Epidermal growth factor attenuates tubular necrosis following mercuric chloride damage by regeneration of indigenous, not bone marrow-derived cells.

To assess effects of epidermal growth factor (EGF) and pegylated granulocyte colony-stimulating factor (P-GCSF; pegfilgrastim) administration on the cellular origin of renal tubular epithelium regenerating after acute kidney injury initiated by mercuric chloride (HgCl2). Female mice were irradiated and male whole bone marrow (BM) was transplanted into them. Six weeks later recipient mice were assigned to one of eight groups: control, P-GCSF+, EGF+, P-GCSF+EGF+, HgCl2, HgCl2+P-GCSF+, HgCl2+EGF+ and HgCl2+P-GCSF+EGF+. Following HgCl2, injection tubular injury scores increased and serum urea nitrogen levels reached uraemia after 3 days, but EGF-treated groups were resistant to this acute kidney injury. A four-in-one analytical technique for identification of cellular origin, tubular phenotype, basement membrane and S-phase status revealed that BM contributed 1% of proximal tubular epithelium in undamaged kidneys and 3% after HgCl2 damage, with no effects of exogenous EGF or P-GCSF. Only 0.5% proximal tubular cells were seen in S-phase in the undamaged group kidneys; this increased to 7–8% after HgCl2 damage and to 15% after addition of EGF. Most of the regenerating tubular epithelium originated from the indigenous pool. BM contributed up to 6.6% of the proximal tubular cells in S-phase after HgCl2 damage, but only to 3.3% after additional EGF. EGF administration attenuated tubular necrosis following HgCl2 damage, and the major cause of this protective effect was division of indigenous cells, whereas BM-derived cells were less responsive. P-GCSF did not influence damage or regeneration.

Endogenous EGF maintains Sertoli germ cell anchoring junction integrity and is required for early recovery from acute testicular ischemia/reperfusion injury

Administration of exogenous epidermal growth factor (EGF) improves testicular injury after acute ischemia-reperfusion (IR) stress, but the molecular basis is poorly understood. The role of endogenous EGF in testicular recovery and the underlying intracellular signaling pathways involved were herein investigated. In mice, testicular IR injury significantly enhanced the expression level of endogenous Egf at the very beginning of reperfusion. Expression of EGF receptor (Egfr (ErbB1)) was accordingly upregulated 3 h after reperfusion. Deprivation of majority of circulated EGF by sialoadenectomy aggravated testicular detriment (especially in pachytene spermatocytes), enhanced germ cell apoptosis, and thereafter resulted in impaired meiotic differentiation after IR insult. Mechanistically, endogenous EGF signaling appeared to be indispensable for the proper maintenance of Sertoli germ cells anchoring junction dynamics during the early testicular recovery. We also provided the in vitro evidences in a well-established rat Sertoli germ cell co-cultures model that the pro-survival effect of endogenous EGF on germ cells in response to testicular IR insult is mediated, at least in part, via the phosphatidylinositol 3-kinase/pAkt pathway. Collectively, our results suggest that the augment of endogenous EGF during the early testicular recovery may act on top of an endocrinous cascade orchestrating the intimate interactions between Sertoli cells and germ cells and may operate as indispensable defensive mechanism in response to testicular IR stress. Future studies in this field would shed light on this complicated pathogenesis.

Anti-inflammatory effects of epidermal growth factor on the immature human intestine

The inflammatory response of the preterm infants' intestine underlines its inability to respond to hemodynamic stress, microbes, and nutrients. Recent evidence suggests that exogenous epidermal growth factor (EGF) exerts a therapeutic influence on neonatal enteropathies. However, the molecular mechanisms underlying the beneficial effects of EGF remain to be clarified. The purpose of this study was to evaluate the impact of EGF on the gene expression profiles of the developing human small and large intestine at midgestation in serum-free organ cultures using microarrays. The gene expression profiles of cultured human fetal ileal and colonic explants were investigated in the absence or presence of a physiological concentration of 50 ng/ml EGF for 48 h. Data were analyzed with the Ingenuity Pathway Analysis (IPA) software and confirmed by qPCR. We found a total of 6,474 differentially expressed genes in the two segments in response to EGF. IPA functional analysis revealed that in addition to differentially modulating distinct cellular, molecular, and physiological functions in the small and large intestine, EGF regulated the inflammatory response in both intestinal segments in a distinct manner. For instance, several intestinal-derived chemokines such as CCL2, CCL25, CXCL5, and CXCL10 were found to be differentially regulated by EGF in the immature ileum and colon. The findings showing the anti-inflammatory influence of exogenous EGF suggests a mechanistic basis for the beneficial effects of EGF on neonatal enteropathies. These results reinforce growing evidence that by midgestation, the human small intestine and colon rely on specific and distinct regulatory pathways.

Effect of exogenous epidermal growth factor (EGF) on nonenzymatic antioxidant capacities and MPO activity of wound tissue

Growth factors and cytokines are key regulators of the wound-healing process. Epidermal growth factor (EGF) plays a vital role during the process of wound healing. There are limited numbers of studies related to EGF implantation effects on oxidative stress in oral wound healing. The objective of this study was to investigate the effect of EGF on myeloperoxidase (MPO) activity and nonenzymatic antioxidant levels of oral wound tissue on different days. In this study, New Zealand male albino rabbits were used. After submucosal incisions, the rabbits were divided into two equal groups: untreated wounds, and EGF-implanted wounds. The levels of glutathione (GSH), ascorbic acid (AA), and MPO activity were measured spectrophotometrically. As a result, MPO activity of oral wound tissue strips was increased by exogenous EGF treatment compared with controls. In addition, GSH and AA levels of oral wound tissue strips were not changed during oral wound-healing phases for both controls and experimental groups.

Epidermal growth factor preserves intestinal integrity and decreases mortality in a murine model of Pseudomonas aeruginosa pneumonia

Exogenous epidermal growth factor (EGF) attenuates intestinal injury and decreases mortality in a peritonitis model. However, the role of EGF in sepsis caused by extraabdominal infection is unknown.We hypothesized that EGF preserves intestinal integrity and decreases mortality in P. aeruginosa pneumonia‐induced sepsis.FVB/N mice were intratracheally injected with 40 μl of 0.6 McF P. aeruginosa and treated with or without EGF (150 ug/kg/d i.p.; n=9/group). Controls were injected with saline (n=5). All mice received antibiotics. At 24 hr, intestines were evaluated for apoptosis by caspase‐3 staining, proliferation by BrdU staining, and villus length. Survival studies were done in a separate cohort of mice (n=5–10/group).Compared to shams, septic mice had increased intestinal apoptosis (36 ± 3 vs. 17 ± 3 cells/100 crypts; p<0.001) and decreased proliferation (502 ± 51 vs. 959 ± 43 cells/100 crypts; p<0.001), while EGF normalized both to sham levels (21 ± 1 vs. 17 ± 3 and 886 ± 64 vs. 959 ± 43 cells/100 crypts, respectively; p=ns). Septic mice had shorter villi compared to shams, and EGF treatment resulted in normalization to sham villi lengths (333 ± 15 vs. 415 ± 30 vs. 432 ± 19 μ m; p<0.01 and p=ns). At 7 days, septic EGF treated mice had a survival advantage compared to septic mice (92% vs. 50%; p=0.05).EGF preserves intestinal integrity and improves survival in pneumonia‐induced sepsis; thus, the protective effects of EGF are not specific to the anatomical site or type of infection. EGF may be a novel therapeutic agent for the treatment of sepsis.

Decreased mitogenic response to epidermal growth factor in human squamous cell carcinoma lines overexpressing epidermal growth factor receptor owing to limiting amounts of the adaptor protein Grb2: rescue by retinoic acid treatment.

Growth factor receptors of the tyrosine kinase family regulate proliferation of a variety of cell types. In some human cancers, the epidermal growth factor receptor (EGFR) and its ligands often are overexpressed, leading to both constitutive and autocrine activation. Intracellular signaling via this receptor takes place through several mechanisms of action, including activation of ras and the mitogen-activated protein kinase (MAPK) pathway. Our previous studies have shown that human squamous cell carcinoma (SCC) lines overexpress EGFR and do not increase proliferation in response to exogenous epidermal growth factor (EGF). The vitamin A metabolite retinoic acid (RA) has been used as a chemotherapeutic drug in the treatment of SCC. RA decreases proliferation of SCC lines, in part owing to inhibition of EGFR expression. However, we previously found that treatment of SCC lines with inhibitory doses of RA sensitized cells to the proliferative effects of EGF. We now present a mechanism of action for this effect. RA inhibited expression of EGFR and proteins in the MAPK signaling pathway. Expression of these molecules returned to basal levels within 24 h after RA withdrawal. RA also inhibited autocrine secretion of EGF, which returned to basal levels with slower kinetics. During this time, addition of exogenous EGF stimulated mitosis in SCC lines. These data suggested that signaling proteins downstream of overexpressed EGFR may have limited the mitotic response in SCC lines. In support of this hypothesis, overexpression of the EGFR adaptor protein Grb2 increased cell proliferation and restored EGF-induced mitosis.

Epidermal growth factor accelerates pancreatic recovery after caerulein-induced pancreatitis

We examined the influence of endogenous and exogenous epidermal growth factor (EGF) on pancreatic repair after acute pancreatitis. Caerulein-induced pancreatitis was evoked in rats with intact or removed salivary glands and EGF (10 μg/kg) was administered starting 24 h after cessation of caerulein infusion. The dose of EGF 10 μg/kg was chosen because it was the most effective in preliminary experiments when 1, 10 or 50 μg/kg of EGF was used. Caerulein administration caused acute edematous pancreatitis with biochemical and histological manifestation of pancreatic damage, followed by spontaneous regeneration. The effect of salivectomy on the course of acute pancreatitis was slight, resulting in additional reduction in pancreatic blood flow, DNA synthesis and in an increase in plasma interleukin 1β level. Treatment with EGF accelerated the healing of pancreatic damage, causing an increase in pancreatic blood flow and DNA synthesis. EGF caused faster normalization of plasma amylase and lipase activity and plasma interleukin 1β concentration, as well as, this peptide accelerated the restoration of pancreatic amylase activity. On histological examination, EGF caused reduction of pancreatic damage and acceleration of tissue repair. We conclude that EGF reduces the severity of pancreatic damage evoked by caerulein-induced pancreatitis-related pancreatic damage and accelerates tissue repair. The beneficial effects of EGF appear to depend, at least in part, on the improvement of pancreatic blood flow, as well as on an increase of pancreatic cell growth and limitation of the activation cytokine release.

The distribution of endogenous epidermal growth factor after small bowel resection suggests increased intestinal utilization during adaptation

Background/Purpose: Although exogenous epidermal growth factor (EGF) amplifies adaptation after massive small bowel resection (SBR), the role for endogenous EGF is unclear. The authors sought to determine whether SBR was associated with changes in the levels of EGF in the serum, saliva, or urine and EGF receptor (EGF-R) signaling in the ileum. Methods: Male ICR mice underwent 50% proximal SBR or sham surgery bowel transection/reanastomosis). After 3 days, levels of EGF were measured by enzyme-linked immunosorbent assay (ELISA) in the serum, saliva, and urine. EGF-R activation was measured in isolated ileal enterocytes by probing an EGF-R immunoprecipitate with an antibody to phosphotyrosine. Results: When compared with sham, SBR resulted in no change in serum, increased salivary (2209 ± 266 nmol SBR v 1183 ± 119 nmol sham, P < .05) and decreased urinary (417 ± 58 nmol SBR v 940 ± 143 nmol sham; P < .05) EGF levels. EGF-R activation increased 2.5-fold after SBR. Conclusions: Increased salivary and reduced urinary EGF linked with enhanced EGF-R activation suggests increased ileal utilization of EGF during adaptation. This observation, along with the known beneficial effects of exogenous EGF, infers a crucial role for endogenous EGF in the pathogenesis of intestinal adaptation after SBR.

Interactions between epidermal growth factor and the Tabby mutation in skin.

Mutations of the X-linked genes Tabby (Ta) in mice and EDA in humans result in developmental and functional abnormalities, primarily in the skin and hair follicles. Although both genes are believed to encode membrane-associated proteins, it has been suggested that, in the mouse, the mutation is linked to a deficiency of epidermal growth factor (EGF). This study investigated relationships between the skin abnormalities of Ta mice and the EGF signal pathway. The distribution of endogenous EGF in tissues of Ta/Y and +/Y animals was examined and, because of its reported morphogenetic actions and ability to overcome receptor signalling defects in vivo, the effects of exogenous EGF on the hair follicle population were determined. EGF levels were similar in a number of tissues of Ta/Y and +/Y mice, but amounts in Ta/Y submaxillary glands were reduced, probably due to a smaller gland size. Exogenous EGF inhibited hair follicle development and decreased follicle density in both genotypes. It was concluded from comparisons of the distributions of EGF and its effects in skin with those in mice bearing mutations in the EGF signal pathway that the normal phenotype results from interactions between EGF and the Ta peptide in skin.

Epidermal Growth Factor Augments Adaptation Following Small Bowel Resection: Optimal Dosage, Route, and Timing of Administration

Background.In assorted animal models of small bowel resection (SBR), exogenous epidermal growth factor (EGF) has been shown to augment intestinal adaptation. This study was designed to elucidate the optimal dose, route, and timing of exogenous EGF to boost adaptation in our murine model of SBR.Methods.Male ICR mice underwent either 50% proximal SBR or sham surgery (bowel transection with reanastomosis) and then randomized to receive either saline or human recombinant EGF (5, 50, 150, or 300 μg/kg/day) by twice daily intraperitoneal (i.p.) injection or orogastric gavage (o.g.). At 7 days, protein and DNA content, crypt depth, and villus height were determined in the ileum. The premium dose and route was then given for 1 week either during (1 week after SBR) or after the adaptive phase (1 month after SBR). Differences between group means were analyzed using ANOVA. AP< 0.05 was considered significant.Results.EGF enhanced DNA and protein content, crypt depth, and villus height to the greatest extent at a dosage of 50 μg/kg/day by the o.g. route. EGF had no significant effect on enhancing adaptation when given after the adaptive response had already occurred.Conclusions.Intestinal adaptation is optimally enhanced by a specific dose and route of EGF. Exogenous EGF enhances adaptation only during the adaptive response to SBR and not after it has already taken place. Determination of the best circumstances for EGF administration will permit a systematic approach toward understanding a mechanism for the beneficial effect of EGF during intestinal adaptation.

Effect of Epidermal Growth Factor, Hepatocyte Growth Factor, and Keratinocyte Growth Factor, on Proliferation, Motility and Differentiation of Human Corneal Epithelial Cells

We sought to determine the effects of exogenous epidermal growth factor (EGF), heparin-binding EGF (HB-EGF), transforming growth factor alpha (TGF-alpha), single-chain precursor hepatocyte growth factor (SC-HGF), double-chain mature HGF (DC-HGF), and keratinocyte growth factor (KGF) on proliferation, motility, and differentiation of first passage cultures of human corneal epithelial cells in serum-free chemically defined medium. The effect of EGF, HB-EGF, TGF-alpha, SC-HGF, DC-HGF, KGF or combinations of the growth factors on proliferation was measured by counting cells present after 3 weeks of culture and by immunostaining for the cell-cycle-specific nuclear proliferation antigen Ki-67. The effect of the factors on epithelial cell motility was assessed by morphometric analysis of photographs of cells migrating from confluent islands of cells. The effect of growth factors on differentiation of epithelial cells were determined by immunostaining epithelial cell islands for the keratin K3 and by Western blotting for keratin K3. EGF, alone or in combination with KGF and SC-HGF, significantly stimulated motility of epithelial cells at the periphery of confluent islands of cells and induced an elongated cell morphology. TGF-alpha, HB-EGF and DC-HGF produced motility effects similar to EGF. There was diminished proliferation of the migrating cells in response to EGF, HB-EGF, TGF-alpha or DC-HGF, while non-migrating epithelial cells in the center of confluent islands continued to proliferate in response to the growth factors. EGF, HB-EGF, TGF alpha or DC-HGF inhibited expression of the differentiation-related marker keratin K3 in epithelial cells, both at the edge and at the center of the islands. KGF stimulated proliferation of corneal epithelial cells at low density and in confluent islands of cells. KGF did not affect expression of keratin K3 or migration of epithelial cells. SC-HGF had no effect on corneal epithelial cells. results indicate that the effects of EGF, HB-EGF, TGF-alpha and DC-HGF on corneal epithelial cell proliferation, motility and differentiation vary from those of KGF and SC-HGF, EGF, HB-EGF, TGF-alpha and DC-HGF induced changes in epithelial cell morphology and motility in cells plated at low cell density or in cells located at the edge of a confluent island. Thus, these effects appear to be dependent on the extent of cell-cell contact. The inhibitory effect of EGF, HB-EGF, TGF-alpha or DC-HGF on corneal epithelial cell differentiation, however, is independent of cell density. In contrast, KGF stimulates proliferation of human corneal epithelial cells regardless of the cell density, does not significantly affect motility, and has no effect on differentiation in serum-free defined medium in vitro.

Secretion of epidermal growth factor and related polypeptides by the DU 145 human prostate cancer cell line.

Epidermal growth factor (EGF)-like proteins may be important in regulating the growth of human prostate cancer cells and in the development of hormone independence. In the present study, we demonstrated that the DU 145 human prostate cancer cell line secretes EGF-like polypeptides into serum-free culture medium. The production of both authentic EGF and transforming growth factor-alpha was demonstrated by specific radioimmunoassays. In addition to 6-7k monomers, immunoreactive higher molecular weight species were isolated by gel chromatography. The DU 145 cells also specifically bound human EGF, with both high- (Kd 1.8 x 10(-10) M) and low- (Kd 1.1 x 10(-9) M) affinity binding sites. Exogenous EGF stimulated 3H-thymidine incorporation when cells were plated at low density in serum-free culture medium, while at higher cell density neither cell division nor 3H-thymidine incorporation was significantly altered. We postulate that DU 145 cells may be autologously stimulated by endogenously produced EGF and related polypeptides, which, under normal culture conditions, precludes any further effect of exogenous EGF.
