PTP1B regulates neurite extension mediated by cell-cell and cell-matrix adhesion molecules.

N-cadherin and beta1-integrin adhesion and signaling play important roles in growth cone adhesion and guidance. Each of these adhesion receptor systems is composed of multiprotein complexes, and both adhesion and downstream signaling events are regulated through the interaction of protein tyrosine kinases and phosphatases with many of the proteins that make up these complex systems. Work from our laboratory reported that the nonreceptor protein tyrosine phosphatase PTP1B is localized to adherens junctions and focal adhesion complexes and regulates both N-cadherin- and beta1-integrin-mediated adhesion. PTP1B appears to modulate integrin-mediated adhesion through regulation of src activation and cadherin-mediated adhesion through dephosphorylation of beta-catenin. We have continued these studies and report that PTP1B is localized to the tips of growing neurites and that introduction of a noncatalytic mutant of PTP1B into PC12 cells results in inhibition of N-cadherin- and beta1-integrin-mediated neurite outgrowth but is without effect on neurite outgrowth on poly-L-lysine. Moreover, suppressing the level of PTP1B in primary embryonic chick neural retina cells using antisense oligonucleotides also inhibits N-cadherin- and beta1-integrin-mediated neurite outgrowth. Neither of these techniques reduces the levels of expression of either adhesion receptor. We conclude that PTP1B is a regulatory component of the molecular complex required for both N-cadherin and beta1-integrin-mediated axon growth.

Antibodies to the retina N-acetylgalactosaminylphosphotransferase modulate N-cadherin-mediated adhesion and uncouple the N-cadherin transferase complex from the actin-containing cytoskeleton.

Embryonic chick neural retina cells have at their surface an N-Acetylgalactosaminylphosphotransferase (GalNAcPTase) which is associated with, and glycosylates, the calcium-dependent cell-cell adhesion molecule, N-cadherin (Balsamo, J., and J. Lilien. 1990. J. Biol. Chem. 265:2923-2928). In this manuscript, we demonstrate that antibodies directed against the GalNAcPTase, as well as anti-N-cadherin antibodies, are able to inhibit adhesion of chick neural retina cells to a cell monolayer, to immobilized N-cadherin, or to immobilized anti-N-cadherin antibody. These results indicate that anti-GalNAcPTase antibodies modulate the function of N-cadherin, interfering with the formation of N-cadherin-mediated adhesions. We also demonstrate that actin is associated with the N-cadherin/GalNAcPTase complex and that binding of anti-GalNAcPTase antibodies to intact cells results in dissociation of actin from the complex. We suggest that the GalNAcPTase modulates N-cadherin function by altering its interaction with the cytoskeleton.

N-cadherin is stably associated with and is an acceptor for a cell surface N-acetylgalactosaminylphosphotransferase.

Calcium-dependent cell-cell adhesion among embryonic chick neural retina cells is mediated by N-cadherin, a 130,000-Da integral membrane protein. We have reported that the ability of chick neural retina cells to form calcium-dependent cell-cell adhesion is also correlated with the presence of an N-acetylgalactosaminyltransferase at the cell surface (Balsamo, J., and Lilien, J. (1982) J. Biol. Chem. 257, 349-354). This enzyme transfers N-acetylgalactosaminephosphate to endogenous acceptors (Balsamo, J., Pratt, R. S., and Lilien, J. (1986) Biochemistry 25, 5402-5407) and is tightly associated with these acceptors forming a complex which can be isolated from neutral detergent extracts of plasma membranes. We now report that N-cadherin is present in the complex and is an acceptor for the N-acetylgalactosaminylphosphotransferase. Antibodies prepared against N-cadherin precipitate the transferase containing complexes from neutral detergent extracts; however, when the complexes are dissociated by treatment with sodium dodecyl sulfate prior to immunoprecipitation, only N-cadherin is precipitated. Similarly, anti-transferase antibodies immunoprecipitate N-cadherin containing complexes prior to disruption of the complex with sodium dodecyl sulfate. Catalysis of the transferase reaction in the complex results in the transfer of N-acetylgalactosamine phosphate to oligosaccharide chains on N-cadherin. Glycosylation of N-cadherin by the transferase also occurs in situ; labeling of retina cells with [32P] orthophosphate results in N-cadherin molecules containing terminal N-acetylgalactosamine linked to an oligosaccharide chain via a phosphodiester bond.

Expression of calcium-dependent cell adhesion during ocular development: A biochemical, histochemical and functional analysis

Previous studies of the adhesive properties of embryonic chick neural retina cells indicate a gradual decrease in the expression of calcium-dependent adhesions during retinal histogenesis, a function which has been attributed in part to gp130/4.8, a retinal calcium-dependent adhesion-associated cell surface membrane glycoprotein with a molecular weight of approximately 130 kDa and an isoelectric point of 4.8 ( G. B. Grunwald, R. Pratt, and J. Lilien, 1982, J. Cell Sci. 55, 69–83 ). The experiments described here were done to define the relationship of gp130/4.8 to N-cadherin, another calcium-dependent adhesion molecule found in chick retina, which has a reported molecular weight of 127 kDa and which is recognized by monoclonal antibody NCD-2 ( K. Hatta and M. Takeichi, 1986, Nature (London) 320, 447–449 ). Using two-dimensional gel electrophoresis followed by Western blotting as well as quantitative solid-phase immunoassays, polyspecific antisera recognizing gp130/4.8 were compared with monoclonal antibody NCD-2 for reactivity with proteins of retina and other tissues. The data lead us to conclude that retinal calcium-dependent adhesion proteins gp130/4.8 and N-cadherin are likely to be the same molecule. In order to obtain evidence for a direct correlation of changes in expression of these adhesion proteins with changes in retinal cell adhesivity and related morphogenetic events, parallel studies were carried out with cells from various ocular tissues to examine the functional, biochemical, and immunohistochemical expression of N-cadherin during ocular development. Immunohistochemical mapping of N-cadherin in the developing chick eye reveals three modes of N-cadherin expression which occur simultaneously in different ocular tissues: (1) down-regulation, (2) up-regulation, and (3) steady-state expression. These patterns of expression correlate with changes in the adhesive behavior of cells as well as with discrete stages in the morphogenesis of several ocular tissues. The results suggest that N-cadherin is a versatile cell adhesion protein with a role in both the development of several ocular tissues and the maintenance of specialized structures in the mature eye.

Retina cognin does not bind to itself during membrane interaction in vitro

Retina cognin (R-cognin) is an intrinsic membrane protein of vertebrate retinal cells which supports tissue-specific cell adhesion and mediates cell type-specific associations during development. As a first step in understanding how R-cognin mediates specific adhesion of retinal cell membranes, we asked if cognin bound to another cognin molecule or to a different macromolecule, a possible cognin-binding protein. To do this, we constructed an affinity column with retinal cell membrane proteins (enriched for cognin) bound to the matrix. Proteins in a detergent extract of retinal cell membranes were exposed to this matrix and those which bound specifically eluted and identified by immunoelectrophoresis. Most prominent among these was a protein with an apparent mass of 64 kDa. The binding of this material to the column was blocked by cognin antibody. To eliminate possible artifacts of molecular interactions in vitro, we sought independent confirmation that 64 kDa protein actually bound R-cognin. Using a modified retina membrane vesicle system, we asked what proteins could be photoaffinity cross-linked to cognin during vesicle aggregation. Cross-linking produced a 114 kDa complex on gels which could be resolved into a 50 kDa (cognin) and a 64 kDa band under reducing conditions. Identification of a 64 kDa protein by independent techniques suggests that cognin promotes association of embryonic chick neural retina cells by binding to this macromolecule or these molecules. Identification of a second component in the mechanism should allow elucidation of cognin's role in mediating cell-cell interactions in developing neural retina.

Pp60c-src expression in transdifferentiating cultures of embryonic chick neural retina cells.

Chick embryo neural retinal cells transdifferentiate extensively into lens cells when cultured in Eagle's MEM containing horse and fetal calf sera (FHMEM). Such cultures express elevated levels of pp60c-src-associated tyrosine kinase activity relative to parallel cultures prevented from transdifferentiating by the addition of supplementary glucose (FHGMEM) or replacement of MEM by medium 199 (F199). Northern blotting and in vitro translation studies suggest that c-src mRNA levels are only slightly higher in late transdifferentiating (FHMEM) cultures as compared to parallel blocked (FHGMEM or F199) cultures. By immunocytochemical staining, we show that pp60c-src protein is largely localized in cell groups undergoing conversion into lens (i.e. expressing delta crystallin) in late FHMEM cultures. Initial studies of pp60c-src in chick lens tissues during development indicate that higher kinase activity is found in the epithelial cells relative to mature lens fibres. Thus pp60c-src may be expressed both during the differentiation of lens cells in vivo and during the transdifferentiation of neural retina cells into lens in vitro.

Production of monoclonal antibodies selective for aggregation-competent chick neural retina cells: An immunosuppressive approach

We present here a rapid means of preparing mouse monoclonal antibodies which bind selectively to aggregation-competent, embryonic chick neural retina cells. The approach couples two existing technologies: well-defined procedures for manipulating the adhesive properties of embryonic retinal cells through control of the dissociation conditions, and an immunosuppressive technique designed to reduce the immune response against unwanted ‘background’ antigens. This approach greatly increases the likelihood of generating monoclonal antibodies directed against cell surface molecules mediating intracellular adhesion in this tissue. Fab prepared from the combined supernates of all IgG-producing clones obtained in this manner inhibited retinal Ca 2+-dependent adhesion, suggesting that at least one clone in the group was reactive with elements of the Ca 2+-dependent adhesion mechanism.

A chick neural retina adhesion and survival molecule is a retinol-binding protein.

A 20,000-D protein called purpurin has recently been isolated from the growth-conditioned medium of cultured embryonic chick neural retina cells (Schubert, D., and M. LaCorbiere, 1985, J. Cell Biol., 101:1071-1077). Purpurin is a constituent of adherons and promotes cell-adheron adhesion by interacting with a cell surface heparan sulfate proteoglycan. It also prolongs the survival of cultured neural retina cells. This paper shows that purpurin is a secretory protein that has sequence homology with a human protein synthesized in the liver that transports retinol in the blood, the serum retinol-binding protein (RBP). Purpurin binds [3H]retinol, and both purpurin and chick serum RBP stimulate the adhesion of neural retina cells, although the serum protein is less active than purpurin. Purpurin and the serum RBP are, however, different molecules, for the serum protein is approximately 3,000 D larger than purpurin and has different silver-staining characteristics. Finally, purpurin supports the survival of dissociated ciliary ganglion cells, indicating that RBPs can act as ciliary neurotrophic factors.

Gangliosides support neural retina cell adhesion.

Cell surface carbohydrates and complementary carbohydrate receptors may mediate cell-cell recognition during neuronal development. To model such interactions, we developed a technique to test the ability of cell surface lipids (particularly glycosphingolipids) to mediate specific cell recognition and adhesion (Blackburn, C.C., and Schnaar, R.L. (1983) J. Biol. Chem. 258, 1180-1188). When cells were incubated on plastic microwells adsorbed with various glycolipids, carbohydrate-specific cell adhesion was readily detected. We report here the use of this method to study adhesion of embryonic chick neural retina cells to purified cell surface lipids. Rapid and specific cell adhesion was observed when the neural retina cells were incubated on surfaces adsorbed with gangliosides (an important class of neuronal cell surface glycoconjugates) but not on surfaces adsorbed with various neutral glycosphingolipids, phospholipids, or sulfatide. This suggests that the observed cell adhesion was specific for the carbohydrate moiety of the adsorbed ganglioside and was not due to nonspecific ionic or hydrophobic interactions. Although the surface density of adsorbed lipid required to support cell adhesion was the same for all gangliosides examined, the extent of adhesion varied when different purified gangliosides were used. Ganglioside-specific adhesion was not dependent on the presence of calcium (at 37 degrees C) and was attenuated by pretreatment of the cells with trypsin. The extent of ganglioside-directed neural retinal cell adhesion varied with embryonic age. These results imply that gangliosides may play a role in cell-cell recognition in the developing nervous system.

Identification of the chick neural retina cell surface N-acetylgalactosaminyltransferase using monoclonal antibodies.

Intact embryonic chick neural retina cells have at their surface an N-acetylgalactosaminyltransferase which catalyzes the incorporation of N-acetylgalactosamine from UDP-N-acetylgalactosamine into endogenous macromolecular acceptors. The enzyme along with its endogenous acceptors can be isolated as a particulate complex following treatment of membrane-enriched fractions with Triton X-100. In this paper we report on two separate fusions generating monoclonal antibodies: one using as immunogen the particulate complex and the second using as immunogen a soluble N-acetylgalactosaminyltransferase found in tissue-culture-conditioned medium which lacks endogenous acceptor activity. Antibodies from both fusions recognize an antigen which is tightly associated with the particulate transferase/acceptor complex and a soluble antigen having N-acetylgalactosaminyltransferase activity toward exogenously added acceptors. The antibodies recognize a component of ca Mr 220,000, which shows N-acetylgalactosaminyltransferase activity after SDS-gel electrophoresis and transfer to nitrocellulose. This component comigrates on two-dimensional gel electrophoresis with an iodinatable cell surface component whose presence at the cell surface correlates with endogenous transferase activity. We conclude that the antibodies recognize the transferase enzyme itself. Immunohistochemical analysis shows that the enzyme is initially localized throughout the embryonic neural retina in a pattern indicative of a cell surface disposition but becomes restricted to the outer plexiform layer and to outer segments in the adult.

Cell-substratum adhesion in chick neural retina depends upon protein-heparan sulfate interactions.

Embryonic chick neural retina cells in culture release complexes of proteins and glycosaminoglycans, termed adherons, which stimulate cell-substratum adhesion when adsorbed to nonadhesive surfaces. Two distinct retinal cell surface macromolecules, a 170,000-mol-wt glycoprotein and a heparan sulfate proteoglycan; are components of adherons that can independently promote adhesion when coated on inert surfaces. The 170,000-mol-wt polypeptide contains a heparin-binding domain, as indicated by its retention on heparin-agarose columns and its ability to bind [3H]heparin in solution. The attachment of embryonic chick retinal cells to the 170,000-mol-wt protein also depends upon interactions between the protein and the heparan sulfate proteoglycan, since heparan sulfate in solution disrupts adhesion of chick neural retina cells to glass surfaces coated with the 170,000-mol-wt protein. This adhesion is not impaired by chondroitin sulfate or hyaluronic acid, which indicates that inhibition by heparan sulfate is specific. Polyclonal antisera directed against the cell surface heparan sulfate proteoglycan also inhibit attachment of retinal cells to the 170,000-mol-wt protein, which suggests that cell-adheron binding is mediated in part by interactions between cell surface heparan sulfate proteoglycan and 170,000-mol-wt protein contained in the adheron particles. Previous studies have indicated that this type of cell-substratum adhesion is tissue-specific since retina cells do not attach to muscle adherons. Schubert D., M. LaCorbiere, F. G. Klier, and C. Isolation of a cell-surface receptor for chick neural retina adherons.

Embryonic chick neural retina cells release glycoprotein complexes, termed adherons, into their culture medium. When absorbed onto the surface of petri dishes, neural retina adherons increase the initial rate of neural retina cell adhesion. In solution they increase the rate of cell-cell aggregation. Cell-cell and adheron-cell adhesions of cultured retina cells are selectively inhibited by heparan-sulfate glycosaminoglycan, but not by chondroitin sulfate or hyaluronic acid, suggesting that a heparan-sulfate proteoglycan may be involved in the adhesion process. We isolated a heparan-sulfate proteoglycan from the growth-conditioned medium of neural retina cells, and prepared an antiserum against it. Monovalent Fab' fragments of these antibodies completely inhibited cell-adheron adhesion, and partially blocked spontaneous cell-cell aggregation. An antigenically and structurally similar heparan-sulfate proteoglycan was isolated from the cell surface. This proteoglycan bound directly to adherons, and when absorbed to plastic, stimulated cell-substratum adhesion. These data suggest that a heparan-sulfate proteoglycan on the surface of chick neural retina cells acted as a receptor for adhesion-mediating glycoprotein complexes (adherons).

Stress Protein and Crystallin Synthesis During Heat Shock and Transdifferentiation of Embryonic Chick Neural Retina Cells: (chicken/retina/transdifferentiation/heat shock).

Embryonic chick neural retina responds to heat shock by the synthesis of "stress" polypeptides with molecular weights of 85 and 70 kd. Both stress proteins are synthesised from newly-transcribed messenger RNA. Sodium arsenite induces an additional stress protein of MW 25 kd. The heat shock response does not change during culture and subsequent transdifferentiation, and crystallin synthesis is not coinducible with the heat-shock proteins. We have also examined the pattern of protein synthesis at various stages of culture in both monolayer and aggregate systems; although changes in the protein synthetic profine are evident, there is no stress protein induction above basal levels at any time. Whilst mammalian α crystallin (B2 chain) exhibits considerable homology to four small Drosophila heat-shock proteins, no significant antigenic similarity is apparent between δ crystallin and the major avian heat shock proteins. Thus during transdifferentiation, (a) the crystallin proteins do not behave in a manner analogous to stress proteins; moreover (b) crystallin production is not mediated by stress proteins resulting from a culture-induced stress response.

Expression of Toxin Receptors on Cell Surfaces in Transdifferentiating Cultures of Neural Retina: (chick embryo neural retina/transdifferentiation/neuronal cell surface toxin receptors/δ crystallin/double fluorescent labelling).

It has often been asked which of the cell types found during the early stages of culturing embryonic chick neural retina can undergo transdifferentiation into lens in vitro. Since neuronal cell-surface toxin receptors are maintained in NR cultures for much longer than internal neuronal enzymes (e.g. choline acetyltransferase), and since the transdifferentiation process can be greatly accelerated by preparing reaggregates of neural retina cells after about 10 days of preculture as "monolayers", a direct test of this question became feasible. 7 or 9 day embryonic chick neural retina cells, precultured for 10-12 days as monolayers, were dissociated and reaggregated under continuous gyration. Reaggregates were maintained for 8 days in the presence of either tetanus toxin or FITC-conjugated α-bungarotoxin, to permit surface-bound toxins to become internalised via receptor turnover. The reaggregates were then dissociated, stained with rabbit antitoxin and FITC-conjugated anti-antibody in the case of tetanus toxin-labelled material, and restained with a rat or mouse antibody against chick δ crystallin followed by the appropriate rhodamine-conjugated anti-antibody. Although both FITC/toxin-labelled cells (putative neurones) and rhodamine/δ crystallin-labelled cells (transdifferentiated lens cells) were abundant, no examples of double-labelled cells were observed with 9 day starting material, and only a very few with 7 day starting material. We conclude that the vast majority of differentiated neuronal cells expressing surface receptors for these toxins do not transdifferentiate directly into lens cells.

A role for adherons in neural retina cell adhesion.

Embryonic chick neural retina cells release glycoprotein complexes, termed adherons, into their culture medium. When absorbed onto the surface of petri dishes, neural retina adherons increase the initial rate of neural retina cell adhesion; they also stimulate the rate of cell-cell aggregation. Adheron-stimulated adhesion is tissue specific, and the spontaneous aggregation of neural retina cells is inhibited by monovalent Fab' fragments prepared from an antiserum against neural retina adherons. Therefore cell surface antigenic determinants shared with adherons are involved in normal cell-cell adhesions. The particles from the heterogeneous neural retina population contain many proteins and several glycosaminoglycans. The adherons migrate as a symmetrical 12S peak on sucrose gradients and are predominantly 15-nm spheres when examined by electron microscopy. Finally, the specific activity of neural retina adherons increases from embryonic days 7 through 12 and then declines. These results suggest that glycoprotein particles may be involved in some of the adhesive interactions between neural retina cells and between the cells and their environment.

Repair of a calcium-dependent adhesive mechanism of embryonic neural retina cells: kinetic and molecular analysis.

In this paper, we examine the restitution of a Ca2+-dependent adhesive system of embryonic chick neural retina cells following trypsinization in the absence of divalent cations. The processes involved in the restitution of this system are dissected by monitoring the effects of various drugs, alone and in specific combinations, on the kinetics of adhesion and the morphology of 24-h aggregates. In agreement with prior observations, aggregate formation can be separated into two distinct phases: the formation of adhesions and their ultimate stabilization. The formation of adhesions appears to involve two processes, de novo synthesis of protein and mobilization of an endogenous pool, while stabilization requires de novo synthesis of RNA and protein glycosylation. We also examine the appearance of two cell surface molecules previously implicated in the function of the Ca2+-dependent adhesive system. The cells repaired in the presence or absence of the inhibitors and surface-iodinated at 3 and 5 h. The labelled molecules are then separated by two-dimensional polyacrylamide gel electrophoresis. The appearance of the two molecules, gp130/4.8 and gp70/4.8, is affected by the various drugs in a manner consistent with the effects of these agents on the kinetics of adhesion. These molecules appear to exist in endogenous pools and they are also synthesized de novo during the repair period.

Enzymic dissection of embryonic cell adhesive mechanisms. III. Immunological identification of a component of the calcium-dependent adhesive system of embryonic chick neural retina cells.

Immunological probes and cell-surface biochemical analysis are combined to identify a cell-surface glycoprotein, which appears to be involved in the formation of calcium-dependent adhesions among embryonic chick neural retina cells. Comparison of radioiodinated cells shows that adhesive-competent cells have at their surface two glycoproteins not present on non-adhesive cells. These two components (with Mr 130 x 10(3), pI 4.8 and Mr 70 x 10(3), pI 4.8) are immunoprecipitable with adhesion-blocking antisera. A fraction derived from retina-conditioned medium, containing one major immunoprecipitable component, neutralizes the inhibitory activity of aggregation-inhibiting Fab' fragments and competes in immunoprecipitation for the two glycoprotein components. The evidence suggests that one or both of these components is involved in the formation of calcium-dependent adhesions among embryonic chick neural retina cells.

The accessibility of certain proteins on embryonic chick neural retina cells to iodination and tryptic removal is altered by calcium.

We have used cell-surface-specific labelling techniques and two-dimensional gel electrophoresis to identify proteins on embryonic chick neural retina cells and to determine the effects of Ca2+ on their accessibility to labelling and tryptic removal. A number of proteins on these cells are, in the presence of Ca2+, relatively inaccessible to iodination and/or tryptic removal. Of these, a glycoprotein of Mr approx. 130 x 10(3), with a pI of approx. 4.8, is the major cell-surface-iodinatable species that is retained during trypsinization in the presence of Ca2+. The removal of Ca2+ renders this glycoprotein much more accessible to both procedures. Its accessibility to these probes decreases on re-addition of Ca2+. The accessibility of its oligosaccharide moiety to galactose oxidase is, however, unaltered by the removal of Ca2+. These characteristics, together with immunological data presented elsewhere suggest that this glycoprotein may be a component of the Ca2+-dependent adhesive system that can be demonstrated on these cells.

General inhibition of embryonic cell aggregation by neoplastic cells

The presence of small numbers of cancer cells cause an inhibition in the aggregation of embryonic chick neural retina cells. Experiments were made to investigate the inhibitory action of malignant mouse and virus-transformed chick neural retina cells on the aggregation and adhesion of several embryonic cell types. Malignant mouse cells, and their conditioned medium, inhibited the aggregation and adhesion of embryonic skeletal muscle heart and liver cells. The transformed retinal cells inhibited the aggregation of neural retina cells. These results suggest that the effect of cancer cells on the behavior of embryonic cells is a general one not related to the origin of the cancer cells.

Calcium-dependent and calcium-independent adhesive mechanisms are present during initial binding events of neural retina cells.

The hypothesis that intercellular adhesion can be subdivided into two separable phenomena, an initial recognition event and a subsequent stabilization, is supported by the use of a new cell binding assay that provides a quantitative measure of intercellular binding strengths. Radioactive single cells are brought into contact with cell monolayers at 4 degrees C in sealed compartments. The compartments are inverted and a centrifugal force is then applied tending to dislodge the probe cells from the monolayers. By varying the speed of centrifugation, the force maintaining association between embryonic chick neural retina cells was determined to be on the order of 10(-5) dynes after incubation at 4 degrees C. Brief incubations at 37 degrees C resulted in significant strengthening of the intercellular bond. Using this cell binding assay, neural retina cells were shown to exhibit both a Ca++-independent and a Ca++-dependent mechanism in their initial binding to one another.
