Simulated nitrogen deposition alters the ectomycorrhizal fungal community structure in northern Korean pine plantations

Northern temperate forests are sensitive to nitrogen deposition. Korean pines are the zonal dominant vegetation in these forest ecosystems and a typical obligate ectomycorrhizal (ECM) tree. However, the impact of varying nitrogen additions on the ectomycorrhizal fungal (ECMF) community structure in Korean pine plantations remains unclear. In this experiment, nitrogen deposition was simulated (CK = 0 kg N ha−1 yr−1, LN = 20 kg N ha−1 yr−1, MN = 40 kg N ha−1 yr−1, HN = 80 kg N ha−1 yr−1), and soil biochemical indicators, ECMF community composition, and diversity were measured. The results showed that high nitrogen addition led to increased ECMF colonization, decreased ECMF community diversity and richness, and altered the occurrence of biomarkers. The MN treatment significantly increased the Simpson index compared to the LN treatment (P < 0.05), with Amphinema sp, Tuber sp, and Tylospora asterophora identified as biomarkers in the CK, LN, and HN treatments, respectively. The HN treatment significantly reduced the Chao1 index (P < 0.05), with Inocybe quietiodor, Tarzetta catinus, Inocybe giacomi, and Russula viscida serving as biomarkers in the CK, LN, MN, and HN treatments, respectively. Nitrogen addition led to decreased soil pH and available phosphorus content, increased ammonium nitrogen content, and affected ectomycorrhiza colonization by altering the soil ECMF community structure. Among these factors, ammonium nitrogen was the most influential in changing the ECMF community structure. This research offers valuable data to help predict changes in ECMF community structure in Korean pine plantations as nitrogen additions increase.

Effects of resin tapping on ectomycorrhizal fungal community composition and structure of Pinus massoniana in subtropical mountain forest ecosystems in southwestern China

Although resin tapping is an important management practice in Pinus massoniana plantation, our understanding of its influence on symbiotic ectomycorrhizal fungal (EMF) communities associated with P. massoniana remains limited. To determine the effect of resin tapping on the EMF community, this study observed the EMF community in resin-tapped (RT) and resin-untapped (NRT) P. massoniana plantation using community surveys, soil property analysis, and mycorrhizal identification. A total of 49 operational taxonomic units (OTUs), belonging to 15 families and 18 genera, were identified. At the phylum level, the relative abundance of Basidiomycota and Ascomycota accounted for 82.46% and 17.54% of the total population, respectively. Resin tapping significantly altered the composition and structure of the EMF community of P. massoniana, and the number of endemic OTUs was higher than that of the shared OTUs. Lactarius and Tomentella were dominant in RT plantation, and Russula and Tomentella were dominant in NRT plantations. There was no significant difference in the alpha diversity of EMF communities between RT and NRT Plantation. Principal coordinates analysis revealed marked differences in the composition of EMF communities between R and NR plantation. Canonical correspondence analysis showed that available K, total N, and urease were important factors influencing the EMF community of P massoniana. In conclusion, resin tapping affects the composition and structure of EMF communities associated with P. massoniana plantation in subtropical mountain ecosystems.

Soil nutrients and plant diversity affect ectomycorrhizal fungal community structure and functional traits across three subalpine coniferous forests.

The symbiotic relationship between ectomycorrhizal fungi (EMF) and the roots of host plants is significantly important in regulating the health and stability of ecosystems, especially of those such as the climate warming affected subalpine forest ecosystems. Therefore, from the coniferous forest systems located in the Southern Qinghai-Tibetan Plateau, root tips from three forest tree species: Pinus wallichiana, Abies spectabilis and Picea spinulosa, were collected to look for the local causes of EMF community composition and diversity patterns. The EMF colonization rate, diversity and taxonomic community structure were determined by morphotyping and sanger sequencing of the fungal ITS gene from the root tip samples. Soil exploration types were identified based on the morphologies of the ectomycorrhizas, coupled with soil properties analysis and plant diversity survey. Contrasting patterns of EMF community and functional diversity were found across the studied three forests types dominated by different coniferous tree species. In terms of associations between soil and EMF properties, the total phosphorus (TP) and nitrate (NO3−) contents in soil negatively correlated with the colonization rate and the Shannon diversity index of EMF in contrast to the positive relationship between TP and EMF richness. The soil total nitrogen (TN), ammonium (NH4+) and plant diversity together caused 57.6% of the total variations in the EMF taxonomic community structure at the three investigated forest systems. Whereas based on the soil exploration types alone, NH4+ and TN explained 74.2% of variance in the EMF community structures. Overall, the findings of this study leverage our understanding of EMF dynamics and local influencing factors in coniferous forests dominated by different tree species within the subalpine climatic zone.

Traits and tradeoffs among non-native ectomycorrhizal fungal symbionts affect pine seedling establishment in a Hawaiian coinvasion landscape.

Pine invasions lead to losses of native biodiversity and ecosystem function, but pine invasion success is often linked to coinvading non-native ectomycorrhizal (EM) fungi. How the community composition, traits, and distributions of these fungi vary over the landscape and how this affects pine success is understudied. A greenhouse bioassay experiment was performed to test the effects of changes in EM fungal community structure from a pine plantation, to an invasion front to currently pine-free areas on percent root colonization and seedling biomass. Soils were also analysed by qPCR to determine changes in inoculum and spore density over distance for a common coinvading EM fungus, Suillus pungens. Percent colonization increased with distance from the plantation, which corresponded with an increase in seedling biomass and stark changes in EM fungal community membership where Suillus spp. dominated currently pine-free areas. However, there was a negative relationship between S.pungens inoculum potential versus root colonization over distance. We conclude that the success of pine invasions is facilitated by specific traits of Suillus spp., but that the success of Suillus is contingent on a lack of competition with other ectomycorrhizal fungi.

Land‐use history alters the diversity, community composition and interaction networks of ectomycorrhizal fungi in beech forests

Abstract Forests have expanded across Europe over the last centuries as a consequence of farmland (agricultural and pasture) abandonment. Agricultural practices usually increase soil fertility and reduce the diversity and abundance of ectomycorrhizal (ECM) fungi, essential mutualists of many woody species in temperate and Mediterranean forests. The recovery of this biotic interaction after the cessation of human activities is, thus, crucial for the re‐establishment of functional forest ecosystems. Here we addressed the legacies of past land use and the recovery of the mutualism between ECM fungi and Fagus sylvatica trees in recent beech forests (&lt;50 years) in Northeast Spain. Soil and root samples were collected in six long‐established and eight recent beech forests to analyse soil abiotic properties and the ECM fungi associated with beech roots (Illumina DNA metabarcoding of ECM tips). Up to 609 amplicon sequence variants (ASVs) of ECM fungi were identified, with 220 ASVs found in both forest types. Recent forests had a significantly lower soil organic matter and phosphorus content, which had a significant effect on the community structure of ECM fungi in beech roots. Moreover, beech trees in recent forests interacted with less fungal taxa but had a higher relative abundance of Ascomycota. Tarzetta spp. (Ascomycota, Pezizales) and Lactarius blennius (Basidiomycota, Russulales) emerge as the particular taxa associated with recent and long‐established forests respectively. More specialized mutualistic networks with a lower species normalized degree were found in recent forests, which might result in a lower quality and resilience of the ECM mutualism. Synthesis. Land‐use history modulated the mycorrhizal symbiosis in regenerating beech forests through changes in soil organic matter and nitrogen, which were the main drivers of the differences in fungal community composition and functional types associated with beech trees in recent forests.

Diversity of Ectomycorrhizal Fungal Communities in Four Types of Stands in Pinus massoniana Plantation in the West of China

Ectomycorrhizal (ECM) fungi can form symbioses with plant roots, which play an important role in regulating the rhizosphere microenvironment. As a broad-spectrum ECM tree species, Pinus massoniana forms symbiotic relationship called mycorrhiza with various ECM fungal species. In this study, four types of forests were selected from a 38-year-old Pinus plantation in eastern Sichuan, namely, pure P. massoniana forest (MC), P. massoniana mixed with Cunninghamia lanceolata forest (MS), P. massoniana–Cryptomeria fortunei forest (ML), and P. massoniana–broadleaved forest (MK), the species mixture ratio of all forests was 1:1. The ITS2 segment of ECM root tip sequenced by high-throughput sequencing using the Illumina MiSeq sequencing platform. (1) The ECM fungi of these four P. massoniana forests showed similar dominant genera but different relative abundances in community structure during the three seasons. (2) The alpha diversity index of ECM fungi was significantly influenced by season and forest type. (3) Soil pH, soil organic matter (SOM), total nitrogen (TN), C/N ratio, and total phosphorus (TP) influenced the ECM fungal community structure in different seasons. In summary, there were significant differences in ECM fungal communities among different forest types and different seasons; the colonization rate of ECM fungal in P. massoniana–Cunninghamia lanceolata was the highest, so we infer that Cunninghamia lanceolata is the most suitable tree species for mixed with P. massoniana in three mixture forests.

Diversity and community structure of ectomycorrhizal fungi in Pinus thunbergii coastal forests bordering the Yellow Sea of China.

Ectomycorrhizas play a fundamental role in the function of forest ecosystems, being essential for plant nutrition absorption and soil quality. Many afforestation and reforestation programmes have begun to recover and maintain coastal forests in China, using pine species including Pinus thunbergii. We investigated the ectomycorrhizal colonization status of P. thunbergii in coastal pine forests of the Yellow Sea of China. We identified a total of 53 ectomycorrhizal fungal species in 74 soil samples collected from three sites and found that Thelephoraceae (10 spp.) and Russulaceae (8 spp.) were the most species-rich ectomycorrhizal fungal lineages. Russula sp. 1 was the most abundant species, accounting for 15.3% of the total ectomycorrhizal tips identified. Most of the remaining species were rare. At this small scale, host identity had no significant effect on the ectomycorrhizal fungal community composition (A = 0.036, P = 0.258), but sampling sites did (A = 0.135, P = 0.041). In addition, Na+ and K+ content and soil pH had significant effects on the ectomycorrhizal fungal community. The ectomycorrhizal fungal community associated with different host plants will become an important new direction for research, as ectomycorrhiza may have the potential to improve host capacity to establish in salt-stressed environments. This will provide a theoretical basis and technical support for saline soil reforestation and rehabilitation using pine species with compatible, native ectomycorrhizal fungi in Yellow Sea coastal areas.

Characterizing Ectomycorrhizal Fungal Community Structure and Function of Two Varieties of Pinus clausa That Differ in Disturbance History

Despite the immense amount of diversity present in the soil biota, the ecological and evolutionary processes that regulate species diversity and abundance of ectomycorrhizal (ECM) fungi across space and time remain elusive. In forest ecosystems, ECM fungal diversity may be maintained by periodic disturbances which operate at different time scales due to their effects on host genetic and phenotypic characteristics and the associated environment. To investigate the degree to which these factors shape ECM fungal community composition and function, I sampled 10 independent sites for a pine species indicative of an endangered ecosystem, the Florida scrub, where disturbance history has driven the divergence of a single species into two genetically distinct varieties (Pinus clausa var. immuginata and var. clausa). A total of 300 ECM fungal species were identified based on rDNA ITS sequences, but each variety harbors different ECM species composition and function. A follow-up greenhouse experiment, in which the seed from each variety was grown in its own soil (“home”) and in the soil of the other variety (“away”), suggests these communities differentially impact the growth of their host seedlings. While var. clausa seedlings had the same total biomass regardless of soil origin, var. immuginata had higher biomass in their own soil compared to var. clausa. This is likely due to an increased number of ECM colonized tips in the home soil compared to in away soil. Taken together, these results may suggest different evolutionary histories where structure host genetic and phenotypic characteristics may be important for structuring their dynamics with ECM fungi.

Ectomycorrhizal fungal community structure in a young orchard of grafted and ungrafted hybrid chestnut saplings

Ectomycorrhizal (ECM) fungal community of the European chestnut has been poorly investigated, and mostly by sporocarp sampling. We proposed the study of the ECM fungal community of 2-year-old chestnut hybrids Castanea × coudercii (Castanea sativa × Castanea crenata) using molecular approaches. By using the chestnut hybrid clones 111 and 125, we assessed the impact of grafting on ECM colonization rate, species diversity, and fungal community composition. The clone type did not have an impact on the studied variables; however, grafting significantly influenced ECM colonization rate in clone 111. Species diversity and richness did not vary between the experimental groups. Grafted and ungrafted plants of clone 111 had a different ECM fungal species composition. Sequence data from ITS regions of rDNA revealed the presence of 9 orders, 15 families, 19 genera, and 27 species of ECM fungi, most of them generalist, early-stage species. Thirteen new taxa were described in association with chestnuts. The basidiomycetes Agaricales (13 taxa) and Boletales (11 taxa) represented 36% and 31%, of the total sampled ECM fungal taxa, respectively. Scleroderma citrinum, S. areolatum, and S. polyrhizum (Boletales) were found in 86% of the trees and represented 39% of total ECM root tips. The ascomycete Cenococcum geophilum (Mytilinidiales) was found in 80% of the trees but accounted only for 6% of the colonized root tips. These results could help to unveil the impact of grafting on fungal symbionts, improving management of chestnut agro-ecosystems and production of edible fungal species.

Profiling the community structure of ectomycorrhizal fungi at endemic pine (Pinus greggii) stands of northeastern Mexico

The biological communities of endemic, relictual ecosystems are poorly known and the rate of destruction of these ecosystems is high worldwide. Pinus greggii is a near-threatened Mexican pine endemic species with a highly restricted distribution. In healthy tree stands, the ectomycorrhizal (ECM) symbiosis of trees and fungi is of primary importance; however, the community structure of ECM associated with natural stands of P. greggii has not been studied. Using next-generation sequencing (NGS) (= high-throughput sequencing, HTS), the community of ECM at four natural P. greggii var. greggii stands in the northern Sierra Madre Oriental of Mexico (states of Coahuila and Nuevo Leon) was analysed. A total of 47 516 sequences (runs) and 139 operational taxonomic units (OTUs) of ECM associated with all stands were detected. These ECM communities indicate a community pattern similar to those of boreal forests (albeit with some remarkable absences of taxa), and different from ECM species structures from Neotropical forests. The near absence of Amanitaceae, Boletaceae and Clavulinaceae, and the broad occurrence of gasteroid and corticioid, effuse ECM fungi probably indicates a ECM community adapted to aridity. Many OTUs not assignable to species were detected, which possibly correspond to undescribed taxa. Three genera and 20 species associated with these P. greggii stands are new records for Mexico, including species not previously detected in the Western hemisphere. The most prevalent OTUs across stands belonged to species of Tomentella (25.5%). Taxa in the families Inocybaceae, Rhizopogonaceae, Russulaceae, Sebacinaceae and Tuberaceae were also frequent. NGS-HTS is a powerful and sensitive suite of techniques that yields critical information on ECM communities in forests. This report provides baseline information for future research on biogeography, ecology and applied forestry of pine stands in Mexico, especially P. greggii and associated ECM.

Drivers of Ectomycorrhizal Fungal Community Structure Associated with Pinus sylvestris var. mongolica Differ at Regional vs. Local Spatial Scales in Northern China

Pinus sylvestris var. mongolica, a widely planted tree species, is facing long-lasting, unresolved degradation in desertified Northern China. Ectomycorrhizal fungi (EMF) are closely related to the stand status, because they substantially participate in ecological processes of terrestrial forest ecosystems. EMF may be key to solving the introduction recession. Therefore, we performed DNA sequencing of P. sylvestris root samples from plantations and natural forests as control to characterize the EMF from semi-arid and dry sub-humid regions, using ITS Illumina sequencing and conventional soil physicochemical index determination. The results indicated that (1) the dominant EMF genera were Suillus, Rhizopogon, and Wilcoxina in the Hulunbuir, Mu Us, and Horqin Sandy Lands, respectively. Their dominance retained with stand ageing. (2) Plantation EM fungal diversity differs significantly among the three sandy lands and was significantly lower than in natural forest. The diversity varied with stand age, showing distinct trends at the local scale. (3) At the regional scale, the mean annual sunshine times and the soil organic carbon content affect EMF diversity. The community composition and structure were more characterized by temperature and precipitation. At the local scale, besides the soil organic carbon content, the EM fungal community composition and structure were correlated with total nitrogen and phosphorus content (Hulunbuir), the total phosphorus content (Mu Us), and the pH and total soil porosity (Horqin). The EM fungal community composition and structure have the obvious geographical distribution variation; they were strongly correlated with the meteorological elements and soil nutrients at the regional scale. At the local scale, they were jointly driven by stand age and soil properties. This improved information contributes to increasing the understanding of the interaction between EMF and forest ecosystems and guides sustainable forest management of degraded P. sylvestris plantations.

Large-scale insect outbreak homogenizes the spatial structure of ectomycorrhizal fungal communities.

Ectomycorrhizal fungi (plant symbionts) are diverse and exist within spatially variable communities that play fundamental roles in the functioning of terrestrial ecosystems. However, the underlying ecological mechanisms that maintain and regulate the spatial structuring of ectomycorrhizal fungal communities are both complex and remain poorly understood. Here, we use a gradient of mountain pine beetle (Dendroctonus ponderosae) induced tree mortality across eleven stands in lodgepole pine (Pinus contorta) forests of western Canada to investigate: (i) the degree to which spatial structure varies within this fungal group, and (ii) how these patterns may be driven by the relative importance of tree mortality from changes in understory plant diversity, productivity and fine root biomass following tree death. We found that the homogeneity of the ectomycorrhizal fungal community increased with increasing tree death, aboveground understory productivity and diversity. Whereas, the independent effect of fine root biomass, which declined along the same gradient of tree mortality, increased the heterogeneity of the ectomycorrhizal fungal community. Together, our results demonstrate that large-scale biotic disturbance homogenizes the spatial patterns of ectomycorrhizal fungal communities.

Common garden experiments disentangle plant genetic and environmental contributions to ectomycorrhizal fungal community structure.

The interactions among climate change, plant genetic variation and fungal mutualists are poorly understood, but probably important to plant survival under drought. We examined these interactions by studying the ectomycorrhizal fungal (EMF) communities of pinyon pine seedlings (Pinus edulis) planted in a wildland ecosystem experiencing two decades of climate change-related drought. We established a common garden containing P.edulis seedlings of known maternal lineages (drought tolerant, DT; drought intolerant, DI), manipulated soil moisture and measured EMF community structure and seedling growth. Three findings emerged: EMF community composition differed at the phylum level between DT and DI seedlings, and diversity was two-fold greater in DT than in DI seedlings. EMF communities of DT seedlings did not shift with water treatment and were dominated by an ascomycete, Geopora sp. By contrast, DI seedlings shifted to basidiomycete dominance with increased moisture, demonstrating a lineage by environment interaction. DT seedlings grew larger than DI seedlings in high (28%) and low (50%) watering treatments. These results show that inherited plant traits strongly influence microbial communities, interacting with drought to affect seedling performance. These interactions and their potential feedback effects may influence the success of trees, such as P.edulis, in future climates.

Effects of simulated nitrogen deposition on ectomycorrhizae community structure in hybrid larch and its parents grown in volcanic ash soil: The role of phosphorous

With the rapid industrial development and modern agricultural practices, increasing nitrogen (N) deposition can cause nutrient imbalance in immature volcanic ash soil commonly found in Japan. Larch species, widely distributed in northeast Eurasia, are associated with ectomycorrhizal (ECM) fungi which play a critical role in nutrient acquisition for their hosts. In this study, we investigated species richness and diversity of ECM fungi associated with a hybrid larch (F1) and its parents, Dahurian larch (Larix gmelinii var. japonica) and Japanese larch (L. kaempferi), under simulated N deposition (0 and 100kgha−1yr−1) with/without phosphorous (P) (0 and 50kgha−1yr−1). Seedlings planted in immature volcanic ash with low nutrient availability were subjected to the N and P treatments for fifteen months. We found that response of ECM community structure to the increased nutrient availability depended on host genotypes. Nutrient addition significantly affected ECM structure in Japanese larch, but no such significant effect was found for Dahurian larch. Effects of the nutrient addition to ECM fungal community in F1 were intermediate. F1 was tolerant to high N loading, which was due to consistent, relatively high association with Suillus sp. and Hebeloma sp. F1 showed heterosis in relative biomass, which was most apparent under high N treatments. This co-variation of ECM fungal community structure and F1 biomass in response to N loading suggest that ECM community structure might play an important role in host growth. The present findings indicate effects of N deposition on ECM fungal community structure can depend on larch species, thus it is challenging to predict general trends.

Diversity and community structure of ectomycorrhizal fungi associated with Larix chinensis across the alpine treeline ecotone of Taibai Mountain.

Alpine treeline ecotones represent ecosystems that are vulnerable to climate change. We investigated the ectomycorrhizal (ECM) community, which has potential to stabilize alpine ecosystems. ECM communities associated with Larix chinensis were studied in four zones along a natural ecotone from a mixed forest stand over pure forest stands, the timberline, and eventually, the treeline (3050-3450m) in Tabai Mountain, China. Sixty operational taxonomic units (OTUs) of ECM fungi were identified by sequencing the rDNA internal transcribed spacer of ECM tips. The richness of ECM species increased with elevation. The soil C/N ratio was the most important factor explaining ECM species richness. The treeline zone harbored some unique ECM fungi whereas no unique genera were observed in the timberline and pure forest zone. Elevation and topography were equally important factors influencing ECM communities in the alpine region. We suggest that a higher diversity of the ECM fungal community associated with L. chinensis in the treeline zone could result from niche differentiation.

Ectomycorrhizal fungal response to warming is linked to poor host performance at the boreal-temperate ecotone.

Rising temperatures associated with climate change have been shown to negatively affect the photosynthetic rates of boreal forest tree saplings at their southern range limits. To quantify the responses of ectomycorrhizal (EM) fungal communities associated with poorly performing hosts, we sampled the roots of Betula papyrifera and Abies balsamea saplings growing in the B4Warmed (Boreal Forest Warming at an Ecotone in Danger) experiment. EM fungi on the root systems of both hosts were compared from ambient and +3.4°C air and soil warmed plots at two sites in northern Minnesota. EM fungal communities were assessed with high-throughput sequencing along with measures of plant photosynthesis, soil temperature, moisture, and nitrogen. Warming selectively altered EM fungal community composition at both the phylum and genus levels, but had no significant effect on EM fungal operational taxonomic unit (OTU) diversity. Notably, warming strongly favored EM Ascomycetes and EM fungi with short-contact hyphal exploration types. Declining host photosynthetic rates were also significantly inversely correlated with EM Ascomycete and EM short-contact exploration type abundance, which may reflect a shift to less carbon demanding fungi due to lower photosynthetic capacity. Given the variation in EM host responses to warming, both within and between ecosystems, better understanding the link between host performance and EM fungal community structure will to clarify how climate change effects cascade belowground.

Edaphic Selection Pressures as Drivers of Contrasting White Spruce Ectomycorrhizal Fungal Community Structure and Diversity in the Canadian Boreal Forest of Abitibi-Témiscamingue Region.

Little is known about edaphic selection pressures as drivers of contrasting white spruce ectomycorrhizal fungal community structure and diversity in the Canadian boreal forest. We hypothesized that community composition differs among the four sites sampled–nursery, mining site, forest edge, and natural forest. Ectomycorrhizal (ECM) fungal community structure and diversity was studied at the four locations with soil fertility gradient through morpho-molecular and phylogenetic analyses in relationships with rhizospheric soil chemical properties. 41 different species were identified. Mining site had a significantly different species composition than the surrounding environments. Soil pH and percentage of roots colonized by ECM fungi increased while soil P, N, Fe, C, K, Mg, Al, Ca, and Na contents declined across the soil fertility gradient: nursery → natural forest → forest edge → mining site. Contrary to the preference of acid soils by ECM fungi, a few ecologically adapted to high pH, poor soil chemical fertility, and low organic matter content colonize white spruce roots on the non-acidogenic mining site, allowing natural regeneration of white spruce seedlings. Other ECM fungi are adapted to high fertigation level of commercial nursery. This study clearly shows the contrasting difference in white spruce ectomycorrhizal fungal community structure and diversity driven by edaphic selection pressures.

The effect of environmental contamination on the community structure and fructification of ectomycorrhizal fungi.

Ectomycorrhizal fungi are an essential component of forest ecosystems, most of which can form edible and medical fruiting bodies. Although many studies have focused on the fructification of ectomycorrhizal fungi in phenology, the impact of environmental contamination, especially living garbage, on the formation of fruiting body is still unknown. A field investigation, combined with a high‐throughput sequencing method, was used to study the effect of living garbage pollution on the fructification and hypogeous community structure of ectomycorrhizal fungi symbiosing with cedar (Cedrus deodara (Roxb.) G. Don). The results showed that garbage significantly altered soil abiotic and biotic properties, increasing soil urease activity, decreasing the soil exchangeable metal content and phosphatase activity, and ultimately inhibiting the formation of fruiting bodies. The pollution of garbage also changed the community structure of hypogeous ectomycorrhizal fungi where ectomycorrhizal ascomycetes dominated. In unpolluted sites, the relative abundance of ectomycorrhizal ascomycetes and basidiomycetes were almost equal. Although no fruiting bodies were observed in that soil polluted by living garbage, the sequencing result showed that various ectomycorrhizal fungi were present underground, suggesting that these taxonomic fungi had the potential to cope with adverse conditions. This study not only provided a deeper understanding of the relationship between ectomycorrhizal fungal communities and prevailing environmental conditions, but provided a new pathway for the excavation and utilization of the resource of antistress ectomycorrhizal fungi.

Tree genotype influences ectomycorrhizal fungal community structure: Ecological and evolutionary implications

Abstract Although the eco-evolutionary dynamics of multicellular organisms are intertwined with the microorganisms that colonize them, there is only a rudimentary understanding of how a host's genotype influences its microbiome. We utilize Populus angustifolia to test whether communities of essential symbionts, ectomycorrhizal fungi (EMF), vary among host genotypes. Further, we test whether EMF communities covary among tree genotypes with the chemistry of senescent leaves and aboveground biomass, traits important to tree fitness, and carbon and nutrient cycling. We found: 1) EMF composition, colonization and the Basidiomycota to Ascomycota ratio varied among tree genotypes (broad-sense heritability = 0.10–0.25). 2) EMF composition did not covary among genotypes with aboveground biomass but it did covary with senescent leaf chemistry ( rho = 0.29), primarily due to a single genotype. These findings demonstrate a link between tree genotype and EMF communities, which has implications for fungal diversity, host-symbiont interactions and aboveground-belowground linkages in ecological and evolutionary contexts.

Phosphorus availabilities in beech (Fagus sylvatica L.) forests impose habitat filtering on ectomycorrhizal communities and impact tree nutrition

Phosphorus (P) is an important nutrient, whose concentrations are declining in many European forest ecosystems. Here, we selected five old-aged temperate beech (Fagus sylvatica) forests that represented a sequence of decreasing soil P resources. We addressed the following hypotheses: (i) root P concentrations correspond to soil P concentrations, when P availability is suboptimal for tree nutrition, (ii) decreasing soil P concentrations, and increasing host P demand foster increasing ectomycorrhizal fungal (EMF) species richness and lead to a shift in the EMF community structure towards increasing soil exploration. We found that the decrease in P concentrations along the geosequence was less steep in the organic layer than that in the mineral topsoil. P concentrations in roots showed a positive relationship with P concentrations in soil, with a stronger correlation in coarse than in fine roots. This finding indicates that low P availability mainly affected P storage of the host. The root tips were completely colonized with EMF. In the organic layer EMF biomass was higher than that of saprophytic fungi, and correlated with inorganic P (Pi). In the mineral topsoil EMF biomass was about 10-fold lower than in the organic layer and biomass of saprophytes and microbial P, but not that of EMF, was correlated with Pi and phosphatase activities. Based on these results, we propose that beech P nutrition was mainly achieved by EMF in the organic layer. Variation in EMF species richness was unrelated to P in soil and decreased with increasing N in the organic layer. The EMF community structures were taxonomically divergent and filtered by habitat soil chemistry in the mineral layer and Pi in the organic layer between the P-rich forest and the P-poor forest. Changes in the taxonomic structures of the EMF did not result in corresponding changes in soil exploration. In conclusion, our results support a relationship between soil P concentrations and P storage in roots, but do not support mono-causal relationships between soil P and EMF species richness or hyphal soil exploration. Our results suggest that the taxonomic dissimilarities of the EMF along the P gradient were mainly driven by Pi concentrations in the organic layer and by the nutrient resources in the mineral layer.
