Magneto-thermal-acoustic imaging with magnetic nanoparticles (MNPs) is a novel medical imaging modality based on the thermoacoustic (TA) effect of MNPs. We proposed a new dual-modality imaging strategy that combines the TA and echo signals. Based on the synthetic aperture method, we used TA signals to image cancer tissues labeled with MNPs and applied echo signals to achieve tissue structure imaging. In the simulation, we constructed a breast cancer model and assumed that cancer tissue was labeled with MNPs. Under a limited field of view, the TA and echo signals were collected using a multi-element transducer array. By fusing the imaging results of the TA and echo signals, we obtained a boundary distribution map of the breast cancer model, and accurately located the tumor tissue. In this experiment, we built a multi-channel acquisition system. Pork tissue labeled with MNPs was used as the breast cancer phantom. We collected a total of 120 TA and echo signals at 1 mm intervals using a multi-element transducer array and faithfully imaged the boundary distribution map of the phantom. Compared with the traditional linear back projection method based on focused transducers, we demonstrated that the proposed method not only improves the lateral resolution of the image, but also makes accurate cancer identification easier. This provides a promising medical imaging technology for clinical applications.

Effects of Magnetic Nanoparticles on the Functional Activity of Human Monocytes and Dendritic Cells.

The use of nanoparticles in medicine is sometimes hampered by their potential to activate immune cells, eliciting inflammation or allergy. We investigated whether magnetic nanoparticles (MNPs) or biomimetic magnetic nanoparticles (BMNPs) affect relevant activities of human monocytes. We found that the nanoparticles neither elicited the production of pro-inflammatory mediators IL-6 and TNFα by resting monocytes (when BMNP dose < 300 μg/mL) nor enhanced their secretion induced by R848, a molecule engaging virus-recognizing receptors, or bacterial lipopolysaccharide (LPS). MNPs and BMNPs neither induced the generation of reactive oxygen species (ROS), nor affected the ROS production elicited by the NADPH oxidase activator phorbol myristate acetate (PMA) or the fungal derivative β-glucan. BMNPs, but not MNPs, caused an up-regulation of the maturation markers CD80, CD83, and CD86 in immature monocyte-derived dendritic cells (DCs), whereas both nanoparticles did not affect the LPS-induced expression of these markers. Moreover, the nanoparticles were greedily ingested by monocytes and DCs without altering their viability. Therefore, these nanoparticles are candidates for medical applications because they do not activate pro-inflammatory activities of monocytes. Furthermore, their ability to stimulate DC maturation could be used for the design of vaccines. Moreover, harmlessly engulfed nanoparticles could be vehicles to carry molecules inside the immune cells to regulate the immune response.

Effect of Magnetic Nanoparticles on Hormone Level Changes During Perimenopausal Period and Regulation of Bone Metabolism.

This work aimed to explore the effect of nerve magnetic stimulation based on superparamagnetic Fe3O4 nanoparticle (NP) on bone metabolism during the perimenopausal period. First, the multifunctional water-soluble polymer PTMP-PMAA was utilized as the ligand. PTMP-MAA@ Fe3O4 NP with high magnetization was prepared by the co-precipitation method, and NPX diffraction pattern analysis and in vitro stability analysis were implemented. Then, NPs were co-cultured with 293T cells, and the cytotoxicity was detected by the CCK-8 method. Subsequently, 3-month-old female young SD rats and 11~15-month-old natural menopausal SD rats were taken as the research objects. According to the vaginal smear, the rats were randomly rolled into a young control, perimenopausal period model, estrogen treatment, and osteoporosis prevention groups. Rats in the estrogen treatment group were given Premarin suspension by gavage. Rats in the osteoporosis prevention group were injected stereotaxically with PTMP-MAA@ Fe3O4 NP suspension, and a rotating magnetic field was applied to the brain for nerve magnetic stimulation. The rats were sacrificed three days after treatment and brain tissues were taken for pathological analysis. Rat humerus was weighted and dual-energy X-ray was utilized to determine bone density and bone mineral content. Serum was collected and radioimmunoassay and ELISA were employed to detect estradiol (E2), osteocalcin (Boneglaprotein, BGP), oxytocin (OT), bone alkaline phosphatase (BALP), type I collagen carboxy-terminated cross-linked peptide (CTX-I), and tartrate-resistant acid phosphatase (TRACP-5b) in the serum of rats in each group. The results showed that PTMP-MAA@ Fe3O4 NP had good biocompatibility, and the CCK-8 test results showed that PTMP-MAA@ Fe3O4 NP had low cytotoxicity. Compared with the young control group, the humeral dry weight, wet weight, bone density, and bone mineral content, serum E2, OT, and BGP content in the perimenopausal period model group were reduced, while the serum BALP, CTX-I, and TRACP -5b content was increased (P<0.05). It was verified that nerve magnetic stimulation based on PTMP-MAA@ Fe3O4 NP increased the serum estrogen level of female rats during the perimenopausal period, increased the bone density of rats, promoted bone formation, and regulated bone metabolism.

Microalgae liquefaction in ethanol to produce high-quality fuels: Effect of magnetic nanoparticles on nitrogen transformation

As the representative of the 3rd generation feedstock of biodiesel, microalgae show extraordinary advantages for energy demands. Magnetic separation is a proven and effective method for microalgae harvesting. This study aims to investigate the effect of unrecovered magnetite-based nanoparticles (Fe3O4 NPs) after microalgae harvesting on the bio-oil quality, especially nitrogen content in the subsequent production. Co-liquefaction of Chlorella Vulgaris mixed with three residual amounts (1 wt%, 5 wt%, and 10 wt% of microalgae addition) Fe3O4 NPs in ethanol at 220 and 260 °C were performed. Ethanol can provide hydrogen in situ during liquefaction, and the catalytic activity of Fe3O4 to hydrogenation reaction enhances the yield and quality of bio-oil. At 260 °C and 5 wt% NPs dosage, the bio-oil yield can reach up to 79.71%. The addition of NPs reduces the relative N content of bio-oil by more than 16.90%. GC/MS analysis shows that the nitrogen heterocyclic compounds are significantly reduced about 60%; instead, hydrocarbons are increased in the bio-oil. It implies that Fe3O4 NPs are beneficial to the denitrogenation of bio-oil during microalgae liquefaction in ethanol.

The effect of magnetic nanoparticles containing hyaluronic acid and methotrexate on the expression of genes involved in apoptosis and metastasis in A549 lung cancer cell lines

Lung cancer has been shown to be resistant to treatment with some chemotherapy drugs due to epithelial-mesenchymal transmission (EMT). Because the rate of cytotoxicity and induction of apoptosis by methotrexate (MTX) is negligible in A549 lung cancer cells, a CD44 positive cell line, we decided to synthesize magnetic nanoparticles (MNPs) containing hyaluronic acid (HA) and MTX to evaluate the effect of CD44 receptor targeting on the expression of genes involved in apoptosis. The TNF genes can modulate the expression of CD44 and implicate carcinogenesis and metastases. Therefore, inhibition of the TNF gene and study of its interaction with the CD44 receptor can determine the success of a treatment method. The results of the MTT assay confirmed that the MNPs-HA-MTX offered better cellular cytotoxic effects on cell viability than free MTX. The real-time PCR test also showed that the Bak1/Bclx ratio was 52.5 times higher than the control. On the other hand, the expression of the TNF gene was severely reduced, which could be due to the binding of HA-moiety of the MNPs-HA-MTX to the receptor and endocytosis. All the results gave us hope that we could increase the effectiveness of methotrexate in lung cancer by targeting the CD44 receptor.

Synthesis of magnetic nanoparticles and their effects on growth and physiological parameters of Calotropis procera seedlings.

The present study was carried out to elucidate effects of synthesized magnetic nanoparticles (MNPs) on morphological and physiological parameters and main essential oil components of Calotropis procera seedlings. For this purpose, 21-day-old seedlings grown under hydroponic conditions were treated by the different MNP concentrations (0, 50, 100, 150, and 200 mg L-1). The results showed that the growth parameters, chlorophyll pigments, soluble sugars, and total proteins significantly increased in leaf under MNP treatment, except for the root length. As compared to the control, MNPs induced a substantial change in the activities of antioxidant enzymes, H2O2, and malondialdehyde contents. Ascorbate peroxidase activity showed a meaningful increase in leaf treated with 200 mg L-1 MNPs, while superoxide dismutase activity and concentration of H2O2 conspicuously decreased relative to the control. Moreover, MNPs enhanced geranial, 1,8-cineol, a-phellandrene, citronellal, camphor, and terpinen-4-ol contents as major components. These results suggest that MNPs could be a promising method of iron application in agricultural systems. Regarding the effects of MNPs, 200-mg L-1 MNPs were most effective on the production of main essential oils and plant growth that could serve as a favorable elicitor for plant improvement.

Effects of Magnetite Nanoparticles and Static Magnetic Field on Neural Differentiation of Pluripotent Stem Cells.

Neurodevelopmental processes of pluripotent cells, such as proliferation and differentiation, are influenced by external natural forces. Despite the presence of biogenic magnetite nanoparticles in the central nervous system and constant exposure to the Earth's magnetic fields and other sources, thereisscant knowledge regarding the role of electromagnetic stimuli in neurogenesis. Moreover, emerging applications of electrical and magnetic stimulation to treat neurological disorders emphasize the relevance of understanding the impact and mechanisms behind these stimuli. Here, the effects of magnetic nanoparticles (MNPs)in polymeric coatings and the static external magnetic field (EMF) were investigated on neural induction of murine embryonic stem cells (mESCs) and human induced pluripotent stem cells (hiPSCs). The resultsshowthat the presence of 0.5% MNPs in collagen-based coatings facilitates the migration andneuronal maturationof mESCs and hiPSCs in vitro. Furthermore, theapplication of 0.4 TeslaEMF perpendicularly to the cell culture plane, discernibly stimulates proliferationand guide fate decisions of thepluripotent stem cells, depending on the origin of stem cells and their developmental stage. Mechanistic analysis revealsthat modulation of ionic homeostasis and theexpression of proteins involved in cytostructural,liposomaland cell cycle checkpoint functions provide a principal underpinning for the impact of electromagnetic stimuli on neural lineage specification andproliferation. These findings not only explore the potential of the magnetic stimuli as neural differentiation and functionmodulator but also highlight the risks that immoderate magnetic stimulation may affect more susceptible neurons, such as dopaminergic neurons.

MAGNETIC NANOPARTICLES AND IONIC LIQUID EFFECT ON ELECTROCHEMICAL SENSORS PERFORMANCE

Magnetic nanoparticles and ionic liquid (IL, 1-hexyl-3-methyl-imidazolium bromide) based on graphene oxide (GO) composite provide unique physical and chemical properties in electrochemical sensors performance. Magnetic nanoparticles can cover active sites that increase chemical reactions with easy separation. IL increases the scan rate of electron transfer between the modified electrode and solution because it includes conductive adhesion properties. Also, IL in the next steps of design carbon paste electrodes (CPE) increases the cohesion. The study aims are to study the effects of magnetic nanoparticles and IL on the electrochemical detection of dopamine (DA). DA has a vital role in the mammalian central nervous system and change its value from the standard range leads broad mental diseases. But magnetic graphene oxide (MGO) may not exist capable of enhancing electrochemical signs alone. In this regard, after the synthesis of MGO, IL was established on composite. Then gold nanoparticles (AuNPs) and molecularly imprinted polymer (MIP) were modified MGO nanocomposite. MIP polymerization was continued by methacrylic acid (MAA) in the presence of DA as a template molecule. The developed sensor with modified nanocomposite studied by cyclic voltammetry (CV) and differential pulse voltammetry (DPV) techniques. The modified sensor based on nanocomposite with a broad concentration linear range, between 1×10-7 to 1×10-4 M and a limit of detection of 1×10-8 M (S/N=3) was used for the detection of DA in biological samples. Furthermore, these results prove that MGO was improved active sites of surface nanocomposite and IL was increased conductivity in the based electrochemical sensor for DA detection.

Kosmotropic and chaotropic effect of biocompatible Fe3O4 nanoparticles on egg white lysozyme; the key role of nanoparticle-protein corona formation

Accurate understanding of the interaction between different types of nanoparticles and proteins is very important in various fields, especially in Nano-biotechnology. Concentration-dependent nanoparticles can cause structural changes in proteins. There are several procedures to increase the efficiency of nanoparticles in these interactions. In this study, the effect of magnetic nanoparticles (MNPs) including Fe3O4 and Fe3O4@RA (Rosmarinic acid coated with Fe3O4) was estimated on the stability and activity of egg white Lysozyme (EWL). The synthesis of MNPs was done using co-precipitation method; then they were characterized by X-ray diffraction (XRD), Fourier-transform infrared (FT-IR), vibrating sample magnetometer (VSM), CHNS elemental analysis, dynamic light scattering (DLS) and field emission scanning electron microscopy (FE-SEM). The results of fluorescence analysis showed that both MNPs can have two opposite effects on the tertiary structure of EWL, depending on their concentration. Accordingly, at the concentrations below a concentration threshold (0.0083 and 0.0067 for Fe3O4 NPs and Fe3O4@RA, respectively), they improve the structure of the EWL (kosmotropic-like effect) and at the concentrations above it, they gradually cause its degradation (chaotropic-like effect). Changes in the protein activity against Micrococcus luteus bacteria based on UV analysis data were also fully consistent with the fluorescence analysis. In order to study the secondary structure of EWL, CD Spectra showed an increase in the helicity of EWL after interaction with both MNPs (3.7% and 1.1% increase in the helicity for 0.02 and 0.1 mg.mL−1 of Fe3O4 respectively, and also 5.5% and 2% for 0.02 and 0.1 mg.mL−1 of Fe3O4@RA respectively) and a decrease in this factor after interaction with pure RA (0.9% and 4.1% decrease in the helicity for 0.02 and 0.1 mg.mL−1 of pure RA respectively).

Effect of magnetic nanoparticles containing ropivacaine on ankle nerve block in rats

Ropivacaine (RVC), a common pain management drug used for clinical anesthesia and postoperative analgesia, inhibits peripheral nociceptive pain stimulation. However, the potential neurological damage resulting from RVC use must be considered. Developing a strategy to enhance the local anesthetic effect of RVC while reducing its potential acute toxicity to the central nervous system is urgently needed. In this study, a novel RVC nanocomposite drug, magnetic iron oxide/polyethylene glycol-carboxymethyl chitosan/ropivacaine nanoparticle (mCMCS-PEG/RVC NPs), was synthesized with magnetic iron oxide. The inherent shell–core structure of mCMCS-PEG retained core magnetic properties, improved the stability and biocompatibility of magnetic nanoparticles, and avoided excessive degradation. Thus, mCMCS-PEG/RVC NPs are expected to provide a new pain management strategy for perioperative patients.

Non-Thermal Intervention of Lung Tumor by Core-Shell Magnetic Nanoparticles in a Magnetic Field

K-Ras mutations result in normal cells dividing uncontrollably and becoming cancerous. The prognosis is currently poor for patients due to the lack of drugs that can effectively target these mutations. In this study, magnetic nanoparticles (MNPs) were prepared, characterized, and cooperated with a magnetic field to intervene in the growth of lung tumor cells. The rise in temperature of a stimulation coil was studied by numerical calculation. The non-thermal effects of MNPs under a magnetic force were analyzed. The cell experiments showed that the growth of A549 tumor cells slowed down. The result of a wound-healing assay also indicated that the migration of tumor cells was suppressed. Compared with magnetic stimulation without MNPs, MNPs enhanced the inhibitory effects of a magnetic field. This study suggests a new way to treat K-Ras driven lung tumors using non-thermal effects of MNPs without the side effects caused by thermal effects.

Effective production of multifunctional magnetic-sensitive biomaterial by an extrusion-based additive manufacturing technique

A calcium phosphate (CaP)-based scaffold used as synthetic bone grafts, which smartly combines precise dimensions, controlled porosity and therapeutic functions, presents benefits beyond those offered by conventional practices, although its fabrication is still a challenge. The sintering step normally required to improve the strength of the ceramic scaffolds precludes the addition of any biomolecules or functional particles before this stage.This study presents a proof of concept of multifunctional CaP-based scaffolds, fabricated by additive manufacturing from an innovative ink composition, with potential for bone regeneration, cancer treatment by local magnetic hyperthermia and drug delivery platforms. Highly loaded inks comprising iron-doped hydroxyapatite and β-tricalcium phosphate powders suspended in a chitosan-based solution, in the presence of levofloxacin (LEV) as model drug and magnetic nanoparticles (MNP), were developed. The sintering step was removed from the production process, and the integrity of the printed scaffolds was assured by the polymerization capacity of the ink composite, using genipin as a crosslinking agent. The effects of MNP and LEV on the inks’ rheological properties, as well as on the mechanical and structural behaviour of non-doped and iron-doped scaffolds, were evaluated. Magnetic and magneto-thermal response, drug delivery and biological performance, such as cell proliferation in the absence and presence of an applied magnetic field, were also assessed. The addition of a constant amount of MNP in the iron-doped and non-doped CaP-based inks enhances their magnetic response and induction heating, with these effects more pronounced for the iron-doped CaP-based ink. These results suggest a synergistic effect between the iron-doped CaP-based powders and the MNP due to ferro/ferrimagnetic interactions. Furthermore, the iron presence enhances human mesenchymal stem cell metabolic activity and proliferation.

In vitro and ex vivo relaxometric properties of ethylene glycol coated gadolinium oxide nanoparticles for potential use as contrast agents in magnetic resonance imaging

Magnetic nanoparticles (MNPs) have widely demonstrated their applicability in many biomedical applications including magnetic resonance imaging (MRI), hyperthermia, and drug delivery. However, the effectiveness of MNPs can be limited for in vivo applications due to their hydrophobic surfaces leading to nanoparticle agglomeration and thus requires appropriate surface modification to enhance colloidal stability. Glycols are widely used coating material for surface modifications of MNPs to improve their physicochemical properties and biocompatibility. The present work reports the preparation of two different sized ethylene glycol coated gadolinium oxide nanoparticles (EG@Gd2O3 NPs) using two different synthesis approaches and their applicability as contrast agents in MRI. Thermo-gravimetric analysis and Fourier transform infrared spectroscopy confirmed the successful coating of ethylene glycol on the surface of NPs. We found that independent of the size of NPs, the globular shaped EG@Gd2O3 NPs exhibited similar crystal structures, magnetic properties, and cellular cytotoxicity behavior. However, a significant impact of size on MRI contrast enhancement properties was seen. It was revealed that the relaxivity of EG@Gd2O3 NPs increases with a decrease in particle size. Small sized EG@Gd2O3 NPs (∼12 nm) exhibited a high specific in vitro and ex vivo longitudinal relaxivity of 3.7 and 1.5 mM−1 s−1, respectively, thus clearly elucidating the potential of these NPs for use as local contrast enhancement agents. The present study gives insights into the intrinsic dependence of magnetic resonance contrast effects of NPs on particle size and surface coating layer mass ratio and thus demonstrates the development of efficient magnetic nanoparticles based contrast agents by fine tuning of particle size and surface properties.

Magnetofection: Magic magnetic nanoparticles for efficient gene delivery

Magnetic nanoparticles (MNPs) have become a research hotspot and widely used in the biomedical field in recent decades due to their unique magnetic properties. This minireview summarizes the specific gene transfection of magnetic particles (magnetofection) during eversy dynamic process of gene delivery (gene binding, cellular uptake, endosomal escape, intracellular trafficking and in vivo targeting). Meanwhile, the synergistic biomedical application of magnetofection and the effects of MNPs have also been discussed, including magnetic resonance imaging (MRI), magnetic mediated hyperthermia (MMH), Fenton reaction and autophagy. Finally, the clinical prospect of magnetofection was briefly expected.

Analytical and Numerical Model of High Intensity Focused Ultrasound Enhanced With Nanoparticles.

High intensity focused ultrasound (HIFU) is a new noninvasive therapeutics that allows local treatment of solid tumors through a hyperthermal mechanism using ultrasonic energy. One promising strategy to increase the thermal efficiency of HIFU is to employ nanoparticles (NPs) as ultrasound agents for the hyperthermia procedure. However, the interaction mechanism between NPs and ultrasonic waves has not been well understood. In an effort to investigate the heating process of NPs-enhanced HIFU, we derived a set of HIFU equations governing the temperature variation during the thermal ablation based on the principle of conservation of energy for heat transfer mechanism. A numerical model was developed to solve the HIFU equations to simulate the absorption mechanism of HIFU in the presence of NPs, the consequent heat transfer process, and the temperature rise profile during the sonication period. The accuracy of numerical model was verified by performing a series of experiments on tissue-mimicking phantoms embedded with magnetic NPs (MNPs). The transport processes taking place at the boundaries between NPs and surrounding medium played the major role in the temperature rise during HIFU sonication. Besides, the effects of MNPs on rising temperature were improved by amplifying the ultrasonic power and frequency as well as by increasing the MNP concentration. A quantitative comparison with experimental results demonstrated the potential of the numerical model to accurately predict the heating mechanism of HIFU mediated by NPs. The proposed method can help with simulation of HIFU when NPs are employed as ultrasound agents.

The role of faceting and elongation on the magnetic anisotropy of magnetite Fe3O4 nanocrystals

Fe3O4 nanoparticles are one of the most promising candidates for biomedical applications such as magnetic hyperthermia and theranostics due to their bio-compatibility, structural stability and good magnetic properties. However, much is unknown about the nanoscale origins of the observed magnetic properties of particles due to the dominance of surface and finite size effects. Here we have developed an atomistic spin model of elongated magnetite nanocrystals to specifically address the role of faceting and elongation on the magnetic shape anisotropy. We find that for faceted particles simple analytical formulae overestimate the magnetic shape anisotropy and that the underlying cubic anisotropy makes a significant contribution to the energy barrier for moderately elongated particles. Our results enable a better estimation of the effective magnetic anisotropy of highly crystalline magnetite nanoparticles and is a step towards quantitative prediction of the heating effects of magnetic nanoparticles.

Effect of magnetic nanoparticles and silver-loaded magnetic nanoparticles on advanced wastewater treatment and disinfection

BackgroundStrict discharge standards necessitate the advanced treatment and disinfection of wastewater treatment plant (WWTP) effluents for environmental protection and water reuse purposes. This study aimed to apply magnetic nanoparticles (MNPs) and silver-loaded magnetic nanoparticles (Ag-MNPs) in the advanced treatment and disinfection of the secondary effluent from a municipal WWTP. MethodsThe as-prepared nanoparticles were employed under various conditions of nanoparticle dose, contact time, and effluent contamination level. Total coliforms (TC), fecal coliforms (FC), heterotrophic bacteria (HB), and chemical oxygen demand (COD) were examined in the control and experimental samples to determine the treatment effectiveness. ResultsThe findings showed that MNPs and Ag-MNPs were successfully synthesized with a mean size of 41 and 34 nm and the saturation magnetization of 62 and 67 emu/g, respectively. The results of advanced wastewater treatment showed that 105 mg/L nanoparticle dose, 70 min contact time, and 55% contamination level led to 0.65 TC, 0.48 FC, and 0.58 HB log reductions and 30.40% COD removal, on average. Increasing nanoparticle dose and contact time and decreasing contamination level enhanced the treatment efficacy. Switching from MNPs to Ag-MNPs significantly resulted in a 0.06 increase in TC, FC, and HB log reductions and a 6.16% increase in COD removal. ConclusionBased on the findings, MNPs and Ag-MNPs are promising in the advanced wastewater treatment and disinfection.

Exploring the potential of the dynamic hysteresis loops via high field, high frequency and temperature adjustable AC magnetometer for magnetic hyperthermia characterization

Aim The Specific Absorption Rate (SAR) is the key parameter to optimize the effectiveness of magnetic nanoparticles in magnetic hyperthermia. AC magnetometry arises as a powerful technique to quantify the SAR by computing the hysteresis loops' area. However, currently available devices produce quite limited magnetic field intensities, below 45mT, which are often insufficient to obtain major hysteresis loops and so a more complete and understandable magneticcharacterization. This limitation leads to a lack of information concerning some basic properties, like the maximum attainable (SAR) as a function of particles' size and excitation frequencies, or the role of the mechanical rotation in liquid samples. Methods To fill this gap, we have developed a versatile high field AC magnetometer, capable of working at a wide range of magnetic hyperthermia frequencies (100 kHz – 1MHz) and up to field intensities of 90mT. Additionally, our device incorporates a variable temperature system for continuous measurements between 220 and 380 K. We have optimized the geometrical properties of the induction coil that maximize the generated magnetic field intensity. Results To illustrate the potency of our device, we present and model a series of measurements performed in liquid and frozen solutions of magnetic particles with sizes ranging from 16 to 29 nm. Conclusion We show that AC magnetometry becomes a very reliable technique to determine the effective anisotropy constant of single domains, to study the impact of the mechanical orientation in the SAR and to choose the optimal excitation parameters to maximize heating production under human safety limits.
