Effects of Dietary Nucleotide Supplementation on Performance, Profitability, and Disease Resistance of Litopenaeus vannamei Cultured in Indonesia under Intensive Outdoor Pond Conditions.

Simple SummaryIn Indonesia, there is an increasing tendency to use vegetable protein sources as a replacement for fish meal in diets for Pacific white shrimp (PWS), Litopenaeus vannamei. This; however, involves a negative impact on shrimp health given the high content of anti-nutritional factors in such protein sources. As dietary nucleotides modulate the immune response, they might be able to counteract this effect by enhancing PWS immunity and improving their performance and resistance to diseases. In this study, we evaluated the effects of nucleotide supplementation in PWS receiving diets in which fish meal had been partially replaced with soybean meal. Our data show that dietary nucleotide supplementation leads to improved growth performance and profitability, as well as to higher resistance to Vibrio harveyi. These positive results indicate that dietary nucleotides could constitute a useful tool in the production of shrimp cultured under intensive open-pond systems.This study evaluated the effects of dietary nucleotide supplementation in Pacific white shrimp, Litopenaeus vannamei, cultured in Indonesia. A total of 22,500 shrimp receiving diets in which fish meal (FM) had been partially replaced with vegetable protein sources were classified into five study groups (4500 shrimp/group) and received different diets for 110 days: 10FM (control group; 10% FM), 6FM (6% FM—low FM and no nucleotide supplementation), 10FMN (10% FM; 0.1% nucleotides), 8FMN (8% FM; 0.1% nucleotides) and 6FMN (6% FM; 0.1% nucleotides). Growth performance, body composition, total hemocyte count (THC), lysozyme activity, and hepatopancreas histopathology were assessed. Organoleptic evaluation and profitability assessments were also performed. In addition, shrimp resistance to a Vibrio harveyi challenge was studied in shrimps after having received the diets for 30 days. Results showed that reducing FM had a negative impact on growth performance and hepatopancreas morphology. Adding nucleotides resulted in better performance and profitability, a healthier histomorphological appearance of the hepatopancreas, and significantly higher survival rates upon challenge with V. harveyi, while it did not negatively affect organoleptic parameters. In conclusion, nucleotide supplementation could be useful for optimizing performance, profitability, and disease resistance in shrimp cultured under intensive outdoor pond conditions.

An Overlooked Prebiotic: Beneficial Effect of Dietary Nucleotide Supplementation on Gut Microbiota and Metabolites in Senescence-Accelerated Mouse Prone-8 Mice.

Nucleotides (NTs) are regulatory factors in many biological processes and play important roles in the growth, development, and metabolism of living organisms. We used senescence-accelerated mouse prone-8 (SAMP8) to investigate the effects of NTs on the gut microbiota and metabolites. And the promoting effect of NTs on the growth of a probiotic (Lactobacillus casei) was explored through in vitro experiments. The results showed that the sequencing depth of 16S rDNA covered all microbial species in the feces of SAMP8. Supplementation with exogenous NTs to the diet enhanced the diversity of the gut microbiota, reduced the abundance of bacteria with negative effects on the body (such as Verrucomicrobia, Ruminococcaceae, Akkermansia and Helicobacter), and increased the abundance of the microbiota, which had beneficial effects on the mice (such as Lactobacillus, Candidatus saccharimonas and Lachnospiraceae_NK4A136_group). Metabonomic analysis showed that NT deficiency in the diet significantly affected metabolites in the mouse feces. The metabolites in mice supplemented with NTs tended to be normal (SAMR1). The differentially expressed metabolites caused by NT addition are involved in various pathways in the body, including linoleic acid metabolism, vitamin B6 metabolism, and histidine metabolism. Correlation analysis revealed a significant correlation between the gut microbiota and differentially expressed metabolites caused by the addition of NTs. In vitro experiments showed that NTs significantly promoted the growth, secretion of biofilm and extracellular polymeric substance of L. casei. NTs also promoted the ability of the crude extract of L. casei to resist the secretion of Shigella biofilm. Thus, NTs can regulate the abundance of the gut microbiota and alter the metabolic expression of the intestinal microbiome.

Effect of dietary nucleotide supplementation on broiler intestinal histomorphology

The present experiment was performed to investigate the influence of two different products of nucleotides as a feed additive for broiler chicks in terms of improvement of intestinal histomorphology. A total number of two hundred and seven-one-day-old broiler chicks (Ross 308) were randomly distributed into three different treatment groups (A, B, and C), of three replicates (23 chicks / replicate). The chicks in control group (A) were fed negative control diet, group B were fed diet containing (Nucleoforce®) (0.025%) while group C were fed diet containing (Immune force®) (0.1%). Results showed that, groups fed diet supplemented with nucleotides had expressed significant increase in length of intestinal villi when compared with the control one, although group B that was fed Nucleoforce® showed highest values among the three different groups. It was concluded that, inclusion of nucleotides (Nucleoforce® and Immune-force®) in broiler diets had a positive role in improvement of intestinal histomorphology.

Effect of dietary nucleotide supplementation on broiler performance and economic efficiency

A B S T R A C T A total number of two hundred forty-one-day-old broiler chicks (Ross 308) were randomly distributed into four different treatment groups (A, B, C and D), of three replicates (20 chicks / replicate). The chicks in control group (A) were fed negative control diet, group B were fed diet containing (Nucleoforce®) (0.025%) in first 10 days of age, group B were fed diet containing (Nucleoforce®) (0.025%) in first 25 days of age. While group D was fed diet containing Nucleoforce® (0.025%) from zero day till slaughtering. This experiment was designed to investigate the effect of nucleotide as an immunostimulant for broiler chicks in terms of chick performance and economic efficiency. Results showed that, groups fed diet supplemented with Nucleoforce recorded improvements in live body weight, body weight gain and relative growth rate with no significant difference in feed intake and feed conversion index than the control group. Economic efficiency showed non-significant difference between control group and other groups but group (D) recorded high value 0.94 more than control group (A) 0.84. It was concluded that, inclusion of nucleotide 0.025% in broiler diets had a positive role in improvement of final body weight, body weight gain, feed conversion ratio and economic efficiency.

Effects of dietary nucleotide supplementation on growth in infants: a meta-analysis of randomized controlled trials.

Dietary nucleotides are thought to be conditionally essential nutrients in infancy. However, studies have reported inconsistent findings regarding the association between nucleotide supplementation and infant physical growth. We conducted this meta-analysis to examine the efficacy of nucleotide supplementation of infant formula in promoting early infant growth. Randomized controlled trials that evaluated the association between nucleotide supplementation and infant growth through June 2017 were included. Study quality was assessed using the Cochrane Collaboration's Risk of Bias tool. Standardized mean differences (SMD) with 95% confidence intervals (CIs) were calculated. Heterogeneity was assessed using Q and I2 tests. Nucleotide supplementation significantly increased the rate of weight gain (SMD 0.26; 95% CI 0.06-0.47), but had no effect on weight (SMD -0.16; 95% CI -0.55-0.23), weight Z score (SMD, -0.42; 95% CI -1.64-0.81), length (SMD 0.01; 95% CI -0.18-0.21) and length Z score (SMD 0.15; 95% CI -0.10-0.40). Occipitofrontal head circumference (OFC) at 7-8 weeks (SMD 0.30; 95% CI 0.10-0.50) and the rate of OFC gain (SMD 0.34; 95% CI 0.09-0.58) were significantly improved with nucleotide supplementation, whereas, 16- and 20-week OFC values did not differ. Our meta-analysis indicated that nucleotide supplementation can increase the rate of weight gain, OFC and rate of OFC gain; however, we cannot conclude that it affects weight, weight Z score, length or length Z score. Large-scale randomized controlled trials of long-term nucleotide supplementation are needed to reach definitive conclusions.

Dietary yeast-based nucleotides as an alternative to in-feed antibiotics in promoting growth performance and nutrient utilization in weaned pigs

Effect of dietary nucleotide supplementation without or with in-feed antibiotics on piglet performance and energy and nutrient digestibility was determined. Supplementing antibiotics without or with nucleotides (1000 ppm) increased weight gain (P < 0.05), whereas increasing nucleotide levels in antibiotic-supplemented diets linearly decreased (P < 0.05) energy and nutrient digestibility.

The effects of pure nucleotides on performance, humoral immunity, gut structure and numbers of intestinal bacteria of newly weaned pigs1

Weaning is often stressful for piglets and accompanied by morphological, histological, microbial, and immunological changes along the digestive tract. Dietary nucleotides are bioactive compounds which have the potential to diminish weaning-associated challenges. The experiment was carried out with 5 litters each of 7 pigs (mixed sex), weaned at 20 d of age. One baseline pig per litter was slaughtered at d 0. The remaining 30 pigs were housed individually and randomly allocated to 2 dietary treatments: the control diet or the control diet supplemented with a mixture of nucleotides. Measurements of growth performance traits included ADFI, ADG, G:F, and BW. At d 17, fresh fecal samples were taken to determine bacterial numbers. On d 19 and 20, pigs were slaughtered and blood samples were analyzed for plasma immunoglobulins and intestinal samples were assessed for morphological traits. Digesta from the jejunum and cecum were collected for analysis of the microbiome. The ADFI was greater in the nucleotide treatment compared with the control treatment (P < 0.05), but ADG, G:F, and BW did not differ between treatments. Plasma IgA concentrations increased with age and were greater in the nucleotide (P < 0.05) compared with the control group. There were no treatment differences in plasma IgG and IgM, gut morphology, or intestinal and fecal bacterial counts. Supplemental nucleotides may increase ADFI but without having any impact on growth performance of the pigs. Greater plasma IgA concentrations indicate that adding nucleotides in the weaning diet supported humoral immunity. However, there was no effect of dietary nucleotide supplementation on the composition of the bacterial community in parts of the small and large intestine. Further research is warranted before the use of nucleotide as a feed additive in pig diet can be recommended.

Effect of dietary nucleotide supplementation on performance and development of the gastrointestinal tract of broilers

1. Two experiments were conducted to determine the effects of dietary nucleotide supplementation on broiler performance, and physical and morphological development of the gastrointestinal tract. 2. Experiment 1: A total of 180 one-d-old male chicks were placed in battery brooders in 3 × 6 replicate pens containing 10 chicks each. Chicks were randomly assigned to one of the three dietary treatments; a maize-soyabean meal based diet supplemented with 0, 0·25, and 0·50% Torula yeast RNA (as a source of nucleotides) from 0 to 16 d of age. 3. Experiment 2: A total of 1344 one-d-old male chicks were placed in floor pens and reared on recycled wood shavings (two flocks) under a high stocking density (0·068 m2/bird). Chicks were randomly assigned to one of the 4 dietary treatments (0, 0·25% Torula yeast RNA, 2% and 6% Nupro®) for the starter period (0 to 14 d of age) with 6 replicate pens containing 56 chicks each. All the birds were fed on the same common grower diet with no supplementation of nucleotides from 15 to 32 d of age. 4. Experiment 1: Supplementing the diets with up to 0·50% Torula yeast RNA did not affect broiler performance, or relative intestinal tract weight and length of broilers at any periods measured. 5. Experiment 2: From 0 to 14 d of age, broilers fed on the diets supplemented with 0·25% Torula yeast RNA and 2 and 6% Nupro® were significantly heavier and had improved feed conversion (feed:gain) ratios as compared with the birds fed on the control diet. Supplementing the starter diet only with 2% Nupro® supplementation significantly improved body weight (BW) gain as compared with the control diet over the entire experiment (0 to 32 d of age). Broilers fed on the diets supplemented with 2 and 6% Nupro® from 0 to 14 d of age had better feed conversion (feed:gain) ratios over the entire experiment (0 to 32 d of age) as compared with the birds fed on the control diet, even though the birds were only fed on the diets supplemented with Nupro® from 0 to 14 d of age. The broilers fed on the diets supplemented with 0·25% Torula yeast RNA and 2% Nupro® had higher villus height and an improved villus height-to-crypt depth ratio as compared with birds fed on the control or 6% Nupro® diet at 14 d of age. 6. It is generally assumed that nucleotides are not an essential nutrient; thus there is no need to supplement the diets of broilers reared under normal conditions. However, dietary nucleotide supplementation may be important to maintain maximum growth performance when birds are exposed to stress conditions, such as high stocking density combined with dirty litter.

Effects of dietary nucleotide supplementation on growth performance and hormonal and immune responses of piglets

The effects of dietary nucleotide supplementation from 9 days of age until the end of post-weaning on piglets hormonal and immune responses and on growth performance were investigated. During lactation (days 9 to 21) and post-weaning (days 22 to 55) 10 [HBI Fomeva11 × (Large White × Landrace)] litters (n = 108 piglets) had ad libitum access to two standard diets, both supplemented with 0% (T0 group) or 0.1% (T1 group) of yeast extract nucleotides. BW of piglets at days 21 (P < 0.10), 35 and 55 (P < 0.05) was greater in T1 compared with T0. Feed intake was not different between groups (P > 0.05). Cortisol content was lower in T1 than in T0 at days 28 and 35 (P < 0.05), whereas growth hormone was lower at day 35 (P < 0.05). Levels of IGF-1 were similar across groups (P > 0.05). Nucleotide-supplemented diets increased lymphocyte subpopulation CD4−CD8+high at days 21 and 35 (P < 0.05), whereas CD4+CD8− cells were higher in T1 than in T0 at day 21 (P < 0.05). Peripheral blood mononuclear cells cytokine expression was influenced by dietary nucleotide supplementation. At weaning, interleukin (IL)-6 and IL-1β expression was lower (P < 0.05) in T1 compared with T0, whereas the expression of interferon (IFN)-γ and IL-10 was higher (P < 0.05). At day 28, piglets in T1 showed higher values of tumor necrosis factor (TNF)-α expression than T0 and lower values of IL-10 expression (P < 0.05). Dietary nucleotide supplementation had a suppressive effect on IL-6 and IL-10 expression (P < 0.05) at day 35. On the contrary, the expression of IFN-γ, TNF-α and IL-1β was enhanced (P < 0.05). In conclusion, these results suggest that starting a dietary nucleotide supplementation before weaning can improve the adaptive capabilities of weaned piglets to the stressors, enhancing the growth performance.

Dietary Nucleotide Supplements in Infant Formula Modify the Expression of P-glycoprotein in the Intestinal Epithelium in vitro

The effect of dietary nucleotides at concentrations found in supplemented infant formula on P-glycoprotein (P-gp) expression in colon cells was examined. We report that P-gp expression in colon cells was significantly decreased in a time- and concentration-dependent manner. When colon cells were co-cultured with lymphocytes, so as to mimic the involvement of gut-associated lymphoid tissue in normal gut pathophysiology, we observed a reversal of this effect with a demonstrated increase in P-gp expression. These findings have important implications on effects of nucleotide exposure on increasing drug bioavailability, reducing the capacity for xenobiotic efflux, and increasing the risk of inflammatory bowel disease in susceptible infants. Future studies are directed at defining both the mechanisms underlying these findings and effects of dietary nucleotide supplementation in vivo.

Lack of effect of dietary nucleotide supplementation on erythrocyte 2,3-diphosphoglycerate concentration. A study on preterm neonates

Background. Recently we demonstrated an increased 2,3-diphosphoglycerate (2,3-DPG) erythrocyte concentration in rat pups subjected to nucleotide-enriched artificial feeding.Design. The present study was carried out to test the hypothesis that a possible increase in 2,3-DPG concentration can also be obtained in human neonates who are fed nucleotide-enriched formula. Preterm neonates born or referred to the neonatal intensive care unit of the G. Gaslini Hospital, Genoa University, with a gestational age >30 weeks and <37 weeks were enrolled in our randomized trial. Recruitment took place within 48–72 hours from birth. Only newborns of mothers deciding not to breast-feed were eligible to be randomized for the supplemented group (FN) or non-supplemented group (RF). Breast-fed newborns were considered the control group (C). The study window (for supplementation and blood samples) was restricted to the first two weeks following birth (from the 2nd (t1) to the 16th (t2) day of life). At the end of our study, only 21 neonates were eligible for statistical analysis.Results. The stimulating action of dietary nucleotides on 2,3-DPG concentration failed to be demonstrated; increases in 2,3-DPG concentration that were observed in newborns fed with nucleotide supplemented formula (FN) were comparable to those observed in newborns fed with regular formula (RF) and breast-fed newborns.Conclusions. The EC recommendation for the amount of nucleotides allowed in formula milk does not seem to be high enough to have positive effects on 2,3-DPG synthesis. Whether this possible ‘pharmacological’ effect can be achieved by a higher intake of ingested nucleotides and/or a change in the proportions of single nucleotides contained in milk formulas remain interesting end points to be elucidated.

Effect of dietary nucleotide supplementation on growth and immune function in term infants: a randomized controlled trial

To examine the effect of nucleotide (NT)-supplemented cow's milk-based formula on growth and biochemical indices of immune function in healthy infants. Randomized controlled trial (RCT) of formula-fed term infants allocated to control formula with an innate level of NT at 10 mg/l (n = 102), or formula fortified with NT at 33.5 mg/l (n = 98). A parallel group of 125 breastfed infants followed the same protocol as a reference. Growth was assessed at enrolment, 7 weeks, 4 months and 7 months of age. Natural killer cell activity, cytokine production and lymphocyte subpopulations were assessed at 7 weeks of age. Antibody responses to diphtheria toxoid, tetanus toxoid and Haemophilus influenzae type b (Hib) immunizations were measured at 7 months of age. NT supplementation did not influence the growth of formula fed infants or any markers of immunity measured at 7 weeks of age. Antibody responses to tetanus toxoid were higher in the NT-supplemented group (n = 68) compared with the control group (n = 70) at 7 months of age (median (5th, 95% percentile): 1.57(0.42, 3.43) vs 1.01(0.41, 4.66) IU/ml, P < 0.03). A difference between treatments was seen in response to diphtheria toxoid but this effect disappeared when adjusted for hepatitis B immunization at birth. There was no effect of treatment on antibody responses to Hib immunization. Supplementation of formulas with NT at 33.5 mg/l resulted in a modest improvement in antibody response consistent with RCTs that used higher levels of NT supplementation. Whether this translates to clinical benefits in well-nourished infants requires further study. Supported by a grant from Wyeth Nutrition. Effect of dietary nucleotides on degree of fibrosis and steatosis induced by oral intake of thioacetamide.

The administration of thioacetamide in rats induces nodular cirrhosis of the liver, characterized by fibrous septae, parenchymal nodules, proliferation of the bile ducts, and excessive deposition of connective tissue elements. Nodular cirrhosis is also associated with changes in lipid metabolism, as shown by the accumulation of lipid droplets in the hepatocyte cytoplasm. Adequate nutritional support during cirrhosis is important to sustain liver function and promote recovery after the lesions have been induced. Supplementation with nucleotides may increase cellular proliferation and thus optimize hepatic recovery. The aim of this study was to investigate the effects of dietary nucleotide supplementation on the degree of fibrosis and steatosis in rats with liver cirrhosis induced by four months of oral intake of thioacetamide. The use of dietary nucleotides after thioacetamide administration was found to decrease the percentage area of fibrous septae. In animals with liver cirrhosis fed the nucleotide-supplemented diet for two weeks, the total area of fibrosis was reduced. Withdrawal of the hepatotoxic agent led to a decrease in the degree of steatosis in cirrhotic animals, which was significant in rats given the nucleotide-supplemented diet during a two-week recovery period. In conclusion, dietary nucleotides may be an important factor in the histological recovery of damaged liver in experimental cirrhosis.

Effect of dietary nucleotide supplementation on diarrhoeal disease in infants.

The effects of a nucleotide-supplemented formula on diarrhoeal disease was studied in 141 infants (group 1) who belonged to the low socioeconomic stratum; 148 controls (group 2) received the same formula but unsupplemented. Group 1 experienced less episodes of diarrhoea (109 versus 140), including less first episodes (74 versus 102; chi-square = 8.19, p < 0.004; odds ratio 2.01) and for a lesser number of days (807 versus 996 days); 45.0% and 31.1% of infants in groups 1 and 2, respectively, never developed episodes of diarrhoea. There were no differences in the clinical characteristics of the episodes or in the enteropathogens isolated from symptomatic or asymptomatic infants. The mechanisms through which nucleotides decrease the incidence of diarrhoeal disease in infants remain unclear.
