Electrospray Ionization Tandem Mass Spectrometric Study of Selected Phosphine-Based Ligands for Catalytically Active Organometallics.

Selected organometallic compounds are nowadays extensively used as highly efficient catalysts in organic synthesis. A great variety of different ligand systems exists, of which phosphine-based ligands are a significant subgroup. While mass spectrometry, predominantly electrospray ionization mass spectrometry (ESI-MS), is a standard analytical technique for the identification of new ligands and their metal complexes, there is little information on the behavior of phosphine-based ligands/molecules by electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS) at low collision energies (<100 eV) in the literature. Here, we report a study on the identification of typical product ions occurring in tandem mass spectra of selected phosphine-based ligand systems by ESI-CID-MS/MS. The influence on the fragmentation behavior of different backbones (pyridine, benzene, triazine) as well as different spacer groups (amine, methylamine, methylene), which are directly linked to the phosphine moiety, is investigated by tandem mass spectrometry. In addition, possible fragmentation pathways are elaborated based on the assigned masses in the tandem mass spectra with high-resolution accurate mass determination. This knowledge may be particularly useful in the future for the elucidation of fragmentation pathways for coordination compounds by MS/MS, where the studied compounds serve as building blocks.

Structure and Biological Activity Analysis of Fucoidan Isolated from Sargassum siliquosum.

Fucoidans are heterologous polysaccharides commonly seen in brown macroalgae and are known for their biological activity including anticancer, antiangiogenic, immunomodulation, and antiviral properties. The brown macroalga Sargassum siliquosum was used for the extraction and analysis of fucoidan in this study. The S. siliquosum fucoidan was indicated as a galactofucoidan composed of sugars, uronate, and sulfate at a ratio of 12:1:4 and its purity was 85% based on the abovementioned three major components. Structural analysis by electrospray ionization collision-induced dissociation tandem mass spectroscopy revealed that the purified fucoidan consisted of a carbohydrate chain composed of (1→3)-linked or (1→4)-linked l-fucose residues, with sulfate groups at C-2 and C-4 positions. Galactose residues with (1→4)-linkages function as the branch points and they are located at the C-3 or C-4 position of fucose residues. Galactose residues are sulfated mainly at C-4 and C-6, while some sulfation can also be seen at C-2. The fucoidan purified from S. siliquosum demonstrated antioxidant, anti-inflammatory, and antilipogenic activities.

1,4-D-Glucan block copolymers: synthesis and comprehensive structural characterization

Multi-block glucans comprising permethylated and partially methylated blocks are compounds of interest. In order to monitor their formation by transglycosylation of corresponding starting glucans, a method has been developed and applied to model compounds. This method allows determining the average length of the blocks and the progress of incorporation of methyl blocks in partially methylated sequences with a random distribution. The method, comprising liquid chromatography mass spectrometry (LC-MS) and electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MSn) measurements of two types of peralkylated glucans representing derivatives of the target compounds, is comprehensively described and discussed. ESI-MSn allows looking into the sequences of oligomeric domains. In addition, transglycosylation is followed by attenuated total reflection FTIR and NMR spectroscopy.Graphical abstract

Structural analysis and anti-complement activity of polysaccharides extracted from Grateloupia livida (Harv.) Yamada

Polysaccharides were extracted from Grateloupia livida (Harv.) Yamada using hot water (extracted product denoted WGW) and then degraded in dilute sulfuric acid (degraded product denoted WGWD). The degraded mixture was then separated into four fractions through anion exchange chromatography on 2-diethylaminoethanol (DEAE)-Bio-Gel Agarose FF gel. Electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS) was performed to elucidate the structural features of all fractions. In combination with nuclear magnetic resonance spectroscopy (NMR) and infrared spectroscopy (IR) data, the major polysaccharide structures were concluded to be µ-carrageenan and κ-carrageenan. µ-Carrageenan usually has a backbone of alternating 1,3-linked s-D-galactopyranose residues sulfated at C-4 and 1-4-linked α-D-galactopyranose residues sulfated at C-6, while κ-carrageenan consists of alternating 1,3-linked s-D-galactopyranose residues sulfated at C-4 and 1,4-linked α-D-3,6-anhydrogalactopyranose residues. Trace v-carrageenan- composed of 1,3-linked s-D-galactopyranose residues sulfated at C-4 and 1,4-linked α-D-galactopyranose residues sulfated at C-2 and C-6, was also detected. Furthermore, the polysaccharide had a backbone comprising 1,3-linked s-D-galactopyranose and 1,4-linked α-L-galactopyranose sulfated at C-6, which is the agarose precursor. The hydroxy groups in the galactopyranose were partially substituted by methyl and pyruvic acid acetal (PA) groups. The anti-complementary activities of WGW and its derivatives against classical pathways were measured. The native polysaccharides in WGW had higher activities, while the derivatives had much weaker activities. This indicated that the molecular weight and sulfate content were important factors affecting the anti-complement activity.

Electron transfer dissociation of synthetic and natural peptides containing lanthionine/methyllanthionine bridges.

The modes of cleavage of lanthionine/methyllanthionine bridges under electron transfer dissociation (ETD) were investigated using synthetic and natural lantipeptides. Knowledge of the mass spectrometric fragmentation of lanthionine/methyllanthionine bridges may assist in the development of analytical methods for the rapid discovery of new lantibiotics. The present study strengthens the advantage of ETD in the characterization of posttranslational modifications of peptides and proteins. Synthetic and natural lantipeptides were obtained by desulfurization of peptide disulfides and cyanogen bromide digestion of the lantibiotic nisin, respectively. These peptides were subjected to electrospray ionization collision-induced dissociation tandem mass spectrometry (CID-MS/MS) and ETD-MS/MS using an HCT ultra ETDII ion trap mass spectrometer. MS3 CID was performed on the desired product ions to prove cleavage of the lanthionine/methyllanthionine bridge during ETD-MS/MS. ETD has advantages over CID in the cleavage of the side chain of lanthionine/methyllanthionine bridges. The cleavage of the N-Cα backbone peptide bond followed by C-terminal side chain of the lanthionine bridge results in formation of c•+ and z+ ions. Cleavage at the preceding peptide bond to the C-terminal side chain of lanthionine/methyllanthionine bridges yields specific fragments with the cysteine/methylcysteine thiyl radical and dehydroalanine. ETD successfully cleaves the lanthionine/methyllanthionine bridges of synthetic and natural lantipeptides. Diagnostic fragment ions of ETD cleavage of lanthionine/methyllanthionine bridges are the N-terminal cysteine/methylcysteine thiyl radical and C-terminal dehydroalanine. Detection of the cysteine/methylcysteine thiyl radical and dehydroalanine in combined ETD-CID-MS may be used for the rapid identification of lantipeptide natural products.

Dissociation Behavior of a TEMPO-Active Ester Cross-Linker for Peptide Structure Analysis by Free Radical Initiated Peptide Sequencing (FRIPS) in Negative ESI-MS.

We have synthesized a homobifunctional amine-reactive cross-linking reagent, containing a TEMPO (2,2,6,6-tetramethylpiperidine-1-oxy) and a benzyl group (Bz), termed TEMPO-Bz-linker, to derive three-dimensional structural information of proteins. The aim for designing this novel cross-linker was to facilitate the mass spectrometric analysis of cross-linked products by free radical initiated peptide sequencing (FRIPS). In an initial study, we had investigated the fragmentation behavior of TEMPO-Bz-derivatized peptides upon collision activation in (+)-electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS) experiments. In addition to the homolytic NO-C bond cleavage FRIPS pathway delivering the desired odd-electron product ions, an alternative heterolytic NO-C bond cleavage, resulting in even-electron product ions mechanism was found to be relevant. The latter fragmentation route clearly depends on the protonation of the TEMPO-Bz-moiety itself, which motivated us to conduct (-)-ESI-MS, CID-MS/MS, and MS3 experiments of TEMPO-Bz-cross-linked peptides to further clarify the fragmentation behavior of TEMPO-Bz-peptide molecular ions. We show that the TEMPO-Bz-linker is highly beneficial for conducting FRIPS in negative ionization mode as the desired homolytic cleavage of the NO-C bond is the major fragmentation pathway. Based on characteristic fragments, the isomeric amino acids leucine and isoleucine could be discriminated. Interestingly, we observed pronounced amino acid side chain losses in cross-linked peptides if the cross-linked peptides contain a high number of acidic amino acids. Graphical Abstract ᅟ.

Accurate mass MS/MS/MS analysis of siderophores ferrioxamine B and E1 by collision-induced dissociation electrospray mass spectrometry.

Siderophores are bacterially secreted, small molecule iron chelators that facilitate the binding of insoluble iron (III) for reuptake and use in various biological processes. These compounds are classified by their iron (III) binding geometry, as dictated by subunit composition and include groups such as the trihydroxamates (hexadentate ligand) and catecholates (bidentate). Small modifications to the core structure such as acetylation, lipid tail addition, or cyclization, make facile characterization of new siderophores difficult by molecular ion detection alone (MS(1)). We have expanded upon previous fragmentation-directed studies using electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS/MS) and identified diagnostic MS(3) features from the trihydroxamate siderophore class for ferrioxamine B and E1 by accurate mass. Diagnostic features for MS(3) include C-C, C-N, amide, and oxime cleavage events with proposed losses of water and -CO from the iron (III) coordination sites. These insights will facilitate the discovery of novel trihydroxamate siderophores from complex sample matrices. Graphical Abstract ᅟ.

Structural Study of Sulfated Fuco-Oligosaccharide Branched Glucuronomannan from Kjellmaniella crassifolia by ESI-CID-MS/MS

GRAPHICAL ABSTRACTA polysaccharide was purified from brown alga Kjellmaniella crassifolia, and its precise structure was deduced on the basis of sequence analysis of oligosaccharide fragments obtained from mildly and strongly acidic hydrolysis of the polysaccharide. Electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS) was used to identify the structural features of the resultant oligosaccharides. The side chain oligosaccharides were composed of α-1,3-linked 4-O-sulfated fucose and α-1,3-linked 2,4-O-disulfated fucose, while the backbone oligosaccharides consisted of the disaccharide repeating unit of β-1,4-linked glucuronic acid and α-1,2-linked mannose. Thus, the fucoidan was determined to be a sulfated fuco-oligosaccharide branched glucuronomannan.

Structural analysis and anti-complement activity of polysaccharides from Kjellmaniella crsaaifolia.

Two polysaccharides, named KCA and KCW, were extracted from Kjellmaniella crassifolia using dilute hydrochloric acid and water, respectively. Composition analysis showed that these polysaccharides predominantly consisted of fucose, with galactose, mannose and glucuronic acid as minor components. After degradation and partial desulfation, electrospray ionization mass spectrometry (ESI-MS) was performed, which showed that the polysaccharides consisted of sulfated fucooligosaccharides, sulfated galactofucooligosaccharides and methyl glycosides of mono-sulfated/multi-sulfated fucooligosaccharides. The structures of the oligomeric fragments were further characterized by electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS2 and ESI-CID-MS3). Moreover, the activity of KCA and KCW against the hemolytic activity of both the classical and alternative complement pathways was determined. The activity of KCA was found to be similar to KCW, suggesting that the method of extraction did not influence the activity. In addition, the degraded polysaccharides (DKCA and DKCW) displayed lower activity levels than the crude polysaccharides (KCA and KCW), indicating that molecular weight had an effect on activity. Moreover, the desulfated fractions (ds-DKCA and ds-DKCW) showed less or no activity, which confirmed that sulfate was important for activity. In conclusion, polysaccharides from K. crassifolia may be good candidates for the treatment of diseases involving the complement pathway.

Electrospray ionization collision‐induced dissociation tandem mass spectrometry of amoxicillin and ampicillin and their degradation products

Detailed analysis of the literature results on the electrospray ionization mass spectrometry (ESI-MS) fragmentation of amoxicillin and ampicillin, and their comparison with our results, have revealed some incorrect suggestions or incomplete interpretations of mass spectra of these compounds. Therefore, this paper contains a comprehensive discussion devoted to the ESI-MS/MS of ampicillin and amoxicillin as well as their degradation products, namely products of hydrolysis and methanolysis. Electrospray ionization collision-induced dissociation tandem mass (ESI-CID-MS/MS) spectra and accurate mass measurements were made on a quadrupole time-of-flight (Q-tof) mass spectrometer. Hydrolysis of the antibiotics was performed by heating, for a few hours, their aqueous solutions adjusted to pH 10. Methanolysis of the antibiotics was performed by heating their methanol solutions for a few minutes. Additionally, mass spectra of isotope-labeled compounds were also obtained. A number of fragment ions, previously wrongly interpreted or not interpreted, have been rationalized. For example, formation of an abundant fragment at m/z 208 originating from the protonated amoxicillin molecule (ion [Amox + H](+)) was previously rationalized as a result of breaking of two bonds of the β-lactam ring. We found that this fragment ion had to be formed by the loss of ammonia and breaking of three bonds of the bicyclic system. The discussion presented enables a better understanding of the MS decompositions of amoxicillin and ampicillin as well as their degradation products. MS decomposition is used for the determinations of these compounds, when the so-called multiple-reaction monitoring is applied during liquid chromatography (LC)/ESI-MS analysis. Thus, better understanding of MS decompositions of the above compounds seems to be important.

Differentiation of isomeric pentose disaccharides by electrospray ionization tandem mass spectrometry and discriminant analysis

The structural characterization of unknown oligosaccharides remains a big challenge since a large number of isomeric structures are possible even for disaccharides. In this work, electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MS/MS) was used for the differentiation of isomeric pentose disaccharides, α-(1 → 5)-L-arabinobiose (Ara(2)) and β-(1 → 4)-D-xylobiose (Xyl(2)). ESI-MS/MS spectra of [M + Li](+) and [M + Na](+) ions of Ara(2) and Xyl(2), as well as these precursor ions of (18)O-labelled disaccharides, were acquired using two mass spectrometers equipped with different analyzers: LIT (linear ion trap) and Q-TOF (quadrupole time-of-flight). Product ions observed in MS/MS spectra arise from the cleavage at the nonreducing side of the glycosidic bond (Y(1)(+)) and from cross-ring cleavages (0,1)A(2)(+), (0,2)A(2)(+), and (0,3)A(2)(+) at the reducing residue. Statistically significant differences were observed between the relative abundance of specific product ions, when comparing both disaccharides. These differences allowed discriminant models to be built and to propose a criterion using the relative abundances of selected ions capable of discriminating between the isomers for both adduct ions and spectrometers. Isomeric pentose disaccharides can be distinguished based on the fragmentation of both [M + Li](+) and [M + Na](+) ions and using different mass spectrometers. However, LIT instrument has a better discriminant power.

Negative-ion fast atom bombardment and electrospray ionization tandem mass spectrometry for characterization of sulfated and sialyl Lewis-type glycosphingolipids.

Structural characterization of sulfated and sialyl Lewis (Le)-type glycosphingolipids performed by fast atom bombardment (FAB) and electrospray ionization (ESI) mass spectrometry is described. Both FAB and ESI collision-induced dissociation tandem mass spectrometry (CID-MS/MS) of acidic glycosphingolipids allowed identification of the sulfated or sialyl sugar, and provided information on the saccharide chain sequence. The negative-ion tandem FABMS of sulfated Le-type glycosphingolipids having the non-reducing end trisaccharide ion as the precursor can be used to differentiate the Le(a)- and Le(x)-type oligosaccharides. The ESI CID-MS/MS of multiple-charged ions provided even more detailed structural information, and some of the useful daughter ions appeared with higher m/z values than the precursor because of a lower charge-state. These methodologies can be applied to the structural analyses of glycoconjugates with much larger molecular masses and higher polarity, such as the poly-sulfated and sialyl analogues.
