Larval feeding activity and use of embryonic resources determine juvenile performance of the common prawn Palaemonserratus

Phenotypic links are the potential for “carryover” of effects of experience during one life history stage into performance and selection at subsequent stages. They reflect plastic responses to the environment experienced during an early phase on the phenotype of subsequent phases. We are studying these effects by following individuals of the shrimp Palaemon serratus from the embryonic (eggs carried by females) through the larval phase (pelagic) to the juvenile phase (benthic). In experiment 1, we investigated the effects of larval prey concentration (10, 4 and 2 Artemia/mL) and larval incubation temperature (16 and 22 °C) on larval performance (metamorphosis rate, developmental duration and growth) and then on juvenile performance (survival and Specific Growth Rate, SGR, at 18 and 24 °C in 14 days). In experiment 2, we investigated the effects of embryonic incubation temperature (larval biomass and lipid content of newly hatched larvae from embryos incubated at 12 and 18 °C) and larval prey concentration on larval performance and then on juvenile performance. In both experiments, the larvae plastically increased their development time in response to the reduction in temperature and prey concentration, whereas their survival decreased with temperature and prey concentration. The quantity of lipids available at hatching decreased with decreasing embryonic incubation temperature, which reduced the larval performance, particularly with a low concentration of prey. Survival at 14 days post-metamorphosis was significantly reduced when the embryos were incubated at 12 °C compared with those incubated at 18 °C, regardless of the subsequent larval incubation conditions, revealing phenotypic links between overconsumption of embryonic yolk reserves and post-metamorphic fitness. Overall, juveniles had a better SGR at 24 than at 18 °C, and even better when incubated under stressful embryo-larval conditions (temperature and prey concentration). This study highlighted phenotypic links between developmental stages and over developmental periods of several months.

The carryover effects of embryonic incubation temperature on subsequent growth and thermal tolerance in white sturgeon

Environmental variation experienced during early periods of development can lead to persistent phenotypic alteration, known as carryover effects. Such effects increase concern for threatened or endangered species such as the white sturgeon (Acipenser transmontanus), particularly considering expected thermal changes due to climate change. We evaluated how temperature during embryonic development affects physiological parameters such as larval and early juvenile growth and thermal tolerance. Nechako River white sturgeon embryos were incubated at different environmental temperatures (Te) of 12 °C (the natural spawning temperature of this population), 15 °C (the hatchery incubation temperature), and 18 °C (representing potential increases in river temperatures given global climate change). After hatch, fish were reared at a common 15 °C for 80 days post-hatch (dph). Individuals from each temperature treatment were tested for thermal tolerance using the critical thermal maximum method (CTmax), euthanized, and measured. Fish were examined at regular intervals from 13 to 80 dph, which bridged the time from the start of exogenous feeding through the transition into early juveniles. We found carryover effects of high embryonic Te in the short term for both thermal tolerance and growth. Fish that developed at 18 °C had the lowest thermal tolerance during the start of exogenous feeding. However, differences in thermal tolerance were small for early juveniles and were unlikely to be ecologically relevant in the longer term. Fish that developed at 18 °C were smallest over the observation period, indicating a possible cost for survival from increasing environmental temperatures during embryonic development. This research represents a window into a critical period of development during which fish are particularly vulnerable to climatic variation, and shows that cooler temperatures (12 °C) during incubation are optimal for this population. The results can inform environmental managers on the best strategies to help conserve current white sturgeon populations across their range.

Immediate and Persistent Effects of Temperature on Oxygen Consumption and Thermal Tolerance in Embryos and Larvae of the Baja California Chorus Frog, Pseudacris hypochondriaca.

The developmental environment has significant immediate effects on phenotypes, but it may also persistently or permanently shape phenotypes across life history. This study examined how developmental temperature influenced embryonic and larval phenotypes of Baja California chorus frog (Pseudacris hypochondriaca), an abundant amphibian in southern California and northern Baja California. We collected egg clutches from native ponds in northern San Diego County within 24 h of fertilization, and clutches were separated and distributed between constant temperatures of 10, 15, 20, and 25°C for incubation. Oxygen consumption rate (O2), developmental stage, and embryo and yolk masses were measured throughout development. Time to 50% hatch, survival at 50% hatch, and hatch duration were determined. Development rate was strongly affected by temperature, with warmer temperatures reducing time to hatch and hatch duration. Survival to hatch was high across all temperatures, >90%. Mass-specific O2 of embryos either remained constant or increased throughout development, and by hatching energy demand was significantly increased at higher temperatures. There were limited temperature effects on growth, with embryo and yolk dry mass similar between temperatures throughout embryonic development. To examine long-term effects of embryonic temperature, we reared hatchlings from each temperature until onset of larval feeding. Once feeding, larvae were acclimated to 20 or 25°C (>2 weeks). Following acclimation to 20 or 25°C, we measured larval mass-specific O2 and critical thermal maximum (CTMax) at a common developmental stage (Gosner stages 32–36, “hindlimb toe differentiation”). Embryonic temperature had persistent effects on larval O2 and CTMax, with warmer temperatures generally resulting in similar or higher O2 and CTMax. This partially supported a “warmer is better” effect of embryonic incubation temperature. These results suggest that in a thermally robust amphibian species, temperature may program the phenotype during early development to construct traits in thermal tolerance and energy use that may persist. Overall, P. hypochondriaca displays a thermally robust phenotype, and it is possible that amphibians that possess a wider range of phenotypic plasticity will be relatively more successful mitigating effects of climate change.

Small noncoding RNA profiles along alternative developmental trajectories in an annual killifish

Embryonic development of Austrofundulus limnaeus can occur along two phenotypic trajectories that are physiologically and biochemically distinct. Phenotype appears to be influenced by maternal provisioning based on the observation that young females produce predominately non-diapausing embryos and older females produce mostly diapausing embryos. Embryonic incubation temperature can override this pattern and alter trajectory. We hypothesized that temperature-induced phenotypic plasticity may be regulated by post-transcriptional modification via noncoding RNAs. As a first step to exploring this possibility, RNA-seq was used to generate transcriptomic profiles of small noncoding RNAs in embryos developing along the two alternative trajectories. We find distinct profiles of mature sequences belonging to the miR-10 family expressed in increasing abundance during development and mature sequences of miR-430 that follow the opposite pattern. Furthermore, miR-430 sequences are enriched in escape trajectory embryos. MiR-430 family members are known to target maternally provisioned mRNAs in zebrafish and may operate similarly in A. limnaeus in the context of normal development, and also by targeting trajectory-specific mRNAs. This expression pattern and function for miR-430 presents a potentially novel model for maternal-embryonic conflict in gene regulation that provides the embryo the ability to override maternal programming in the face of altered environmental conditions.

Daily, repeating fluctuations in embryonic incubation temperature alter metabolism and growth of Lake whitefish (Coregonus clupeaformis)

Lake whitefish (Coregonus clupeaformis) utilize overwintering embryonic development (up to 180 days), and such stenothermic, cold-water embryos may be particularly susceptible to thermal shifts. We incubated whitefish embryos in temperature treatments that were constant temperature (2.0 ± 0.1 °C, 5.0 ± 0.1 °C, and 8.0 ± 0.1 °C; mean ± SD) or variable temperature (VT, mean = 5.0 ± 0.3 °C). In the VT, a daily 2 °C temperature change followed a continuous pattern throughout development: 2-4-6-8-6-4-2 °C. Hatchling survival proportion from fertilization to hatch was significantly impacted by incubation temperature (P < 0.001): 2 °C (0.88 ± 0.01) and 5 °C (0.91 ± 0.01) showed higher survival than both the VT (0.83 ± 0.02) and 8 °C groups (0.15 ± 0.06), which were statistically distinct from each other. Time to hatch (dpf) was significantly different across all treatments (P < 0.001): 8 °C (68 ± 2 dpf), VT (111 ± 4 dpf), 5 °C (116 ± 4 dpf), 2 °C (170 ± 3 dpf). Likewise, hatchling yolk-free dry mass (mg) and total body length (mm) were significantly different across all treatments (P < 0.001): 8 °C (0.66 ± 0.08 mg; 11.1 ± 0.08 mm), VT (0.97 ± 0.06 mg; 11.7 ± 0.05 mm), 5 °C (1.07 ± 0.03 mg; 12.0 ± 0.02 mm), 2 °C (1.36 ± 0.04 mg; 12.8 ± 0.05 mm). Oxygen consumption rate (V̇o2) was significantly affected by the interaction between treatment and measurement temperature (P < 0.001). Hatchling VT whitefish showed mean V̇o2 that was higher compared to the 2 °C group measured at 2 °C, and lower compared to the 2 °C and 5 °C group measured at 8 °C. This study demonstrates that the VT incubation treatment produced fewer (increased mortality), smaller embryos that hatched earlier than 2 °C and 5 °C embryos. The plasticity of V̇o2 for this stenothermic-incubating fish species under variable incubation conditions reveals a metabolic cost to cycling thermal incubation conditions.

MiRNAs regulate acute transcriptional changes in broiler embryos in response to modification of incubation temperature

MicroRNAs are post-transcriptional regulators that play critical roles in diverse biological processes. We hypothesize that miRNAs may be involved in regulating transcriptome responses to changes in embryonic incubation temperature in chickens affecting differentiation and proliferation processes during tissue development. Therefore, we conducted comparative transcriptome profiling of miRNAs to examine altered expression in breast and hind muscle of embryos and day 35 chickens experiencing high (38.8 °C), control (37.8 °C), or low (36.8 °C) embryonic incubation temperature during embryonic day (ED) 7–10 or ED10–13. The results revealed differential expression of miRNAs due to modification of embryonic incubation temperature in a muscle type-specific and a developmental stage-specific manner. The immediate effects of thermal change observed in embryos were substantial compared to the subtle long-term effects in chickens at day 35 post-hatch. Upregulation of miR-133 in breast muscle and downregulation of miR-199a-5p, miR-1915, and miR-638 in hind muscle post ED7–10 high-temperature treatment are functionally associated with myogenesis and body size. ED10–13 low-temperature treatment led to downregulation of let-7, miR-93, and miR-130c that are related to proliferation and differentiation. The results provide insight into the dynamics of miRNA expression at variable embryonic incubation temperatures during developmental processes and indicate a major regulatory role of miRNAs in acute responses to modified environmental conditions that affect remodelling of cells and tissues.

Low incubation temperature during early development negatively affects survival and related innate immune processes in zebrafish larvae exposed to lipopolysaccharide

In many fish species, the immune system is significantly constrained by water temperature. In spite of its critical importance in protecting the host against pathogens, little is known about the influence of embryonic incubation temperature on the innate immunity of fish larvae. Zebrafish (Danio rerio) embryos were incubated at 24, 28 or 32 °C until first feeding. Larvae originating from each of these three temperature regimes were further distributed into three challenge temperatures and exposed to lipopolysaccharide (LPS) in a full factorial design (3 incubation × 3 challenge temperatures). At 24 h post LPS challenge, mortality of larvae incubated at 24 °C was 1.2 to 2.6-fold higher than those kept at 28 or 32 °C, regardless of the challenge temperature. LPS challenge at 24 °C stimulated similar immune-related processes but at different levels in larvae incubated at 24 or 32 °C, concomitantly with the down-regulation of some chemokine and lysozyme transcripts in the former group. Larvae incubated at 24 °C and LPS-challenged at 32 °C exhibited a limited immune response with up-regulation of hypoxia and oxidative stress processes. Annexin A2a, S100 calcium binding protein A10b and lymphocyte antigen-6, epidermis were identified as promising candidates for LPS recognition and signal transduction.

Temperature during early development has long-term effects on microRNA expression in Atlantic cod.

BackgroundEnvironmental temperature has serious implications in life cycle of aquatic ectotherms. Understanding the molecular mechanisms of temperature acclimation and adaptation of marine organisms is of the uttermost importance for ecology, fisheries, and aquaculture, as it allows modeling the effects of global warming on population dynamics. Regulatory molecules are major modulators of acclimation and adaptation; among them, microRNAs (miRNAs) are versatile and substantial contributors to regulatory networks of development and adaptive plasticity. However, their role in thermal plasticity is poorly known. We have asked whether the temperature and its shift during the early ontogeny (embryonic and larval development) affect the miRNA repertoire of Atlantic cod (Gadus morhua), and if thermal experience has long-term consequences in the miRNA profile.ResultsWe characterized miRNA during different developmental stages and in juvenile tissues using next generation sequencing. We identified 389 putative miRNA precursor loci, 120 novel precursor miRNAs, and 281 mature miRNAs. Some miRNAs showed stage- or tissue-enriched expression and miRNAs, such as the miR-17 ~ 92 cluster, myomiRs (miR-206), neuromiRs (miR-9, miR-124), miR-130b, and miR-430 showed differential expression in different temperature regimes. Long-term effect of embryonic incubation temperature was revealed on expression of some miRNAs in juvenile pituitary (miR-449), gonad (miR-27c, miR-30c, and miR-200a), and liver (let-7 h, miR-7a, miR-22, miR-34c, miR-132a, miR-192, miR-221, miR-451, miR-2188, and miR-7550), but not in brain. Some of differentially expressed miRNAs in the liver were confirmed using LNA-based rt-qPCR. The effect of temperature on methylation status of selected miRNA promoter regions was mostly inconclusive.ConclusionsTemperature elevation by several degrees during embryonic and larval developmental stages significantly alters the miRNA profile, both short-term and long-term. Novel evolutionary pathways of sex‐determining mechanisms

Evolutionary transitions between sex-determining mechanisms (SDMs) are an enigma. Among vertebrates, individual sex (male or female) is primarily determined by either genes (genotypic sex determination, GSD) or embryonic incubation temperature (temperature-dependent sex determination, TSD), and these mechanisms have undergone repeated evolutionary transitions. Despite this evolutionary lability, transitions from GSD (i.e. from male heterogamety, XX/XY, or female heterogamety, ZZ/ZW) to TSD are an evolutionary conundrum, as they appear to require crossing a fitness valley arising from the production of genotypes with reduced viability owing to being homogametic for degenerated sex chromosomes (YY or WW individuals). Moreover, it is unclear whether alternative (e.g. mixed) forms of sex determination can persist across evolutionary time. It has previously been suggested that transitions would be easy if temperature-dependent sex reversal (e.g. XX male or XY female) was asymmetrical, occurring only in the homogametic sex. However, only recently has a mechanistic model of sex determination emerged that may allow such asymmetrical sex reversal. We demonstrate that selection for TSD in a realistic sex-determining system can readily drive evolutionary transitions from GSD to TSD that do not require the production of YY or WW individuals. In XX/XY systems, sex reversal (female to male) occurs in a portion of the XX individuals only, leading to the loss of the Y allele (or chromosome) from the population as XX individuals mate with each other. The outcome is a population of XX individuals whose sex is determined by incubation temperature (TSD). Moreover, our model reveals a novel evolutionarily stable state representing a mixed-mechanism system that has not been revealed by previous approaches. This study solves two long-standing puzzles of the evolution of sex-determining mechanisms by illuminating the evolutionary pathways and endpoints.

Incubation Conditions Are More Important in Determining Early Thermoregulatory Ability than Posthatch Resource Conditions in a Precocial Bird

Recent research in birds suggests that investing in incubation is one mechanism by which parents can enhance the phenotype of their offspring. Posthatch environmental conditions can also shape an individual's phenotype, and it is thus possible for pre- and posthatch conditions to have interactive effects on an individual's phenotype. In this study, we examined the individual and interactive effects of prehatch incubation temperature and posthatch food availability on growth, food consumption, and thermoregulatory ability in wood duck (Aix sponsa) ducklings. Eggs were incubated at one of three temperatures (35.0°, 35.9°, or 37.0°C), and then ducklings were reared on an either ad lib. or time-restricted diet for 12 d after hatching. We found that food availability influenced duckling growth, with the slowest growth occurring in ducklings fed the restricted diet. Incubation temperature also interacted with food conditions to influence duckling growth: ducklings fed ad lib. from the lowest incubation temperature grew slower than ducklings fed ad lib. from the higher incubation temperatures. Most importantly, we found that the improvement in a duckling's ability to maintain body temperature in the face of a thermal challenge was influenced by embryonic incubation temperature but not feeding conditions. Ducklings from the highest incubation temperature experienced the greatest improvement in thermoregulatory performance with age. Our findings suggest that the prehatch environment is more important than posthatch resource conditions in determining some physiological functions and underscores the important role that incubation temperature plays in determining offspring phenotype in birds.

Larval development rate of the mosquitoes Culex quinquefasciatus and Aedes aegypti (Diptera: Culicidae) varies between clutches: implications for population ecology

AbstractEgg clutches from two mosquito species were incubated under controlled conditions and the larval development time was recorded. Both the egg raft‐laying mosquito Culex quinquefasciatus and the individual egg layer Aedes aegypti displayed significant intraspecific variation between clutches when it came to larval development at the same temperature. Furthermore, embryonic incubation temperature was found to influence subsequent larval growth rate in some cases. These hitherto undocumented phenomena are indicative of development rate plasticity that appears to be conserved across mosquito genera. The derived development rate data were used to calculate the minimum number of egg clutches required to give a representative mean day‐degrees value for a population of each species. These values were six and five clutches, respectively. A simple population simulation was given to illustrate the impact of different larval development rates when overlapping generations of problem mosquitoes breed. The implication of this work for mosquito population modelling is that the extent of developmental rate variation needs to be taken into account. Furthermore, such conserved variation in larval development rate illustrates the extent of selective pressures on this life history trait.

Effects of acclimation and egg-incubation temperature on selected temperature by hatchling western painted turtles ( Chrysemys picta bellii)

The ability of hatchling turtles to detect environmental temperature differences and to effectively select preferred temperature is a function that critically impacts survival. In some turtle species, temperature preference may be influenced by embryonic and post-hatching conditions, such as egg-incubation and acclimation temperature. We tested for effects of embryonic incubation temperature (27.5 °C, 30 °C) and acclimation temperature (20 °C, 25 °C) on the selected temperature and movement patterns of 32 Chrysemys picta bellii (Reptilia: Emydidae) hatchlings in an aquatic thermal gradient of 14–34 °C and in single-temperature (20 °C, 25 °C) control tests. Among 10–11 month old hatchlings, acclimation temperature and egg-incubation temperature influenced temperature selection and movement patterns. Acclimation temperature affected activity and movement: in thermal gradient and single-temperature control tests, 25 °C-acclimated turtles relocated between chambers significantly more frequently than individuals acclimated to 20 °C. Acclimation temperature also affected temperature selection: 20 °C-acclimated turtles selected a specific temperature during gradient tests, but 25 °C-acclimated turtles did not. Among 20 °C-acclimated turtles, egg-incubation temperature was inversely related to selected temperature: hatchling turtles incubated at 27.5 °C selected the warmest temperature available (34 °C); individuals incubated at 30 °C selected the coldest temperature (14 °C). These results suggest that interactions of environmental conditions may influence post-hatching thermoregulatory behavior in C. picta bellii, a factor that ultimately affects fitness.

Influence of Egg Shell Embryonic Incubation Temperature and Broiler Breeder Flock Age on Posthatch Growth Performance and Carcass Characteristics

A study was conducted to examine the posthatch growth performance of high-yielding broilers when eggs were incubated at 3 different embryo temperatures from 2 flocks of breeders at different ages (different egg size). Two thousand, four hundred eggs from 2 broiler breeder flocks (29 and 57 wk of age) of the same high-yielding strain (Cobb x Cobb) were incubated in the same incubator for 16 d at 37.5 degrees C. Following candling, the eggs from the 2 flocks were transferred into 3 hatcher cabinets at starting temperatures of 36.5 degrees C (low, L), 37.6 degrees C (middle, M), and 38.7 degrees C (high, H) and adjusted to achieve a shell temperature of 37.5 degrees C (L), 38.6 degrees C (M), and 39.7 degrees C (H) using an infrared thermometer. All chicks were taken off at 21 d of incubation, randomized into floor pens, and reared for 44 d. Body weights, feed intake, and feed conversion were determined at 21, 35, and 44 d of age. Body weight of birds from the H treatment was significantly less at 21, 35, and 44 d compared with the M birds. Birds in the L group weighed significantly less at 35 and 44 d compared with the M birds. Progeny from the older breeder flock had significantly greater BW at 1, 21, and 35 d of age, but had only numerically greater BW at 44 d when compared with birds from the younger flock. Feed conversion for the H birds was significantly higher from 0 to 21 d of age compared with the M and L birds. Broilers from the 29-wk-old breeder flock had lower cumulative feed conversion values than the birds from the 57-wk-old flock. No significant differences in mortality were observed. Posthatch performance appears to be affected by hatcher environment as determined by embryo shell temperature.

Changes in zoeae of the snow crab, Chionoecetes opilio, with variation in incubation temperature

The purpose of this study was to evaluate the effect of embryonic incubation temperature on the morphology, energetic content, and individual weights of C. opilio zoeae. Ovigerous female C. opilio were collected by trawl from the eastern Bering Sea in the summer of 2002 and were held in the laboratory at five temperature treatments (− 1, 0, 1, 3, and 6 °C) from embryo stage five to hatching. The mean duration of incubation ranged from 240 d at 6 °C to 353 d at − 1 °C. No relationship between larval weight and caloric content with incubation temperature was detected, indicating that increased development time at cooler temperatures may not have an energetic cost. However, several morphological features varied with temperature. Most larval characters including rostro-dorsal length, rostral spine length, lateral spine length, and carapace width were smaller at the warmest incubation temperature. Protopodite length did not differ with temperature. The length of the 3rd abdominal somite consistently increased with increased temperature and may serve as a suitable indicator that could be used in field populations to infer temperature of incubation. Variation in morphology with temperature could confound identification of C. opilio zoeae from those of a congener, Chionoecetes bairdi because identification of C. bairdi is based on the length of the 3rd abdominal somite.

Embryonic temperature shapes behavioural change following social experience in male leopard geckos, Eublepharis macularius

Individual variation in behavioural expression and behavioural plasticity exist in all species, and early experiences are critical determinants of both. The leopard gecko is a lizard with temperature-dependent sex determination, and in this species, embryonic incubation temperature (IncT) affects the display of social behaviours. For example, adult males hatched from eggs incubated at an IncT that produces predominantly males (male-biased IncT, 32.5°C) are more aggressive and less sexually active than males hatched from eggs at an IncT that produces predominantly females (female-biased IncT, 30°C). It is not known, however, whether IncT influences behavioural plasticity in adulthood. We assessed whether adult males hatched from eggs incubated at female- and male-biased IncTs showed different changes in territorial behaviour (scent marking in an empty test arena), anticipatory behaviours (activity, scent marking and tail vibrations in response to cues that predict the introduction of a female) and courtship behaviour following social interactions with females. We found that heterosexual social experiences increased territorial behaviour and the display of anticipatory behaviours in males from the female-biased IncT but not in males from the male-biased IncT. This difference was not due to differences in the amount of social experiences acquired, and the greater change in anticipatory behaviour in males from the female-biased IncT suggest that they acquired sexual conditioning more readily. Differences in sex steroid concentrations or neural metabolism caused by IncT could underlie this difference. These results highlight the profound implications of maternal nest site selection for offspring phenotype and plasticity in this species.Copyright 2003 The Association for the Study of Animal Behaviour. Phenotypic Plasticity for Growth in the Common Snapping Turtle: Effects of Incubation Temperature, Clutch, and Their Interaction

Abstrcact.-We examined a critical component of the Charnov-Bull hypothesis of temperaturedependent sex determination (TSD) by determining the reaction norms of hatchling growth to embryonic incubation temperature in the common snapping turtle, Chelydra serpentina. Hormone manipulations of eggs produced females at male temperatures and vice versa, which thereby permitted same-sex comparisons of hatchling growth across a range of incubation temperatures. In this way, the normally confounded effects of incubation temperature and sex were dissociated experimentally. The resultant hatchlings, including controls and experimentals, exhibited normal gonadal structure and sex steroid profiles. The subsequent growth of hatchlings monitored for 6 mo was strongly affected by embryonic incubation temperature but not by sex. As predicted, growth was enhanced at incubation temperatures that produced males. Clutch effects and interaction effects (clutch by incubation temperature) on growth were significant. In addition, there was a positive genetic covariance among incubation temperatures, but incubation temperature effects varied among clutches.
