Nano-Sized Graphene Oxide Attenuates Ovalbumin/Alum-Induced Skin Inflammation by Down-Regulating Th2 Immune Responses in Balb/c Mice.

Graphene oxide (GO), a carbon-based material with oxygen-containing functional groups, can be applied in biomedicine for drug delivery, cancer therapy, and tissue regeneration. We have previously shown that nanoscale-sized graphene oxide (NGO), an oxidized graphene derivative, exhibits effective anti-inflammatory activity in a murine model of sepsis mediated by T helper (Th)1-promoting cytokines such as IFNγ and TNFα. However, whether NGO influences Th2-induced skin inflammation remains unclear. To address this issue, we employed an ovalbumin (OVA) plus aluminum hydroxide (Alum)-induced Th2-mediated skin inflammation model in conjunction with OVA-specific DO11.10 T cell receptor transgenic Balb/c mice. In vivo NGO injection upon OVA/Alum sensitization down-regulated OVA-elicited antigen-specific Th2 cells and GATA3-expressing Th2-type regulatory T cells. Next, we examined the effect of NGO injection on OVA/Alum-induced atopic dermatitis (AD)-like skin inflammation. NGO-injected mice exhibited significantly decreased Th2 disease phenotypes (e.g., a lower clinical score, decreased epidermal thickness and Th2 cell differentiation, and fewer infiltrated mast cells and basophils in skin lesions) compared with vehicle-injected control mice. Overall, our results suggest that NGOs are promising therapeutic materials for treating allergic diseases such as AD.

Metabolomic screening of anti-inflammatory compounds in Acanthopanax sessiliflorus fruit (Ogaza) extract

This study investigated the anti-inflammatory compounds in Ogaza, Acanthopanax sessiliflorus fruit, and their extracts using metabolomic screening. Ogaza extracts were obtained in various solvents, such as 70% ethanol, 70% methanol, and water. The anti-inflammatory activity was estimated by evaluating nitric oxide production in lipopolysaccharide (LPS)-induced RAW 264.7 cells treated with the extracts. The 70% ethanol extract (EO) showed the most effective anti-inflammatory activity, inhibiting nitric oxide production by approximately 50% and downregulating iNOS expression. The 70% ethanol extract was further fractionated into three partial subfractions by preparative LC to identify the anti-inflammatory compounds. Assessment of the anti-inflammatory activity of each subfraction revealed that the third subfraction (E-F3) showed the highest inhibitory activity against nitric oxide. E-F3 effectively suppressed iNOS expression. Subsequently, liquid chromatography-tandem mass spectrometry (LC-MS/MS) and multivariate statistical analyses were performed to identify the compounds that significantly contributed to the anti-inflammatory activity of the Ogaza extract. Fourteen and 16 compounds in the negative- and positive-ion modes, respectively, were identified as significant constituents of Ogaza. Compounds like quercetin, hyperoside, acanthoside D, oleanolic acid, and scopoletin were identified as potential anti-inflammatory components in Ogaza extract. This study characterized the functional properties of Acanthopanax sessiliflorus fruit and indicated the possibility that other compartments of Acanthopanax sessiliflorus may also serve as natural sources of nutraceuticals.

Formulation and Evaluation of Ketoprofen Emulgel for Topical Drug Delivery System

Emulgel have emerged as an auspicious drug delivery system for the delivery of hydrophobic drugs. The impartial of the study was to prepare emulgel of ketoprofen, a non-steroidal anti-inflammatory drug, using Carbopol 940 as a gelling agent. Mentha oil and clove oil were used as diffusion enhancers. The emulsion was prepared and it was combined in a gel base. The formulations were evaluated for spreading coefficient studies, rheological studies, and ex vivo release studies. Formulations F2 and F3 show maximum release. So, it can be concluded that topical emulgel of ketoprofen possesses an effective anti-inflammatory and analgesic activity.

Evaluation of Antioxidant, Anti-inflammatory, and Antimicrobial Activities of Raspberry Fruit Extract: An In Vitro Study.

Introduction Rubus idaeus (R. idaeus)is an edible fruit thatcontains numerous significant bioactive compounds that hold important biological properties and are categorized as nutraceuticals owing to the health benefits it imparts including decreasing the risk of chronic diseases like diabetes mellitus, cancer, heart disease, and many other diseases.The objective of the present research was to explorethe antimicrobial, antioxidant, and anti-inflammatory characteristics of the aqueous raspberry extractthrough in vitro assays. Materials and methods R. idaeus aqueous extract was prepared and examined for its antimicrobial activity against Streptococcus mutans (S. mutans) bacteria and Candida albicans (C. albicans) fungi using the agar-well diffusion method, and the antioxidant activity was evaluated using the DPPH (2, 2-Diphenyl-1-picrylhydrazyl hydrate) radical scavenging assay and the hydrogen peroxide radical scavenging assay. The anti-inflammatory activity of the prepared extract was investigated using bovine serum albumin (BSA) and egg albumin denaturation assays. Results R. idaeus fruit extract displayed strong antimicrobial activity at a higher concentration of 100 µL with a 26 mm zone of inhibition against Streptococcus mutans and 24 mm for Candida albicans. The extract showed 87.42% hydrogen peroxide free radical scavenging activity and inhibited 91.12% DPPH free radicals at the highest concentration of 50 µg/mL. The extract showed effective anti-inflammatory activity by preventing the denaturation of bovine serum albumin (80%) and egg albumin proteins (77%) at the highest concentration of 50 μg/mL.The free radical scavenging activity positively correlates with the increased concentration of the prepared extract against DPPH and hydrogen peroxide free radicals, thus showing the raspberry extract's potent antioxidant activity. Similarly, the anti-inflammatory assay result shows that the prepared raspberry aqueous extract has excellent anti-inflammatory activity by preventing the denaturation of bovine serum albumin and egg albumin protein in a dose-dependent manner. Conclusion The meticulously prepared raspberry extract exhibited noteworthy antimicrobial, antioxidant, and anti-inflammatory characteristics, and owing to its astounding therapeutic benefits it holds a tremendous promise as a natural alternative in the field of oral medicine especially in the management of oral mucosal lesions, oral potentially malignant lesions such as lichen planus and leukoplakia, candidiasis, oral cancer and oral mucositis. Further animal studies and clinical trials are recommended to fully reap the therapeutics potential of raspberry extract.

Rutaecarpine ameliorates imiquimod-induced psoriasis-like dermatitis in mice associated with alterations in the gut microbiota.

Psoriasis is accepted as a chronic, inflammatory, immune-mediated skin disease triggered by complex environmental and genetic factors. For a long time, disease recurrence, drug rejection, and high treatment costs have remained enormous challenges and burdens to patients and clinicians. Natural products with effective immunomodulatory and anti-inflammatory activities from medicinal plants have the potential to combat psoriasis and complications. Herein, an imiquimod (IMQ)-induced psoriasis-like dermatitis model is established in mice. The model mice are treated with 1% rutaecarpine (RUT) (external use) or the oral administration of RUT at different concentrations. Furthermore, high-throughput 16S rRNA gene sequencing is applied to analyze the changes in the diversity and composition of the gut microbiota. Based on the observation of mouse dorsal skin changes, RUT can protect against inflammation to improve psoriasis-like skin damage in mice. Additionally, RUT could suppress the expression levels of proinflammatory cytokines (IL-23, IL-17A, IL-22, IL-6, and IFN-α) within skin tissue samples. Concerning gut microbiota, we find obvious variations within the composition of gut microflora between IMQ-induced psoriasis mice and RUT-treated psoriasis mice. RUT effectively mediates the recovery of gut microbiota in mice induced by IMQ application. Psoriasis is linked to the production of several inflammatory cytokines and gut microbiome alterations. This research shows that RUT might restore gut microbiota homeostasis, reduce inflammatory cytokine production, and ameliorate psoriasis symptoms. In conclusion, the gut microbiota might be a therapeutic target or biomarker for psoriasis that aids in clinical diagnosis and therapy.

Green Synthesis of Aluminum Oxide Nanoparticles Using Clerodendrum phlomidis and Their Antibacterial, Anti-inflammatory, and Antioxidant Activities.

Clerodendrum phlomidisplays a significant role in many indigenous medical systems, and it can be mostly found in Southeast Asia. The objective of the study was to synthesize and characterize the biosynthesized aluminum oxide nanoparticles (AlO-NPs) usingC. phlomidisand analyze their antibacterial (bactericidal), antioxidant, and anti-inflammatory activities. The extract was prepared by the autoclave-assisted method, and the AlO-NPs were synthesized by the green synthesis method. The biosynthesized AlO-NPs were characterized by ultraviolet-visible (UV-Vis) spectroscopy, Fourier transform infrared (FT-IR), field emission scanning electron microscopy (FE-SEM), and energy dispersive X-ray (EDX) analysis. The antibacterial property was assessed by the Kirby-Bauer well diffusion method, and the antioxidant activity was checked by DPPH (2,2-diphenyl-1-picrylhydrazyl) activity compared with the control L-ascorbic acid. Anti-inflammatory activity was evaluated by an albumin denaturation assay, and diclofenac was used as a control. IBM SPSS Statistics for Windows, Version 21.0 was used for the statistical analysis. Results: An absorption peak at a wavelength of 380 nm was detected by UV-Vis spectroscopy analysis. It proves that AlO-NPs have been successfully produced by the green synthesis method. The results of the FT-IR study demonstrated the existence of numerous chemicals and functional groups in the 500-3500 cm-1range. AlO-NPs from the plant extract were subjected to FE-SEM analysis, which revealed an aggregated or spherically cluster-like structure. The sample's elemental makeup, which revealed that it included 38% aluminum and 28% oxygen, was identified with the help of the EDX, and this verified the high purity of the AlO-NPs. The results of the antibacterial activity of AlO-NPs revealed that there was a zone of inhibition forEnterococcus faecalis;however, there was no zone of inhibition forStreptococcus mutans. The synthesized AlO-NPs exhibit strong antioxidative (DPPH activity) and anti-inflammatory (albumin denaturation assay) action. In this work, the in vitro antioxidant activity ofC. phlomidiswas assessed using the standard, L-ascorbic acid, as a measure of DPPH activity. At a maximum concentration of 500 µg/ml, the obtained results showed the incredible antioxidant properties of the investigated AlO-NPs synthesized from the plant extracts and demonstrated 90% inhibition. AlO-NPs that were biosynthesized showed effective anti-inflammatory activity at a higher concentration of 100 µg/ml and demonstrated 89% inhibition in contrast to the drug diclofenac sodium. According to the study's findings, AlO-NPs made using a greener synthesis approach have the potential to be used in a variety of industries and are also an affordable and sustainable way to effectively act as anti-inflammatory and antioxidant agents.

Development of a Palm Kernel-Based Emulgel Loaded with Mitracarpus scarber Leaf Extracts for Effective Anti-Inflammatory Activity

Mitracarpus scaber is a tropical herb used in ethnomedicine for the treatment of inflammation and a variety of other diseases. The aim of this study was to develop a palm kernel-based emulgel containing Mitracarpus scarber and examine the product's physicochemical characteristics and anti-inflammatory efficacy on Wister rats. Different batches of emulgel comprising Mitracarpus scarber leaf extracts (MSE), different proportions of oils (palm kernel oil and/or liquid paraffin), and surfactants (tween-80 and/or Cremophor EL-30) were prepared. The freshly prepared and 5-month-old formulations, stored at room temperature (28 ± 2 °C), were evaluated for rheology, syneresis, and extract/excipient compatibility using Fourier transform infrared (FT-IR) spectroscopy. The spreadability, extrudability, and anti-inflammatory activity of the optimal formulation were compared with those of a commercial anti-inflammatory emulgel. Even after five months of storage, the created emulgels remained olive green and creamy, with a homogeneous texture, consistency, and glossy appearance. The optimized batch containing palm kernel oil, tween-80, and xanthan gum had the most stable characteristics, with no significant change in pH (P = 1.000) or FT-IR spectrum after 5 months of storage. It exhibited spreadability and extrudability of 12.75 cm2 and 600 g/cm2, respectively, compared to 16.82 cm2 and 400 g/cm2 for the commercial formulation. Four hours post-treatment, the anti-inflammatory effect of the optimized batch was significantly (P < 0.05) higher than that of the aqueous MSE dispersion. The obtained results demonstrate the prospects of palm kernel-based emulgel loaded with Mitracarpus scarber leaf extracts as an innovative therapeutic approach to inflammation treatment. Heterocyclic compounds have diverse structure, which makes them broad and economical therapeutic agents. Pyrazole is a five-membered aromatic ring that contains two nitrogen atom and is responsible for the diverse functions of heterocyclic compounds. This implies the need of reviewing the work that already has been done on the potential of pyrazole scaffolds in the field of medicinal chemistry for the synthesis of new compounds with more effective antimicrobial, antioxidant, anticancer, anti-tuberculosis, and anti-inflammatory activities.

Exploring the therapeutic potential of Prinsepia utilis Royle seed oil: A comprehensive study on chemical composition, physicochemical properties, anti-inflammatory, and analgesic activities

Ethnopharmacological relevancePrinsepiautilis (PU) Royle, native to the Himalayan region, is a deciduous thorny shrub with numerous traditional uses of its roots, leaves and seeds for treatment of conditions such as rheumatic pain, joint pain, arthritis, and inflammation. Aim of the studyKeeping in mind the growing demand of products of natural origin as alternate medicine, the present study was undertaken to scientifically validate for the first time the traditional claims of healing pain and inflammation by evaluating the fatty oil isolated from the seeds using established in vitro and in vivo models. Materials and methodsPU Seeds were Soxhlet extracted using n-hexane and fatty oil was isolated. Chemical composition of the oil was established with the aid of Gas Chromatography-Flame Ionization Detection (GC-FID) and Gas Chromatography-Mass Spectrometry (GC-MS). The oil was then subjected to in vitro anti-inflammatory activity by following the established protocols of trypsin inhibitory and bovine serum albumin denaturation assays. The acute toxicity of the oil was also studied using OECD guidelines 423. The anti-inflammatory property of the oil was further evaluated using carrageenan-induced and formalin-induced edema in the rat paw. Moreover, hot plate latency and tail immersion assay were employed to evaluate analgesic activity of the oil. To establish the quality of the oil, various physicochemical properties were also studied. ResultsGC-FID and GC-MS analysis of the oil revealed the presence of linoleic acid (59.06 ± 0.00%), oleic acid (28.11 ± 0.01%), palmitic acid (9.51 ± 0.01%) and stearic acid (3.32 ± 0.01%). In vitro trypsin inhibitory and bovine serum albumin denaturation assay revealed dose-dependent notable activity of the oil with IC50 value of 63.57 μg/mL and 518.14 μg/mL, respectively. The physico-chemical characterization demonstrated that the oil possesses a low acidity and a high oxidative stability index. The oil was found to be non-toxic and displayed effective anti-inflammatory activities with significant inhibition till 4 h in carrageenan-induced and formalin-induced rat paw edema at maximum tested dose of 200 mg/kg b.w. The oil also exhibited significant results in hot plate latency and tail immersion assay with positive effects showing up to 4 h after dose administration. ConclusionThese findings, besides supporting the traditional claims, suggest that P. utilis seed oil has potential therapeutic applications as a natural anti-inflammatory and analgesic agent. Further studies are warranted to explore its mechanisms of action and potential use in pharmaceutical and nutraceutical industries.

Synthesis and In-vivo Bioactivity Studies of Some New Hydrazide Schiff Bases and Mannich Bases of Indole Derivatives

The anti-inflammatory, analgesic, and antiulcer activities of eight new hydrazide Schiff bases and eight new Mannich bases of indole derivatives were investigated. 2-Methyl-1H-indole-3-carboxylate (III) on hydrazinolysis gave 2-methyl-1H-indole-3-carbohydrazide (IV), which on reaction with aldehydes gave corresponding hydrazide Schiff bases Va–Vh. The reaction of IV with formaldehyde, different aliphatic, aromatic, and heterocyclic amines afforded the Mannich bases of indole derivatives (VIa–VIe). The results of biological studies revealed that Schiff bases with 2,4-dichlorobenzaldehyde (Vh) and 2-chlorobenzaldehyde (Vb) were potent anti-inflammatory, analgesic, and antiulcer activities. Mannich bases (VIh and VIc) having -NO2 and -Cl groups in the paralocation of the heterocyclic nucleus displayed effective anti-inflammatory, analgesic, and antiulcer activities.

Formulation, characterization and evaluation of gelatin-syringic acid/zinc oxide nanocomposite for its effective anticancer, antioxidant and anti-inflammatory activities

BackgroundHepatocellular carcinoma (HCC) is the most typical form of liver cancer. Apart from modern therapies, various natural chemical constituents have been tested for the treatment of HCC. However, the usage of the latter has declined due to their low bioavailability and stability. ObjectivesConsidering the above drawback’s, in this study, we describe a gelatin-syringic acid/ zinc oxide (ZnO) nanocomposite for liver cancer treatment, which showed antioxidant, anti-inflammatory and anticancer activities. MethodsThe hydrothermal method was used to synthesize ZnO nanoparticle and it was encapsulated with gelatin-syringic acid by coacervation technique. The nanocomposites were characterized by UV spec, FTIR, SEM, XRD, EDX, and DLS. The drug release profile of nanocomposite was studied by dialysis bag method, which exhibits a sustained drug release. ResultsThe antioxidant ability of nano-composite was studied by performing an ABTS and DPPH assay and the IC50 was recorded as 76.5 and 47.63 µg/mL, respectively. The anti-inflammatory potential was studied by assessing the ability of nanocomposite on denaturation of protein and the results exhibited a dose-dependent inhibition. The acute toxicity of nanocomposite tested on zebrafish liver and heart showed that it is non-toxic. Moreover, the nanocomposite inhibits the Hep G2 cell viability with an increasing concentration and it increases oxidative stress caused by the mitochondrial damage, which leads to cell death. ConclusionBased on the findings of the present study, gelatin-syringic acid/ZnO nanocomposite has been shown to possess antioxidant, and anti-inflammatory properties, and thus can be used against HCC.

Highly Water-Dispersed and Stable Deinoxanthin Nanocapsule for Effective Antioxidant and Anti-Inflammatory Activity.

Deinoxanthin (DX), a carotenoid, has excellent antioxidant and anti-inflammatory properties. However, owing to its lipophilicity, it is unfavorably dispersed in water and has low stability, limiting its application in cosmetics, food, and pharmaceuticals. Therefore, it is necessary to study nanoparticles to increase the loading capacity and stability of DX. In this study, DX-loaded nanocapsules (DX@NCs) were prepared by nanoprecipitation by loading DX into nanocapsules. The size, polydispersity index, surface charge, and morphology of DX@NCs were confirmed through dynamic light scattering and transmission electron microscopy. The loading content and loading efficiency of DX in DX@NCs were analyzed using high-performance liquid chromatography. The antioxidant activity of DX@NCs was evaluated by DPPH assay and in vitro ROS. The biocompatibility of DX@NCs was evaluated using an in vitro MTT assay. In vitro NO analysis was performed to determine the effective anti-inflammatory efficacy of DX@NCs. DX@NCs exhibited increased stability and antioxidant efficacy owing to the improved water solubility of DX. The in situ and in vitro antioxidant activity of DX@NCs was higher than that of unloaded DX. In addition, it showed a strong anti-inflammatory effect by regulating the NO level in an in vitro cell model. This study presents a nanocarrier to improve the water-soluble dispersion and stability of DX. These results demonstrate that DX@NC is a carrier with excellent stability and has a high potential for use in cosmetic and pharmaceutical applications owing to its antioxidant and anti-inflammatory effects.

A reactive oxygen/nitrogen species-eliminating natural product-based nanomedicine for prevention and treatment of inflammatory liver injury

Reactive oxygen and nitrogen species (RONS) are key signaling molecules that play an important role in the progression of inflammatory diseases. Although various antioxidants have RONS scavenging properties, they are difficult to be used to cope with acute inflammation which requires inflammatory site-targeting, effective anti-inflammatory activity, and low toxicity. In this work, we developed a safe and effective nano-antioxidant ZMF formed by simply self-assembly of natural plant protein zein and natural drug mangiferin for the synergistic treatment of inflammation-related acute liver injury. ZMF could eliminate RONS, inhibit the activation of macrophages, and modulate the level of inflammatory cytokines through the synergistic effects of mangiferin and zein. In addition, in the mouse models of inflammation-related acute liver injury, ZMF showed superior efficacy in reducing inflammatory reactions. Importantly, the preliminary in vitro and in vivo experiments proved the good safety of ZMF. This study proved the remarkable anti-inflammatory activities of nanomedicine self-assembled by RONS-eliminating natural products, which could provide a reference for the green development of efficient nano-antioxidant from natural products for the treatment of inflammation-related diseases.

Associations between biomarkers and the psychophysiological impacts of teacher stress: a systematic review

Extensive literature demonstrates associations between occupational stress and adverse mental and physical health outcomes. Findings are inconsistent regarding the impacts of occupational stress on specific biomarkers/physiological systems among teachers. We conducted a PRISMA systematic electronic search to evaluate associations between biomarkers and occupational stress in teachers across education levels and identify gaps in the field. 38 papers met inclusion criteria from the initial search (N=3,290). Most papers were cross-sectional, with a subset using longitudinal or experimental designs (n=12). For Hypothalamic Pituitary Adrenal (HPA) axis biomarkers (n =29; salivary or hair cortisol), a third of studies found no association between stress and physiological alterations; the remaining HPA axis studies found associations between higher chronic stress (burnout) and blunted/hyporeactive HPA axis activation, and positive associations between acute stress (job strain, perceived stress) and increased HPA axis activation (higher diurnal cortisol, higher reactivity). Cardiovascular findings were mixed, with inconsistent relations between stress and cardiac function (n=10; HR, BP, HRV). For immunological studies (n=6; immune assays), chronic work stress was associated with increased pro-inflammatory and less effective anti-inflammatory activity, and mixed associations between burnout and inflammatory markers. Both acute and chronic occupational stress (burnout, perceived stress, job strain), negatively impact teacher health processes. More research will clarify associations between facets of occupational stress and physiology. We conclude with recommendations for improved methodology, a call for more experimental and intervention work, and more focus placed on teacher health and how teacher health impacts both teacher and child outcomes.

V4 C3 MXene Immune Profiling and Modulation of T Cell-Dendritic Cell Function and Interaction.

Although vanadium-based metallodrugs are recently explored for their effective anti-inflammatory activity, they frequently cause undesired side effects. Among 2D nanomaterials, transition metal carbides (MXenes) have received substantial attention for their promise as biomedical platforms. It ishypothesized that vanadium immune properties canbe extended to MXene compounds. Therefore, vanadium carbide MXene (V4 C3 ) is synthetized, evaluating its biocompatibility and intrinsic immunomodulatory effects. By combining multiple experimental approaches in vitro and ex vivo on human primary immune cells, MXene effects on hemolysis, apoptosis, necrosis, activation, and cytokine production are investigated. Furthermore, V4 C3 ability is demonstrated to inhibit T cell-dendritic cell interactions, evaluating the modulation of CD40-CD40 ligand interaction, two key costimulatory molecules for immune activation. The material biocompatibility at the single-cell level on 17 human immune cell subpopulations by single-cell mass cytometry is confirmed. Finally, the molecular mechanism underlying V4 C3 immune modulation is explored, demonstrating a MXene-mediated downregulation of antigen presentation-associated genes in primary human immune cells. Thefindings set the basis for further V4 C3 investigation and application as a negative modulator of the immune response in inflammatory and autoimmune diseases.

Screening for autophagy/hypoxia/ferroptosis/pyroptosis-related genes of tendon injury and repair in a rat model after celecoxib and lactoferrin treatment

BackgroundTendon injuries are among the most common musculoskeletal disorders. Celecoxib possesses an effective anti-inflammatory activity in the tendon injury treatment. Lactoferrin has a great potential for the tendon regeneration. However, the efficacy of celecoxib combined with lactoferrin in the treatment of tendon injury has not been reported. In this study, we aimed to investigate the effect of celecoxib and lactoferrin on tendon injury and repair, and screen for the crucial genes associated with the tendon injury and repair.MethodsThe rat tendon injury models were established and divided into four groups: normal control group (n = 10), tendon injury model group (n = 10), celecoxib treatment group (n = 10), and celecoxib + lactoferrin treatment group (n = 10). Then, RNA sequencing was performed to identify differentially expressed lncRNAs (DElncRNAs), miRNAs (DEmiRNAs) and mRNAs (DEmRNAs) in celecoxib treatment group and celecoxib + lactoferrin treatment group. Next, autophagy/hypoxia/ferroptosis/pyroptosis-related DEmRNAs were further identified. Subsequently, functional enrichment, protein–protein interaction (PPI) network and transcriptional regulatory network construction for these genes were performed.ResultsThe animal study demonstrated that combinational administration of celecoxib with lactoferrin rescued the harmful effects caused by celecoxib in the treatment of tendon injury. Compared to tendon injury model group, 945 DEmRNAs, 7 DEmiRNAs and 34 DElncRNAs were obtained in celecoxib treatment group, and 493 DEmRNAs, 8 DEmiRNAs and 21 DElncRNAs were obtained in celecoxib + lactoferrin treatment group, respectively. Subsequently, 376 celecoxib + lactoferrin treatment group-specific DEmRNAs were determined. Then, 25 DEmRNAs associated with autophagy/hypoxia/ferroptosis/pyroptosis were identified.ConclusionsSeveral genes, such as, Ppp1r15a, Ddit4, Fos, Casp3, Tgfb3, Hspb1 and Hspa8, were identified to be associated with tendon injury and repair.

Repurposing sarecycline for osteoinductive therapies: an in vitro and ex vivo assessment.

Tetracyclines (TCs) embrace a class of broad-spectrum antibiotics with unrelated effects at sub-antimicrobial levels, including an effective anti-inflammatory activity and stimulation of osteogenesis, allowing their repurposing for different clinical applications. Recently, sarecycline (SA)-a new-generation molecule with a narrower antimicrobial spectrum-was clinically approved due to its anti-inflammatory profile and reduced adverse effects verified with prolonged use. Notwithstanding, little is known about its osteogenic potential, previously verified for early generation TCs. Accordingly, the present study is focused on the assessment of the response of human bone marrow-derived mesenchymal stromal cells (hBMSCs) to a concentration range of SA, addressing the metabolic activity, morphology and osteoblastic differentiation capability, further detailing the modulation of Wnt, Hedgehog, and Notch signaling pathways. In addition, an ex vivo organotypic bone development system was established in the presence of SA and characterized by microtomographic and histochemical analysis. hBMSCs cultured with SA presented a significantly increased metabolic activity compared to control, with an indistinguishable cell morphology. Moreover, RUNX2 expression was upregulated 2.5-fold, and ALP expression was increased around sevenfold in the presence of SA. Further, GLI2 expression was significantly upregulated, while HEY1 and HNF1A were downregulated, substantiating Hedgehog and Notch signaling pathways' modulation. The ex vivo model developed in the presence of SA presented a significantly enhanced collagen deposition, extended migration areas of osteogenesis, and an increased bone mineral content, substantiating an increased osteogenic development. Summarizing, SA is a promising candidate for drug repurposing within therapies envisaging the enhancement of bone healing/regeneration.

In-Vitro Anti-inflammatory Activity of Traditionally reported Poly Herbal Combination

Inflammatory diseases are still one of the most important heath-problems in the world. Most of anti-inflammatory drugs such as non–steroidal anti-inflammatory drugs (NSAIDS) have number of adverse effects hence there are a need to develop safe and new anti-inflammatory formulation with minimum side effects. Present study focused on investigating and comparing anti-inflammatory potential of hydroalcoholic extracts of Tinospora cordifolia, Boerhavia diffusa, Tribulus terrestris and their combinations. Inhibition of heat induced HRBC membrane lysis was taken as a measure of the anti-inflammatory activity. The combination ratio of hydroalcoholic extracts of Tinospora cordifolia, Boerhavia diffusa, Tribulus terrestris (2:5:3) showed the most significant membrane stabilizing action on HRBC membrane as compare tohydroalcoholic extracts of individual drugs and theirother combinations. The present study revealed that the proper combination of Tinospora cordifolia, Boerhavia diffusa, Tribulus terrestris gives more effective anti-inflammatory activity than any of the individual drug.

Anti-Inflammatory Effect and Toxicological Profile of Pulp Residue from the Caryocar Brasiliense, a Sustainable Raw Material.

Caryocar brasiliense Cambess is a plant species typical of the Cerrado, a Brazilian biome. The fruit of this species is popularly known as pequi, and its oil is used in traditional medicine. However, an important factor hindering the use of pequi oil is its low yield when extracted from the pulp of this fruit. Therefore, in this study, with aim of developing a new herbal medicine, we an-alyzed the toxicity and anti-inflammatory activity of an extract of pequi pulp residue (EPPR), fol-lowing the mechanical extraction of the oil from its pulp. For this purpose, EPPR was prepared and encapsulated in chitosan. The nanoparticles were analyzed, and the cytotoxicity of the encapsu-lated EPPR was evaluated in vitro. After confirming the cytotoxicity of the encapsulated EPPR, the following evaluations were performed with non-encapsulated EPPR: in vitro anti-inflammatory activity, quantification of cytokines, and acute toxicity in vivo. Once the anti-inflammatory activity and absence of toxicity of EPPR were verified, a gel formulation of EPPR was developed for topical use and analyzed for its in vivo anti-inflammatory potential, ocular toxicity, and previous stability assessment. EPPR and the gel containing EPPR showed effective anti-inflammatory activity and lack of toxicity. The formulation was stable. Thus, a new herbal medicine with anti-inflammatory activity can be developed from discarded pequi residue.
