Direct Three-Dimensional Mass Spectrometry Imaging with Laser Ablation Remote Atmospheric Pressure Photoionization/Chemical Ionization.

The laser ablation remote atmospheric pressure photoionization/chemical ionization (LARAPPI/CI) platform coupled to an ultrahigh resolution quadrupole-time-of-flight (QToF) mass spectrometer was developed and employed for the first direct three-dimensional (3D) mass spectrometry imaging (MSI) of metabolites in human and plant tissues. Our solution for 3D MSI does not require sample modification or cutting into thin slices. Ablation characteristics of an optical system based on a diffraction optical element are studied and used for voxel stacking to directly remove layers of tissues. Agar gel, red radish, kiwi, human kidney cancer, and normal tissue samples were used for the tests of this new system. The 2D and 3D ion images vividly illustrate differences in the abundances of selected metabolites between cancerous and noncancerous regions of the kidney tissue and also between different parts of plant tissues. The LARAPPI/CI MSI setup is also the first example of the successful use of combined dopant-assisted atmospheric pressure photoionization (DA-APPI) and atmospheric pressure chemical ionization (APCI) ion source for mass spectrometry imaging.

Highly sensitive and selective sulfur hexafluoride detection based on DAPPI-FAIMS

This paper proposed a novel detection method of sulfur hexafluoride (SF6) by combining dopant-assisted atmospheric pressure photoionization (DAPPI) with high-field asymmetric waveform ion mobility spectroscopy (FAIMS). Acetone was employed as a dopant to achieve high sensitivity, and the negative ion mode of FAIMS was selected to achieve high selectivity. The impact of dopant concentrations on detection sensitivity and linearity was studied, and the influence of relative humidity on the detection of sulfur hexafluoride was studied. Ammonia, benzene, and hydrogen sulfide were selected as interferences to verify the selectivity. A linearity between the concentration and the signal was studied, and the detection limit for sulfur hexafluoride was determined. Experimental results demonstrated that the DAPPI source effectively enhanced the ionization efficiency of sulfur hexafluoride with a linear coefficient of 96.1 %. The sensitivity of sulfur hexafluoride is inversely proportional to relative humidity. Sulfur hexafluoride has a characteristic fingerprint spectrum different from air background and typical interfering, and can be distinguished from interfering substances in mixtures containing ammonia, benzene, and hydrogen sulfide. The concentration of sulfur hexafluoride has excellent linearity with signal intensity, the linear coefficient was 99.8 %, and the detection limit was 0.02 ppm.

Emissions of Formamide and Ammonia from Foam Mats: Online Measurement Based on Dopant-Assisted Photoionization TOFMS and Assessment of Their Exposure for Children

Formamide has been classified as a Class 1B reproductive toxicant to children by the European Union (EU) Chemicals Agency. Foam mats are a potential source of formamide and ammonia. Online dopant-assisted atmospheric pressure photoionization time-of-flight mass spectrometry (DA-APPI-TOFMS) coupled with a Teflon environmental chamber was developed to assess the exposure risk of formamide and ammonia from foam mats to children. High levels of formamide (average 3363.72 mg/m3) and ammonia (average 1586.78 mg/m3) emissions were measured from 21 foam mats with three different raw material types: ethylene-vinyl acetate (EVA: n = 7), polyethylene (PE: n = 7), and cross-linked polyethylene (XPE: n = 7). The 28 day emission testing for the selected PE mat showed that the emissions of formamide were 2 orders of magnitude higher than the EU emission limit of 20 μg/m3, and formamide may be a permanent indoor contaminant for foam mat products during their life cycle. The exposure assessment of children aged 0.5-6 years showed that the exposure dose was approximately hundreds of mg/kg-day, and the age group of 0.5-2 years was subject to much higher dermal exposures than others. Thus, this study provided key relevant information for further studies on assessing children's exposure to indoor air pollution from foam mats.

Analysis of Dechlorane Plus and related compounds in gull eggs by GC-HRMS using a novel atmospheric pressure photoionization source

Here, a new gas chromatography-atmospheric pressure photoionization-high-resolution mass spectrometry (GC-APPI-HRMS) method combined with selective pressurized liquid extraction (sPLE) has been developed for the selective determination of Dechlorane Plus (DP) and its related compounds in gull egg samples used as a bioindicator of contamination. To the best of our knowledge, this is the first time these compounds have been analyzed by GC-MS using atmospheric pressure photoionization (APPI). Negative ion dopant-assisted APPI using vapors of diethyl ether and a source temperature of 250°C provided high ionization efficiencies and mass spectra characterized by intense in-source fragment ions as well as the presence of molecular ion and characteristic cluster ions containing oxygen atoms in their chemical structure. This made it possible to improve the selectivity in the determination of these compounds compared to that obtained with traditional GC-MS ion sources. Under optimized conditions, the sPLE GC-APPI-HRMS (Orbitrap) method provided high recoveries (> 91%), good precisions (RSD% < 12%), and low method limits of detection (0.1-3.5pgg-1 wet weight). The developed methodology has been applied to the determination of DP and related compounds in eggs of two gull species (L. michahellis and L. audouinii) from several Spanish protected areas. The results obtained showed significant differences in the DP concentration profiles in eggs from different gull breeding locations and between gull species of the same protected area. These results demonstrated the good performance of the GC-APPI-HRMS system to achieve a selective and sensitive determination of DP and related compounds in complex environmental samples.

Deciphering Dissolved Organic Matter: Ionization, Dopant, and Fragmentation Insights via Fourier Transform-Ion Cyclotron Resonance Mass Spectrometry.

Fourier transform-ion cyclotron resonance mass spectrometry (FT-ICR MS) has been increasingly employed to characterize dissolved organic matter (DOM) across a range of aquatic environments highlighting the role of DOM in global carbon cycling. DOM analysis commonly utilizes electrospray ionization (ESI), while some have implemented other techniques, including dopant-assisted atmospheric pressure photoionization (APPI). We compared various extracted DOM compositions analyzed by negative ESI and positive APPI doped with both toluene and tetrahydrofuran (THF), including a fragmentation study of THF-doped riverine DOM using infrared multiple photon dissociation (IRMPD). DOM compositions followed the same trends in ESI and dopant-assisted APPI with the latter presenting saturated, less oxygenated, and more N-containing compounds than ESI. Between the APPI dopants, THF-doping yielded spectra with more aliphatic-like and N-containing compounds than toluene-doping. We further demonstrate how fragmentation of THF-doped DOM in APPI resolved subtle differences between riverine DOM that was absent from ESI. In both ionization methods, we describe a linear relationship between atomic and formulaic N-compositions from a range of DOM extracts. This study highlights that THF-doped APPI is useful for uncovering low-intensity aliphatic and peptide-like components in autochthonous DOM, which could aid environmental assessments of DOM across biolability gradients.

Rapid and highly sensitive measurement of trimethylamine in seawater using dynamic purge-release and dopant-assisted atmospheric pressure photoionization mass spectrometry

Trimethylamine (TMA) is ubiquitous in the marine systems and may affect atmospheric chemistry as a precursor and strong stabilizer of atmospheric secondary aerosol, influencing cloud formation. Rapid and accurate measurement of the concentration of TMA in seawater is challenging due to their polarity, aqueous solubility, volatility and existence at low concentrations in marine environments. In this study, a dopant-assisted atmospheric pressure photoionization time-of-flight mass spectrometry (DA-APPI-TOFMS) coupled with a dynamic purge-release method was developed for rapid and sensitive analysis of TMA in seawater. A novel three-zones ionization source has been developed for improving the ionization efficiency of analyte molecules and minimizing the influence of high-humidity of the sample gas, which allowed direct analysis of high-humidity (RH> 90%) gas samples from microbubble purging process by the mass spectrometer. At atmospheric pressure, the three-zones ionization source allows the use of high-speed purge gas to quickly purge all organic amines dissolved in the water into the gas phase, ensuring quantitative accuracy. The limit of quantification (LOQ) for TMA down to 0.1 μg L−1 was obtained in less than 2 min by consuming only 2 mL seawater sample. This method was applied for the determination of the concentrations of TMA in the coastal seawater to validate its practicability and reliability for analysis of trace amines in marine environments.

Charge Exchange Reaction in Dopant-Assisted Atmospheric Pressure Chemical Ionization and Atmospheric Pressure Photoionization.

The efficiencies of charge exchange reaction in dopant-assisted atmospheric pressure chemical ionization (DA-APCI) and dopant-assisted atmospheric pressure photoionization (DA-APPI) mass spectrometry (MS) were compared by flow injection analysis. Fourteen individual compounds and a commercial mixture of 16 polycyclic aromatic hydrocarbons were chosen as model analytes to cover a wide range of polarities, gas-phase ionization energies, and proton affinities. Chlorobenzene was used as the dopant, and methanol/water (80/20) as the solvent. In both techniques, analytes formed the same ions (radical cations, protonated molecules, and/or fragments). However, in DA-APCI, the relative efficiency of charge exchange versus proton transfer was lower than in DA-APPI. This is suggested to be because in DA-APCI both dopant and solvent clusters can be ionized, and the formed reagent ions can react with the analytes via competing charge exchange and proton transfer reactions. In DA-APPI, on the other hand, the main reagents are dopant-derived radical cations, which favor ionization of analytes via charge exchange. The efficiency of charge exchange in both DA-APPI and DA-APCI was shown to depend heavily on the solvent flow rate, with best efficiency seen at lowest flow rates studied (0.05 and 0.1 mL/min). Both DA-APCI and DA-APPI showed the radical cation of chlorobenzene at 0.05-0.1 mL/min flow rate, but at increasing flow rate, the abundance of chlorobenzene M(+.) decreased and reagent ion populations deriving from different gas-phase chemistry were recorded. The formation of these reagent ions explains the decreasing ionization efficiency and the differences in charge exchange between the techniques. Graphical Abstract ᅟ.

Analysis of gasoline contaminated water samples by means of dopant-assisted atmospheric pressure photoionization differential ion mobility spectrometry

In this study the influence of aromatic dopant benzene on the sensitivity of GC-APPI-DMS to gasoline related aromatic compounds was investigated. This influence was investigated on example of four gasolin related fingerprints (toluene, ethylbenzene, o-xylene, and 1,2,4-trimethylbenzene), which were found in high relative abundance in the water-soluble gasoline fraction. The analysis of calibration curves slopes demonstrats that the GC-APPI-DMS sensitivity to gasoline fingerprints can be improved by up to seven times when benzene concentration in nitrogen carrier gas is less than 10 ppmv/v. The estimated detection limits (S/N = 3) for the analyzed in this study compounds were found to be within the range of 33–105 μg L−1 at benzene concentration in the carrier gas of 2.27 ppmv/v (10 μL injection volume). These limits of detection may be reduced (at the cost of lower resolution) using the larger injection volumes. For example, increase of injection volume to 100 μL at benzene concentration in the carrier gas of 2.27 ppmv/v leads to reduction of LOD values for toluene, ethylbenzene, and o-xylene to 11.1, 13.3, and 5.3 μg L−1, respectively.

Nucleophilic Aromatic Substitution Between Halogenated Benzene Dopants and Nucleophiles in Atmospheric Pressure Photoionization.

In a preceding work with dopant assisted-atmospheric pressure photoionization (DA-APPI), an abundant ion at [M + 77](+) was observed in the spectra of pyridine and quinoline with chlorobenzene dopant. This contribution aims to reveal the identity and route of formation of this species, and to systematically investigate structurally related analytes and dopants. Compounds containing N-, O-, and S-lone pairs were investigated with APPI in the presence of fluoro-, chloro-, bromo-, and iodobenzene dopants. Computational calculations on a density functional theory (DFT) level were carried out to study the reaction mechanism for pyridine and the different halobenzenes. The experimental and computational results indicated that the [M + 77](+) ion was formed by nucleophilic aromatic ipso-substitution between the halobenzene radical cation and nucleophilic analytes. The reaction was most efficient for N-heteroaromatic compounds, and it was weakened by sterical effects and enhanced by resonance stabilization. The reaction was most efficient with chloro-, bromo-, and iodobenzenes, whereas with fluorobenzene the reaction was scarcely observed. The calculated Gibbs free energies for the reaction between pyridine and the halobenzenes were shown to increase in the order I < Br < Cl < F. The reaction was found endergonic for fluorobenzene due to the strong C-F bonding, and exergonic for the other halobenzenes. For fluoro- and chlorobenzenes the reaction was shown to proceed through an intermediate state corresponding to [M + dopant](+), which was highly stable for fluorobenzene. For the bulkier bromine and iodine, this intermediate did not exist, but the halogens were shown to detach already during the approach by the nucleophile.

Ionization of EPA contaminants in direct and dopant-assisted atmospheric pressure photoionization and atmospheric pressure laser ionization.

Seventy-seven EPA priority environmental pollutants were analyzed using gas chromatography-mass spectrometry (GC-MS) equipped with an optimized atmospheric pressure photoionization (APPI) and an atmospheric pressure laser ionization (APLI) interface with and without dopants. The analyzed compounds included e.g., polycyclic aromatic hydrocarbons (PAHs), nitro compounds, halogenated compounds, aromatic compounds with phenolic, acidic, alcohol, and amino groups, phthalate and adipatic esters, and aliphatic ethers. Toluene, anisole, chlorobenzene, and acetone were tested as dopants. The widest range of analytes was ionized using direct APPI (66/77 compounds). The introduction of dopants decreased the amount of compounds ionized in APPI (e.g., 54/77 with toluene), but in many cases the ionization efficiency increased. While in direct APPI the formation of molecular ions via photoionization was the main ionization reaction, dopant-assisted (DA) APPI promoted ionization reactions, such as charge exchange and proton transfer. Direct APLI ionized a much smaller amount of compounds than APPI (41/77 compounds), showing selectivity towards compounds with low ionization energies (IEs) and long-lived resonantly excited intermediate states. DA-APLI, however, was able to ionize a higher amount of compounds (e.g. 51/77 with toluene), as the ionization took place entirely through dopant-assisted ion/molecule reactions similar to those in DA-APPI. Best ionization efficiency in APPI and APLI (both direct and DA) was obtained for PAHs and aromatics with O- and N-functionalities, whereas nitro compounds and aliphatic ethers were the most difficult to ionize. Halogenated aromatics and esters were (mainly) ionized in APPI, but not in APLI.

Exploring the peptide fragmentation mechanisms under atmospheric pressure photoionization using tunable VUV synchrotron radiation

Intensive fragmentations into b/y- and c-sequence ions have been reported for Substance P ionized under dopant-assisted atmospheric pressure photoionization (APPI) conditions. It was postulated that the fragmentation comes from the reaction of the doubly charged peptide with photoelectrons, released upon photoionization of the dopant, through an electron capture or transfer dissociation-type mechanism. In order to investigate further the origin of each fragment ion type, a tunable VUV synchrotron radiation source coupled to a modified APPI source has been used. Using this set-up, the photon energy can be scanned continuously below and above the photoionization threshold of the dopant, i.e. toluene, probing the ionization/fragmentation processes occurring on Substance P peptide at the atmospheric pressure interface of the mass spectrometer.

The ionization mechanisms in direct and dopant-assisted atmospheric pressure photoionization and atmospheric pressure laser ionization.

A novel, gas-tight API interface for gas chromatography-mass spectrometry was used to study the ionization mechanism in direct and dopant-assisted atmospheric pressure photoionization (APPI) and atmospheric pressure laser ionization (APLI). Eight analytes (ethylbenzene, bromobenzene, naphthalene, anthracene, benzaldehyde, pyridine, quinolone, and acridine) with varying ionization energies (IEs) and proton affinities (PAs), and four common APPI dopants (toluene, acetone, anisole, and chlorobenzene) were chosen. All the studied compounds were ionized by direct APPI, forming mainly molecular ions. Addition of dopants suppressed the signal of the analytes with IEs above the IE of the dopant. For compounds with suitable IEs or Pas, the dopants increased the ionization efficiency as the analytes could be ionized through dopant-mediated gas-phase reactions, such as charge exchange, proton transfer, and other rather unexpected reactions, such as formation of [M + 77](+) in the presence of chlorobenzene. Experiments with deuterated toluene as the dopant verified that in case of proton transfer, the proton originated from the dopant instead of proton-bound solvent clusters, as in conventional open or non-tight APPI sources. In direct APLI using a 266 nm laser, a narrower range of compounds was ionized than in direct APPI, because of exceedingly high IEs or unfavorable two-photon absorption cross-sections. Introduction of dopants in the APLI system changed the ionization mechanism to similar dopant-mediated gas-phase reactions with the dopant as in APPI, which produced mainly ions of the same form as in APPI, and ionized a wider range of analytes than direct APLI.

Dopant-Assisted Atmospheric Pressure Photoionization of Patulin in Apple Juice and Apple-Based Food with Liquid Chromatography–Tandem Mass Spectrometry

A dopant-assisted atmospheric pressure photoionization (APPI) with liquid chromatography tandem mass spectrometry (LC-MS/MS) method was developed to determine patulin in apple juice and apple-based food. Different dopants, dopant flow rates, and LC separation conditions were evaluated. Using toluene as the dopant, the LC-APPI-MS/MS method achieved a linear calibration from 12.5 to 2000 μg/L (r(2) > 0.99). Matrix-dependent limits of quantitation (LOQs) were from 8 μg/L (solvent) to 12 μg/L (apple juice). [(13)C]-Patulin-fortified apple juice samples were directly analyzed by the LC-APPI-MS/MS method. Other apple-based food was fortified with [(13)C]-patulin, diluted using water (1% formic acid), centrifuged, and filtered, followed by LC-APPI-MS/MS analysis. In clear apple juice, unfiltered apple cider, applesauce, and apple-based baby food, average recoveries were 101 ± 6% (50 μg/kg), 103 ± 5% (250 μg/kg), and 102 ± 5% (1000 μg/kg) (av ± SD, n = 16). Using the suggested method, patulin was detected in 3 of 30 collected market samples with concentrations ranging from <LOQ to 18 μg/L. The use of [(13)C]-patulin allowed quantitation using solvent calibration standards with satisfactory precision and accuracy.

Fully automated trace level determination of parent and alkylated PAHs in environmental waters by online SPE-LC-APPI-MS/MS

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous compounds that enter the environment from natural and anthropogenic sources, often used as markers to determine the extent, fate, and potential effects on natural resources after a crude oil accidental release. Gas chromatography-mass spectrometry (GC-MS) after liquid-liquid extraction (LLE+GC-MS) has been extensively used to isolate and quantify both parent and alkylated PAHs. However, it requires labor-intensive extraction and cleanup steps and generates large amounts of toxic solvent waste. Therefore, there is a clear need for greener, faster techniques with enough reproducibility and sensitivity to quantify many PAHs in large numbers of water samples in a short period of time. This study combines online solid-phase extraction followed by liquid chromatography (LC) separation with dopant-assisted atmospheric pressure photoionization (APPI) and tandem MS detection, to provide a one-step protocol that detects PAHs at low nanograms per liter with almost no sample preparation and with a significantly lower consumption of toxic halogenated solvents. Water samples were amended with methanol, fortified with isotopically labeled PAHs, and loaded onto an online SPE column, using a large-volume sample loop with an auxiliary LC pump for sample preconcentration and salt removal. The loaded SPE column was connected to an UPLC pump and analytes were backflushed to a Thermo Hypersil Green PAH analytical column where a 20-min gradient separation was performed at a variable flow rate. Detection was performed by a triple-quadrupole MS equipped with a gas-phase dopant delivery system, using 1.50mL of chlorobenzene dopant per run. In contrast, LLE+GC-MS typically use 150mL of organic solvents per sample, and methylene chloride is preferred because of its low boiling point. However, this solvent has a higher environmental persistence than chlorobenzene and is considered a carcinogen. The automated system is capable of performing injection, online SPE, inorganic species removal, LC separation, and MS/MS detection in 28min. Selective reaction monitoring was used to detect 28 parent PAHs and 15 families of alkylated PAHs. The methodology is comparable to traditional GC-MS and was tested with surface seawater, rainwater runoff, and a wastewater treatment plant effluent. Positive detections above reporting limits are described. The virtual absence of sample preparation could be particularly advantageous for real-time monitoring of discharge events that introduce PAHs into environmental compartments, such as accidental releases of petroleum derivates and other human-related events. This work covers optimization of APPI detection and SPE extraction efficiency, a comparison with LLE+GC-MS in terms of sensitivity and chromatographic resolution, and examples of environmental applications.

Determination of 17 illicit drugs in oral fluid using isotope dilution ultra-high performance liquid chromatography/tandem mass spectrometry with three atmospheric pressure ionizations

The collection of oral fluid for drug testing is easy and non-invasive. This study developed a drug testing method using ultra-high performance liquid chromatography/tandem mass spectrometry (UHPLC–MS/MS) in selected-reaction monitoring (SRM) mode. We tested the method on the analysis of four opiates and their metabolites, five amphetamines, flunitrazepam and its two metabolites, and cocaine and its four metabolites in oral fluid. 100-μL samples of oral fluid were diluted with twice the amount of water then spiked with isotope-labeled internal standards. After the samples had undergone high-speed centrifugation for 20 min, we analyzed the supernatant. The recovery of the sample preparation ranged from 81 to 108%. We compared the performance of electrospray ionization (ESI), atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization (APPI). The ion suppression of most analytes on ESI (28–78%) was lower than that of APCI and APPI. A post-column flow split (5:1) did not reduce the matrix effect on ESI. Direct APPI performed better than dopant-assisted APPI using toluene. ESI, APCI and APPI limits of quantitation mostly ranged from 0.11 to 1.9 ng/mL, 0.02 to 2.2 ng/mL and 0.02 to 2.1 ng/mL, respectively, but were much higher on amphetamine and ecgonine methyl ester (about 2.7–4.7 ng/mL, 8.7–14 ng/mL, and 10–19 ng/mL, respectively). Most of the bias percentages (accuracy) and relative standard deviations (precision) on spiked samples were below 15%. This method greatly simplifies the process of sample preparation and shortens the chromatographic time to only 7.5 min per run and is able to detect analytes at sub-ppb levels.

Dopant-Assisted Atmospheric Pressure Photoionization Mass Spectrometry of Polyisobutylene Derivatives Initiated by Mono- and Bifunctional Initiators

Nine polyisobutylene (PIB) derivatives with different end groups (chlorine, vinyl, isobutenyl, 2,2-diphenylvinyl, and carboxyl) and molecular weights (1000 to 4500 g/mol), initiated by monofunctional and aromatic bifunctional initiators were studied by atmospheric pressure photoionization mass spectrometry (APPI-MS) in both negative and positive ion modes. Consistent with previous findings, negative ion APPI-MS revealed end-group identities through the formation of PIB adducts with chloride ions formed in situ from a chlorinated solvent (e.g., CCl4) in the presence of a dopant (toluene). In positive ion mode, considerable fragmentation of these PIB derivatives was observed, rendering end-group determinations very difficult. The M(n) values obtained by APPI(-)-MS were considerably lower than those determined by SEC for PIB derivatives with M(n) higher than 2000 g/mol. PIBs containing carboxyl termini can undergo collision-induced dissociation, yielding structurally important product ions. The resulting APPI-MS/MS intensities were found to reflect the "arm-length" distribution for PIBs with bifunctional aromatic moieties. In positive ion mode, [M + COCl]+ adducts were observed for PIBs with an aromatic initiator moiety. The origin of the COCl+ species is also discussed.

MECHANISM OF IONIZATION OF POLYCYCLIC AROMATIC HYDROCARBONS BY A TOLUENE/ANISOLE MIXTURE AS A DOPANT IN LIQUID CHROMATOGRAPHY/DOPANT-ASSISTED ATMOSPHERIC-PRESSURE PHOTOIONIZATION/MASS SPECTROMETRY

Ionization mechanisms of 16 polycyclic aromatic hydrocarbons (PAHs) by dopant-assisted atmospheric-pressure photoionization (DA-APPI) with a toluene/anisole mixture as a dopant were examined in the reversed-phase liquid chromatography mode with a methanol/water to acetonitrile linear gradient mobile phase. When toluene/anisole mixtures were used as dopants, their characteristics were governed by anisole. Since the IE of anisole (8.20 eV) is below that of toluene (8.83 eV), anisole is ionized not only by photons from the krypton lamp but also by charge transfer from toluene radical cations. In the latter half of chromatogram, when acetonitrile was dominant in the eluent, ions originating from both toluene and anisole decreased while acetonitrile-cluster ions began to increase. Because acetonitrile-cluster ions are not formed with anisole alone, but are formed with toluene alone, toluene radical cations would be consumed to form acetonitrile-cluster ions.

Dopant assisted-atmospheric pressure photoionization (DA-APPI) liquid chromatography–mass spectrometry for the quantification of 27-hydroxycholesterol in plasma

27-Hydroxycholesterol (27OH-Chol) is of potential diagnostic interest due to its role in maintaining whole-body cholesterol homeostasis. Dopant assisted-atmospheric pressure photoionization (DA-APPI) has improved the sensitivity of 27OH-Chol analysis, in comparison to the published LC-APCI–MS method, allowing quantification from a very low amount of sample (≤50 μL plasma). The method was validated for quantification from 50 μL and 15 μL plasma, with the limit of quantification (LOQ) of 10 ng/mL and 40 ng/mL plasma, respectively. A further advantage is that no prior derivatization was needed, unlike the LC-ESI–MS or the standard GC–MS method. The method validation also resulted in good linearity and recovery of 91–106%. The within-day and between-day coefficient of variation were less than 15% while giving the accuracy of 90.9–113.4%. This report summarizes the effects of some critical parameters on the ionization of 27OH-Chol using APPI, as well as the validation of the method. The sensitivity achieved with DA-APPI broadens the usefulness of the LC–MS method in clinical applications.

Mechanism of [M + H] + Formation in Atmospheric Pressure Photoionization Mass Spectrometry: Identification of Propionitrile in Acetonitrile with High Mass Accuracy Measurement and Tandem Mass Spectrometry and Evidence for Its Involvement in the Protonation Phenomenon

The role of propionitrile in the production of [M+H]+ under atmospheric pressure photoionization (APPI) was investigated. In dopant-assisted APPI using acetone and anisole, protonated acetone and anisole radical cations were the most prominent ions observed. In dopant-free or direct APPI in acetonitrile, however, a major ion in acetonitrile was detected and identified as propionitrile, using high accuracy mass measurement and collision induced dissociation studies. Vaporizing ca. 10(-5) M althiazide and bendroflumethazide under direct APPI in acetonitrile produced their corresponding protonated species [M+H]+. In addition to protonated acetonitrile, its dimers, and acetonitrile/water clusters, protonated propionitrile, propionitrile dimer, and propionitrile/water clusters were also observed. The role of propionitrile, an impurity in acetonitrile and/or a possible product of ion-molecule reaction, in the production of [M+H]+ of althiazide and bendroflumethazide was further investigated in the absence of dopant using propionitrile-d5. The formation of [M+D]+ species was observed, suggesting a possible role of propionitrile in the protonation process. Additionally, an increase in the [M+H]+ signal of althiazide and bendroflumethazide was observed as a function of propionitrile concentration in acetonitrile. Theoretical data from the literature supported the assumption that one possible mechanism, among others, for the formation of [M+H]+ could be attributed to photo-initiated isomerization of propionitrile. The most stable isomers of propionitrile, based on their calculated ionization energy (IE) and relative energy (DeltaE), were assumed to undergo proton transfer to the analytes, and mechanisms were proposed.

High-performance liquid chromatography-atmospheric pressure photoionization/tandem mass spectrometry for the detection of 17alpha-ethinylestradiol in hepatocytes

Atmospheric pressure photoionization (APPI) as an interface for the high-performance liquid chromatography (HPLC)–tandem mass spectrometry (MS/MS) system was employed for the direct determination of 17alpha-ethinylestradiol (EE 2) in the incubation mixtures to support in vitro hepatic clearance studies. For the APPI source, the radical cation of the analyte via charge exchange with the dopant radical cation was used for the detection of EE 2 in the positive ion mode. It was demonstrated that the major signals of EE 2 in the acetonitrile/water mobile phase were substantially increased by replacing toluene with anisole as the dopant. The effects of several experimental conditions on the photoionization efficiency of EE 2 in the dopant-assisted APPI source were explored. Electrospray ionization (ESI) source was also suitable for the analysis of the analyte; however, ESI required a derivatization step prior to analysis. The applicability of the proposed HPLC–APPI–MS/MS approach following a protein precipitation procedure for the determination of EE 2 at low nano-mole levels was examined with respect to assay specificity and linearity. The assay results obtained by both HPLC–APPI–MS/MS and HPLC–ESI–MS/MS methods were in good agreement.
