Chemical synthesis of per-selenocysteine human epidermal growth factor.

Human seleno-epidermal growth factor (seleno-EGF), a 53-residue peptide where all six cysteine residues of the parent human EGF sequence were replaced by selenocysteines, was synthesized and the oxidative folding of a polypeptide containing three diselenide bonds was compared to that of the parent cysteine peptide. The crude high performance liquid chromatography (HPLC) profiles clearly showed that both the native EGF and its selenocysteine-analogue fold smoothly, yielding a single sharp peak, proving that even in the case of three disulfide-bonded polypeptides the disulfide-to-diselenide bond substitution is highly isomorphous, as confirmed by conformational circular dichroism measurements and particularly by the biological assays.

Distinct Disulfide Isomers of μ-Conotoxins KIIIA and KIIIB Block Voltage-Gated Sodium Channels

In the preparation of synthetic conotoxins containing multiple disulfide bonds, oxidative folding can produce numerous permutations of disulfide bond connectivities. Establishing the native disulfide connectivities thus presents a significant challenge when the venom-derived peptide is not available, as is increasingly the case when conotoxins are identified from cDNA sequences. Here, we investigate the disulfide connectivity of μ-conotoxin KIIIA, which was predicted originally to have a [C1-C9,C2-C15,C4-C16] disulfide pattern based on homology with closely related μ-conotoxins. The two major isomers of synthetic μ-KIIIA formed during oxidative folding were purified and their disulfide connectivities mapped by direct mass spectrometric collision-induced dissociation fragmentation of the disulfide-bonded polypeptides. Our results show that the major oxidative folding product adopts a [C1-C15,C2-C9,C4-C16] disulfide connectivity, while the minor product adopts a [C1-C16,C2-C9,C4-C15] connectivity. Both of these peptides were potent blockers of Na(V)1.2 (K(d) values of 5 and 230 nM, respectively). The solution structure for μ-KIIIA based on nuclear magnetic resonance data was recalculated with the [C1-C15,C2-C9,C4-C16] disulfide pattern; its structure was very similar to the μ-KIIIA structure calculated with the incorrect [C1-C9,C2-C15,C4-C16] disulfide pattern, with an α-helix spanning residues 7-12. In addition, the major folding isomers of μ-KIIIB, an N-terminally extended isoform of μ-KIIIA identified from its cDNA sequence, were isolated. These folding products had the same disulfide connectivities as μ-KIIIA, and both blocked Na(V)1.2 (K(d) values of 470 and 26 nM, respectively). Our results establish that the preferred disulfide pattern of synthetic μ-KIIIA and μ-KIIIB folded in vitro is 1-5/2-4/3-6 but that other disulfide isomers are also potent sodium channel blockers. These findings raise questions about the disulfide pattern(s) of μ-KIIIA in the venom of Conus kinoshitai; indeed, the presence of multiple disulfide isomers in the venom could provide a means of further expanding the snail's repertoire of active peptides.

Isolation and characterization of factor I of the bovine complement system.

To isolate and characterize factor I of the bovine complement system. Sample Population-Serum obtained from the blood of beef cattle. Serum samples were fractionated to yield factor I by means of sequential precipitation, ion-exchange, and gel-filtration chromatography. The protein was identified throughout the procedure on the basis of its ability to degrade the alpha'-chain of bovine C3b in the presence of bovine factor H. Electrophoresis in polyacrylamide gels was used to assess the degradation of C3b and determine the molecular weights of factor I and its component polypeptide chains. Bovine factor I had an apparent molecular weight of 94 kd and consisted of 2 disulfide-bonded polypeptides that had apparent molecular weights of 51 and 42 kd (under reducing conditions). Factor H was required for the factor I cleavage of the alpha'-chain of bovine C3b into iC3b. A similar cofactor effect was provided by trypsinized bovine erythrocytes or erythrocyte ghosts. Bovine properdin was prepared and shown to be a single polypeptide chain of 58 kd in the reduced form. Bovine factor I can be purified from serum by a simple 4-step procedure. It is structurally and functionally comparable to factor I of other species, and its purification completes the isolation and characterization of all the soluble components of the bovine alternative complement pathway.

Sizing the protein translocation pathway of colicin Ia channels.

The bacterial toxin colicin Ia forms voltage-gated channels in planar lipid bilayers. The toxin consists of three domains, with the carboxy-terminal domain (C-domain) responsible for channel formation. The C-domain contributes four membrane-spanning segments and a 68-residue translocated segment to the open channel, whereas the upstream domains and the amino-terminal end of the C-domain stay on the cis side of the membrane. The isolated C-domain, lacking the two upstream domains, also forms channels; however, the amino terminus and one of the normally membrane-spanning segments can move across the membrane. (This can be observed as a drop in single-channel conductance.) In longer carboxy-terminal fragments of colicin Ia that include ≤169 residues upstream from the C-domain, the entire upstream region is translocated. Presumably, a portion of the C-domain creates a pathway for the polar upstream region to move through the membrane. To determine the size of this translocation pathway, we have attached “molecular stoppers,” small disulfide-bonded polypeptides, to the amino terminus of the C-domain, and determined whether they could be translocated. We have found that the translocation rate is strongly voltage dependent, and that at voltages ≥90 mV, even a 26-Å stopper is translocated. Upon reduction of their disulfide bonds, all of the stoppers are easily translocated, indicating that it is the folded structure, rather than some aspect of the primary sequence, that slows translocation of the stoppers. Thus, the pathway for translocation is ≥26 Å in diameter, or can stretch to this value. This is large enough for an α-helical hairpin to fit through.

Characterization of seed storage proteins and their synthesis during seed development in Olea europaea

SDS-PAGE analysis showed that seed storage proteins (SSPs) in olive mainly consisted of two hydrogen-bonded subunits, Solea I (41 kDa) and Solea II (47.5 kDa). When reduced, Solea I and II yielded 3 and 2 disulfide-bonded polypeptides, respectively. N-terminal sequencing confirmed that both Solea I and II shared a high similarity with II S proteins. Additionally, time-course of protein synthesis in olive seeds was investigated from 25 days after anthesis (DAA) until seed ripening. Before 105 DAA, young seeds contained only trace amounts of detectable proteins. Only from 105 DAA onwards, seed proteins began to be massively synthesized. Seeds of 105 DAA and more exhibited no significant difference in protein profiles with mature seeds, especially in terms of Solea I and II.

Preparation of Disulfide-Bonded Polypeptide Heterodimers by Titration of Thio-Activated Peptides with Thiol-Containing Peptides

Titration of 2-pyridinesulfenyl (SPyr)-protected cysteine-containing peptides with thiol-unprotected peptides has proven to be a superior procedure to selectively generate disulfide-linked peptide heterodimers preventing the formation of homodimers. This procedure was used to synthesize large amounts of highly purified peptide heterodimers consisting of intracellularly active moieties, that were coupled to the third α-helix of the transcription factor Antennapedia, which serves as a ‘shuttle tag’.

The glutenin macropolymer of wheat flour doughs: structure–function perspectives

Gluten is the main functional component of wheat, and is the main source of the viscoelastic properties in a dough. Dough properties are primarily governed by the structure of gluten, and interactions within the protein complex. While starch and water are the main components of doughs, the physical properties of a dough arise from interactions between gluten proteins, particularly the disulfide-bonded glutenin macropolymer. The primary structures of glutenin subunits are well characterized, but information on the specificity and pattern on their interactions has been lacking, despite the fact that dough is one of the key food systems where the likely link between molecular protein structure and function is compelling. The structure of polymeric glutenin has been difficult to study since it comprises several dozen different disulfide-bonded polypeptides in the molecular mass range of millions to hundreds of millions. Recently, a range of techniques has been used to help unravel structural information. These include biochemical methods, particularly chromatography and electrophoresis systems which are suitable for examining polydisperse high molecular weight protein complexes of limited solubility; small-scale rheological testing systems, in which the function of individual components (either isolated or from expressed genes) can be determined; and a range of microscopy-based techniques, including confocal light scanning microscopy, transmission electron microscopy, and scanning electron microscopy. Utilizing these techniques, an improved understanding of the relationship between polymeric glutenin structure and dough function has been obtained. It has been proposed that glutenin subunits provide a structural backbone to the glutenin macropolymer through the formation of disulfide bonds that are highly resistant to cleavage [1]. The inherent ability of glutenin subunits to form disulfide bonds is thought to be determined by the primary and secondary structure of the proteins, which determines whether cysteine residues are present and available to form disulfide bonds, the capacity of a subunit to fold in the manner that would be required to form the bond, and the elasticity of the subunit once in the polymer to provide visco-elastic properties to a dough [2]. In this review, we discuss the structure of the glutenin macropolymer of wheat flour and dough.

Biosynthesis and processing of type XVI collagen in human fibroblasts and smooth muscle cells.

The alpha 1(XVI) collagen chain, recently identified by cDNA cloning, exhibits structural similarity to a subgroup of collagens that associate with collagen fibrils. Recombinant alpha 1(XVI) collagen chains produced in embryonic kidney cells are able to form stable homotrimers, which are rapidly converted into smaller polypeptides after secretion into the culture medium. In this study, we investigated the biosynthesis of native type XVI collagen by immunoprecipitation of metabolically labeled human cells. Dermal fibroblasts and arterial smooth muscle cells were precipitated with three antibodies raised against distinct regions in the N- and C-terminal part of the human alpha 1(XVI) collagen chain. A disulfide-bonded polypeptide of 220 kDa was obtained from the culture medium, cells and extracellular matrix with all three antibodies. This polypeptide is sensitive to bacterial collagenase digestion and partially resistant to pepsin digestion, suggesting that it is the endogenous alpha 1(XVI) collagen chain. Pulse/chase experiments showed that the newly synthesized alpha 1(XVI) chains are secreted into the medium and deposited in the extracellular matrix in a time-dependent manner. Unlike the recombinant chain, the native type XVI collagen does not undergo extensive proteolytic processing upon secretion. Both cell types deposit a substantial amount of the newly synthesized alpha 1(XVI) chain into the extracellular matrix, in which the 220-kDa polypeptide is the only product immunoprecipitated. There is little evidence for the presence of another constituent chain. The data are consistent with a nomotrimeric chain composition for type XVI collagen. No apparent difference exists in the rate of synthesis and secretion between fibroblasts and smooth muscle cells. Indirect immunofluorescence microscopy showed an extracellular distribution of type XVI collagen, which is located close to cells but not associated with fibrillar structures.

Expression in Escherichia coli of Sin a 1, the Major Allergen from Mustard

Sin a 1, the major yellow mustard allergen, is a seed storage protein that belongs to the 2S albumin family. It is composed of two disulfide-bonded polypeptide chains. The cloning of this allergen has been carried out by means of the polymerase chain reaction using non-degenerate oligonucleotides encoding the N-terminal and C-terminal regions of the mature protein as primers. Five genomic nucleotide sequences have been analyzed, encoding both mature polypeptide chains linked by the internal processed fragment. The sequence data show the existence of microheterogeneities at ten positions, demonstrating the polymorphism exhibited by the natural protein. One of the genomic clones was expressed in Escherichia coli by fusion to glutathione S-transferase from Schistosoma japonicum. The resulting chimeric protein was purified by affinity chromatography on a glutathione-Sepharose 4B matrix, and digested with thrombin to release the recombinant allergen. The recombinant Sin a 1 is recognized by rabbit polyclonal and mouse monoclonal antisera raised against natural Sin a 1, as well as by the IgE of mustard-sensitive human sera. In addition, recombinant Sin a 1 possesses a high resistance to trypsin digestion, like the native mustard allergen.

Recombinant Analysis of Human alpha1(XVI) Collagen. Evidence for Processing of the N-Terminal Globular Domain

The N-terminal non-collagenous domain NC11 of the human collagen α1(XVI) chain was obtained as a recombinant 35-kDa protein from stably transfected kidney cell clones. This form had undergone proteolytic trimming at a basic cleavage motif indicating a similar release in vivo. Domain NC11 showed a globular shape after rotary shadowing and was resistant to neutral proteases. Specific antibodies could be raised against recombinant NC11 and were used for the analysis of other cell clones transfected with the full-length α1(XVI) chain. Immunoprecipitation of detergent extracts of metabolically labelled cells demonstrated the presence of disulfide-bonded 200-kDa polypeptides possessing NC11 epitopes. This material was partially resistant to pepsin, indicating the formation of α1(XVI) chain homotrimers with a triple-helical conformation. Yet a substantial proportion of these homotrimers was degraded to fragments of variable size (35–150 kDa) when secreted into the culture medium. Several of these fragments could be obtained on a semi-preparative scale from cells grown in hollow fiber cassettes and showed substantial hydroxylation of proline, consistent with triple-helix formation. Edman degradation demonstrated the origin of some from the N-terminal and of one from a more C-terminal position of collagen XVI. This extensive degradation may be explained by the release of NC11 and by further cleavages within some of the nine interruptions of the triple-helical domain of the α1(XVI) chain. Whether this process also occurs in situ remains to be shown.

Platelet-derived growth factor expression in a transgenic model.

The migration, proliferation and organization of cells in tissues are controlled by several forms of intercellular communication. One form of control involves a network of diffusible polypeptide growth factors that activate membrane receptors and transduce proliferative signals. One of these growth factors, platelet-derived growth factor (PDGF), has provided a framework for understanding the cellular and molecular events that underlie the biological effects of a number of growth factors. PDGF is a family of several closely related 30 kDa dimers consisting of two disulfide-bonded polypeptide chains, designated A and B [reviewed in 1]. All three isoforms of PDGF (AA-, AB- and BB) are biologically active. Two distinct PDGF receptors have been identified: one that binds either to the A- or the B-chain (the α subunit) and one that binds only the B-chain (the β subunit) [reviewed in 2]. The capacity of cells to respond to PDGF depends upon the presence of a particular dimeric form of PDGF as well as expression of appropriate receptors. PDGF is both mitogenic and chemotactic for cells such as fibroblasts, smooth muscle cells and glial cells [reviewed in 3]. Because of these properties, PDGF has been implicated in stimulating the proliferation and cell movement of cells in normal developmental and reparative responses (for example, wound healing), as well as in pathologic processes, such as atherosclerosis and neoplasia. However, there is little compelling evidence that PDGF actually plays a critical role in any of these processes in vivo. To fully evaluate the possible functions of PDGF, a detailed description of ligand production, as well as PDGF receptor expression, is required in each of these settings. Surprisingly, such documentation is only preliminary. Standard approaches to localize PDGF expression in vivo have employed either immunohistochemistry or in situ hybridization. However, these methods have several potential disadvantages: first, PDGF is a secreted protein, which implies that cytoplasmic levels of the protein in intact animals may be low; second, isoform-specific antibodies which cross species are not readily available; and third, transcript levels of PDGF B-chain may be low in vivo because the mRNA has a short half-life. To circumvent these potential problems, a different approach was taken.

Separation of disulfide-bonded polypeptides using two-dimensional diagonal gel electrophoresis

A simple method for the separation of disulfide-bonded polypeptides using two-dimensional diagonal gel electrophoresis is described. The first dimension involves standard SDS-PAGE of nonreduced proteins. The separated proteins are then subjected to reduction before electrophoresis in the second dimension. This technique has several applications. It can be utilized to detect the presence of disulfide-bonded polypeptides within a protein, as an assay to detect the presence of a protein known to contain disulfide-bonded polypeptides in a mixture of proteins, and as a means of separating disulfide-bonded polypeptides from proteins of similar molecular weight.

Structural analysis of bovine pancreatic thread protein

Pancreatic thread protein (PTP) forms double helical threads in the neutral pH range after purification, undergoing freely reversible, pH-dependent globule-fibril transformation. The purified bovine PTP consists on SDS gels of two carbohydrate-free polypeptide chains (Gross et al., 1985). Plasma desorption mass spectrometry and amino acid sequence analysis now confirm that bovine PTP contains two disulfide-bonded polypeptides, an A chain of 101 amino acid residues with a molecular weight of 11,073 and a B chain of 35 residues with a molecular weight of 3970. The intact protein exhibits a molecular weight of 15,036, agreeing greater than 99.9% with the molecular weight calculated from the sequence. The B chain sequence was determined by gas-phase Edman degradation of the intact polypeptide. The A chain sequence was determined from overlapping peptides generated by cleavage at lysyl, tryptophanyl, and aspartyl-prolyl residues. Based upon the bovine PTP cDNA structure, the two chains of the protein result from cleavage of a single polypeptide with removal of a dipeptide between the NH2-terminal A chain and COOH-terminal B chain. Comparison of bovine PTP with other proteins reveals significant structural relatedness with the single-chain homologues from human and rat pancreas and with the motif associated with Ca2(+)-dependent carbohydrate recognition domains. The physiological role of PTP has not yet been resolved. The protein is present in very high concentration in pancreatic secretion and it has been detected in brain lesions in Alzheimer's disease and Down syndrome and in regenerating rat pancreatic islets. The present results provide a firm protein base for ongoing molecular, physical-chemical, and structure-function studies of this unusual protein.

The structure of human collagen type IX and its organization in fetal and infant cartilage fibrils.

Human collagen type IX was isolated from the media of organ cultures of fetal or infant hyaline cartilage. It consisted of three distinct, disulfide-bonded polypeptides of 115, 84, and 72 kDa, respectively. Digestion with chondroitinase ABC reduced the apparent molecular mass of the 115-kDa chain to about 65 kDa demonstrating that also human collagen type IX is a proteoglycan. In the electron microscope, the molecule had a rigid rod-like structure with characteristic kinks and with a globular domain at one end. Digestion of human collagen type IX with pepsin leads to somewhat heterogeneous fragments. Affinity-purified antibodies to the mixture of fragments specifically reacted with the fragment HMW without cross-reaction with chicken HMW. LMW of both species were recognized to the same low extent. Mechanically generated fibril fragments from human fetal cartilage were heterogeneous in diameter. Significantly, they could be immunostained for collagen type IX in a D-periodic pattern and regardless of the fibril diameter. Some fibrils were poorly labeled, again independently of the diameter. Therefore, the role of collagen type IX in cartilage probably is not to control directly the lateral growth during fibrillogenesis but rather to stabilize the fibril network.

Plasma cell membrane glycoprotein PC-1. Primary structure deduced from cDNA clones.

The PC-1 protein is a membrane glycoprotein that is selectively expressed on the surface of antibody-secreting cells. Previous work has shown that it consists of two apparently identical disulfide-bonded polypeptides, each of molecular weight approximately 120,000. We now describe the sequence of PC-1 mRNA and protein. The PC-1 protein is shown to consist of 905 amino acids and to have an uncommon transmembrane orientation. The NH2-terminal 58 residues are intracellular and the COOH-terminal 826 residues are extracellular. A cysteine-rich region of 85 amino acids lies adjacent to the extracellular surface of the membrane and appears to have arisen by exon duplication. In common with other membrane glycoproteins with this orientation, there is no obvious signal sequence other than the transmembrane segment. The PC-1 protein therefore has an overall structure and membrane orientation that resembles those of the transferrin receptor, the asialoglycoprotein receptor, and the Ia invariant chain.

Structure of proteoheparan sulfates from fibroblasts. Confluent and proliferating fibroblasts produce at least three types of proteoheparan sulfates with functionally different core proteins.

[3H]Leucine- and [35S]sulfate-labeled proteoheparan sulfates were isolated from postconfluent or proliferating cultures of human skin fibroblasts. Cell layers were solubilized by Triton X-100, and transferrin-binding macromolecules were isolated by affinity chromatography. Proteoglycans with no affinity for transferrin were purified by using ion-exchange and gel permeation chromatography. Postconfluent cells synthesize a proteoheparan sulfate of Mr 350,000 (as determined by gel permeation chromatography) which has affinity for transferrin as well as for octyl-Sepharose. Its core protein (Mr 180,000) consists of two disulfide-bonded polypeptides of Mr 90,000. This species was not detected in cultures of proliferating cells. Proliferating and confluent cells also synthesize other forms of proteoheparan sulfates (Mr 200,000-400,000) which have no affinity for transferrin. However, most of them have affinity for octyl-Sepharose. The core protein of proteoheparan sulfates made by proliferating cells has Mr 50,000. A smaller form (Mr 250,000) of this proteoglycan was solubilized by Triton X-100, whereas a larger form (Mr 400,000) remained associated with the pericellular matrix. A third type of proteoheparan sulfate (Mr 200,000) without affinity for transferrin nor octyl-Sepharose was associated with postconfluent cell layers but not with proliferating ones. Its core protein has Mr 35,000. Heparan sulfate oligosaccharides (Mr 6,000 or higher) were found in proliferating cells but not in postconfluent ones.

The murine plasma cell antigen PC-1: purification and partial amino acid sequence.

The murine plasma cell alloantigen PC-1 is selectively expressed on B lymphocytes in their terminal phase of differentiation into antibody-secreting cells. Previous work on an analytical scale has shown that PC-1 consists of two apparently identical disulfide-bonded polypeptides, each of Mr 115,000. In this paper, we describe the generation of a monoclonal antibody to PC-1 and its use in the preparative isolation of PC-1 by affinity chromatography. Final purification to apparent homogeneity was achieved by preparative polyacrylamide gel electrophoresis. It was estimated that NS-1 myeloma cells possess 1 to 4 X 10(5) PC-1 monomers per cell on their surface. The yield of PC-1 after purification was approximately 10(5) monomers per cell. Purified PC-1 was digested with trypsin, and the resulting peptides were separated by reversed-phase high-performance liquid chromatography. Purified peptides were sequenced with a gas-phase sequencer.

Unique surface phenotype of T cells in lymphoproliferative autoimmune MRL/Mp-lpr/lpr mice.

MRL/l mice, which carry the mutant gene lpr, develop massive T cell lymphoproliferation and an autoimmune syndrome. The surface antigen profile of MRL/l lymphocytes was analyzed with rat monoclonal antibodies. They included YE1/9.9, anti-transferrin receptor; YE1/7.1, which reacts with a population of proliferating immature T cells; and YE1/19.1, a newly identified antibody reactive with a surface antigen on EL-4 cells, which consists of two disulfide-bonded polypeptide chains, each of 115,000 m.w. Flow cytometric analysis of lymphocytes from the enlarged lymph nodes of MRL/1 mice showed the majority of them to be Lyt-1+, YE1/7.1+, YE1/19.1+, YE1/9.9-. In contrast, YE1/7.1 and YE1/19.1 antibodies did not significantly react with lymphocytes from the congenic MRL/n mice, which lack the lpr gene and do not develop lymphoproliferation. Lymphocytes from younger MRL/l mice, which have almost normal size lymph nodes, were also YE1/7.1-, YE1/19.1-. A small proportion of mitogen-stimulated MRL/n lymph node cells appeared to express these antigens at low densities, but it is not known whether they represent a unique cell population. Among the several lymphoid cell lines tested, only the T lymphoma line EL-4 had the Lyt-1+, YE1/7.1+, YE1/19.1+ phenotype. These results suggest that the lpr mutation induces the expansion of a unique T cell subset.
