Essential oils from fennel plants as valuable chemical products: gas chromatography–mass spectrometry, FTIR, quantum mechanical investigation, and antifungal activity

AbstractIn the present study, the biomass produced by fennel plants (Foeniculum vulgare Mill.) was converted to yield bioactive chemicals, and the hydrodistillation method was used to extract the essential oils (EOs) from both the leaves and the umbels. The antifungal activity of the EOs was tested using bioassay against the development of Fusarium oxysporum MW854649 and Alternaria solani MT279570. Molecular spectroscopic detection techniques were used to evaluate the EO products using gas chromatography–mass spectrometer (GC–MS) and Fourier transform infrared spectroscopy (FTIR). GC–MS equipped with single quadruple analyzers have been used to measure the electron ionization (EI) mass spectra of the primary constituents of fennel EOs at 70 eV. The main chemical compounds in the EO from leaves were anethole, estragole, D-limonene, trans-β-ocimene, and fenchone, with percentages of 37.94, 35.56, 17.46, 1.53, and 1.49%, respectively. The abundant compounds in the EO from umbels were estragole, anethole, D-limonene, fenchone, and γ-terpinene, with percentages of 51.18, 25.08, 12.22, 6.57, and 2.86%, respectively. EI mass spectral fragmentation of the major compounds D-limonene, estragole, anethole, and fenchone has been investigated. Umbels and leaf EOs at 5000 mg/L displayed the strongest suppression of fungal growth against A. solani, with values of 87.78% and 79.63%, respectively, compared to the positive control (94.44%). The EOs from umbels and leaves at 5000 mg/L showed the highest inhibition of fungal growth against F. oxysprium as compared to the positive control (94.44%), with values of 77.77% and 72.96%, respectively. All of the important ions—including a few distinctive fragment ions—have comprehensive fragmentation pathways defined. Based on EI, the main routes of fragmentation for the primary compounds have been identified. The existence of alkenes, aliphatic alcohols, ethers, carboxylic acids, ester compounds, alkanes, hydrogen-bonded alcohols, and phenols was demonstrated by the FTIR analysis of fennel EOs. On the other hand, the reactive behavior of the studied molecules has been investigated using two quantum mechanics method: the modified neglect of diatomic overlap (MNDO), a semi-empirical method, and the density functional theory (DFT)/B3LYP hybrid density functional method with the 6-311G (d, p) basis set in the ground state for gas phase. The optimum geometries have been obtained through the execution of computations and electrostatic potential. The obtained analytical and calculated results were then used to understand the activity of the studied EOs in further medical applications.

Change of the favored routes of EI MS fragmentation when proceeding from N1, N1-dimethyl-N2-arylformamidines to 1,1,3,3-tetraalkyl-2-arylguanidines: substituent effects

Although series of N(1), N(1)-dimethyl-N(2)-arylformamidines and of 1,1,3,3-tetraalkyl-2-arylguanidines are structurally analogous and similar electron-ionization mass spectral fragmentation may be expected, they display important differences in the favored routes of fragmentation and consequently in substituent effects on ion abundances. In the case of formamidines, the cyclization-elimination process (initiated by nucleophilic attack of the N-amino atom on the 2-position of the phenyl ring) and formation of the cyclic benzimidazolium [M-H](+) ions dominates, whereas the loss of the NR(2) group is more favored for guanidines. In order to gain information on the most probable structures of the principal fragments, quantum-chemical calculations were performed on a selected set. A good linear relation between log{I[M-H](+)I [M](+*)} and sigma(R)(+) constants of substituent at para position in the phenyl ring occurs solely for formamidines (r = 0.989). In the case of guanidines, this relation is not significant (r = 0.659). A good linear relation is found between log{I[M-NMe(2)](+)/I [M](+*)} and sigma(p)(+) constants (r = 0.993).

Chemical ionization mass spectral analysis of pinacolyl alcohol and development of derivatization method using p-tolyl isocyanate

A simple and routine chemical ionization method (CI) providing molecular weight information is developed for the unambiguous identification of 3,3-dimethyl-2-butanol known as pinacolyl alcohol (PA), a potential precursor for prohibited chemical weapons such as Soman, a nerve agent. This method meets the CI criteria used for the evaluation of proficiency tests conducted by OPCW avoiding use of reference chemical. In addition, a derivatization method using p-tolyl isocyanate for the analysis of PA using GC-MS has also been developed for the identification. This derivatization method facilitates the easy analysis of PA at low concentration present in environmental matrices. The efficiency of the derivatization is >99% with 10 fold increase in the sensitivity of the GC-MS signal. The electron ionization mass spectral fragmentation of the p-tolyl isocyanate (PTI) derivatives of three isomeric alcohols is also established. The positive CI of PTI derivatives of these alcohols resulted in molecular ion information and meets the OPCW criteria for CI method of analysis. Further, the retention indices for all the studied alcohols after derivatization help the unambiguous identification of these compounds present in complex environmental matrices.

Substituted 3-phenylpropenoates and related analogs: electron ionization mass spectral fragmentation and density functional theory calculations

Analysis of ethyl 3-(2-chlorophenyl)propenoate by electron ionization mass spectrometry showed the distinct loss of an ortho chlorine. To characterize the structural requisites for the observed mass fragmentation, a series of 30 halogen-substituted 3-phenylpropenoate-related structures were examined. All ester-containing alkene derivatives exhibited loss of the distinctive chlorine from the 2-position of the phenyl ring. Analogous derivatives with the halogen (chlorine or bromine) in the para position did not evidence selective halogen loss. Results demonstrated that substituted 3-phenylpropenoates and their analogs fragment via the formation of a previously reported benzopyrylium intermediate. To understand the correlation between the intramolecular radical substitution and the abundance and selectivity of the chlorine (or other halogen) displacement, density functional theory calculations were performed to determine the charge on the principal cation involved in the chlorine loss (in the ortho, meta, and para positions), the charge for the neutral radical (noncation), the excess alpha-electron density on the relevant atom and the energy to form the cation from the neutral atom (ionization energy). Results showed that the selectivity and extent of halogen displacement correlated highly to the electrophilicity of the radical cation as well as the neutral radical. These data further support the proposed fragmentation mechanism involving intramolecular radical elimination.

Hydrogen and Trimethylsilyl Transfers During EI Mass Spectral Fragmentation of Hydroxycarboxylic and Oxocarboxylic Acid Trimethylsilyl Derivatives

This paper, describing electron ionization mass spectral fragmentation of some hydroxycarboxylic and oxocarboxylic acid trimethylsilyl derivatives, focuses on the formation of fragment ions resulting from the interactions between the two functionalities of these compounds. These interactions result in the formation of fragment ions at [CH2=C(OTMS)2]+., [CH2=CHC(OTMS)=OTMS]+, [M-31]+, [M-105]+, and [M-RCHO]+. in the case of hydroxycarboxylic acid trimethylsilyl derivatives of formula RCHOTMS(CH2)nCOOTMS and at [RC(OTMS)=CH2]+., [RC(=OTMS)CH=CH2]+, and [M-RC(=O)CH2]+ in the case of oxocarboxylic acid trimethylsilyl esters of formula RC(=O)(CH2)nCOOTMS. Some of these fragmentations appeared to be sufficiently specific to be used to characterize these compounds. Several fragmentation pathways involving trimethylsilyl and hydrogen transfers were proposed to explain the formation of these different fragment ions and were substantiated by deuterium labeling.

Electron ionization mass spectral fragmentation of new (E)-4-chalconylothioalkyl substituted derivatives of piperidinie, 4-methylpiperidine, morpholine and piperazine as well as (E)-4-bromoalkylthiochalcones being substrates of the former compounds have been studied. Fragmentation pathways have been proposed on the basis of the accurate mass and metastable transition measurements.

The Conversion of Stilbenols to Stilbenethiols via N,N-Dimethylthiocarbamates

A series of O-stilbenyl N,N-dimethylthiocarbamates has been prepared by the reaction of N,N-dimethylthiocarbamoyl chloride with stilbenols in the presence of DABCO and DMF. The thermal rearrangement of O-stilbenyl N,N-dimethylthiocarbamates into S-stilbenyl analogues has also been studied. The S-stilbenyl N,N-dimethylthiocarbamates obtained have been converted into the corresponding stilbenethiols. Electron ionization mass spectral fragmentation of these compounds has been studied. Fragmentation pathways have been proposed on the basis of the accurate mass and metastable transition measurements.

Trimethylsilyl transfer during electron ionization mass spectral fragmentation of some omega-hydroxycarboxylic and omega-dicarboxylic acid trimethylsilyl derivatives and the effect of chain length.

The electron ionization (EI) mass spectral fragmentation of omega-hydroxycarboxylic and omega-dicarboxylic acid trimethylsilyl derivatives was investigated. The mass spectra of these compounds exhibited fragment ions resulting from classical fragmentation of the trimethylsilyl ether and ester groups, and others resulting from the interactions between the two functionalities (m/z 147, 204, 217, [M-31](+) and [M-105](+) in the case of omega-hydroxycarboxylic acid derivatives and m/z 147, 204, 217 and [M-131](+) in the case of omega-dicarboxylic acid derivatives). Several fragmentation pathways were proposed to explain the formation of these different fragment ions. It is proposed that the ions at m/z 204 and 217 are formed via an initial trimethylsilyl transfer between the ether and the ester group or between the two ester groups. This transfer appeared to be more favoured in the case of omega-dicarboxylic acid derivatives and to be dependent on the chain length. A more efficient transfer was in fact observed for compounds with a relatively long alkyl chain. In the case of shorter omega-hydroxycarboxylic and omega-dicarboxylic acid trimethylsilyl derivatives the formation of the ions at m/z 204 and 217 suffers strongly from competition from production of the ion at m/z 147.
