Solubility Enhancement of Poorly Water-Soluble Aceclofenac by Amalgamation Micronization and Solid Dispersion Techniques

Aceclofenac is a poorly aqueous soluble Biopharmaceutical Classification System (BCS) class II drug that has bioavailability problems after oral administration due to low aqueous solubility. There are numerous solubility modification approaches and they also have process limitations. Hence, the present research work aims to use two different solubility enhancement techniques (Micronization with Solid dispersion) simultaneously to counter individual limitations of techniques. The micronization technique increases the solubility of pure aceclofenac through increasing particle surface area, but it produces charged micronized material, leading to segregation and clumping of micronized material. Hence, micronized material handling during dosage form manufacturing is quite difficult. In the present research work, aceclofenac material is micronized by air jet mill to produce materials of below 25 μm size ranges. This micronized aceclofenac material is further encapsulated in a solid dispersion technique to produce amorphous aceclofenac materials. Solid dispersion of micronized aceclofenac was prepared by various techniques like physical kneading, solvent evaporation, melting and a combination thereof using different polymers (Polyox 301, HPMC, cetostearyl alcohol and glyceryl behenate). The prepared solid dispersion formulation (SD-31, SD-22, SD-23 and SD-30) exhibited an increase in the dissolution of aceclofenac as compared to pure aceclofenac. Various characterization techniques (Differential scanning calorimetry (DSC), X-ray Diffraction (XRD) and scanning electron microscope (SEM)) also reveal that aceclofenac crystallinity is significantly minimized in Solid dispersion formulation of SD-31, SD-22, SD-23 and SD-30. This selected solid dispersion (SD-31 and SD-22) also exhibited good stability (Photostability and stability at 40°C/75% RH after 3 months).

Phototoxicity in vitro and safety in vivo of the emulsion photosensitizer based on furanocoumarins of Heracleum sosnowskyi

The use of plants such as giant hogweed as raw materials for the manufacture of dosage forms has been little explored. In this study, we utilized furanocoumarins from the Heracleum sosnowskyi plant to create an experimental emulsion dosage form (EmFHS). The EmFHS was finely dispersed (481.8 nm ± 71.1 nm), shelf-stable, and contained predominantly 8-methoxypsoralen at a concentration of 1 mg/ml.Phototoxicity analysis of EmFHS for THP-1 cells under UV (365 nm) irradiation showed an IC50 of 19.1 µg/ml (24 h) and 6.3 µg/ml (48 h). In relation to spheroids (L929), EmFHS exhibited a phototoxic effect in the concentration range of 31.25–125 µg/ml8-MOP. A full phototoxic effect was observed 48 h after UV irradiation. The phototoxic effect of EmFHS in vitro was dose-dependent and comparable to the effect of emulsion synthetic 8-methoxypsoralen and chlorin e6 solution. EmFHS cytotoxicity was caused solely by UV radiation, and toxicity in the dark was minimal.EmFHS, administered at a dose of 3 mg/kg8-MOP, was found to be safe after a single intravenous administration to rats. It had a photosensitizing effect in the form of local photodermatitis when exposed to UV irradiation at a dose of 44 J/cm2.The biokinetics of emulsion furanocoumarins showed that the phototoxic effect of EmFHS is due to the high penetration ability of the emulsion into cells of spheroids. At the same time, it has a low degree of cumulation when administered intravenously.The obtained data suggest that EmFHS may be a promising treatment for PUVA therapy of various dermatological diseases. Additionally, the plant Heracleum sosnowskyi shows potential as a basis for creating new dosage forms with phototherapeutic effects.

Drug Polymorphism: An Important Pre-formulation Tool in the Formulation Development of a Dosage Form

Abstract: Polymorphism permits solids to have two or multiple crystal structures with varying orientations or conformations, and polymorph drug dissolution and solubility vary. Polymorphism influences drug efficacy, bioavailability and toxicity. Various parameters can regulate polymorph crystallization, including supersaturation phase, temperature, stirring rate, solvent addition rate, seed crystal, additives and pH. To characterize and monitor polymorphs, various analytical approaches are available, including powder X-Ray diffraction / X-Ray diffraction (PXRD/XRD), also called as Gold Standard method, differential scanning calorimetry (DSC), Infrared spectroscopy (IR), and microscopical studies. To prevent polymorphic change during production, distribution, and storage, formulation with the most stable form in the marketed product is considered. In addition, by monitoring each phase of raw material processing, polymorphisms can be controlled during dosage form manufacturing. The objective of this review is to provide concise information on drug polymorphism, their characterization process, their effect on the stability of dosage forms, factors controlling polymorphism in the crystallization process, some case studies on polymorphs, control of polymorphism during the formulation of successful dosage forms and some of the regulatory considerations regarding polymorphs. Various databases like Pubmed/Medline, Google Scholar and Web of Science of all English language articles were searched, and relevant information was collected regarding the importance of polymorphism in formulation development. From a vast literature survey, it was found that polymorphism is a very important tool in a pre-formulation study that provides information about the fate of a drug molecule in its journey. From the present study, it was concluded that the polymorphism property of a drug should be taken into consideration at the pre-formulation stage since it is a very important tool for the formulation of a successful dosage form with stability and efficacy.

Polyvinyl Alcohol, a Versatile Excipient for Pharmaceutical 3D Printing.

Three-dimensional (3D) printing in the pharmaceutical field allows rapid manufacturing of a diverse range of pharmaceutical dosage forms, including personalized items. The application of this technology in dosage form manufacturing requires the judicious selection of excipients because the selected materials must be appropriate to the working principle of each technique. Most techniques rely on the use of polymers as the main material. Among the pharmaceutically approved polymers, polyvinyl alcohol (PVA) is one of the most used, especially for fused deposition modeling (FDM) technology. This review summarizes the physical and chemical properties of pharmaceutical-grade PVA and its applications in the manufacturing of dosage forms, with a particular focus on those fabricated through FDM. The work provides evidence on the diversity of dosage forms created using this polymer, highlighting how formulation and processing difficulties may be overcome to get the dosage forms with a suitable design and release profile.

A case study to investigate the influence of extrusion temperature, 3D printing parameters and the use of antioxidants on the degradation of dexamethasone

The use of 3D printing techniques such as fused deposition modelling (FDM) for the manufacturing of dosage forms has become an interesting research field in pharmaceutics. When using FDM 3D printing, there are usually two temperature events that can have influence on temperature-sensitive drugs and excipients. The aim of this work was to investigate the influence of various parameters on the stability of the active ingredient dexamethasone. For this purpose, active ingredient-containing filaments were produced by hot melt extrusion and the influence of temperature and the presence of antioxidants on drug stability were investigated. Vitamin C, vitamin E and butylated hydroxytoluene were used as antioxidants. If suitable filaments were obtained, these filaments were used to 3D print test objects. In addition to the presence of antioxidants, the influence of printing parameters such as printing temperature, printing speed and print bed temperature on the active ingredient content was investigated. The variation of the temperature during the extrusion process showed an influence on the degradation of dexamethasone. Furthermore, during the investigation of the 3D printed objects, the presence of the antioxidants did not show any positive influence on the drug stability. The printing temperature was identified as a critical process parameter during 3D printing of dexamethasone, while other printing settings like the temperature of the print bed and the printing speed had no detectable influence.

Precise Fabrication of Ocular Inserts Using an Innovative Laser-Driven CaliCut Technology: In Vitro and in Vivo Evaluation

Ocular inserts offer distinct advantages, including a preservative-free drug delivery system, the ability to provide tailored drug release, and ease of administration. The present research paper delves into the development of an innovative ocular insert using CaliCut technology. Complementing the hot melt extrusion (HME) process, CaliCut, an advanced technology in ocular insert development, employs precision laser gauging to achieve impeccable cutting of inserts with desired dimensions. Its intelligent control over the stretching process through auto feedback-based belt speed adjustment ensures unparalleled accuracy and consistency in dosage form manufacturing. Dry eye disease (DED) poses a significant challenge to ocular health, necessitating innovative approaches to alleviate its symptoms. In this pursuit, castor oil has emerged as a promising therapeutic agent, offering beneficial effects by increasing the thickness of the lipid layer in the tear film, thus improving tear film stability, and reducing tear evaporation. To harness these advantages, this study focuses on the development and comprehensive characterization of castor oil-based ocular inserts. Additionally, in-vivo irritancy evaluation in rabbits has been undertaken to assess the inserts' safety and biocompatibility. By harnessing the HME and CaliCut techniques in the formulation process, the study demonstrates their instrumental role in facilitating the successful development of ocular inserts.

Analysis of time costs in the manufacture of non-sterile drugs in the form of liquid dosage forms in a production pharmacy

AIM: to determine the time spent on pharmaceutical manufacturing of non-sterile medicines in the form of liquid dosage forms and assess their compliance with current standards. MATERIALS AND METHODS: the objects of the study were the technological pro- cesses of manufacturing non-sterile liquid dosage forms in pharmacies of the State Unitary Enterprise “Tattechmedpharm” No. 57 and No. 361 (Kazan), No. 277 (central district pharmacy of Almetyevsk), performing a manufacturing function (license L042-01181-16/00269222 dated 16/09/2020). During the study, methods of grouping, systematization, structural and logical analysis, comparative analysis, direct observation method, measurement method, time-study method, methods of statistical data processing using the SPSS Statis- tics program were used. RESULTS AND DISCUSSION: during the study, the time-study of technological operations in the manufacturing of non-sterile liquid dosage forms was used by direct measurement using a stopwatch. Time measurements were carried out for dosage forms, which were prepared daily, with different numbers of components (from 1 to 6). The most labor-intensive operations were identified, namely: weigh- ing of pharmaceutical substances, filtration of a water solution, and performing calculations in a written control passport. The obtained results were also compared with current standards. During the pro- cess, it was revealed that the time spent was compliant with the use of concen- trated solutions, as well as when preparing glycerin solutions that don’t require filtration. However, when performing such technological operations as weighing of ingredients, dissolving and filtering the dosage form, the results exceeded the recommended standards. CONCLUSION: in modern conditions, the time spent on manufacturing of liquid dosage forms according to individual prescriptions, exceeds established stan- dards, due to the lack of burette systems in pharmacies and the reduction of the practice of using concentrated solutions. Based on the results of the study, re- serves were identified for increasing the productivity of pharmaceutical employees in the manufacture of liquid dosage forms: the use of electronic scales, concen- trated solutions and the development of standard operating procedures for the pharmaceutical manufacturing of liquid dosage forms.

Selective laser sintering additive manufacturing of solid oral dosage form: Effect of laser power and hatch spacing on the physico-technical behaviour of sintered printlets

Selective laser sintering is a laser-based powder bed fusion 3D printing technique in which, the laser parameters plays a vital role in the fusion of polymeric powder particles and the behavior of the final printed object. Hence, the objective of the current study was to investigate the impact of laser power and hatch spacing on the fusion of particles, followed by their physicochemical and mechanical behavior investigations of the final sintered object made with dapsone and poly (1-vinylpyrrolidone-co-vinyl acetate) as a model drug and polymer, respectively. An increase in laser power and a decrease in hatch spacing improved particle fusion, resulting in smoother surface, increased density, and better mechanical properties. Interestingly, there was a loss of drug crystallinity was experienced in respect to varied laser parameters, highlighting the potential of laser-based powder bed fusion technique in developing amorphous solid dispersions. Despite extensive laser parameters, drug entrapment efficiency was found to be greater than 90 % for all sintered batches. No notable differences were observed in the dissolution behavior of all performed samples although it was anticipated to achieve as there was differences witnessed in porosity and disintegration time. Finally, the oral pharmacokinetics study revealed the comparable performance of the sintered dosage form with the tablet prepared with conventional direct compression techniques. Overall, these findings demonstrate that with varying laser parameters, various properties of the final sintered object can be manipulated which further showed the flexibility of this 3D printing technique to fabricate dosage form with a more personalized and customized needs.

Computer numerical control (CNC) carving as an on-demand point-of-care manufacturing of solid dosage form: A digital alternative method for 3D printing

Computer numerical control (CNC) carving is a widely used method of industrial subtractive manufacturing of wood, plastics, and metal products. However, there have been no previous reports of applying this approach to manufacture medicines. In this work, the novel method of tablet production using CNC carving is introduced for the first time. This report provides a proof-of-concept for applying subtractive manufacturing as an alternative to formative (powder compression) and additive (3D printing) manufacturing for the on-demand production of solid dosage forms. This exemplar manufacturing approach was employed to produce patient-specific hydrocortisone (HC) tablets for the treatment of children with congenital adrenal hyperplasia. A specially made drug-polymer cast based on polyethene glycol (PEG 6,000) and hydroxypropyl cellulose was produced using thermal casting. The cast was used as a workpiece and digitally carved using a small-scale 3-dimensional (3D) CNC carving. To establish the ability of this new approach to provide an accurate dose of HC, four different sizes of CNC carved tablet were manufactured to achieve HC doses of 2.5, 5, 7.5 and 10 mg with a relative standard deviation of the tablet weight in the range of 3.69–4.79%. In addition, batches of 2.5 and 5 mg HC tablets met the British Pharmacopeia standards for weight uniformity. Thermal analysis and X-ray powder diffraction indicated that the model drug was in amorphous form. In addition, HPLC analysis indicated a level of purity of 96.5 ± 1.1% of HC. In addition, the process yielded mechanically strong cylindrical tablets with tensile strength ranging from 0.49 to 1.6 MPa and friability values of <1%, whilst maintaining an aesthetic look. In vitro, HC release from the CNC-carved tablets was slower with larger tablet sizes and higher binder contents. This is the first report on applying CNC carving in the pharmaceutical context of producing solid dosage forms. The work showed the potential of this technology as an alternative method for the on-demand manufacturing of patient-specific dosage forms.

Risks of Powder Flow Obstruction in Hopper and Bin Discharge in Solid Dosage Form Manufacture should be Predicted Under The Active Stress State

Discharge of powder from a hopper or bin is a common operation in solid dosage form manufacture. Powder flow obstruction during hopper/bin discharge, such as arching or ratholing, remains an outstanding risk and cannot be reliably diagnosed using the existing flow function coefficient-based method. In this study, we showed that the major principal stress (σ1) at the bin outlet is required for an accurate prediction of powder flow obstruction risks. We noted that powder is susceptible to flow obstruction when the unconfined yield strength exceeds the stress facilitating powder failure. We presented a complete model to calculate the stress conditions and subsequently predict flow obstruction risks in hopper/bin discharge based on this criterion. The method was experimentally verified by hopper/bin discharge experiments encompassing 10 powder blends and 2 equipment systems. Importantly, we showed that the active stress state assumption should be employed for the powder flow obstruction prediction because σ1 is high and powder is more susceptible to flow obstruction. Prediction under the passive stress state can lead to significant under-estimation of flow obstruction risks. Therefore, the hopper design protocol, which assumes the passive stress state in arching prediction, should not be indiscriminately used toward pharmaceutical powder flow applications.

Could ginger extract be a therapeutic drug for migraine?

Migraine is a common neurological disorder that may be accompanied by vascular disturbances, Migraine is one of the most causes of disability worldwide. Zingiber officinale is a medicinal herb that has an analgesic effect on many disorders such as headaches, migraine, muscle tension, stomach spasm, and dysmenorrhea. Also, ginger has many pharmacological actions used to treat and prevent various common symptoms and diseases. This review aims to evaluate the potential of ginger to treat or prevent migraine episodes. Especially nowadays, Patients prefer herbal and complementary medicine to avoid the hard side effects of chemical drugs. The author searched several databases including PubMed, Science Direct, Wiley Online, and Scopus through February 2023 for recent articles with good quality evaluating the potential of ginger to treat migraine patients. The author made investigations and Interpretations depending on the results of the authors' experiments in previous articles included in my review. It is suggested that the bioactive compounds in ginger have the potential to treat and prevent acute migraine episodes effectively and safely. The author recommends encouraging the manufacturing of different pharmaceutical dosage forms of ginger extract to be used worldwide in a safe way and to render a higher absorption rate, and pharmacological response.

In situ thermal image analysis of selective laser sintering for oral dosage form manufacturing

Additive Manufacturing (AM) is a fast-growing approach to produce personalized oral dosage forms. Even though some AM technologies are promising as alternative to conventional compounding with resulting dosage manipulation, they still suffer from a lack of quality control. Due to the high regulatory demands and standards applied to dosage forms in the case of dose accuracy and tablet properties such as friability, effective quality control is a key feature in promoting AM as a valid technology for patient-tailored medications. One of the AM techniques used is selective laser sintering, which allows for capturing the surface state layer-by-layer during the printing process. It provides the opportunity to apply non-destructive quality control based on image analysis extracting essential data at each layer of the sintering process. This work is devoted to establishing the value of data gathered via thermal image analysis for the subsequent quality control.

Finite-Temperature Mechanical Properties of Organic Molecular Crystals from Classical Molecular Simulation

The mechanical properties of crystalline materials can have an impact on different aspects of solid dosage form design and manufacturing. Experimental determination of these quantities is known to pose difficulties and can be time-consuming. Instead, first-principles predictions of such materials have the potential for accelerating drug formulation development. We calculated elastic constants and several derived mechanical properties for the crystalline phase of a set of 18 small organic molecules with available experimental data, using molecular dynamics simulations based on the widely used GAFF2 force field. Attempts to further improve the accuracy of the predicted mechanical properties were made by fitting the force field parameters to the energies and forces determined by DFT calculations. We determine that DFT-based optimization of the force field parameters were unable to improve the accuracy of the mechanical property predictions over the currently established GAFF2 force field, which already provides reasonably accurate results with mean unsigned errors of individual elastic constants of around 2–3 GPa, or lower, in the majority of cases. Also, the elastic anisotropy is correctly predicted, at least qualitatively. The method used here can be a simple and fast means for the approximate determination of mechanical properties of a crystalline drug powder─material properties that can be used to guide decisions during formulation development.

An Overview of Recent Patents and Future Perspective Based on Cyclodextrin Complexation.

The majority of drugs taken orally have limited aqueous solubility and dissolution rate. Cyclodextrin (CD) and its derivatives are used as pharmaceutical adjuvants, contributing to the development of safe and high bioavailability formulations. CDs have a unique structure with a variety of physicochemical features that aid pharmaceutical scientists in solving drug delivery issues for poorly water-soluble drugs (PWS). This article covers information about cyclodextrin and its various derivatives, its different manufacturing process, physicochemical properties, advantages, and recent advancements. There are various advantages of CD-based inclusion complexes, such as enhancement of solubility, bioavailability, and stability and reduction of irritation caused by the drug. Moreover, they are used as odor and taste enhancers and also prevent incompatibility by physically isolating the incompatible drug components in drug formulation. CD and its derivatives are extensively employed as solubilizers in the manufacturing of parenteral and oral dosage forms. Inclusion complexes formed by CDs with appropriately sized guest molecules improve drug water solubility, physical-chemical stability, and bioavailability. Simultaneously CDs prevent the drugs from degradation like oxidation, hydrolysis, and photodegradation and extend the shelf life of the drug. The manuscript also highlights patents and exclusive branded formulations of modified CDs. It also discusses the different examples of chemically modified CDs, i.e., captisol, sulfobutyl ether-β-CD, hydroxy propyl betadex, randomly methylated β-CD, methyl β-CD, and hydoxy propyl γ-CD, all are used in the various dosage forms.

Selective laser sintering additive manufacturing of dosage forms: Effect of powder formulation and process parameters on the physical properties of printed tablets

Large batches of placebo and drug-loaded solid dosage forms were successfully fabricated using selective laser sintering (SLS) 3D printing in this study. The tablet batches were prepared using either copovidone (N-vinyl-2-pyrrolidone and vinyl acetate, PVP/VA) or polyvinyl alcohol (PVA) and activated carbon (AC) as radiation absorbent, which was added to improve the sintering of the polymer. The physical properties of the dosage forms were evaluated at different pigment concentrations (i.e., 0.5 and 1.0 wt%) and at different laser energy inputs. The mass, hardness, and friability of the tablets were found to be tunable and structures with greater mass and mechanical strength were obtained with increasing carbon concentration and energy input. Amorphization of the active pharmaceutical ingredient in the drug-loaded batches, containing 10 wt% naproxen and 1 wt% AC, was achieved in-situ during printing. Thus, amorphous solid dispersions were prepared in a single-step process and produced tablets with mass losses below 1 wt%. These findings show how the properties of dosage forms can be tuned by careful selection of the process parameters and the powder formulation. SLS 3D printing can therefore be considered to be an interesting and promising technique for the fabrication of personalized medicines.

Investigating the Use of Magnetic Nanoparticles As Alternative Sintering Agents in Selective Laser Sintering (SLS) 3D Printing of Oral Tablets.

Selective laser sintering (SLS) is a single-step, three-dimensional printing (3DP) process that is gaining momentum in the manufacturing of pharmaceutical dosage forms. It also offers opportunities for manufacturing various pharmaceutical dosage forms with a wide array of drug delivery systems. This research aimed to introduce carbonyl iron as a multifunctional magnetic and heat conductive ingredient for the fabrication of oral tablets containing isoniazid, a model antitubercular drug, via SLS 3DP process. Furthermore, the effects of magnetic iron particles on the drug release from the SLS printed tablets under a specially designed magnetic field was studied. Optimization of tablet quality was performed by adjusting SLS printing parameters. The independent factors studied were laser scanning speed, hatching space, and surface/chamber temperature. The responses measured were printed tablets' weight, hardness, disintegration time, and dissolution performance. It has been observed that, for the drug formulation with carbonyl iron, due to its inherent thermal conductivity, sintering tablets required relatively lower laser energy input to form the tablets of the same quality attributes as the other batches that contained no magnetic particles. Also, printed tablets with carbonyl iron released 25% more drugs under a magnetic field than those without it. It can be claimed that magnetic nanoparticles appear as an alternative conductive material to facilitate the sintering process during SLS 3DP of dosage forms.

Particle Property Characterization and Data Curation for Effective Powder Property Modeling in the Pharmaceutical Industry.

Computational modeling, machine learning, and statistical data analysis are increasingly utilized to mitigate chemistry, manufacturing, and control failures related to particle properties in solid dosage form manufacture. Advances in particle characterization techniques and computational approaches provide unprecedented opportunities to explore relationships between particle morphology and drug product manufacturability. Achieving this, however, has numerous challenges such as producing and appropriately curating robust particle size and shape data. Addressing these challenges requires a harmonized strategy from material sampling practices, characterization technique selection, and data curation to provide data sets which are informative on material properties. Herein, common sources of error in particle characterization and data compression are reviewed, and a proposal for providing robust particle morphology (size and shape) data to support modeling efforts, approaches for data curation, and the outlook for modeling particle properties are discussed.
