Here, we examined the role of two single nucleotide polymorphisms regulating prefrontal cortex (PFC) and striatal DA activity (COMTMet108/158Val, and ANKK1/DRD2TaqIA) on the stimulus-driven control of attention. Participants performed a distraction protocol and a set-switching task. Behavioral analysis was complemented with phase-coherence of gamma-band oscillations and the analysis of novelty-P3 component. COMTMet individuals, related to higher PFC DA concentrations, combined with a high DRD2 density (without ANKK1/DRD2TaqIA1 allele) and COMTVal participants (low PFC DA availability) combined with low DRD2 density (with ANKK1/DRD2TaqIA1 allele) displayed a delay in response time to the task following novel events relative to standard ones. They also displayed a task switch cost paralleled by an increase in P3 amplitude in task-switch trials, as updating the context when required. Differently, individuals with other combination of polymorphisms shifted the attention toward novel events without a time cost to the task through a mechanism of phase-resetting of gamma oscillations to novel events. The reported electrophysiological measures might constitute an endophenotypic marker of cognitive deficits associated to DA-related disorders.
