Characterization of Hippo Signaling Components in the Early Dorsal Pancreatic Bud.

YAP1 expression in the early mouse pancreatic anlagen is low until approximately E11.5, when it becomes localized to cell nuclei in multipotent progenitor cells. At E14.5, we find nuclear YAP1 in ductal cells.YAP1 is not expressed in early and midgestation endocrine cells. By contrast, TAZ is expressed in first transition endocrine cells.Hippo regulators MERLIN and LATS1/2 kinases are robustly expressed in the early pancreatic bud by E10.5. Both MERLIN and LATS1/2 exhibit strong apical localization in epithelial cells at nascent microlumens. Using in vitro models of de novo pancreas lumen formation, we show that YAP1 nuclear localization is high in early phases of lumen formation and gradually decreases as lumens matures.

Effect of Organic Substrates on Macro-propagation of Banana under Island Conditions

Banana is an important tropical fruit crop grown globally and is the world’s most important agricultural food commodity. Plantation crops take a major share of cultivated area in Andaman and Nicobar Islands. Banana is one of the important fruit crops that is highly suitable to grow as intercrop in plantation based cropping system. Local varieties of banana like Korangi, Mitta Champa, Khatta Champa and Cheenakela are cultivated by the farmers traditionally to meet the local demand. However, in recent past growing commercial varieties of banana has become popular in the Island due to consumer demand. Traditionally banana is propagated by suckers and wide variability was observed in sucker production among the commercial varieties. Due to this large scale multiplication of banana through suckers could not meet the planting material demand in the Island. Hence, a study was undertaken during 2020-2022 to standardize the macro-propagation technology using different organic substrates in the commercial variety ‘Poovan’. Nine different organic substrate combinations were used in the present study to enhance the efficacy of lateral bud development and plantlet production. Suckers weighing 1.0 to 1.5 kg were used as the propagule. Among all the organic media combinations tested, significant differences were observed in terms of plantlet production, root and shoot growth. The treatment T8 (Coircompost: sawdust: FYM: vermicompost (1:1:1:1) with Arka microbial consortia @10 kg) produced maximum number of primary (5.60) and secondary buds (12.07) during primary and secondary capitation of decorticated mother rhizome. Early bud emergence (11.5 days), maximum number of plantlets (12-13 per mother rhizome), maximum shoot length (46.43 cm), maximum shoot girth (3.03 cm) and maximum number of leaves (4.50) were also observed in the treatment T 8. Minimum number of days was taken (52.70 days) for plantlet separation during secondary capitation in the same treatment T 8 suggesting that the treatment combination (Coircompost: sawdust: FYM: vermicompost (1:1:1:1) with Arka microbial consortia @10 kg) was effective for macro-propagation of banana Cv. Poovan under Island ecosystem.

Impact of Zinc Application on Growth, Yield, and Quality of African Marigold in Semi-Arid Conditions

Marigold is a widely cultivated, hardy flower crop known for its adaptability. Among the various intercultural practices in marigold cultivation, foliar application of zinc plays a critical role in enhancing yield and quality, however, standardization of this practice is necessary to optimize commercial production. Hence, the investigation aimed to standardize foliar zinc application for improving productivity in African marigold (Pusa Narangi Gainda) at Rajasthan Agricultural Research Institute, Jaipur, during 2020-21, and 2021-22. Two concentrations of zinc sulphate (ZnSO₄), Z1 (0.2%), and Z2 (0.5%), were applied at 35 and 65 days after transplanting (DAT) in a factorial randomized block design with three replications. Results indicated that Z1 (ZnSO₄ 0.2%) was most effective in increasing plant height, and promoting early bud initiation, and flower opening, while Z2 (ZnSO₄ 0.5%) significantly enhanced plant spread, primary branching, leaf dimensions, plant biomass, root-to-shoot ratio, flower diameter, and yield parameters such as flower mass and number. Z2 also improved quality traits like chlorophyll and xanthophyll content. Statistical analysis revealed significant differences among treatments, suggesting that the application of ZnSO₄ at 0.5% may be recommended for optimizing growth, yield, and quality in commercial marigold cultivation, thereby enhancing productivity and profitability.

Effect of Pinching on Growth, Yield, and Quality of African Marigold (Tagetes erecta L.) cv. Pusa Narangi Gainda

Marigold is a very common and a hardy flower crop cultivated throughout the world. Pinching is an important intercultural practice commonly followed in cultivation of marigold however, this needs to be standardized for getting higher yields in commercial cultivation. Hence, the present investigation was conducted at Rajasthan Agricultural Research Institute, Durgapura, Jaipur, Division of Horticulture, during 2020-21 and 2021-22 to standardize the commercial pinching practice for getting higher productivity. Three levels of pinching (no pinching, single at 20 days and double at 20 and 40 days after transplanting) were used in the course of the research arranged using a factorial randomized block layout with three replications applied to African marigold variety Pusa Narangi Gainda. The results revealed that treatment P0 (No pinching) was discovered to be the finest in increment in height of plant, early flower bud initiation flower opening and treatment P2 (Double pinching) in boosting spread of plant, primary branches, length of leaf, leaf width, leaf area, plant fresh weight, plant dry weight, total biomass, root- shoot ratio, 50% flowering, flower diameter, stalk and ray floret length, fresh and dry flower weight, mass and number of flower per plant, flower yield additionally improved quality characteristics like chlorophyll and xanthophyll content in marigold. Hence, it might be concluded that application of double pinching can be helpful and recommended in improving yield as well as quality characteristics in commercial cultivation of marigold.

The BnaBPs gene regulates flowering time and leaf angle in Brassica napus.

The flowering time and plant architecture of Brassica napus were significantly associated with yield. In this study, we found that the BREVIPEDICELLUS/KNAT1(BP) gene regulated the flowering time and plant architecture of B. napus. However, the precise regulatory mechanism remains unclear. We cloned two homologous BP genes, BnaBPA03 and BnaBPC03, from B. napus Xiaoyun. The protein sequence analysis showed two proteins containing conserved domains KNOX I, KNOX II, ELK, and HOX of the KONX protein family. The CRISPR/Cas9 knockout lines exhibited early budding and flowering time, coupled with floral organ abscission earlier and a larger leaf angle. On the contrary, overexpression plants displayed a phenotype that was the inverse of these characteristics. Furthermore, we observed upregulation of gibberellin and ethylene biosynthesis genes, as well as floral integrator genes in knocked-out plants. The results revealed that BnaBPs play a role in flowering time, floral organ abscission, and leaf angle as well as germination processes mediated. Additionally, BnaBPs exerted an impact on the biosynthesis pathways of ethylene and GA.

Nutrition for commercial flower production of Longiflorum asiatica (LA) hybrid under northern plains

The study was conducted during the winter seasons, comprising the various combinations of inorganic fertilizers (N, P2O5, and K2O) and organic manure (FYM). Treatment T24 [(25 t FYM+160 kg N+120 kg P2O5+100 kg K2O)/ha] emerged as the statistically significant treatment, outperforming others in respect of tallest plant (122.43 cm), leaf count per plant (79.33), leaf length (7.90 cm), and early flower bud initiation (28.01 days), bud initiation to colour shown (47.11 days), and colour shown to flower opening (6.10 days). This treatment also tended to show the longest flower retention on plant (15.30 days), number of flower buds per plant (4.20), bud length (8.96 cm), stalk length (110.10 cm), stalk diameter (0.96 cm), flower diameter (22.98 cm), total chlorophyll content (6.50 mg/g), leaf nitrogen (4.70%), potassium (4.10%), and net assimilation ratio (NAR) (0.0065 mg/cm2/ day). Treatment T23 [(25 t FYM+120 kg N+90 kg P2O5+75 kg K2O)/ha] proved next best treatment. The lowest value was recorded in T25 (Control). Overall, treatment T24 proved to be the most effective treatment statistically for the commercial flower production of LA hybrid lilium for northern Indian plains.

Impact of planting density and shoot thinning on alstroemeria flowering, soil attributes and cost economics

A field experiment was conducted to assess the impact of various planting densities and levels of shoot thinning on cut flower yield, quality, soil chemical properties and cost economics in the commercially cultivated alstroemeria cultivar 'Capri'. The experiment involved three planting densities (4, 6 and 8 plants/m2) along with three shoot thinning levels (10 %, 20 % and 30 %). It was observed that higher planting density (4 plants/m2) promoted the length of cut stem, early bud formation, and flowering, optimal quality parameters, including maximum stem thickness, cut stem weight, floret quantity/stem, floret size, number of flowering stems/plant (yield) and vase life. Exclusive use of higher vegetative mass removal (30 % shoot thinning) was found to improve quality parameters such as the length of cut stem, number of flowering stems, stem thickness, weight of cut stem, floret quantity/stem, floret size, vase life, and early flowering. Soil samples were collected from each treatment and the consequent lab analysis revealed that the lower planting density and shoot thinning level resulted in maximum soil organic carbon, nitrogen, phosphorous and potassium content. In conclusion, planting density of 4 plants/m2 and 30 % shoot thinning emerged as the optimal combination for enhancing quantitative parameters in commercial alstroemeria cultivation. The study underscores the importance of strategic planting practices and vegetative mass management for maximizing yield and quality in alstroemeria production, along with ensuring higher economic returns.

Human iPSC-liver organoid transplantation reduces fibrosis through immunomodulation.

Donor organ shortages for transplantation remain a serious global concern, and alternative treatment is in high demand. Fetal cells and tissues have considerable therapeutic potential as, for example, organoid technology that uses human induced pluripotent stem cells (hiPSCs) to generate unlimited human fetal-like cells and tissues. We previously reported the in vivo vascularization of early fetal liver-like hiPSC-derived liver buds (LBs) and subsquent improved survival of recipient mice with subacute liver failure. Here, we show hiPSC-liver organoids (LOs) that recapitulate midgestational fetal liver promote de novo liver generation when grafted onto the surface of host livers in chemical fibrosis models, thereby recovering liver function. We found that fetal liver, a hematopoietic tissue, highly expressed macrophage-recruiting factors and antifibrotic M2 macrophage polarization factors compared with the adult liver, resulting in fibrosis reduction because of CD163+ M2-macrophage polarization. Next, we created midgestational fetal liver-like hiPSC-LOs by fusion of hiPSC-LBs to induce static cell-cell interactions and found that these contained complex structures such as hepatocytes, vasculature, and bile ducts after transplantation. This fusion allowed the generation of a large human tissue suitable for transplantation into immunodeficient rodent models of liver fibrosis. hiPSC-LOs showed superior liver function compared with hiPSC-LBs and improved survival and liver function upon transplantation. In addition, hiPSC-LO transplantation ameliorated chemically induced liver fibrosis, a symptom of liver cirrhosis that leads to organ dysfunction, through immunomodulatory effects, particularly on CD163+ phagocytic M2-macrophage polarization. Together, our results suggest hiPSC-LO transplantation as a promising therapeutic option for liver fibrosis.

Effect of photoperiod and chemical retardants on growth and flowering of pot mums (<i>Dendranthema grandiflora</i> Ramat)

Potted plants of chrysanthemum varieties namely Arun Kumar and Sweta Singar, were exposed to 8 h light and 16 h darkness and natural photoperiod, and spayed with maleic hydrazide (1000 and 2000 ppm), cycocel (1500 and 3000 ppm) and control (distilled water) in factorial complete block design with three replications. The results revealed that controlled photoperiod of 8 h light and 16 h darkness was found significantly superior over natural photoperiod with regard to minimum internodal length (2.85 cm), days to floral bud initiation (98.88), maximum flowering duration (23.14 days), flowers plant-1 (189.50), flower diameter (2.72 cm), fresh weight of flower (1.16 g) and vase life (14.86 days). Among growth retardants, maleic hydrazide (2000 ppm) significantly recorded minimum plant height, maximum branches plant-1 (18.85), flowering duration (25.25 days), flower diameter (2.95 cm), flowers per plant-1 (194.90) and weight of flower (1.20 g), while, maleic hydrazide (1000 ppm) recorded minimum internodal length (2.65 cm) and maximum vase life (18.00 days). The cv. Arun Kumar under controlled photoperiod recorded minimum internodal length (2.38 cm) and early floral bud initiation (91.04 days), however, maleic hydrazide (2000 ppm) recorded maximum flower diameter (4.05 cm), fresh weight of flower (1.53 g) and vase life (18.50 days). cv. Arun Kumar under controlled photoperiod treated with CCC-3000 ppm showed the best performance in recording the maximum number of flowers plants-1 (138.00) during first fortnight of flowering.

Influence of planting dates and fertilizer modules on yield of chrysanthemum and soil health

BackgroundOptimum planting date and appropriate fertilizer module are essential facets of chrysanthemum cultivation, to enhance quality yield, and improve soil health. A field-based study was undertaken over multiple growing seasons in 2022 and 2023, where six different planting dates, viz., P1:June 15, P2:June 30, P3:July 15, P4:July 30, P5:August 15 and P6:August 30 and two fertilizer modules, FM1:Jeevamrit @ 30 ml plant−1 and FM2:NPK @ 30 g m−2 were systematically examined using a Randomized Block Design (factorial), replicated thrice.ResultsP6 planting resulted in early bud formation (44.03 days) and harvesting stage (90.78 days). Maximum plant height (79.44 cm), plant spread (34.04 cm), cut stem length (68.40 cm), flower diameter (7.83 cm), stem strength (19.38˚), vase life (14.90 days), flowering duration (24.08 days), available soil N (314 kg ha−1), available P (37 kg ha−1), available K (347 kg ha−1), bacterial count (124.87 × 107 cfu g−1 soil), actinomycetes count (60.72 × 102 cfu g−1 soil), fungal count (30.95 × 102 cfu g−1 soil), microbial biomass (48.79 µg g−1 soil), dehydrogenase enzyme (3.64 mg TPF h−1 g−1 soil) and phosphatase enzyme (23.79 mol PNP h−1 g−1 soil) was recorded in P1 planting. Among the fertilization module, minimum days to bud formation (74.94 days) and days to reach the harvesting stage (120.95 days) were recorded with the application of NPK @30 g m−2. However, maximum plant height (60.62 cm), plant spread (23.10 cm), number of cut stems m−2 (43.88), cut stem length (51.34 cm), flower diameter (6.92 cm), stem strength (21.24˚), flowering duration (21.75 days), available soil N (317 kg ha−1), available P (37 kg ha−1) and available K (349 kg ha−1) were also recorded with the application of NPK @300 kg ha−1. Maximum vase life (13.87 days), OC (1.13%), bacterial count (131.65 × 107 cfu g−1 soil), actinomycetes count (60.89 × 102 cfu g−1 soil), fungal count (31.11 × 102 cfu g−1 soil), microbial biomass (51.27 µg g−1 soil), dehydrogenase enzyme (3.77 mg TPF h−1 g−1 soil) and phosphatase enzyme (21.72 mol PNP h−1 g−1 soil) were observed with the application of Jeevamrit @ 30 ml plant−1.ConclusionEarly planting (P1) and inorganic fertilization (NPK @ 30 g m−2) resulted in improved yield and soil macronutrient content. The soil microbial population and enzymatic activity were improved with the jeevamrit application. This approach highlights the potential for improved yield and soil health in chrysanthemum cultivation, promoting a more eco-friendly and economically viable agricultural model.

TBX3 is essential for establishment of the posterior boundary of anterior genes and upregulation of posterior genes together with HAND2 during the onset of limb bud development.

During limb bud formation, axis polarities are established as evidenced by the spatially restricted expression of key regulator genes. In particular, the mutually antagonistic interaction between the GLI3 repressor and HAND2 results in distinct and non-overlapping anterior-distal Gli3 and posterior Hand2 expression domains. This is a hallmark of the establishment of antero-posterior limb axis polarity, together with spatially restricted expression of homeodomain and other transcriptional regulators. Here, we show that TBX3 is required for establishment of the posterior expression boundary of anterior genes in mouse limb buds. ChIP-seq and differential gene expression analysis of wild-type and mutant limb buds identifies TBX3-specific and shared TBX3-HAND2 target genes. High sensitivity fluorescent whole-mount in situ hybridisation shows that the posterior expression boundaries of anterior genes are positioned by TBX3-mediated repression, which excludes anterior genes such as Gli3, Alx4, Hand1 and Irx3/5 from the posterior limb bud mesenchyme. This exclusion delineates the posterior mesenchymal territory competent to establish the Shh-expressing limb bud organiser. In turn, HAND2 is required for Shh activation and cooperates with TBX3 to upregulate shared posterior identity target genes in early limb buds.

An Efficient Propagation Approach to Forcing Softwood Shoots from Epicormic Buds and Subsequent Rooting of Paulownia elongata S. Y. Hu.

The current research describes the multiplication of Paulownia elongata S. Y. Hu, a timber plant, through the forcing of softwood shoots from epicormic buds under glasshouse conditions in spring and fall seasons. Different growth media were used to compare their effect on the forcing potential of epicormic buds. For this, 25-30-cm-long and 1.2-2-cm-diameter stem segments taken from the lower juvenile portion of a mother plant were placed horizontally in flat trays containing media, i.e., sterilized well-moistened sand, peat moss, perlite, and vermiculite individually. Furthermore, 4-6-cm-long forced softwood shoots were detached and treated with various concentrations of IBA (indole-3-butyric acid) and NAA (α-naphthyl acetic acid) either individually or in combinations for subsequent rooting. The response of shoot forcing was better in spring as compared to fall in terms of shoot length (cm), and number of shoots or leaves; however, an earlier bud break was observed during fall after 30 days of the initial experiment. The use of peat moss and vermiculite proved to be equally suitable for early bud break in both seasons, whereas in terms of shoot and leaf number as well as the shoot length (cm), the best outcome was observed in sand. Best rooting was observed at 3 gL-1 IBA + 3 gL-1 NAA in terms of root number per shoot, root length (cm), and days to root initiation while using sand as the growth medium after 50 days of the rooting experiment. The successfully established plantlets were further shifted to soil at Botanical Garden, University of the Punjab, Lahore, Pakistan, exhibiting an 87.5% survival rate. On the basis of the results obtained, it may be concluded that reasonable softwood shoot forcing in P. elongata may further be exploited for its mass scale nursery propagation as well as use in future in vitro studies.

Rice-Based Cropping System Effect on Yield and Yield Attributes on Promising Released Sesame Varieties

A field experiment was carried out at Agricultural College farm, Bapatla district, Andhra Pradesh, during the period from March 2023 to June 2023 to assess the performance of Sesame (Sesamum indicum L.) varieties under different fertilizer management practices in rice-based cropping system in Krishna Zone of AP. The experiment was laid out in a randomized block design with the factorial concept and three replications. The results showed that YLM 146 recorded significantly the highest seed yield (748 kg ha-1) at harvest. The yield attributing characters like the number of capsules plant-1 (67) and the number of seeds capsules-1 (45) were found significantly to be the highest in the variety YLM 146. Concerning different fertilizer management practices, 125% RDF + 1% foliar spray of 19:19:19 at the early budding and capsule formation stages recorded significantly higher seed yields (827 kg ha-1). Yield attributes like number of capsules plant-1 (96) and number of seeds capsules-1 (51) were significantly highest in 125% RDF + 1% foliar spray of 19:19:19 at the early budding and capsule formation stages. Overall, the results showed that sowing of YLM 146 variety with 125% RDF + 1% foliar spray of 19:19:19 at the early budding stage and early capsule formation stage seems to be appropriate for getting higher seed yield in Krishna Zone of Andhra Pradesh.

Optimization of Flowering and Fruit Development in Ber (Ziziphus mauritiana L.) cv. Umran using Fertilizers, Micronutrients, and Bioregulators

In order to investigate the effect of fertilizers, micronutrients and bioregulators on flowering and fruit growth and developments on ber (Ziziphus mauritiana L.) cv. Umran, an experiment was carried out during 2019/2020 cropping season at the Fruit Research Station, Imalia, Department of Horticulture JNKVV Jabalpur. The study used a factorial design using randomized complete block design (RCBD), with three replications and a total of twelve treatment combinations that included bioregulators, micronutrients, and fertilizers either separately or in various combinations. The application of treatment T4 [RDF (N:P:K)+Foliar spray of GA3 + NAA (50 ppm+50 ppm)] was found to be significantly superior as compared to other treatments for flowering parameter in respect to induce early flower bud formation, based on the results obtained in this study. It takes 22 days to induce the earliest flowering after spray, and it takes 8.67 days to achieve 50% flowering. The earliest date of flower bud initiation was found with the T11 [RDF (N:P:K)+ NPK -18:18:18 (1.0%) + ZnSO4 (0.5%) + Borax (0.3%)] which showed to be the best for fruit growth and development characteristics, including specific gravity (1.047), fruit weight (36.52 g), fruit volume (36.56 ml), fruit length (4.53 cm), and fruit diameter (3.50 cm).With regard to practically every fruit growth and development attribute, the treatment T10 was found to be the second best.

Knockdown of vitamin D receptor affects early stages of pectoral fin development in zebrafish.

The vitamin D receptor (VDR) signalling has been implicated in vertebrate limb or fin formation. However, the involvement of VDR signalling in the early stages of limb/fin development remains to be elucidated. In this study, the role of VDR signalling in pectoral fin development was investigated in zebrafish embryos. Knockdown of vdr induced the severe impairment of pectoral fin development. The zebrafish larvae lacking vdr exhibited reduced pectoral fins with no skeletal elements. Insitu hybridization revealed depletion of vdr downregulated fibroblast growth factor 24 (fgf24), a marker of early pectoral fin bud mesenchyme, in the presumptive fin field even before fin buds were visible. Moreover, a perturbed expression pattern of bone morphogenetic protein 4 (bmp4), a marker of the pectoral fin fold, was observed in the developing fin buds of zebrafish embryos that lost the vdr function. These findings suggest that VDR signalling is crucial in the early stages of fin development, potentially influencing the process by regulating other signalling molecules such as Fgf24 and Bmp4.

The Stimulation of Chemical Male Sterility for F1 Hybrid Lettuce (Lactuca Sativa Var. Longifolia) Production

Male sterility is a unique application in the production of F1 hybrid seeds of some important species. Today, while F1 hybrid lettuce varieties of abroad origin take their place in the Türkiye seed market, unfortunately, we do not have a native F1 hybrid variety. Besides, F1 hybrid seed production has become a prestige for multi-national companies regardless of the type of vegetable. Within this perspective, the effects of some chemical hybridizing agents (CHAs) such as E4FO, Ethrel and GA3 on male sterilizing activity in lettuce cultivars (Maylight352 F1, Presidential and Yedikule) were evaluated. Therefore, pollen presence in the early bud stage, seed formation and seed viability (germination) were examined. The applications had different effects on pollen presence, seed formation and seed germination, and thus male sterility. Ethrel was not effective at low doses, but at high doses, it caused flower bud deformation and growth retardation. E4FO is partially effective, but the application doses are low. Therefore, E4FO should be used at higher than 1500 ppm and 2000 ppm. GA3 applications produced the best results in stimulating male sterility, and full sterility (% 100) was achieved from 200, 250 and 300 ppm in all cultivars. As a result, 200 ppm GA3 was determined as the recommendable dose in the production of Fı hybrid lettuce.

Spectral-temporal traits in Sentinel-1 C-band SAR and Sentinel-2 multispectral remote sensing time series for 61 tree species in Central Europe

Tree species maps derived from satellite imagery increasingly support forest administrations and nature conservation authorities with large-scale and up-to-date information. However, many species are often excluded or aggregated in classification tasks due to a limited knowledge of the most suitable predictors. Our study aims to gain a better understanding of optical and polarimetric traits for tree species mapping by examining Sentinel-1 and Sentinel-2 time series from 61 tree species in temperate Europe. For a selection of 32 optical, polarimetric and structural variables, the principal component analysis revealed that Sentinel-2 variables mainly explain the variance in the data by contributing to the “seasonality” and “foliage color” components. Sentinel-1 contribute most to the “texture” component. The Normalized Difference Vegetation Index (NDVI), Tasseled Cap Greenness (TCG) and Radar Vegetation Index (RVI) were chosen as key variables for further analysis. Seasonality was found to be the most dominant aspect in all vegetation indices. Furthermore, the TCG was found to be useful to distinguish between early and late budding species. The RVI showed a large potential to discriminate conifers, which is attributed to the crown volume effect of C-band SAR. Using exploratory data analysis, we further examined the influence of management, biogeographical and meteorological factors on the time series from Fagus sylvatica, Pinus sylvestris, and Picea abies. The NDVI and TCG are relatively robust to different conditions. For the two conifer species however, we found strong spatial variations of the RVI which are presumably caused by different crown conditions across the study area. Using Sentinel-1 data could therefore lead to uncertainties in tree species mapping across large biogeographical gradients. This study contributes to the improvement of tree species mapping based on optical and dual-polarimetric data and thus benefits forest authorities and other stakeholders in their monitoring tasks and decision-making.

Transcriptional Regulation and Gene Mapping of Internode Elongation and Late Budding in the Chinese Cabbage Mutant lcc.

Two important traits of Chinese cabbage, internode length and budding time, destroy the maintenance of rosette leaves in the vegetative growth stage and affect flowering in the reproductive growth stage. Internodes have received much attention and research in rice due to their effect on lodging resistance, but they are rarely studied in Chinese cabbage. In Chinese cabbage, internode elongation affects not only the maintenance of rosette leaves but also bolting and yield. Budding is also an important characteristic of Chinese cabbage entering reproductive growth. Although many studies have reported on flowering and bolting, studies on bud emergence and the timing of budding are scarce. In this study, the mutant lcc induced by EMS (Ethyl Methane Sulfonate) was used to study internode elongation in the seedling stage and late budding in the budding stage. By comparing the gene expression patterns of mutant lcc and wild-type A03, 2280 differentially expressed genes were identified in the seedling stage, 714 differentially expressed genes were identified in the early budding stage, and 1052 differentially expressed genes were identified in the budding stage. Here, the transcript expression patterns of genes in the plant hormone signaling and clock rhythm pathways were investigated in relation to the regulation of internode elongation and budding in Chinese cabbage. In addition, an F2 population was constructed with the mutants lcc and R500. A high-density genetic map with 1602 marker loci was created, and QTLs for internode length and budding time were identified. Specifically, five QTLs for internode length and five QTLs for budding time were obtained. According to transcriptome data analysis, the internode length candidate gene BraA02g005840.3C (PIN8) and budding time candidate genes BraA02g003870.3C (HY5-1) and BraA02g005190.3C (CHS-1) were identified. These findings provide insight into the regulation of internode length and budding time in Chinese cabbage.

Integrative Metabolomic and Transcriptomic Analysis Elucidates That the Mechanism of Phytohormones Regulates Floral Bud Development in Alfalfa.

Floral bud growth influences seed yield and quality; however, the molecular mechanism underlying the development of floral buds in alfalfa (Medicago sativa) is still unclear. Here, we comprehensively analyzed the transcriptome and targeted metabolome across the early, mid, and late bud developmental stages (D1, D2, and D3) in alfalfa. The metabolomic results revealed that gibberellin (GA), auxin (IAA), cytokinin (CK), and jasmonic acid (JA) might play an essential role in the developmental stages of floral bud in alfalfa. Moreover, we identified some key genes associated with GA, IAA, CK, and JA biosynthesis, including CPS, KS, GA20ox, GA3ox, GA2ox, YUCCA6, amid, ALDH, IPT, CYP735A, LOX, AOC, OPR, MFP2, and JMT. Additionally, many candidate genes were detected in the GA, IAA, CK, and JA signaling pathways, including GID1, DELLA, TF, AUX1, AUX/IAA, ARF, GH3, SAUR, AHP, B-ARR, A-ARR, JAR1, JAZ, and MYC2. Furthermore, some TFs related to flower growth were screened in three groups, such as AP2/ERF-ERF, MYB, MADS-M-type, bHLH, NAC, WRKY, HSF, and LFY. The findings of this study revealed the potential mechanism of floral bud differentiation and development in alfalfa and established a theoretical foundation for improving the seed yield of alfalfa.

Transcriptome Analysis of Intermittent Light Induced Early Bolting in Flowering Chinese Cabbage.

In flowering Chinese cabbage, early booting is one of the most important characteristics that is linked with quality and production. Through fixed light intensity (280 μmol·m-2·s-1) and fixed intermittent lighting in flowering Chinese cabbage, there was early bolting, bud emergence, and flowering. Moreover, the aboveground fresh weight, blade area, dry weight of blade, and quantification of the leaves in flowering Chinese cabbage were significantly reduced, while the thickness of tillers, tillers height, dry weight of tillers, and tillers weight were significantly increased. The chlorophyll contents and soil-plant analysis and development (SPAD) value decreased in the early stage and increased in the later stage. The nitrate content decreased, while the photosynthetic rate, vitamin C content, soluble sugar content, soluble protein content, phenolic content, and flavonoid content increased, and mineral elements also accumulated. In order to explore the mechanism of intermittent light promoting the early bolting and flowering of '49d' flowering Chinese cabbage, this study analyzed the transcriptional regulation from a global perspective using RNA sequencing. A total of 17,086 differentially expressed genes (DEGs) were obtained and 396 DEGs were selected that were closely related to early bolting. These DEGs were mainly involved in pollen wall assembly and plant circadian rhythm pathways, light action (34 DEGs), hormone biosynthesis and regulation (26 DEGs), development (21 DEGs), and carbohydrate synthesis and transport (6 DEGs). Three hub genes with the highest connectivity were identified through weighted gene co-expression network analysis (WGCNA): BrRVE, BrLHY, and BrRVE1. It is speculated that they may be involved in the intermittent light regulation of early bolting in flowering Chinese cabbage. In conclusion, intermittent light can be used as a useful tool to regulate plant growth structure, increase planting density, enhance photosynthesis, increase mineral accumulation, accelerate growth, and shorten the breeding cycle.
