One micro, small, and medium enterprise (MSME) in the Payakumbuh City area is Andika Farm, which focuses on broiler chicken production. This MSME has distributed a lot of broiler chickens and is collaborating with other parties to increase its production. One of the problems faced by Andika Farm MSMEs is capacity planning, empty cages due to a shortage of seeds, disrupted distribution of chickens, and a lack of planning to meet needs. The research aims to help plan capacity by determining the amount of production, constraints that must be considered, optimal profits, and sensitivity values to the optimum solution achieved in MSMEs. This research was conducted using two methods: Linear Programming and Decision Trees. The results of optimizing cage profit 2 produce an EMV of IDR 104,500. Compared with cage 1, cage 2 has the best use of production factors. The quality of the seeds is greatly influenced by the cage's location, where production factors could be more efficient.Keywords: capacity planning; decision tree; linear Programming

A Molecular Dynamics Study on Xe/Kr Separation Mechanisms Using Crystal Growth Method.

The separation of xenon/krypton gas mixtures is a valuable but challenging endeavor in the gas industry due to their similar physical characteristics and closely sized molecules. To address this, we investigated the effectiveness of the hydrate-based gas separation method for mixed Xe-Kr gas via molecular dynamics (MD) simulations. The formation process of hydrates facilitates the encapsulation of guest molecules within hydrate cages, offering a potential strategy for gas separation. Higher temperatures and pressures are advantageous for accelerating the hydrate growth rate. The final occupancy of guest molecules and empty cages within 512, 51264, and all hydrate cages were thoroughly examined. An increase in the pressure and temperature enhanced the occupancy rates of Xe in both 512 and 51264 cages, whereas elevated pressure alone improved the occupancy of Kr in 51264 cages. However, the impact of temperature and pressure on Kr occupancy within 512 cages was found to be minimal. Elevated temperature and pressure resulted in a reduced occupancy of empty cages. Predominantly, 51264 cages were occupied by Xe, whereas Kr showed a propensity to occupy the 512 cages. With increasing simulated pressure, the final occupancy of Xe molecules in all cages rose from 0.37 to 0.41 for simulations at 260 K, while the final occupancy of empty cages decreased from 0.24 to 0.2.

Interplay between the Formation of Colloidal Clathrate and Cubic Diamond Crystals.

Controlling the valency of directional interactions of patchy particles is insufficient for the selective formation of target crystalline structures due to the competition between phases of similar free energy. Examples of such are stacking hybrids of interwoven hexagonal and cubic diamonds with (i) its liquid phase, (ii) arrested glasses, or (iii) clathrates, all depending on the relative patch size, despite being within the one-bond-per-patch regime. Herein, using molecular dynamics simulations, we demonstrate that although tetrahedral patchy particles with narrow patches can assemble into clathrates or stacking hybrids in the bulk, this behavior can be suppressed by the application of external surface potential. Depending on its strength, the selective growth of either cubic diamond crystals or empty sII clathrate cages can be achieved. The formation of a given ordered network depends on the structure of the first adlayer, which is commensurate with the emerging network.

Optimization of an electric field calibration chamber for high-precision measurements for imaging applications

The DEVCOM Army Research Laboratory (ARL) electric field “cage” generates a uniform E-field over a large working volume, along the lines of the IEEE-Std 1308–1994. The end plates are spaced farther apart than the IEEE standard field source, and the fringing fields are controlled by the addition of “guard tubes.” This chamber was originally constructed to calibrate and characterize electric field sensors, and it has been redesigned to support quasi-static electric field imaging applications. A planar array of sensors forms a grounded end plate of the cage and is used to measure distortions in the uniform field generated by the cage due to objects placed inside. However, the array itself distorts this field and introduces significant errors. Several modifications were made to mitigate the errors, including adding nonfunctional “dummy” elements, a border around the array, and a back plane behind it. The parameter space for these additions is very large, since the number of nonfunctional elements, the width of the border, and the size and placement of the back plane can all be tuned independently. Extensive computer modeling was used to explore this parameter space and test thousands of possible designs. The design chosen yields modeled absolute field errors over a 1.2-m × 0.8-m sensing plane that are less than 0.5 % for a uniform ambient field (empty cage), and less than 1 % for a sphere with a 0.5-m radius in an ambient field.

79 Flexible Support Materials Maintain Disc Height and Support the Formation of Hydrated Tissue Engineered Intervertebral Discs in Vivo

OBJECTIVES/GOALS: We evaluated the long-term success of tissue engineered intervertebral discs (TE-IVDs) cultured in flexible (FPLA) or stiff (PLA) support materials, hypothesizing that FPLA would maintain disc height and tissue hydration in the minipig spine. METHODS/STUDY POPULATION: TE-IVD: NP cells were encapsulated in alginate and NP plugs were placed in the center of FPLA cages. AF cells were encapsulated in type I collagen and pipetted around NP plugs. Implantation: Empty FPLA cages (n=4), and TE-IVDs cultured in FPLA (n=4) were implanted at C3-4 or C5-6 following complete discectomy (DX) in skeletally mature minipigs (n=4). Imaging and Quantification: Terminal disc height indices (DHI) were calculated from weekly x-rays using a previously described method, and results were compared to the PLA pilot study. T2 MRI scans were taken of levels treated with TE-IVDs to quantify disc hydration as previously described. RESULTS/ANTICIPATED RESULTS: FPLA cages restored DHIs to native levels until endpoint. In contrast, PLA cages fractured, and terminal DHIs were statistically similar to DX levels. Of the four levels treated with TE-IVDs, 2 were displaced from the disc space. Stabilized levels yielded DHIs which were statistically similar to native IVD and greater than displaced and DX levels. Displaced levels yielded DHIs which were significantly lower than native and stabilized levels, but greater than DX levels (P<0.05). T2 MRIs of stabilized TEIVDs revealed that levels treated with a construct maintained tissue hydration which was significantly greater than levels treated with an empty cage or DX levels (P<0.0001), but which was about half the hydration of native disc tissue. DISCUSSION/SIGNIFICANCE: Implanting TE-IVDs with FPLA support cages leads to disc height maintenance and the stabilization of hydrated tissues in the spine, enhancing the long term success of TE-IVD implants and providing a basis for clinical translation.

Correlations between components of the immune system

Background No evidence of the possibility of altering a constituent of the immune system without directly affecting one of its associated components has yet been shown. Methods A schematic model was developed in which two triggers, fasting and splenectomy, were studied for their ability to affect the expression of cell membrane epitopes and the cytokine secretion of out-of-body autogeneic and syngeneic lymphocytes. Results The effect of fasting and/or splenectomy on promoting correlations between immune systems was studied by determining the alterations in expressions of cell membrane epitopes and in cytokine secretion by out-of-body autogeneic and syngeneic lymphocytes. The effect of fasting as a trigger decreased expression of CD8 and CD25 and increased TNFα levels. The effect of splenectomy as a trigger was investigated in non-fasting mice by comparing splenectomized and non-splenectomized mice. An increase in the CD8 expression and in TNFα, IFNg, and IL10 secretion was noted. The effect of splenectomy as a trigger in fasting mice was determined by comparing splenectomized and non-splenectomized mice. Splenectomy significantly affected the expression of CD25 and CD4 CD25 and on secretion of TNFα, IFNg, and IL10. To determine the effect of keeping the cells in an out-of-body location on the expression of lymphocyte epitopes, tubes kept on top of the cages of the fasting mice were compared with tubes kept on top of empty cages. The results showed a significant change in the CD8 expression was noted. To determine the effect of keeping cells in an out-of-body location on cytokine secretion, tubes kept on cages were tested for cytokine levels significant decrease was noted in the secretion of TNFα and IFNg. Conclusions The study showed that a mouse could affect cells at a distance and alter the expression of surface markers and cytokine secretion following two types of triggers: fasting and/or splenectomy. The data characterized a system for the induction of correlations between two’s immune system components without a transfer of mediators. It suggests that an out-of-body correlation can be induced between two components of the immune system.

Abstract WMP75: Neurobehavioral Outcomes Following Left- and Right-Sided Intracerebral Hemorrhage in Mice

Background: Injury location is an established outcome predictor in intracerebral hemorrhage (ICH), with most studies focusing on outcomes relating to motor deficits. However, location may also affect other neurobehavioral deficits following ICH, such as mood and cognitive decline. To address this gap, we studied neurobehavioral outcomes following left and right-sided ICH in mice. Methods: Three-month-old male mice (C57BL/6) were given a left (n=5) or right-sided (n=5) ICH. In short, type VII collagenase was injected into the left or right striatum (0.0 mm anterior, +/- 2.3 mm lateral, and 3.6 mm deep from bregma). Naïve mice (n=5) were used as controls. Behavior was assessed at day 28 post-ICH using a video-recorded three-chamber sociability task in which one chamber had an enclosed male stranger mouse. Distance traveled, time in each chamber, and time exploring the stranger mouse were quantified using automated software (AnyMaze). Data were analyzed by one-way ANOVA and Tukey post-hoc tests. Results: ICH location had differential effects on behavioral outcomes. Compared to naïve and right-sided ICH mice, left-sided ICH mice traveled a greater overall distance exploring the chambers (both p<0.05), spent a greater overall time immobile in the stranger mouse zone (both p<0.001) and less time oriented towards the stranger mouse (both p<0.05). Furthermore, when comparing naive and left-sided ICH mice, left-sided mice had a higher frequency of investigations of the empty cage (p<0.05). No significant differences were found between the naïve and right-sided ICH mice. Conclusions: These results reveal differences in behavioral phenotypes associated with left- and right-sided ICH.

Fluid–Structure Interactions of Net Cages—Full-Scale Pushing Tests in the Field

Abstract This paper presents field tests on a full-scale cage, with and without fish, being pushed by a boat in Masfjorden at various speeds. The purpose was to imitate the exposure of net cages to different currents. The tests involved measuring cage deformations, fish behaviors, and the corresponding flow upstream, downstream, and inside the cage. The study found that the experimental setup used can achieve predictable and stable upstream flow for a full-scale net cage. Based on pressure tag data, the volume reductions of the cage, both with and without fish, were estimated at different speeds. Both cases show a similar trend of cage volume reduction with respect to flow speeds as the previous studies. Moreover, the presence of fish had limited the influence on the net volume change. The reduction in speed inside and downstream from the cage was within the range reported in previous literature. Notably, when the cage becomes significantly deformed, it not only reduces flow speed but also alters flow directions, as evidenced by the high variability of flow direction inside the empty cage, particularly at high speeds. The measured flow speed inside the stocked cage also exhibited high variability, but the pattern of variation differed significantly from that of the empty cage, indicating the influence of fish. These findings suggest that traditional flow speed models might oversimplify the flow field in and around fish cages, especially in studies concerning the dispersion of particles, pathogens, and dissolved matter in and out of fish cages.

Hemicryptophane: A Viable Supramolecular Approach to Improve Chlordecone Removal from Water

AbstractThe persistent organic pollutant chlordecone massively contaminates water resources of the French West Indies. Hemicryptophanes are cages capable of complexing chlordecone in organic solvents. Here we demonstrate that a hemicryptophane cage is not only able to recognize this pollutant in chloroform but is also efficient for the removal of chlordecone from water. This evidences that, beyond our previously reported proof of concept, this receptor could be apply as a direct and efficient depolluting agent, making it a promising system for applied supramolecular chemistry. A simple pH switch (basic washing) enables the release of chlordecone from the host, under its deprotonated form, allowing the empty supramolecular cage to be recycled and reused. This method for recycling the receptor, is to the best of our knowledge unprecedented.

Polymerase Chain Reaction on In-cage Filter Paper at Different Time Points to Detect Helicobacter spp.

Helicobacter spp. infections in mice can have broad-ranging effects on gastrointestinal, reproductive, and immune systems. This can introduce significant confounding variables for research and may reduce scientific rigor. Screening mouse colonies for Helicobacter species can be accomplished via noninvasive PCR testing on filter paper placed in animal-free dirty bedding sentinel cages. In our facility, one tablespoon of dirty bedding from each cage on a rack is added to a designated sentinel cage every 3 wk at cage change, and PCR testing is performed on in-cage filter paper quarterly. We hypothesized that cages that received Helicobacter spp.-positive bedding at later time points would have a lower detection rate of Helicobacter spp. with PCR testing compared with cages that received positive bedding at earlier time points due to the filter paper becoming saturated. To determine if screening would be able to detect one positive row of cages on a rack, 9 tablespoons of Helicobacter- positive bedding and 71 tablespoons of negative bedding were added at the 3-, 6-, or 9-wk time points to 14 empty sentinel cages per time point. Negative bedding was added every 3 wk to cages not scheduled to receive positive bedding. Negative controls received 80 tablespoons of negative bedding and positive controls received 80 tablespoons of positive bedding at each time point. Filter paper was tested via PCR for Helicobacter spp. at 12 wk. All positive controls tested positive, and all negative controls tested negative. Two 3-wk cages, two 6-wk cages, and three 9-wk cages were positive, indicating no difference between time points. This resulted in a 16.7% Helicobacter spp. detection rate. These results indicate that PCR on in-cage filter paper may not be reliable in detecting low levels of Helicobacter spp. nucleic acid in dirty bedding.

Let's stick together: Infection enhances preferences for social grouping in a songbird species.

Acute infections can alter foraging and movement behaviors relevant to sociality and pathogen spread. However, few studies have directly examined how acute infections caused by directly transmitted pathogens influence host social preferences. While infected hosts often express sickness behaviors (e.g., lethargy) that can reduce social associations with conspecifics, enhanced sociality during infection might be favored in some systems if social grouping improves host survival of infection. Directly assaying social preferences of infected hosts is needed to elucidate potential changes in social preferences that may act as a form of behavioral tolerance (defined as using behavior to minimize fitness costs of infection). We tested how infection alters sociality in juvenile house finches (Haemorhous mexicanus), which are both highly gregarious and particularly susceptible to infection by the bacterial pathogen Mycoplasma gallisepticum (MG). We inoculated 33 wild-caught but captive-held juvenile house finches with MG or media (sham control). At peak infection, birds were given a choice assay to assess preference for associating near a flock versus an empty cage. We then repeated this assay after all birds had recovered from infection. Infected birds were significantly more likely than controls to spend time associating with, and specifically foraging near, the flock. However, after infected birds had recovered from MG infection, there were no significant differences in the amount of time birds in each treatment spent with the flock. These results indicate augmented social preferences during active infection, potentially as a form of behavioral tolerance. Notably, infected birds showed strong social preferences regardless of variation in disease severity or pathogen loads, with 14/19 harboring high loads (5-6 log10 copies of MG) at the time of the assay. Overall, our results show that infection with a directly transmitted pathogen can augment social preferences, with important implications for MG spread in natural populations.

Eco-saving and green anode materials for Li-ion batteries utilizing oxide-decorated biocarbon substrates proceeded from cassava residues

In this work, an eco-saving and green anode structure (CRC) of MnO/Mn3O4 nanoparticles (NPs) anchored on biocarbon substrates derived from cassava residues (CRs) was designed and fabricated successfully by an in-situ method from MnCl2 and CRs. The amount of MnO/Mn3O4 NPs in the CRC can be effectively modulated by altering the mass ratio of MnCl2 to CRs. In Li-ion batteries, CRC anodes exhibit outstanding electrochemical performances such as high Coulombic efficiency, high capacity, excellent rate capacity, and stable cycling. The high reversible capacity of 698 mAh g−1 at 100 mA g−1 can be achieved even after 100 cycles. This could be accredited to the special structure of MnO/Mn3O4 NPs and biocarbon substrates. First, CR-derived biocarbon favors the alleviation of volume change and prevents the aggregation of MnO/Mn3O4 NPs during cycling. Then, an appropriate amount of MnO/Mn3O4 NPs formed on the surface and empty cages of biocarbon generate various free contacting channels between the electrolyte and active materials. Finally, MnO/Mn3O4 NPs intensify the electrochemical activity of biocarbon, leading to an increase in Li-storage capacities. Therefore, the facile in-situ method in this work is quite promising to produce cost-effective and eco-friendly anodes for Li-ion batteries utilizing recycled CRs and simple chemicals.

Fear response of rat pups to a non-aversive social stimulus: Evidence for the involvement of memory processes.

Learning processes in rats during early development are importantly mediated by the mother, which represents the primary source of environmental information. This study aimed to determine whether aversive early experiences can induce the expression of pups' fear responses toward a non-aversive stimulus as a consequence of a memory process. First, we determined pups' fear responses toward an anesthetized female after being exposed to this stimulus or an empty cage together with their mothers from Postnatal Day (PNDs) 1 to 4. Second, we evaluated if the administration of the protein synthesis inhibitor cycloheximide (CHX; 0.2mg/kg, subcutaneously (sc).) disrupted the reconsolidation processes and abolished the fear response on PND 9. Only female pups previously exposed to the female intruder expressed fear responses toward an anesthetized female on PND 8. CHX administration to female pups immediately after exposure to an anesthetized female on PND 8 suppressed fear responses on PND 9, indicating that the fear expression was the result of a memory process, probably mediated by the mother. These findings demonstrated that early experiences can shape responses to social stimuli in a sex-dependent manner and emphasize the critical role of the mother in influencing fear learning in a social context.

Kinetics studies of methane hydrate formation and dissociation in the presence of cyclopentane

The work systematically investigates the kinetic of methane(CH4) hydrate formation in the presence of cyclopentane（CP）by measuring induction time, hydrate formation rate, and gas uptake. The results showed that there are two steps for CH4 hydrate formation in the presence of CP: Step 1, pure CP hydrate formation. Step 2, gas enters the empty cages of pure CP hydrates to form CP/CH4 mixed hydrates. Besides, the rapid hydrate formation rate and high CH4 uptake are observed from hydrate formation in this work. At the optimum conditions of 288.15 K and CP mole fractions of 5.6 mol%, the gas uptake and the time required for completion of hydrate formation are 131.80 mmol/mol H2O and 138 min, respectively. Relative to the highest reported gas uptake obtained in the presence of thermodynamic promoter (72.0 mmol/mol H2O), the value increased by 83%. More strikingly, the work shows that high gas uptake hydrates formed at relatively low temperatures can be stored and transported at relatively high temperatures, as high as 298.15 K, without significantly reducing the gas uptake, thereby providing one of the cost-effective methods for hydrate storage and transportation, that is forming hydrates at relatively low temperatures and storing and transporting hydrates at relatively high temperatures. Further, we proposed a schematic of the process chain of SNG(storing natural gas via solid hydrate) technology in vehicular and detailed discuss the current status of SNG technology development. It can be concluded that using the strategy that storage and transportation hydrates at a low-pressure condition instead of at ambient pressure (1.0 atm) may be a more realistic choice to industrialize SNG technology.

Molecular basis for inhibition of methane clathrate growth by a deep subsurface bacterial protein.

Methane clathrates on continental margins contain the largest stores of hydrocarbons on Earth, yet the role of biomolecules in clathrate formation and stability remains almost completely unknown. Here, we report new methane clathrate-binding proteins (CbpAs) of bacterial origin discovered in metagenomes from gas clathrate-bearing ocean sediments. CbpAs show similar suppression of methane clathrate growth as the commercial gas clathrate inhibitor polyvinylpyrrolidone and inhibit clathrate growth at lower concentrations than antifreeze proteins (AFPs) previously tested. Unlike AFPs, CbpAs are selective for clathrate over ice. CbpA3 adopts a nonglobular, extended structure with an exposed hydrophobic surface, and, unexpectedly, its TxxxAxxxAxx motif common to AFPs is buried and not involved in clathrate binding. Instead, simulations and mutagenesis suggest a bipartite interaction of CbpAs with methane clathrate, with the pyrrolidine ring of a highly conserved proline residue mediating binding by filling empty clathrate cages. The discovery that CbpAs exert such potent control on methane clathrate properties implies that biomolecules from native sediment bacteria may be important for clathrate stability and habitability.

CH3 CN@open-C60 : An Effective Inner-Space Modification and Isotope Effect Inside a Nano-Sized Flask.

Despite several small molecules being encapsulated inside cage-opened fullerene derivatives, such species have not considerably affected the structures and properties of the outer carbon cages. Herein, we achieved an effective inner-space modification for an open-cage C60 derivative by insertion of a neutral CH3 CN molecule into the cavity. The CH3 CN@open-C60 thus obtained showed an enhanced polarity, thus affording an easy separation from a mixture containing the empty cage by column chromatography on silica gel, without the preparative HPLC that was needed for previous cases. The less negative reduction potentials with respect to those of empty cage reflect the decreased energy level of the LUMO, which is supported by the DFT calculations. NMR spectroscopy, single-crystal X-ray analysis, and theoretical calculations revealed that both the presence of the encapsulated CH3 CN and cage deformation caused by the CH3 CN play an essential role in the change of the electronic properties. Furthermore, the favored binding affinity of deuterated acetonitrile CD3 CN with internal C60 surface is discussed.

Evaluation of Parameters Which Influence Voluntary Ingestion of Supplements in Rats.

Drug safety and efficacy studies frequently use oral gavage, but repetitive usage may cause problems. Administration through voluntary ingestion represents an opportunity for refinement. We aimed to develop a protocol for voluntary ingestion of gelatin-based supplements in rats, assessing the influence of age, sex, fasting (4 h), and additives (vanilla, VF; sucralose, S), and to test it in lactating dams. Three-week-old and 5-month-old Sprague-Dawley rats were placed individually in an empty cage containing a gelatin cube and trained daily (5 days/week), recording the day the whole cube was consumed (latency). Rats trained prior to gestation were offered a gelatin containing 250 mg/kg cocoa shell extract (CSE) during lactation. Rats that did not eat the cube after 8 training days were considered non-habituated, with a proportion similar in young males (7.1%), young females (11.1%), and adult females (10.3%), but significantly higher in adult males (39.3%). Excluding non-habituated rats, latency was 2-3 days, without differences between young and adult rats (p = 0.657) or between males and females (p = 0.189). VF or VF + S in the gelatin did not modify latency, while fasting significantly reduced it in females (p = 0.007) but not in males (p = 0.501). During lactation, trained females ate the CSE-gelatin within 1-5 min without litter problems. Conclusions: Acceptance of a gelatin-based supplement is negatively influenced by male sex, facilitated by fasting, and not modified by additives. Training is remembered after 2 months and does not interfere with lactation. Gelatin-based voluntary ingestion is suitable to administer drugs that need to pass through the digestive system, ensuring adequate dosage, and is important to detect non-habituated rats prior to the study. The current protocol may be implemented by training the rats in their own cage.

Study on Carbon Storage by CO<sub>2</sub> Hydrate Formation in the Ion-Containing Environment

The current greenhouse effect caused by massive CO₂ accumulation must be addressed urgently. Because of the ocean's enormous capacity, the high density of CO₂ hydrate, and the high stability of CO₂ sequestration in hydrate form, the process of hydrate generation can trap CO₂ molecules into the lattice of water molecules and is considered for the sequestration of CO₂ in hydrate form in the ocean. In this paper, molecular dynamics (MD) simulations are used to investigate the kinetic behavior of CO₂ hydrate growth in ion-containing electrolyte solutions. The mechanism of salt ion effect on hydrate growth during pressure changes is investigated. The simulations show that in the presence of salt ions, the hydrate growth rate fluctuates significantly, despite the fact that a greater driving force can facilitate the hydrate growth process. Higher pressures increase guest molecule occupancy in the hydrate cage, while the saline environment causes competition for water molecules between the hydrate cage and salt ions, increasing the proportion of empty cages. Lower pressures and the presence of salt ions reduce CO₂ molecules' selectivity for the smaller 5¹² cages, creating a barrier to hydrate growth. The findings of the study look at the microscopic mechanism of CO₂ hydrate generation in seawater and serve as a guide for future implementation of hydrate method CO₂ sequestration technology in the marine environment.

Molecular insights into the dissociation of carbon dioxide hydrates in the presence of an ionic liquid, [BMIM][PF6

Methane production from hydrate reservoirs using combined CO2and chemical inhibitor injection is a potential method for simultaneous methane recovery and CO2sequestration. Molecular dynamics simulations have been performed to study the dissociation of CO2hydrates in the presence and absence of the ionic liquid, viz., [BMIM][PF6], at 250, 255 and 260 K and pressures ranging from 20 to 60 bar. Effect of [BMIM][PF6] on hydrate thickness has been demonstrated to be not significant by hydrogen bonding and order parameter analyses. Local number density profiles of guest and host molecules show that in presence of this ionic liquid, deviation from hydrate-like behaviour is less than in its absence. This is due to preferential association of CO2with [PF6]- anion. The asymmetry in the density profiles of CO2 and H2O brings out the role of filled and empty partial cages at the interfaces on hydrate dissociation in the presence of [BMIM][PF6].
