Abstract Background: Prostate cancer is the second most frequently occurring carcinoma in males worldwide and one of the leading causes of death in men around the world. Recent studies estimate that over 1.4 million males are diagnosed with prostate cancer on an annual basis, with approximately 375 000 succumbing to the disease annually. With current treatments continuing to show severe side effects, there is a need for new treatments. In this study we looked at the effect of cannabis sativa extract, cannabidiol and cisplatin on prostate cancer cells, PC3. Methods: In addressing the above questions, we employed the MTT assay to measure the antiproliferative effect on PC3 cells following treatment with varying concentrations of Cannabis sativa extract, cisplatin and cannabidiol. xCELLigence was also used to confirm the IC50 activity in which cells were grown in a 16 well plate coated with gold and monitor cell attachment. Caspase 3/7 activity was also measured using 96 well-plate following treatment. Western-blot and qRT-PCR was also used to measure the gene expression of tumor suppressor genes, p53, Bax and Bcl2. Animal studies were employed to measure the growth of PC3-mouse derived cancer to evaluate the effect of compounds in vivo. Results: From the treatment with varying concentrations of Cannabis sativa extract, cannabidiol and cisplatin, we have observed that the three compounds induced antiproliferation of PC3 cancer cell lines through the activation of caspase 3/7 activity. We also observed induction of apoptosis in these cells following silencing of retinoblastoma binding protein 6 (RBBP6), with upregulation of p53 and bax mRNA expression, and a reduction in Bcl2 gene expression. The growth of tumors in the mouse models were reduced following treatment with cisplatin and cannabidiol. Conclusion: We demonstrated that cannabidiol is a viable therapy to treat prostate cancer cells, in combination with silencing of RBBP6. Cannabis sativa Increases Seizure Severity and Brain Lipid Peroxidation in Pentylenetetrazole-Induced Kindling in Rats

The effect of Cannabis sativa extract on chemical kindling induced in rats by the repeated intraperitoneal (ip) injections of pentylenetetrazole (PTZ) was studied. Rats were treated with PTZ (35 mg/kg) once every 48 hours for 12 times alone or with ip Cannabis sativa (20 mg/kg expressed as Δ9-THC content) 30 min prior to PTZ injection. Seizures were recorded for 20 minutes. Control rats received ip saline. Cannabis treatment caused significant elevation of mean seizure score as compared to PTZ only group after the 5th, 6th and 7th PTZ repeated injections during seizure development. In particular, cannabis caused significant elevation in the frequency of myoclonic jerks, rearing (stage 3), turn over onto one side position and back position (stage 4), and generalized tonic-clonic seizures (stage 5) compared with the PTZ only group. PTZ caused significant elevations in brain lipid peroxidation (malondialdehyde), and nitric oxide along with deceased reduced glutathione level. In addition, brain acetylcholinesterase (AChE) activity significantly decreased compared to control value after PTZ treatment. Cannabis given to PTZ treated rats caused significant increase in brain malondialdehyde and AChE activity compared to PTZ only group. Reduced glutathione level was restored by cannabis. Histopathological studies indicated the presence of spongiform changes, degenerated and necrotic neurons, inflammatory cells, and gliosis in cerebral cortex and degeneration of some Purkinje cells in the cerebellum in both PTZ- and cannabis-PTZ-treated groups. It is concluded that in an epilepsy model induced by repeated PTZ administration, cannabis increased lipid peroxidation and mean seizure score.

The effect of hydroalcoholic extract of Cannabis Sativa on morphology and growth of bone marrow mesenchymal stem cells in rat

Background:One of the most important issues in cellular therapies is the examination of stem cells in different conditions. Given the increasing use of herbal drugs and herbal medicines and the potential effects of herbs on various diseases, the study of stem cells in the mediums of these compounds seems to be necessary.Aims:The current paper was to evaluate the effect of cannabis sativa extract on morphology, growth and maintenance of the characteristics of rat bone marrow stem cells.Methods:In this study, rat bone marrow mesenchymal stem cells were used. The effect of cannabis extract in different concentrations on cell morphology was investigated through microscopic observations and cytogenetic methods. In the context of the study of bioavailability and growth process, we used the mtt assay test and calculated the cell population doubling time and the growth curve plotting.Results:The results show that cannabis extract increases the proliferation of rat bone marrow mesenchymal stem cells compared to control group. Also, the effect of cannabis extract is dependent on its dose. The treated cells with a concentration of 100 ng / ml cannabis extract had the highest growth rate compared to control (P˂.050).Conclusion:Given the increasing impact of low doses of cannabis extract on the growth of bone marrow mesenchymal stem cells, it seems that the use of effective ingredients in this plant extract is beneficial for cell-therapeutic purposes in autoimmune diseases. On the other hand, higher doses cause cell death and an increase in apoptosis, so, in people consuming these substances, they will decrease the proliferation of stem cells due to increased concentration of substances in the body.

Preventive effects of cannabis on neurotoxic and hepatotoxic activities of malathion in rat

Objective: To investigate the effect of Cannabis sativa extract on the development of neuro- and hepato-toxicity caused by malathion injection in rats. Methods: The extract of Cannabis sativa was obtained from the plant resin by chloroform treatment. Δ-Tetrahydrocannabinol content of the extract (20%) was quantified using gas chromatography–mass spectrometry. The doses of cannabis extract were expressed as Δ -tetrahydrocannabinol content of 10 or 20 mg/kg. Malathion (150 mg/kg) was intraperitoneally administered followed after 30 min by the cannabis extract (10 or 20 mg/kg, subcutaneously). Rats were euthanized 4 h later. Acute Effects of Cannabis sativa on Ischaemia/Reperfusion Injury in the Rat Brain

We investigated the effect of Cannabis sativa extract on brain damage, oxidative stress and inflammation in rats with transient global cerebral ischaemia. Rats were subjected to bilateral common carotid artery (CCA) occlusion for 45 minutes followed by 4 h of reperfusion. Rats were treated with cannabis at a dose of 20 mg/kg (expressed as Δ9-THC) intraperitoneally (i.p) either before CCA, at time of reperfusion or after reperfusion. Alternatively, cannabis was given i.p. daily for 2 days before surgery. Markers of oxidative stress (malondialdehyde, reduced glutathione, nitric oxide) and the proinflammatory cytokine tumour necrosis factor-alpha (TNF-α) were determined in brain tissue. Histopathological evaluation was also done. Compared with the sham-treatment group, CCA occlusion resulted in increased brain malondialdehyde (54.5 ± 2.0 vs. 26.0 ± 1.45 nmol/g. tissue; p<0.05) and nitric oxide (75.2 ± 3.2 vs. 31.3 ± 3.0 mmol/g. tissue; p<0.05) contents along with decreased brain reduced glutathione (6.6 ± 0.14 vs. 8.28 ± 0.31mmol/g. tissue; p<0.05). There was also a pronounced rise in brain TNF-α concentrations (2248 ± 105 vs. 51.42 ± 3.21 pg/g. tissue; p<0.05). Cannabis sativa significantly increased reduced glutathione (by 30.3%-60.6%; p<0.05) and alleviated the increase in nitric oxide levels (by 51.5%- 58.5%; p<0.05) in the ischaemic brain tissue. Cannabis given before or at time of CCA occlusion significantly reduced brain TNF-α by 24.4% and 26.7%, respectively (1699 ± 80 and 1647 ± 54 vs. 2248 ± 105 pg/g. tissue; p<0.05). Histopathological examination of the cerebral cortex from rats subjected to CCA occlusion revealed gliosis, vacuolation and widespread neuronal degeneration. Cannabis given as a single dose 1h prior to CCA ligation or as 2 days pretreatment conferred protection against the ischaemic neuronal injury. It is concluded that in cerebral ischaemia the prior administration of cannabis exerted neuroprotective effects which could be accounted for by a decrease in nitric oxide and in the inflammatory response.

BACKGROUND AND OBJECTIVE: Epilepsy is one of the most common neurologic disorders that presents in a large number of drug-resistant patients. Given the presence of the anticonvulsant compounds in Cannabis and its use in traditional medicine, this study aims to evaluate the effect of Cannabis sativa extract on the treatment of seizures METHODS: In this experimental study, 40 Syrian male rats were selected and randomly divided into five groups, including one control, one sham, and three experimental groups. The control group only received pentylentetrazol (PTZ), the sham group was administered distilled water, and the experimental groups were gavaged with 400, 600, and 800 mg/kg of body weight of the aqueous extract of Cannabis sativa for four weeks. Moreover, 45 mg/kg of PTZ was intraperitoneally administered 30 minutes later in the experimental and sham groups. Finally, the convulsive behaviors and the relevant parameters were recorded using a camera inside a specific cage for 25 minutes. FINDINGS: Aqueous extract of Cannabis sativa increased the delay of seizure onset at the dose of 800 mg/kg compared to the control group (232.87±33.76 sec vs. 103.84±7.50 sec; p<0.05) and inhibited the progression of epilepsy phases equally at each three doses compared to the control group (1.91±0.5 vs. 5±0; p<0.00). CONCLUSION: The results showed that cannabis extract had the maximum anticonvulsant effect on PTZ-induced seizures at the dose of 800 mg.kg of body weight. Therefore, the anticonvulsant properties of aqueous extract of cannabis are dose-dependent.

Effect of Cannabis sativa extract on gastric acid secretion, oxidative stress and gastric mucosal integrity in rats

Studies were conducted in pylorus-ligated rats to investigate the effect of Cannabis sativa extract on gastric acid secretion, experimental gastric ulcer and on oxidative stress and inflammatory markers in the gastric mucosa. C. sativa (5, 10 and 20 mg/kg, expressed as Δ9-tetrahydrocannabinol) was administered subcutaneously daily for 4 weeks prior to pylorus ligation and different treatments. Under basal conditions, pretreatment with cannabis extract at doses of 5 and 10 mg/kg increased gastric acid secretion and induced minimally visible gastric mucosal lesions in the 4 h pylorus-ligated rat. Malondialdehyde and nitric acid concentration increased, while reduced glutathione decreased by cannabis at doses of 5 and 10 mg/kg in gastric mucosa. TNF-α increased by cannabis extract at doses of 5 and 10 mg/kg but decreased following the high dose of 20 mg/kg. On the other hand, the gastric acid secretory responses stimulated by pentagastrin or carbachol (but not histamine) were inhibited in rats pretreated with cannabis extract. Under these conditions, cannabis decreased pepsin content after pentagastrin and carbachol but not histamine stimulation. Cannabis also decreased lipid peroxidation and nitric oxide content, and increased both reduced glutathione and catalase activity in mucosa. Moreover, cannabis decreased mucosal inflammation (level of TNF-α) and the development of gastric mucosal lesions. Cannabis administered for 1 month prior to pylorus-ligation and either acidified aspirin or ethanol (96 %) decreased the development of gastric mucosal damage in a dose-dependent manner, along with reduction in gastric acid output, gastric mucosal oxidative stress and inflammation (TNF-α). Sections of gastric mucosa stained with periodic acid Schiff showed increased mucus secretion by cannabis in basal conditions and after treatment with aspirin or ethanol. Results indicate that: (1) the effect of cannabis differs in basal conditions and after exposure of the gastric mucosa to high acid concentrations or other chemical noxious agents; (2) cannabis administered systemically exerts gastric mucosal protective effects against mucosal damage evoked by stimulation of gastric acid secretion, acidified aspirin or ethanol. These effects of cannabis are likely to involve inhibition of gastric acid and pepsin secretion, increased mucus, decreased oxidative stress and inflammation in gastric mucosa.

Effect of Cannabis sativa on oxidative stress and organ damage after systemic endotoxin administration in mice

The effect of Cannabis sativa extract on oxidative stress and organ tissue damage during systemic inflammation was studied. For this purpose, Swiss mice were challenged with a single intraperitoneal dose of lipopolysaccharide (LPS; 200 μg/kg) to mimic aspects of mild systemic infection. Cannabis resin extract (5, 10, or 20 mg/kg) (expressed as Δ9-tetrahydrocannabinol) was given via subcutaneous route for 2 days prior to and at the time of endotoxin administration. Mice were euthanized 4 h after LPS injection. Malondialdehyde (MDA), reduced glutathione (GSH), and nitric oxide (nitrite/nitrate) in the brain, liver, kidney, lung, and heart as well as brain glucose were measured. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) were measured in liver homogenates. Histopathological examination of different organs was performed, and immunohistochemical techniques were used to evaluate expression levels of inducible nitric oxide synthase (iNOS) and caspase-3 in the brain and liver. The administration of only cannabis (20 mg/kg) decreased MDA, increased GSH, and decreased glucose level in the brain. No significant effects were observed for cannabis alone on MDA, GSH, or nitric oxide in other organs or on liver enzymes. The administration of LPS increased MDA and nitric oxide, while GSH decreased in different organs. Brain glucose increased by endotoxin. AST, ALT, and ALP were markedly increased in the liver tissue. In LPS-treated mice, cannabis (20 mg/kg) decreased MDA. GSH increased in the brain, kidney, and lung, nitric oxide decreased in the brain and lung while brain glucose decreased after the highest dose of cannabis. Cannabis failed to alter the level of liver enzymes. Histological damage in the brain, kidney, heart, lung, and liver due to endotoxin is increased by cannabis. Increased immunoreactivity of caspase-3 in the cytoplasm of the hepatocytes was observed after LPS and cannabis cotreatment compared with the LPS only group. Caspase-3 immunoreactivity markedly increased in degenerating neurons of the cortex following cannabis and LPS cotreatment. iNOS inmmunoreactivity increased after LPS and more intense iNOS expression was detected in hepatocytes after cannabis and LPS cotreatment. iNOS expression increased after cannabis and LPS treatment especially in the cerebral cortex. Thus, the administration of cannabis decreased tissue oxidative stress but increased organ damage after endotoxin injection in mice.

Effects of Cannabis sativa extract on haloperidol-induced catalepsy and oxidative stress in the mice.

Haloperidol is a classic antipsychotic drug known for its propensity to cause extrapyramidal symptoms due to blockade of dopamine D2 receptors in the striatum. Interest in medicinal uses of cannabis is growing. Cannabis sativa has been suggested as a possible adjunctive in treatment of Parkinson's disease. The present study aimed to investigate the effect of repeated administration of an extract of Cannabis sativa on catalepsy and brain oxidative stress induced by haloperidol administration in mice. Cannabis extract was given by subcutaneous route at 5, 10 or 20 mg/kg (expressed as Δ(9)-tetrahydrocannabinol) once daily for 18 days and the effect on haloperidol (1 mg/kg, i.p.)-induced catalepsy was examined at selected time intervals using the bar test. Mice were euthanized 18 days after starting cannabis injection when biochemical assays were carried out. Malondialdehyde (MDA), reduced glutathione (GSH) and nitric oxide (the concentrations of nitrite/nitrate) were determined in brain and liver. In saline-treated mice, no catalepsy was observed at doses of cannabis up to 20 mg/kg. Mice treated with haloperidol at the dose of 1 mg/kg, exhibited significant cataleptic response. Mice treated with cannabis and haloperidol showed significant decrease in catalepsy duration, compared with the haloperidol only treated group. This decrease in catalepsy duration was evident on days 1-12 after starting cannabis injection. Later the effect of cannabis was not apparent. The administration of only cannabis (10 or 20 mg/kg) decreased brain MDA by 17.5 and 21.8 %, respectively. The level of nitric oxide decreased by 18 % after cannabis at 20 mg/kg. Glucose in brain decreased by 20.1 % after 20 mg/kg of cannabis extract. The administration of only haloperidol increased MDA (22.2 %), decreased GSH (25.7 %) and increased brain nitric oxide by 44.1 %. The administration of cannabis (10 or 20 mg/kg) to haloperidol-treated mice resulted in a significant decrease in brain MDA and nitric oxide as well as a significant increase in GSH and glucose compared with the haloperidol-control group. Cannabis had no significant effects on liver MDA, GSH, nitric oxide in saline or haloperidol-treated mice. It is concluded that cannabis improves catalepsy induced by haloperidol though the effect is not maintained on repeated cannabis administration. Cannabis alters the oxidative status of the brain in favor of reducing lipid peroxidation, but reduces brain glucose, which would impair brain energetics.

Cannabis sativa exacerbates hepatic injury caused by acetaminophen or carbon tetrachloride in rats

The effect of Cannabis sativa extract on acute liver injury caused by acetaminophen or carbon tetrachloride (CCl4) was studied in rats. Cannabis sativa was given at doses of 5 or 10 mg/kg (expressed as Δ9-tetrahydrocannabinol) once daily intraperitoneally (i.p.) for 2 days and simultaneously with acetaminophen or CCl4. Rats were killed 24 h after acetaminophen or CCl4 administration. Reduced glutathione (GSH), lipid peroxidation (malondialdehyde; MDA) and nitric oxide (nitrite/nitrate) concentrations were measured in the liver. Alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were determined in serum. Hepatic injury was also determined via histological examination of liver sections. The administration of only cannabis for 2 days had no significant effect on serum liver enzymes or on the hepatic levels of GSH, MDA or nitric oxide. However, in rats intoxicated with acetaminophen, Cannabis sativa at 5 or 10 mg/kg resulted in a significant increase in serum GOT by 17.6% and 19.5%, respectively, compared with the acetaminophen control group. In the CCl4-induced acute liver injury, the levels of AST, ALT and ALP in serum were significantly elevated by Cannabis sativa extract in a dose-dependent manner by 23.7–29.1%, 14.4–21.3% and 17.6–22.1%, respectively. In both models of hepatic injury, Cannabis sativa resulted in a significant increase in the level of liver MDA and nitric oxide and a significant decrease in GSH compared with the corresponding acetaminophen or CCl4 control group. These changes were dose dependent. Histological examination showed an increase in centrilobular necrotic areas in acetaminophen or CCl4-treated rats administered with Cannabis sativa. Histochemical investigation revealed a decrease in intracellular protein contents caused by CCl4 or acetaminophen, and these were further decreased by Cannabis sativa. It is concluded that short-term administration of Cannabis sativa enhances acute hepatic damage caused by CCl4 or acetaminophen in rats.

Effects of Cannabis sativa extract on conditioned fear

Male and female rats were chronically or acutely treated with marihuana extract before or after being subjected to a conditioned emotional response (CER) paradigm. Acute administration of the marihuana compound partially blocked both acquisition and retention of conditioned fear. After chronic administration, however, only retention of the conditioned suppression was affected.

Extinction of operant responses by rats under the effects of Cannabis sativa extract

Rats previously trained in T- and Lashley III mazes and injected with cannabis extract during extinction showed an equal trend to extinguish when compared to control animals. On the other hand, rats which had been trained in the T-maze under marihuana action extinguished as controls when extinction sessions were carried out without drugs. These data speak against the hypothesis, put forth to explain the effects of cannabis in reducing CER of rats, that marihuana compounds potentiate dominant responses.
