Predicting resting-state brain functional connectivity from the structural connectome using the heat diffusion model: a multiple-timescale fusion method

Objective. Understanding the intricate relationship between structural connectivity (SC) and functional connectivity (FC) is pivotal for understanding the complexities of the human brain. To explore this relationship, the heat diffusion model (HDM) was utilized to predict FC from SC. However, previous studies using the HDM have typically predicted FC at a critical time scale in the heat kernel equation, overlooking the dynamic nature of the diffusion process and providing an incomplete representation of the predicted FC. Approach. In this study, we propose an alternative approach based on the HDM. First, we introduced a multiple-timescale fusion method to capture the dynamic features of the diffusion process. Additionally, to enhance the smoothness of the predicted FC values, we employed the Wavelet reconstruction method to maintain local consistency and remove noise. Moreover, to provide a more accurate representation of the relationship between SC and FC, we calculated the linear transformation between the smoothed FC and the empirical FC. Main results. We conducted extensive experiments in two independent datasets. By fusing different time scales in the diffusion process for predicting FC, the proposed method demonstrated higher predictive correlation compared with method considering only critical time points (Singlescale). Furthermore, compared with other existing methods, the proposed method achieved the highest predictive correlations of 0.6939 ± 0.0079 and 0.7302 ± 0.0117 on the two datasets respectively. We observed that the visual network at the network level and the parietal lobe at the lobe level exhibited the highest predictive correlations, indicating that the functional activity in these regions may be closely related to the direct diffusion of information between brain regions. Significance. The multiple-timescale fusion method proposed in this study provides insights into the dynamic aspects of the diffusion process, contributing to a deeper understanding of how brain structure gives rise to brain function.

Construction and Analysis of a New Resting-State Whole-Brain Network Model.

Mathematical modeling and computer simulation are important methods for understanding complex neural systems. The whole-brain network model can help people understand the neurophysiological mechanisms of brain cognition and functional diseases of the brain. In this study, we constructed a resting-state whole-brain network model (WBNM) by using the Wendling neural mass model as the node and a real structural connectivity matrix as the edge of the network. By analyzing the correlation between the simulated functional connectivity matrix in the resting state and the empirical functional connectivity matrix, an optimal global coupling coefficient was obtained. Then, the waveforms and spectra of simulated EEG signals and four commonly used measures from graph theory and small-world network properties of simulated brain networks under different thresholds were analyzed. The results showed that the correlation coefficient of the functional connectivity matrix of the simulated WBNM and empirical brain networks could reach a maximum value of 0.676 when the global coupling coefficient was set to 20.3. The simulated EEG signals showed rich waveform and frequency-band characteristics. The commonly used graph-theoretical measures and small-world properties of the constructed WBNM were similar to those of empirical brain networks. When the threshold was set to 0.22, the maximum correlation between the simulated WBNM and empirical brain networks was 0.709. The constructed resting-state WBNM is similar to a real brain network to a certain extent and can be used to study the neurophysiological mechanisms of complex brain networks.

A phenomenological model of whole brain dynamics using a network of neural oscillators with power-coupling

We present a general, trainable oscillatory neural network as a large-scale model of brain dynamics. The model has a cascade of two stages - an oscillatory stage and a complex-valued feedforward stage - for modelling the relationship between structural connectivity and functional connectivity from neuroimaging data under resting brain conditions. Earlier works of large-scale brain dynamics that used Hopf oscillators used linear coupling of oscillators. A distinctive feature of the proposed model employs a novel form of coupling known as power coupling. Oscillatory networks based on power coupling can accurately model arbitrary multi-dimensional signals. Training the lateral connections in the oscillator layer is done by a modified form of Hebbian learning, whereas a variation of the complex backpropagation algorithm does training in the second stage. The proposed model can not only model the empirical functional connectivity with remarkable accuracy (correlation coefficient between simulated and empirical functional connectivity- 0.99) but also identify default mode network regions. In addition, we also inspected how structural loss in the brain can cause significant aberration in simulated functional connectivity and functional connectivity dynamics; and how it can be restored with optimized model parameters by an in silico perturbational study.

Comparing individual and group-level simulated neurophysiological brain connectivity using the Jansen and Rit neural mass model.

Computational models are often used to assess how functional connectivity (FC) patterns emerge from neuronal population dynamics and anatomical brain connections. It remains unclear whether the commonly used group-averaged data can predict individual FC patterns. The Jansen and Rit neural mass model was employed, where masses were coupled using individual structural connectivity (SC). Simulated FC was correlated to individual magnetoencephalography-derived empirical FC. FC was estimated using phase-based (phase lag index (PLI), phase locking value (PLV)), and amplitude-based (amplitude envelope correlation (AEC)) metrics to analyze their goodness of fit for individual predictions. Individual FC predictions were compared against group-averaged FC predictions, and we tested whether SC of a different participant could equally well predict participants' FC patterns. The AEC provided a better match between individually simulated and empirical FC than phase-based metrics. Correlations between simulated and empirical FC were higher using individual SC compared to group-averaged SC. Using SC from other participants resulted in similar correlations between simulated and empirical FC compared to using participants' own SC. This work underlines the added value of FC simulations using individual instead of group-averaged SC for this particular computational model and could aid in a better understanding of mechanisms underlying individual functional network trajectories.

P13.03.B PREDICTING FUNCTIONAL CONNECTIVITY FROM STRUCTURAL CONNECTIVITY IN GLIOMA PATIENTS

Abstract BACKGROUND The mechanism by which activity in the brain relates to its anatomical structure is a central topic of research in neuroscience. In recent years, this relation has been studied in the context of network science, where anatomical white-matter connections are captured in Structural Connectivity (SC) and statistical dependence between grey-matter activity in Functional Connectivity (FC). The relation between SC and FC is poorly understood at an individual level, especially in glioma patients. Developing methods to predict FC from SC in these patients could improve our understanding of how gliomas affect brain function, and consequently cognition. Furthermore, a model accurate at individual patient level could aid in clinical decision-making by identifying eloquent areas and connections in individuals. MATERIAL AND METHODS Building on earlier work predicting FC from SC in healthy subjects, we trained a deep learning model on tractography and resting-state fMRI data of 288 glioma patients to predict FC at an individual level. Since our sample size is modest in the context of deep learning, we attempted to improve accuracy by pre-training the model on a large set of healthy subjects. This way, the model should learn baseline patterns in SC and FC before learning the specific patterns associated with glioma patients. We used data from 1052 subjects of the Human Connectome Project and 633 subjects of the Cambridge Centre for Ageing and Neuroscience. Finally, we analyzed the effect of several factors on prediction accuracy: tumor volume, location and grade and patient age. RESULTS Training on only the patient set, the prediction model achieved an accuracy, measured as correlation between predicted and empirical FC, of 0.53 ± 0.03 at individual patient level. This is comparable with accuracy achieved in studies on healthy subjects. Pre-trained models did not improve accuracy. The model had more difficulty predicting FC in patients with a glioma in the insular region (p = 0.0014). No other factors were associated with accuracy. CONCLUSION Although further research is required to investigate the utility of FC predictions made by our model, our work is a first step towards predicting activity from brain structure in glioma patients at an individual level, since earlier research in patient populations has focused on group-level effects. We observed that predictions are less accurate in patients with a glioma in the insular region, which indicates that the relation between the structural and functional organization of the brain in this group is different from that in patients with tumors in other locations. Improving our methods to achieve higher accuracy could provide us, as a next step, with a tool to predict functional consequences of resective surgery on glioma patients, and consequently improve decision making by more accurately identifying eloquent regions at an individual level.

Modeling the role of the thalamus in resting-state functional connectivity: Nature or structure.

The thalamus is a central brain structure that serves as a relay station for sensory inputs from the periphery to the cortex and regulates cortical arousal. Traditionally, it has been regarded as a passive relay that transmits information between brain regions. However, recent studies have suggested that the thalamus may also play a role in shaping functional connectivity (FC) in a task-based context. Based on this idea, we hypothesized that due to its centrality in the network and its involvement in cortical activation, the thalamus may also contribute to resting-state FC, a key neurological biomarker widely used to characterize brain function in health and disease. To investigate this hypothesis, we constructed ten in-silico brain network models based on neuroimaging data (MEG, MRI, and dwMRI), and simulated them including and excluding the thalamus, and raising the noise into thalamus to represent the afferences related to the reticular activating system (RAS) and the relay of peripheral sensory inputs. We simulated brain activity and compared the resulting FC to their empirical MEG counterparts to evaluate model's performance. Results showed that a parceled version of the thalamus with higher noise, able to drive damped cortical oscillators, enhanced the match to empirical FC. However, with an already active self-oscillatory cortex, no impact on the dynamics was observed when introducing the thalamus. We also demonstrated that the enhanced performance was not related to the structural connectivity of the thalamus, but to its higher noisy inputs. Additionally, we highlighted the relevance of a balanced signal-to-noise ratio in thalamus to allow it to propagate its own dynamics. In conclusion, our study sheds light on the role of the thalamus in shaping brain dynamics and FC in resting-state and allowed us to discuss the general role of criticality in the brain at the mesoscale level.

Multi-modal and multi-model interrogation of large-scale functional brain networks

Existing whole-brain models are generally tailored to the modelling of a particular data modality (e.g., fMRI or MEG/EEG). We propose that despite the differing aspects of neural activity each modality captures, they originate from shared network dynamics. Building on the universal principles of self-organising delay-coupled nonlinear systems, we aim to link distinct features of brain activity - captured across modalities - to the dynamics unfolding on a macroscopic structural connectome.To jointly predict connectivity, spatiotemporal and transient features of distinct signal modalities, we consider two large-scale models - the Stuart Landau and Wilson and Cowan models - which generate short-lived 40 Hz oscillations with varying levels of realism. To this end, we measure features of functional connectivity and metastable oscillatory modes (MOMs) in fMRI and MEG signals - and compare them against simulated data.We show that both models can represent MEG functional connectivity (FC), functional connectivity dynamics (FCD) and generate MOMs to a comparable degree. This is achieved by adjusting the global coupling and mean conduction time delay and, in the WC model, through the inclusion of balance between excitation and inhibition. For both models, the omission of delays dramatically decreased the performance. For fMRI, the SL model performed worse for FCD and MOMs, highlighting the importance of balanced dynamics for the emergence of spatiotemporal and transient patterns of ultra-slow dynamics. Notably, optimal working points varied across modalities and no model was able to achieve a correlation with empirical FC higher than 0.4 across modalities for the same set of parameters. Nonetheless, both displayed the emergence of FC patterns that extended beyond the constraints of the anatomical structure. Finally, we show that both models can generate MOMs with empirical-like properties such as size (number of brain regions engaging in a mode) and duration (continuous time interval during which a mode appears).Our results demonstrate the emergence of static and dynamic properties of neural activity at different timescales from networks of delay-coupled oscillators at 40 Hz. Given the higher dependence of simulated FC on the underlying structural connectivity, we suggest that mesoscale heterogeneities in neural circuitry may be critical for the emergence of parallel cross-modal functional networks and should be accounted for in future modelling endeavours.

Network Hyperexcitability in Early Alzheimer’s Disease: Is Functional Connectivity a Potential Biomarker?

Network hyperexcitability (NH) is an important feature of the pathophysiology of Alzheimer’s disease. Functional connectivity (FC) of brain networks has been proposed as a potential biomarker for NH. Here we use a whole brain computational model and resting-state MEG recordings to investigate the relation between hyperexcitability and FC. Oscillatory brain activity was simulated with a Stuart Landau model on a network of 78 interconnected brain regions. FC was quantified with amplitude envelope correlation (AEC) and phase coherence (PC). MEG was recorded in 18 subjects with subjective cognitive decline (SCD) and 18 subjects with mild cognitive impairment (MCI). Functional connectivity was determined with the corrected AECc and phase lag index (PLI), in the 4–8 Hz and the 8–13 Hz bands. The excitation/inhibition balance in the model had a strong effect on both AEC and PC. This effect was different for AEC and PC, and was influenced by structural coupling strength and frequency band. Empirical FC matrices of SCD and MCI showed a good correlation with model FC for AEC, but less so for PC. For AEC the fit was best in the hyperexcitable range. We conclude that FC is sensitive to changes in E/I balance. The AEC was more sensitive than the PLI, and results were better for the thetaband than the alpha band. This conclusion was supported by fitting the model to empirical data. Our study justifies the use of functional connectivity measures as surrogate markers for E/I balance.

Detecting Changes in Correlation Networks with Application to Functional Connectivity of fMRI Data.

Research questions in the human sciences often seek to answer if and when a process changes across time. In functional MRI studies, for instance, researchers may seek to assess the onset of a shift in brain state. For daily diary studies, the researcher may seek to identify when a person's psychological process shifts following treatment. The timing and presence of such a change may be meaningful in terms of understanding state changes. Currently, dynamic processes are typically quantified as static networks where edges indicate temporal relations among nodes, which may be variables reflecting emotions, behaviors, or brain activity. Here we describe three methods for detecting changes in such correlation networks from a data-driven perspective. Networks here are quantified using the lag-0 pair-wise correlation (or covariance) estimates as the representation of the dynamic relations among variables. We present three methods for change point detection: dynamic connectivity regression, max-type method, and a PCA-based method. The change point detection methods each include different ways to test if two given correlation network patterns from different segments in time are significantly different. These tests can also be used outside of the change point detection approaches to test any two given blocks of data. We compare the three methods for change point detection as well as the complementary significance testing approaches on simulated and empirical functional connectivity fMRI data examples.

Using The Virtual Brain to study the relationship between structural and functional connectivity in patients with multiple sclerosis: a multicenter study.

The relationship between structural connectivity (SC) and functional connectivity (FC) captured from magnetic resonance imaging, as well as its interaction with disability and cognitive impairment, is not well understood in people with multiple sclerosis (pwMS). The Virtual Brain (TVB) is an open-source brain simulator for creating personalized brain models using SC and FC. The aim of this study was to explore SC-FC relationship in MS using TVB. Two different model regimes have been studied: stable and oscillatory, with the latter including conduction delays in the brain. The models were applied to 513 pwMS and 208 healthy controls (HC) from 7 different centers. Models were analyzed using structural damage, global diffusion properties, clinical disability, cognitive scores, and graph-derived metrics from both simulated and empirical FC. For the stable model, higher SC-FC coupling was associated with pwMS with low Single Digit Modalities Test (SDMT) score (F=3.48, P$\lt$0.05), suggesting that cognitive impairment in pwMS is associated with a higher SC-FC coupling. Differences in entropy of the simulated FC between HC, high and low SDMT groups (F=31.57, P$\lt$1e-5), show that the model captures subtle differences not detected in the empirical FC, suggesting the existence of compensatory and maladaptive mechanisms between SC and FC in MS.

Strength-dependent perturbation of whole-brain model working in different regimes reveals the role of fluctuations in brain dynamics.

Despite decades of research, there is still a lack of understanding of the role and generating mechanisms of the ubiquitous fluctuations and oscillations found in recordings of brain dynamics. Here, we used whole-brain computational models capable of presenting different dynamical regimes to reproduce empirical data's turbulence level. We showed that the model's fluctuations regime fitted to turbulence more faithfully reproduces the empirical functional connectivity compared to oscillatory and noise regimes. By applying global and local strength-dependent perturbations and subsequently measuring the responsiveness of the model, we revealed each regime's computational capacity demonstrating that brain dynamics is shifted towards fluctuations to provide much-needed flexibility. Importantly, fluctuation regime stimulation in a brain region within a given resting state network modulates that network, aligned with previous empirical and computational studies. Furthermore, this framework generates specific, testable empirical predictions for human stimulation studies using strength-dependent rather than constant perturbation. Overall, the whole-brain models fitted to the level of empirical turbulence together with functional connectivity unveil that the fluctuation regime best captures empirical data, and the strength-dependent perturbative framework demonstrates how this regime provides maximal flexibility to the human brain.

Reliability and subject specificity of personalized whole-brain dynamical models

Dynamical whole-brain models were developed to link structural (SC) and functional connectivity (FC) together into one framework. Nowadays, they are used to investigate the dynamical regimes of the brain and how these relate to behavioral, clinical and demographic traits. However, there is no comprehensive investigation on how reliable and subject specific the modeling results are given the variability of the empirical FC. In this study, we show that the parameters of these models can be fitted with a ”poor” to ”good” reliability depending on the exact implementation of the modeling paradigm. We find, as a general rule of thumb, that enhanced model personalization leads to increasingly reliable model parameters. In addition, we observe no clear effect of the model complexity evaluated by separately sampling results for linear, phase oscillator and neural mass network models. In fact, the most complex neural mass model often yields modeling results with ”poor” reliability comparable to the simple linear model, but demonstrates an enhanced subject specificity of the model similarity maps. Subsequently, we show that the FC simulated by these models can outperform the empirical FC in terms of both reliability and subject specificity. For the structure-function relationship, simulated FC of individual subjects may be identified from the correlations with the empirical SC with an accuracy up to 70%, but not vice versa for non-linear models. We sample all our findings for 8 distinct brain parcellations and 6 modeling conditions and show that the parcellation-induced effect is much more pronounced for the modeling results than for the empirical data. In sum, this study provides an exploratory account on the reliability and subject specificity of dynamical whole-brain models and may be relevant for their further development and application. In particular, our findings suggest that the application of the dynamical whole-brain modeling should be tightly connected with an estimate of the reliability of the results.

Towards an efficient validation of dynamical whole-brain models

Simulating the resting-state brain dynamics via mathematical whole-brain models requires an optimal selection of parameters, which determine the model’s capability to replicate empirical data. Since the parameter optimization via a grid search (GS) becomes unfeasible for high-dimensional models, we evaluate several alternative approaches to maximize the correspondence between simulated and empirical functional connectivity. A dense GS serves as a benchmark to assess the performance of four optimization schemes: Nelder-Mead Algorithm (NMA), Particle Swarm Optimization (PSO), Covariance Matrix Adaptation Evolution Strategy (CMAES) and Bayesian Optimization (BO). To compare them, we employ an ensemble of coupled phase oscillators built upon individual empirical structural connectivity of 105 healthy subjects. We determine optimal model parameters from two- and three-dimensional parameter spaces and show that the overall fitting quality of the tested methods can compete with the GS. There are, however, marked differences in the required computational resources and stability properties, which we also investigate before proposing CMAES and BO as efficient alternatives to a high-dimensional GS. For the three-dimensional case, these methods generated similar results as the GS, but within less than 6% of the computation time. Our results contribute to an efficient validation of models for personalized simulations of brain dynamics.

Learning to Model the Relationship Between Brain Structural and Functional Connectomes

Recent neuroimaging advances along with algorithmic innovations in statistical learning from network data offer a unique pathway to integrate brain structure and function, and thus facilitate revealing some of the brain's organizing principles at the system level. In this direction, we develop a supervised graph representation learning framework to model the relationship between brain structural connectivity (SC) and functional connectivity (FC) via a graph encoder-decoder system, where the SC is used as input to predict empirical FC. A trainable graph convolutional encoder captures direct and indirect interactions between brain regions-of-interest that mimic actual neural communications, as well as to integrate information from both the structural network topology and nodal (i.e., region-specific) attributes. The encoder learns node-level SC embeddings which are combined to generate (whole brain) graph-level representations for reconstructing empirical FC networks. The proposed end-to-end model utilizes a multi-objective loss function to jointly reconstruct FC networks and learn discriminative graph representations of the SC-to-FC mapping for downstream subject (i.e., graph-level) classification. Comprehensive experiments demonstrate that the learnt representations of said relationship capture valuable information from the intrinsic properties of the subject's brain networks and lead to improved accuracy in classifying a large population of heavy drinkers and non-drinkers from the Human Connectome Project. Our work offers new insights on the relationship between brain networks that support the promising prospect of using graph representation learning to discover more about brain function.

Numerical Simulation of Higher-Order Nonlinearity of Human Brain Functional Connectivity Using Hypergraph p-Laplacian

Unravelling how the human brain structure gives rise to function is a central question in neuroscience and remains partially answered. Recent studies show that the graph Laplacian of the human brain’s structural connectivity (SC) plays a dominant role in shaping the pattern of resting-state functional connectivity (FC). The modeling of FC using the graph Laplacian of the brain’s SC is limited, owing to the sparseness of the Laplacian matrix. It is unable to model the negative functional correlations. We extended the graph Laplacian to the hypergraph p-Laplacian in order to describe better the nonlinear and high-order relations between SC and FC. First we estimated those possible links showing negative correlations between the brain areas shared across subjects by statistical analysis. Then we presented a hypergraph p-Laplacian model by embedding the two matrices referring to the sign of the correlations between the brain areas relying on the brain structural connectome. We tested the model on two experimental connectome datasets and evaluated the predicted FC by estimating its Pearson correlation with the empirical FC matrices. The results showed that the proposed diffusion model based on hypergraph p-Laplacian can predict functional correlations more accurately than the models using graph Laplacian as well as hypergraph Laplacian.

Parcellation-induced variation of empirical and simulated brain connectomes at group and subject levels.

Recent developments of whole-brain models have demonstrated their potential when investigating resting-state brain activity. However, it has not been systematically investigated how alternating derivations of the empirical structural and functional connectivity, serving as the model input, from MRI data influence modeling results. Here, we study the influence from one major element: the brain parcellation scheme that reduces the dimensionality of brain networks by grouping thousands of voxels into a few hundred brain regions. We show graph-theoretical statistics derived from the empirical data and modeling results exhibiting a high heterogeneity across parcellations. Furthermore, the network properties of empirical brain connectomes explain the lion’s share of the variance in the modeling results with respect to the parcellation variation. Such a clear-cut relationship is not observed at the subject-resolved level per parcellation. Finally, the graph-theoretical statistics of the simulated connectome correlate with those of the empirical functional connectivity across parcellations. However, this relation is not one-to-one, and its precision can vary between models. Our results imply that network properties of both empirical connectomes can explain the goodness-of-fit of whole-brain models to empirical data at a global group level but not at a single-subject level, which provides further insights into the personalization of whole-brain models.

Virtual Connectomic Datasets in Alzheimer's Disease and Aging Using Whole-Brain Network Dynamics Modelling.

Large neuroimaging datasets, including information about structural connectivity (SC) and functional connectivity (FC), play an increasingly important role in clinical research, where they guide the design of algorithms for automated stratification, diagnosis or prediction. A major obstacle is, however, the problem of missing features [e.g., lack of concurrent DTI SC and resting-state functional magnetic resonance imaging (rsfMRI) FC measurements for many of the subjects]. We propose here to address the missing connectivity features problem by introducing strategies based on computational whole-brain network modeling. Using two datasets, the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset and a healthy aging dataset, for proof-of-concept, we demonstrate the feasibility of virtual data completion (i.e., inferring "virtual FC" from empirical SC or "virtual SC" from empirical FC), by using self-consistent simulations of linear and nonlinear brain network models. Furthermore, by performing machine learning classification (to separate age classes or control from patient subjects), we show that algorithms trained on virtual connectomes achieve discrimination performance comparable to when trained on actual empirical data; similarly, algorithms trained on virtual connectomes can be used to successfully classify novel empirical connectomes. Completion algorithms can be combined and reiterated to generate realistic surrogate connectivity matrices in arbitrarily large number, opening the way to the generation of virtual connectomic datasets with network connectivity information comparable to the one of the original data.

Accurately modeling the human brain functional correlations with hypergraph Laplacian

The relationship between the structural connectivity (SC) of human brain networks and their functional connectivity (FC) is of immense importance in understanding brain cognition and disorders and has gained significant attention in the neuroscience over the past decade. However, the underlying mechanism of how SC gives rise to the whole pattern of FC especially the negative correlations still remains poorly understood. In this paper, we propose a model that can accurately simulate the resting-state human brain functional correlations based on hypergraph Laplacian, including the negative correlations. We firstly assume that, for each brain region, there are some links showing positive correlations and some other links showing negative correlations. Then we derive a hypergraph model with the two matrices indicating positive and negative correlations respectively using the SC of human brain network. We apply the model to two empirical datasets and the results show that the simulated FC can achieve higher Pearson correlations with the empirical FC, outperforming the state-of-art graph diffusion (GD) model.

Modeling brain dynamics after tumor resection using The Virtual Brain

Brain tumor patients scheduled for tumor resection often face significant uncertainty, as the outcome of neurosurgery is difficult to predict at the individual patient level. Recently, simulation of the activity of neural populations connected according to the white matter fibers, producing personalized brain network models, has been introduced as a promising tool for this purpose. The Virtual Brain provides a robust open source framework to implement these models. However, brain network models first have to be validated, before they can be used to predict brain dynamics. In prior work, we optimized individual brain network model parameters to maximize the fit with empirical brain activity. In this study, we extend this line of research by examining the stability of fitted parameters before and after tumor resection, and compare it with baseline parameter variability using data from healthy control subjects. Based on these findings, we perform the first “virtual neurosurgery”, mimicking patient’s actual surgery by removing white matter fibers in the resection mask and simulating again neural activity on this new connectome.We find that brain network model parameters are relatively stable over time in brain tumor patients who underwent tumor resection, compared with baseline variability in healthy control subjects. Concerning the virtual neurosurgery analyses, use of the pre-surgery model implemented on the virtually resected structural connectome resulted in improved similarity with post-surgical empirical functional connectivity in some patients, but negligible improvement in others. These findings reveal interesting avenues for increasing interactions between computational neuroscience and neuro-oncology, as well as important limitations that warrant further investigation.

Adaptive frequency-based modeling of whole-brain oscillations: Predicting regional vulnerability and hazardousness rates.

Whole-brain computational modeling based on structural connectivity has shown great promise in successfully simulating fMRI BOLD signals with temporal coactivation patterns that are highly similar to empirical functional connectivity patterns during resting state. Importantly, previous studies have shown that spontaneous fluctuations in coactivation patterns of distributed brain regions have an inherent dynamic nature with regard to the frequency spectrum of intrinsic brain oscillations. In this modeling study, we introduced frequency dynamics into a system of coupled oscillators, where each oscillator represents the local mean-field model of a brain region. We first showed that the collective behavior of interacting oscillators reproduces previously shown features of brain dynamics. Second, we examined the effect of simulated lesions in gray matter by applying an in silico perturbation protocol to the brain model. We present a new approach to map the effects of vulnerability in brain networks and introduce a measure of regional hazardousness based on mapping of the degree of divergence in a feature space.
