Effects of low-dose X-ray irradiation on melanin-derived radicals in mouse hair and skin

To clarify a possible index for long-term and low-dose irradiation, the effects of repeated low-dose X-ray irradiation on the amount of melanin-derived radicals in mouse hair and tail skin were investigated. Eight-week-old female C3H/HeSlc mice were irradiated by X-rays at a dose of 100 mGy/day 5 days/week for 12 weeks. Similarly, a 4-week irradiation experiment was carried out at 500 mGy/day for C3H/HeSlc mice, or at 10, 100, and 500 mGy/day for 8-week-old female C57BL/6NCrSlc mice. The hair sample (~10 mg) was weighed accurately and stuffed into a plastic tube. The 2-cm tip of the tail was sampled and lyophilized. Melanin-derived radicals in hair and tail samples were measured by X-band electron paramagnetic resonance spectrometry. After X-ray irradiation at 100 mGy/day for 12 weeks, no difference was found in the amount of melanin-derived radicals in the hair of the irradiated and non-irradiated groups. X-ray irradiation at 500 mGy/day for 4 weeks increased the amount of melanin-derived radicals in hair compared with the non-irradiated group, but the baseline amount of melanin-derived radicals in hair was varied. The amount of melanin-derived radicals in the tail skin dose-dependently increased. Melanin-derived radicals in skin may be an endogenous marker for long-term and low-dose irradiation.

First Insights into the Effect of Low-Dose X-Ray Irradiation in Adipose-Derived Stem Cells.

(1) Background: Emerging interest of physicians to use adipose-derived stem cells (ADSCs) for regenerative therapies and the fact that low-dose irradiation (LD-IR ≤ 0.1 Gy) has been reported to enhance the proliferation of several human normal and bone-marrow stem cells, but not that of tumor cells, lead to the idea of improving stem cell therapies via low-dose radiation. Therefore, the aim of this study was to investigate unwanted side effects, as well as proliferation-stimulating mechanisms of LD-IR on ADSCs. (2) Methods: To avoid donor specific effects, ADSCs isolated from mamma reductions of 10 donors were pooled and used for the radiobiological analysis. The clonogenic survival assay was used to classify the long-term effects of low-dose radiation in ADSCs. Afterwards, cytotoxicity and genotoxicity, as well as the effect of irradiation on proliferation of ADSCs were investigated. (3) Results: LD (≤ 0.1 Gy) of ionizing radiation promoted the proliferation and survival of ADSCs. Within this dose range neither geno- nor cytotoxic effects were detectable. In contrast, greater doses within the dose range of >0.1–2.0 Gy induced residual double-strand breaks and reduced the long-term survival, as well as the proliferation rate of ADSCs. (4) Conclusions: Our data suggest that ADSCs are resistant to LD-IR. Furthermore, LD-IR could be a possible mediator to improve approaches of stem cells in the field of regenerative medicine.

Effects of low-dose X-ray irradiation on activated macrophages and their possible signal pathways.

Low-dose irradiation (LDI) has been used in clinics to treat human diseases, including chronic inflammation. This study assessed the effects of LDI on the inflammatory response of activated mouse primary peritoneal macrophages, and the underlying signal pathways. Primary peritoneal macrophages were isolated from mice and then incubated with lipopolysaccharide (LPS)-coated Ti microparticles (Ti-positive control) with or without brief exposure to LDI (X-ray, 0.5 Gy) 1 h later (Ti-LDI group) or left untreated in culture medium (Ti-negative control). The macrophages were then subjected to qRT-PCR, Western blot, cell viability CCK-8 assay, and ELISA. qRT-PCR analysis revealed the Ti-LDI group expressed significantly lower levels of IL-1β, IL-6, and TNF-α mRNA than those of the Ti-positive control group, while the ELISA data showed that Ti-LDI group had significantly lower secretion of IL-1β, IL-6, and TNF-α proteins. The most significant reduction associated with LDI was the secretion TNF-α protein, which barely increased from 13 to 25 h after treatment. Western blot data demonstrated that phosphorylation of p65 and ERK was much lower in the Ti-LDI group than in the controls. The data from the current study suggests that LDI of activated mouse macrophages was associated with significantly lower inflammation responses, compared with non-exposed activated macrophages, which was possibly through inhibition of the NF-κB and ERK pathways.

Effect of low-dose X-ray irradiation on micronucleus formation in human embryo, newborn and child cells

Purpose: It is well known that a high-dose of ionizing radiation is sufficient to break DNA strands, which leads to elevated genotoxic risks; however, the risks associated with low doses of ionizing radiation remain unclear. In addition, there is little data about the effect of low-dose ionizing radiation on human-derived embryo, newborn and child cells. We investigated the frequency of micronucleus (MN) formation in these cells to understand the genotoxic effects of ionizing radiation.Materials and methods: We irradiated the cells with X-rays from 0.02–2 Gy at a rate of 0.0635 Gy/min. After irradiation, we investigated the effect of low-dose X-ray irradiation on cellular viability and frequency of MN formation.Results: Increases in MN formation were largely dose-dependent; however, there were no differences between controls and doses lower than 0.2 Gy, except in KMST-6 human transformed embryo cells.Conclusion: We could not detect an obvious effect of low-dose X-ray irradiation at doses lower than 0.1 Gy. The embryonic cells were more sensitive to X-ray irradiation than newborn and child cells. The threshold for X-ray-induced MN formation appears to be in the range of 0.05–0.1 Gy in cultured human embryo, newborn and child cells.

Effect of low-dose X-ray irradiation and Ti particles on the osseointegration of prosthetic.

Low-dose irradiation (LDI) exhibits a positive effect on osteoblasts and inhibitory effect of inflammation. Here, we test the hypothesis that LDI can promote osseointegration and inhibit the inflammatory membrane formation in the presence of titanium (Ti) particles. Endotoxin-free titanium particles were injected into rabbit, prior to the insertion of a Ti6-Al-4-V sticks pre-coated with hydroxyapatite. Two days after operation, both distal femurs of the animal were exposed to 0.5 Gy X-ray irradiation. All ani-mals were euthanized 8 weeks after the operation. The PINP concentration was determined at day 0, 2, 4, and 8 weeks after operation. Trabecular morphology around the implants 8 weeks after operation was assessed using micro-CT, then the maximum push out force of simples was assessed using biomechanics test. Five samples in each group were chosen for bone histomorphology study without decalcification 8 weeks after operation. The results confirmed that the LDI can significantly improve ingrowth of bone into the prosthetic interface and stability of the prosthesis when there was no wear particles. Although promotion effects for bone formation induced by LDI can be counteracted by wear particles, LDI can significantly inhibit the interface membrane formation around the implant induced by wear particles. Based on these results, we conclude that LDI may be useful for enhancing the stability of prosthesis when there are no wear particles and for inhibiting the interface membrane formation during the early stage of aseptic loosening in the presence of wear particles. © 2016 Orthopaedic Research Society. Effects of low-dose X-ray irradiation of eggshells on radical production

We investigated effects of low-dose X-ray irradiation on eggshells radical production. Eggshells irradiated by low-dose X-ray produced stable radicals. The stable radicals at the central region of the spectrum are the carbonates and CO2•− signals. The electron spin resonance (ESR) signals increase with increasing absorbed dose. The estimated radical concentration (spin number) was approximately 3.6 × 1012 (spins/g) at 10-Gy dose using various known concentrations of TEMPOL (4-hydroxy-2, 2, 6, 6-tretramethylpiperidin-1-oxyl) aqueous solutions. The stable radicals in irradiated eggshells were recognized at the low-level dose (0.10 Gy). In addition, no significant dose rate (0.5–1.5 Gy/min) dependence was found for the radical production.

Microbial Quality and Safety of X‐Ray Irradiated Fresh Catfish (Ictalurus punctatus) Fillets Stored under CO2 Atmosphere

The effect of low-dose X-ray irradiation (0, 2, and 3 kGy) on pathogens, spoilage bacteria (psychrotrophic, anaerobic, and lactic acid bacteria), and physicochemical parameters of iced catfish fillets stored under carbon dioxide (CO(2) ) atmosphere were studied. Irradiation of 2 or 3 kGy eliminated all Listeria monocytogenes (4.8 log CFU/g) and Salmonella Typhimurium (4.7 log CFU/g) while CO(2) atmosphere alone showed less than 1 log reduction on Salmonella Typhimurium. There was no recovery of either pathogen for the experimental period. Regardless of dose, irradiation showed 2 log reduction on psychrotrophic, anaerobic, and lactic acid bacteria for 24 d. Further post-irradiation reductions of spoilage bacteria were observed. Spoilage bacteria on catfish fillets treated with 2 kGy irradiation regained their growth after 16 d of storage. However, catfish fillets exposed to 3 kGy did not show growth of spoilage bacteria throughout the experimental period (24 d). Irradiation (2 or 3 kGy) increased pH and TBARS, and decreased Hunter "b" values but produced no effect on water holding capacity and texture. Shelf life of catfish fillets exposed to 2 or 3 kGy irradiation could be extended by more than 24 d at 2 to 4 °C, with the added safety of possibly eliminating pathogens.

Response to low-dose X-irradiation is p53-dependent in a papillary thyroid carcinoma model system

The link between high doses of radiation and thyroid cancer has been well established in various studies, as opposed to the effects of low doses. In this study, we investi-gated the effects of low-dose X-ray irradiation in a papillary thyroid carcinoma model with wild-type and mutated p53. A low dose of 62.5 mGy was enough to cause an upregulation of p16 and a decrease in the number of TPC-1 cells in the S phase, but not in the number of BCPAP p53-mutant cells. At a dose of 0.5Gy, visible signs of senescence appeared only in the TPC-1 cells. We conclude that low doses of X-rays are enough to cause a change in cell cycle distribution, possibly p53-dependent p16 activation, but no significant apoptosis. Senescence requires higher doses of X-irradiation via a mechanism involving both p16 and p21.

The Effect of Low-dose X-ray Irradiation on the Quality of Fresh-cut Asparagus in Microwaveable Vacuum Skin Packs

Low-dose x-ray irradiation was used on vacuum skin-packaged fresh-cut asparagus to extend its shelf life and improve its microbial safety. Freshly imported Peruvian asparagus (Asparagus officinalis L.) spears were obtained from a local source, cleaned, trimmed, and sanitized and then vacuum skin-packaged (VSP) in a tray pack system. A breathable lid film was vacuum-sealed to protect the product from contamination and allow for gas exchange. Asparagus were packaged in VSP, which modified the package atmosphere, and exposed to a target dose of 1.0 kGy or left untreated. Asparagus were also placed in low-density polyethylene (LDPE) bags as a control. All samples were then stored at 4 °C for 24 days. To determine the effectiveness of the irradiation treatment, sample packages were periodically removed from storage and several measurements were used to evaluate the product, including headspace gas content, microbial growth, water soluble sugar content, and enzyme activity. A dose of 1.0 kGy reduced initial microbial population up to 3.8 log colony-forming units (cfu)/g and maintained 20% more of the initial sugar content relative to the non-irradiated VSP asparagus. Phenylalanine ammonia-lyase (PAL) activity in VSP asparagus was lower than in the control package (LDPE bag) regardless of irradiation treatment (P < 0.05). However, x-ray treatment increased the PAL activity of asparagus in VSP up to Day 8 (P < 0.05).

Change of glutathione peroxidase synthesis along with that of superoxide dismutase synthesis in mice spleens after low-dose X-ray irradiation

We have previously demonstrated that the activity of superoxide dismutase (SOD), an antioxidant, is enhanced by low-dose X-ray irradiation in various organs of animals such as rats. Since SOD is an enzyme that mediates the dismutation of O 2 − to H 2O 2, the question as to whether the resultant H 2O 2 is further detoxicated into H 2O and O 2 or not must still be evaluated. Hence, we studied the effect of low-dose X-ray irradiation on the synthesis of glutathione peroxidase (GSHPx), which is an antioxidant that catalyzes this reaction. The results suggest that H 2O 2 produced by increased SOD activity can be detoxicated into H 2O and O 2 due to simultaneous enhancement of the GSHPx activity by X-ray irradiation at 20 cGy, in contrast to irradiation at 400 cGy. The results also show the enhancement in enzyme activities by induction of their synthesis shortly after irradiation at 20 cGy. Moreover, as this phenomenon was observed in BALB/c mice (which are more radiation-sensitive compared to other mouse strains) and radiation-resistant C57BL/6NJcl mice, it was considered to be a common phenomenon in the rat spleen.

Effects of low-dose X-ray irradiation on biomembrane in brain cortex of aged rats

We previously found that low-dose X-ray irradiation or radon (weak α-ray) inhalation increases SOD activities and reduces lipid peroxide levels in various organs of 7-week-old rats or rabbit. In this study, we examined how the changes of SOD activity, lipid peroxide level, and membrane fluidity of the cerebral cortex in aged male Wistar rats (65 and 91 weeks old) were affected by low-dose X-ray irradiation (100 cGy or under) compared with those in 7-week-old rats, to elucidate the mechanism of aging inhibition. The following results were obtained: Although radiation sensitivity was observed to decreases with age, low-dose irradiation changed the Mn-SOD activity, lipid peroxide level, and membrane protein fluidity parameter of the cerebral cortex in the aged rats to be closer to those in the youth. These finding suggest that the increased SOD activity induced by low-dose irradiation enhances biomembrane functions, and that the decrease of lipid peroxide level enhances the membrane protein fluidity.
