Spatial Metabolomic Profiling of Pinelliae Rhizoma from Different Leaf Types Using Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry Imaging.

Pinelliae Rhizoma (PR), a highly esteemed traditional Chinese medicinal herb, is widely applied in clinical settings due to its diverse pharmacological effects, including antitussive, expectorant, antiemetic, sedative-hypnotic, and antitumor activities. Pinellia ternata exhibits morphological variation in its leaves, with types resembling peach, bamboo, and willow leaves. However, the chemical composition differences among the corresponding rhizomes of these leaf phenotypes remain unelucidated. This pioneering research employed Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry Imaging (MALDI-MSI) to conduct the in situ identification and spatial profiling of 35 PR metabolites in PR, comprising 12 alkaloids, 4 organic acids, 12 amino acids, 5 flavonoids, 1 sterol, and 1 anthraquinone. Our findings revealed distinct spatial distribution patterns of secondary metabolites within the rhizome tissues of varying leaf types. Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) effectively differentiated between rhizomes associated with different leaf morphologies. Furthermore, this study identified five potential differential biomarkers-methylophiopogonanone B, inosine, cytidine, adenine, and leucine/isoleucine-that elucidate the biochemical distinctions among leaf types. The precise tissue-specific localization of these secondary metabolites offers compelling insights into the specialized accumulation of bioactive compounds in medicinal plants, thereby enhancing our comprehension of PR's therapeutic potential.

Influencing Factors of Street Vitality in Historic Districts Based on Multisource Data: Evidence from China

Amid urban expansion, historic districts face challenges such as declining vitality and deteriorating spatial quality. Using the streets of Xi’an’s historical and cultural district as examples, this research utilizes multisource data, including points of interest (POIs), street view images, and Baidu heatmaps, alongside analytical techniques such as machine learning. This study explores the determinants of street vitality from the dual perspectives of its external manifestation and spatial carriers. A quantitative framework for measuring street vitality in historic districts is established, thoroughly examining the driving factors behind street vitality. Additionally, the relationship between built environment indicators and street vitality is elucidated through statistical analysis methods. The findings reveal significant, time-varying influences of these spatial carriers on human vitality, with distinct spatial distribution patterns of human activity across different times, and the significance of the influence of external representations of human vitality and various types of spatial carriers varies over time. Based on these insights, this paper proposes strategies for enhancing the vitality of historic streets, aiming to rejuvenate and sustain the diverse and dynamic energy of these districts. It provides a foundation for revitalizing the vigor of cultural heritage zones and offers strategies applicable to similar urban contexts.

Contamination Characteristics and Source Apportionment of Heavy Metal in the Topsoil of a Small Watershed in South Taihang

With the rapid development of industrialization and urbanization, the issue of soil environmental pollution is becoming more and more prominent, especially concerning heavy metal contamination, which has garnered significant scholarly attention. The surface watershed formed by waterline is influenced by various factors such as topography, industrial emissions, and agricultural runoff, resulting in a complex process of migration and accumulation of heavy metal elements from multiple sources. In this study, the pollution characteristics and sources of heavy metal elements Hg, As, Pb, Ni, Cd, Cr, Cu and Zn in 165 surface soil samples from the Manghe River watershed in Jiyuan City were comprehensively analyzed using a variety of methods, including statistics, geostatistics, enriched factor analysis and the Positive Matrix Factorization Model (PMF). The results showed that the concentrations of Hg, Cd, As, Cu, Pb and Zn exceeded their corresponding background values with varying degrees of enrichment. Notably, the average contents of Cd, Hg and Pb were 26.70 times, 3.69 times and 4.49 times higher than those in Chinese soils on average, respectively, showing obvious enrichment characteristics. Moreover, there were distinct spatial distribution patterns for each heavy metal element; Ni and Cr exhibited similar trends mainly controlled by the parent material, while human activities significantly affect the other six elements forming high-value areas around mining and related industries. It is noteworthy that Cu, Hg and Zn were influenced by dominant wind direction in autumn and winter, forming sub-high-value zones in southern forested areas; meanwhile, Cu and Zn were also influenced by agricultural fertilizer application as well as surface runoff, leading to secondary high-value areas in the dryland areas. Further analysis revealed a significant positive correlation among these heavy metal elements, suggesting that they may share common sources. Through the PMF Model, four main factors were identified, with factor 2 (36.25%), factor 1 (23.00%), factor 3 (21.20%) and factor 4 (19.55%) ranked in descending order of contribution rate. The heavy metal pollution in the study area was attributed to anthropogenic activities and natural factors, accounting for 63.75% and 36.25%, respectively. Coal mining, chemical industry smelting, vehicle emissions and excessive use of agrochemicals were identified as the main sources of heavy metal pollution. These pollutants entered the soil through direct emissions, atmospheric deposition, transportation and agricultural activities, exerting a significant impact on the soil environment. Therefore, delving into the spatial distribution pattern of soil heavy metal pollution and precise analysis of its sources are of great importance for effective treatment and remediation of soil heavy metal pollution in small watersheds, maintaining healthy soil ecology and safeguarding human health.

Influence of boundary layer jets on the vertical distribution of ozone in Guangdong, China

The planetary boundary layer (PBL) characteristics during ozone (O3) episodes in China have been extensively studied; however, knowledge of the impact of boundary layer jets (BLJs) on O3 vertical distribution is limited. This study conducted a field campaign from 1 to 8 December 2020 to examine the vertical structure of the O3 concentration and wind velocity within the boundary layer at two sites (Foshan: FS, Maoming: MM) in Guangdong. Utilising lidar observations and the Weather Research and Forecasting model coupled with Chemistry (WRF-Chem), distinct spatial distribution patterns of O3 over FS and MM influenced by BLJs were identified. The BLJs at both locations exhibited pronounced diurnal variations with a nocturnal maximum exceeding 11 m/s at a height of approximately 500 m. The nocturnal enhancement of BLJs resulted from inertial oscillations coupled with diurnal thermal forcing over sloping terrain. A stronger BLJ at FS induced an evident uplift of O3 and the prevailing northeasterly wind facilitated the transport of O3 in the nocturnal residual layer from FS to MM. After sunrise, surface heating and the development of the PBL caused the air mass with elevated O3 levels in the residual layer to descend to ground level. At MM, calm surface winds, a weaker BLJ at 500 m height, and strong downdrafts collectively contributed to a significant increase in surface O3 concentration in subsequent days. These findings contribute to our understanding of the interactions between BLJs and variations in surface air pollutant concentrations, thereby providing important insights for future regional emissions control measures.

Abstract 1150: Unveiling the spatial landscape of tumor and stroma heterogeneity in colorectal cancer with spatial transcriptomics

Abstract Heterogeneity in colorectal cancer (CRC) manifests in the diversity and interactions among tumor cells, support stroma, and immune components, critically impacting patient prognosis and therapeutic efficacy. In this study, spatial transcriptomics (ST, 10x Genomics Visium Spatial Gene Expression) was performed on 40 retrospectively collected CRC samples, along with detailed clinicopathological parameters and long-term clinical outcomes, with the goal of unraveling the molecular topography of the tumor and surrounding stromal compartments, and associating their spatial interactions to clinical outcomes. Five primary RNA groups were identified through unsupervised clustering on ST data, correlating with tissue classes detected in H&E-stained images via a pre-trained deep learning model. Markers and pathways specific to these tissues were identified. Furthermore, immune cell infiltration in these regions was deconvoluted using reference single-cell RNA sequencing (scRNA-seq) data. Sub-clustering within the tumor groups revealed five major tumor subtypes, each characterized by a distinct set of differentially expressed marker genes. Enrichment analysis across these subtypes highlighted distinct activation and suppression in pathways including KRAS, p53, MAPK, and Integrin. Tumors exhibited intra- and inter-tumoral heterogeneity. Location-specific tumor subtypes were observed, with those in right-sided CRC displaying increased immune cell infiltration, linked to enriched pathways such as Type II Interferon Signaling and Cytokine Signaling. Conversely, left-sided CRC subtypes had reduced immune cell presence, and were characterized by enhanced pathways related to vascular development, Hypoxia, and p53 Signaling. Inferred clonal lineages from copy number variation (CNV) using ST suggested a spatially structured phylogenetic organization corresponding to transcriptional clusters. Additionally, the ST data unveiled distinct spatial distribution patterns across various cancer-associated fibroblast (CAF) phenotypes, such as matrix, inflammatory, vessel-associated, tumor-promoting, and antigen-presenting CAFs. Transcriptional patterns uncovered through scRNA-seq, including epithelial-mesenchymal transition (EMT) signatures and consensus molecular subtypes (CMS), displayed unique spatial layouts within the tumor microenvironment. To our knowledge, this is the most comprehensive study to date on spatial transcriptomic heterogeneity within CRC patients, revealing distinct tumor, stroma and immune compositions and spatial structures. Evaluation, Driving Mechanism and Spatial Correlation Analysis of Atmospheric Environmental Efficiency in the “2+26” Cities Based on the Nonradial MEA Model

The “2+26” cities are 26 cities in Beijing, Tianjin and the surrounding cities, constituting a region characterized by economic prosperity and diverse industries but plagued by severe atmospheric pollution. As a focal area for atmospheric pollution control, a scientific assessment of atmospheric environmental efficiency in the “2+26” cities that measures the degree of coordination between the economy and air pollution is very important for winning the battle of blue sky defense. Based on this, this study comprehensively used the nonradial multi-directional efficiency analysis (MEA) model, Global Reference Malmquist Model and spatial correlation analysis to evaluate the atmospheric environmental efficiency, calculate the driving factors and explore the spatiotemporal distribution characteristics of the “2+26” cities from 2009 to 2018. The research findings indicate the following: (1) Atmospheric environmental efficiency showed a trend of first decreasing and then increasing, with a significant improvement potential of 26.7% in the future. (2) There was a significant discrepancy between the best- and worst-performing cities, with the best being 0.910 and the worst being 0.573, demonstrating imbalanced development between cities. The relatively low-efficiency cities were mainly located in Hebei, Shanxi and Henan provinces. (3) A value of technological efficiency change (EC) less than 1 was the main restrictive factor for improving atmospheric environmental efficiency, whereas a value of technological change (TC) greater than 1 enhanced it. (4) The atmospheric environmental efficiency presented a distinct spatial distribution pattern of high–high and low–low aggregation, forming high-value areas centered in the Beijing–Tianjin region and along the Zibo–Zhengzhou line. The western and central regions were relatively low, whereas the northern and eastern regions were relatively high, with significant regional differences in spatial distribution. The conclusions from this article’s empirical analysis can help concerned developing countries determine key factors to improve their atmospheric environmental efficiency and then formulate policies for sustainable economic and environmental development.

Aerodynamic Analysis of the Opening Hood Structures at Exits of High-Speed Railway Tunnels

As train operating speeds increase, the aerodynamic characteristics of the train within the tunnel become more pronounced, and effectively addressing the issue of micro-pressure wave (MPW) over-limits becomes especially crucial. This paper utilized the control volume method to investigate the key influencing parameters of tunnel exit hoods on the mitigation effectiveness of MPWs. Additionally, numerical simulation methods were used to validate these crucial parameters. The analysis considered various opening ratios, different opening forms, and the influence of hoods at tunnel entrances and exits on the amplitude and spatial distribution patterns of MPWs. A design methodology that comprehensively takes into account the advantages of tunnel entrance and exit hoods was proposed. The results showed that a higher opening ratio of tunnel exit hoods led to lower MPW amplitudes. Compared to without opening in the hood, when the opening ratio of the exit hood reached 90%, the maximum amplitude of MPWs at a distance of 20 m from the hood outlet decreased by 48.7%. Various opening forms of exit hoods resulted in distinct spatial distribution patterns of MPW amplitudes, with amplitudes near the openings notably higher than in other areas. There were differences in the mitigation mechanisms between entrance and exit hoods. In comparison to entrance hoods, exit hoods exhibited higher mitigation efficiency within a specific range of MPW amplitudes. Additionally, when both entrance and exit hoods were installed, they achieved the most effective mitigation of MPWs.

Analysis of Water Conservation Trends and Drivers in an Alpine Region: A Case Study of the Qilian Mountains

The water conservation service of an ecosystem reflects the sustainability of regional water resources and is significant to human survival and sustainable development. However, global climate warming and intensified human activities pose substantial challenges to regional water conservation services, especially in an alpine region with a fragile ecological environment, which is more sensitive to climate factors and land use pattern changes. In this study, the Qilian Mountains (QLM) region was chosen as the study area to investigate water conservation trends and drivers in an alpine region. The InVEST model was used to estimate water conservation in the QLM from 2000 to 2020. In addition, the characteristics of the spatiotemporal variation in the water conservation were analyzed using a combination of the Theil–Sen median trend and Mann–Kendall method, coefficient of variation, and Hurst exponent, and the main driving factors affecting these changes were determined using partial correlation analysis and contribution analysis. The main conclusions are as follows: (1) The predicted water conservation in the QLM based on the InVEST model’s water yield module had a relative inaccuracy of 5.96%, and the mean water conservation in the QLM from 2000 to 2020 was approximately 78.08 × 108 m3. (2) The water conservation showed a slight increase over the whole QLM region, with a change rate of 0.565 mm/a; yet, it showed a distinct spatial distribution pattern characterized by “more in the east than in the west”. (3) The contribution of the various land use categories to the total water conservation, from highest to lowest, was according to the following: grassland (62.44%) > unutilized land (15.99%) > forest (11.44%) > cultivated land (9.86%) > construction land (0.45%) > water (0.03%). (4) Precipitation exhibited a significant positive correlation, with contribution ratios of approximately 58.50% to the variation in the water content, whereas potential evapotranspiration and surface temperature showed a nonsignificant negative correlation with contribution ratios of approximately 2.17% and 2.08%, respectively. The results can provide scientific reference for ecological protection in the QLM and other similar alpine environment areas.

Impact of microplastics on the in situ, high-resolution of key nutrient dynamics at the soil-water interface in rice fields

Introduction: Microplastics are characterized by their small size, widespread distribution, and durability, present a significant environmental risk. Despite their omnipresence in terrestrial and aquatic systems, the potential consequences on nutrient cycling remain under-investigated. Microplastics have emerged as a focal point of current research, presenting both a challenge and a frontier in environmental science.Methods: This study explores the effects of microplastics on the high-resolution, in situ distribution and exchange dynamics of key nutrients, nitrogen (N) and phosphorus (P), at the soil-water interface in rice paddies, utilizing the Diffusive Gradients in Thin-films (DGT) technique.Results: Our results reveal distinct spatial distribution patterns for N and P across the soil-water interface. Labile phosphorus (P) concentrations were significantly higher in the soil than in the overlying water, whereas DGT-NO3− concentrations exhibited the inverse trend. Different microplastic concentrations notably impacted DGT-NO3− (P = 0.022) and DGT-NH4+ (P = 0.033), with an increase between 27.79% and 150.68%. Moreover, different particle sizes significantly influenced NH4+. Interestingly, paddy soil acted as a “source” for labile P and a “sink” for NH4+ and NO3−.Discussion: These insights provide valuable insights into the interactions between microplastics and nutrient cycles at the soil-water interface, and assess the effects on nutrient migration and transformation. The outcomes of this study will contribute to an improved understanding of the broader ecological implications of microplastic pollution in agricultural settings. It will also provide a foundation for the development of strategies to manage and mitigate the impacts of microplastic pollution in agricultural soils, particularly in rice dominated agroecosystems.

Abstract 4483: Distinct spatial distribution patterns of ALK-inhibitor naïve versus ALK-inhibitor treated ALK-positive NSCLC brain metastases

Abstract Background: Non-small cell lung cancer (NSCLC) with anaplastic lymphoma kinase rearrangement (ALK+) has a high affinity to form brain metastases (BrM). The cumulative incidence of BrMs in ALK+ lung cancer is over 50%, despite highly effective ALK tyrosine kinase inhibitors (TKIs) with CNS activity. Pharmacokinetic (PK) data from other CNS-active lung cancer TKIs (e.g., osimertinib) have revealed major brain white vs. gray matter drug concentration differences, raising the possibility of a PK-driven effect on BrM formation and response. This study aims to compare the size and distribution of ALK+ NSCLC BrMs at diagnosis in a TKI-naïve and TKI-exposed cohort. Methods: We retrospectively reviewed brain MRIs from the date of BrM diagnosis for patients with ALK+ NSCLC at Johns Hopkins. Demographic and clinical information were collected by chart review. Each tumor was marked in a standard space brain model in the corresponding anatomic location represented by a sphere of corresponding diameter using 3D Slicer 4.11. FreeSurfer white-gray matter atlases were used to assess BrM distribution. The data for patients who were on TKI vs TKI-naïve at the time of BrM diagnosis were then analyzed separately. T-tests were used to compare the metastatic burden (sum of BrM diameters), mean BrM diameter per patient, number of BM per patient, per individual mean of white matter exclusive (defined as no overlap with gray matter) and deep white matter (≥5mm away from gray matter) BrMs between patient groups. Results: 429 BrMs were identified in 39 patients, with 25 patients being TKI-naïve at the time of BrM diagnosis while 14 patients were on TKI therapy. TKI-exposed patients had significantly smaller BrM diameters than those in the TKI-naïve group (6.1±3.8 vs 10.2±5.5mm, p=0.02). While metastatic burden was very similar between the groups, the mean number of BrM per patient was numerically higher in the TKI-exposed group (10.6±11.9 vs 6.2±9.5; p=0.22). Notably, patients in the TKI-exposed group also had higher numbers of white matter exclusive (3.5±4.4 vs 1.4±2.0, p=0.05) and deep white matter metastases (3.2±4.3 vs 1.3±2.0, p=0.06) than those who were TKI-naïve. Conclusion: Our data highlight the differences in BrM characteristics among ALK+ NSCLC exposed to ALK TKI. TKI therapy was associated with similar BrM burden but smaller individual lesions that were more likely to be exclusive to the white matter where drug concentrations might be significantly lower. These findings suggest that suboptimal drug CNS distribution in the white matter may underly brain progression of ALK+ NSCLC despite TKI therapy. Abstract 6763: Spatial profiling of cancer-associated fibroblasts in non-small cell lung cancer

Abstract Cancer-associated fibroblast (CAF) is an unique component of the tumor microenviroment (TME). CAFs heterogeneous have been identified by recent single-cell studies. However, their spatial distribution, orgnization patterns and neighbors remain incompletely understood. Deep profiling of CAFs in tissue context may provides new insights into improved understanding of their functions and identify targets for the development of CAF-targeted therapies.We obtained the open-source CosMx SMI dataset generated on 8 tissue sections from 5 non-small-cell lung cancer (NSCLC) tissues (He, S. et al. Nat Biotechnol. 2022). We extracted the cells’ spatial locations and transcriptome data for subsequent analyses. Quanlity filtering, data normalization, batch correction and clustering analysis was performed by Seurat package. Cell type was defined based on canonical immune markers and curated gene sets. In addition, we applied NMF clustering approach to define the spatial subtypes of CAFs based on the composition of neighboring cells. After that, Four spatial subtypes of CAFs were defined, exhibiting distinct spatial distribution patterns and compositions of neiboring cells, including CAFs that are clustered with tumor cells, CAFs localized within stromal, CAFs co-localized with B cells and T cells within the tertiary lymphoid structures, and CAFs localized within myeloid-enriched areas. Notably, we observed an intimate link between their spatial location and tarnscriptioanl states, i.e., these different spatial CAF subtypes showed clearly distinct expression profiles, may suggest their heterogeneous functions in the tumor microenviroment. For example, tumor-enriched CAFs showed high expression of contractile (e.g., ACTA2, TAGLN) and extracellular matrix (COL1A1) related genes, resembling myofibroblast (myCAFs), whereas the TLS-niches CAFs showed high expression of genes involved in antigen presentation such as major histocompatibility complex class II molecules and CD74, suggesting their antigen presentation function and were identified as apCAFs. Consistently, we observed high expression of inflammation related genes such as interleukin and PDGFRA in the myeloid-enriched CAFs suggesting an inflammatory phenotype (iCAFs). In addition, these CAFs in different spatial subtypes showed differential expression of molecular pathways regulating stress and inflammation response, likely reflecting their complex interactions with other components of the tunmor microenvironment. We also included single-cell RNA sequencing (scRNA-seq) and spatially resolved whole transcriptomics datasets for cross validation of our novel discoveries in additional cohorts.In summary, by applying a novel approach to classify CAFs in the spatial context, this study provided novel insights into an improved understanding of the phenotypic and functional heterogeneity of CAFs in the tunmor microenvironment of NSCLC. Baleen whales in the Scotia Sea during January and February 2003

Different species of baleen whales display distinct spatial distribution patterns in the Scotia Sea during the austral summer. Passive acoustic and visual surveys for baleen whales were conducted aboard the RRS James Clark Ross in the Scotia Sea and around South Georgia in January and February 2003. Identified calls from four species were recorded during the acoustic survey including southern right (Eubalaena australis), blue (Balaenoptera musculus), fin (B. physalus) and humpback whales (Megaptera novaeangliae). These acoustic data included up calls made by southern right whales, downswept D and tonal calls by blue whales, two possible types of fin whale downswept calls and humpback whale moans and grunts. Visual detections included southern right, fin, humpback and Antarctic minke whales (B. bonaerensis sp.). Most acoustic and visual detections occurred either around South Georgia (southern right and humpback whales) or south of the southern boundary of the Antarctic Circumpolar Current (ACC) and along the outer edge of the ice pack (southern right, blue, humpback and Antarctic minke whales). Fin whales were the exception, being the only species acoustically and visually detected primarily in the central Scotia Sea, along the southern ACC front. In addition to identifiable calls from these species, two types of probable baleen whale calls were detected: 50Hz upswept and pulsing calls. It is proposed that minke whales may produce the pulsing calls, based on their similarities with minke whale calls recorded in the North Atlantic Ocean. There was an overlap between locations of fin whale sightings and recordings and locations of 50Hz upswept calls in the central Scotia Sea, but these calls were most similar to calls attributed to blue whales in other parts of Antarctica. More study is required to determine if baleen whales produce these two call types, and if so, which species. The efficiency of acoustics and visual surveys varied by species, with blue whales being easier to detect using acoustics, Antarctic minke whales being best detected during visual surveys and other species falling in between these two extremes.

Genetic and Morphological Variation of Belgian Cyperus esculentus L. Clonal Populations and Their Significance for Integrated Management

Cyperus esculentus is an invasive troublesome neophyte in many arable crops across the globe. Analysis of the genetic and morphological profile of local C. esculentus clonal populations may be useful in explaining differential herbicide sensitivity found among distinct clonal populations and spatial distribution patterns. In this study, 35 Belgian C. esculentus clonal populations, evenly spread across the entire infestation area (30,689 km2) and covering a great diversity of farm and soil types, and hydrological and environmental conditions, were genetically characterized using amplified fragment length polymorphism (AFLP) profiling. These clonal populations were also grouped into different morphological clusters using data from shoot, tuber, and inflorescence number, fresh tuber biomass, individual fresh tuber weight, and number of germinable seeds collected in three consecutive years. Of the 271 AFLP markers, 207 were polymorphic. The gene diversity among clonal populations was 0.331 and three genetically distinct clusters were identified. Depending on observation year, clonal populations were grouped in four to five morphologically distinct clusters that closely aligned with the genetic clusters. The genetically distinct clusters differed in their geographical distribution pattern and range as well as in their morphological characteristics. Clonal populations belonging to clusters with broad distribution ranges produced numerous viable seeds. Clusters with clonal populations that produced large tubers were less widespread than the cluster with clonal populations producing many small tubers. The results suggest that tuber size, tuber number, and fecundity may all play an important role in the spread of C. esculentus. Morphotyping may be very useful in designing effective preventive and curative C. esculentus management strategies.

Pemanfaatan Citra Sentinel untuk Pemetaan Perubahan Kerapatan Vegetasi di Desa Jambudipa Kecamatan Cisarua

Vegetation is a group of plants, or all plant species found in a given area, with distinct patterns of temporal and spatial distribution. The presence of plants in a certain area helps to maintain the balance of the ecosystem. This study aims to determine the level of vegetation density in Jambudipa Village, Cisarua District, Bandung Regency in 2019 and 2022 using the Resistant Vegetation and Atmospheric Index (MSARVI) method. In 2019 there is still a lot of vegetation, but in 2022 the density will decrease. The amount of vegetation decreases due to several factors, such as the construction of settlements and public facilities such as urban developments and public services, which contribute to the decline in vegetation.

Mass spectrometry-based profiling and imaging strategy, a fit-for-purpose tool for unveiling the transformations of ginsenosides in Panax notoginseng during processing

BackgroundPanax notoginseng, a valuable medicinal plant, is traditionally used to treat trauma, body pain, and cardiovascular diseases in two clinical forms including raw (crude) and processed form. Processing-triggered compound transformation is responsible for the distinct bioactivity between raw and processed Panax notoginseng. Nevertheless, investigating the chemical diversity and dynamic transformation pattern of processed Panax notoginseng is challenging. MethodsA new approach, which integrates multi-components characterization, processing trajectory depiction, discovery of differential markers, transformation mechanism of metabolites, in situ spatial distribution and transformation of metabolites, was established to elucidate the role of processing on the holistic chemical transformations of Panax notoginseng (PN). ResultsIn this study, 136 ginsenosides (mainly rare ginsenosides) were identified or tentatively characterized and the temperature-dependent chemical variation trajectory was depicted via principal component analysis (PCA). Nineteen processing-associated markers were confirmed by orthogonal partial least squares-discriminant analysis (OPLS-DA). For the first time, the transformation pathway of ginsenosides during processing were elucidated by integrating the precursor ion scan (PIS) and mimic processing strategy that involves with deglycosylation, dehydration, hydration, acetylation, and isomerization. Results of mass spectrometry imaging (MSI) revealed the major ginsenosides M-Rb1, R1, Rg1, Rb1, Rd, and Re exhibited distinct spatial distribution pattern that are highly abundant in the xylem and showed a downward trend during processing. We firstly depicted the spatial distribution of processing-triggered rare ginsenosides (Rg3, Rk1, Rg5, etc.), and in situ transformation of ginsenosides was discovered in the process of steaming. Additionally, this variation trend was consistent with untargeted metabolomics results. ConclusionThis study comprehensively revealed chemical diversity and dynamic transformation pattern and depicted the spatial distribution of ginsenosides of PN during processing. It could provide a clue for the distinct bioactivities between raw and processed PN and elucidate the role of processing on the holistic chemical transformations of natural products, more importantly, the proposed strategy is valuable for the quality evaluation and control of the processing of natural product.

Spatial and temporal variations of rainfall erosivity in the middle Yellow River Basin based on hourly rainfall data

Understanding spatial and temporal variations in rainfall erosivity is essential for estimating the variability in soil erosion. However, rainfall erosivity in the middle Yellow River Basin is usually calculated based on daily rainfall data, which fails to consider the impact of the changing rainfall intensity at the subdaily scale. In this study, we collected hourly rainfall data from 355 meteorological stations within and around the region to calculate rainfall erosivity during 1960–2017. We found that the rainfall erosivity calculated based on hourly data had much larger variability than that calculated based on daily data, and the calculated spatial and temporal variability were substantially different in periods when the rainfall intensity was highly variable. Based on our new estimation, we found that the spatial distribution of rainfall erosivity was highly heterogeneous, with high erosivity values in the Loess Regions and Rocky Mountain Regions with high soil erodibility and low erosivity values in the Aeolian Regions and Soft Sandstone Regions with low soil erodibility. This heterogeneity was mainly due to the distinct spatial distribution patterns of rainfall amount and rainfall intensity. Although the long-term increasing trend of rainfall erosivity was statistically insignificant from 1960 to 2017, its rate of increase was higher than that of the annual rainfall amount. Decadal changes showed a slight decrease from the 1960 s to the 1980 s and an increase since the 1990 s, which was substantially different from the decadal changes in the annual rainfall amount. These differences in temporal variability were mainly caused by the contribution of changing rainfall intensity, which was greater in the Aeolian Regions and Soft Sandstone Regions than in the other subregions. Our study highlighted the importance of considering the changing rainfall intensity at a subdaily scale in semiarid and semihumid climate zones and provided the first estimation of the long-term variability in rainfall erosivity based on hourly rainfall data in this region.

Unraveling microbe-mediated degradation of lignin and lignin-derived aromatic fragments in the Pearl River Estuary sediments

Estuaries are one of the most crucial areas for the transformation and burial of terrestrial organic carbon (TerrOC), playing an important role in the global carbon cycle. While the transformation and degradation of TerrOC are mainly driven by microorganisms, the specific taxa and degradation processes involved remain largely unknown in estuaries. We collected surface sediments from 14 stations along the longitudinal section of the Pearl River Estuary (PRE), P. R. China. By combining analytical chemistry, metagenomics, and bioinformatics methods, we analyzed composition, source and degradation pathways of lignin/lignin-derived aromatic fragments and their potential decomposers in these samples. A diversity of bacterial and archaeal taxa, mostly those from Proteobacteria (Deltaproteobacteria, Gammaproteobacteria etc.), including some lineages (e.g., Nitrospria, Polyangia, Tectomicrobia_uc) not previously implicated in lignin degradation, were identified as potential polymeric lignin or its aromatic fragments degraders. The abundance of lignin degradation pathways genes exhibited distinct spatial distribution patterns with the area adjacent to the outlet of Modaomen as a potential degradation hot zone and the Syringyl lignin fragments, 3,4-PDOG, and 4,5-PDOG pathways as the primary potential lignin aromatic fragments degradation processes. Notably, the abundance of ferulic acid metabolic pathway genes exhibited significant correlations with degree of lignin oxidation and demethylation/demethoxylization and vegetation source. Additionally, the abundance of 2,3-PDOG degradation pathways genes also showed a positive significant correlation with degree of lignin oxidation. Our study provides a meaningful insight into the microbial ecology of TerrOC degradation in the estuary.

Analysis of Spatial and Temporal Distribution of Purinergic P2 Receptors in the Mouse Hippocampus

ATP and other nucleotides are important glio-/neurotransmitters in the central nervous system. They bind to purinergic P2X and P2Y receptors that are ubiquitously expressed in various brain regions modulating various physiological and pathophysiological processes. P2X receptors are ligand-gated ion channels mediating excitatory postsynaptic responses whereas P2Y receptors are G protein-coupled receptors mediating slow synaptic transmission. A variety of P2X and P2Y subtypes with distinct neuroanatomical localization provide the basis for a high diversity in their function. There is increasing evidence that P2 receptor signaling plays a prominent role in learning and memory and thus, in hippocampal neuronal plasticity. Learning and memory are time-of-day-dependent. Moreover, extracellular ATP shows a diurnal rhythm in rodents. However, it is not known whether P2 receptors have a temporal variation in the hippocampus. This study provides a detailed systematic analysis on spatial and temporal distribution of P2 in the mouse hippocampus. We found distinct spatial and temporal distribution patterns of the P2 receptors in different hippocampal layers. The temporal distribution of P2 receptors can be segregated into two large time domains, the early to mid-day and the mid to late night. This study provides an important basis for understanding dynamic P2 purinergic signaling in the hippocampal glia/neuronal network.

Bioinspired adhesive and tumor microenvironment responsive nanoMOFs assembled 3D-printed scaffold for anti-tumor therapy and bone regeneration

Tumor-induced bone loss is the main reason causing bone tumor therapy failure. Rational design of implants with both anti-tumor and bone tissue regeneration functions is urgently needed. This study presents a 3D-printed implant that simultaneously releases anti-cancer drugs and growth factors for anti-tumor therapy and osteogenesis. Such an implant was realized by alternatively assembling polydopamine (PDA)-hybridized nanosized zeolitic imidazolate framework-8 (pZIF-8 nanoMOFs) and PDA-decorated-hydroxyapatite nanoparticles (pHA NPs) on the surfaces of the 3D-printed gelatin-based scaffolds through PDA-assisted layer-by-layer (LbL) assembly strategy. The synthesis of the pZIF-8 nanoMOFs was based on mussel-inspired catechol chemistry, which endowed the nanoMOFs with versatile adhesiveness, high drug loading efficiency, good physiological stability, and tumor environment-sensitive degradability. By using the pZIF-8 nanoMOFs as drug nanocarriers, it was possible to define the distinct spatial distribution and environmental-adaptive release patterns for BMP-2 and cisplatin from the scaffold. In vitro and in vivo studies confirmed that the scaffold possessed good osteoinductivity to induce osteogenic differentiation and to promote new bone formation. By responding to stimuli in the tumor microenvironment, the scaffolds efficiently released cisplatin and inhibited tumor growth. In short, this PDA-hybridized nanoMOF offers a new avenue to functionalize biomaterials with smart and responsive therapeutic ability for diverse biomedical applications.

Spatial analysis of the incidence of Dengue, Zika and Chikungunya and socioeconomic determinants in the city of Rio de Janeiro, Brazil.

In 2015-2016, simultaneous circulation of dengue, Zika and chikungunya in the municipality of Rio de Janeiro (Brazil) was reported. We conducted an ecological study to analyse the spatial distribution of dengue, Zika and chikungunya cases and to investigate socioeconomic factors associated with individual and combined disease incidence in 2015-2016. We then constructed thematic maps and analysed the bivariate global Moran indices. Classical and spatial models were used. A distinct spatial distribution pattern for dengue, Zika and chikungunya was identified in the municipality of Rio de Janeiro. The bivariate global Moran indices (P < 0.05) revealed negative spatial correlations between rates of dengue, Zika, chikungunya and combined arboviruses incidence and social development index and mean income. The regression models (P < 0.05) identified a negative relationship between mean income and each of these rates and between sewage and Zika incidence rates, as well as a positive relationship between urban areas and chikungunya incidence rates. In our study, spatial analysis techniques helped to identify high-risk and social determinants at the local level for the three arboviruses. Our findings may aid in backing effective interventions for the prevention and control of epidemics of these diseases.
