Influence of propionic acid microorganisms on the quality indicators of the produced sausage

The article considered the effect of probiotic bacteria on the quality of meat for semi-smoked sausages. The biochemical activity of propionic acid bacteria was studied at the stage of the production process of semi-smoked sausages after the treatment of raw materials with starter microorganisms. We also studied the effect of salt, and sodium nitrite on the development of propionic acid bacteria in raw beef. The results of the technology of production of semi-smoked sausages treated with propionic acid bacteria in the amount of 0.1%, and the quality characteristics of the finished sausage products are presented. On the basis of experimental studies optimal technological parameters of salting and precipitation in the production of fermented semi-smoked sausages were selected. The use of liquid propionic acid bacteria (2 strains of propionic acid bacteria Propionibacterium shermani) in the amount of 0.1% will give a decrease in the production time of half-smoked sausages by two times. As a result of the research it was proved that the introduction of 2 different strains of propionic acid bacteria concentrate into raw meat accelerates biochemical changes in the process of salting and provides functional and technological properties. Also, the results of the study showed that propionic acid bacteria used in the technology of semi-smoked sausages are resistant to the amount of food salts and sodium nitrite used. As a result of research, a better way of using liquid propionate microorganism in the production of semi-smoked fermented sausages was adopted.

The Effect of Probiotic Bacteria on Composition and Metabolite Production of Faecal Microbiota Using In Vitro Batch Cultures.

Probiotic supplements are increasingly being used to target the gut microbiome with a view to improving cognitive and psychological function via the gut-brain axis. One possible mechanism behind the effect of probiotics is through alterations to microbially-derived metabolites including short-chain fatty acids (SCFA) and neurotransmitters. However, research to date has largely been conducted in animal models or under conditions irrelevant to the human gastrointestinal tract (GIT). The aim of the current work was therefore to use anaerobic, pH controlled in vitro batch cultures to (a) assess the production of neuroactive metabolites in human faecal microbiota under conditions relevant to the human GIT, and (b) to explore how several pre-selected probiotic strains may affect bacterial composition and metabolite production. Enumeration of bacteria was assessed using fluorescence in situ hybridisation with flow cytometry, and concentrations of SCFAs and neurotransmitters were measured using gas chromatography and liquid chromatography mass spectroscopy, respectively. GABA, serotonin, tryptophan, and dopamine were successfully detected, suggesting some level of microbial derivation. The addition of Lactococcus lactis W58 and Lactobacillus rhamnosus W198 resulted in a significant increase in lactate after 8 h of fermentation, while no significant effect of probiotics on bacterial composition or neurotransmitter production was found.

Therapeutic Role of Probiotics In Managing Various Diseases

Since last few decades, probiotics gained the attention of clinicians for their use in the prevention and treatment of multiple diseases. Probiotic bacteria play a critical and functional role in clinical and nutritional applications. In the last decades, a wide number of scientific works and studies shed light on the crucial role of probiotic microorganisms and their potential to cure various ailments and disorders based on the promising findings and results of different in vitro and in vivo investigations, suggesting a powerful connection flanked by these so-called probiotics and the human immuno-modulatory responses. The issues that were noticed are included: fibrocystic, diabetes, acne, colon cancer, cardiovascular, urinary tract infections, atopic eczema syndrome, food allergies and obesity. Probiotics main mechanisms of action include enhanced mucosal barrier function, direct antagonism with pathogens inhibition of bacterial adherence and invasion capacity, boosting of the immune system and regulation of the central nervous system. Enhancement in using drug treatment has led to the appearance of drug-resistance concern, thus probiotics can be a suitable choice. There is a mutual communication between the central nervous system and the liver, the so-called “microbiota-gut-liver axis” as well as a reciprocal communication between the intestinal microbiota and the central nervous system through the “microbiota-gut-brain axis”. This review focuses on the effect of probiotic bacteria and their metabolites on immune-boosting, prevention and treatment of these diseases.

The effect of probiotic bacteria on toll-like receptor-2 and -4 expression by spermatozoa in couples with unexplained recurrent spontaneous abortion

The disturbance of microbiota composition in the female reproductive tract (FRT) can result in several reproductive disorders. Spermatozoa express toll like receptors (TLRs) and may encounter many types of microbiota in the FRT, however no study has been performed regarding the interaction between spermatozoa TLRs and FRT microbiota in unexplained recurrent spontaneous abortion (URSA) and fertile couples. In this study, we investigate the interaction of vaginal lactobacillus casei probiotic as a representative of FRT microbiota with TLR2 and 4 on spermatozoa. Ten fertile couples and ten URSA couples were involved in this study. Untreated and lactobacillus casei probiotic treated purified spermatozoa were evaluated for TLR2 and 4 expression by flow cytometry. Vaginal lactobacillus casei probiotic treatment of spermatozoa led to increased expression of TLR4 and decreased expression of TLR2 on spermatozoa in both URSA and fertile couples. Vaginal lactobacillus casei probiotic led to an increase in TLR4 expression and a decrease in TLR2 expression on spermatozoa in fertile and URSA groups. However, the disturbed expression of TLR2 and 4 was not completely correct, and further studies with other types of vaginal lactobacilli are needed. In contrast to our expectation, vaginal lactobacillus casei probiotic could not improve the disturbed expression of TLR2 and TLR4 in the RSA group. This could be due to small sample size and the use of one type of lactobacillus. Therefore, further study needs to be performed with other types of lactobacilli to determine the effect of microbiota and probiotics on spermatozoa function such as motility, acrosome reaction, sperm capacitation, sperm and egg fusion and spermatozoa motility and apoptosis and etc. Nevertheless, this study can provide a first step to investigate the effectiveness of vaginal microbiota on spermatozoa, and consequently design new strategies for RSA couples.

Evaluation of Probiotic Properties of Novel Brazilian Lactiplantibacillus plantarum Strains.

Beneficial effects of Lactiplantibacillus plantarum strains have been widely reported. Knowing that the effects of probiotic bacteria are strain-dependent, this study aimed to characterize the probiotic properties and investigate the gastrointestinal protective effects of nine novel L. plantarum strains isolated from Bahia, Brazil. The probiotic functionality was first evaluated in vitro by characterizing bile salt and acidic tolerance, antibacterial activity, and adhesion to Caco-2 cells. Antibiotic resistance profile, mucin degradation, and hemolytic activity assays were also performed to evaluate safety features. In vivo analyses were conducted to investigate the anti-inflammatory effects of the strains on a mouse model of 5-Fluorouracil-induced mucositis. Our results suggest that the used L. plantarum strains have good tolerance to bile salts and low pH and can inhibit commonly gastrointestinal pathogens. Lp2 and Lpl1 strains also exhibited high adhesion rates to Caco-2 cells (13.64 and 9.05%, respectively). Phenotypical resistance to aminoglycosides, vancomycin, and tetracycline was observed for most strains. No strain showed hemolytic or mucolytic activity. Seven strains had a protective effect against histopathological and inflammatory damage induced by 5-FU. Gene expression analysis of inflammatory markers showed that five strains upregulated interleukin 10 (Il10), while four downregulated both interleukin 6 (Il6) and interleukin 1b (Il1b). Additionally, all strains reduced eosinophilic and neutrophilic infiltration; however, they could not prevent weight loss or reduced liquid/ food intake. Altogether, our study suggests these Brazilian L. plantarum strains present good probiotic characteristics and safety levels for future applications and can be therapeutically adjuvant alternatives to prevent/treat intestinal mucositis.

Effect of probiotic bacteria on physicochemical, microbiological, textural, sensory properties and fatty acid profile of sour cream.

Sour cream is a dairy product with good potential to act as a food carrier for probiotic bacteria. The effect of probiotic cultures on the quality of sour cream from dry dairy ingredients was assessed during 5 weeks of storage. The treatments design was as follows: Bifidobacterium, Bifidobacterium+ Lactobacillus casei, Lactobacillus acidophilus, Lactobacillus acidophilus+ Lactobacillus casei and Lactobacillus casei. Fatty acids profile, texture, microbial load and sensory characteristics were analyzed. Comparison of fatty acids of different creams produced showed that there is a significant difference between the produced creams. There are a wide range of fatty acids in the structure of sour cream fat. Also, with the addition of these bacteria, the texture of the samples became firmer during storage. The sensory analyses showed that the addition of all three bacteria increased the overall acceptance of this product by the panelists. It is suggested that sour cream fortified with probiotics well orientated functional food had accepted composition and sensory properties.

Effects of Probiotic Bacteria on Central Neuronal Activation in Experimental Colitis.

Brain-gut axis dysregulation is observed in inflammatory bowel disease. However, the effect of altered gut flora on neuro- immunomodulation and its role in the pathogenesis of inflammatory bowel disease are unknown. The aims of this study are to determine (i) whether colitis modifies the expression of c-fos, a marker of general neuronal activation in the brain and (ii) whether this activation could be modulated by probiotic bacteria. In this study, 28 Sprague-Dawley rats were divided into 4 groups: colitis-probiotic group, non-colitis-fed-control group receiv- ing probiotic Lactobacillus delbrueckii subsp. Bulgaricus B3 strain for 7 days, colitis group, and sham group receiving only sodium chlo- ride. Colitis was induced by intracolonic administration of trinitrobenzene sulfonic acid-ethanol. The expression of c-fos was detected by immunohistochemistry in the brain tissue. Cytokines and inflammatory mediators were analyzed in the plasma. Histological scores and oxidative status were analyzed in the colon samples. The inflammatory response was accompanied by increased levels of cytokines, lipid peroxidation activities, c-fos expression in the medial nucleus of the amygdala, and decreased levels of antioxidant enzymes in the colitis (P < .001). Probiotic treatment reversed those effects. Also, histopathologic scores were significantly lower in the probiotic-treated groups compared to the colitis group (P = .035). In contrast, the expression of c-fos was significantly increased in the paraventricular nucleus of hypothalamus in the probiotic- treated rats (P < .001). Colitis and intestinal inflammation are associated with the activation of neurons in the limbic system creating stress-like effects in the brain. Probiotics diversely modulate limbic response and hypothalamic axis activity in addition to protective effects in inflammation.

Oral feed-based administration of Lactobacillus plantarum enhances growth, haematological and immunological responses in Cyprinus carpio

The effect of probiotic bacteria (Lactobacillus plantarum isolate FLB1) on the immunological responses of Cyprinus carpio (Common carp) was tested in a feeding trial. Fingerlings were divided into five treatment groups and fed with graded probiotic diets [@ 0 (T1), 106 colony-forming unit (CFU)/g (T2), 107 CFU/g (T3), 108 CFU/g (T4), and 109 CFU/g (T5) of test meal] in triplicate. Fish fed probiotic supplemented diets had significantly (P < 0.05) improved growth performance, haematological parameters, blood Haemoglobin (Hb), Total Erythrocyte Count (TEC), Total Leucocyte Count (TLC), Packed Cell Volume (PCV)/ Hematocrit (Ht), Mean Corpuscular Haemoglobin (MCH), and Mean Corpuscular Haemoglobin Concentration (MCHC), with the greatest improvement in the treatment T5. Significant (P < 0.05) increase in total serum protein and globulin, as well as lower Albumin (Alb)/ Globulin (Glb) ratio values were also reported. These values indicated the overall improvements in non-specific immune markers. In all the probiotic fed fish groups, significantly (P < 0.05) reduced values of blood glucose levels, total cholesterol, Triglycerides (TG), and High-Density Lipoprotein (HDL) were observed. Increased numbers of lactic acid bacteria (LAB) were detected in the gastrointestinal tract of probiotic-fed groups indicating the viability and survival of L. plantarum in the fish gut. The results showed that supplementing the diet with L. plantarum @ 109CFU/g feed during the growth period of the fish in a semi-intensive culture system could improve growth and health status of C. carpio.

The Effect of Probiotics on Various Diseases and their Therapeutic Role: An Update Review

Probiotic bacteria play a critical and functional role in clinical and nutritional applications. In the present study, the ability of various probiotics and their metabolites in the prevention and treatment of different diseases, infections and disorders was reviewed. The issues that were noticed are included: Fibrocystic, diabetes, acne, colon cancer, cardiovascular, urinary tract infections, atopic eczema syndrome, food allergies and obesity. Enhancement in using drug treatment has led to the appearance of drug-resistance concern, thus probiotics can be a suitable choice. This review focuses on the effect of probiotic bacteria and their metabolites on immune-boosting, prevention and treatment of these diseases. For this purpose, after a short glance at each disease, infection and disorder, the mechanism of probiotic action and recent studies about that disease are reviewed. It could be recommended that probiotics consumption, perhaps from birth to all stages of life, would be effective in the life-long, development of health effects and disease treatments.

Inhibition of Clostridium (C.) botulinum and its toxins by probiotic bacteria and their metabolites: An update review

Clostridium (C.) botulinum is the causative agent of foodborne poisoning such as botulism, which includes high mortality rates in animals and humans. Probiotic bacteria play critically functional roles in food matrices, as well as agricultural, clinical and nutritional fields. In this review, potentials of various probiotic bacteria and their metabolites to prevent C. botulinum toxicity are reviewed. For this purpose, an introduction about C. botulinum and its mechanisms of action is provided. After a short introduction of probiotic bacteria and their beneficial health effects on humans, the bacterial mechanisms of their action are reviewed. Then bacteriocin production by probiotic bacteria is described. After description of C. botulinum and its neurotoxins, effects of probiotic bacteria on C. botulinum are reviewed with a special focus on effects of the bacterial bacteriocins on this pathogen. Furthermore, physicochemical factors, which show great effects on potential of nisin to prevent growth and toxin production of the bacteria, are introduced. This study has shown that probiotic bacteria and their bacteriocins can be effective on growth, toxin formation and toxicity of C. botulinum. In conclusion, probiotic use in food safety studies can be effective in preventing or treating toxicity of C. botulinum.

Effects of probiotic bacteria of Lactobacillus acidophilus and Lactobacillus casei on aflatoxin B1 detoxification within a simulated gastrointestinal tract model

The present study was objected to assess the ability of lactic acid bacteria in the reduction of aflatoxin B1 (AFB1) in a simulated human gastrointestinal tract. In this work, eight treatments were prepared to compare the ability of noted probiotic bacteria in AFB1 reduction in different simulated gastrointestinal conditions. The results of the present study revealed a significant reduction in AFB1 in all of the treatments in which the highest reduction rate was observed by L. acidophilus in the absence of sterilized milk. Moreover, the findings showed that bacteria, gastric juice, and small intestine may contribute to the reduction of AFB1.

GABA potentiate the immunoregulatory effects of Lactobacillus brevis BGZLS10-17 via ATG5-dependent autophagy in vitro

The characterization of mechanisms involved in the positive effects of probiotic bacteria in various pathophysiological conditions is a prerogative for their safe and efficient application in biomedicine. We have investigated the immunological effects of live bacteria-free supernatant collected from GABA-producing Lactobacillus brevis BGZLS10-17 on Concanavalin A-stimulated mesenteric lymph node cells (MLNC), an in vitro model of activated immune cells. We have shown that GABA containing and GABA-free supernatant of Lactobacillus brevis BGZLS10-17 have strong immunoregulatory effects on MLNC. Further, GABA produced by this strain exhibit additional inhibitory effects on proliferation, IFN-γ and IL-17 production by MLNC, and the expression of MHCII and CD80 on antigen presenting cells. At the other hand, GABA-containing supernatants displayed the strongest stimulatory effects on the expression of immunoregulatory molecules, such as Foxp3+, IL-10, TGF-β, CTLA4 and SIRP-α. By looking for the mechanisms of actions, we found that supernatants produced by BGZLS10-17 induce autophagy in different MLNC, such as CD4+ and CD8+ T lymphocytes, NK and NKT cells, as well as antigen presenting cells. Further, we showed that the stimulation of Foxp3+, IL-10 and TGF-β expression by BGZLS10-17 produced GABA is completely mediated by the induction of ATG5 dependent autophagy, and that other molecules in the supernatants display GABA-, ATG5-, Foxp3+-, IL-10- and TGF-β- independent, immunoregulatory effects.

The effect of probiotic bacteria in culture media using organic fertilizer for population density, biomass production and nutrient quality of Phronima sp . as natural feed

Phronima sp. is a natural feed with high nutrient content, and it can be an alternative substitute for Artemia. The existence of probiotic bacteria in culture media aims to improve water quality through biodegradation, maintain microbial balance and control pathogenic bacteria, so that nutrients in culture media can be utilized by Phronima sp. This study aimed to determine the effect of fermented organic fertilizer by probiotic bacteria in mass culture media on the population density, biomass production and nutrient content of Phronima sp. and to find the best culture media composition for mass culture of Phronima sp. The study was conducted with a completely randomized design with five treatments and three replications. The treatments were as follows: A: 0% chicken manure, 50% rice bran, 50% tofu waste; B: 25% chicken manure, 37.5% rice bran, 37.5% tofu waste; C: 50% chicken manure, 25% rice bran, 25% tofu waste; D: 75% chicken manure, 12.5% rice bran, 12.5% tofu waste and E: 100% chicken manure. The test animals were Phronima sp., and during cultivation, fertilization was carried out every 3 days. Population density and biomass were observed, and proximate analysis, amino acid profile analysis and fatty acid profile analysis were performed. The highest population density was at treatment B, with a peak population of 98 individuals/L that occurred on the 16th day of maintenance culture, and the highest biomass of 0.51 g was at the same treatment. The highest nutrient content was obtained at treatment B, for which the highest protein proximate analysis value was 58.90%, the proportion of the fatty acids comprised of eicosapentaenoic acid was 7.53%, and lysine amino acids were found at 44.16 ppm.

Effect of Probiotic Bacteria on Legumes Purees

In this study three common legumes (Fababean, Chickpea and Soybean) were fermented using probiotic bacteria after isolated and identified by morphological, biochemical and molecular characteristics. The cultures containing Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium lactis as probiotic bacteria were used to study their effect on the properties of some food products prepared from legumes. Results revealed that P Soy bean (puree) recorded the highest scores in appearance (8.5 ± 0.2), acceptable odor and mouth feeling scores were (8.4 ± 0.2) respectively .However P Faba bean (puree) and P Chickpea (puree) had the best color (8.5 ± 0.2). While,taste was the best for P Faba bean (8.2 ± 0.2). Results indicated that the highest values of chemical parameters were recorded for P Faba bean in moisture and ash which were (4.7±0.1, (4.6±0.2) respectively. At the same time, P Soy bean in lipids had (19.0±0.0), and in protein was (39.3±0.1).Results obtained of P Chickpea in carbohydrates was( 62.6±0.1). Also, results obtained shows the effect of all three bacterial isolates (Lactobacillus acidophilus , Bafidobacterhum lactis and Strentococcus thermophilus ) as probiotic -fermented Faba bean, Chick pea and Soy bean. shows that ten storage days value at (5.5 ± 0.3) for the Soy bean (5.5± 0.3)for the Faba bean and (5.8 ± 0.5) for the Soy bean. Finaly the results concluded that the counts of probiotic bacterial isolates were improved the sensory and chemicaly evaluation of fermentated legumes when increased.

Evaluation of the effect of probiotic bacteria on growth performance and survival rate of carp, Cyprinus carpio

The objective of this research was to obtain probiotic bacteria isolated from the digestive tract of catfish Clarias batracus, to evaluate its effect on growth performances and survival rate of carp. After purified, the bacteria was identified through biochemical tests and selection was carried out through proteolytic, amylolytic, cellulolytic, tests as well as pathogenicity test. Probiotic candidate was administered orally to fish at different concentrations. The fish were fed for 30 days respectively as much as 3% of body weight per day with feeding frequency of two times a day. The results showed that addition of probiotics into feed significantly increased growth performance but not survival rate.Keywords: Probiotic, Clarias batracus, growth performance, aquaculture. ABSTRAKTujuan dari penelitian ini adalah untuk mendapatkan bakteri probiotik yang diisolasi dari saluran pencernaan ikan lele Clarias batracus, dan mengevaluasi pengaruhnya terhadap kinerja pertumbuhan dan kelangsungan hidup benih ikan mas. Bakteri probiotik diidentifikasi melalui uji biokimia dan seleksi dilakukan melalui proteolitik, amilolitik, tes selulolitik dan patogenisitas. Kandidat probiotik diberikan pada ikan secara oral dengan konsentrasi berbeda Pemberian pakan berprobiotik berlangsung selama 30 hari sebanyak 3% dari berat badan per hari dengan frekuensi pemberian dua kali sehari. Hasil penelitian menunjukkan bahwa penambahan probiotik ke dalam pakan secara signifikan meningkatkan kinerja pertumbuhan namun tidak memengaruhi kelangsungan hidup benih ikan mas.Kata kunci : Probiotik, Clarias batracus, kinerja pertumbuhan, budidaya

Effect of probiotic bacteria in composition of children's saliva

Probiotics are widely used in the food industry and may affect the oral microbiota. This study aimed to evaluate the effect of petit-suisse plus probiotic on the microbiota of children's saliva. Strawberry flavor petit-suisse cheese plus green banana flour without Lactobacillus casei (control) and with addition of Lactobacillus casei (probiotic) were prepared and used in the experiments. The feasibility of adding probiotic to petit-suisse was assessed over 28 days. Saliva samples from volunteers who consumed the petit-suisses produced were collected before the period of consumption of the products (T0), on the last day of consumption of petit-suisse (T1) and two weeks after the consumption stopped (T2 - post treatment). The samples were immediately fixed with paraformaldehyde in a final concentration of 2%, after which the bacteria of interest were identified by Fluorescent In Situ Hybridization (FISH) and counted with the aid of an epifluorescence microscope. Lactic bacteria were viable between 108 and 109 CFU·g−1 for 28 days of storage at 4 °C. Both products (control and with L. casei added) were able to significantly reduce (p < 0.05) the total number of microorganisms and Streptococcus mutans in the volunteers' saliva. However, only the product plus L. casei decreased density of Agreggatibacter actinomycetemcomitans, as well as being able to maintain the low density of Porphyromonas gingivalis post treatment. Therefore, the petit-suisse cheese developed showed the ability to carry probiotic microorganism, being a potential alternative for reducing potentially pathogenic microbiota in the oral cavity.

Probiotics for life – Part I general health perspectives

Scientific evidence is expanding continuously during the past few decades, pointing to the beneficial effects of probiotic bacteria on human health. Food industry has been very active in studying and promoting various probiotic products. The WHO has advocated the reduced use of antibiotics and microbial interference therapy (use of nonpathogens to eliminate pathogens), observing the increasing development of antimicrobial resistance. Initially, probiotics were traditional used for the treatment of gastrointestinal disorders and allergies; later, there has been an expansion in therapeutic uses of these agents in urogenital infections, carcinomas, etc. Due to the increasing mountain of literature on this topic, brief user-friendly information is provided in lucid language for busy clinicians. This Part I article gives an overview of the history and properties of probiotics, mechanisms of action, and uses to promote general health. The Part II article, coming in next issue of this journal, highlights on oral health. To understand oral effects of probiotics, their medical background knowledge is essential.
