Haematological parameters and biochemical indices of African catfish (Clarias gariepinus) exposed to glyphosate-based herbicide (Force up®) for 96 hours

IntroductionGeometric aquaculture growth has resulted in exponentially increasing use of agrochemicals as either parasiticides or herbicides in the aquaculture environment. This study determines some of the toxicological (haematological and biochemical) effects of glyphosate-based herbicides on non-target aquatic animals using Clarias gariepinus as the animal model.MethodSeventy-five apparently healthy adult C. gariepinus (300 g) were sourced from a local farmer and acclimatised for 2 weeks; of these, sixty subjects were divided into four treatment groups (fifteen fish per group and five replicates per unit) by simple randomisation and labelled as T0 (control), T1 (first treatment), T2 (second treatment), and T3 (third treatment). The treatments were replicated thrice. Four concentrations of Force up® [0 mL, 0.15 mL (0.003 mL/L or 5.1 mg/L), 0.225 mL (0.0045 mL/L or 7.65 mg/L), and 0.3 mL (0.006 mL/L or 10.2 mg/L) were added to a 50-L tank of water for T0, T1, T2, and T3, respectively. Approximately 5 mL of blood was collected from the fish in each treatment group 96 h post-exposure for measurement of the blood parameters and biochemical indices using standard analytical methods as well as calculation of the mean values.ResultThe mean values of the packed cell volume, haemoglobin concentration, red blood cell count, and white blood cell count compared to the control group showed an initial increase at T1 but decreased as the glyphosate concentrations increased at T2 (0.0045 mL/L) and T3 (0.006 mL/L). The platelet mean values decreased at T1, increased at T2, and decreased at T3, while the mean values of the corpuscular volume, corpuscular haemoglobin, and corpuscular haemoglobin concentration increased with glyphosate concentration, with the mean corpuscular haemoglobin concentration decreasing at T2. Only the platelet value was statistically significant at a p-value of <0.05 using ANOVA and post hoc Tukey test. The biochemical indices showed decreases in the mean values of aspartate transaminase, blood urea nitrogen, creatinine, and triglycerides at T1, increases at T2, and decreases at T3, while the total protein (g/dL), cholesterol, alanine transaminase, and alkaline phosphatase values showed increases at T1 and decreases at T2 and T3. All these values were not statistically significant based on ANOVA and had p-values >0.05.DiscussionBased on the results of this study, it is deduced that glyphosate-based herbicide (Force up®) has genotoxic, hepatotoxic, and nephrotoxic effects on C. gariepinus even at sublethal doses, with more adverse effects at increasing concentrations.

Effects of glyphosate-based herbicide on gut microbes and hepatopancreatic metabolism in Pomacea canaliculata

Roundup®, a prominent glyphosate-based herbicide (GBH), holds a significant position in the global market. However, studies of its effects on aquatic invertebrates, including molluscs are limited. Pomacea canaliculata, a large freshwater snail naturally thrives in agricultural environments where GBH is extensively employed. Our investigation involved assessing the impact of two concentrations of GBH (at concentrations of 19.98 mg/L and 59.94 mg/L, corresponding to 6 mg/L and 18 mg/L glyphosate) during a 96 h exposure experiment on the intestinal bacterial composition and metabolites of P. canaliculata. Analysis of the 16 S rRNA gene demonstrated a notable reduction in the alpha diversity of intestinal bacteria due to GBH exposure. Higher GBH concentration caused a significant shift in the relative abundance of dominant bacteria, such as Bacteroides and Paludibacter. We employed widely-targeted metabolomics analysis to analyze alterations in the hepatopancreatic metabolic profile as a consequence of GBH exposure. The shifts in metabolites primarily affected lipid, amino acid, and glucose metabolism, resulting in compromised immune and adaptive capacities in P. canaliculata. These results suggested that exposure to varying GBH concentrations perpetuates adverse effects on intestinal and hepatopancreatic health of P. canaliculata. This study provides an understanding of the negative effects of GBH on P. canaliculata and may sheds light on its potential implications for other molluscs.

The effects of glyphosate-based herbicide on the hypothalamic-pituitary thyroid axis are tissue-specific and dependent on age exposure

The significant increase in glyphosate-based herbicide (GBH) use raises concerns about residues in the environment and food, potentially jeopardizing human health. The involvement of GBHs in the increased incidence of thyroid disorders is speculated, since glyphosate has been linked to an increased risk of thyroid disease in farmers. In this sense, this study aims to investigate the potential effects of low levels of GBH exposure (0, 0.5 or 5 mg/kg) from weaning (postnatal day PND23) to adult life (PND60 and PND90) in male Wistar rats on hypothalamic-pituitary-thyroid (HPT) axis function. The serum levels of T4 were increased. The hypothalamus showed reduced expression of Dio2, Thra1, and Thra2. The pituitary showed reduced expression of Mct8 and Dio2 and increased expression of Thra1. The thyroid showed increased expression of Tshr and Thra1. The heart showed increased expression of Mct8 and Myh6. The liver showed reduced expression of Mct8 and Thra2 and increased expression of Thra1. In thyroid morphometry, a decrease in both follicular diameter and area and decreased follicular and colloid diameters and areas were observed. These results suggested that GBH may affect several steps of HPT axis regulation at the transcriptional level in an age-dependent manner and alter the morphometric parameters of the thyroid gland and TH synthesis, with potential repercussions in the TH-target organs.

The effects of short-term glyphosate-based herbicide exposure on insect gene expression profiles

Glyphosate-based herbicides (GBHs) are the most frequently used herbicides worldwide. The use of GBHs is intended to tackle weeds, but GBHs have been shown to affect the life-history traits and antioxidant defense system of invertebrates found in agroecosystems. Thus far, the effects of GBHs on detoxification pathways among invertebrates have not been sufficiently investigated. We performed two different experiments—1) the direct pure glyphosate and GBH treatment, and 2) the indirect GBH experiment via food—to examine the possible effects of environmentally relevant GBH levels on the survival of the Colorado potato beetle (Leptinotarsa decemlineata) and the expression profiles of their detoxification genes. As candidate genes, we selected four cytochrome P450 (CYP), three glutathione-S-transferase (GST), and two acetylcholinesterase (AChE) genes that are known to be related to metabolic or target-site resistances in insects. We showed that environmentally relevant levels of pure glyphosate and GBH increased the probability for higher mortality in the Colorado potato beetle larvae in the direct experiment, but not in the indirect experiment. The GBHs or glyphosate did not affect the expression profiles of the studied CYP, GST, or AChE genes; however, we found a large family-level variation in expression profiles in both the direct and indirect treatment experiments. These results suggest that the genes selected for this study may not be the ones expressed in response to glyphosate or GBHs. It is also possible that the relatively short exposure time did not affect gene expression profiles, or the response may have already occurred at a shorter exposure time. Our results show that glyphosate products may affect the survival of the herbivorous insect already at lower levels, depending on their sensitivity to pesticides.

Effects of Glyphosate-Based Herbicide on Primary Production and Physiological Fitness of the Macroalgae Ulva lactuca

The use of glyphosate-based herbicides (GBHs) worldwide has increased exponentially over the last two decades increasing the environmental risk to marine and coastal habitats. The present study investigated the effects of GBHs at environmentally relevant concentrations (0, 10, 50, 100, 250, and 500 μg·L−1) on the physiology and biochemistry (photosynthesis, pigment, and lipid composition, antioxidative systems and energy balance) of Ulva lactuca, a cosmopolitan marine macroalgae species. Although GBHs cause deleterious effects such as the inhibition of photosynthetic activity, particularly at 250 μg·L−1, due to the impairment of the electron transport in the chloroplasts, these changes are almost completely reverted at the highest concentration (500 μg·L−1). This could be related to the induction of tolerance mechanisms at a certain threshold or tipping point. While no changes occurred in the energy balance, an increase in the pigment antheraxanthin is observed jointly with an increase in ascorbate peroxidase activity. These mechanisms might have contributed to protecting thylakoids against excess radiation and the increase in reactive oxygen species, associated with stress conditions, as no increase in lipid peroxidation products was observed. Furthermore, changes in the fatty acids profile, usually attributed to the induction of plant stress response mechanisms, demonstrated the high resilience of this macroalgae. Notably, the application of bio-optical tools in ecotoxicology, such as pulse amplitude modulated (PAM) fluorometry and laser-induced fluorescence (LIF), allowed separation of the control samples and those treated by GBHs in different concentrations with a high degree of accuracy, with PAM more accurate in identifying the different treatments.

A Glyphosate-Based Herbicide Cross-Selects for Antibiotic Resistance Genes in Bacterioplankton Communities.

ABSTRACTAgrochemicals often contaminate freshwater bodies, affecting microbial communities that underlie aquatic food webs. For example, the herbicide glyphosate has the potential to indirectly select for antibiotic-resistant bacteria. Such cross-selection could occur if the same genes (encoding efflux pumps, for example) confer resistance to both glyphosate and antibiotics. To test for cross-resistance in natural aquatic bacterial communities, we added a glyphosate-based herbicide (GBH) to 1,000-liter mesocosms filled with water from a pristine lake. Over 57 days, we tracked changes in bacterial communities with shotgun metagenomic sequencing and annotated metagenome-assembled genomes (MAGs) for the presence of known antibiotic resistance genes (ARGs), plasmids, and resistance mutations in the enzyme targeted by glyphosate (enolpyruvyl-shikimate-3-phosphate synthase; EPSPS). We found that high doses of GBH significantly increased ARG frequency and selected for multidrug efflux pumps in particular. The relative abundance of MAGs after a high dose of GBH was predictable based on the number of ARGs in their genomes (17% of variation explained) and, to a lesser extent, by resistance mutations in EPSPS. Together, these results indicate that GBHs can cross-select for antibiotic resistance in natural freshwater bacteria.IMPORTANCE Glyphosate-based herbicides (GBHs) such as Roundup formulations may have the unintended consequence of selecting for antibiotic resistance genes (ARGs), as demonstrated in previous experiments. However, the effects of GBHs on ARGs remain unknown in natural aquatic communities, which are often contaminated with pesticides from agricultural runoff. Moreover, the resistance provided by ARGs compared to canonical mutations in the glyphosate target enzyme, EPSPS, remains unclear. Here, we performed a freshwater mesocosm experiment showing that a GBH strongly selects for ARGs, particularly multidrug efflux pumps. These selective effects were evident after just a few days, and the ability of bacteria to survive and thrive after GBH stress was predictable by the number of ARGs in their genomes and, to a lesser extent, by mutations in EPSPS. Intensive GBH application may therefore have the unintended consequence of selecting for ARGs in natural freshwater communities.

Proteomic profiling of royal jelly produced by Apis mellifera L. exposed to food containing herbicide-based glyphosate

Royal jelly (RJ) is rich in protective elements associated with collective immune defenses in the hive of Apis mellifera. Exposure of bees to glyphosate-based herbicides causes ultrastructural changes in the hypopharyngeal glands and a reduction in the production of RJ. However, the effects of glyphosate-based herbicides on the protein composition of RJ and consequences for the hive are unknown. Thus, we performed proteomic profiling of royal jelly produced in hives of A. mellifera exposed to food containing 1,5 μL of Roundup® (2.16 mg. g−1 of glyphosate). The production of RJ was carried out in six hives, following the method of artificial production of queens. The combs containing 80 grafting cells were introduced into the hives, and the collection of royal jelly was performed after 72 h. Two treatments were determined based on hive feeding and the hive as the experimental unit: Control and “Roundup®”. Royal jelly from the Roundup® treatment hives was compared to the Control hives. Proteins with differences in expression were identified by mass spectrometry. Only the proteins present in all three biological replicates were considered in the differential abundance analysis, using Student's t-test (p-value < 0.05, two-tailed). Hives that received food containing Roundup®, analysis showed alterations in protein profile in the RJ produced therein. In total, 24 proteins were identified, and the accumulation of Major royal jelly protein 3 (MRJP3) was downregulated, showing a significant reduction in hives exposed to food containing Roundup® in relation to control hives (t = 0.0017). MRJP3 acts analogously to polyclonal antigen-antibody reactions, performing functions related to immunity in bees of different ages and castes. To the best of our knowledge, this is the first study to demonstrate changes in the proteomic profile of RJ caused by glyphosate-based herbicides, indicating its negative effects on the nutrition and social immunity of bees.

Carcinogenic evaluation of the herbicide glyphosate

Glyphosate, an herbicidal derivative of the amino acid glycine, was introduced to agriculture in the 1970s. Glyphosate is widely considered by regulatory authorities and scientific bodies to have no carcinogenic potential. These have been also reviewed by numerous regulatory agencies including the US Environmental Protection Agency, the European Commission, and the Canadian Pest Management Regulatory Agency; however, The International Agency for Research on Cancer (IARC) published a monograph in 2015 concluding that glyphosate is “probably carcinogenic to humans”. In this review, we evaluated the carcinogenicity of the herbicide glyphosate, based on analyses of case control or cohort epidemiology studies that determinate the association between glyphosate and cancer. There are fourteen case-control studies; the assessment found that the data do not support a causal relationship between glyphosate exposure and cancer. As a result, the Panels concluded that glyphosate is unlikely to pose a carcinogenic risk to humans. Despite this results, future studies could be improved by more careful attention to validating exposure to glyphosate, thus we need for research on the health effects of glyphosate-based herbicides.

Alterations in blood parameters, DNA damage, oxidative stress and antioxidant enzymes and immune-related genes expression in Nile tilapia (Oreochromis niloticus) exposed to glyphosate-based herbicide

In this study, effects of different concentrations (0, 5, 10, 20, 30, and 40 mg/L) of glyphosate-based herbicide (GBH) on Nile tilapia (Oreochromis niloticus) were investigated after a 14-day of exposure. After determination of LC50 value, effects of GBH on hematological and serum biochemical parameters in blood samples, DNA damage, lipid peroxidation and catalase activity in liver tissues, expression levels of antioxidant enzyme-related genes (SOD, CAT, GPx, and GST) and immune-related genes (TGF-β, TGF-α and IL1-β) were evaluated. The LC50 value has been found as 44.4 mg/L for GBH. GBH exposure at all concentrations caused alterations in blood parameters. GBH induced oxidative stress in liver and DNA damage in blood. Antioxidant enzyme-related genes were significantly up-regulated to suppress oxidative stress. On the other hand, the expression levels of immune-related genes decreased in fish exposure to particularly ≥20 mg/L GBH.

Effects of glyphosate-based herbicides and their active ingredients on earthworms, water infiltration and glyphosate leaching are influenced by soil properties

BackgroundGlyphosate-based herbicides (GBHs) are among the most often used pesticides. The hundreds of GBHs used worldwide consist of the active ingredient (AI) glyphosate in form of different salts, possibly other AIs, and various mostly undisclosed co-formulants. Pesticide risk assessments are commonly performed using single AIs or GBHs at standard soil conditions without vegetation. In a greenhouse experiment, we established a weed population with common amaranth (Amaranthus retroflexus) to examine the effects of three GBHs (Roundup LB Plus, Roundup PowerFlex, Touchdown Quattro) and their corresponding AIs (salts of glyphosate isopropylammonium, potassium, diammonium) on the activity and physiological biomarkers (glutathione S-transferase, GST; acetylcholine esterase, AChE) of an ecologically relevant earthworm species (Lumbricus terrestris). GBHs and AIs were applied at recommended doses; hand weeding served as control. Experiments were established with two soil types differing in organic matter content (SOM; 3.0% vs. 4.1%) and other properties.ResultsEarthworm activity (casting and movement activity) decreased after application of glyphosate formulations or active ingredients compared to hand weeding. We found no consistent pattern that formulations had either higher or lower effects on earthworm activity than their active ingredients; rather, differences were substance-specific. Earthworm activity was little affected by soil organic matter levels. Biomarkers remained unaffected by weed control types; GST but not AChE was decreased under high SOM. Water infiltration after a simulated heavy rainfall was interactively affected by weed control types and SOM. Leachate amount was higher after application of formulations than active ingredients and was higher under low SOM. Glyphosate concentrations in soil and leachate were strongly affected by application of formulations or active ingredients and varied with SOM (significant weed control type x SOM interaction).ConclusionsWe found that both commercial formulations and pure active ingredients can influence earthworms with consequences on important soil functions. Glyphosate products showed increased, reduced or similar effects than pure glyphosate on particular soil functions; soil properties can substantially alter this. Especially at lower SOM, heavy rainfalls could lead to more glyphosate leaching into water bodies. A full disclosure of co-formulants would be necessary to further decipher their specific contributions to these inconsistent effects.

Effects of glyphosate-based herbicide on royal jelly production of Apis mellifera (Hymenoptera: Apidae) in field conditions

To evaluate the effect of Roundup® on royal jelly production, Apis mellifera hives were exposed to food containing sublethal doses of the herbicide. The bioassay was carried out in field conditions with exposed and not exposed hives to Roundup®, three replicates per treatment. The Roundup® diet decreases the amount of royal jelly produced and the number of grafted cells.

Glyphosate-based herbicide formulations and reproductive toxicity in animals

The adoption of genetically engineered (GE) crops in agriculture has increased dramatically over the last few decades. Among the transgenic plants, those tolerant to the herbicide glyphosate are among the most common. Weed resistance to glyphosate-based herbicides (GBHs) has been on the rise, leading to increased herbicide applications. This, in turn, has led to increased glyphosate residues in feed. Although glyphosate has been considered to be generally safe to animal health, recent studies have shown that GBHs have potential to cause adverse effects in animal reproduction, including disruption of key regulatory enzymes in androgen synthesis, alteration of serum levels of estrogen and testosterone, damage to reproductive tissues and impairment of gametogenesis. This review emphasizes known effects of GBHs on reproductive health as well as the potential risk GBH residues pose to animal agriculture.

Effects of low glyphosate-based herbicide concentrations on endocrine-related gene expression in the decapoda Macrobrachium potiuna.

Glyphosate-based herbicides (GBH) are the most used herbicides worldwide and are considered as endocrine-disrupting compounds (EDC) for non-target organisms. However, effects of GBH on their endocrine systems remain poorly understood. Thus, the aim of this study was to assess the effects of low concentrations of Roundup WG® on growth and reproduction process molecules in both males and females of the decapod crustacean Macrobrachium potiuna, by the relative transcript expression levels of the ecdysteroid receptor (EcR), the molt-inhibiting hormone (MIH), and the vitellogenin (Vg) genes. Prawns were exposed to three concentrations of GBH (0.0065, 0.065, and 0.28mgL-1) for 7 and 14days. The results revealed that only in males the three genes transcript levels were influenced by the GBH concentration, time of exposure, and the interaction between the concentrations and time of exposure, suggesting that males were more sensitive to GBH than females. For males, after 7days of exposure at 0.065mgL-1, EcR and MIH were over-expressed, while the Vg expression was only over-expressed after 14days. The present study highlighted that GBH impacted endocrine systems of M. potiuna. Moreover, EcR and MIH gene expressions could be promising EDC biomarkers of exposure in crustaceans. These results also indicate that GBH concentrations, considered secure by regulatory agencies, should be reviewed to minimize the effects on non-target organisms. Effect of glyphosate-based herbicide on hematological and hemopoietic parameters in common carp (Cyprinus carpio L).

The effects of Roundup (glyphosate-based herbicide) on peripheral blood parameters and hematopoietic tissue cellular composition and activity in common carp juveniles were evaluated. The fish were exposed for 7days at 0.0, 0.1, 0.5, or 5.0mg/L of glyphosate. In fish exposed to Roundup, hematological alterations were observed; however, most of them were not directly related to the herbicide concentration. An increase in Ht and MCV, and decrease in Hb, MCH, and MCHC compared to the control were observed. Fish exposed to Roundup showed also a reduction in WBC and oxidative metabolic activity of phagocytes (NBT) compared to the control. The fish exposed to 0.1 and 5.0mg/L showed increased glucose values, whereas in those subjected to 0.5mg/L blood glucose concentration declined compared to the control. Cholesterol significantly increased at 0.1mg/L and decreased at 5.0mg/L. Analysis of head kidney hematopoietic tissue revealed that Roundup at concentrations 0.5 and 5.0mg/L caused a significant increase in the rate of cell proliferation accompanied by an increase in frequency of early blast cells. No significant differences occurred in percentages of most cell lineages but the frequency of monocytoid, eosinophilic, and basophilic lineage cells significantly increased in the herbicide-exposed fish compared to the control. The obtained results revealed that sublethal concentrations of Roundup that may occur in polluted natural waters caused a slight anemic and significant immunosuppressive response in common carp juveniles. On the other hand, they indicate a considerable compensatory potential of carp hematopoietic system.

Effects of glyphosate-based herbicides on survival, development and growth of invasive snail (Pomacea canaliculata)

This study tests the hypotheses that whether environmental relevance of glyphosate would help control spread of the invasive snail Pomacea canaliculata, or benefit its population growth worldwide. Our results showed that glyphosate induced acute toxicity to the snail only at high concentrations (96h LC50 at 175mg/L) unlikely to occur in the environment. Long-term exposures to glyphosate at sublethal levels (20 and 120mg/L) caused inhibition of food intake, limitation of growth performance and alterations in metabolic profiles of the snail. It is worth noting that glyphosate at 2mg/L benefited growth performance in P. canaliculata. Chronic exposures of glyphosate significantly enhanced overall metabolic rate and altered catabolism from protein to carbohydrate/lipid mode. Cellular responses in enzyme activities showed that the exposed snails could increase tolerance by their defense system against glyphosate-induced oxidative stress, and adjustment of metabolism to mitigate energy crisis. Our study displayed that sublethal concentrations of glyphosate might be helpful in control of the invasive species by food intake, growth performance and metabolic interruption; whether environmental relevance of glyphosate (≤2mg/L) benefits population growth of P. canaliculata is still inconclusive, which requires further field study.

Low Concentrations of Glyphosate-Based Herbicide Affects the Development of Chironomus xanthus

Glyphosate is an herbicide commonly used worldwide for weed control and generally applied as part of a formulated product, such as Roundup. Contamination of surface water by glyphosate-based herbicides can cause deleterious effects in organisms, mainly in aquatic systems near to intensive agricultural areas (e.g., transgenic soybean crops). Given the lack of toxicological information concerning effects of glyphosate-based herbicides on tropical aquatic ecosystems, we aimed to evaluate the lethal and sub-lethal effects of Roundup Original® on the dipteran Chironomus xanthus. The endpoints evaluated included survival, growth, and emergence. The results showed that the 48 h LC50 for glyphosate to C. xanthus was 251.5 mg a.e./L. Larval growth of C. xanthus was reduced under glyphosate exposure (LOEC for body length = 12.06 mg/L; LOEC for head capsule width = 0.49 mg/L). No effects were observed in terms of cumulative percentage of imagoes emergence. However, low concentrations of glyphosate caused delayed emergence of females (at 1.53 mg/L) and induced fast emergence of males (at 0.49 mg/L), compared to control treatment. The deleterious effects of environmental relevant concentrations of glyphosate (0.7 mg/L) observed in terms of C. xanthus growth and development suggest that glyphosate-based herbicides can have negative consequences for aquatic non-target invertebrates such as Chironomus. Multigerational assays are needed to assess the long term effects of glyphosate on C. xanthus populations. Finally, our study adds ecotoxicological data on the effects of glyphosate-based herbicides on tropical freshwater invertebrates.

Integrative assessment of biomarker responses in teleostean fishes exposed to glyphosate-based herbicide (Excel Mera 71)

Abstract Present study deals with the effects of glyphosate-based herbicide, Excel Mera 71 on Anabas testudineus , Heteropnestes fossilis and Oreochromis niloticus in field conditions (1.85 kg/ha) based on anti-oxidative, metabolic and digestive responses. For this study following biomarkers viz., acetylcholinesterase (AChE), lipid peroxidation (LPO), catalase (CAT), glutathione-S-transferase (GST), alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), amylase, lipase and protease were investigated in gill, stomach, intestine, liver, kidney, brain, muscle and spinal cord of the concerned fish species. Enzyme activities were significantly altered by glyphosate exposure after 30 days, these activities were tissue as well as species specific. The results suggested that these biomarkers could be used to assess the ecological risks of glyphosate on fish. Bioaccumulation factor (BAF) studied in different aquatic natural macrophytes showed order of Alternanthera philoxeroides > Azolla pinnata > Lemna sp. (Minor) > Lemna sp. Bioconcentration factor (BCF) study showed maximum accumulation of glyphosate in liver, kidney or intestine, and minimum either in bone or stomach irrespective of fish species. An integrated biomarker response (IBR), which uses a battery of biomarkers to calculate the standardized scores for each biomarker responses ranging from physiological to biochemical/molecular responses, was evaluated by combining the multiple biomarkers into a single value to evaluate quantitatively the toxicological effects of glyphosate. In general, the multiple indices exhibited variations and A. testudineus was more affected than other fish species; maximum IBR value was observed for LPO and minimum in case of ALT. The order of integrated biomarkers caused by glyphosate treatment was recorded as follows: LPO > Amylase > CAT > AST > Protease > Lipase > ALP > GST > AChE > ALT for A. testudineus , LPO > AChE > AST > Protease > CAT > Amylase > Lipase > GST > ALP > ALT for H. fossilis and AChE > CAT > LPO > AST > Amylase > GST > Protease > ALP > Lipase > ALT for O. niloticus . Finally, IBR analysis is able to distinguish the variations between different parameters and might be a useful tool for the quantification of integrated responses induced by glyphosate toward fish.

Concerns over use of glyphosate-based herbicides and risks associated with exposures: a consensus statement.

The broad-spectrum herbicide glyphosate (common trade name “Roundup”) was first sold to farmers in 1974. Since the late 1970s, the volume of glyphosate-based herbicides (GBHs) applied has increased approximately 100-fold. Further increases in the volume applied are likely due to more and higher rates of application in response to the widespread emergence of glyphosate-resistant weeds and new, pre-harvest, dessicant use patterns. GBHs were developed to replace or reduce reliance on herbicides causing well-documented problems associated with drift and crop damage, slipping efficacy, and human health risks. Initial industry toxicity testing suggested that GBHs posed relatively low risks to non-target species, including mammals, leading regulatory authorities worldwide to set high acceptable exposure limits. To accommodate changes in GBH use patterns associated with genetically engineered, herbicide-tolerant crops, regulators have dramatically increased tolerance levels in maize, oilseed (soybeans and canola), and alfalfa crops and related livestock feeds. Animal and epidemiology studies published in the last decade, however, point to the need for a fresh look at glyphosate toxicity. Furthermore, the World Health Organization’s International Agency for Research on Cancer recently concluded that glyphosate is “probably carcinogenic to humans.” In response to changing GBH use patterns and advances in scientific understanding of their potential hazards, we have produced a Statement of Concern drawing on emerging science relevant to the safety of GBHs. Our Statement of Concern considers current published literature describing GBH uses, mechanisms of action, toxicity in laboratory animals, and epidemiological studies. It also examines the derivation of current human safety standards. We conclude that: (1) GBHs are the most heavily applied herbicide in the world and usage continues to rise; (2) Worldwide, GBHs often contaminate drinking water sources, precipitation, and air, especially in agricultural regions; (3) The half-life of glyphosate in water and soil is longer than previously recognized; (4) Glyphosate and its metabolites are widely present in the global soybean supply; (5) Human exposures to GBHs are rising; (6) Glyphosate is now authoritatively classified as a probable human carcinogen; (7) Regulatory estimates of tolerable daily intakes for glyphosate in the United States and European Union are based on outdated science. We offer a series of recommendations related to the need for new investments in epidemiological studies, biomonitoring, and toxicology studies that draw on the principles of endocrinology to determine whether the effects of GBHs are due to endocrine disrupting activities. We suggest that common commercial formulations of GBHs should be prioritized for inclusion in government-led toxicology testing programs such as the U.S. National Toxicology Program, as well as for biomonitoring as conducted by the U.S. Centers for Disease Control and Prevention.

Histopathological and Ultrastructural Alterations in Anabas testudineus Exposed to Glyphosate-Based Herbicide, Excel Mera 71 under Field and Laboratory Conditions

Present study aimed to investigate the effects of glyphosate-based herbicide, Excel Mera 71 on histopathological and ultrastructural changes in freshwater teleostean fish, Anabas testudineus (Bloch) under field (750 g/acre) and laboratory (17.20 mg/l) conditions for 30 days. In field experiment, fish were reared in special type of cage submerged in pond. Cellular alterations in stomach and intestine were observed through light, scanning and transmission electron microscopy (SEM and TEM). Lesions at the cellular and subcellular levels under field and laboratory conditions were compared. Responses under light microscopy depicted that the symptoms of lesions were more pronounced under laboratory condition than field. Ultrastructural examination also confirmed the observations through SEM and TEM study and the degree of responses in concerned tissues were different under these two conditions. Therefore, evaluation of these histopathological lesions in the concerned tissues may be established as symptomatic indicators for toxicity study in aquatic ecosystem.
