sugar transporters that respond to the ABA signalling pathway, especially OsSWEET4.

Exogenous preculture with sucrose and abscisic acid improves post-cryopreservation survival of eastern bracken fern gametophytes

Cryopreservation is an important technique used in the conservation of various plant tissues. This study proposes a cryopreservation method for the long-term conservation of eastern bracken fern gametophytes (Pteridium aquilinum var. latiusculum). Encapsulation–dehydration of the gametophytes was performed, and the exogenous sucrose and abscisic acid (ABA) preculture conditions were investigated. Gametophytes are sensitive to dehydration and drying, and the following treatment conditions were applied: encapsulation by alginate containing 0.75 M sucrose, 18-h loading treatment with 0.75 M sucrose, and 6-h drying treatment. The survival rate following cryopreservation was determined. The water content of < 27.5% in the alginate beads after dehydration and drying was found to be appropriate for ensuring survival. Additionally, performing an exogenous sucrose and ABA preculture was essential before encapsulation to achieve a survival of ≥ 90%. The high stress induced by cryopreservation and exogenous preculture regulated the expression of PaSuSy, PaLEA14, and PaABI1b and the endogenous ABA content. In eastern bracken gametophytes, ABI1 appears to be a negative regulator of ABA signaling. These results indicate that the encapsulation–dehydration method is effective for the long-term conservation of eastern bracken fern gametophytes, and exogenous preculture alleviates abiotic stress and increases the survival rate.

ScAREB4 promotes potato constitutive and acclimated freezing tolerance associated with enhancing trehalose synthesis and oxidative stress tolerance.

Cold is a major environmental factor that restrains potato production. Abscisic acid (ABA)can enhance freezing tolerance in many plant species, but powerful evidence of the ABA-mediatedsignalling pathway related to freezing tolerance is still in deficiency. In the present study, cold acclimation capacity of the potato genotypes was enhanced alongside with improved endogenous content of ABA. Further exogenous application of ABA and its inhibitor (NDGA)could enhance and reduce potato freezing tolerance, respectively. Moreover, expression pattern of downstream genes in ABA signalling pathway was analysed and only ScAREB4 was identified with specifically upregulate in S. commersonii (CMM5)after cold and ABA treatments. Transgenic assay with overexpression of ScAREB4 showed that ScAREB4 promoted freezing tolerance. Global transcriptome profiling indicated that overexpression of ScAREB4 induced expression of TPS9 (trehalose-6-phosphate synthase) and GSTU8 (glutathione transferase), in accordance with improved TPS activity, trehalose content, higher GST activity and accumulated dramatically less H2 O2 in the ScAREB4 overexpressed transgenic lines. Taken together, the current results indicate that increased endogenous content of ABA is related to freezing tolerance in potato. Moreover, ScAREB4 functions as a downstream transcription factor of ABA signalling to promote cold tolerance, which is associated with increased trehalose content and antioxidant capacity.

Antagonistic or Compensatory: Crosstalk between ABA and Ethylene in Regulating Stomatal Behavior under High CO2 and Progressive Soil Drying.

Increasing atmospheric CO2 concentrations accompanied by intensified drought stress have a significant impact on plant growth and physiology. The aim of this study was to explore the role of abscisic acid (ABA) in mediating the response of stomata to elevated CO2 (e[CO2]) and soil water deficits. Tomato plants with different endogenous ABA concentrations (Ailsa Craig (AC), its ABA-deficient mutant (flacca) and ABA-overproduction transgenic tomato (SP5)) were grown in ambient (a[CO2], 400 μmol mol -1) and elevated (800 μmol mol -1) CO2 environments and were subjected to progressive soil drying at six-leaf stage. Results showed that, compared to plants grown under a[CO2], e[CO2]-plants had significantly lower stomatal conductance in AC and SP5 but not in flacca. Under drought, e[CO2]-plants had better water status and higher water use efficiency at both leaf and whole plant levels. e[CO2] had no effects on the ABA concentration of plants under well-watered conditions but promoted the accumulation of ABA in leaves of plants subjected to drought stress, which coincided with the upregulated expression of ABA biosynthetic genes and downregulated expression of ABA metabolic genes. Although in flacca, the increase of ABA induced by drought was much less than in AC and SP5, it accumulated large amounts of ethylene, suggesting that in plants with ABA deficiency, ethylene may play a compensatory role in inducing stomatal closure during soil drying. Collectively, these findings improve our understanding of plant performances to a future drier and CO2-enriched environment.

A holistic and sustainable approach linked to drought tolerance of Mediterranean crops.

The rapid increase in average temperatures and the progressive reduction in rainfalls caused by climate change is reducing crop yields worldwide, particularly in regions with hot and semi-arid climates such as the Mediterranean area. In natural conditions, plants respond to environmental drought stress with diverse morphological, physiological, and biochemical adaptations in an attempt to escape, avoid, or tolerate drought stress. Among these adaptations to stress, the accumulation of abscisic acid (ABA) is of pivotal importance. Many biotechnological approaches to improve stress tolerance by increasing the exogenous or endogenous content of ABA have proved to be effective. In most cases the resultant drought tolerance is associated with low productivity incompatible with the requirements of modern agriculture. The on-going climate crisis has provoked the search for strategies to increase crop yield under warmer conditions. Several biotechnological strategies, such as the genetic improvement of crops or the generation of transgenic plants for genes involved in drought tolerance, have been attempted with unsatisfactory results suggesting the need for new approaches. Among these, the genetic modification of transcription factors or regulators of signaling cascades provide a promising alternative. To reconcile drought tolerance with crop yield, we propose mutagenesis of genes controlling key signaling components downstream of ABA accumulation in local landraces to modulate responses. We also discuss the advantages of tackling this challenge with a holistic approach involving different knowledge and perspectives, and the problem of distributing the selected lines at subsidized prices to guarantee their use by small family farms.

Regulation of reactive oxygen species and phytohormones in osmotic stress tolerance during seed germination in indica rice.

Climate change due to global warming is now affecting agricultural production worldwide. In rice, one of the most important crops, water limitation due to irregular rainfall in rainfed lowlands during crop growth limits yield. Dry direct-sowing has been proposed as a water-efficient approach to cope with water stress during rice growth, but poor seedling establishment due to drought during germination and emergence is a problem. Here, we germinated indica rice cultivars Rc348 (drought tolerant) and Rc10 (drought sensitive) under osmotic stress induced by PEG to elucidate mechanisms of germination under drought. Rc348 had higher germination rate and germination index under severe osmotic stress of -1.5 MPa, above those of Rc10. Rc348 showed up-regulated GA biosynthesis, down-regulated ABA catabolism, and up-regulated α-amylase gene expression in imbibed seeds under PEG treatment compared to that of Rc10. During germination, reactive oxygen species (ROS) play important roles in antagonism between gibberellic acid (GA) and abscisic acid (ABA). Embryo of Rc348 treated with PEG had significantly greater expression of NADPH oxidase genes and higher endogenous ROS levels, together with significantly increased endogenous GA1, GA4 and ABA contents compared to that of Rc10. In aleurone layers treated with exogenous GA, expression of α-amylase genes was higher in Rc348 than in Rc10, and expression of NADPH oxidase genes was enhanced with significantly higher ROS content in Rc348, suggesting higher sensitivity of GA to ROS production and starch degradation in aleurone cells of Rc348. These results suggest that the osmotic stress tolerance of Rc348 is due to enhancement of ROS production, GA biosynthesis, and GA sensitivity, resulting in a higher germination rate under osmotic stress.

Thioredoxin TRXo1 is involved in ABA perception via PYR1 redox regulation

Abscisic acid (ABA) plays a fundamental role in plant growth and development processes such as seed germination, stomatal response or adaptation to stress, amongst others. Increases in the endogenous ABA content is recognized by specific receptors of the PYR/PYL/RCAR family that are coupled to a phosphorylation cascade targeting transcription factors and ion channels. Just like other receptors of the family, nuclear receptor PYR1 binds ABA and inhibits the activity of type 2C phosphatases (PP2Cs), thus avoiding the phosphatase-exerted inhibition on SnRK2 kinases, positive regulators which phosphorylate targets and trigger ABA signalling. Thioredoxins (TRXs) are key components of cellular redox homeostasis that regulate specific target proteins through a thiol-disulfide exchange, playing an essential role in redox homeostasis, cell survival, and growth. In higher plants, TRXs have been found in almost all cellular compartments, although its presence and role in nucleus has been less studied. In this work, affinity chromatography, Dot-blot, co-immunoprecipitation, and bimolecular fluorescence complementation assays allowed us to identify PYR1 as a new TRXo1 target in the nucleus. Studies on recombinant HisAtPYR1 oxidation-reduction with wild type and site-specific mutagenized forms showed that the receptor underwent redox regulation involving changes in the oligomeric state in which Cys30 and Cys65 residues were implied. TRXo1 was able to reduce previously-oxidized inactive PYR1, thus recovering its capacity to inhibit HAB1 phosphatase. In vivo PYR1 oligomerization was dependent on the redox state, and a differential pattern was detected in KO and over-expressing Attrxo1 mutant plants grown in the presence of ABA compared to WT plants. Thus, our findings suggest the existence of a redox regulation of TRXo1 on PYR1 that may be relevant for ABA signalling and had not been described so far.

Effects of exogenous glycine betaine and cycloleucine on photosynthetic capacity, amino acid composition, and hormone metabolism in Solanum melongena L.

Although exogenous glycine betaine (GB) and cycloleucine (Cyc) have been reported to affect animal cell metabolism, their effects on plant growth and development have not been studied extensively. Different concentrations of exogenous glycine betaine (20, 40, and 60 mmol L−1) and cycloleucine (10, 20, and 40 mmol L−1), with 0 mmol L−1 as control, were used to investigate the effects of foliar spraying of betaine and cycloleucine on growth, photosynthesis, chlorophyll fluorescence, Calvin cycle pathway, abaxial leaf burr morphology, endogenous hormones, and amino acid content in eggplant. We found that 40 mmol L−1 glycine betaine had the best effect on plant growth and development; it increased the fresh and dry weight of plants, increased the density of abaxial leaf hairs, increased the net photosynthetic rate and Calvin cycle key enzyme activity of leaves, had an elevating effect on chlorophyll fluorescence parameters, increased endogenous indoleacetic acid (IAA) content and decreased abscisic acid (ABA) content, and increased glutamate, serine, aspartate, and phenylalanine contents. However, cycloleucine significantly inhibited plant growth; plant apical dominance disappeared, plant height and dry and fresh weights decreased significantly, the development of abaxial leaf hairs was hindered, the net photosynthetic rate and Calvin cycle key enzyme activities were inhibited, the endogenous hormones IAA and ABA content decreased, and the conversion and utilization of glutamate, arginine, threonine, and glycine were affected. Combined with the experimental results and plant growth phenotypes, 20 mmol L−1 cycloleucine significantly inhibited plant growth. In conclusion, 40 mmol L−1 glycine betaine and 20 mmol L−1 cycloleucine had different regulatory effects on plant growth and development.

Abscisic Acid and Cytokinins Are Not Involved in the Regulation of Stomatal Conductance of Scots Pine Saplings during Post-Drought Recovery

Delayed or incomplete recovery of gas exchange after water stress relief limits assimilation in the post-drought period and can thus negatively affect the processes of post-drought recovery. Abscisic acid (ABA) accumulation and antagonistic action between ABA and cytokinins (CKs) play an important role in regulation of stomatal conductance under water deficit. Specifically, in pine species, sustained ABA accumulation is thought to be the main cause of delayed post-drought gas exchange recovery, although the role of CKs is not yet known. Therefore, we aimed to study the effects of ABA and CKs on recovery of stomatal conductance in greenhouse-grown 3-year-old Scots pine saplings recovering from water stress. We analysed both changes in endogenous ABA and CK contents and the effects of treatment with exogenous CK on stomatal conductance. Drought stress suppressed stomatal conductance, and post-drought stomatal conductance remained suppressed for 2 weeks after plant rewatering. ABA accumulated during water stress, but ABA levels decreased rapidly after rewatering. Additionally, trans-zeatin/ABA and isopentenyladenine/ABA ratios, which were decreased in water-stressed plants, recovered rapidly in rewatered plants. Spraying plants with 6-benzylaminopurine (0.1-100 µM) did not influence recovery of either stomatal conductance or needle water status. It can be concluded that the delayed recovery of stomatal conductance in Scots pine needles was not due to sustained ABA accumulation or a sustained decrease in the CK/ABA ratio, and CK supplementation was unable to overcome this delayed recovery.

Abscisic-Acid-Modulated Stomatal Conductance Governs High-Temperature Stress Tolerance in Rice Accessions

Rising air temperature is a major constraint for crop productivity under the current climate change scenario. Rice crops are known to be sensitive to high-temperature (HT) stress at anthesis and post-anthesis stages. Photosynthesis is an important metabolic process and is affected by HT stress. A pot study was planned to screen a set of seventy-three Indian rice accessions based upon changes in the rate of photosynthesis (Pn) and related gas exchange traits under HT, and to characterize the contrasting rice accessions for component traits of HT stress tolerance. All accessions were raised under ambient temperature (AT) until the booting stage and exposed to HT using controlled chambers at anthesis and post-anthesis. HT exposure led to a large reduction (up to 50%) in Pn, but stomatal conductance (gs) and the rate of transpiration (E) increased significantly across the rice accessions. Based on the photosynthetic response under HT, two contrasting rice accessions (IRGC 135883, tolerant, and IRGC 127222, sensitive) were selected and characterized for HT tolerance, along with an NL-44 check. Among them, Pn decreased marginally but gs and E showed significant increases under HT in the tolerant accession, while sensitive accession showed an up to 50% reduction in Pn and marginal increase in gs and E. No significant changes were recorded for chlorophyll fluorescence (Fv/Fm) in both the genotypes, but tissue temperature depression (TTD) was higher in IRGC 135883 accession under HT. Endogenous abscisic acid (ABA) content increased under HT in the flag leaf of both the accessions, and the highest increase was observed in the sensitive accession. Similarly, spikelet fertility and grain yield showed large reductions in sensitive rice accession under HT. A large increase in ABA concentration in the leaves of the sensitive rice accession might be affecting its gs and cooling capacity under an HT environment. Finally, the study concludes that tolerant rice accessions can be recommended as donors and exploited in future rice breeding programs for developing climate-resilient rice genotypes.

Elucidating Biological Functions of 9-cis-Epoxycarotenoid Dioxygenase Genes Involved in Seed Dormancy in Paeonia lactiflora

Abscisic acid (ABA) is a major phytohormone affecting seed dormancy and germination in plants. ABA is synthesized mainly through the C40 carotenoid pathway. In the ABA biosynthesis pathway, 9-cis-epoxycarotenoid dioxygenase (NCED) is a key rate-limiting enzyme that regulates the accumulation and content of ABA. However, the role of the NCED gene in perennial plants with complex seed dormancy remains largely unknown. Here, we cloned two differentially expressed paralogs of herbaceous peony NCED genes, named PlNCED1 and PlNCED2, and further identified their involvement in seed dormancy from perennial herbaceous peony experiencing complex double seed dormancy. The deduced PlNCED amino acid sequences had high sequence homology with NCED sequences from other plants and contained the typical conserved RPE65 domain of the NCED family. Phylogenetic analysis showed that PlNCED1 and PlNCED2 have a close relationship with PoNCED in Paeonia ostii and VvNCED6 in Vitis vinifera, respectively. A subcellular localization assay demonstrated that the PlNCED1 protein resided within the nucleus, while the PlNCED2 protein was located in the cytoplasm, indicating their different roles in the biosynthesis of ABA. Furthermore, the content of endogenous ABA in transgenic calluses showed that PlNCEDs were positively correlated with ABA content. Both PlNCED transgenic Arabidopsis lines and the functional complementation of Arabidopsis NCED mutants found that PlNCEDs promoted seed dormancy and delayed seed germination. These results reveal that PlNCEDs participate in the seed dormancy of herbaceous peony by regulating the accumulation of endogenous ABA.

Structural And Functional Relationship Study In Plant Salinity Stress

In today’s world the increased use of low-quality water for irrigation and soil salinization, saltness is a huge biotic problem that effecting factory growth and product in numerous corridors in the world. In repercussion to salt stress, plant cells accumulate compatible solutes and redistribute ions, all of which contribute them to acclimate to a low soil water potential. In addition, the endogenous abscisic acid (ABA) content increases daily, followed by changes in principle genetic expression in salt stress conditions. Here we are going to discuss about the proteins involved in plant salinity stress and their interactions with the salt tolerant factor which has a negative impact with the plant growth system. Some highly evaluated proteins that were observed under this stress are Plant ferredoxin-like protein (PFLP), Arabidopsis thaliana Salt Tolerance Related Protein (STRP), OsEXPA7, ribulose-1, 5-bisphosphate carboxylase (RuBP), etc. This report is also going to focus on various transcription factors (tf), responsible for the enhanced tolerance under biological activity of plant development. TFs bZIP, MYBs, bHLH, C2H2-Dof, G2-like, AP2-EREBP, NAC, GATA, ARF, ERF, bZIP and MYB which were highly up- regulated in salt stress response.
